Echinococcus granulosus, the aetiological agent of cystic hydatid disease, exists as a series of strains or genotypes which differ in biological features. Pig strain (G7 genotype) has been shown to differ from sheep strain (G1 genotype) in phenotypical characters such as intermediate host range, geographical distribution and rate of development of the adult worm. Since in vivo studies of different parasite genotypes can provide insights into host–parasite relationship we analysed for the first time the behaviour of E. granulosus G7 genotype protoscoleces in the murine experimental model. Our results show that G7 protoscoleces were unable to establish a regular infection in mice in contrast to G1 protoscoleces which developed intraperitoneal hydatid cysts. This inability was observed in co-infection experiments, i.e. even in the presence of a controlled immune response that allows G1 genotype protoscoleces establishment. In addition, the implantation of in vitro obtained E. granulosus G7 genotype microcysts resulted in a low percentage of hydatid cysts establishment. These results show a difference in the biological ability of both E. granulosus strains to develop secondary hydatid cysts in mice. We suggest that the comparison of infective and non infective genotypes of E. granulosus in the experimental host can be regarded as a new model to study the mechanisms of infection of Echinococcus spp. This knowledge could provide helpful information for the development of therapies, drugs and/or vaccines against cystic hydatid disease. The Piperaceae family is represented by herbaceous plants, shrubs, and, more rarely, trees. Among the aromatic varieties of this genus, Piper hispidinervum C. DC. stands out. In this work, volatile and non‐volatile extracts from P. hispidinervum were obtained through the application of sequential extraction methods. The results of steam distillation experiments indicated that optimal conditions were achieved when the extractions were conducted at 101 kPa and an initial bed porosity of 0.91. Chemical analysis showed that safrole is the major component present in the essential oil. NMR analysis revealed the presence of 6‐methoxy eugenol for P. hispidinervum extracts obtained by supercritical fluid extraction using water as co‐solvent at 20 MPa and 313.15 K. Mathematical models adjusted well to the experimental data in all conditions investigated. The percentage of respondents who gave affirmative responses for each of the items of the ELCSA was determined. Exploratory factor analysis was carried out to determine the ELCSA’s factor structure. A probit model was used to determine the impact of some individual and household sociodemographic characteristics on the identified dimensions of food insecurity. Metropolitan area centred on Montevideo, the capital city of Uruguay, April–September 2014. Adults aged between 18 and 93 years (n 742). The percentage of affirmative responses to the items of the ELCSA ranged from 4·4 to 31·7 %. Two factors were identified in the exploratory factor analysis performed on data from households without children under 18 years old, whereas three factors were identified for households with children. The identified factors were associated with different severity levels of food insecurity. Likelihood of experiencing different levels of food insecurity was affected by individual characteristics of the respondent as well as characteristics of the household. The influence of sociodemographic variables varied among the ELCSA dimensions. Household income had the largest influence on all dimensions, which indicates a strong relationship between income and food insecurity. Persicaria maculosa (Polygonaceae) (known as lady’s thumb) is an annual morphologically variable weed that is widely distributed in Chile. The purpose of this study was to investigate the antifeedant potential of methanolic (MeOH), ethanolic (EtOH), and dichloromethane (DCM) extracts from the aerial parts of this plant collected in the Valparaíso and Curicó provinces (Chile) and relate this activity to the antioxidant capacity and the presence of phenolic compounds in the extracts. When developing new products to be used in honeybee colonies, further than acute toxicity, it is imperative to perform an assessment of risks, including various sublethal effects. The long-term sublethal effects of xenobiotics on honeybees, more specifically of acaricides used in honeybee hives, have been scarcely studied, particularly so in the case of essential oils and their components. In this work, chronic effects of the ingestion of Eupatorium buniifolium (Asteraceae) essential oil were studied on nurse honeybees using laboratory assays. Survival, food consumption, and the effect on the composition of cuticular hydrocarbons (CHC) were assessed. CHC were chosen due to their key role as pheromones involved in honeybee social recognition. While food consumption and survival were not affected by the consumption of the essential oil, CHC amounts and profiles showed dose-dependent changes. All groups of CHC (linear and branched alkanes, alkenes and alkadienes) were altered when honeybees were fed with the highest essential oil dose tested (6000 ppm). The compounds that significantly varied include n-docosane, n-tricosane, n-tetracosane, ntriacontane, n-tritriacontane, 9-tricosene, 7-pentacosene, 9-pentacosene, 9-heptacosene, tritriacontene, pentacosadiene, hentriacontadiene, tritriacontadiene and all methyl alkanes. All of them but pentacosadiene were up-regulated. On the other hand, CHC profiles were similar in healthy and Nosema-infected honeybees when diets included the essential oil at 300 and 3000 ppm. Our results show that the ingestion of an essential oil can impact CHC and that the effect is dose-dependent. Changes in CHC could affect the signaling process mediated by these pheromonal compounds. Besides, xanthoxylin was also<br />found in the aerial parts of S. schottiana and the honeydew excreted by E. cestri feeding on this tree. A larva feeding assay<br />where xanthoxylin-enriched diets were offered to 1st<br />instar larvae showed that larvae died in the same proportion as larvae did when offered enriched diets with nectars from RD-colonies. These findings demonstrate that a xenobiotic can mimic the RD syndrome in honeybee larvae and provide evidence of an interspecific flow of xanthoxylin among three trophic levels. Here we devised a platform for high throughput identification of nanobodies to cell receptor based on the use of a biotin handle.<br />Methods<br /><br />Using a biotin-acceptor peptide tag, the in vivo biotinylation of nanobodies in 96 well culture blocks was optimized allowing their parallel analysis by flow cytometry and ELISA, and their direct use for pull-down/MS target identification.<br />Results<br /><br />The potential of this strategy was demonstrated by the selection and characterization of panels of nanobodies to Mac-1 (CD11b/CD18), MHC II and the mouse Ly-5 leukocyte common antigen (CD45) receptors, from a VHH library obtained from a llama immunized with mouse bone marrow derived dendritic cells. Recombinant single domain antibodies (nanobodies) constitute an attractive alternative for the production of neutralizing therapeutic agents. Their small size warrants rapid bioavailability and fast penetration to sites of toxin uptake, but also rapid renal clearance, which negatively affects their performance. In this work, we present a new strategy to drastically improve the neutralizing potency of single domain antibodies based on their fusion to a second nanobody specific for the complement receptor CD11b/CD18 (Mac-1). These bispecific antibodies retain a small size (~30 kDa), but acquire effector functions that promote the elimination of the toxin-immunocomplexes. The principle was demonstrated in a mouse model of lethal toxicity with tetanus toxin. Three anti-tetanus toxin nanobodies were selected and characterized in terms of overlapping epitopes and inhibition of toxin binding to neuron gangliosides. Bispecific constructs of the most promising monodomain antibodies were built using anti Mac-1, CD45 and MHC II nanobodies. When co-administered with the toxin, all bispecific antibodies showed higher toxin-neutralizing capacity than the monomeric ones, but only their fusion to the anti-endocytic receptor Mac-1 nanobody allowed the mice to survive a 10-fold lethal dose. In a model of delayed neutralization of the toxin, the anti- Mac-1 bispecific antibodies outperformed a sheep anti-toxin polyclonal IgG that had shown similar neutralization potency in the co-administration experiments. This strategy should have widespread application in the development of nanobody-based neutralizing therapeutics, which can be produced economically and more safely than conventional antisera. Además de los anticuerpos convencionales, los camélidos poseen anticuerpos carentes de cadena liviana en los que un único dominio forma el sitio de unión al antígeno. Este dominio, denominado Nanobody o VHH, es uno de los fragmentos de reconocimiento de antígeno más pequeños conocidos en la naturaleza (~15 kDa) y se caracterizan por su alta estabilidad, solubilidad y especificidad. Estas características únicas de los VHHs junto a su facilidad de manipulación mediante biología molecular convierten a los VHH en una prometedora alternativa a los anticuerpos convencionales o fragmentos de anticuerpo para numerosas aplicaciones biotecnológicas. Este trabajo de tesis tuvo como horizonte establecer tecnología local para la generación de bibliotecas de nanobodies en fagos filamentosos a partir de linfocitos B de llamas (Lama glama). Asimismo se desarrollaron metodologías que facilitan la selección y caracterización de los nanobodies de interés contra una variedad de antígenos que van desde pequeñas moléculas hasta complejas estructuras celulares. La funcionalidad de los nanobodies seleccionados se demostró en diferentes aplicaciones biotecnológicas. En la primera parte del trabajo se presenta el desarrollo de un vector de alto nivel de expresión y un sistema de biotinilacion in vivo de los nanobodies que facilita el proceso de screening y caracterización paralela de cientos de clones en forma sistematizada y rápida. El sistema tiene gran versatilidad y facilita el estudio de la reactividad de los nanobodies mediante citometría de flujo, ELISA, microscopía, inmunoprecipitación, etc. Como primer aplicación de este sistema se desarrolló un método de ranqueo de afinidad relativa por el antígeno de la población de nanobodies aislados en la etapa de selección inicial (panning) y un sistema que permite la búsqueda de pares de nanobodies para la detección del antígeno en inmunoensayos tipo sándwich, lo que permitió el desarrollo de un ELISA ultrasensible para la cuantificación de la epoxi-hidrolasa humana. 2 En la segunda parte del trabajo se describe la preparación de una biblioteca de nanobodies contra células dendríticas murinas, y la adaptación de la plataforma de descubrimiento de nanobodies para aislar anticuerpos contra receptores celulares nativos utilizando células enteras para las etapas de aislamiento y caracterización. De esta forma se aislaron anticuerpos contra el componente CD11b del receptor Mac-1, la región no polimórfica del MHC-II y contra el receptor CD45, constituyendo un desarrollo de gran relevancia debido a que, a diferencia de los anticuerpos convencionales, los nanobodies pierden drásticamente la reactividad contra su antígeno ante pequeños cambios conformacionales. Debido a eso los nanobodies seleccionados contra receptores recombinantes inmovilizados sobre poliestireno (placa de ELISA) muy raramente reaccionan en forma cruzada con el receptor nativo en las células. A pesar de esto último, y dada la importancia de dirigir la selección utilizando receptores recombinantes, se mostró que la cuidadosa elección de la forma de inmovilización del receptor recombinante, sí permite aislar nanobodies contra receptores nativos. Esto fue demostrado seleccionando nanobodies contra la cadena CD11c del receptor CDR4 que es un marcador de células dendríticas. Por último se realizó una prueba de concepto sobre la utilidad de los nanobodies como dominios de funcionalización, en este caso para conferir funciones efectoras a nanobodies con capacidad de neutralizar la toxina tetánica. Una vez aislados los nanobodies anti-toxina, se prepararon anticuerpos biespecíficos fusionándolos a los nanobodies anti- CD11b, MHCII y CD45, y se estudió su poder de neutralización y biodistribución in vivo, encontrando un dramático incremento del poder neutralizante cuando los anticuerpos neutralizantes se funcionalizaron con el nanobody anti-CD11b (que reacciona con un receptor endocítico), pero no con el anti-MHC-II o anti-CD45. Two different Echinococcus granulosus antigen B subunits (AgB8:1 and AgB8:2) were characterized and the structure of the genes encoding these two proteins were compared. DNA sequences were expressed in Escherichia coli and the antigens’ diagnostic value was then assessed. The genomic sequence of AgB8:1 has a 92 bp intron in the position corresponding to amino acid 16; the AgB8:2 genomic sequence presents a 68 bp intron in the position corresponding to amino acid 20. Both introns are located between the putative N-terminal hydrophobic sequence and the secreted peptide. A comparison between the AgB8:1 and AgB8:2 nucleotide sequences showed a 53.5% identity among exons and a 50% identity between introns. According to the molecular diversity analysis, the elapsed time since both genes shared a common ancestor would be around 4.2�107 years. When the native AgB and the two recombinant antigens (rAgB8:1 and rAgB8:2) were tested in an anti-IgG ELISA, the sensitivity of the native antigen B was 77.41% and its specificity was 81.9%, while rAgB8:1 showed 54.84% of sensitivity and 80.17% of specificity and rAg138:2 had an 83.87% sensitivity and a 98.28% specificity. BACKGROUND: The global beef market demands the meat industry to ensure product quality and safety in markets that are often very distant. The present study aimed to evaluate the effects of chilled (CH, 120 d) and chilled-then-frozen (CHF, 28 d + 92 d) storage conditions of beef vacuum packaged (VP) and vacuum packaged with antimicrobial (VPAM) on meat quality, oxidative status and microbial loads. Treatments resulted from the combination of storage condition and packaging type: VP + CH, VP + CHF, VPAM + CH and VPAM + CHF. RESULTS: Warner–Bratzler shear force values decreased in all treatments after 28 d of chilling. Except for VP + CH, L* values (lightness) of meat color did not differ in each treatment as the storage time increased. Meat from VP + CH had greater a* values than CHF treatments on day 120 of storage. A consumer panel did not detect differences in tenderness, flavor and overall liking between VP and VPAM beef, but they preferred CHF steaks rather than CH beef. TBARS values did not differ between VP and VPAM and between CH and CHF at any time during the storage period. At the end of storage time, all treatments except VP + CHF presented a greater concentration of thiols than at 48 h post-mortem. On day 120 of storage, VP + CH had greater cat alase enzyme activity than CHF treatments while VP + CH and VP + CHF showed a greater superoxide dismutase activity than VPAM + CHF. Storage condition (CH or CHF) had a greater impact on microbial counts than the type of packaging. CONCLUSION: Freezing meat after an ageing period represents a suitable strategy to extend beef storage life without a detri mental impact on its quality. This cross-sectional study examines the association between blood lead level (BLL) and two OS markers-urinary F2-8α isoprostane or isoprostane (a marker of lipid peroxidation) and 8-hydroxy-2-deoxy-Guanosine or 8-OH-dG (a marker of DNA damage) in 211 children, aged 5-8 years, from Montevideo, Uruguay. The role of dietary intakes of vitamin C and zinc in modifying the relation between BLL and OS was also examined. The mean (SD) BLL of the study children was 4.7 (2.2) µg/dL, with 30.2% children having BLL ≥5 µg/dL, the current reference level set by the US Centre for Disease Control for identifying, monitoring and management of children with elevated BLL. In covariate-adjusted analysis, there was a weak positive association between BLL and urinary isoprostane (adjusted for specific gravity) [β=0.09, p<0.1]. No association was found between children's BLL and urinary 8-OH-dG. Interactions between dietary intakes of vitamin C or zinc and BLL on OS biomarkers were not consistent. However, when BLL and vitamin C or BLL and zinc were modeled together, BLL was independently associated with isoprostane concentration [β=0.10, p<0.05] but vitamin C or zinc intake was not. These findings suggest that there may be a potential adverse effect of BLL on OS in children with low-level Pb exposure. There is a need to study the effects of Pb on other OS measures, as well as the role of OS in mediating low-level Pb toxicity on functional outcomes. Calcination was considered for the first time as an alternative to recover inorganic bone component from spent glucoamylase-bone derivatives. The subsequent adsorption of glucoamylase (GA) onto calcined bone particles was assessed. Adsorption capacity of the calcined matrix was found to be from 1.1- to 1.4-fold superior to that of non-calcined supports, and it was dependent on the applied load. Moreover, the expressed activity of GA derivatives on calcined matrix was, at least, 2-fold higher than that of biocatalysts onto non-calcined support. The optimization of the loading allowed the preparation of derivatives with 139 GA units per gram of support, which preserve 52% of the immobilized activity. Additionally, calcination of spent GA biocatalysts on calcined bone particles was performed, and adsorption of glucoamylase onto the bone particles calcined a second time was also found to be efficient. In addition to the improved catalytic properties, the half-life at 55°C of the GA biocatalysts on calcined bone was increased 1.7- fold in comparison with that of soluble GA and GA adsorbed onto non-calcined bone particles. Furthermore, the same cassava starch conversion can be achieved batchwise in a stirred-tank reactor using less insoluble biocatalyst, 37% of the GA-bone derivative, which represents an important saving for industrial applications. Nitroalkenes derivatives of free as well as esterified unsaturated fatty acids are present in human plasma and tissue, representing novel pluripotent cell signaling mediators. Lipid nitration occurs in response to pro-inflammatory stimuli as an adaptative mechanism to downregulate inflammatory responses. This chapter first discusses the generation of nitroalkenes during macrophage activation following chemical and biological characterization. In particular, it describes procedures for (a) synthesizing and characterizing esterified (cholesteryl-nitrolinoleate, CLNO2) as well as free (nitroarachidonate, AANO2) nitroalkenes, (b) determining nitration of cholesteryl linoleic acid during macrophage activation by inflammatory stimuli, (c) examining the modulatory effects of nitroalkenes on the expression of inducible enzymes by activated macrophages, and (d) discussing the signaling pathways involved in nitroalkene-mediated anti-inflammatory actions.
