Abstract
A summary of the resource.
Se estudió la marcación con 99Tcm de derivados del ácido iminodiacético (dietil-IDA diisopropil-IDA p-butil-IDA y trimetilbromo-IDA) por vía electrolítica. Los rendimientos de marcación en función del tiempo y la estabilidad in vitro (a temperatura ambiente) de las correspondientes moléculas marcadas se evaluaron por procedimientos fisicoquímicos y por distribución biológica en animales de laboratorio. Después de evaluados los resultados se concluye que el método electrolítico constituye una vía alternativa de marcación de los derivados del IDA brindando radiofármacos de calidad comparable a los resultantes de la utilización de juegos de reactivos con SnCl2 como agente reductor.

Abstract
A summary of the resource.
The synthesis of a ligand containing a nitrobenzyl group as bioreductive pharmacophore and the preparation of the corresponding technetium and rhenium complexes are presented. 99mTc labelling was performed in high yield (>90%) by ligand substitution using fac-[99mTc(CO)3(H2O)3]+ as precursor. The structure of the technetium complex was established by chromatographic comparison with the analogous rhenium compound which was fully characterized by elemental analysis, spectroscopic methods and X-ray crystallography. Reduction potential of the rhenium complex was in the characteristic range for bioreductive compounds. Biodistribution in normal mice was characterized by fast blood and soft tissue depuration and combined excretion via the hepatobiliary and urinary systems. Tumour uptake was low, probably due to low lipophilicity but tumour/muscle ratios were favourable as a consequence of high excretion

The [Tc(phen)3] (PF6)3 complex has been synthesized by ligand substitution on [Tc(tu)6]3+. The reaction proceeds quickly in aqueous solution. A pure compound is obtained with 40% yield. It was characterized by Tc elemental analysis, cerimetric titration, conductometry and electrophoresis. The corresponding spectroscopic properties (UV, Vis, IR and NMR) are also reported and discussed.

A summary of the resource.
Samarium complexes with glutamic acid containing cationic [Sm2(Glu)2(H2O)8]4þ units have been prepared and characterized by using elemental analysis, infrared absorption spectra and thermal analysis. In addition, the crystal and molecular structure of [Sm2(Glu)2(H2O)8](ClO4)4·3H2O was determined by X-ray diffraction. The presence of glutamic acid in the Sm coordination sphere allows the formation of a singular extended 2D arrangement. Sm atoms are nine coordinated, in a monocapped square antiprism geometry. They are connected by means of different m-COO2 bridges, involving a- and gcarboxylate groups of the amino acid.

OBJECTIVE: The aim of the present study was to evaluate the influence of the incorporation of caffeine as a model active ingredient on the quality attributes of a bioadhesive emulgel formulation previously optimized by Quality by Design (QbD) tools. Emulgels are emerging topical drug delivery systems for cosmetic or pharmaceutical uses, which combine the advantages of both emulsions and gels. METHODS: In this work, the observed and predicted values for spreadability, phase separation by centrifugation and detachment forces performed by texturometer, were compared with those of the control and active-containing emulgel formulations. In addition, rheological properties, release of caffeine and comparative in vitro/ex vivo bioadhesion properties were evaluated using human skin. RESULTS: The flow curves of emulgel formulations showed the typical pseudoplastic and no thixotropic flow with yield stress. The incorporation of active ingredient did not produce significant changes. All emulgels were uniformly spread and no significant differences in spreadability values between control and caffeine containing formulations and neither respect to those predicted values from experimental design optimization were found. Emulgel formulations showed appropriate detachment forces values and no significant differences between caffeine loading and control emulgel formulations were observed. CONCLUSION: Caffeine was successfully vehiculized in this optimized bioadhesive emulgel formulation, which showed high robustness regarding the process variability. Bisthiazolidines (BTZ) are bicyclic compounds considered as penicillin analogs that inhibit the full range of Metallo‐β‐Lactamases (MBLs) and potentiate β‐lactam activity against resistant bacteria. Herein, we present a new methodology to prepare 2‐substituted bisthiazolidines by aldehyde exchange. Thirteen new bisthiazolidines were prepared using this methodology, with yields ranging from 31 to 75 %. The reaction is based on in situ imines formation, which are able to exchange side chains. The reaction intermediates were studied based on NMR experiments, and a key imine 1b‐II could be detected in the reaction mixture. Furthermore, a DFT computational analysis was performed to gain insights into the reaction mechanism, allowing us to unveil the different pathways and their activation barriers within the synthetic route. The results suggest that the most favorable route involve the formation of the thiazolidine 1b‐III by i) a N‐assisted N–C bond cleavage, and ii) a thiol‐mediated 5 endo‐trig cyclization followed by a C–N bond cleavage. In contrast with previously reported evidence, the imine metathesis was discarded as a plausible pathway. Finally, the reaction of 1b‐III with aldehyde 2a leads to bicycle 4a via the iminium ion 1b‐V.

<p>2-fluoro-2-deoxy-D-glucose (FDG) is considered of great interest. Herein,we present the synthesis and preliminary bioevaluation of a 99mTc(CO)3-glucose derivative. Derivatization of glucose at C2 was achieved using the so-called ‘click chemistry’, forming a histidine-like, 1,4-disubstituted triazole adequate as donor atom system for Tc(I)-tricarbonyl complexes. Synthesis of the intermediate azide derivative was achieved by conventional organic chemistry. Because of the efficiency and selectivity of this cycloaddition, labelling was performed in ‘one pot’, adding the tricarbonyl precursor fac-[99mTc(CO)3(H2O)3]+ to the same vial where the click reaction has taken place without any further purification. A single product with radiochemical purity higher than 90%was obtained. It was stable for at least 4 h in reaction milieu and exhibited high hidrophilicity (log P of �1.2) and low binding to plasma proteins (5�1%). Biodistribution in C57BL/6 mice bearing induced Lewis murine lung carcinoma is characterized by low blood and liver uptake and rapid urinary excretion. 99mTc complex showed moderate tumour uptake but significant retention until 2 h post-injection. Soft tissue clearance was fast leading to significantly higher uptake in tumour in comparison to muscle (p = 0.05) at all time points. Overall, biodistribution of our compound was very similar to that of [18F]-FDG. However, tumour uptake was significantly higher for [18F]-FDG, probably because of high hidrophilicity of our derivative that may hinder cell penetration. Similarity to biodistribution of FDG is a promising outcome, and modifications of the chelator and linkermight improve biological results</p>

The enzyme penicillin G acylase (PGA) is currently employed at an industrial scale in the hydrolysis of penicillin and cephalosporin G. Here, we describe the preparation of a new immobilized preparation of the enzyme that yields derivatives that are very thermostable and resistant in the presence of organic solvents. The stabilization is obtained via a double strategy. First, using a new epoxy support (Sepabeads), a high degree of multipoint covalent attachment is achieved. A threestep methodology is proposed: (1) immobilization on the support at high ionic strength, (2) promotion of an intense multipoint covalent attachment by incubation for a long time at alkaline pH, and (3) blocking hydrophylization of the support surface. These derivatives are more rigid and therefore more resistant to inactivation by both temperature or organic solvents. Second, the contact of the enzyme with the organic solvent molecules is reduced by generating a highly hydrophilic microenvironment using polymers (polyethylenimine, aldehyde-dextran, and sulfate-dextran). The exact protocol for the production of this environment has been found to be critical to achieve the desired results (final derivatives remain active in 90% of organic solvents). This derivative could be used in the hydrolysis of antibiotics even in media fully saturated with organic solvents, making simpler the current processes of production of 6-aminopenicillanic acid (6-APA) or 7-aminodeacetoxycephalosporanic acid (7-ADCA).

A summary of the resource.
In order to prepare the imine derived from condensation of glycine ethyl ester and 5-nitrothiophene-2-carboxaldehyde, another compound was obtained instead. This product was the enamine, ethyl 2-amino-3-(5-nitro-2-thienyl)-2-propenoate, resulting from the condensation between the methylene of the glycine derivative and the heterocyclic carbonyl group. The reactivity of related amines and heterocyclic aldehydes was studied in order to get more insight about the mechanism of enamine formation. It was found that the nitro-heterocyclic group and the easy of enolization of the aminoacidic ester played an important role.

A summary of the resource.
Preparation of Concanavalin A-adsorbents by immobilization on Sepharose activated with 1-cyano-4-(dimethylamino)-pyridinium tetrafluoroborate (CDAP-reagent) is reported. High immobilization yields of lectin (above 90%) were attained using an optimized CDAP-activating protocol. The effect of ligand density on the performance of the adsorbent for specific binding of glycoproteins was studied using horseradish peroxidase (HRP) as a model. Adsorption yields of pure HRP exceeding 90% were obtained with Con A-derivatives containing not < 20 mg of immobilized Con A/ml of packed gel. With lectin content of 2 mg/(ml of packed gel), only 20% of HRP was adsorbed. Purification of peroxidase from horseradish roots extract was successfully accomplished on Con A-Sepharose with high Con A content.

The Pechini-type sol-gel (PSG) process has been used for the preparation of doped oxides due to its capability to overcome most of the difficulties that frequently occur by using other producing methods. In this work we analyze the case of samples of pure and In-doped yttria (Y2O3) prepared by the PSG process. We experimentally characterize the synthesized samples by x-ray diffraction, micro-Raman spectroscopy, electrochemical impedance spectroscopy (EIS), and time-differential perturbed γ-γ angular correlation (PAC) spectroscopy, and we compare these results with those obtained starting from commercial oxide powders. We found that the PSG process can be used to successfully produce doped yttria in the cubic phase, with the impurities substitutionally located at the cationic sites of the structure. By the proposed PSG route, the inclusion of impurities does not affect the particle size nor the resistivity. However, when we compare the PSG samples with other samples produced from commercial powder, we found that the first have lower resistivities at grain interiors. On the other hand, PAC spectroscopy in 111In(→111Cd)-doped yttria allows the study of the dynamic hyperfine interactions observed by the radioactive 111Cd impurity-probe, which can be used to “sense” the host electron availability near the impurities after the electron-capture decay of 111In. Differences between PAC spectra for PSG samples and the commercial powder suggest that the PSG process introduces additional donor defects into the yttria electronic structure, which is consistent with the lower resistivity observed in the PSG samples by EIS spectroscopy.

The aim of the present work is to illustrate basic issues relating to the antibody response through the preparation reagents for blood group serology. Two rabbits were immunised with the stromata of A and B cells. The antibody response was analysed by ELISA and Haemagglutination. The appropriate bleeding was selected in order to prepare the reagents. They were tested by blood grouping a panel of volunteers and comparing the results with those obtained with a commercial kit.

The photoisomerization of the Diels–Alder adduct formed between tetramethylpurpurogallin and maleic anhydride is described. The process is highly stereoselective, involves the b,c-unsaturated ketone chromophore present in the adduct, proceeds through 15 the formation of allyl and aroyl radical intermediates, and leads to a novel substituted dihydrofluorenone in good yields.

Abstract
A summary of the resource.
Ligand exchange is one of the possible synthetic routes to obtain99mTc coordination compounds. However, the success of this route depends on the availability of good precursors. The objective of this work is the preparation of the complex [99mTc(tu)6]3+ (tu = thiourea), as a potential precursor for99mTc(III) coordination compounds. The preparation was successfully performed in acidic conditions, the excess of tu serving as reducing agent. At pH values higher than 3, the compound becomes unstable and on addition of polydentate ligands new Tc(III) complexes are formed. With edta, the complex99mTc(III)-edta was obtained in high yield.

Abstract
A summary of the resource.
Optimized procedures for the affinity purification of soybean agglutinin (SBA) from soybean flour, and its further immobilization, were developed. Lectin purification on galactosyl-Sepharose yielded 44.563.5 mg of pure SBA/50 g of flour. To prepare SBA adsorbents, the lectin was immobilized onto 1-cyano-4-(dimethylamino)pyridinium tetrafluoroborate (CDAP) activated Sepharose with high yields (77%). Feasibility of the use of this improved SBA adsorbent for affinity purification of Streptococcus pneumoniae capsular polysaccharides from strain 14 (CPS-14) at laboratory scale was demonstrated. Using SBA-Sepharose adsorbent (7.0 mg lectin per ml), amounts of 6.3 mg of pure CPS-14 per cycle were produced, the adsorbent being reused up to four times without loss of capacity.

A summary of the resource.
In order to characterize GATA transcription factors in Echinococcus granulosus, a PCR-based cloning strategy was developed. Degenerate oligonucleotides were designed for the most conserved sequence in GATA proteins that include 20 amino acids of the zinc domain. A 60 bp fragment was isolated that had high homology among this sequence and those reported in other species. An analogous sequence was obtained by performing the same procedure with DNA from the free living platyhelminth Dugesia tigrina. High stringency Southern blotting experiments confirmed the presence of this sequence in the parasite genome.

The nirS nitrite reductase genes were studied in two strains (strains 27 and 28) isolated from two denitrifying reactors and characterized as Thauera according to their 16S rRNA gene sequences. Strain 28 contains a single nirS sequence, which is related to the nirS of Thauera mechernichensis, and strain 27 contains two nirS sequences; one is similar to the nirS sequence from Thauera mechernichensis (gene 2), but the second one (gene 8) is from a separate clade with nirS from Pseudomonas stutzeri, Azoarcus species, Alcaligenes faecalis, and other Thauera species. Both genes were expressed, but gene 8 was constitutively expressed while gene 2 was positively regulated by nitrate.
