Background Microbial oils produced by diverse microorganisms are being considered as alternative sources of triglycerides for biodiesel production. However, the standalone production of biodiesel from microorganisms is not currently economically feasible. In case of yeasts, the use of low-value nutrient sources in microbial production and the implementation of cost-efficient downstream processes could reduce costs and make microbial lipids competitive with other commodity-type oils in biodiesel production. Industrial biodiesel synthesis from oleaginous seeds is currently based on a multistep process. However, a simple process called in situ transesterification (ISTE), which takes place within the biomass without a previous lipid extraction step, is receiving increasing interest. In this work, the optimal conditions for an ISTE process to obtain biodiesel from previously selected oleaginous yeast (Rhodotorula graminis S1/S2) were defined using the response surface methodology (RSM). Results Using the RSM approach, the optimal conditions for the maximum yield with minimum reaction time included a methanol-to-biomass ratio of 60:1, 0.4 M H2SO4, and incubation at 70°C for 3 h. The optimized in situ process yield was significantly higher (123%) than that obtained with a two-step method in which fatty acids from saponifiable lipids were first extracted and then esterified with methanol. The composition of the fatty acid methyl ester mixture obtained from R. graminis S1/S2 by ISTE met Uruguayan standards for biodiesel. Conclusion The characteristics achieved by the optimized method make microbial oil a potential alternative for biodiesel production from yeast at an industrial scale. Tissue engineering is an interdisciplinary field that applies the principles of engineering and life sciences toward the development of biological substitutes that restore, maintain, or improve tissue function. The aims of this work were to compare chemically and physically processed human Amniotic Membranes (hAM) and analyze the cytocompatibility and proliferation rate (PR) of two primary human mesenchymal stromal cell lines, from different sources and donor conditions seeded over these scaffolds. The evaluated hAM processes were: cold shock to obtain a frozen amniotic membrane (FEAM) with remaining dead epithelial cells, denudation of hAM with trypsin for 20/10 min (DEAM20/10) or treatment with sodium dodecyl sulfate to decellularized hAM (DAM). All samples were sterilized with gamma radiation. The selection of the treated hAM to then generate composites was performed by scanning and transmission electron microscopy and characterization by X-ray diffraction, selecting DEAM10 and FEAM as scaffolds for cell seeding. Two sources of primary human stromal cells were used, both developed by our researchers, human Dental Pulp Stem Cells (hDPSC) from living donors and human Mesenchymal Stromal Cells (hMSC) from bone marrow isolated from brain dead donors. This last line of cells conveys a novel source of human cells that, to our knowledge, have not been tested as part of this type of construct. We developed four in vitro constructs without cytotoxicity signs and with different PR depending on the scaffolds and cells. hDPSC and hMSC grew over both FEAM and DEAM10, but DEAM10 allowed higher PR.

Se realizó un estudio preliminar de biodisponibilidad de furosemida vs. una prodroga de furosemida: 4-cloro-N-furfuril-5-sulfamoilantranilato de acetiloxi- metilo, en seis voluntarios sanos, a través de un diseño aleatorio, cruzado y compensado. Se monitoreó por un lapso de 24 horas, los niveles de Furosemida en orina por Cromatografía Líquida de Alta Resolución (HPLC). Se calculó el porcentaje acumulado de la dosis de furosemida recuperada en orina al cabo de las 24 hs.(%E24), las constantes cinéticas de eliminación (β) y de absorción (Ka), y el tiempo para la velocidad máxima de excreción de furosemida (Tmax). Se analizó estadísticamente cada parámetro mediante el test t-Student para series apareadas, previa verificación de normalidad en la distribución de cada serie y homogeneidad de varianzas

Los colorantes sintéticos son ampliamente utilizados en la industria textil. La presencia de estos compuestos recalcitrantes en los efluentes industriales representa un problema ambiental si no se realiza un tratamiento previo para su eliminación antes de su vertido en los cursos de agua. Estos xenobióticos presentan una elevada resistencia a la degradación química y/o biológica, causan la disminución de entrada de luz en ecosistemas acuáticos retardando la fotosíntesis y modifican la solubilidad de gases en el agua afectando el desarrollo de organismos acuáticos. Adicionalmente, muchos son tóxicos, carcinógenos o mutagénicos. La aplicación de lacasas en el tratamiento enzimático de colorantes utilizados en la industria textil surge como alternativa a los tratamientos con métodos tradicionales, los cuales no siempre son ambientalmente amigables, accesibles desde el punto de vista económico, ni completamente efectivos. Estas oxidorreductasas son muy versátiles debido a su baja especificidad, pueden oxidar diversos compuestos fenólicos y aminas aromáticas, así como compuestos más complejos utilizando mediadores redox como co-sustrato. En esta Tesis se trabajó con dos lacasas producidas por cepas nativas de los basidiomicetes Trametes villosa y Pycnoporus sanguineus aislados de zonas forestadas con Eucalyptus globulus en Uruguay. Se optimizó la producción de ambas enzimas en un medio de cultivo semi-sólido utilizando como sustrato un sub-producto de la industria forestal y las mismas fueron purificadas y caracterizadas. Estas lacasas presentaron una alta capacidad de degradación de distintos colorantes sintéticos, incluso en algunos casos sin necesidad de un mediador redox. Los estudios realizados representan una contribución importante al área ya que no se ha reportado previamente el tratamiento de efluentes coloreados con lacasas producidas por hongos de colecciones locales. Por otro lado, dadas las ventajas que ofrecen los procesos de inmovilización de proteínas, las lacasas fueron inmovilizadas para la obtención de nuevos biocatalizadores insolubles. La inmovilización hace posible la separación del derivado enzimático de los productos de reacción así como la disminución de los costos globales del proceso al permitir el re-uso del biocatalizador. Como método de inmovilización se seleccionó el covalente reversible. El mismo combina una unión fuerte que evita la Resumen XX pérdida de enzima durante la aplicación del biocatalizador insoluble y posibilita regenerar el soporte por reversibilidad de la unión, lo cual contribuye a la reducción del costo global del proceso. Es importante remarcar que el desarrollo de un biocatalizador insoluble en base a lacasa inmovilizada en el soporte tiolsulfinato-agarosa es totalmente novedoso y de gran potencial para futuras aplicaciones biotecnológicas. En particular, con el derivado obtenido por inmovilización de lacasa de T. villosa se logró alcanzar altos rendimientos de degradación de colorantes tipo azoico y antraquinónico, los cuales representan las principales clases de colorantes empleados a nivel industrial. Por último, se debe tener en cuenta que si bien la disminución del color del efluente puede ser el principal objetivo de los tratamientos realizados en la industria no debe ser el único, ya que se debe considerar asimismo la toxicidad de los productos obtenidos. En este sentido, los estudios realizados aportan información significativa acerca de la ecotoxicidad de dos colorantes azoicos (Acid Red 88 y Acid Black 172) tratados con el sistema lacasa-mediador. De esta forma, el trabajo realizado contribuye al desarrollo de técnicas para el tratamiento de efluentes industriales coloreados que contemplen diferentes aspectos relacionados a la preservación del medio ambiente.

Individual differences in food-related consumer behaviour are well documented, but lack thorough exploration in relation to product-elicited emotional associations. In this research, focus is directed to product involvement as a factor that modulates emotional associations to tasted products (dried fruit, n = 4) and written descriptions of consumption situations (drinking red wine, cooking dinner using seafood). Emoji questionnaires were used (as check-all-that-apply questions: CATA), and across two studies with consumers in New Zealand (n = 352) and China (n = 450), higher levels of involvement were associated with more positive emotional associations. For example, consumers with higher involvement for dried fruit used emoji with positive meanings (e.g., face savouring delicious food (Image 1), smiling face with heart-shaped eyes (Image 2) and smiling face with smiling eyes (Image 3) more frequently than those with lower levels of involvement. Conversely, emoji with negative or neutral meanings (e.g., confused face (Image 4), confounded face (Image 5), neutral face (Image 6)), were more frequently used by consumers with lower levels of product involvement. The number of significant differences between the samples of dried fruit were lower in the less involved consumer segment, and these consumers, on average, used less emoji to characterise the samples. A similar pattern of results were established for the written stimuli, which were used with Chinese consumers. For example, in the segment with greater involvement with seafood, associations to emoji with positive meanings were higher when responding to the situation “cooking dinner using frozen seafood as one of the ingredients.” In the case of “drinking French red wine,” the strategy used to define segments (median vs. triadic split of summed involvement scores) additionally influenced the results, and bigger differences were established when comparing more discrete segments (two extreme groups following triadic split).

The reformulation of the food products available in the marketplace to improve their nutritional quality has been identified as one of the most cost-effective policies for controlling the global obesity pandemic. Front-of-pack (FOP) nutrition labelling is one of the strategies that has been suggested to encourage the food industry to reformulate their products. However, the extent to which certain FOP labels can encourage product reformulation is dependent on consumer reaction. The aim of the present work was to assess consumers' perception towards product reformulation in the context of the implementation of nutritional warnings, an interpretive FOP nutrition labelling scheme. Three product categories were selected as target products: bread, cream cheese and yogurt, each associated with high content of one target nutrient. For each category, six packages were designed using a 3 × 2 experimental design with the following variables: product version (regular, nutrient-reduced and nutrient-free) and brand (market leader and non-market leader). A total 306 Uruguayan participants completed a choice experiment with 18 choice sets. Reformulated products without nutritional warnings were preferred by participants compared to regular products with nutritional warnings. No apparent preference for products reformulated into nutrient-reduced or nutrient-free product versions was found, although differences depended on the product category and the specific reformulation strategy. Preference for reformulated products without nutritional warnings was more pronounced for non-market leaders. Results from the present work suggest that reformulation of foods in the context of the implementation of nutritional warnings holds potential to encourage consumers to make more healthful food choices and to cause a reduction of their intake of nutrients associated with non-communicable diseases.

Methodological research is needed to support the continued popularity of sensory product characterization by consumers. The present research considers stability and reliability of such product spaces. Using data from 13 studies (n = 1332) involving check-all-that-apply questions (CATA) and projective mapping tasks replicated assessments and increasing the sample size are evaluated for their impact on the product spaces derived from a base group of consumers. In each study, a group of consumers (41–95 participants) completed a sensory characterization task twice under identical conditions (“replication” condition), and additional consumers (49–93 participants) completed the task (“increased sample size” condition). The stability of product spaces was evaluated using a bootstrapping resampling approach. Both experimental conditions had a small but positive impact as evidenced by small increases in average RV coefficients across simulations. Additionally, in studies using projective mapping, the confidence ellipses around sample positions were smaller than those established in product spaces with the base group of consumers. This pointed to increased sample discrimination.

Abstract
A summary of the resource.
We studied the production and the potential use of a purple-pigment produced by an Antarctic bacterial isolate. This pigment was identified as violacein, a metabolite produced by many bacterial strains and reported that it has antiproliferative activity in many cell lines. We analyzed the effect of temperature and the composition of the growth medium on pigment production, achieving the highest yield at 20 °C in Tryptic Soy Broth medium supplemented with 3.6 g/L glucose. We doubled the yield of the pigment production when the process was scaled up in a 5 L bioreactor (77 mg/L of crude pigment). The pigment was purified and identified by mass spectrometry (DI–EI–MS) and Nuclear Magnetic Resonance (NMR) spectroscopy as violacein. We performed survival assays that showed that the pure pigment has antiproliferative activity and sensitize HeLa cells (cervix cell carcinoma) to cisplatin. Besides, the pigment did not show genotoxic activity in HeLa cells as found performing micronucleus assays. These results suggest that this pigment may be used as anticancer or sensitizer to cisplatin drug in cervix cancer.

New enzymes isolated from the biodiversity of native wine ecosystems could contribute to increase the varietal character of regional wines. This study reports on the production and characterization of Issatchenkia terricola, beta-glucosidase and its potential to release red-wine aromatic compounds. The enzyme, a monomer of 48 kDa with an isoelectric point of 3.5 is tolerant to glucose and ethanol, properties compatible with enological use. Although fed-batch is usually the most suitable system for enzyme production in submerged culture, in this case the yield was practically the same as in batch culture. Enzyme productivity was increased 2-fold in synthetic medium with glucose with respect to the YPG and 3–8-fold with respect to other media assayed. After enzymatic treatment, GC-MS analysis of the released aglycones demonstrated significant increases in the concentration of phenols (83%) and norisoprenoids (65%). According to the judges of the sensory panel, the treatment resulted in a wine with dried fruits and raisins notes, as compared to the control, which was found more sweet and fruity. This, together with the lack of activity on anthocyanin glycosides, highlights the potential of this enzyme in enology, since its high selectivity allowed the development of aroma without compromising wine color.

Aims: To characterize the bioemulsifier produced by a nonfluorescent strain of Pseudomonas putida isolated from a polluted sediment and to determine the influence of pH, temperature, media composition, and carbon and nitrogen source on growth and emulsifying activity. Methods and Results: Different indexes were employed to determine the emulsifying properties of culture supernatants of P. putida ML2 in defined and complex media. Surface tension of cell-free supernatants was measured. Purification and chemical analysis of the emulsifier was performed. Confirmed results indicate that a polysaccharide with hexasaccharide repeating units is responsible for the emulsifying activity in a mineral medium with glucose as sole carbon source. Moreover, an emulsifier is produced when growing on naphthalene. Conclusions: Culture media composition influences the amount and the properties of the emulsifier produced by this P. putida strain. Under nitrogen limiting conditions, a polysaccharide is responsible for the emulsifying activity in defined mineral media. In complex nitrogen rich medium, a different kind of emulsifier is produced. The exopolymer may contribute to hydrocarbons solubilization. Significance and Impact of the Study: This is the first exopolysaccharide with emulsifying properties produced by a Pseudomonas strain reported to the present. Also chemical composition is significantly different from previous reports. This strain has potential use in bioremediation and the purified polysaccharide may be used in food and cosmetic industry. Moreover, the production of the exopolymer may play a role on biofilm formation.

ß-Glucosidases (ßGL)—widely used in the enological field—are enzymes that catalyze the liberation of aromatic volatiles from their glycosidic precursors during winemaking. In the present study, a ßGL obtained from the Antarctic yeast Mrakia sp. LP 7.1.2016 was produced on a bioreactor scale, purified, characterized, and the enzyme’s properties studied for a potential enological application. Sodium-dodecylsulfate–polyacrylamide-gel electrophoresis indicated a high molecular weight for this enzyme, it having at least two subunits of 134 and 14 kDa. ßGL exhibited a pH optimum of 5.0 and retained substantial activity and stability at pH 4.0. The temper ature range for optimal activity was 50–55 ◦C, with the enzyme demonstrating thermostability up to 50 ◦C and retaining 87% of residual activity at that temperature after 3 h. The respective kinetic constants determined with p-nitrophenyl-ß-D-glucopyranoside and cellobiose were 0.38 and 1.79 mM for Km and 20.1 and 5.65 μmol− 1 mg− 1 min.1 for Vmax, ß-Glucosidase manifested high activity in 10–25% (v/v) ethanol, 30.0 mg L− 1 sulfur di oxide, and 10–200 g L− 1 fructose; but it was strongly inhibited by glucose, retaining only 6% of residual activity in the presence of 20 g L− 1 . Upon investigating the influence of the enzyme on Muscat-wine glycosidic pre cursors, we found significant differences in terpene content after 14 days of ßGL treatment at an eightfold in crease over control-wine levels. The enzyme was more active toward the precursors of the monoterpenes nerol and geraniol and oxides of trans- and cis-linalool. These findings contribute to our understanding of the potential of cold-active enzymes in advantageous biotechnological applications to enology.

Chitosan is derived from chitin and is commonly extracted from the arthropod’s exoskeleton and the cell wall of filamentous fungi. The production of chitosan from fungi has recently emerged as a more sustainable alternative to using crustacean exoskeletons. Fungal chitosan production offers several benefits, including reduced dependence on seasonal factors, milder extraction conditions, more homogeneous physicochemical properties, and fewer allergic reactions. In the present work, the production of fungal chitosan from Rhizopus stolonifer was investigated. Chitosan was extracted from the biomass obtained at different incubation times using two different alkali treatments. Remarkably, we achieved a maximum chitosan yield of 56.9 mg per gram of dry biomass, higher than those previously reported for this fungus. Characterization analyses by FT- IR, XRD, and TGA showed substantial similarities with commercial chitosan, confirming the presence of 2chitosan in all samples. These findings demonstrate that R. stolonifer has the potential as an alternative source of chitosan.

In the present study we describe the production and characterization of a panel of monoclonal antibodies (MoAbs) directed against cruzipain (Crz), the major cysteine proteinase from Trypanosoma cruzi. The five MoAbs, BD6, BF2, CG2, CH8, and DC10 were analysed with respect to affinity and specificity. None of the MoAbs cross-reacted with papain, which has regions of high homology with Crz. Treatment of the antigen with periodate did not affect the binding of the MoAbs. suggesting that they bind to the polypeptide moiety of Crz. CH8 recognized a continuous epitope located at the C-terminal extension of the proteinase that appeared to be highly immunogenic. Although the rest of the MoAbs recognized epitopes located in the catalytic domain, the enzymatic activity of Crz was not impaired by the binding of the MoAbs. Characterization of the antibody-binding sites revealed the presence of at least four separate epitopes.

In this work, we have constructed two functional mouse/human chimeric antibodies (IgMκ and IgG1κ isotypes) by inserting genomic DNA fragments encoding VH and Vκ variable regions of the murine monoclonal antibody IgMκ-83D4 into mammalian expression vectors containing human μ, γ1, and κ constant exons, and by transfecting them into the nonsecreting mouse myeloma X-63 cell line. In previous works, we have demonstrated that 83D4 murine mAb reacts with Tn determinant (GalNAcα-O-Ser/Thr) expressed in 90% of breast, ovary, and colon carcinomas. Both expressed chimeric antibodies were purified from the transfected cell line supernatant by affinity chromatography, and their reactivities against Tn antigen were confirmed by ELISA on asialo ovine submaxilar mucin and immunofluorescence studies on MCF-7 breast carcinoma cell line. We have demonstrated by gel filtration chromatography, that the principal secreted forms were monomers for IgG1κ and pentamers for IgMκ. The binding affinities of these chimeric antibodies against synthetic Tn glycopeptides, were evaluated by surface plasmon resonance showing an affinity constant similar to that of 83D4 native antibody for IgMκ and a lower affinity constant for IgG1κ chimeric antibody. On the other hand, the replacement of mouse C regions with human C regions confers both chimeric antibodies the ability to activate human complement. These mouse/human chimeric antibodies should be much less immunogenic and could play an important role in the lysis of tumor cell expressing Tn-antigen. Therefore, these anti-Tn chimeric antibodies could be considered as potential tools for human in vivo studies.

Proteins in whey from casein production were hydrolyzed with proteolytic extracts from the latex of Araujia sericifera var. hortorum. Hydrolysate fractions that were partially purified by RP-HPLC and evaluated using the ORAC method resulted in an antioxidant capacity of 112.4 ± 4.2 Trolox equivalents (μM). LC–MS/MS analysis of peptide fractions showed that peptides derived from β-lactoglobulin could be responsible for antioxidant activity. The sum of these results along with the fact that a controlled and reproducible production process was used, involving the use of natural resources and reaction conditions without excessive costs at an industrial scale, makes a promising contribution to improving antioxidant dairy products.
