In this article, we show the adaptation of a method for predicting the consumer acceptability of mandarins, originally reported using a high-field 400 MHz NMR spectrometer, to benchtop 60 MHz NMR systems. However, evidence on CD6’s role in the physiological response to bacterial infection was missing. Steady state analyses ofCd6 / mice show decreased levels of natural polyreactive antibodies, concomitant with decreased cell counts of spleen B1a and marginal zone B cells. Time-dependent Density Functional Theory (TD-DFT) calculations are performed to understand the optical properties of complexes and their response upon deprotonation. TD-DFT studies are conducted with PBE1PBE in combination with LANL2TZ on rhenium and 6–311++G(d,p) on non-metal atoms within the CPCM approach. This methodology shows a reasonable match between the measured and simulated spectra keeping the computational cost low. The nature of all absorption bands is ascribed to a ligand-metal-to-ligandmetal charge transfer (LMLMCT), in which negligible contribution from carboxylic units is detected. Environmental exposure to active pharmaceutical ingredients (APIs) can have negative effects on the health of ecosystems and humans. While numerous studies have monitored APIs in rivers, these employ different analytical methods, measure different APIs, and have ignored many of the countries of the world. This makes it difficult to quantify the scale of the problem from a global perspective. Furthermore, comparison of the existing data, generated for different studies/regions/continents, is challenging due to the vast differences between the analytical methodologies employed. Here, we present a global-scale study of API pollution in 258 of the world’s rivers, representing the environmental influence of 471.4 million people across 137 geographic regions. Samples were obtained from 1,052 locations in 104 countries (representing all continents and 36 countries not previously studied for API contamination) and analyzed for 61 APIs. Highest cumulative API concentrations were observed in sub-Saharan Africa, south Asia, and South America. The most contaminated sites were in low- to middle-income countries and were associated with areas with poor wastewater and waste management infrastructure and pharmaceutical manufacturing. The most frequently detected APIs were carbamazepine, metformin, and caffeine (a compound also arising from lifestyle use), which were detected at over half of the sites monitored. Concentrations of at least one API at 25.7%of the sampling sites were greater than concentrations considered safe for aquatic organisms, or which are of concern in terms of  selection for antimicrobial resistance. Therefore, pharmaceutical pollution poses a global threat to environmental and human health, as well as to delivery of the United Nations Sustainable Development Goals. Bull’s-eye rot, produced by Phlyctema vagabunda, is an important postharvest disease in apples. Current measures to control infection include synthetic fungicides, in addition to the application of copper hydroxide and potassium phosphite. However, growing public concern regarding fungicide residues in food has generated interest in developing non-chemical alternative control methods; biological control is one of the most promising alternatives. In this research, native endophytic yeasts were isolated and evaluated for the biocontrol of P. vagabunda in apples. The mechanisms of action involved were also determined. Our research found 2 isolates, Vishniacozyma victoriae EPL4.5 and EPL29.5, which exhibited biocontrol activity against P. vagabunda at 20 �C in apples, the incidence of bull’s-eye rot was reduced by 39% and 61%, respectively, and the severity of the disease was decreased by 67% and 70%, respectively, when apples were inoculated with these yeasts 24 h before applying the pathogen. The main mechanisms that could be involved in the observed biocontrol activity are the ability to form biofilms and the production of volatile organic compounds. Design: In-depth interviews with parents of children aged from 6 months to 5 years. Setting: Montevideo, Uruguay. Participants: Forty-two parents (aged 19−44 years, 90% female). Phenomenon of Interest: Parents’ personal experiences, feelings, and opinions related to how they choose foods for their children. Analysis: Content analysis based on deductive-inductive coding. Results: Parents’ accounts confirmed they intended to pursue different goals when choosing foods for their children. Health-related goals mainly motivated the selection of healthy foods, but they also drove the selection of some ultra-processed products because of misconceptions about their healthiness. Pleasure and enjoyment motivated the selection of ultra-processed products, whereas stress avoidance was associated with selecting convenient foods. The selection of unhealthy foods raised conflicts between goals, which were solved using goal shielding, changes in risk perception and/or compensatory health beliefs. Conclusions and Implications: Results point to the need for communication campaigns and community- based interventions to introduce changes in how ultra-processed products are conceptualized and increase the associations between healthy foods and children’s pleasure and enjoyment to promote healthier eating patterns during infancy and early childhood. Porphyrin and perylene diimide have great application potential in organic optoelectronic devices. However, large plane frames of porphyrin and PDI tend to aggregate, resulting in low power conversion efficiency. In this work, four side chain modified porphyrins (H2TCPP, ZnTCPP, H2TCPP(CH3)4, ZnTCPP(CH3)4), and one perylene diimide (PDI-C5) were synthesized. The blended films of the donor and acceptor were prepared by a simple and low-cost liquid deposition method. Femtosecond transient absorption spectroscopy and global analysis revealed that ZnTCPP(CH3)4:PDI-C5 showed a higher rate of charge transfer and radical ions formation compared to the other porphyrin donors, which means that the ZnTCPP(CH3)4 has good compatibility with PDIC5. In addition, we also characterized these blended films by ultraviolet–visible absorption spectroscopy, cyclic voltammetry (CV), and AFM techniques. The CV results show that the difference between the LUMO of each porphyrin donor and PDI-C5 acceptor is about 0.3 eV, which is sufficient for exciton dissociation. A telephone survey (n = 601) was conducted in Uruguay to explore participants’ mental associations with meat and the underlying reasons changes in the quantity and type of meat consumed. Participants mentioned mostly positive associations with meat’s culinary and cultural aspects and its nutritional value as source of high-quality protein. Other associations were related to meat production, specific types of meat and cuts, meat consumption and pleasure, while price was perceived negatively. Animal welfare concerns emerged as a relevant association, but minor references were made to the effects of meat production on the environment. Changes in the amount and type of meat consumed (higher pork, chicken and cheaper cuts and beef) in the last year were mainly due to price followed by health reasons. Results emphasize the strong meat culture among Uruguayans and reveal that changes in the amount and type of meat consumed last year relate to external (price) or egoistic (health) rather than altruistic motives (ethical or environmental concerns). Antimicrobial resistance is a growing concern in public health and current research shows an important role for bacterial biofilms in recurrent or chronic infections. New strategies, therefore, are necessary to overcome antimicrobial resistance, through the development of new therapies that could alter or inhibit biofilm formation. In this sense, antibiofilm natural products are very promising. In this work, a bioprospection of antimicrobial and antibiofilm extracts from Uruguayan soil bacteria and insect gut bacteria was carried out. Extracts from extracellular broths were tested for their ability to inhibit planktonic cell growth and biofilm formation. Genomic analysis of Bacillus cereus ILBB55 was carried out. All extracts were able to inhibit the growth of, at least, one microorganism and several extracts showed MICs lower than 500 µg mL−1 against microorganisms of clinical relevance (Staphylococcus aureus, Pseudomonas aeruginosa, and Enterobacter cloacae). Among the extracts evaluated for biofilm inhibition only ILBB55, from B. cereus, was able to inhibit, S. aureus (99%) and P. aeruginosa (62%) biofilms. Genomic analysis of this strain showed gene clusters similar to other clusters that code for known antimicrobial compounds. Our study revealed that extracts from soil bacteria and insect gut bacteria, especially from B. cereus ILBB55, could be potential candidates for drug discovery to treat infectious diseases and inhibit S. aureus and P. aeruginosa biofilms. Penicillin is one of the most widely used antibiotics to treat bacterial infections in clinical practice. The antibiotic undergoes degradation under physiological conditions to produce reactive compounds that in vivo bind self proteins. These conjugates might elicit an immune response and trigger allergic reactions challenging to di agnose due to the complex immunogenicity. Penicillin allergy delabeling initiatives are now part of antibiotic stewardship programs and include the use of invasive and risky in vivo tests. Instead, the in vitro quantification of specific IgE is highly useful to confirm immediate allergy to penicillins. However, discrepant results associated with the low sensitivity and accuracy of penicillin allergy in vitro tests have limited their routine diagnostic use for delabeling purposes. We aimed to develop a homologous chemiluminescence-based immunochemical method for the reliable determination of specific IgE to penicillin G, using unprecedented synthetic human-like standards. The synthetic standard targets the major antigenic determinant of penicillin G and the paratope of Omalizumab, acting as human-like specific IgE. It is a potent calibrator, highly stable, easy, and inexpensive to produce, overcoming the limitations of the pooled human serum preparations. The developed method achieved a good agreement and strong positive relationship, reaching a detection limit below 0.1 IU mL− 1 and excellent reproducibility (RSD <9%). The clinical sensitivity of the assay significantly increased (66%), doubling the accuracy of the reference method with an overall specificity of 100%. The new diagnostic strategy compares favorably with results obtained by the standard procedure, paving the way towards the standardization of penicillin allergy testing, and enhancing the detection sensitivity of specific IgE in serum to tackle reliably β-lactam allergy delabeling. The sensing of fluoride ion through convenient and inexpensive methods has become significant analytical goals. The state-of-the-art involves the supporting of urea/thiourea optical discrete anion sensors into a polymeric scaffold. Nevertheless, high levels of selectivity are difficult to achieve, since the colorimetric/fluorometric response is also quenched by other anions such as dihydrogenphosphate. In this work we present, for the first time, the design of a polymeric optical fluoride chemosensor by using the anion-imprinting technique. First, four polymerizable anion receptors containing a urea-type binding site and different fluorophores as signaling units were prepared. One of them (1-(4-vinylphenyl)-3-(1-naphtyl)-urea) shows the most intense fluorescence emis sion in solution, exhibiting high optical sensitivity only for F– and H2PO4– . The electronic and structural features behind the optical response and anion affinity were elucidated through experimental and computational tools. To enhance the optical sensitivity and selectivity, a fluoride-imprinted polymeric optical sensor was prepared starting from this soluble sensor. The fluorescent emission is 13 times higher for the polymer and it is switched off only in the presence of fluoride, while for the rest of the anions, including H2PO4– , there was almost no optical response. To the best of our knowledge, this is the first fluoride-imprinted polymeric optical sensor reported so far. CRISPR-Cas systems in prokaryotic cells provide an adaptive immunity against invading nucleic acids. For example, phage infection leads to addition of new immunity (spacer acquisition) and DNA cleavage (interference) in the bacterial model species Streptococcus thermophilus, which primarily relies on Cas9-containing CRISPR-Cas systems. Phages can counteract this defense system through mutations in the targeted protospacers or by encoding anti-CRISPR proteins (ACRs) that block Cas9 interference activity. Here, we show that S. thermophilus can block ACR-containing phages when the CRISPR immunity specifically targets the acr gene. This in turn selects for phage mutants carrying a deletion within the acr gene. Glutathione transferases (GSTs) perform catalytic and non-catalytic activities, mostly involved in stress-response and cell detoxification. Helminth parasites express several GSTs of multiple classes that are involved in the neutralization of potentially harmful oxidants, and in the inactivation or removal of xenobiotics. Additionally, GSTs participate in immunomodulatory processes that facilitate the parasite establishment and survival within its host. In Echinococcus granulosus sensu lato (s.l.) -the cestode parasite responsible for cystic echinococcosis- only one Mu-class GST has been reported. In the present work, by using bioinformatic and proteomic approaches we searched for novel Mu-class GSTs potentially involved in the parasite oxidative-stress metabolism. In the genome of E. granulosus s.l., 6 GST-related sequences were found to constitute a strongly conserved phylogenetical clade with Mu-class members. Among them, 5 displayed conserved gene structure (exon/intron), as well as specific residues and motifs characteristic of Mu-class enzymes. By proteomic analysis, 3 Mu-GSTs were identified to be expressed in the protoscolex parasite stage, 2 of them being firstly described as Mu-class GSTs here. The existence of more than one productive Mu-GST gene expands the parasite xenobiotic phase II metabolism, which might have beneficial roles on E. granulosus s.l. ability to successfully infect its host. Thus, the objective of this work was to provide a fast and robust method for the analysis of infusions of yerba mate to evaluate teas with different particle sizes, degree of processing and type. A simple extraction using an espresso machine and ultra-high performance liquid chromatography with diode array detection (UHPLC-DAD) was developed and validated for the determination of theobromine, caffeine, caf feoylquinic and dicaffeoylquinic acids in yerba mate infusions. The chemical profiles of twenty-six yerba mate samples reflected their industrial processes. The dietary intake and the ratio of chlorogenic acids to caffeine of yerba mate was evaluated for different products (aged, green, and roasted yerba mate). The new method proved to be a fast, simple, and fit-for-purpose procedure for the aqueous extraction and determination of bioactive compounds in yerba mate products. Herbal teas and yerba mate infusions are an important part of the human diet that may contain toxic contaminants due to the industrial processing of the herbal material. The presence of the lipophilic compounds polycyclic aromatic hydrocarbons (PAHs) was investigated in dry yerba mate samples and how they transfer into the infusions. Samples were selected among eight different brands, and three types of yerba mate products (aged, green, and roasted). Synchronized fluorescence scanning (SFS) was used as primary screening method for PAHs in dried yerba mate, and high-per formance liquid chromatography with fluorescence detection for the ana lysis of the infusions. SFS screening showed presence of PAHs in the dry herbal material, and the PAHs ratios suggest that the contamination origi nates from pyrogenic sources. Using an experimental setup to simulate the traditional yerba mate consumption (successive extractions with small amounts of water), the sum of the six PAHs in each infusion of yerba mate sample ranged from 4.8 to 30.2 ng g 1 . Benzo [a] pyrene concentration ranged from n.d. to 1.1 ng g 1 while phenanthrene reached a maximum of 59 ng g 1 in the yerba mate samples tested. An estimated 0.11% to 0.54% of the six PAH analytes were transferred into the infusion and the SFS method can be used to screen the presence of PAHs in the dry mater ial. The chemical profile of the most abundant PAHs found in the infusions indicate a low total equivalent exposure index. Cystic echinococcosis (CE) is a zoonotic disease worldwide distributed, caused by the cestode Echinococcus granulosus sensu lato (E. granulosus), with an incidence rate of 50/100,000 person/year and a high prevalence in humans of 5-10%. Serology has variable sensitivity and specificity and low predictive values. Antigens used are from the hydatid fluid and recombinant antigens have not demonstrated superiority over hydatid fluid. A cell line called EGPE was obtained from E. granulosus sensu lato G1 strain from bovine liver. Serum from CE patients recognizes protein extracts from EGPE cells with higher sensitivity than protein extracts from hydatid fluid. In the present study, EGPE cell protein extracts and supernatants from cell colonies were eluted from a protein G affinity column performed with sera from 11 CE patients. LC-MS/MS proteomic analysis of the eluted proteins identified four E. granulosus histones: one histone H4 in the cell extract and supernatant, one histone H2A only in the cell extract, and two histones H2A only in the supernatant. This differential distribution of histones could reflect different parasite viability stages regarding their role in gene transcription and silencing and could interact with host cells. Bioinformatics tools characterized the linear and conformational epitopes involved in antibody recognition. The three-dimensional structure of each histone was obtained by molecular modeling and validated by molecular dynamics simulation and PCR confirmed the presence of the epitopes in the parasite genome. The three histones H2A were very different and had a less conserved sequence than the histone H4. Comparison of the histones of E. granulosus with those of other organisms showed exclusive regions for E. granulosus. Since histones play a role in the host-parasite relationship they could be good candidates to improve the predictive value of serology in CE. The present research primarily contributes to the latter by showcasing consumer segmentation based on product-elicited emotional associations obtained using the circumplex-inspired valence × arousal single-response emotion word questionnaire (CEQ). This gives rise to three-way data, where responses from each consumer can be represented by a matrix of p products × 12 emotion word pairs. A multi study (n = 6) strategy was pursued in the present research, with 197 to 1014 people per study. Four of the six studies used tasted products and two studies used written stimuli (i.e., names of foods and beverages). For each study two or three emotion-based clusters were retained, and the differences between these could be classified into three patterns: i) differences in the emotional associations with specific sensory characteristics (tasted products); ii) differences in the emotional associations of the category (tasted products); and iii) differences mainly driven by liking (written stimuli). Fitting expectations, differences between emotion-based clusters within a study extended beyond the insights gained from liking scores. Taken together, the results from the six case studies suggest that consumer segmentation based on emotional responses deserves further exploration. The proposed sample preparation procedure avoids the use of perchloric acid and provided accurate results even for silicon determination using an acid digestion with hydrofluoric acid. The determination of major elements in rocks is required for the design of classification diagrams that provides relevant information for geochemical analysis. • MP-AES showed to be an adequate technique to measure major and some trace elements that are relevant for classification of igneous rocks. • This method is in better agreement with the principles of the Green Analytical Chemistry and constitutes a reliable alternative to classical analytical and tedious procedures used for geochemical analysis. • The methodology was successfully applied to chemical classification of rocks from Valle Chico (Uruguay) using a Total Alkali-Silica Diagram (TAS). Composition and antioxidant properties of sixteen pecan [Carya illinoinensis (Wangenh) K. Koch] cultivars grown simultaneously in a single experimental orchard in Uruguay were evaluated to characterize their nutritional properties and nutraceutical potential. The percentage of oil, moisture, ash, minerals, and proteins were determined and also the fatty acid profile. Total phenolic compounds [18–41 mg gallic acid equivalents (GAEs)/g], condensed tannin [2–12 mg catechin equivalents (CEs)/g], and tocopherols (110–163 µg/g) contents were estimated in nut kernels. Total phenolic compounds (32–117 mg GAE/g), condensed tannins (130–357 mg CE/g), and total anthocyanins (1–3 mg 3-glucoside cyanidin/g) were also determined for pecan shells. The antioxidant activity in shells [57.15–578.88 µmol Trolox equivalents (TEs)/g] was 5 times higher compared with the kernels (23.15–156.60 µmol TEs/g) measured with hydrophilic ORAC. Bioactive compounds concentrations present statistically significant genetic variability between cultivars studied (p < 0.05). The presence of phenolic compounds was related with high-antioxidant capacity in kernels and shells, and a strong correlation between content of total phenolic compounds and condensed tannins in pecan shells was found. Principal component analysis (PCA) and hierarchical cluster analysis (HCA) show association between cultivars and the observed variables. The nutritional profile in the different cultivars showed the trends described in other countries, but this work shows some significant differences that could be attributed to the specific edaphoclimatic conditions of cultivation in Uruguay.
