En Uruguay, las condiciones climáticas favorecen el desarrollo de Botrytis cinerea Pers, pudiendo afectar más del 50% de la producción de un viñedo. En un contexto donde cada vez son menos los fungicidas permitidos, y B cinerea ha presentado resistencia a Benzimidazoles y Dicarboximidas, nace como necesidad desarrollar estrategias de control no químico. Surge como alternativa el empleo de prácticas culturales como el deshojado, comúnmente utilizada en variedades tintas. Con el objetivo de introducir esta práctica en variedades blancas, durante las temporadas 2005/06 y 2006/07, se probaron sobre el cv. Sauvignon blanc., distintas intensidades y momentos de deshojado en combinación con aplicaciones de Caolinita (Sorround®), en un diseño en bloques al azar, con cuatro repeticiones. Todos los deshojados mostraron una incidencia significativamente menor de Botrytis sp, que el testigo sin deshojar. El porcentaje de quemado de sol en racimos, fue significativamente mayor en los tratamientos deshojados sin aplicación de caolinita.

The contribution of yeast fermentation metabolites to the aromatic profile of wine is well documented; however, the biotechnological application of this knowledge, apart from strain selection, is still rather limited and often contradictory. Understanding and modeling the relationship between nutrient availability and the production of desirable aroma compounds by different strains must be one of the main objectives in the selection of industrial yeasts for the beverage and food industry. In order to overcome the variability in the composition of grape juices, we have used a chemically defined model medium for studying yeast physiological behavior and metabolite production in response to nitrogen supplementation so as to identify an appropriate yeast assimilable nitrogen level for strain differentiation. At low initial nitrogen concentrations, strain KU1 produced higher quantities of esters and fatty acids whereas M522 produced higher concentrations of isoacids, g-butyrolactone, higher alcohols and 3-methylthio-1-propanol. We propose that although strains KU1 and M522 have a similar nitrogen consumption profile, they represent useful models for the chemical characterization of wine strains in relation to wine quality. The differential production of aroma compounds by the two strains is discussed in relation to their capacity for nitrogen usage and their impact on winemaking. The results obtained here will help to develop targeted metabolic footprinting methods for the discrimination of industrial yeasts.

A summary of the resource.
In this study, a preparation of saponins (QB-90U) extracted from leaves of Quillaja brasiliensis collected in Uruguay was evaluated as a vaccine adjuvant by comparison with alum and the well known saponinbased adjuvant, Quil A. The haemolytic activity and cellular toxicity of the saponin preparations were also evaluated. QB-90U was only slightly haemolytic and showed a low cytotoxicity when compared to Quil A. The adjuvant properties of QB-90U were assayed by sub-cutaneous immunization of mice with a preparation of inactivated bovine herpesvirus 5 (BoHV-5) either with no adjuvant or adjuvanted with QB-90U, Quil A or alum. Serum levels of anti-BoHV-5 IgG, IgG1, IgG2a, IgG2b and also IgG3 were significantly increased by QB-90U and were of the same order as those elicited by Quil A. Furthermore, high titres of neutralizing antibodies were found to be present in the serum of immunized animals from both groups. The cellular response induced by QB-90U did also reproduce the one elicited by Quil A. In fact, a robust DTH response was observed in mice immunized with both saponin preparations; as well as increased splenocytes levels of Th1-type cytokines, namely IFN-� and IL-2. Taken together, the above results confirm and extend our previous observation regarding the similarity of the responses elicited by Quil A and the saponin preparation from Q. brasiliensis (Fleck et al., 2006) and indicate that QB-90U is worth of further studies as a safe and potent vaccine adjuvant.

Disulfide-bond formation is a major post-translational modification and is essential for protein folding, stability, and function. This is especially true for secreted proteins, many of which possess great potential for biotechnological applications. Focusing on the use of Escherichia coli for the production of this class of proteins, we describe the mechanisms that maintain redox compartmentalization in the cell, with an emphasis on those that promote the formation and isomerization of disulfide bonds in the bacterial periplasm, while presenting parallel pathways in the eukaryotic endoplasmic reticulum. Based on these concepts, we review the use of E. coli as a cell factory for the production of heterologous disulfide-containing proteins using either peri- or cytoplasmic expression and, in particular, how these compartments can be tuned to improve the yield of correctly folded recombinant proteins. Finally, we describe a few examples of the production of small disulfide- rich proteins (protease inhibitors) to illustrate how soluble, active, and fully oxidized recombinants may be successfully obtained upon peri- or cytoplasmic expression in E. coli.

Abstract
A summary of the resource.
Eukaryotes using trans-splicing for transcript processing incorporate a taxon-specific sequence tag (the spliced leader, SL) to a proportion (either all or a fraction) of their mRNAs. This feature may be exploited for the preparation of full-length-enriched cDNA libraries from these organisms (a diverse group including euglenozoa and dinoflagellates, aswell asmembers from five metazoan phyla: Cnidaria, Rotifera,Nematoda, Platyhelminths and Chordata). The strategy has indeed been widely used to construct cDNA libraries for the generation of ESTs, mainly from parasitic euglenozoa and helminths. We describe a set of optimised protocols to prepare directional SL-cDNA libraries; the method involves PCR-amplification of SL-cDNA and its subsequent cloning in a plasmid vector under a specific orientation. It uses small amounts of total RNA as starting material and may be applied to a variety of samples. The approach permits the selective cloning of mRNAs tagged with a particular SL from mixtures including large amounts of non-trans-spliced mRNAs. Thus, it allows exclusion of host contamination when isolating SL-cDNAs from parasitic organisms, and has other potential applications, such as the characterisation of the trans-spliced transcriptome from organisms in mixed pools of species.

responsables del defecto en el aroma del vino descrito como aroma a “establo”, “sudor de caballo”, “curita medicinal”, debido a la producción de fenoles volátiles. En este trabajo presentamos el primer relevamiento en Uruguay de la presencia de estas levaduras en diferentes ecosistemas de uva y vino, incluyendo suelos y orujos, barricas, tanques de hormigón con epoxi, válvulas y tanques de acero inoxidable, utilizando un método microbiológico basado en un medio de cultivo selectivo y diferencial. De un total de 241 muestras analizadas en bodega por los propios enólogos, resultaron con presencia de Brettanomyces 86 vinos (36%). Sin embargo solo el 3% de las muestras presentó una población mayor de 10000 cel/mL y 6% de las muestras una población entre 10 y 10000 cel/mL. El resto de las muestras positivas presentó poblaciones de menos de 10 cel/mL. Para detectar otros posibles orígenes de esta levadura fueron analizados otros insumos de bodega como ser: chips de roble, polvo de roble, polvo de taninos, los que no presentaron estas levaduras. En cambio muestras de escobajo, compostaje y orujos si las presentaron. El método y el medio empleado en este estudio presentan importantes ventajas para la industria del vino, por su simpleza, rápida y sencilla lectura, bajo costo y su capacidad para detectar bajas concentraciones de Dekkera/Brettanomyces en las muestras analizadas.

Abstract
A summary of the resource.
A major fraction of monoterpenes and norisoprenoids in young wines is conjugated to sugars representing a significant reservoir of aromatic precursors. To promote their release, b-glucosidase, a-arabinosidase, and a-rhamnosidase from a commercial Aspergillus niger preparation, were immobilized onto acrylic beads. The aim of this work was the development and application of an immobilized biocatalyst, due to the well-known advantages over soluble enzyme preparations: control of the reaction progress and preparation of enzyme-free products. In addition, the obtained derivative showed increased stability in simile wine conditions. After the treatment of Muscat wine with the biocatalyst for 20 days, free monoterpenes increased significantly (from 1119 to 2132 lg/L, p < 0.01) with respect to the control wine. Geraniol was increased 3,4-fold over its flavor thresholds, and accordingly its impact on sensorial properties was very relevant: nine of ten judges considered treated wine more intense in fruit and floral notes

The aim of this study is to confirm the factor structure of a self-report measure of achievement goal tendencies (Hayamizu & Weiner, 1991) described in previous research, and examine the reliability and validity of scores in a sample of Uruguayan university students. A theoretical three-factor model is proposed (learning, performance and outcome goals), as well as one-factor models for each goal. Confirmatory factor analyses show that all models are identified, and solutions are conceptually correct. Our results suggest that outcome goals could be split into two correlated sub-dimensions related to achievement time (short and long term goals). Concurrent and predicted validity were established through significant correlations with perceived ability and academic achievement respectively. Conceptualisation of these findings and suggestions for future research are discussed. En el presente trabajo se procura confirmar la estructura factorial de una escala de metas académicas (Hayamizu y Weiner, 1991) descrita en investigaciones previas y se valoran la fiabilidad y la validez en una muestra de estudiantes universitarios uruguayos. Se propone un modelo teórico de tres factores relacionados (metas de aprendizaje, de ejecución y de resultado), así como tres modelos unifactoriales separados para cada tipo de meta. Todos los modelos se encuentran identificados y las soluciones son conceptualmente correctas. Nuestros resultados sugieren que las metas de resultado podrían eventualmente desdoblarse en dos subdimensiones correlacionadas, vinculadas al tiempo de concreción de los logros a los que hacen referencia (resultados a corto y a largo plazo). La validez concurrente y la validez predictiva fueron establecidas mediante correlaciones significativas con la percepción de la capacidad y el rendimiento académico respectivamente. Se proponen conceptualizaciones para estos hallazgos y sugerencias para futuras investigaciones.

El objetivo de este trabajo es estudiar el comportamiento diferencial de dos indicadores de rendimiento académico mediante modelos de regresión lineal multivariante: el indicador tradicional –promedio de calificaciones– y un indicador novedoso –el avance en la carrera– definido como la razón entre los créditos acumulados por el alumno durante cierto período de tiempo y los créditos que teóricamente debió acumular en dicho período de acuerdo con el plan de estudios programado. Se trata de una investigación correlacional, de corte transversal, de variables no manipuladas experimentalmente. Participaron 587 estudiantes matriculados en carreras de Química de la Universidad de la República (UdelaR) durante 2000-2003, con al menos una asignatura aprobada durante el período comprendido entre marzo de 2003 y marzo de 2005. El grupo de participantes fue dividido aleatoriamente en dos muestras: estimación –se construyeron los modelos de regresión lineal incluyendo análisis de interacciones y diagnóstico de anomalías– y validación –los modelos fueron validados mediante contrastes de hipótesis de igualdad de parámetros. Los factores explicativos considerados fueron el rendimiento previo, las metas académicas de los estudiantes, la percepción de su propia capacidad y variables sociodemográficas. Los dos indicadores están notablemente correlacionados entre sí (r=0,615, p<0,001) y ninguno correlaciona significativamente con las metas de resultado (promedio, p=0,612; avance, p=0,358). Los modelos multivariantes fueron satisfactoriamente validados y muestran que los indicadores se ven igualmente afectados por el rendimiento previo, que resultó la variable dominante (promedio, p=0,493, p<0,001; avance, p=0,484, p<0,001). Se diferencian en que el promedio además resulta afectado por la capacidad percibida (p=0,175, p<0,001) y, de manera indirecta, por las metas de aprendizaje (p=0,16, p<0,05, controlando por capacidad percibida). El comportamiento de ambos indicadores es similar, siendo los estudiantes que obtienen las peores calificaciones los más susceptibles de presentar retraso curricular. Hay una dominancia del rendimiento previo como predictor de ambas formas de rendimiento futuro.

Este artículo describe las actividades que la Cátedra de Radioquímica realiza en docencia, investigación y extensión. En docencia se describen los objetivos, requisitos y principales características de los actuales curso de grado : Química Nuclear, Radioquímica, Radiotrazadores en Ciencias Biológicas, Radiofarmacia y Tópicos avanzados en Radioquímica. En investigación la Cátedra desarrolla una intensa actividad en 2 áreas principales: las aplicaciones médicas de los radionucleidos y el desarrollo de los materiales para detección de radiaciones ionizantes. En extensión se apoya a la comunidad educativa, de Medicina Nuclear y se realizan asesoramientos para varias industrias nacionales. En los palanes a futuro se destaca la implementación de un Diploma de Especialista en Radiofarmacia y un proyecto de asistencia técnica destinada a alcanzar los más altos estándares internacionales en la infraestructura, equipamiento y capacitación de la Radiofarmacia del Hospital Universitario.

Introduction: Hypoxia imaging is an important field in radiopharmaceutical research since hypoxic cells are very resistant to radiation treatment and diffusional limitations restrict the efficacy of chemotherapy. Gallium-68 is a widely used radionuclide for positron emission tomography (PET) due to the availability of the 68Ge/68Ga-generator. With the aim to develop new potential [68Ga]-radiopharmaceuticals for imaging hypoxia, we have synthesized and evaluated two novel 68Ga-labelled 5-nitroimidazole derivatives. Methods: Two 5-nitroimidazole derivatives, 10-[2-(2-methyl-5-nitro-1 H-imidazole-1-yl)ethylaminocarbonylmethyl]- 1,4,7,10-tetraazacyclododecane-1,4,7-triacetic acid (Nit1) and 10-{[N-methyl-1-[1-(2-(2-methyl- 5-nitro-1 H-imidazole-1-yl)ethyl)-1 H-1,2,3-triazole-4-yl] methylaminocarbonylmethyl}-1,4,7,10- tetraazacyclododecane-1,4,7-triacetic acid (Nit2) were synthesized. Preparation of [68Ga]complexes [68Ga]- Nit1 and [68Ga]-Nit2 was performed at pH 4.5 and 95 °C during 15 minutes and radiochemical purity (RP) was evaluated by reverse phase HPLC. Stability, lipophilicity and plasma protein binding were studied. Biological behaviour in HCT-15 cells both in normoxia and hypoxia has been assessed. Biodistribution in animals bearing induced 3LL Lewis murine lung carcinoma was studied. Comparison with [18F]FMISO is also presented. Results: Nit1 and Nit2 have been successfully synthesized. Labelling in high radiochemical purity was achieved for both ligands. Complexes are stable in labellingmilieu for at least four hours and in human plasma or in the presence of an excess of DTPA for at least two hours. Both compounds showed high uptake in hypoxic cells in vitro and a very favourable biodistribution profile in mice bearing induced tumours. Results are comparable to those obtained for [18F]FMISO. Conclusions: Selective uptake and retention in tumour together with favourable tumour/muscle ratio make these compounds promising candidates for further evaluation as potential hypoxia imaging agents.

The synthesis of a ligand containing a nitrobenzyl group as bioreductive pharmacophore and the preparation of the corresponding technetium and rhenium complexes are presented. 99mTc labelling was performed in high yield (>90%) by ligand substitution using fac-[99mTc(CO)3(H2O)3]+ as precursor. The structure of the technetium complex was established by chromatographic comparison with the analogous rhenium compound which was fully characterized by elemental analysis, spectroscopic methods and X-ray crystallography. Reduction potential of the rhenium complex was in the characteristic range for bioreductive compounds. Biodistribution in normal mice was characterized by fast blood and soft tissue depuration and combined excretion via the hepatobiliary and urinary systems. Tumour uptake was low, probably due to low lipophilicity but tumour/muscle ratios were favourable as a consequence of high excretion

The objective of this work was to develop a novel 99mTc complex bearing the 5-nitroimidazol-1-yl moiety with recognised selectivity towards hypoxic tissue, as a potential radiopharmaceutical for imaging tumour hypoxia. The new metronidazole derivative (2-amine-3-[2-(2-methyl-5-nitro-1H-imidazol-1-yl)ethylthio]propanoic acid) (L) containing adequate groups to coordinate technetium through the formation of a Tc(I)-tricarbonyl complex was synthesised with adequate yield (33%) and characterised by spectroscopy. Labelling was performed by substitution of three labile water molecules of the technetium tricarbonyl precursor, fac-[99mTc(CO)3(H2O)3]+ with the ligand. A radiochemical purity higher than 90% was achieved and remained unchanged for more than 4 h. The complex has a high stability in plasma, a moderate plasma protein binding and a moderate hydrophilicity. In vitro cell uptake studies showed a ratio between the activity taken up by cells in hypoxia/normoxia of 1.6 ± 0.4 (p < 0.5). Biodistribution in normal mice showed rapid depuration and low uptake in all organs and tissues except liver. Biodistribution in mice bearing induced tumours showed a low tumour uptake, but tumour/muscle ratio was favourable thanks to depuration. Comparison with biological results of other metronidazole derivatives clearly shows that modifications of the chelator are very important and contribute to improve the biological behaviour.

<p>2-fluoro-2-deoxy-D-glucose (FDG) is considered of great interest. Herein,we present the synthesis and preliminary bioevaluation of a 99mTc(CO)3-glucose derivative. Derivatization of glucose at C2 was achieved using the so-called ‘click chemistry’, forming a histidine-like, 1,4-disubstituted triazole adequate as donor atom system for Tc(I)-tricarbonyl complexes. Synthesis of the intermediate azide derivative was achieved by conventional organic chemistry. Because of the efficiency and selectivity of this cycloaddition, labelling was performed in ‘one pot’, adding the tricarbonyl precursor fac-[99mTc(CO)3(H2O)3]+ to the same vial where the click reaction has taken place without any further purification. A single product with radiochemical purity higher than 90%was obtained. It was stable for at least 4 h in reaction milieu and exhibited high hidrophilicity (log P of �1.2) and low binding to plasma proteins (5�1%). Biodistribution in C57BL/6 mice bearing induced Lewis murine lung carcinoma is characterized by low blood and liver uptake and rapid urinary excretion. 99mTc complex showed moderate tumour uptake but significant retention until 2 h post-injection. Soft tissue clearance was fast leading to significantly higher uptake in tumour in comparison to muscle (p = 0.05) at all time points. Overall, biodistribution of our compound was very similar to that of [18F]-FDG. However, tumour uptake was significantly higher for [18F]-FDG, probably because of high hidrophilicity of our derivative that may hinder cell penetration. Similarity to biodistribution of FDG is a promising outcome, and modifications of the chelator and linkermight improve biological results</p>

A summary of the resource.
Activated carbons obtained from rice husk exhibit low specific surface areas when physical activation is applied due to its high silica content. The purpose of this work was to improve textural parameters of powdered activated carbons obtained from rice husk. To avoid the negative influence of the raw material ash content, a leaching step was included in the preparation process. Hydrofluoric acid, in two concentrations (25 and 50 wt %), was used as leaching agent and applied in different stages of the process. Physical activation using water vapor as activating agent was applied. Specific surface area and porosity were evaluated from nitrogen adsorption data. When a leaching step was included, specific surface area values between 700 and 1200 m2/g were obtained. These values are higher than that corresponding to the activated carbon prepared from rice husk not treated with acid (290 m2/g). Pore size distribution depends on the step sequence of the preparation process and on the HF concentration.

Abstract
A summary of the resource.
An increasing global concern regarding energy saving and a growing pharmaceutical interest towards the adoption of industrial continuous processing generate an appropriate scenario for the development of solid dosage forms by moisture activated dry granulation (MADG) processes, which eliminate granulate drying and sizing, and minimize granulation end point control by making it less sensitive to shear and processing time. The aim of this study was to contribute to further enlighten formulation aspects of an attractive granulation process such as MADG, using statistical techniques. A 2^(4-1) fractional factorial design was introduced to study the effect of formulation variables on granulates obtained and on their tablets. Multivariate techniques were applied in the evaluation of the products. Lactose monohydrate and mannitol, povidone K15 and copovidone, and microcrystalline cellulose, partially pregelatinized corn starch and amorphous silica were selected, as diluents, binders and moisture absorbents, respectively. Contact angle and drop penetration time of diluents and moisture absorbents were used to study their wettability. All of the granulates yielded tablets with appropriate aspect and tensile strength, low friability, short disintegration time and in most of the cases, good weight uniformity. The statistical techniques showed the influence of excipient selection in the granulation process. Lactose produced granulates which presented lower median size and more rounded granules, leading to tablets which showed greater weight uniformity than the other diluent mannitol. The binder PVP K15 increased granule mechanical strength, producing finished tablets with higher tensile strength and longer disintegration time than copovidone. The use of a second moisture absorbent improved granulate flowability and contributed to prevent granulate caking. Wettability studies provided useful information for guidance on preliminary excipient selection. In summary, MADG is an innovative and economical process which facilitates quality by design approach.
