A summary of the resource.
The accurate estimation of the free energy changes in solution for supramolecular complexes is one of the most important challenges for quantum chemistry in this century. In general, there is a good correlation between the association constants determined experimentally and the gas phase binding energies calculated using the most popular density functional methods. However, when the receptors adopt very different structures in the gas and condensed phase, this approach does not work, leading to unreliable predictions. We discuss this problem in detail using three popular fullerene receptors: Aida's dimeric porphyrins, buckycatchers I, and II. We found that it is possible to obtain a reliable order of affinities using gas phase binding energies when a special attention is put on analyzing the structure adopted by the receptor in the gas and condensed phases.

Granulomas are responses to persistent nonliving bodies or pathogens, centrally featuring specialized macrophage forms called epithelioid and multinucleated giant cells. The larval stages of the cestode parasites of the Taeniidae family (Taenia, Echinococcus) develop for years in fixed tissue sites in mammals. In consequence, they are targets of granulomatous responses. The information on tissue responses to larval taeniids is fragmented among host and parasite species and scattered over many decades. We attempt to draw an integrated picture of these responses in solid tissues. The intensity of inflammation around live parasites spans a spectrum from minimal to high, parasite vitality correlating with low inflammation. The low end of the inflammatory spectrum features collagen capsules proximal to the parasites and moderate distal infiltration. The middle of the spectrum is dominated by classical granulomatous responses, whereas the high end features massive eosinophil invasions. Across the range of parasite species, much observational evidence suggests that eosinophils are highly effective at killing larval taeniids in solid tissues, before and during chronic granulomatous responses. The evidence available also suggests that these parasites are adapted to inhibit host granulomatous responses, in part through the exacerbation of host regulatory mechanisms including regulatory T cells and TGF-β.

The results from the synthesis of peptides by Fmoc/SPPS on a 2-CTC resin and then lactamization in solution or solid phase for the preparation of cyclopeptides are presented. Both procedures allow the synthesis of the desired compounds in good to very good yield and with high cyclization efficiency for on-resin macrocyclization. In addition, the activities of the corresponding cyclopeptides against the chloroquine-resistant K1 strain of Plasmodium falciparum were evaluated. Cyclo-Cys(Trt)-Gly-Thr(tBu)-Gly-Cys(Trt)-Gly showed potent in vitro and selective activity against this parasite, EC50 = 28 nM.

The role of α‐ and β‐d‐glucose, and d‐gluconate anion, acting as capping agent, on silver nanoparticles (AgNPs) was evaluated using both Raman and density functional theory studies. The particles were synthetized employing glucose, as reducing and blocking agent, in alkaline conditions. Capping agents were characterized through surface enhancement Raman spectroscopy together with modeling performed using silver clusters. Several geometries were optimized, and its energies and wavenumbers were calculated for each of them. Vibration normal modes from d‐glucose isomers and d‐gluconate were obtained and compared with those reported in bibliography, showing a satisfactory correlation. Comparison between experimental and calculated frequencies reveals a preference for d‐gluconate anion acting as capping agent under the necessary presence of d‐glucose on the surface of the AgNPs. The study of energies shows a greater affinity of d‐gluconate for the silver atoms (above −1.3 eV for both geometries) than for other configurations of the glucose molecule (about −0.66 eV in the case of α‐d‐glucose).

La adicción es un desorden neurobiológico en el que el uso repetido de sustancias de abuso corrompe los circuitos neuronales normales de recompensa y de conductas adaptativas, provocando cambios neuroplásticos en dicho circuito. Esta enfermedad crónica tiene graves y duraderas consecuencias para la salud del individuo y la sociedad.[1] El tratamiento farmacológico actual para la adicción a drogas de abuso está enfocado principalmente a disminuir los severos síntomas desarrollados durante la abstinencia o incluso a evitar la recaída. Sin embargo, hasta el momento no existen estrategias exitosas que logren revertir los cambios neurobiológicos implicados en el consumo crónico de una sustancia. En este sentido, sería deseable contar con fármacos que promuevan la generación de neuroplasticidad orientados a restablecer los circuitos neuronales a un estado de no-adicción

Uno de los principales problemas de gestión ambiental de las pequeñas y medianas industrias lácteas, es el destino de los subproductos generados por su actividad industrial, como el suero lácteo. A causa de esto, en las últimas décadas ha aumentado el interés en el aprovechamiento de dicho subproducto (Ganju & Gogate, 2017). El suero lácteo es definido como la sustancia líquida obtenida por separación del coágulo de leche en la elaboración de queso luego de que la caseína es precipitada (Foegeding & Luck, 2002). Existen dos tipos de suero lácteo dependiendo de la metodología utilizada en la elaboración de queso a la hora de la coagulación. El denominado dulce, basado en la acción de la enzima renina a pH 6,5 y el llamado ácido, resultante del proceso de fermentación o adición de ácidos orgánicos o minerales para coagular la caseína, como sucede por ejemplo en la elaboración de quesos frescos

A summary of the resource.
Methotrexate (MTX), a structural analog of folic acid, is widely employed in the treatment of different cancers and autoimmune diseases. Despite the successful results observed, the main disadvantage lies in interpatient variability in the pharmacokinetic and pharmacodynamic parameters. In particular, adverse events and toxicities induced by MTX are a matter of concern and can be the cause of dose reduction or treatment discontinuation. Among the different approaches to reduce MTX therapeutic limitations, pharmacogenomics contributes by considering the effect of inherited genetic differences on those parameters. This review provides an update on MTX pharmacogenomics. It reports the contribution of main gene polymorphisms involved in the influx, efflux, cellular effect, and elimination on MTX toxicity and efficacy, on all the diseases treated with this drug. From the analysis of the data presented in this review, we concluded that only gene polymorphisms MTHFR rs1801133, SLC19A1 rs1051266, and TYMS rs34743033 could influence clinical decision-making.

Searching for prospective vanadium-based agents against Trypanosoma cruzi, the parasite causing Chagas disease, four new [VVO(8HQ–H)(L–2H)] compounds, where 8HQ is 8-hydroxyquinoline and L are tridentate salicylaldehyde semicarbazone derivatives L1–L4, were synthesized and characterized in the solid state and in solution. The compounds were evaluated on T. cruzi epimastigotes (CL Brener) as well as on VERO cells, as mammalian cell model. Compounds showed activity against T. cruzi (IC50 6.2–10.5 μM) of the same order than Nifurtimox and 8HQ, and a four- to sevenfold activity increase with respect to the free semicarbazones. For comparison, [VVO2(L–H)] series was prepared and the new [VVO2(L3–H)] was fully characterized. They showed negligible activity and low selectivity towards the parasite. The inclusion of 8HQ as ligand in [VVO(8HQ–H)(L–2H)] compounds led to good activities and increased selectivity towards the parasite with respect to 8HQ. 51V NMR experiments, performed to get insight into the nature of the active species, suggested partial decomposition of the compounds in solution to [VVO2(L–H)] and 8HQ. Depending on the dose, the compounds act as trypanocide or trypanostatic. A high uptake of vanadium in the parasites (58.51–88.9% depending on dose) and a preferential accumulation in the soluble protein fraction of the parasite was determined. Treated parasites do not seem to show a late apoptotic/necrotic phenotype suggesting a different cell death mechanism. In vivo toxicity study on zebrafish model showed no toxicity up to a 25 µM concentration of [VVO(8HQ–H)(L1–2H)]. These compounds could be considered prospective anti-T. cruzi agents that deserve further research.
