A summary of the resource.
A dynamic development of methodologies of analytical flow injection measurements during four decades since their invention has reinforced the solid position of flow analysis in the arsenal of techniques and instrumentation of contemporary chemical analysis. With the number of published scientific papers exceeding 20 000, and advanced instrumentation available for environmental, food, and pharmaceutical analysis, flow analysis is well established as an extremely vital field of modern flow chemistry, which is developed simultaneously with methods of chemical synthesis carried out under flow conditions. This review work is based on almost 300 original papers published mostly in the last decade, with special emphasis put on presenting novel achievements from the most recent 2–3 years in order to indicate current development trends of this methodology. Besides the evolution of the design of whole measuring systems, and including especially new applications of various detections methods, several aspects of implications of progress in nanotechnology, and miniaturization of measuring systems for application in different field of modern chemical analysis are also discussed.

Las proteasas o peptidasas, son enzimas que catalizan la hidrólisis del enlace peptídico de las proteínas. Se describen cinco clases de acuerdo a su mecanismo catalítico: proteasas aspárticas, de cisteína, metálicas, de serina y de treonina. Estas enzimas desempeñan papeles clave en todos los organismos vivos. A pesar de catalizar la misma reacción, la especificidad por el sustrato suele ser marcadamente diferente para cada proteasa, haciéndolas moléculas únicas. El estudio de nuevas peptidasas, aporta información valiosa permitiendo una mayor comprensión de las propiedades que definen la especificidad y la relación estructura-función, además de generar nuevas herramientas biotecnológicas. En este contexto, se trabajó con frutos maduros de Bromelia antiacantha Bertol., una especie autóctona, perteneciente a la familia Bromeliaceae. Partiendo del extracto de estos frutos y conociendo los puntos isoeléctricos de sus componentes proteicos, se empleó una metodología de purificación simple que permitió separar tres nuevas peptidasas que denominamos: Antiacanthaína A (AntA), Antiacanthaína B (AntB) y Antiacanthaína C (AntC). La caracterización bioquímica de las tres enzimas mostró que ellas tienen propiedades similares y típicas de las cisteín peptidasas

A lectin was isolated from fruiting bodies of the mushroom Gymnopilus spectabilis (GSL) by ionic exchange chromatography. The lectin agglutinates mouse red cells exhibiting broad specificity towards several monosaccharides including the N-acetylneuraminic acid. Agglutination was also inhibited by the glycoproteins: fetuin, lactoferrin, and recombinant erythropoietin. GSL is a glycoprotein possessing 16 % of carbohydrates; the SDS-PAGE showed two bands with molecular mass of 52.1 and 64.4 kDa. Isoelectric focusing displayed microheterogeneity, with two bands at pIs 5.1 and 5.3. The lectin was stable between pH 2 and pH 8 while at pH 10, the agglutination decayed to 50%of initial activity. Incubation at 40 and 80 °C led to 50 and 100 % loss in activity of the lectin, respectively. Synthesized GSL-Sepharose interacts with serum pregnant mare gonadotropin, and at least two subpopulations of this glycoprotein were separated. There was no interaction between transferrin and soluble GSL while a partial recognition was achieved with GSL-Sepharose. The terminal sialic acid seems to play an active role in modifying the interaction with GSL, depending if the lectin is in a soluble or immobilized form. The purified lectin inhibited in vitro the growth of Staphylococcus aureus and Aspergillus niger.

Biological glycans mediate several physiological processes, thus altered glycosylation patterns can lead to different diseases such as autoimmune, infectious, chronic anti-inflammatory diseases, or even cancer. In fact, alterations in fucosylation in either N- or O-glycans are among the most frequent changes in glycosylation patterns associated with cancer. Therefore, elucidation of the role of glycoconjugate glycans is essential for understanding the development of pathologies where they are involved. In this sense glycosidases are excellent tools, since they catalyse the selective removal of sugar residues, allowing the evaluation of changes in their biological role due to glycan removal. This work describes the purification and characterization of a α-fucosidase from the fungus Dichostereum sordulentum 1488. It is a homodimer with a molecular weight of 214 kDa and optimum pH and temperature of 4.0 and 70 ◦C respectively. It has a KM of 0.27 mM and VMax of 3.3 μmoles PNP/ min per mg for the substrate p-nitrophenyl-α-L-fucopyranoside, showing a substrate inhibition profile. It showed high specificity for the hydrolysis of fucose linked by α-(1,2) bonds. The identification, purification, and characterization of this new α-fucosidase is highly relevant for enlarging the availability of glycosidases for use as tools for glycan elucidation.

Sinorhizobium meliloti CE52G strain produces a periplasmic laccase that has been purified by a two-step procedure involving heat treatment and immobilized metal affinity chromatography (IMAC). The fraction with laccase activity retained its original activity after 24 h of incubation at pH between 4.0 and 8.0 and after 3 h of incubation at 70 ◦C, pH 7.2 and supplemented with 1.3M(NH4)2SO4. It proved to be a homodimeric protein with an apparent molecular mass of 45 kDa each subunit and an isoelectric point of 6.2. CE52G laccase was inhibited by halides (NaF and NaCl), ions (Fe3+, Mn2+, and Cu2+), sulfhydryl organic compounds (�-mercaptoethanol and reduced glutathione), and electron flow inhibitors (NaCN and NaF). Laccase activity was strongly enhanced by (NH4)2SO4, Na2SO4, and K2SO4. The effects of all these agents, as well as the probability of a partially unfolding polypeptide chain to enhance the interaction between the substrate and the active site, are discussed. CE52G laccase is a pH- and thermo-stable protein with promising biotechnological applications.

A protease classified as trypsin was isolated and purified from the pyloric caeca of two species of fishes: Brevoortia spp (menhaden) and Mugil spp. (mullet). The characterization of both enzymes as trypsin was based on their molecular weight (24 kDa) determined by SDS-PAGE, their inhibition by some known trypsin inhibitors (PMSF, SBTI, TLCK and benzamidine), and their ability to hydrolyze the synthetic trypsin substrate N-α-Benzoyl-DL-arginine-p-nitroanilide (BAPA). Menhaden trypsin had maximum activity at pH 9.5 and was stable for 30 min between pH 6.0 and 10.0 at 0, 10 and 25C. On the other hand, mullet trypsin showed maximum activity at pH's from 7.8 to 9.0, and was stable over a wider pH range (7.0–10.0). The optimum temperature for menhaden and mullet trypsin was 63C and 60C respectively. Thermostability for menhaden trypsin was up to 50C, whereas mullet trypsin was stable up to 60C. The Km(BAPA) values for menhaden trypsin were conserved in the temperature range from 10 to 30C, while for mullet trypsin the Km(BAPA) was conserved between 10 and 25C.

An NADPH-dependent enzyme that reduces ethyl 2-methylacetoacetate stereoselectively to ethyl (2 R)-methyl-(3 S)-hydroxybutanoate was purified 730-fold from Escherichia coli. The N-terminal amino acid sequence data obtained from the purified reductase were used to search the E. coli genome, and a single match was found at the start of the yqhE open reading frame. The YqhE protein had been identified previously by Yum et al. as a 2,5-diketo-d-gluconate reductase on the basis of sequence similarity to other bacterial homologues [Yum, D.-Y.; Lee, B.-Y.; Pan, J.-G. Appl.Environ. Microbiol.1999, 65, 3341–3346]; however, it had not been examined for β-keto ester reductions. Our results thus link a key enzyme in the microbial production of ascorbate with stereoselective β-keto ester reductions, two important fields in biocatalysis. The purified YqhE reductase accepts ethyl acetoacetate and a variety of 2-substituted derivatives, and its sequence is similar to other aldose reductase superfamily members that also reduce α-substituted β-keto esters to syn-(2 R,3 S) alcohols.<br /><br />

l-Arabinose isomerase is an intracellular enzyme that can convert l-arabinose to l-ribulose and d-galactose to d-tagatose, a promising but rare nutraceutical. Most of l-arabinose isomerases purified up to date employed the combination between DNA recombinant technology and affinity chromatography based on poly-histidine tail recognition, but few of the enzymes were obtained and purified in a non-recombinant way. For these reasons, a specific affinity bioadsorbent containing l-arabitol as ligand, a competitive inhibitor of the enzyme, was designed and synthesized for achieving pure preparations of the enzyme l-arabinose isomerase from wild-type Enterococcus faecium DBFIQ E36 strain, isolated from raw cow milk. The two-step purification procedure consisted in fractionation by ammonium sulphate precipitation followed by affinity chromatography with obtained bioadsorbent, allowing the purification, to electrophoretical homogeneity, of target enzyme. Characterization studies were performed with purified l-arabinose isomerase in order to increase knowledge of their physicochemical properties. In this sense, enzyme exhibited an optimum temperature of 50 °C and optimum pH of 7.0, maintaining good stability in the ranges 20–45 °C and pH 6.5–8. Ki were calculated, employing d-galactose as substrate, for l-arabitol and l-ribitol, achieving values of 7.9 mM and 183 mM, respectively. Km and Vmax values obtained were 35 mM and 81 U mg−1 at 50 °C, respectively. Mass spectrometry assay revealed a 48 kDa monomer whereas gel permeation chromatography achieved a 187 kDa molecular weight for native enzyme. Finally, 2D-electrophoresis and isoelectrofocusing analysis revealed an isoelectric point value of 3.80. Results have unveiled both an acidic nature and promising properties for l-arabinose isomerase isolated from E. faecium DBFIQ E36.

Microtubules are non-covalent cylindrical polymers formed by alpha- and beta-tubulin heterodimer units, crucial for cell division, intracellular transport, motility and differentiation. This makes them very attractive pharmacological targets exploited to develop different drugs such as anthelmintics, antifungals, and antineoplastics. In this work, in order to establish an in vitro target-based screen to integrate to the search for new anthelmintics, we explored the extraction of native assembly-competent tubulin from two helminth parasites: Mesocestoides vogae tetrathyridia (syn. corti, Cestoda: Cyclophyllidea), a useful cestode biological model, and Haemonchus contortus, a sheep gastrointestinal nematode of interest in livestock production. For this purpose, a novel tubulin affinity chromatography procedure was employed, based on the binding capacity of TOG (Tumor Overexpressed Gene) domain from MAPs (microtubule-associated proteins). The TOG domain of the protein Stu2 from Saccharomyces cerevisiae fused to GST (glutathione S- transferase) were produced in E. coli, and the immobilized recombinant proteins allowed for native tubulin extraction from parasites. The binding capacity of TOG1 affinity column (3.6%) was estimated using commercial porcine brain tubulin. A total amount of up to 126 μg of M. vogae tubulin was purified, whereas H. contortus tubulin co-eluted with glutamate dehydrogenase enzyme. The identity of tubulins was confirmed by western blotting and mass spectrometry. The abundance of tubulin estimated in M. vogae was 10% soluble extract, which probably could explain differences observed between tubulin purification results of both helminth parasites.

<p>Arthrospira platensis has stood out, especially due to its high protein content and its potential for food application. However,<br />the exploration of protein concentration techniques of this microalgae still presents a great challenge. Thus, the objective<br />of this study was to evaluate the purification of A. platensis proteins by applying two different aqueous two-phase systems<br />(ATS) composed of potassium phosphate and ethanol, and sodium citrate and ethanol, in addition to the association with<br />membrane separation process (MSP), and the characterization of the fractions obtained. ATS under the optimised conditions<br />(19% potassium phosphate and 30% ethanol, and 19.5% sodium citrate and 29% ethanol) resulted in 1.27 and 1.31 purity, and 77.45% and 78% recovery, respectively. The sodium citrate and ethanol ATS showed significantly greater purity; thus, the upper phase was submitted to MSP. The association of ATS and MSP increased the recovery (~ 87%) and the purity (1.5). The top phase of ATS showed higher protein content (61%) than the bottom (5.9%), confirming the protein preference to the top. When the MSP was evaluated, the protein was increased about 8.0%, and antioxidant activity was kept (42 and 40% to top phase and ATS/MSP concentrate, respectively). Emulsions formed by ATS top phase showed a lower droplet size (33.69 μm D4.3 and 6.02 μm D3.2), and a significantly lower degree of overall destabilization (0.04 × 10–<br />6 s−1 kh and 3.54 × 10–3 s−1 ks) than those stabilized with bottom phase and MSP/ATS concentrate. ATS associated with membrane process showed efficiency in the concentration A. platensis protein, being able to be an alternative to be used in industrial systems.</p>

Paramyosin, a vaccine candidate in different helminthiases, was purified from the adult liver fluke Fasciola hepatica using two different procedures. The first started with a crude extraction of paramyosin in high-salt buffer followed by gel filtration chromatography and two precipitation-solubilization cycles; in the second, anion exchange chromatography replaced the gel filtration step. In both cases, the apparent molecular weight of the purified protein determined by sodium dodecyl sulfate gel electrophoresis under reducing and non-reducing conditions was 97 kDa and 200 kDa, respectively. The molecular weights were consistent with the presence of a dimeric protein linked by disulfide bridges. Western blot analysis showed that the dimeric and monomeric forms were both recognized by an antiserum raised against the F. hepatica 97 kDa band (α-FhPmy), and by an anti-Schistosoma mansoni paramyosin immune serum. Immunohistochemistry using α-FhPmy demonstrated the localization of paramyosin within the subtegumental muscle and in muscle cells surrounding the gut of adult parasites. We also observed labeling of extramuscular structures like testes, surface lamellae of the gut and the tegument of adult flukes. Achyrocline satureioides (Lam) D.C (Compositae) is a native medicinal plant of South America traditionally utilized for its anti-inflammatory, sedative and anti-atherosclerotic properties among others. Neuroprotective effects have been reported in vivo and could be associated to its elevated content of flavonoid aglycones. In the present study we performed the isolation and structure elucidation of the major individual flavonoids of A. satureioides along with the in vitro characterization of their individual antioxidant and neuroprotective properties in order to see their putative relevance for treating neurodegeneration. Exact mass, HPLC-MS/MS and 1H NMR identified dicaffeoyl quinic acid isomers, quercetin, luteolin, isoquercitrin, and 3-O-methylquercetin as the mayor polyphenols. Flavonoids intrinsic redox properties were evaluated in the presence of the endogenous antioxidants GSH and Ascorbate. Density Functional Theory (DFT) molecular modeling and electron density studies showed a theoretical basis for their different redox properties. Finally, in vitro neuroprotective effect of each isolated flavonoid was evaluated against hydrogen peroxide-induced toxicity in a primary neuronal culture paradigm. Our results showed that quercetin was more efficacious than luteolin and isoquercitrin, while 3-O-methylquercetin was unable to afford neuroprotection significantly. This was in accordance with the susceptibility of each flavonoid to be oxidized and to react with GSH. Overall our results shed light on chemical and molecular mechanisms underlying bioactive actions of A. satureioides main flavonoids that could contribute to its neuroprotective effects and support the positive association between the consumption of A. satureioides as a natural dietary source of polyphenols, and beneficial health effect.

Two different families of N-oxide containing heterocycles were evaluated as in vitro growth inhibitors of T. cruzi. Both families of heterocycles were selected from our in-house library of compounds as analogues of active anti-T. cruzi N-oxide containing heterocycles. Derivatives from pyrimido[1,2-a]quinoxaline 6-oxide family were poorly active at the assayed doses. However, phenazine 5,10-dioxide derivatives displayed good to excellent anti-T. cruzi activities. The anti-T. cruzi activity of phenazine derivatives was related to substituent' electronic descriptors, σp−. Derivatives 19, 20 and 23 were the most cytotoxic compounds against the protozoan and became excellent hit for further structural modifications.

Changes in climatic and environmental conditions can affect both, plant chemical molecular and atomic composition. Therefore, biomass carbon isotope composition is frequently used to gain information about the environmental conditions at the time a plant thrives. In this field, most studies use isotope values obtained from bulk samples. However, these values are a weighted average of the different plant compounds. The isotopic characterization of individual compounds represents a relevant analytical advance able to differentiate the isotopic composition of the main plant components i.e., polysaccharides, lignin, polypeptides, lipids, waxes, etc. Here, we investigate the link between molecular and isotope composition of bark and lignin extracted from three Eucalyptus species (E. grandis, E. dunnii and a hybrid E. grandis X E. dunnii) using pyrolysis compoundspecific isotope analysis (Py-CSIA). This technique, although not widely used, has been previously developed and applied, with different variants, to characterize plant products including lignin, sugars, and terpenes. Retrieved results for both bark and lignin samples showed the isotopic differences regarding the diverse biogenic groups, accounting for a 13C-enrichment in polysaccharides up to 2.5‰ compared to lignin methoxyphenols, which is consistent with the fractionation during metabolization of the plant. A strong positive correlation was found between the bulk isotope and compound-specific values, especially when applying a weight factor related to the relative abundance of each compound, which support the analytical suitability of this technique. Despite that bark from different origins provide very similar pyrolysis patterns, isotopic composition of relevant specific compounds can make the difference giving additional valuable information.

Eukaryotic diphthine synthase, Dph5, is a promiscuous methyltransferase that catalyzes an extraordinary N,O-tetramethylation of 2-(3-carboxy-3-aminopropyl)-l-histidine (ACP) to yield diphthine methyl ester (DTM). These are intermediates in the biosynthesis of the post-translationally modified histidine residue diphthamide (DTA), a unique and essential residue part of the eukaryotic elongation factor 2 (eEF2). Herein, the promiscuity of Saccharomyces cerevisiae Dph5 has been studied with in silico approaches, including homology modeling to provide the structure of Dph5, protein–protein docking and molecular dynamics to construct the Dph5-eEF2 complex, and quantum mechanics/molecular mechanics (QM/MM) calculations to outline a plausible mechanism. The calculations show that the methylation of ACP follows a typical SN2 mechanism, initiating with a complete methylation (trimethylation) at the N-position, followed by the single O-methylation. For each of the three N-methylation reactions, our calculations support a stepwise mechanism, which first involve proton transfer through a bridging water to a conserved aspartate residue D165, followed by a methyl transfer. Once fully methylated, the trimethyl amino group forms a weak electrostatic interaction with D165, which allows the carboxylate group of diphthine to attain the right orientation for the final methylation step to be accomplished.

Abstract
A summary of the resource.
<p>En el presente trabajo se estudian los distintos tipos de alteraciones de los alimentos clasificadas según dos grupos principales: biológicas y abióticas.</p>
<p>A su vez se consideran posteriormente, en forma detenida, las principales formas de alteración que pueden presentarse en los alimentos.</p>
<p>La alteración de los alimentos, cualquiera sea el tipo considerado, es una función de varios factores que pueden clasificarse en: a) intrínsecos o sea los que dependen del alimento (pH, aw, potencial redox, composición química, componentes antimicrobianos y estructuras biológicas); b) extrínsecos o sea las propiedades del medio ambiente (temperatura de almacenamiento, humedad relativa del ambiente y presencia y concentración y gases); c) tecnológicos o de procesamiento; d) implícitos.</p>
<p>Todo este estudio permite un mejor conocimiento del alimento, un mejor manejo de la tecnología empleada para su almacenaimiento o conservación y el logro de alimentos de una calidad total superior.</p>
