A summary of the resource.
Searching for prospective vanadium-based drugs for cancer treatment, a new series of structurally related [VIVO(L − 2H)(NN)] compounds (1–8) was developed. They include a double deprotonated salicylaldimine Schiff base ligand (L-2H) and different NN-polypyridyl co-ligands having DNA intercalating capacity. Compounds were characterized in solid state and in solution. EPR spectroscopy suggests that the NN ligands act as bidentate and bind through both nitrogen donor atoms in an axial-equatorial mode. The cytotoxicity was evaluated in human tumoral cells (ovarian A2780, breast MCF7, prostate PC3). The cytotoxic activity was dependent on type of cell and incubation time. At 24 h PC3 cells presented low sensitivity, but at 72 h all complexes showed high cytotoxic activity in all cells. Human kidney HEK293 and ovarian cisplatin resistant A2780cisR cells were also included to evaluate selectivity towards cancer cells and potency to overcome cisplatin resistance, respectively. Most complexes showed no detectable interaction with plasmid DNA, except 2 and 7 which depicted low ability to induce single strand breaks in supercoiled DNA. Based on the overall cytotoxic profile, complexes with 2,2´-bipyridine and 1,10-phenanthroline ligands (1 and 2) were selected for further studies, which consisted on cellular distribution and ultrastructural analyses. In the A2780 cells both depicted different distribution profiles; the former accumulates mostly at the membrane and the latter in the cytoskeleton. Morphology of treated cells showed nuclear atypia and membrane alterations, more severe for 1. Complexes induce different cell death pathways, predominantly necrosis for 1 and apoptosis for 2. Complexes alternative mode of cell death motivates the possibility for further developments.

In Latin America Chagas disease is an endemic illness caused by the parasite Trypanosoma cruzi (T. cruzi), killing more people than any other parasitic disease. Current chemotherapies are old and inadequate, thus the development of efficient ones is urgently needed. Vanadium-based complexes have been shown to be a promising approach both against parasitic diseases and cancer and this study aims to achieve significant advances in the pursue of effective compounds. Heteroleptic vanadium complexes of Schiff bases and polypyridine compounds were prepared and their stability in solution evaluated by EPR (Electronic Paramagnetic Resonance) and NMR spectroscopy. Their in vitro activities were evaluated against T. cruzi and a set of cells lines representative of human cancer conditions, namely ovarian, breast and prostate cancer. In T. cruzi, most of the complexes depicted IC50 values in the low μM range, induced changes of mitochondrial membrane potential and apoptosis. In cancer cells, complexes showed good to moderate activity and in metastatic cells (prostate PC3), some complexes inhibited the migratory ability, this suggesting that they display antimetastatic potential. Interestingly, complex 5 seemed to have a dual effect being the most cytotoxic complex on all cancer cells and also the most active anti-T-cruzi compound of the series. Globally the complexes showed promising anticancer and anti T. cruzi activities and also displayed some characteristics indicating they are worth to be further explored as antimetastatic drugs.

Ten structurally related homoleptic complexes, including three new compounds, were synthesized: five [VIVO(L-H)2] and five [VVO(OCH3)(L-H)2] compounds, where L–H = L1-H–L5-H are monodeprotonated L1–L5 8-hydroxyquinoline derivatives. The compounds were characterized in the solid state and in solution. All complexes show inhibitory activity against Trypanosoma cruzi and Leishmania infantum, most within the 0.2–5 μM range, T. cruzi being more sensitive to the compounds than L. infantum. The compounds show high selectivities towards the parasite, evaluated using VERO cells as a mammalian cell model, as well as higher activity against T. cruzi than the reference antitrypanosomal drug nifurtimox. The antitrypanosomal activity depends on the nature of the substituents on the 8-hydroxyquinoline moiety, [VIVO(L1-H)2] (L1 = 5,7-dichloroquinolin-8-ol), with IC50 = 0.21 μM, being the most active and selective compound of the series against T. cruzi, and one of the most active against L. infantum (IC50 = 2.5 μM). A trypanocide effect of [VIVO(L1-H)2] was observed when incubating parasites with 10× the IC50 value and a late apoptotic/necrotic cell death mechanism is suggested. A preferred association with the soluble protein fraction of cells is also observed (49.4% of total vanadium association). [VIVO(L1-H)2] is shown to affect not only the infection potentiality of cell-derived trypomastigotes but also the proliferation of intracellular amastigotes, the two parasite stages more relevant in the mammalian host after infection. By measuring circular dichroism spectra of solutions containing either human serum apo-transferrin (apo-HTF) or bovine serum albumin (BSA) upon adding the compounds, it is shown that both the free ligands and the VIV- and VV-complexes bind to these proteins. From data determined from fluorescence quenching measurements with BSA, the determined Stern–Volmer constants are typically slightly higher for the ligand precursors than for the corresponding [VIVO(L1-H)2] compounds, the calculated conditional binding constants at pH = 7.4 being ca. 105–106. The fluorescence competition measurements with DNA bound to ethidium bromide typically yield relatively small decreases in fluorescence, VIV- and VV-complexes with L1 showing higher quenching.

Trypanosomatid parasites and fungi cause highly prevalent diseases. Ergosterol plays a crucial role in the structure and normal function of the membrane of these microorganisms. It is absent in mammals. Many enzymes of the ergosterol biosynthesis route have been recognized as molecular targets for drugs. Currently, a variety of organic drugs that interfere with sterol biosynthesis are employed to treat fungal infections, and some of them have been proposed as potential treatments for trypanosomiasis. Coordinating these organic drugs to metal ions may positively impact the biological performance by altering relevant physicochemical properties. Additionally, their organometallic derivatization could improve the effectiveness. This comprehensive review aims to highlight the considerable promise of metal-based compounds as inhibitors of essential pathways or key enzymes of these microorganisms, with focus on the ergosterol biosynthesis pathway. Additionally, bioactive metal compounds that have demonstrated experimental efficacy in acting on enzymes within this pathway although not designed for this purpose were included. Cytochrome P450 enzymes and efflux transporters, expressed in the intestine and/or in the liver, play important roles in drug clearance and oral bioavailability. The relative contribution of transporters and enzymes in drug metabolism is still controversial. Some antiepileptic drugs, such as carbamazepine, phenytoin and phenobarbital (phenobarbitone), show time-dependent and dose-dependent pharmacokinetics due to their inductive effect on both efflux transporters and enzymes. However, steady-state plasma drug concentrations for each antiepileptic drug do not relate to oral daily dose in the same way, with decreased or increased apparent clearance according to the drug. A multicompartment pharmacokinetic model was developed in order to explain these different behaviours using a single mechanism of inductive action. The key for solving these apparent dissimilarities was to consider in the model the unique physiological connection that intestine, liver and bloodstream have. Efflux transporters not only enhance enzymatic competition in relation to first-order processes, but also change the predominance of some elimination routes. For instance, the carbamazepine-10,11-epoxide formation increases at the expense of other carbamazepine metabolites, enhancing both the systemic and presystemic elimination of parent drug. A summary of the resource.
Macrocycles possess desirable properties that make them promising candidates for the discovery of novel drugs. They present structural features to favor bioactive conformations, selectivity to the receptors, cell permeability and metabolic stability. More effective and nontoxic drugs to cure human African trypanosomiasis, Leishmaniasis and Chagas disease are needed, especially because resistance has been detected. Therefore, major efforts should be made for investigation in new bioactive compounds exhibiting different mechanisms of action. Macrocycles might fulfill the expectations for the development of new drugs to treat those diseases. In the current review, we focus on macrocycles exhibiting biological activities as antitrypanosomal and/or antileishmanial. The isolation, synthetic and biological studies of this class of compounds published from 2005 to 2014 are summarized.

A summary of the resource.
Distamycin, a natural polyamide containing three heterocycle rings with a polar end, has inspired several groups to prepare synthetic analogues, which proved to have anti-trypanosomal and anti-tumoral activity. We describe the synthesis of bi and tri thiazoles amides that harbor different substitutions at their ends and the evaluation of their anti-Trypanosoma brucei activity. The most active compound 10b showed better biological activity (EC50 310 nM and selectivity index 16) than the control drug nifurtimox (EC50 15 μM and selectivity index 10). Studies on the mode of action show that the parasiticidal activity of 10b originates from disruption of lysosomal homeostasis, which is followed by release of redox active iron, an increase in oxidizing species and collapse of cell membrane integrity. In this respect, our study suggests that non-charged lipophylic distamycins destabilize cell membranes.

OBJECTIVE: The aim of the study was to analyze retrospectively carbamazepine (CBZ) and valproic acid (VPA) salivary data collected from epileptic children during a 3-year period. METHODS: Saliva samples stimulated by citric acid were assayed by FPIA method. One hundred and three patients (aged 1-14 years) were in CBZ or VPA monotherapy or in CBZ-VPA combined therapy. RESULTS: VPA salivary levels were linearly related with daily dose, but a non-linear relationship was found for CBZ, in patients under monotherapy. VPA did not alter saliva CBZ concentration. Conversely, CBZ reduced VPA salivary levels. Non-responsive children displayed higher VPA concentrations. CBZ levels in uncontrolled patients showed non-significant difference in relation with controlled subjects even though their daily doses were higher. CONCLUSION: Citric acid stimulated saliva is reliable enough to perform therapeutic drug monitoring. Saliva drug levels in non-responsive patients would be explained according to the generalized efflux transporter overexpression hypothesis.

The CreA protein mediates carbon catabolite repression in the fungus Aspergillus nidulans. Its DNA-binding domain belongs to the Cys2-His2 class, binding specifically to a 5' SYGGRG 3' nucleotide sequence. EMSA experiments showed that the CreA(G27D) mutation resulted in a 30-fold increase of the Kdiss, and footprinting revealed a altered pattern of protein/DNA contacts. We modeled the CreA and the CreA(G27D) complexes in silico. A 15 ns molecular dynamics simulation of the solvated CreA(G27D) and CreA models was carried out using the MOE 2007.09 suite and the Amber99 force field. We have focused our analysis in residues Arg14, Glu16, His17, and Arg20 and Arg44, Asp46, and Arg50, previously, shown to be responsible for the specific contacts of the two Zn fingers. The electrostatic and the total potential energies showed the CreA(G27D) mutation to decrease the affinity of the complex, in agreement with the Kdiss's values. The in silico approach highlighted the role of the inter-finger linker. We identified several differential structural characteristics of the CreA and CreA(G27D)/DNA complexes and observed that the latter resulted in a lower dynamic flexibility of the complex.

Abstract

Choosing an appropriate treatment for chronic pain remains problematic, and despite the available medication for its treatment, still, many patients complain about pain and appeal to the use of cannabis derivatives for pain control. However, few data have been provided to clinicians about the pharmacokinetic drug-drug interactions of cannabinoids with other concomitant administered medications. Therefore, the aim of this brief review is to assess the interactions between cannabinoids and pain medication through drug transporters (ATP-binding cassette superfamily members) and/or metabolizing enzymes (cytochromes P450 and glucuronyl transferases).

A summary of the resource.
We employ femtosecond laser pulses s80 fs, 1.59 eV, and 80 MHzd to study the optical second-harmonic sSHd response of PbxCd1−xTe ternary alloys sx about 0.2d grown by the vertical Bridgman method. The alloy segregates into a Pb-rich and a Cd-rich phase, the latter dominating the SH response of the ternary alloy by at least two orders of magnitude. Several sample regions show a regular layer-by-layer accommodation of the Pb-rich and Cd-rich phases as seen by a periodic alternation of the alloy’s SH response on a ,10-mm length scale. Furthermore, we employ polarization-resolved SH imaging as well as SH imaging at different azimuthal angles to obtain spatially resolved mappings of the sample, which are sensitive to the composition as well as the growth orientation of the PbxCd1−xTe material system. We observe an azimuthal phase shift of approximately 30° between coherent macroscopic regions sseveral mm2d in the Cd-rich phase of the ternary alloy. We interpret these regions as large area crystalline grains of s111d and s411d crystal orientations and approximately equal composition. Hence, SH imaging is shown to spatially resolve regions of different growth directions within the PbxCd1−xTe sample.

In this study sludge wash-out was evaluated as a strategy to start-up the Anammox process in order to establish it in a shorter period of time. Sludge from a domestic wastewater treatment plant (WTP) was used to seed two (RI and RII) anaerobic sequencing batch reactors (ASBR). During the start-up period RI was operated as a continuous stirred tank reactor (CSTR) using a dilution rate of 0.2d-1, which promoted the sludge wash-out. After this period, the remaining sludge was retained in the reactor. The reactor RII was operated as an ASBR throughout the study period with a high cell retention. The performance of the two reactors in terms of nitrogen removal was compared over a period of 380 days. During the last RI operation phase the specific nitrogen removal rate increased exponentially, attaining values of 85mgN/gTSS.d. However, a rate of 190mgN/gTSS.d in the batch test under optimal conditions was achieved. The specific nitrogen removal rate remained almost constant for RII with a mean value of 6mgN/gTSS.d being observed during the operation period. The rate for the RII batch test was 20mgN/gTSS.d. These results confirm that the higher TSS in RII (reactor with high cell retention) was not effective in terms of N removal improvement. Anammox-like bacteria were found using fluorescence in situ hybridization (FISH) in reactor RI after 225 days and a new Anammox species was identified.

Background The sawfly family Pergidae is best represented in South America, and it is the third largest family in the suborder Symphyta. Tequus is a Neotropical genus that has been reported in association with host plants of the genus Solanum (Solanaceae), with little information about the life history of its members. Tequus schrottkyi (Konow, 1906) was described from Paraguay, without any information about its biology and host plant. New information We report the first record of T. schrottkyi from Uruguay, with information on its host plant and details of its biology. The identification was based on morphology, DNA barcode is provided to allow identification using molecular characters. This sawfly species is associated with Solanum commersonii, a native plant common in Uruguay. Tequus schrottkyi presents several generations between March and July. The larvae feed on leaves and spin a silk cocoon in the soil in which they pupate. The adults exhibit sexual dimorphism, the female being larger than the male and with a different color pattern. The eggs are laid individually in the leaf margins into the leaf tissue. The larvae are unpalatable to a generalist predator, possibly due to defensive compounds sequestered from their host plant, known to contain toxic compounds.

The Brownian kernel is usually assumed to describe pure diffussion-limited cluster-aggregation processes. In this work |b we show that this assumption is correct for simulated data. For experimental data |b however |b significant deviations were observed although the system was aggregated at an electrolyte concentration well above the critical coagulations concentration. This indicates that residual cluster-cluster interactions are not completely absent in real experimental systems. In order to improve the description of the experimental data |b we developed a kernel that considers a monomer-monomer sticking probability explicity and accounts for the possibility of multiple monomer-monomer contacts in the cluster collision area. The proposed kernel agrees excellently with the experimental cluster-size distribution and the corresponding scaling function

This study compares the performance of Hanseniaspora vineae (Hv) under mixed culture fermentation with Saccharomyces cerevisiae, with a conventional control (Sc) for the production of base wine. Hv exhibited fermentation kinetics comparable to controls when its nutritional requirements were satisfied. Volatile acidity in Hv from one batch was significantly lower (–0.3 g/L) compared to the controls, while in the other two batches the levels were equal. Interestingly, Hv trials showed a significant degradation of malic acid (–0.9 g/L on average) without variations of pH. Even though the matrix influenced the volatile composition, the characteristic features of H. vineae remained intact, with 24-fold higher levels of 2-phenylethyl acetate. There was no higher production of 2-aminoacetophenone, albeit tryptophan (known as a 2-aminoacetophenone precursor) is a preferred amino acid for H. vineae. Results suggest the potential of H. vineae to produce base wine with a broad spectrum of aroma compounds that increase flavor complexity, if obtained from nutrient-balanced grape musts.

Family is a major determinant of children’s and adolescents’ eating behavior. The objectives of the present study were to assess diet quality, eating habits, satisfaction with life, family life, and food-related life in mother–father–adolescent triads, and to identify profiles of families according to family members’ diet quality. Questionnaires were administered to a sample of 300 two-parent families with one child over the age of 10 in the city of Temuco (Chile), including the Adapted Healthy Eating Index (AHEI), Satisfaction with Life Scale (SWLS), Satisfaction with Food-related Life (SWFoL) scale, Satisfaction with Family Life (SWFaL) scales, and questions relating to their eating habits. Positive relationships were found between the diet quality of the family members, particularly between mothers and adolescents. Three family profiles with different diet qualities were identified: “families with an unhealthy diet” (39.3%), “families in which mothers and adolescents have healthy diets, but the fathers’ diets require changes” (14.3%), and “families that require changes in their diet” (46.4%). These findings stress the key role of mothers in determining family diet quality and suggest a positive relationship between diet quality and satisfaction with life.
