The reactivity of alkali&ndash;manganese(II) and alkali trifluoroacetates towards amorphous SiO2 (a-SiO2) was studied in the solid-state. K4Mn2(tfa)8, Cs3Mn2(tfa)7(tfaH), KH(tfa)2, and CsH(tfa)2 (tfa = CF3COO&ndash;) were thermally decomposed under vacuum in fused quartz tubes. Three new bimetallic fluorotrifluoroacetates of formulas K4Mn3(tfa)9F, Cs4Mn3(tfa)9F, and K2Mn(tfa)3F were discovered upon thermolysis at 175 &deg;C. K4Mn3(tfa)9F and Cs4Mn3(tfa)9F feature a triangular-bridged metal cluster of formula [Mn3(&mu;3-F)(&mu;2- tfa)6(tfa)3]4&minus;. In the case of K2Mn(tfa)3F, fluoride serves as an inverse coordination center for the tetra hedral metal cluster K2Mn2(&mu;4-F). Fluorotrifluoroacetates may be regarded as intermediates in the trans formation of bimetallic trifluoroacetates to fluoroperovskites KMnF3, CsMnF3, and Cs2MnF4, which crystal lized between 250 and 600 &deg;C. Decomposition of these trifluoroacetates also yielded alkali hexafluorosili cates K2SiF6 and Cs2SiF6 as a result of the fluorination of fused quartz. The ability to fluorinate fused quartz was observed for monometallic alkali trifluoroacetates as well. Hexafluorosilicates and heptafluoro silicates K3SiF7 and Cs3SiF7 were obtained upon thermolysis of KH(tfa)2 and CsH(tfa)2 between 200 and 400 &deg;C. The iboga alkaloids scaffold shows great potential as a pharmacophore in drug candidates for the treatment of neuropsychiatric disorders. Thus, the study of the reactivity of this type of motif is particularly useful for the generation of new analogs suitable for medicinal chemistry goals. In this article, we analyzed the oxidation pattern of ibogaine and voacangine using dioxygen, peroxo compounds, and iodine as oxidizing agents. Special focus was placed on the study of the regio- and stereochemistry of the oxidation processes according to the oxidative agent and starting material. We found that the C16-carboxymethyl ester present in voacangine stabilizes the whole molecule toward oxidation in comparison to ibogaine, especially in the indole ring, where 7- hydroxy- or 7-peroxy-indolenines can be obtained as oxidation products. Nevertheless, the ester moiety enhances the reactivity of the isoquinuclidinic nitrogen to afford C3-oxidized products through a regioselective iminium formation. This differential reactivity between ibogaine and voacangine was rationalized using computational DFT calculations. In addition, using qualitative and quantitative NMR experiments combined with theoretical calculations, the absolute stereochemistry at C7 in the 7- hydroxyindolenine of voacangine was revised to be S, which corrects previous reports proposing an R configuration. Among the topics reviewed are the shift from standardized sensory description to consumer-based feedback; going beyond hedonics toward more complex, interdisciplinary food choice models that take into account context, situation, consumer attitudes and values; the growing interest in emotional research; and the study of the extended eating experience (food behavior, well-being). Also covered are methodological advancements in qualitative and quantitative consumer research, adapting methods from psychology and behavioral economics, and the use of nondirected and implicit methods, as well as considering heuristics in food choice. The chapter also reviews the importance of planning specific studies and applications with special populations from a socioeconomic perspective and from a cognitive perspective. The production, under environmentally benign conditions, of efficient and more cost-effective anti-infective agents (available to the whole mankind) is one of most ambitious dreams of the industrial medicinal chemistry. Semi-synthetic b-lactam antibiotics are very effective anti-infective agents. They are very stable and can be used via oral delivery. They exhibit a very wide spectrum of anti-bacterial activity and minimal side effects after being massively used for a very long time. In this way, we can assume that semi-synthetic b-lactam antibiotics are going to be one of the key anti-infective agents in the next years. The condensation of natural or modified antibiotic nuclei with different acyl donor chains is one of the key steps for the industrial synthesis of these anti-infective agents. Up to now, these condensations are mainly carried out through classical chemical methods and this implies a number of economical, ecological and technological drawbacks (high energy requirements, many protection and deprotection steps, utilization of toxic methylene chloride as solvent, etc). Enzyme biocatalysts may be very useful to catalyse these selective condensations under very mild experimental and environmental conditions. In fact, the possibility of using enzymes to carry out such biotransformations, at laboratory scale, has been discussed and demonstrated a long time ago. However, industrial synthesis of b-lactamic antibiotics is still carried out via unfavorable chemical routes. In fact, enzymes have not been produced in the nature to act in industrial reactors; so they are usually very unstable, inhibited by substrates and products and they may not have ideal catalytic properties for industrial uses (high reaction rates, required selectivity, ability to reach quantitative synthetic yields, stable enough to run a number of reaction cycles, etc). These limitations of enzyme biocatalysis become even more significant mainly when the enzyme is going to be used with non-natural substrates, catalysing non-natural processes and working under non-conventional conditions. However, in the last ten years, a great number of papers have reported substantial improvement of these enzymatic synthetic approaches (new enzymes, better enzyme derivatives, improved reaction designs and so on) and it seems that a massive industrial implementation of enzymes in antibiotic synthesis is approaching. In this review, we would like to make a critical discussion of these very interesting advances in the application of enzyme biocatalysts for the industrial synthesis of semi-synthetic antibiotics. Baccharis is a widespread genus belonging to the Asteraceae family that includes almost 400 species exclusively from the Americas. Even when studied in detail, the taxonomic classification among species from this genus is not yet fully defined. Within the framework of our study of the volatile composition of the Baccharis genus, four species (B. trimera, B. milleflora, B. tridentata, and B. uncinella) were collected from the &lsquo;Campos de Cima da Serra&rsquo; highlands of the Brazilian state of Rio Grande do Sul. The aerial parts were dried and extracted by the simultaneous distillation extraction (SDE) procedure. This is the first time that SDE has been applied to obtain and compare the volatile-extract composition in the Baccharis genus. Characterization of the volatile extracts allowed the identification of 180 peaks with many coeluting components; these latter being detailed for the first time for this genus. The multivariate statistical analyses allowed separating the volatile extracts of the four populations of Baccharis into two separate groups. The first one included the B. milleflora, B. trimera, and B. uncinella volatile extracts. The three species showed a high degree of similarity in their volatile composition, which was characterized by the presence of high contents of sesquiterpene compounds, in particular of spathulenol. The second group comprised the extract of B. tridentata, which contained &alpha;-pinene, &beta;-pinene, limonene, and (E)-&beta;-ocimene in high amounts. La respuesta farmacocin&eacute;tica de un f&aacute;rmaco es variable y depende de m&uacute;ltiples procesos. Esta respuesta puede complejizarse, por ejemplo, debido a procesos de recirculaci&oacute;n plasma &ndash; tracto gastrointestinal &ndash; plasma del f&aacute;rmaco. Las mol&eacute;culas secretadas pueden volverse a absorber, y en dicho caso no se hablar&iacute;a de excreci&oacute;n sino de circulaci&oacute;n entre la sangre y el fluido hacia el cual se pretendi&oacute; eliminar. El proceso m&aacute;s ampliamente reconocido en farmacolog&iacute;a es la recirculaci&oacute;n entero-hep&aacute;tica. Sin embargo, en la presente tesis se profundiza en otros posibles circuitos de recirculaci&oacute;n. Un trayecto del f&aacute;rmaco podr&iacute;a ser a trav&eacute;s del jugo g&aacute;strico, en donde partiendo desde la sangre, alcanzar&iacute;a dicho fluido para luego ingresar al medio interno desde la luz intestinal cuando el mismo se vac&iacute;e desde el est&oacute;mago tras la apertura del p&iacute;loro. Otro podr&iacute;a ser mediante el jugo pancre&aacute;tico, el cual es secretado a trav&eacute;s del ducto pancre&aacute;tico que se conecta con el hep&aacute;tico inmediatamente antes de vaciar su contenido hacia el duodeno. Varios mililitros de ambos jugos son vertidos en pocos minutos hacia el tracto gastrointestinal, activados esencialmente por la llegada de alimentos al est&oacute;mago y al intestino, respectivamente. Estos trayectos adquieren relevancia en mol&eacute;culas ionizables dado el pH que posee cada uno de estos fluidos. Existe un gradiente importante de pH entre ambos y el plasma (pH 7,4). De este modo, las drogas b&aacute;sicas preferir&aacute;n la transferencia desde el plasma hacia el jugo g&aacute;strico de pH entre 1,2 y 2,3, para luego verse impedido su retorno a plasma por ionizaci&oacute;n a este pH &aacute;cido. En tanto las drogas &aacute;cidas, desde el plasma hacia el jugo pancre&aacute;tico de pH 8 aproximadamente. El objetivo de la tesis fue estudiar la circulaci&oacute;n plasma-tracto gastrointestinal-plasma de mol&eacute;culas &aacute;cidas y b&aacute;sicas, que a su vez presenten estereoisomer&iacute;a. Los f&aacute;rmacos estereois&oacute;meros, si bien poseen las mismas propiedades f&iacute;sico- qu&iacute;micas, pueden diferir en su farmacocin&eacute;tica y farmacodinamia. Los procesos de recirculaci&oacute;n pueden afectar directamente la exposici&oacute;n a los diferentes is&oacute;meros ya que una posible ruta de metabolizaci&oacute;n para este grupo de f&aacute;rmacos es justamente la inversi&oacute;n quiral y al atravesar el enterocito puede ocurrir la conversi&oacute;n de un is&oacute;mero en el otro. La investigaci&oacute;n realizada demostr&oacute; la recirculaci&oacute;n a la que son sometidos algunos f&aacute;rmacos. Adem&aacute;s, repetidamente se observ&oacute; que luego de la ingesta de comidas, el cociente de concentraciones de is&oacute;meros modificaba la tendencia que segu&iacute;a. Muchas veces los procesos de recirculaci&oacute;n aumentan el tiempo de residencia de las mol&eacute;culas en el organismo, prolongado su acci&oacute;n, sin embargo, en algunos f&aacute;rmacos, como para Metadona, el is&oacute;mero S podr&iacute;a no favorecerse de este proceso ya que cada vez que atraviesa el enterocito aumenta su eliminaci&oacute;n. Para los AINES, la ingesta de comidas podr&iacute;a en algunos casos promover la relaci&oacute;n entre is&oacute;meros que resulta m&aacute;s beneficiosa para la terapia, al favorecer la conversi&oacute;n R-en-S, pero en otros casos, como para Ibuprofeno, podr&iacute;a no impactar sobre la exposici&oacute;n al is&oacute;mero activo ya que la mayor tasa de conversi&oacute;n no logra compensar el gran aumento de clearance intestinal que se ve para ambos is&oacute;meros cuando se administran comidas frecuentes. Debe considerarse cada caso en particular para poder hacer una recomendaci&oacute;n respecto, por ejemplo, al modo de administraci&oacute;n. Por esto es interesante destacar, que este tipo de 5 investigaciones, si bien aportan valiosa informaci&oacute;n respecto a la farmacocin&eacute;tica de determinados f&aacute;rmacos, principalmente constituyen herramientas poderosas para el entendimiento de la fisiolog&iacute;a, lo que permite la aplicaci&oacute;n del conocimiento a diferentes casos. Otro factor que demostr&oacute; condicionar de manera importante la farmacocin&eacute;tica de los f&aacute;rmacos estudiados, fueron las diferencias fisiol&oacute;gicas entre hombre y mujeres. Se observ&oacute; el m&aacute;s lento vaciado g&aacute;strico de las mujeres respecto los hombres, su mayor pH intestinal y el impacto del tama&ntilde;o corporal sobre procesos de metabolizaci&oacute;n saturables. Resumiendo, se demostr&oacute; la dis&iacute;mil exposici&oacute;n a los is&oacute;meros que puede obtenerse tras la administraci&oacute;n de una mezcla rac&eacute;mica y c&oacute;mo diferentes variables impactan sobre la relaci&oacute;n entre los is&oacute;meros. Por m&aacute;s que la terapia estereoselectiva resulte beneficiosa, por ejemplo, porque solo uno de los is&oacute;meros sea farmacol&oacute;gicamente activo, la mayor&iacute;a de los f&aacute;rmacos estereois&oacute;meros que se encuentran en el mercado, se administran como racematos debido al coste econ&oacute;mico y a la complejidad de la separaci&oacute;n de los is&oacute;meros. Esto introduce la necesidad de considerar el seguimiento de la terapia con estos medicamentos a trav&eacute;s de la determinaci&oacute;n de los is&oacute;meros por separado, ya que, como se mencion&oacute;, el perfil farmacocin&eacute;tico y farmacodin&aacute;mico de los is&oacute;meros puede ser diferente y, por lo tanto, carece de l&oacute;gica el an&aacute;lisis de la mezcla rac&eacute;mica. En el presente trabajo, se pone de manifiesto la ventaja de la determinaci&oacute;n quiral. De otra manera, no hubiese sido posible la detecci&oacute;n de los procesos estudiados y el correcto an&aacute;lisis de los resultados. Por otro lado, la informaci&oacute;n recabada podr&iacute;a ser de utilidad para el desarrollo de formas farmac&eacute;uticas innovadoras que permitan un m&aacute;s eficiente uso de la dosis rac&eacute;mica contenida. The 5&cent;-GCGTGGGCGT-3&cent; (T4) oligomer found in the zinc &reg;nger 268&plusmn;DNA complex was mutated into the sequence 5&cent;-GCGGGGGCGT-3&cent; (G4). A 3D model was constructed from the T4 sequence using an X-ray structure as a template. Molecular dynamics simulations were used to test the thermal stability of the model. A 500-ps trajectory was obtained for the fully charged complex in water using GROMOS87. The complex and the G4 sequence are found to have dynamically stationary behavior. Comparisons made with a previous T4 sequence molecular dynamics simu- lation show both systems have similar thermal stability. The structure of DNA appears to be maintained by its global interactions with the protein although the mutated site does not contribute with its full potential for binding. The protein structure shows some small di€erences compared to the T4 simulation. The simulation provided evidence for the role of a chloride ion interacting with the protein and helping in the recognition process.
