Scavenger receptors participate in a wide range of biological functions after binding to multiple non-self or altered self-ligands. Among them, CD5 and CD6 are lymphocyte scavenger receptors known to interact with different microbial-associated molecular patterns, and the administration of the recombinant soluble ectodomains of human CD5 (rshCD5) and/or CD6 (rshCD6) has shown therapeutic/prophylactic potential in experimental models of fungal, bacterial and echinococcal infections. The latter is a zoonosis caused by the larval stage of the cestode parasite Echinococcus granulosus sensu lato, which in humans can induce secondary cystic echinococcosis (CE) after the spillage of protoscoleces contained within fertile cysts, either spontaneously or during surgical removal of primary hydatid cysts. Herein, we have analysed the mechanisms behind the significant protection observed in the mouse model of secondary CE following prophylactic administration of rshCD5 or rshCD6. Our results show that both molecules exhibit intrinsic antiparasitic activities in vitro, as well as immunomodulatory functions during early secondary CE, mainly through Th1/Th17 cytokine bias and promotion of peritoneal polyreactive antibodies. These data support the relevance of the parasite components bound by rshCD5 and rshCD6, as well as the potential of their prophylactic administration as a useful strategy to reduce secondary CE in patients.

Phage borne peptides isolated from phage libraries have proven to be valuable reagents for the development of small-molecule immunoassays. However, the large size, low diffusion rate, and biological nature of the phage particles create some limitations to their use and require secondary reagents for its detection. In this work, we explore the use of the Nano luciferase (NanoLuc) as a fusion partner to generate recombinant tracers for immunoassay development. The imidaclothiz peptidomimetic C2-15 that specifically binds to the anti-imidaclothiz monoclonal antibody (mAb) 1E7 was fused to NanoLuc, both at the N terminus (C2-15-NanoLuc) and C terminus (NanoLuc-C2-15). NanoLuc-C2-15 showed better performance than C2-15-NanoLuc and was adopted to develop a bioluminescent enzyme immunoassay (BLEIA) and a bioluminescence lateral flow immunoassay (BLLFIA) for imidaclothiz. The luminescence signal of NanoLuc-C2-15 rapidly reaches high intensity with slow attenuation, which enabled one to capture the BLLFIA readout by using a smartphone without an external light source. The IC50 of the BLEIA and BLLFIA were 3.3 ± 0.2 and 6.4 ± 0.4 ng mL-1, respectively. Both immunoassays exhibited good accuracy for the detection of imidaclothiz in environmental and agricultural samples.

Short peptide loops selected from phage libraries can specifically recognize the formation of hapten-antibody immunocomplexes and can thus be used to develop phage anti-immunocomplex assays (PHAIA) for noncompetitive detection of small molecules. In this study, we generated recombinant chimeras by fusing anti-immunocomplex peptides selected from phage libraries to the N- or C-termini of core streptavidin and used them to setup phage-free noncompetitive assays for the herbicide clomazone (MW 240 Da). The best conditions for refolding were optimized by a high throughput screening allowing to obtain tens of mg of purified protein per liter of culture. The noncompetitive assay developed with these chimeras performed with a 50% saturating concentration (SC50) of 2.2 ± 0.3 ng/mL and limit of detection (LOD) of 0.48 ng/mL. Values that are 13- and 8-fold better that those obtained for the SC50 and LOD of the competitive assay setup with the same antibody. Apart from the first demonstration that recombinant peptide-streptavidin chimeras can be used for sensitive immunodetection of small molecules with a positive readout, this new assay component is a highly standardized reagent with a defined stoichiometry, which can be used in combination with the broad option of existing biotinylated reagents offering a great versatility for the development of conventional immunoassay and biosensors. The utility of the test was demonstrated analyzing the clomazone runoff during the rice growing season in northern Uruguay.

Peptidomimetic and anti-immunocomplex peptides, which can be readily isolated from a phage-display library, have shown great potential for small-molecule immunoassay development because they typically improve the sensitivity and avoid the use of chemical haptens as coatings or tracer antigens. However, phage-borne peptides are unconventional immunoassay reagents, which greatly limits their use in commercial applications, and require secondary reagents for detection. In order to overcome these limitations, we used C2-15, a peptidomimetic of imidaclothiz, as a model peptide fused to emerald-green fluorescent protein (EmGFP) at the N-terminus (C2-15-EmGFP) and C-terminus (EmGFP-C2-15) to generate novel fluorescent-peptide tracers. Both recombinant fluorophores reacted with similar affinity to the anti-imidaclothiz monoclonal antibody 1E7, but because of its higher expression, C2-15-EmGFP was chosen to develop a competitive magnetic-separation fluorescence immunoassay (MSFIA). After a competitive step with the analyte, the C2-15-EmGFP–antibody complex bound to the magnetic beads was separated with a magnet, and because of the fast dissociation of the peptide–antibody interaction, the fluorescence signal was detected following the spontaneous dissociation of the complex in fresh buffer. The concentration of imidaclothiz causing the 50% inhibitory concentration (IC50) was 11.00 ng mL–1, and the MSFIA performed with excellent recovery and had a good correlation with high-performance liquid chromatography in different matrices.

Abstract
A summary of the resource.
A class 2 integron was found in an Escherichia coli isolate mediating a urinary tract infection. Unlike other class 2 integrons from pathogens, the encoded IntI2 protein was functional. The integron possessed a dfrA14 cassette, and a second novel cassette in which a lipoprotein signal peptidase gene is predicted.

A summary of the resource.
Among small cellular molecules, certain inositol phosphates (InsPs) excel at complexing Fe3+ in a safe manner in vitro. In particular Ins(1,2,3)P3 is probably an important chelator of iron cations not strongly bound to proteins, which avoids iron redox cycling and the associated production of free radicals. In this work we report the first thermodynamic data of the Fe3+/Fe2+ redox potential in the presence of Ins(1,2,3)P3 which demonstrate the capability of this ligand to inhibit iron(III) reduction in the intracellular media. The Fe3+–Ins(1,2,3)P3 speciation was also studied by 31P NMR measurements, focusing on the structure of the complexes. The work is completed by theoretical calculations which lead us to predict the geometries of the species at different pH values and their relative stabilities. The influence of the ring conformation in the Fe3+ complexed forms is also discussed.

<p>Although redox state is a well-known key process parameter in microbial activity, its impact on wine volatile aroma compounds produced during fermentation has not been studied in detail. In this study we report the effect of reductive and microaerobic conditions on wine aroma compound production using different initial amounts of yeast assimilable nitrogen (YAN: 180 and 400 mg N/l) in a simil grape must defined medium and two S. cerevisiae strains commonly used in wine-making. In batch fermentation culture conditions, reductive conditions were obtained using flasks plugged with Muller valves filled with sulphuric acid; while microaerobic conditions were attained with defined cotton plugs. It was found that significant differences in redox potential were obtained using the different plugs, and with variation of over 100 mV during the main fermentation period. Significant differences in the final concentration of higher alcohols, esters and fatty acids were attributed to differences in the redox state in the medium in both strains. A consistent increase in esters and medium chain fatty acids, as well as a decrease of higher alcohols and isoacids, was seen under reductive fermentation conditions. Interestingly, 1-propanol, d-butyrolactone and ethyl lactate concentrations, showed no significant variation under the different redox conditions. A better understanding of the influence of redox state of the fermentation medium on the composition of volatile compounds in wine could enable improvement of vinification management. From a microbiological standpoint results presented here will contribute to the standardization of data models for the application of metabolic footprinting methods for wine yeast strain phenotyping and characterization</p>

For the first time a critical analysis of the influence that four different graphene oxide reduction methods have on the electrochemical properties of the resulting reduced graphene oxides (RGOs) is reported. Starting from the same graphene oxide, chemical (CRGO), hydrothermal (hTRGO), electrochemical (ERGO), and thermal (TRGO) reduced graphene oxide were produced. The materials were fully characterized and the topography and electroactivity of the resulting glassy carbon modified electrodes were also evaluated. An oligonucleotide molecule was used as a model of DNA electrochemical biosensing. The results allow for the conclusion that TRGO produced the RGOs with the best electrochemical performance for oligonucleotide electroanalysis. A clear shift in the guanine oxidation peak potential to lower values (~0.100 V) and an almost two-fold increase in the current intensity were observed compared with the other RGOs. Abstract

By means of first-principles calculations, we investigate the reduction chemistry of monolayer boron nitride (2D BN) and graphene. The adsorption of alkali atoms dramatically increases the reactivity of 2D BN, Li being the most powerful agent and closely followed by K and Na. Interestingly, the reactivity enhancement in 2D BN is 3.6 times larger in comparison with graphene. On average, Li adsorption enhances the adsorption energies by 66.0 and 18.2 kcal/mol for 2D BN and graphene, respectively. The higher increment in adsorption energies observed for 2D BN can be attributed to the significant difference in alkali charge donation to pristine and functionalized 2D BN. In contrast, for graphene, this difference is much smaller. The increase of reactivity strongly depends on the functional group added as well as on the alkali used. The reduction chemistry facilitates the addition of functional groups to 2D BN which cannot be added without the presence of alkalis. We conclude that reduced 2D BN is more reactive than reduced graphene when alkalis act as reducing agents. We expect that this work will motivate new investigations to attain the covalent functionalization of 2D BN.

Food waste is an issue of global concern. Particularly troublesome is the amount of waste at the consumer level, and a need exists for interventions that draw on a deeper understanding of the underlying factors leading to food waste. The research aims were to (1) study the potential of an information-based strategy to reduce household food waste, (2) explore the impact of information on attitudes towards food waste, and (3) explore the reasons underlying perceived changes in household food waste. A controlled randomized trial was conducted, and participants were assigned to one of two groups (Informed, Control) with data collection at two time points (Baseline, Time 2). At both time points, all participants completed a standardized quantitative survey on food disposal in the past 7 days. At baseline, after the food disposal survey was completed, participants in the informed group read a text about food waste and completed a highlighting task. The key result was a significant reduction in household food waste approximately fifteen days after the Baseline, which was found for both experimental groups, albeit slightly larger in the Informed group. This was found despite the Control group having the same attitudes, on average, to engaging in food waste reduction behaviors. To successfully reduce food waste, participants bought less food, used leftover and suboptimal foods, reduced portion size, checked stored foods to use them before expiration, and planned meals. The main reason for not reducing food waste was related to the belief that household food waste was minimal or inexistent. Abstract
A summary of the resource.
The reference interval for glycated haemoglobin in 174 apparently healthy individuals from Montevideo City(mean 94 |b SD 7umol fructose/10mg haemoglobin) was determined using the thiobarbituric acid spectrophotometric method. In diabetic patients (n=222) the mean was significantly higher than in the control group(mean 117 |b SD 19umol fructose/10 mg haemoglobin). The spectrophotometric method for the determination of glycated haemoglobin was compared to a microcolumn kit method (Boehringer |b Mannheim |b FRG). The correlation for glycated haemoglobins was acceptable (r= 0.85; n=44; P<0.001). The colorimetric reference interval for glycated haemoglobin was converted to an equivalent value in the chromatographic method |b expressed as per cent HbA (mean 8.7 |b SD 1.2) |b using the least squares regression line (y=0.603+0.0857x)

<p>As nutritional recommendations change, new marketing strategies are expected to appear on food labels to convey the idea that products are healthful and aligned with the recommendations. In this context, the objective of the study was to evaluate the effect of references to ‘home-made’, images of natural foods and nutritional warnings on consumers’ healthfulness perception and purchase intention of labels of ultra-processed products with excessive content of nutrients associated with non-communicable diseases (NCDs). The study was conducted with 790 Uruguayan Facebook users (65% female, 18–89 years old), diverse in terms of educational and<br />socio-economic levels. A choice experiment involving labels of four categories of ultra-processed products (burgers, cookies, instant soup and instant vanilla milk custard) was implemented. For each product category, eight sets of two labels differing in the presence of references to home-made, images of natural foods and nutritional warnings were designed. Half of the participants were asked to indicate the healthier product,<br />whereas the other half were asked to indicate the product they would purchase. Data were analyzed using mixed logit models. The inclusion of references to home-made and images of natural foods on the labels had a positive effect on purchase intention and tended to create the belief that products were healthful. Nutritional warnings decreased healthfulness perception and discouraged participants’ choice. In three of the product categories images of natural foods created a health halo that reduced the effect of nutritional warnings on product choice. Results from the present work suggest the need to develop stricter labelling regulations for ultra-processed<br />products with high content of nutrients associated with NCDs.</p>
