La albúmina sérica humana (HSA), es una proteína monomérica compuesta por tres dominios, conocidos como dominios I, II y III. Presenta en su estructura 17 puentes disulfuro y una única cisteína libre (Cys34) que se localiza en el dominio I (DomI). La HSA es la proteína mayoritaria del plasma y su tiol se encuentra principalmente reducido (HSA-SH), constituyendo el 80% de los tioles reducidos en el compartimiento intravascular. Una fracción menor se encuentra formando disulfuros con tioles de bajo peso molecular (HSA-SSR) u oxidado a ácidos sulfínico (HSA-SO2H) y sulfónico (HSA-SO3H). En esta tesis se profundizó en el análisis de las isoformas de la HSA en plasma mediante cromatoenfoque y se trabajó en la expresión, purificación y caracterización del dominio I (DomI) de la HSA producido en Pichia pastoris. En cuanto al cromatoenfoque, se realizó una puesta a punto del análisis de muestras de plasma de donantes sanos mediante controles de precipitación de HSA en el amortiguador de trabajo y estableciendo la cantidad de HSA a inyectar para obtener una buena resolución. El cromatoenfoque es reproducible y puede utilizarse tal como se puso a punto, para estudiar el aumento de las fracciones oxidadas de la HSA de personas con sepsis. Por otra parte, se expresó el DomI en P. pastoris a partir de dos construcciones. Una conteniendo dos mutaciones sinónimas en su secuencia (DomIMS) y otra generada a partir de la secuencia de referencia de la HSA (DomISR). Ambos DomI se expresaron y secretaron correctamente hacia el medio de cultivo, y mostraron la presencia de un pigmento marrón. Respecto al DomIMS, la eliminación de la mayor parte del pigmento se logró con una cromatografía de intercambio catiónico a pH 5.0 y elución con NaCl 1 M. La reducción selectiva de dicha cisteína se logró por incubación con 2-mercaptoetanol, como se vio por reacción con 5,5´-ditiobis(2-nitrobenzoato) (DTNB) y espectrometría de masa. Además se determinó la constante de reacción de la Cys34 de DomI con DTNB. El valor de 229 M-1 s-1 resultó 16 veces mayor que el de la HSA deslipidada, en iguales condiciones experimentales. En relación al DomISR, se transformaron células de la cepa GS115 His- Mut+, se seleccionaron primero transformantes, y luego transformantes multicopia. Se prepararon crioinóculos que expresaron el DomISR correctamente. En un ensayo preliminar se vio que la Cys34 del DomISR se reduce en iguales condiciones que la del DomIMS y reacciona con DTNB con una constante similar, sugiriendo que no existen diferencias estructurales entre ambos

For understanding the electrical and optical properties of electrodeposited Cl-doped Cu2O thin films, we have studied layers with increasing thickness and Cl concentrations of 0.8 and 1.2 at. %. The deposits were characterized by measuring the charge transport, the optical reflectance, and the photoluminescence. No significant decrease of electrical resistivity was observed in doped samples compared to undoped ones. A decrease of about five orders of magnitude was measured and ascribed to the presence of pinholes, as confirmed by scanning electron microscopy analyses. From optical measurements, we concluded that the Cl atoms are incorporated into substitutional sites of Cu2O lattices in agreement with photoluminescence results showing a strong reduction in the peak intensity of VO+2 defects in comparison to undoped layers. Computational calculation using density functional theory has pointed out high formation energy for single Cl related defects, but low formation energy for Cl-defect complexes, such as ClO + VCu, that strongly compensate the carriers generated by the Cl doping.

The biotransformation of (4R)-(-)-carvone by Mentha pulegium (pennyroyal) leaves and its endophytic bacteria was performed in order to search for novel biocatalysts with enoate reductase activity. The obtained results clearly indicated that endophytes play an important role in the biotransformation of (4R)-(-)-carvone with pennyroyal plant tissues. The best activity was associated to the endophytic bacteria Pseudomonas proteolytica FM18Mci1 and Bacillus sp. FM18civ1. Enoate reductase activity for the reduction of (4R)-(-)-carvone and (4S)-(+)-carvone as model substrates was evaluated for each strain. Finally, both isolated strains were evaluated for the kinetic resolution of racemic carvone. The two bacteria gave (1R, 4R) or (1R, 4S)-dihydrocarvone as major products. P. proteolytica FM18Mci1 had preference for the 4S-(-)-carvone, reaching a conversion 95% in 24 h. In contrast, Bacillus sp. FM18civ1 had preference for (4R)-(-)-carvone. The results obtained in the kinetic resolution of carvone indicated that the Bacillus strain could be useful for resolving a racemic mixture of carvone.

Humoral immunity wanes during healthy ageing, increasing susceptibility to infections in the elderly. In this sense, information about parasite infections and human immunosenescence is scarce. Cystic echinococcosis (CE) is an infectious disease caused by the larval stage of the cestode parasite Echinococcus granulosus, whose prevalence in humans shows an increase with host age. Susceptibility to human CE has been associated with humoral immunity to some extent; and therefore, we have here analyzed the influence of host age on the serological profile of young, middle-aged, and aged patients. Our results highlighted the detrimental influence of ageing on the intensity and quality of the anti-parasite antibody response. Remarkable differences in serological profiles between young and aged individuals were observed. In this sense, through Principal Components Analysis we identified aged patients as those exhibiting overall less intense antibody responses, mainly in isotypes/subclasses supposed to exert efficient anti-parasite activities (e.g. IgE and IgG1). Thus, these humoral defects could at least partially explain the reported increase in CE prevalence among older individuals, since a weaker immune response in the elderly might facilitate the establishment and maintenance of the parasite infection. Finally, a possible association between age-dependent susceptibility to CE and host immunosenescence is discussed.

Electronic structure calculations have been performed for the characterization of the different conformers of the herbicide metolachlor (MC), (2-chloro-N-(2-methyl-6-ethylphenyl)-N-(2-methoxy-1-methylethyl) acetamide), the radical structures obtained through the photo chemical breaking of the C–Cl bond and the most important, experimentally identified mono-hydroxylated photo-fragmentation products. Only the S-metolachlor enantiomer that shows herbicide activity was studied. The goal of the study has been twofold. The first target has been the conformational analysis of the neutral molecule and the corresponding radical. Ten different conformers were identified for the S-metolachlor enantiomer, all of them exhibiting a near perpendicular disposition of the amidyl and the phenyl planes. They were classified into two families of minima depending on the orientation of the carbonyl group (either cis or trans with respect to the phenyl ring). All the minima in the S-cis family were found to lie higher than those in the S-trans family. The free energy of isomerization between the lowest minima of each family was found to be ΔG0298 = 6.2 kcal/mol, while the isomerization barrier around the amide C–N bond causing atropisomerism was calculated to be 34.9 kcal/mol (in good agreement with the experimental energy of activation, 36.9 ± 3 kcal/mol) [45]. The second part of the work is devoted to the computational characterization of the most important isomeric forms of the metolachlor radical and the major photodegradation mono-hydroxylated metabolites formed through the coupling of the metolachlor radical with OH. Reaction enthalpies and free energies for the various hydroxylation pathways have been calculated. Based on their order, the relative significance of the various hydroxylation channels is justified and the preference in the phenyl ring hydroxylation is established. The lowest exothermicity value determined, − 5.8 kcal/mol at the M06/6-31 + G(d,p) level, corresponds to phenyl group hydroxylation at the p-position with regard to C–N bond, in excellent agreement with the experimental findings.

Antigen B (EgAgB) is a phosphatidylcholine (PC)-rich lipoprotein of Echinococcus granulosus s.l. larva, potentially capable of modulating the activation of various myeloid cells, including macrophages. As C-reactive protein (CRP) can act as an innate receptor with ability to bind the phosphocholine moiety of PC in lipoproteins, we investigated whether EgAgB and CRP could interact during cystic echinococcosis infection (CE), and how CRP binding could affect the modulation activities exerted by EgAgB on macrophages. To that end, we firstly investigated the occurrence of CRP induction during human CE. We found that 61% of CE patients, but none of healthy donors, exhibited serum CRP levels higher than 10 mg/mL, suggesting that CRP can be induced during the chronic phase of CE. Furthermore, human CRP was capable of binding specifically to EgAgB with high affinity (0.6 ± 0.1 nM); this binding was Ca2+ -dependent and involved the phosphocholine moiety of PC, but not EgAgB8/1, EgAgB8/2 or EgAgB8/3 apolipoproteins. Finally, CRP presence altered the modulation exerted by EgAgB on the cytokine response of LPS-activated macrophages. Overall, our results suggest that CRP presence during CE may contribute to a complex scenario of interactions between EgAgB and myeloid cells, influencing the cytokine response induced during macrophage activation.

Traditional olive oil processing includes batch olive paste kneading for 30–60 min. Microwave (MW) treatment of the olive paste have recently shown to continuously provide faster conditioning, while megasound (MS) treatment have been shown to enhance oil extractability. In the present work the virgin olive oil standard parameters, sensory descriptors, and phenolic and headspace composition upon treatment of oils obtained using novel process enhancement interventions, and their combination in an industrial plant is examined. Standard parameters are within the international specifications. The Control and MW process provide similar phenolic composition but a post MS intervention significantly increased total phenolic content, and decrease total C5 and C6 aldehydes in the MW treated paste. These results encourage further developments for a combined continuous MW and megasonic conditioning technology to fasten the olive oil extraction, to enhance yields, and total phenolic content.

Groundwater reservoirs constitute important freshwater resources. However, these ecosystems are highly vulnerable to contamination and have to rely on the resident microbiota to attenuate the impact of this contamination. Nitrate is one of the main contaminants found in groundwater, and denitrification is the main process that removes the compound. In this study, the response to nutrient load on indigenous microbial communities in groundwater from a low impacted aquifer in Uruguay was evaluated. Denitrification rates were measured in groundwater samples from three different sites with nitrate, acetate and pyrite amendments. Results showed that denitrification is feasible under in situ nitrate and electron donor concentrations, although the lack of readily available organic energy source would limit the attenuation of higher nitrate concentrations. DNA-stable isotope probing, combined with amplicon sequencing of 16S rRNA, nirS and nirK genes, was used to identify the active denitrifiers. Members of the phylum Betaproteobacteria were the dominant denitrifiers in two of three sites, with different families being observed; members of the genus Vogesella (Neisseriaceae) were key denitrifiers at one site, while the genera Dechloromonas (Rhodocyclaceae) and Comamonas (Comamonadaceae) were the main denitrifiers detected at the other sites.

The formation of selenium species in some biological processes involves the generation of ionic and radical intermediates such as RSe●, RSe−, RSeO●, and RSeO−, among others. We performed a theoretical study of the possible mechanisms for the reaction of the two simplest Se radicals—the hydroselenyl (HSe●) and selenenic (HSeO●) radicals, in which the possible products, intermediates, and transition-state structures were investigated. Density functional theory (DFT) was applied at the B3LYP/6–311++G(3df,3pd) level and the Ahlrichs Coulomb fitting basis sets were employed with an effective core potential (ECP) for both Se atoms. The same procedure was used to calculate the electronic density. All calculations were also performed using the M06-2X functional, which describes weaker bonds better than B3LYP does. In the reaction of interest, the so-called CR complex (HSe····SeOH) is formed initially. After passing through the transition state TS1, cis-HSeSeOH is obtained as a product. If a low barrier is then overcome (passing through the transition state TS32), the trans-HSeSeOH species is obtained. The CR complex can also rearrange into the intermediate INT after overcoming the barrier presented by the transition state TS2. Additionally, the decomposition of INT to H2O and 1Se2 is possible through another transition state. This reaction is not included in this study. We also observed a second possible route for the conversion of INT to one of the HSeSeOH species; this route occurs through two pathways (with transition states TS31 and TS32). A comparison of some of the results with those obtained for sulfur analogs along the same pathways is also presented in this work.

The variable VHH domains of camelid single chain antibodies have been useful in numerous biotechnology applications due to their simplicity, biophysical properties, and abilities to bind to their cognate antigens with high affinities and specificity. Their interactions with proteins have been well‐studied, but considerably less work has been done to characterize their ability to bind haptens. A high‐resolution structural study of three nanobodies (T4, T9, and T10) which have been shown to bind triclocarban (TCC, 3‐(4‐chlorophenyl)‐1‐(3,4‐dichlorophenyl)urea) with near‐nanomolar affinity shows that binding occurs in a tunnel largely formed by CDR1 rather than a surface or lateral binding mode seen in other nanobody‐hapten interactions. Additional significant interactions are formed with a non‐hypervariable loop, sometimes dubbed “CDR4”. A comparison of apo and holo forms of T9 and T10 shows that the binding site undergoes little conformational change upon binding of TCC. Structures of three nanobody‐TCC complexes demonstrated there was not a standard binding mode. T4 and T9 have a high degree of sequence identity and bind the hapten in a nearly identical manner, while the more divergent T10 binds TCC in a slightly displaced orientation with the urea moiety rotated approximately 180° along the long axis of the molecule. In addition to methotrexate, this is the second report of haptens binding in a tunnel formed by CDR1, suggesting that compounds with similar hydrophobicity and shape could be recognized by nanobodies in analogous fashion. Structure‐guided mutations failed to improve binding affinity for T4 and T9 underscoring the high degree of natural optimization.

A summary of the resource.
New technologies provide the fruit sector opportunities for the development of new products that meet consumer demands for a healthy diet. These technologies have several advantages over conventional thermal processes as they yield more nutritious food, preserving their sensory characteristics. However, these advantages do not assure consumer acceptance, which is the main determinant of the success of new technologies. This chapter aimed at providing an overview of the main determinants of consumer perception of fruit products manufactured using new technologies and to discuss strategies to increase consumer awareness and acceptance of these products. High hydrostatic pressure, pulsed electric fields, and irradiation were discussed under the perspective of the consumer. The results suggest that providing information about new technologies seems to be a key strategy for increasing consumers’ acceptance of fruit products manufactured by means of these technologies. Information and communication campaigns should be performed at the early phases of the introduction of new technologies into the marketplace.

La academia ha reconocido la necesidad de tener en cuenta tanto las métricas ambientales como las económicas a la hora de desarrollar procesos que, en última instancia, encontrarán aplicación en la fabricación de productos químicos. Las aminas quirales son porciones de moléculas que constituyen un grupo de "estructuras preferidas" (“privileged scaffolds”) que son frecuentes en los productos naturales, fármacos, agroquímicos y otros productos químicos, y que se incluyen habitualmente en el diseño de bibliotecas para el descubrimiento de nuevas drogas. También se conoce que el 80 % de los 20 fármacos más recetados en medicinas son aminas quirales, lo que hace que estos compuestos sean valiosos componentes básicos en la industria farmacéutica. Esta tesis tiene como objetivo principal investigar nuevos biocatalizadores en síntesis para la producción de aminas quirales, utilizando tanto enzimas reportadas como nuevas enzimas aún no identificadas. Mediante análisis de motivos aminoacídicos conservados detectados en secuencias disponibles y análisis de motivos de iminoreductasas reportados en paralelo a nuestro trabajo, se realizó una búsqueda de nuevas IRED. Como resultado, se han encontrado varias secuencias que cumplen con las características para ser potenciales enzimas reductoras de iminas. Además, se aplicó un enfoque de “genome mining and motif análisis”, para la selección de potenciales secuencias R-ω-TA. A partir de 423 candidatos, la secuencia R-ω-TA de Capronia semiimmersa fue seleccionada, clonada y expresada en Escherichia coli . La enzima expresada demostró aceptar D-alanina así como también isopropilamina como grupo aminodonor y tener actividad sobre una variedad importante de cetonas. Ademas, ha sido demostrada la aplicación de una cascada quimioenzimática novedosa para la síntesis regio y estereoselectiva de un panel de pirrolidinas 2,5-disustituidas a partir de las correspondientes 1,4-dicetonas. Al mismo tiempo, se logró establecer un sistema de desracemización de aminas en una misma reacción, con cinco biocatalizadores actuando paralelamente de manera simple y práctica. De esta forma se logran obtener las aminas enantioméricamente puras a partir de una mezcla racémica o a partir de la amina con estereoquímica opuesta. Por otra parte, se logró ampliar los sistemas enzimáticos aplicados a la síntesis de aminas quirales, para ello se utilizó un nuevo tipo de transaminasa bifuncional denominada α,ω- DTAs, perteneciente a la metabolización de la putrescina. Este nuevo sistema, requiere solamente 3-5 equivalentes de aminodonor en el medio de reacción en contraste a los 50-500 equivalentes de grupo aminodonor utilizados normalmente.
