However, the expensive and laborious purifcation processes applied to purify Quillaja saponins used to assemble ISCOM-matrices show an important drawback in the large-scale use of this vaccine adjuvant. Thus, in this study, we describe a protocol to appropriately formulate ISCOM-matrices using the raw aqueous extract (AE) of Quillaja lancifolia leaves. In the presence of lipids, AE was able to self-assemble in nanostructures that resembles immunostimulating complexes (ISCOM). These nega tively charged nanoparticles of approximately 40 nm were characterized by transmission electron microscopy and dynamic light scattering. In addition, well-known saponins with remarkable immunoadjuvant activity, as QS-21, were detected into nanoparticles. Thus, the easier, robust, cheaper, and environmentally friendly method developed here may be an alternative to the classical methods for ISCOM-matrices production that use high-purifed saponins. Increasing the effectiveness of vaccines against respiratory viruses is particularly relevant for the elderly, since they are prone to develop serious infections due to comorbidities and the senescence of the immune system. The addition of saponin-based adjuvants is an interesting strategy to increase the effectiveness of vaccines. We have previously shown that ISCOM matrices from Q. brasiliensis (IMXQB) are a safe and potent adjuvant. In this study, we evaluated the use of IMXQB as an adjuvant for the seasonal trivalent influenza vaccine (TIV) in an aged mice model. Herein, we show that subcutaneous injection of the adjuvanted vaccine promoted higher titers of IgM, IgG (and isotypes), and serum hemagglutination inhibition titers (HAI). Notably, aged mice immunized by intranasal route also produced higher IgG (and isotypes) and IgA titers up to 120 days after priming, as well as demonstrating an improvement in the HAI antibodies against the TIV. Further, experimental infected aged mice treated once with sera from adult naïve mice previously immunized with TIV-IMXQB subcutaneously successfully controlled the infection. Overall, TIV-IMXQB improved the immunogenicity compared to TIV by enhancing systemic and mucosal immunity in old mice conferring a faster recovery after the H1N1pdm09-like virus challenge. Thus, IMXQB nanoparticles may be a promising platform for next-generation viral vaccines. Las experiencias didácticas vividas en formación docente inicial tienen un fuerte impacto en la conformación del perfil profesional del profesor. En estas experiencias se deben ver claramente trabajados los grandes fines en la enseñanza de las ciencias: formar ciudadanos críticos, formar para la participación ciudadana, alfabetizar científicamente. La teniosis por solium, cisticercosis y triquinosis, son parasitosis en los humanos que se adquieren por consumo de carne de cerdo. En la formación inicial de docentes de Biología para la enseñanza media, las especies causantes de las parasitosis se estudian en la asignatura Zoología. Quebrando con el clásico abordaje de temas programáticos siguiendo la lógica de la disciplina, se realizaron en la asignatura trabajos con un enfoque de tipo CTS. Se organizó la conformación de equipos en el grupo clase, uno de los cuales abordó los helmintos parásitos transmitidos al humano por el cerdo. El trabajo incluyó la realización de una investigación bibliográfica, recolección de información en industrias locales que crían cerdo, la producción de una monografía y la presentación del tema a los demás compañeros de clase. Se vio el vínculo existente entre la práctica religiosa judía y la ausencia en dicha comunidad de parasitosis por helmintos que se transmiten con el consumo de carne suina. Se analizaron cuestiones relacionadas con las costumbres alimenticias de la comunidad religiosa (su filosofía, sus libros sagrados), utilizándose los conocimientos de Zoología y Parasitología para dar fundamento científico al vínculo señalado. Con esto se reconocieron diferencias entre preceptos religiosos y construcciones científicas, así como los fundamentos sanitarios de la adecuada tecnología en la cría del cerdo. La experiencia educativa realizada confirma que el tratamiento de temas disciplinares bajo un enfoque CTS es una forma de integrar el conocimiento, promover la interacción e intercambio disciplinar y humanizar la ciencia. El enfoque apunta a la confluencia de propuestas que promueven la participación de los ciudadanos en los problemas sociales y ambientales. The larva of the cestode Echinococcus granulosus (hydatid cyst) is protected by the acellular laminated layer (LL). The mechanisms that make this thick coat a poor activator of host complement are incompletely understood. The structure binds, through unknown motifs, the host regulator of the alternative complement pathway (ACP), factor H. A second potential mechanism of ACP regulation, the inhibition of factor B activation, was detected in assays employing purified components (Immunopharmacology 42 : 91). The inhibitor was subsequently identified as myo-inositol hexakisphosphate (InsP(6)), which in the form of nano-deposits is a major component of the LL (Biochem J 362 : 297; J Cell Biochem 93 : 1272; FEBS J 273 : 3192). In this report we show that colloidal InsP(6 )solids inhibit factor B activation, through adsorption and associated impairment of C3b binding. However, this interaction is not relevant in the presence of serum proteins. In serum, InsP(6) deposits instead bind C1q, and initiate complement activation. This activation is curtailed through efficient C3b inactivation, previously shown to be entirely factor H-dependent, and now observed to be independent of the InsP(6) deposits. Therefore the complement resistance of the LL must be based on functional factor H binding sites present on the mucin-based meshwork that is its other major constituent. Cystic hydatid disease is caused by the multicellular parasite Echinococcus granulosus. The hydatid cyst, being a long-lived, large, antigenic structure lodged in the host's internal organs, could potentially elicit major inflammatory responses. However, in practice, the cyst causes only minimal local inflammation. The complement system is a major pathway to immune-mediated inflammation. Recent results have shown that the host-exposed structure of the cyst, the hydatid cyst wall (HCW), fails to trigger the complement system strongly. We have carried out a wide survey for the mechanisms making the cyst wall relatively complement-inert. The results of those studies are summarised in this work, with emphasis on the most recently identified of the complement inhibitory mechanisms. En los ultimos anos la produccion de papa en el Uruguay se ha visto muy afectada por la bacteria Ralstonia solanacearum, causante de la enfermedad conocida como murchera. Los esfuerzos tendientes al desarrollo de nuevas variedades de papa con caracterlsticas mejoradas, ubican a la especie silvestre Solanum commersonii Dun. como uno de 105 recursos nativos mas valiosos a ser usados en mejoramiento. Nuestro pais constituye un importante reservorio de diversidad de esta especie silvestre, cuyo potencial ha side evaluado en forma preliminar, pero que no se ha integrado en forma sistematica a un banco de germoplasma. Uno de 105 objetivos centrales de esta tesis consisti6 en evaluar la diversidad genetica de S. commersonii mediante la utilizacion de marcadores moleculares RAPD (Random Amplified Polymorphic DNA), AFLP (Amplified Fragment Length Polymorphism) y SSR (Simple Sequence Repeats). Se analizaron 30 accesiones colectadas en distintas regiones del pais. Los tres tipos de marcadores demostraron ser herramientas utiles para el estudio de la diversidad genetica de la especie S. commersoni~ generando resultados con elevada reproducibilidad y confiabilidad. EI analisis SSR fue el metodo que revelo un mayor nivel de polimorfismo, mientras que 105 marcadores AFLP y RAPD presentaron un mayor poder de discriminacion entre las accesiones analizadas. Los resultados de similitud genetica muestran un patron de agrupamiento c1aramente relacionado con el origen geogrclfico de las accesiones, revelando la existencia de dos grupos bien diferenciados provenientes de la zona norte y sur del Uruguay. Tambien se encontre un tercer grupo minoritario de accesiones que presentan marcadas diferencias respecto al resto de la coleccien. Estos conocimientos contribuyen a la racionalizacion en el mantenimiento de la coleccion de germoplasma de esta especie, aSI como al establecimiento de criterios para la seleccion de genotipos durante la planificacion de cruzamientos. Se realize tambien el primer relevamiento de cepas de R. solanacearum lograndose aislar, identificar, caracterizar y mantener ocho cepas provenientes de las principales zonas de produccion papera del Uruguay. Todos 105 aislamientos pertenecen a la Division II, biovar 2-A. No se encontraron diferencias entre 105 perfiles geneticos de las ocho cepas aisladas. Mediante ensayos de inoculacion en invernadero, se confirmo la resistencia de S. commersonii frente a R. solanacearum. Los niveles de resistencia no son uniformes en toda la especie, encontrandose distintas categorlas de resistencia entre las accesiones analizadas. No se encontro ninguna correlacion entre 105 perfiles geneticos de las accesiones de S. commersonii y 105 niveles de resistencia a R. soalanacearum. Sin embargo, a partir del analisis detallado de 105 perfiles geneticos generados, se encontro un marcador RAPD de 800 pb (R66800) presente unicamente en accesiones con fenotipo resistente. Se confirmo la homologla de secuencia de todos 105 fragmentos y se disenaron primers especificos logrando convertir este marcador anenimo RAPD en un marcador de secuencia caracterizada SCAR (Sequence Characterized Amplified Region). Se optimize un ensayo destinado a evaluar la presencia de sustancias con actividad antimicrobiana en extractos vegetales. Se detect6 aSl, que varios extractos acuosos preparados a partir de tuberculos de S. commersonii presentan actividad antimicrobiana frente a R. solanacearum, descartandose la naturaleza proteica del (105) compuesto(s) responsable(s) de esta actividad. Asimismo, se encontro que el contenido de protelnas solubles, 105 perfiles electroforeticos y 105 niveles de actividad hemaglutinante de estes extractos constituyen herramientas adicionales para reafirmar la diversidad que caracteriza a esta especie silvestre. Se confirme la presencia de una lectina en tuberculos de S. commersonii con la misma especificidad que la lectina de tuberculos de S. tuberosum pero diferente peso molecular de la subunidad, determinado mediante SDS-PAGE (90 KDa y 65 KDa, respectivamente). Si bien la lectina aislada de S. tuberosum mostro interaccien con celulas avirulentas de R. solanacearum, este reconocimiento no se observo con la lectina de S. commersoni~ reforzando las diferencias encontradas entre ambas protelnas. Tambien se determino que el exopolisacarido aislado a partir de celulas virulentas de R. solanacearum no es reconocido por ninguna de las dos lectinas. Los resultados generados a partir de este trabajo de tesis sobre el sistema S. commersonii - R. solanacearum establecen bases selidas para el maximo aprovechamiento de esta especie silvestre en la introduccion de resistencia frente a R. solanacearum y validan al sistema como modele para la busqueda de marcadores de resistencia. El objetivo de esta tesis fue desarrollar metodologías simples, rápidas, confiables y validadas que permitan la determinación en miel de dos grupos de analitos de interés por su toxicidad, alcaloides pirrolizidínicos y metales. Con este propósito, se buscó simplificar la etapa de preparación de muestra y emplear técnicas analíticas simples, de bajo costo y fácilmente disponibles. Se exploraron entonces, diversas técnicas tanto para la preparación de muestra como para la determinación. Para la preparación de muestra se emplearon técnicas como la extracción de punto nube, extracción en fase sólida, microextracción líquido-líquido dispersiva, extracción del tipo QuEChERs y QuPPe. Las técnicas empleadas para la determinación de los analitos de interés fueron también diversas dentro de la espectrometría atómica y la cromatografía. Se aplicó espectrometría de absorción atómica de llama, espectrometría de emisión atómica de plasma de acoplamiento inductivo, cromatografía de líquidos acoplada a detector de arreglo de diodos, de fluorescencia y a espectrometría de masas y la cromatografía gaseosa acoplada a espectrometría de masas. Se aplicaron técnicas de análisis en flujo como el análisis por inyección secuencial, el análisis en flujo multiconmutado y plataformas mesofluídicas como LOV. Se desarrolló un método automatizado para la determinación de cadmio y plomo empleando LOV y como método de preparación de muestra extracción en fase sólida empleando una resina de modo mixto, fase reversa e intercambio de cationes. 16 Desarrollo de metodologías analíticas para la determinación de contaminantes traza orgánicos e inorgánicos en miel Desarrollo de metodologías analíticas para la determinación de contaminantes traza orgánicos e inorgánicos en miel Se desarrolló un método mecanizado para la determinación de cadmio empleando microextracción líquido-líquido dispersiva cuyos parámetros de desempeño son adecuados al uso y en comparación con otros métodos reportados de similares características, tiene un mejor desempeño analítico y mayor rapidez. Ambos métodos cumplen con la característica de simplicidad en la preparación de muestra buscada y pueden ser empleados para evaluar la inocuidad de la miel desde el punto de vista de la contaminación por metales. Además, el método desarrollado a partir de la microextracción líquido-líquido dispersiva por su rapidez, puede ser usado para evaluar episodios de contaminación donde las abejas actúen como biomonitor a través de su producto la miel, y un número muy alto de muestras necesita ser procesado. Con respecto a los alcaloides pirrolizidínicos, se obtuvo como resultado de esta tesis un método para su determinación en miel empleando una variante de QuPPe (Quick Polar Pesticides Method) y cromatografía de líquidos acoplada a espectrometría de masas. La preparación de muestra consistió en la simple extracción de los compuestos en acetonitrilo acidificado. Los métodos fueron optimizados aplicando diseño de experimentos como diseño factorial, completo o central compuesto, o de forma univariante. Una vez optimizados los métodos se validaron y aplicaron al análisis de muestras del mercado. El desarrollo de esta tesis permitió entonces generar distintas metodologías que pueden contribuir a garantizar la inocuidad de la miel tanto para su consumo como su comercio. The system is based on the injection of an aliquot of the sample in a stream of organic solvent where it is dispersed, and measurement of the scattered radiation (measured as apparent absorbance) in the visible range (λ = 640 nm). An LED-based photometer was used for this purpose. The whole system including sample injection and data acquisitionwas controlled by a personal computer. Calibration curves exhibited good linearity (h = 0.415±0.016C+ 0.00±0.03, r2 = 0.9995, confidence level of 95%) in the range up to 2.68% (insolubles in pentane). Detection and quantification limits were respectively 0.07% and 0.16% (w/w). The method was validated by analysis of 25 real samples by the proposed method and the FTIR method finding high correlation. Waste generation and reactive consumption is much less than in the official method (ASTM D-893). The proposed method employs 25mL of kerosene per sample while the official method employs 200mL of pentane. An automatic programmable-flow system capitalizing upon mesofluidic Lab-On-Valve (LOV) sample processing coupled on-line to inductively coupled plasma atomic emission spectrometry (ICP-AES) is proposed in this work as a quality control tool for expedient assessment of potential contamination episodes of food safety elements (namely, Cd and Pb) in a variety of undigested ripened honeys, taken as a model of an edible matrix. On-chip micro-solid phase extraction (μSPE) is effected in a bead-injection disposable sorbent mode using a cationic exchange restricted access-like material (viz., Bond Elut Plexa PCX). We have proven that the lifetime of the miniaturized solid reactor on-chip (11 ± 0.6 mg) is limited to processing less than 6 mL of unfiltered sample suspension (5% (w/w) honey buffered at pH 4.5) after which the analytical performance is severely deteriorated because of strong adherence of the sample matrix to the bead surface. Several physicochemical parameters including the nature of the sorbent material and its ionic form were investigated in detail so as to maximize absolute recoveries and enrichment factors of Pb and Cd. Variables for the elution process were explored by means of a full factorial design. Using 4.0 mL of sample and 200 μL of 3.0 mol L−1 HNO3 as an eluent, enrichment factors of ca. 15 with extraction/elution efficiencies close to 80% and relative recoveries ranging from 90–111% in honey were obtained for both Pb and Cd. The limits of detection (LODs), based on the 3sintercept criterion, were 26 ng g−1 and 68 ng g−1 for Cd and Pb, respectively, which are far below those endorsed by current regulatory agencies (namely, 100 and 300–500 ng g−1 for Cd and Pb, respectively). Demonstrated by the analysis of a suite of off-the-shelf honey brands, the proposed LOV-μSPE platform hyphenated to ICP-AES is deemed suitable for the reliable quantitation of trace level concentrations of Pb and Cd in honey and the detection of heavy metal contamination episodes. Despite the success of immune checkpoint blockade for cancer therapy, many patients do not respond adequately. We aimed to improve this therapy by optimizing both the antibodies and their delivery route, using small monodomain antibodies (nanobodies) delivered locally with a self-amplifying RNA (saRNA) vector based on Semliki Forest virus (SFV). We generated nanobodies against PD-1 and PD-L1 able to inhibit both human and mouse interactions. Incorporation of a dimerization domain reduced PD-1/PD-L1 IC50 by 8- and 40-fold for anti- PD-L1 and anti-PD-1 nanobodies, respectively. SFV viral particles expressing dimeric nanobodies showed a potent antitumor response in the MC38 model, resulting in >50% complete regressions, and showed better therapeutic efficacy compared to vectors expressing conventional antibodies. These effects were also observed in the B16 melanoma model. Although a short-term expression of nanobodies was observed due to the cytopathic nature of the saRNA vector, it was enough to generate a strong proinflammatory response in tumors, increasing infiltration of NK and CD8+ T cells. Delivery of the SFV vector expressing dimeric nanobodies by local plasmid electroporation, which could be more easily translated to the clinic, also showed a potent antitumor effect. The alfa(v)beta(3) integrin is involved in angiogenesis and tumor metastasis. Arginine-glycine-aspartic acid (RGD)-peptides bind with high affinity to this integrin. This study compares the influence of (99m)Tc-labeling applying novel Technetium-cores on imaging characteristics of the radiolabeled peptide. Different peptide conjugates based on the cyclic pentapeptide c(RGDyK) (cRGD) were prepared and characterized (HYNIC-, Cys-, L2- and Pz1-cRGD). Radiolabeling experiments using different coligands for HYNIC-cRGD, the (99m)Tc(CO)(3) metal fragment for PZ-1-cRGD (pyrazolyl-derivative), the Tc-nitrido-core using a phosphine-coligand (PNP) for Cys-cRGD and an isonitrile-conjugate (L2-cRGD) together with a NS(3)-coligand (4+1 concept) were performed and showed labeling yields >90% at high specific activities. A high in vitro stability was observed, plasma protein binding and lipophilicity varied considerably between different radiolabeled cRGD conjugates. Experiments on biological activity of the radiolabeled peptides using alfa(v)beta(3) positive (M21) and negative (M21L) tumor cells did show specific uptake of various conjugates. Studies in tumor bearing animals revealed significant differences between different conjugates concerning pharmacokinetic behavior (predominant renal excretion to considerable hepatobiliary clearance) as well as tumor uptake (0.2-2.7%ID/g). Highest specific tumor uptake and tumor/background values were found for [(99m)Tc]EDDA/HYNIC-c(RGDyK), [(99m)Tc]Nitrido-PNP-Cys-c(RGDyK) and [(99m)Tc(CO)(3)]-Pz1-c(RGDyK). Using novel Tc-cores such as the (99m)Tc(CO)(3) metal fragment, Tc-nitrido- and the 4+1 concept peptides could be labeled with [(99m)Tc]technetium at high specific activities resulting in complexes with high stability, but binding moieties have to be optimized especially concerning hydrophilicity resulting in renal rather than hepatobiliary excretion. This comparative study underlines that peptide labeling strategies using (99m)Tc have to be properly selected and optimized. The vapor-phase epitaxy (VPE) deposition of CdTe on III–V heterosubstrates was performed in order to investigate the formation of micro-pixels usable in X-ray and thermo-photovoltaic converters. InSb and (Ga)InSb layers grown by molecular beam epitaxy on semi-insulating GaAs (1 0 0) wafers were employed as substrates. In these layers, the presence of Ga- and In-rich dots (200–500nm in diameter) was detected using AFM and SEM-EDAX techniques. We found that these dots, which are in liquid phase at the VPE growth temperature (450 1C), act as nucleation points during the VPE process and prompt the selective area deposition of CdTe leading to the formation of columnar structures. Two important trends related to the growth morphology were established. First, the group V element does not affect the growth of CdTe columnar structures, i.e. the formation of these structures is independent of As or Sb. The first experimental results about the growth and characterization of Pb1−xCdxTe new high-Z ternary alloy, grown by Vapour Phase Epitaxy (VPE) onto CdTe single crystal substrates are presented. SEM images have shown a well-defined interface between the substrate and the heterostructure bulk, without voids or inclusions. EDX analysis in a line scan mode was done over the layer crossections. The Te, Pb and Cd concentration profiles were found, verifying a strong inter-diffusion in the metallic sublattices and confirming the existence of the Pb1−xCdxTe ternary alloy. This has been also confirmed by X-ray diffraction measurements. For the longest growth times, two types of surface defects arose: PbTe aligned microcrystals and cylindrical-like Te-rich structures. Hall measurements showed that the carrier concentration of the layers decreases and the resistivity increases when the growth temperature increases. The influence of the surface defects on the electrical properties was also demonstrated. MT4-MMP (MMP17) belongs to a unique subset of MT-MMPs that are anchored to the cell surface via a glycosylphosphatidylinositol (GPI) moiety. However, little is known about its biochemical properties. Here we report that MT4-MMP is displayed on the cell surface as a mixed population of monomeric, dimeric, and oligomeric forms. Sucrose gradient fractionation demonstrated that these forms of MT4-MMP are all present in lipid rafts. Mutational and computational analyses revealed that Cys564, which is present within the stem region, mediates MT4-MMP homodimerization by forming a disulfide bond. Substitution of Cys564 results in a more rapid MT4-MMP turnover, when compared to the wild-type enzyme, consistent with a role for dimerization in protein stability. Expression of MT4-MMP in MDCK cells enhanced cell migration and invasion of Matrigel, a process that requires catalytic activity. However, a serine substitution at Cys564 did not reduce MT4-MMP-stimulated cell invasion of Matrigel suggesting that homodimerization is not required for this process. Deglycosylation studies showed that MT4-MMP is modified by N-glycosylation. Moreover, inhibition of Nglycosylation by tunicamycin diminished the extent of MT4-MMP dimerization suggesting that N-glycans may confer stability to the dimeric form. Taken together, the data presented here provide a new insight into the characteristics of MT4-MMP, and highlight the common and distinct properties of the GPIanchored MT-MMPs.
