A summary of the resource.
In this work, we report a novel polymorph of AgBiW2O8, which was synthesized by a simple arc melting method. The structural details of this compound were determined by X-ray powder diffraction, and elemental composition was confirmed with ICP-AES. β-AgBiW2O8 adopts the scheelite structure type with tetragonal space group I41/a and lattice parameters a = b = 5.3019(3) and c = 11.678(2) Å. Structural details and optical properties are discussed in detail. Diffuse reflectance spectroscopy (DRS) measurements of β-AgBiW2O8 showed light absorption profile extended to the visible region, which suggests the material has a smaller band gap energy than Bi2WO6, as expected due to contribution of Ag d-orbitals to the electronic structure.

Enantioselective synthesis of ampelomin B and epi-ampelomin B, D, and E was accomplished starting from toluene, through a chemoenzymatic sequence, in which stereoselective hydrogenation, Mitsunobu reaction, and regio- and stereoselective nucleophilic opening of an epoxide were used as the main transformations. Structural revision and absolute configuration of the natural compounds were carried out.

A summary of the resource.
Dendrotoxins are a group of peptide toxins purified from the venom of several mamba snakes. �- Dendrotoxin (�-DTx, from the Eastern green mamba Dendroaspis angusticeps) is a well-known blocker of voltage-gated K+ channels and specifically of Kv1.1, Kv1.2 and Kv1.6. In this work we show that �- DTx inhibited the ASIC currents in DRG neurons (IC50 = 0.8 �M) when continuously perfused during 25 s (including a 5 s pulse to pH 6.1), but not when co-applied with the pH drop. Additionally, we show that �-DTx abolished a transient component of the outward current that, in some experiments, appeared immediately after the end of the acid pulse. Our data indicate that �-DTx inhibits ASICs in the high nM range while some Kv are inhibited in the low nM range. The �-DTx selectivity and its potential interaction with ASICs should be taken in consideration when DTx is used in the high nM range.

Con el propósito de analizar la flexibilidad en dos diferentes planes de estudio de una misma carrera de grado a nivel universitario, se realizó una búsqueda bibliográfica de indicadores que dieran cuenta de la misma. Como resultado de esa búsqueda, en el presente trabajo se definen una serie de indicadores de flexibilidad curricular y se analizan los resultados de su aplicación. Se trabaja con cinco indicadores que dan cuenta de la comunalidad, de la autonomía y especificidad así como de las posibilidades de movilidad intra e intercarreras. Los resultados se presentan en forma de tablas comparativas para ambos planes y se comentan los aspectos más relevantes en relación a los valores obtenidos.

Introdução: Em 2015 o agente etiológico Mycobacterium tuberculosis (MTB), causador da tuberculose, alcançou 10,4 milhões de pessoas no mundo, ficando em primeiro lugar entre as doenças infecto-contagiosas que mais matam pessoas. O seu difícil combate é devido principalmente ao surgimento das cepas resistentes, por isso o descobrimento de novas moléculas se faz necessário. Neste trabalho determinou-se a concentração inibitória mínima (CIM90), que é a concentração capaz de inibir 90% do crescimento bacteriano dos ligantes hinokitiol (hino) e tropolona (trop) e seus complexos com Pt (platina) e Pd (paládio contra MTB H37Rv (ATCC 27294) e quatro cepas clínicas resistentes a fármacos. Determinou-se também o IC50, que é a concentração capaz de manter 50% da viabilidade celular, através do ensaio de citotoxicidade em células MRC-5 (fibroblasto pulmonar humano). Determinou-se o índice de seletividade (IS) que representa quanto um composto é mais seletivo para a morte do microrganismo sem causar dano celular. Objetivo: Avaliação do potencial anti-MTB dos ligantes hino e trop, seus complexos com Pd e Pt e seus precursores. Metodologia: Tanto o REMA, para determinar a CIM90, quanto o ensaio de citotoxicidade para determinar o IC50, são métodos de microdiluição em placas de 96 poços que utilizam a resazurina como revelador. O inóculo bacteriano utilizado foi de aproximadamente 105 UFC/mL e o intervalo de concentrações foi 25 - 0,098μg/mL. Para o ensaio de citotoxicidade o intervalo de concentrações utilizado foi de 100 - 0,39μg/mL. Para demostrar o IS, calculou-se a divisão dos valores obtidos de IC50/MIC. Resultados e discussão: Com os resultados do REMA nós observamos que a atividade anti-MTB de hino (35.46μM) e trop (29.02μM) sozinhos foram menos ativos que quando complexados com Pd e Pt. Encontramos entre Pd-hino (13.14μM), Pd-trop (9.01μM), Pt-hino (14.73μM), Pt-trop (17.60μM), que Pd-trop é o mais ativo. Já os melhores valores de IS foram para trop (7,02) e hino (7,03), pois os ligantes sozinhos são menos tóxicos para as células. Nos isolados clínicos nós observamos que a atividade foi menor que contra a cepa sensível (H37Rv), mas ainda observamos atividade para trop and Pd-trop confirmando o seu potencial anti-MTB, pois, trop apresentou atividade contra 3 cepas (18,30-99,44μM) e Pd-trop contra 2 (9,05-25,64μM) das 4 cepas testadas. Conclusão: Esses dados demonstram que os ligantes hino e trop apresentam atividade anti-MTB, mas a complexação com Pd e Pt melhora esta atividade. No entanto, a complexação com os metais levou a um aumento na citotoxicidade, apenas hino e trop, quando não complexados, apresentaram IS superiores a 7 enquanto os complexos apresentaram IS menor que 2. A pesquisa de novas moléculas é uma necessidade, visto o aumento da resistência aos antibióticos disponíveis.

A summary of the resource.
Two novel multicomponent crystal forms of the prodrug 5-fluorocytosine (5-FC), an ionic cocrystal and a monohydrate salt, with 2,4,6-trinitrophenol (picric acid, PA) and 3,5-dinitrosalicylic acid (DNSA), respectively, were rationally designed and synthesized. Nitro compounds are used to treat many diseases, such as cancer, one of the 5-FC targets via gene-directed prodrug therapy. In addition, within the wide range of applications for nitro compounds is the formation of charge-transfer complexes with organic molecules. Among all 5-FC crystalline structures depicted, only two contain nitro compounds: a salt with PA and a dihydrate cocrystal with 5-nitrouracil. Therefore, the two new single translucent pale gold prism crystals of 5FC-PA and 5FC-DNSA were successfully obtained by slow evaporation from aqueous solution and characterized by X-ray diffraction (single crystal and powder), Fourier Transform Infrared (FT-IR), and Raman (FT-Raman) spectroscopies. In both crystal structures, the 5-FC pyrimidine ring is protonated at the N3 atom and the respective packings are mainly stabilized by N–H⋯O H-bonds. For the 5FC-PA ionic cocrystal, the formation of triple H-bonds (two N–H⋯O and one charge assisted N–H+⋯N H-bonds) among the 5-FC molecules was observed. This is consistent with the expected stability of the duplex structure, which is considered the strongest association between two species. In addition to the structural study, Hirshfeld surface analysis was carried out based on the single crystal X-ray diffraction data. Thermal stability was assessed through thermogravimetric analysis (TGA), differential scanning calorimetry (DSC), and hot-stage microscopy (HSM), indicating that these compounds are stable up to approximately 200 °C. The results contribute to the diversity of new 5-FC solid forms and introduce two novel compounds, with possible application in fungal/cancer medicine formulations or to better understand the behavior of cytosine in DNA/RNA base pairing.

We report the chemical synthesis route, structural characterization, and physical properties of nanofluid magnetic graphite �NFMG� obtained from the previously synthesized bulk organic magnetic graphite �MG� by stabilizing the aqueous ferrofluid suspension with an addition of active cationic surfactant. The measured magnetization-field hysteresis curves along with the temperature dependence of magnetization confirmed room-temperature ferromagnetism in both MG and NFMG samples.

The optimization and validation study of a qualitative and quantitative multiclass, ethyl acetate (EtOAc) multi-residue method to straightforward monitor 48 compounds in liver (6 veterinary drugs and 42 pesticides) and 54 in muscle (5 veterinary drugs and 49 pesticides) followed by high performance liquid chromatography tandem mass spectrometry (HPLC–MS/MS) and gas chromatography mass-spectrometry determination (GC–MS) is presented. Several clean-up sorbents were evaluated looking for the best strategy for the removal of the matrix co-extractives. A combination of aluminium oxide, (Al2O3), C-18 and magnesium sulphate (MgSO4) yielded the best analytical results in terms of precision and accuracy. The method was validated at three fortification levels: 10, 100 and 250 µg kg−1. The percentages of recovery were between 70 and 114 % for bovine muscle and 70–118 % for liver. Repeatability and intermediate precision percentages were below 20 % for both matrices. Most of the compounds under study presented good linearity and quantification limits below their corresponding European Union (EU) and Codex Alimentarius maximum residue levels (MRLs). Twenty-two randomly taken real samples were analyzed with the validated methodology, trying to prove its effectiveness and suitability for routine analysis. The validated methodology represents a fast and cheap alternative for the simultaneous analysis of pesticides and veterinary drugs which can be easily extend to other compounds and matrices.

New 5-nitroindazole derivatives were developed and their antichagasic properties studied. Eight compounds (14–18, 20, 26 and 28) displayed remarkable in vitro activities against Trypanosoma cruzi (T. cruzi). Its unspecific cytotoxicity against macrophages was evaluated being not toxic at a concentration at least twice that of T. cruzi IC50, for some derivatives. The electrochemical studies, parasite respiration studies and ESR experiment showed that 5-nitroindazole derivatives not be able to yield a redox cycling with molecular oxygen such as occurs with nifurtimox (Nfx). The study on the mechanism of action proves to be related to the production of reduced species of the nitro moiety similar to that observed with benznidazole.

Chia (Salvia hispanica L) is recognized mainly by its oil quality and secondly by the mucilaginous fibre. In the last years, it has become a non-conventional protein source. Besides their nutritional quality, chia proteins show technofunctional properties that encourage their use in food industry. Among the functionality reported previously, superficial properties of chia proteins resulted particularly appropriate and comparable to those for amaranth protein isolates. The pH of the media used for chia proteins extraction from the chia seed flour affects their structural characteristics and profoundly influence their interfacial properties. This chapter deals with the study of both emulsion and foaming capacity of chia proteins isolated by alkaline extraction (at two different pH) followed by isoelectric precipitation at pH 4.5 and the description of the applied methodology. In addition, theory about the protein behaviour at interfaces and also colloidal destabilization process as well as mathematical models used for their characterization is deeply detailed. Since the highest solubility of chia proteins is achieved in alkali, both emulsifying and foaming activities were assayed at pH 8.5. In order to study formation and stabilization of these colloidal systems, chia protein concentration was low enough to allow the destabilization processes to be evaluated in a reasonable period of time.

<p>Metallo-β-lactamase (MBL) production in Gram-negative<br />bacteria is an important contributor to β-lactam antibiotic<br />resistance. Combining β-lactams with β-lactamase inhibitors (BLIs) is a validated route to overcoming resistance, but MBL inhibitors are not available in the clinic. On the basis of zinc utilization and sequence, MBLs are divided into three subclasses, B1, B2, and B3, whose differing active-site architectures hinder development of BLIs capable of “crossclass” MBL inhibition. We previously described 2-mercaptomethyl thiazolidines (MMTZs) as B1 MBL inhibitors (e.g., NDM-1) and here show that inhibition extends to the clinically relevant B2 (Sfh-I) and B3 (L1) enzymes. MMTZs inhibit purified MBLs in vitro (e.g., Sfh-I, Ki 0.16 μM) and potentiate β-lactam activity against producer strains. X-ray crystallography reveals that inhibition involves direct<br />interaction of the MMTZ thiol with the mono- or dizinc centers of Sfh-I/L1, respectively. This is further enhanced by sulfur-π<br />interactions with a conserved active site tryptophan. Computational studies reveal that the stereochemistry at chiral centers is critical, showing less potent MMTZ stereoisomers (up to 800-fold) as unable to replicate sulfur-π interactions in Sfh-I, largely through steric constraints in a compact active site. Furthermore, in silico replacement of the thiazolidine sulfur with oxygen (forming an oxazolidine) resulted in less favorable aromatic interactions with B2 MBLs, though the effect is less than that previously observed for the subclass B1 enzyme NDM-1. In the B3 enzyme L1, these effects are offset by additional MMTZ interactions with the protein main chain. MMTZs can therefore inhibit all MBL classes by maintaining conserved binding modes through different routes.</p>

Ischemic stroke is one of the leading causes of death and disability worldwide. This acute vascular event interferes with blood supply to the brain and induces a burst of free radicals such as nitric oxide and superoxide, producing peroxynitrite, a precursor of strong nitrating agents. Fibrinogen is one of the most abundant plasma proteins; it plays a role in the hemostatic system, mediating clot formation, which can be affected by nitrotyrosine formation. Ongoing research has shown that emoji can be used by children to discriminate food products, but it is unclear if they express emotions and how they are linked to emotional words. Little is known about how children interpret emoji in terms of their emotional meaning in the context of food. This study aimed at investigating the emotional meaning of emoji used to describe food experiences in 9–13‐year‐old pre‐adolescents and to measure related age and gender differences. The meaning of 46 emoji used to describe food experience was explored by: mapping emoji according to similarities and differences in their emotional meaning using the projective mapping technique, and linking emoji with emotion words using a check‐all‐that‐apply (CATA) format. The two tasks gave consistent results and showed that emoji were discriminated along the valence (positive vs. negative) and power (dominant vs. submissive) dimension, and to a lower extent along the arousal dimension (high vs. low activation). In general, negative emoji had more distinct meanings than positive emoji in both studies, but differences in nuances of meaning were found also among positive emoji. Girls and older pre‐adolescents (12–13 years old (y.o.)) discriminated positive emoji slightly better than boys and younger pre‐adolescents (9–11 y.o.). This suggests that girls and older pre‐adolescents may be higher in emotional granularity (the ability to experience and discriminate emotions), particularly of positive emotions. The results of the present work can be used for the development of an emoji‐based tool to measure emotions elicited by foods in pre‐adolescents.

The important role of emotions in consumers’ food-related decisions motivate ongoing research in this domain, and in the present research attention was directed to an area that remains under-investigated: identifying sensory drivers of product-elicited emotions. Assuming that product-elicited emotions depend on the value individuals attach to the sensory characteristics of products, liking was expected to moderate the sensory drivers of emotions. This motivated an exploration of differences in the emotional associations of consumer groups with different preference patterns, as well as the sensory drivers of product-elicited emotions. Across four studies (123–192 consumers per study), using different product categories, emotion questionnaire variants and consumer populations, support for three hypotheses was established: i) consumers with different preference patterns did not largely differ in how they described the sensory characteristics of products, but they differed in their emotional associations, and differences between segments with different liking patterns were larger for emotional product associations than for sensory product perceptions (H1), ii) differences in the emotional associations depended on the size of the difference in liking between the two groups, although some differences in the emotional associations of samples with similar liking scores were also established (H2), and iii) many of the sensory drivers of emotions differed between consumer segments with different preference patterns (H3). The latter has not previously been demonstrated in this level of detail and supports the growing recognition of segmentation analysis in product-focused consumer research.

Abstract

Healthy and sustainable diets are necessary for global development goals, but people struggle to modify their eating behaviours which are often habitual and resistant to change. However, disruption can provide the impetus for change, and in the present research we view the Covid-19 pandemic as a potential catalytic disruption with the potential to induce positive behaviour change. Data were obtained from ~900 USA adults, of which 44% selfreported to have made positive dietary change/s. Those who did so tended to be younger rather than older and with higher educational attainment. They also were less food neophobic than people who did not make positive change/s and placed greater importance on health as a motive of daily decisions about what to eat and drink. The contribution of psychographic variables in differentiating between groups of people who did / did not make positive changes once again show their importance in uncovering and explaining the complex factors that motivate food-related consumer behaviour. It seems this holds also when daily lives are disrupted by unforeseen events.

The structure of the O-antigen polysaccharide (PS) from the Shiga-toxin producing Escherichia coli O63 has been elucidated using a combination of bioinformatics, component analyses and NMR spectroscopy. The O-antigen is comprised of tetrasaccharide repeating units with the following structure: →2)-β-d-Quip3N(d-allo-ThrAc)-(1→2)-β-d-Ribf-(1→4)-β-d-Galp-(1→3)-α-d-GlcpNAc-(1→ in which the N-acetylated d-allo-threonine is amide-linked to position 3 of the 3-amino-3-deoxy-d-Quip sugar residue. The presence of a predicted flippase and polymerase encoded in the O63 gene cluster is consistent with the Wzx/Wzy biosynthetic pathway and consequently the biological repeating unit has likely an N-acetyl-d-glucosamine residue at its reducing end. A bioinformatics approach based on predictive glycosyltransferase function present in ECODAB (E. coli O-antigen database) suggested the structural element β-d-Galp-(1→3)-d-GlcpNAc in the O-antigen. Notably, multiple gene sequence alignment of fdtA and qdtA from E. coli to that in E. coli O63 resulted in discrimination between the two, confirmation of the latter in E. coli O63, and consequently, together with qdtB, biosynthesis of dTDP-d-Quip3N. The E. coli O63 O-antigen polysaccharide differs in two aspects from that of E. coli O114 where the latter carries instead an l-serine residue, and the glycosidic linkage positions to and from the Quip3N residue are both changed. The structural characterization of the O63 antigen repeat supports the predicted functional assignment of the O-antigen cluster genes.

A survey is presented of model building techniques and computer-assisted quantum-chemical and molecular-dynamics simulations, as applied to the study of protease and receptor design, to the determination of the properties of related effectors, agonist and antagonist molecules, and to the design of fatty-acid transport proteins and molecular carriers. Studies covering integral membrane protein design have been reviewed: they include porins, ion channels and G protein coupled receptors. In the transport molecule class, hydrophobic ligand transporters, such as serum and cellular retinoid binding proteins, have been reviewed.
