The palladium catalyzed cross-coupling reaction of phenyltrifluoroborate with a chemoenzymatically derived bromoazidoconduritol, combined with 1,3-dipolar cycloaddition, with a variety of alkynes is described. Fourteen new compounds were synthesized in moderate to good yields. The click chemistry reaction can be effected by using sodium ascorbate and CuSO4 · 5H2O as catalyst in toluene–H2O at room temperature

A summary of the resource.
The aim of this study is to elaborate and evaluate structural characteristics of soybean gelato by varying the content of soybean protein concentrate (2.95 to 17.05 %) and vegetable fat (7.95 to 22.05 %) using experimental design. The replacement of milk by hydrosoluble extract and soybean protein concentrate presented itself as an alternative to gelato production with unique characteristics, especially in terms of protein, solubility, viscosity, melting point, overrun and acceptability. The addition of up to 5 % (m/V) protein concentrate, 14 % (by volume) soybean hydrosoluble extract, and 15 % (by mass) vegetable fat to gelato formulations resulted in better structural characteristics, with viscosity ranging from 0.45–0.70 Pa∙s at 10 °C, a non-Newtonian behaviour and protein stability (total protein content 8.44 % and solubility 41 %). Soybean gelato structural analysis using X-ray diffraction revealed 15° and 35° diffraction angles at 2Θ, characterizing the crystalline part of the product. The thermal analyses showed four bands of mass loss in the temperature range of 40–600 °C, characterizing loss of moisture, decomposition of the soy protein and the fat/emulsifier of the formulations. Thus, the soybean gelato is an innovative product, lactose and milk protein-free with outstanding characteristics for the general public, mainly, for the populations with intolerance to such components.

Total IgG and IgG subclasses recognizing carbohydrate and peptidic epitopes from native and periodate treated partially purified hydatid cyst fluid (ppHCFA) and protoscolex somatic antigens (PSA) were tested by ELISA in hydatid patients sera. Binding of the major cross-reactive antipho- sphorylcholine antibodies was inhibited with free reagent. A predominant anticarbohydrate antibody response against ppHCFA and PSA is shown. Although the main contributing IgG subclass to the antipeptide response against both antigens was IgG4, IgG1 also significantly contributed to the anti-PSA peptidic epitopes response. Western blot showed that IgG1 antibodies strongly recognized in ppHCFA a periodate susceptible 38 kDa antigen. The IgG4 antibodies mainly recognized the periodate-resistant 12, 16 and 24 kDa antigens. In addition, IgG2 antibodies recognized three strongly periodate-susceptible broad bands (116, 55 and 24 kDa antigens). PSA-specific IgG1 and IgG4 antibodies showed similar patterns of antigen recognition as well as no significant reduction of reactivity after periodate treatment while the IgG2 antibody recogni- tion was strongly affected by this treatment. Furthermore, IgG2 showed significantly lower avidities than IgG1 and IgG4 antibodies recognizing both antigens. In conclusion, hydatid patients showed an enhanced production of low avidity anticarbohydrate IgG2 as well as high avidity antipeptide IgG4 antibodies.

The effects of destruction of carbohydrate moieties of Echinococcus granulosus antigen by sodium periodate treatment, and/or blocking phosphorylcholine specific antibodies by the addition of free reagent, were investigated to improve sensitivity and specificity of enzyme-linked immunoabsorbent assay (ELISA) for diagnosis of hydatid disease. Sera were collected, in Uruguay and Sweden, from patients with confirmed hydatidosis, non-hydatid tapeworm infections, other disorders, and healthy donors. ELISA performed with sodium periodate or free phosphorylcholine increased sensitivity, but decreased specificity. Western blot analysis showed reduced recognition of the 38 kDa antigen following both treatments, and of the 15.5 kDa antigen after periodate treatment. Addition of free phosphorylcholine enhanced the recognition of two antigens, 26 and 50 kDa, which may represent new cross-reacting epitopes.

An NADPH-dependent enzyme that reduces ethyl 2-methylacetoacetate stereoselectively to ethyl (2 R)-methyl-(3 S)-hydroxybutanoate was purified 730-fold from Escherichia coli. The N-terminal amino acid sequence data obtained from the purified reductase were used to search the E. coli genome, and a single match was found at the start of the yqhE open reading frame. The YqhE protein had been identified previously by Yum et al. as a 2,5-diketo-d-gluconate reductase on the basis of sequence similarity to other bacterial homologues [Yum, D.-Y.; Lee, B.-Y.; Pan, J.-G. Appl.Environ. Microbiol.1999, 65, 3341–3346]; however, it had not been examined for β-keto ester reductions. Our results thus link a key enzyme in the microbial production of ascorbate with stereoselective β-keto ester reductions, two important fields in biocatalysis. The purified YqhE reductase accepts ethyl acetoacetate and a variety of 2-substituted derivatives, and its sequence is similar to other aldose reductase superfamily members that also reduce α-substituted β-keto esters to syn-(2 R,3 S) alcohols.<br /><br />

<p>Marine natural products1 play an important role in drug development particularly in anticancer, antibiotics and antiparasitics drugs.2 Various bioactive cyanobacterial compounds exhibit unique structural features like cyclic peptides containing azole heterocycles. 3 It is well known that macrocyclic peptides can demonstrate drug-like physicochemical and pharmacokinetic properties such as good metabolic stability, solubility, lipophilicity and bioavailability.4 In addition, asymmetrical cyclopeptides are considered as new scaffolds for supramolecular chemistry and provides an important method for the development and preparation of nanoscale objects.5 Examples of azole cyclopeptides are aerucyclamide B (1),6 dendroamide A (2),7 and venturamide A (3),8 Figure 1. Aerucyclamide B was isolated in 2008 by Gademann and co-workers from the toxic freshwater cyanobacterium Microcystis aeruginosa PCC 7806 and displays potent and selective antiplasmodial activity against Plasmodium falciparum K1. This species is the most virulent of the genus Plasmodium and causes more than 95% of malaria-related morbidity and mortality. According to the World Health Organization 2011 report, there were an estimated 216 million episodes and 655000 deaths of malaria in 2010.9 Various antimalarial drugs are presently used for the treatment of this tropical disease, but the rapid spread of resistance seriously compromises their efficacy.10</p>

In this study we investigated the formation of isomers of lycopene, phytoene, and phytofluene from tomato and their theoretical energy. The results indicated that certain (Z)-isomers are favored thermodynamically and/or kinetically over their (all-E)-counterparts. The relative percentages of (5Z)-lycopene in either thermodynamic or kinetic equilibria were approximately 33%, and those of (all-E)-lycopene were only approximately 22%. Most strikingly (15Z)-phytoene was the major isomer (>90%) when the thermodynamic or the kinetic equilibria were reached. These observations can explain the high levels of lycopene (Z)-isomers found in humans and their rapid formations upon additions of oil to tomato products. In addition, the results can be useful to predict the isomeric forms of lycopene, phytoene, and phytofluene expected in foods as well as in plasma and tissues upon ingestion. In light of the data in the present study, the use of certain geometrical isomers of phytoene, phytofluene and lycopene on their own or as mixtures is recommended in future studies aimed at assessing their possible bioactivity.

A comparative genetic analysis of 42 clinical Klebsiella pneumoniae isolates, resistant to two or more antibiotics belonging to the broad-spectrum �-lactam group, sourced from Sydney, Australia, and three South American countries is presented. The study focuses on the genetic contexts of class 1 integrons, mobilizable genetic elements best known for their role in the rapid evolution of antibiotic resistance among Gram-negative pathogens. It was found that the class 1 integrons in this cohort were located in a number of different genetic contexts with clear regional differences. In Sydney, IS26-associated Tn21-like transposons on IncL/M plasmids contribute greatly to the dispersal of integron-associated multiple-drug-resistant (MDR) loci. In contrast, in the South American countries, Tn1696-like transposons on an IncA/C plasmid(s) appeared to be disseminating a characteristic MDR region. A range of mobile genetic elements is clearly being recruited by clinically important mobile class 1 integrons, and these elements appear to be becoming more common with time. This in turn is driving the evolution of complex and laterally mobile MDR units and may further complicate antibiotic therapy

One hundred four bacterial strains mediating urinary tract infections in separate individuals from a Uruguayan community were isolated. Forty-six strains conferred a multidrug resistance phenotype. All 104 strains were examined for the presence of class 1, 2, and 3 integrons. Class 1 integrons were found in 21 isolates across four distinct bacterial genera. A large class 1 integron in a Klebsiella pneumoniae strain was fully sequenced and was 29,093 bp in length. This integron probably arose by homologous recombination since it was embedded in a hybrid Tn21-like transposon backbone which comprised a Tn5036-like tnp transposition module at the IRi integron end and a Tn21 mer module at the IRt integron end. The parent integron/transposon that contributed the Tn5036 module was not related to Tn1696 since the integron insertion points in the transposon backbones were 16 bases apart. Examination of the other 20 class 1 integron-containing strains revealed further evidence of genetic exchange. This included a strain that possessed a Tn5036 module at the IRt end but not at the IRi end and another that possessed a tnp module beyond IRi that was a hybrid of Tn21 and Tn5051 and that is presumed to have arisen by site-specific recombination. This study highlights the ability of different genetic elements to act cooperatively to spread and rearrange antibiotic resistance in a community.

Several novel class 1 integrons/transposons were found in at least four distinct locations in the chromosome, including genomic islands. These elements seem to be undergoing successful dispersal by lateral gene transfer since integrons were identified across several lineages and more than one clonal line.

Abstract
A summary of the resource.
Substitution of nifurtimox tetrahydrothiazine moiety by triazol-4-yl, benzimidazol-1-yl, pyrazol-1-yl or related aromatic nitrogen heterocycles determines changes in the quantum chemistry descriptors of the molecule, namely, (a) greater negative LUMO energy; (b) lesser electron density on specific atoms, especially on the nitro group atoms, and (c) modification of individual net atomic charges at relevant atoms. These variations correlate with the greater capability of nifurtimox analogues for redox-cycling and oxygen radical production, after one-electron reduction by ascorbate or reduced flavoenzymes. Variation of the nitrofurans electronic structure can also explain the greater activity of nifurtimox analogues as inhibitors of glutathione reductase and Trypanosoma cruzi growth, although other factors, such as molecular hydrophobicity and connectivity may contribute to the latter inhibition.

Caenorhabditis elegans is a free-living nematode that has been validated for anthelmintic drug screening. However, this model has not been used to address anthelmintic dose-response-time and drug-drug interactions through matrix array methodology. Eprinomectin (EPM) and Ivermectin (IVM) are macrocyclic lactones widely used as anthelmintics. Despite being very similar, EPM and IVM are combined in commercial formulations or mixed by farmers, under the assumption that the combination would increase their efficacy. However, there is no data reported on the pharmacological evaluation of the combination of both drugs. In this study, we assessed the pharmacodynamics and drug-drug interactions of these two anthelmintic drugs. Since the action of these drugs causes worm paralysis, we used an infrared motility assay to measure EPM and IVM effects on worm movement over time. The results showed that EPM was slightly more potent than IVM, that drug potency increased with drug time exposure, and that once paralyzed, worms did not recover. Different EPM/IVM concentration ratios were used and synergy and combination sensitivity scores were determined at different exposure times, applying Highest Single Agent (HSA), Loewe additivity, Bliss and Zero Interaction Potency (ZIP) models. The results clearly indicate that there is neither synergy nor antagonism between both macrocyclic lactones. This study shows that it is more relevant to prioritize the exposure time of each individual drug than to combine them to improve their effects. The results highlight the utility of C. elegans to address pharmacodynamics studies, particularly for drug-drug interactions. Models in vitro can be integrated to facilitate preclinical and clinical translational studies and help researchers to understand drug-drug interactions and achieve rational therapeutic regimes.

The serious widespread development of nematode resistance has motivated the use of combined anthelmintic formulations. However, the advantages/disadvantages of the combined use of anthelmintics require further scientific characterization. The goals of the current trial were a) to characterize the pharmacokinetics of closantel (CLO) and moxidectin (MXD) administered both subcutaneously (sc) and orally either separately or co administered (CLO + MXD) to lambs; b) to compare the nematodicidal activity of both molecules given indi vidually or co-administered to lambs infected with resistant nematodes. Seventy (70) Corriedale lambs naturally infected with multiple resistant gastrointestinal nematodes were involved in the pharmacokinetic and efficacy trials. The animals were allocated into six groups (n = 10) and treated with either CLO, MXD, or with the CLO + MXD combined formulation by both the oral and sc routes. Additionally, an untreated control group (n = 10) was included for the efficacy trial. The efficacy was estimated by the faecal egg count reduction test (FECRT). Higher systemic exposure of both CLO and MXD was observed after the sc compared to the oral administration in lambs. The combined administration of CLO + MXD did not markedly alter their disposition kinetics. At 13 days post treatment, the administration of both molecules as a single active principle reached efficacy levels ranging be tween 80% (MXDoral), 84% (CLOoral), 85% (CLOsc), and 92% (MXDsc). The combined oral and sc treatments reached 99% efficacy. No adverse effects were observed after the combined treatment of CLO + MXD, and their co-administration did not show any adverse pharmacokinetic interaction. The combined effect of CLO + MXD successfully restored the maximum efficacy levels, which were not reached by the individual active ingredients.

The goals of the current trial were (a) to characterize the plasma disposition kinetics of levamisole (LEV), albendazole (ABZ) and ivermectin (IVM), each administered either alone (single active ingredient) or as a combined formulation to lambs; (b) to compare the clinical anthelmintic efficacy of the same drugs given either separately or co-administered to lambs infected with resistant nematodes. Fifty Corriedale lambs naturally infected with multiple resistant gastrointestinal nematodes were involved in the following experimental trials: (a) "Pharmacokinetic trial": the animals were allocated into five groups (n=10 each) and intraruminally treated with either LEV (8 mg/kg), ABZ (5mg/kg), IVM (0.2mg/kg), or with a LEV+ABZ+IVM combined formulation, where each active ingredient was administered at the same dose. Blood samples were collected over 15 days post-treatment and drug plasma concentrations measured by HPLC. (b) "Efficacy trial": the same treated groups plus an untreated control group were used to assess the comparative anthelmintic efficacy by the faecal egg count reduction test (FECRT). Although the overall LEV disposition kinetics was unaffected, significantly lower (61%) ABZ-sulphoxide and higher (71%) IVM systemic availabilities were obtained after administration of the combined formulation in comparison to those obtained after treatment with each drug alone. A multiple drug resistance situation was observed for Haemonchus spp. The observed efficacies were 52% (LEV), 72% (ABZ), 80% (IVM) and 87% (triple combined formulation). The results reported here contribute to the pharmaco-therapeutic knowledge on drug combinations. This type of research is crucial before further development of combined anthelmintic preparations reaches the market to deal with resistant nematode control. The co-administration of LEV+ABZ+IVM did not result in a significant advantageous anthelmintic effect compared to the treatment with IVM alone. The simultaneous/combined administration of LEV, ABZ and IVM may account for a drug-drug pharmacological interaction in infected lambs. The pharmacokinetic interaction accounted for a reduced ABZ-sulphoxide and enhanced IVM systemic exposure following the combined treatment.

Abstract
A summary of the resource.
We describe a rapid and efficient method for producing the capsular polysaccharide of Streptococcus pneumoniae by fermentation on tryptic soy broth and purification of this compound by using immobilized soybean lectin as an affinity adsorbent. In principle, the same strategy can be used to produce purified capsular polysaccharides from other streptococcal serotypes by selecting the appropriate lectin adsorbents.

Abstract

En esta Tesis se describe la estructura y comportamiento magnetico de los materiales cenimicos REBaCuCoOs+cS (RE = Y, La, Pr, Nd, Sm, Eu, Dy, Er y Tm) obtenidas por difracci6n de rayos X y neutrones de polvo policristalino y medidas de susceptibilidad magnetica. Se refinaron las estructuras nuclear y magnetica por el metodo de Rietveld combinando diagramas de difracci6n de neutrones y rayos X obteniendose, en algunos casos, panimetros estructurales de alta precisi6n. Los compuestos con RE = Y, Dy, Er y Tm resultaron isoestructurales y fueron refinados en una estructura tetragonal P4/mmm tipica de perovskitas 112 donde el Cu y el Co ocupan sitios distintos en la celda, observandose distancias Cu-O y Co-O consistentes con una piramide CuOs elongada en z y una piramide CoOs mas simetrica. Los momentos magneticos de los Momos de Cu(ll) y Co(Ill) en estado IS se ordenan antiferromagneticamente por debajo de la temperatura de Neel, que se estima esta entre 500 y 600 K. EI ordenamiento puede describirse mediante un grupo espacial bicolor I4'/m'm'm con aM = .J2 a Y CM = 2c. Los momentos magneticos orientados segun c, se ordenan antiferromagneticamente en el plano xy adquiriendo un ordenamiento +-1-+ en z donde el acoplamiento a traves del oxigeno apical de las piramides es antiferromagnetico. Los compuestos con RE = Sm y Eu, presentan una estructura tetragonal identica a los anteriores, excepto por la inclusi6n de cantidades apreciables de oxigeno no estequiometrico que aumenta al pasar de Dy a Eu y de Eu a Sm como 10 hace el radio i6nico de RE. Los compuestos con RE = Pr y Nd se describen mediante una estructura P4/mmm. La aparici6n de defectos de apilamiento en ambos compuestos produce formas de pico inusuales que son imposibles de ajustar con los algoritrnos disponibles en la actualidad. EI alto contenido de oxigeno no estequiometrico encontrado (8 ~ 0.5), produce cambios en el estado de oxidaci6n de Cu(II) que pasa a Cu(III) 10 que se asocian a una estructura magnetica distinta, que puede describirse en base al grupo bicolor I4'm'm con aM = .J2 a Y CM = c. EI compuesto LaBaCuCoOs+cS presenta una estructura diferente, dominada por un arreglo pseudo-cubico de Momos que fue descrita por el grupo espacial ortorr6mbico Pmmm, con una celda 1 xl x3ap siendo ap el parametro de celda de la perovskita cubica y 8 ~ 0.5. EI comportamiento magnetico de este compuesto es aparentemente similar al de los que contienen Pr y Nd, pudiendo observarse una aparente relaci6n entre la temperatura de transici6n y el cambio de tierra rara, reduciendose TN al aumentar el radio i6nico de RE.
