Abstract
A summary of the resource.
The terpenoid benzoquinone, rhodoquinone (RQ), is essential to the bioenergetics of many organisms that survive in low oxygen environments. RQ biosynthesis and its regulation has potential as a novel target for antimicrobial and anti-parasitic drug development. Recent work has uncovered two distinct pathways for RQ biosynthesis which have evolved independently. The first pathway is used by bacteria, such as Rhodospirillum rubrum, and some protists that possess the rquA gene. These species derive their RQ directly from ubiquinone (UQ), the essential electron transporter used in the aerobic respiratory chain. The second pathway is used in animals, such as Caenorhabditis elegans and parasitic helminths, and requires 3-hydroxyanthranilic acid (3-HAA) as a precursor, which is derived from tryptophan through the kynurenine pathway. A COQ-2 isoform, which is unique to these species, facilitates prenylation of the 3-HAA precursor. After prenylation, the arylamine ring is further modified to form RQ using several enzymes common to the UQ biosynthetic pathway. In addition to current knowledge of RQ biosynthesis, we review the phylogenetic distribution of RQ and its function in anaerobic electron transport chains in bacteria and animals. Finally, we discuss key steps in RQ biosynthesis that offer potential as drug targets to treat microbial and parasitic infections, which are rising global health concerns.

A summary of the resource.
Se determinó la incidencia de la infección toxoplásmica en 449 mujeres de 15 a 40 años en Montevideo con la reacción de Sabin y Feldman. Se calculó que entre 2,5 y 4,2 de cada 1000 fetos se infectarian con T.gondii , sufriendo las consecuencias patológicas conocidas. Se comparan los resultados con los obtenidos en otros países. Se discuten los recursos para la prevención o atenuación del daño fetal por Toxoplasma : la educación sanitaria de la embarazada y la determinación del IgM total en sangre del cordón, seguida de la búsqueda de anticuerpos específicos para el grupo de infecciones prenatales (toxoplasmosis, citomegalovirus, rubéola, herpes, sífilis). Se resalta la importancia de proseguir las investigaciones sobre una vacuna para aplicar a las mujeres seronegativas que deseen quedar embarazadas y de la vacunación de los gatos contra la eliminación de ooquistes ya factible.

In the search for new metal-based drugs for the treatment of Chagas disease, the most widespread Latin American parasitic disease, novel complexes of the bioactive ligand risedronate (Ris, (1-hydroxy-1- phosphono-2-pyridin-3-yl-ethyl)phosphonate), [MII(Ris)2]·4H2O, where M Cu, Co, Mn and Ni, and [NiII(Ris)2 (H2O)2]·H2O were synthesized and characterized by using analytical measurements, thermogravimetric analyses, cyclic voltammetry and infrared and Raman spectroscopies. Crystal structures of [CuII(Ris)2]·4H2O and [NiII(Ris)2(H2O)2]·H2O were solved by single crystal X-ray diffraction methods. The complexes, as well as the free ligand, were evaluated in vitro against epimastigotes and intracellular amastigotes of the parasite Trypanosoma cruzi, causative agent of Chagas disease. Results demonstrated that the coordination of risedronate to different metal ions improved the antiproliferative effect against T. cruzi, exhibiting growth inhibition values against the intracellular amastigotes ranging the low micromolar levels. In addition, this strong activity could be related to high inhibition of farnesyl diphosphate synthase enzyme. On the other hand, protein interaction studies showed that all the complexes strongly interact with albumin thus providing a suitable means of transporting them to tissues in vivo.

Abstract
A summary of the resource.
Co-elicitation of hedonic responses and sensory responses is now common. Against<br />this background, this research compared the risk of hedonic bias from sensory checkall-that-apply (CATA) questions and sensory term ratings from unstructured line scales (RQ1) and evaluated if the risk of hedonic bias is independent of the sensory<br />terms of the CATA question (RQ2). Seven studies with 137–293 participants each were conducted, using different product categories. Between-subjects experimental designs were used and allocated consumers to one experimental condition (EC). For RQ1, these were hedonic, hedonic immediately followed by a CATA question, and<br />hedonic immediately followed by a rating of term applicability (“not at all” to “extremely”). In RQ2, both ECs involved hedonic ratings followed by a CATA question, but the sensory terms differed. Results showed a significant effect of EC on<br />hedonic ratings in three of five studies for RQ1, suggesting that hedonic bias may or may not occur. For RQ2, the terms of the CATA question influenced hedonic ratings in two of three studies, and a negative hedonic effect was seen when CATA terms drew attention to particular characteristics of individual samples. Collectively, the results suggest that likelihood of bias depends on interactions between product category, samples, and terms.<br />Practical applications<br />Results from the present work suggest that hedonic bias in sensory co-elicitations<br />using CATA questions and scales may occur depending on the type of sensory attributes<br />considered in the study. The consideration of terms that trigger attention to specific sensory attributes of the samples may lead to changes in hedonic ratings.<br />Rating scales appear more likely to be associated with hedonic bias compared to CATA questions due to their more analytic approach to sample evaluation.

<p>Introducción: Jatropha curcas L. es una especie nativa de México que recientemente se ha considerado un cultivo importante para la producción de biocombustible.</p>
<p>Objetivo: Aislar e identificar rizobacterias de J. curcas con actividad antagónica frente a hongos fitopatógenos (Curvularia lunata y Fusarium equiseti) del cultivo y evaluar la capacidad de estas para promover el crecimiento de plantas de jitomate.</p>
<p>Materiales y métodos: Las cepas bacterianas se obtuvieron por dilución en serie. La actividad antagónica se evaluó mediante compuestos volátiles y difusibles en medio de cultivo PDA; además, se determinó la producción de compuestos antifúngicos y metabolitos que promueven el crecimiento de plantas. Las cepas se identificaron mediante la amplificación de los genes ARNr 16S y sodA; posteriormente, se inocularon en semillas de jitomate para evaluar la germinación de las plantas y la promoción del crecimiento.</p>
<p>Resultados y discusión: Las bacterias antagonistas se identificaron como Bacillus mojavensis, Bacillus subtilis, Bacillus thuringiensis y Pseudomonas aeruginosa. El efecto antifúngico sobre el crecimiento micelial de C. lunata y F. equiseti se debió a la presencia de compuestos volátiles y difusibles con inhibición de entre 30 % y 79 %. Pseudomonas aeruginosa mostró el efecto inhibitorio más fuerte (48.7 a 79.8 %). Bacillus subtilis (A1), B. mojavensis (A4) y B. thuringiensis (A6, A8) mostraron la mayor germinación (70 % a 88 %). El crecimiento de las plantas de jitomate tratadas con cepas bacterianas fue significativamente mayor (P < 0.05) que el de las plantas testigo.</p>
<p>Conclusión: Ocho aislados de rizobacterias de J. curcas mostraron actividad antagónica frente a C. lunata y F. equiseti, y demostraron su capacidad para mejorar la germinación y promover el crecimiento de las plantas de jitomate.</p>

This paper reports on the generation of monoclonal antibodies and the development of a new enzyme-linked immunosorbent assay (ELISA) for the detection of molinate (S-ethyl hexahydroazepine-1-carbothioate). Hybridoma cells were generated using spleen and lymph node cells from a mouse immunized with S-2-carboxyethyl hexahydroazepine-1-carbothioate conjugated to keyhole limpet hemocyanin. After screening with a competitive ELISA, two monoclonal antibodies, mAbs 16C11 and 14D7, with IC50 values of 82 ± 2 and 173 ± 8 ng/mL, respectively, were selected. MAb 16C11 can detect molinate concentrations of 1 ng/mL with no cross-reactivity to any other thiocarbamate pesticides; however, it was susceptible to the presence of organic solvents and to variation in buffer ionic strength. MAb 14D7 tolerated concentrations up to 5% of propylene glycol and 12.5% of acetonitrile in the assay buffer. The sensitivity of mAb 14D7 was further improved by decreasing the amount of coating antigen in the ELISA; the final inhibition assay showed an IC50 of 69.2 ± 1.4 ng/mL. In summary, mAb14D7 provided a more sensitive and robust assay, as compared with previous polyclonal antibody-based assays, with the additional advantage of being based upon a consistent and unlimited source of a defined reagent.

Elevated wine pH is one of the main problems affecting the quality of Vitis vinifera L. Tannat wines in Uruguay. High soil fertility, promoting vigorous vines, and high potassium availability result in high pH grapes and wine. The main objective of this work was to determine the effects of partial leaf and lateral shoot removal practices on the reduction of juice and wine pH over three growing seasons. Partial defoliation in shaded canopies significantly reduced grape potassium accumulation, malic to tartaric acid ratio, and wine pH without negatively affecting other quality parameters of soluble solids, organic acids, anthocyanins, and phenolics.

The aim of the present report is to show the synergistic effect of conducting polyaniline and manganite nanoparticles on the low field magnetoresistance response. We specially focused on the role of the conducting polyaniline interphase in a broad range of temperatures and low applied magnetic fields. Our results showed an increment on the electrical conductivity and enhancement on the magnetoresistance for lower amounts of conducting polyaniline additions with respect to the manganite unloaded samples. We also found that close to room temperature, the changes in magnetoresistance are mostly due to an increment of the spin polarization and spin disorder due to the presence of polyaniline interphase. However, at intermediate temperatures the polyaniline interphase intrinsic magnetoresistance showed an additional contribution to the total negative magnetoresistance response. These results, suggest that conducting polymer interphase shows both passive and active roles in the enhancement of the low field magnetoresistance of inorganic-organic nanocomposites.

Aspergillus spp. and Penicillium spp. are the main producers of ochratoxin A (OTA), a mycotoxin responsible for fatal human diseases. Some authorities have established a maximum of 2 μg/L of OTA in wine. Although the incidence and occurrence of OTA in grapes and wine is highly related to climate conditions, as has been extensively documented, there is no conclusive information on the effects of cultivation systems on the presence of OTA. This study focuses on determining the effect of the trellis system, planting density and cordon height on plant microclimate and thus on Aspergillus spp. contamination and OTA production in Tannat wines in Southern Uruguay. Two experiments were conducted during the 2010–2011 growing season: (1) a strip split plot design with five replicates and two cordon heights (CH) (0.5 m and 1.0 m above the soil) were compared in two planting densities (PD) (0.8 and 1.5 m between plants); (2) a randomised complete block design, vertical shoot positioning (VSP) versus Lyra trellis systems were evaluated. The results suggest that, even the macro- and micro-climate growing conditions play an important part in Aspergillus developing on grapes. Agronomical practices also have an undoubted impact on the risk and control of OTA accumulation in wine.

In this report, we studied the interaction between 9-vinylcarbazole (VK) –i.e. the monomer of poly(9-vinylcarbazole) (PVK)- and a TiO2 (101) surface in order to shed some light in the interface phenomena of this promising fullerene-free donor:acceptor system as active layer for solar cells. Since the role of the TiO2 surface defects is the main concern of our study, we proceeded to simulate the adsorption of the molecule on TiO2 with oxygen or titanium atom vacancies and without any defect. All these systems were studied using First Principles calculations, utilizing Density Functional Theory (DFT). The adsorption energy of the VK monomer-TiO2 systems was favorable in all cases. The presence of vacancies in the TiO2 surface yielded modifications in the electronic properties: the difference between the LUMO level of VK and the conduction band of TiO2-O surface is only ~0.2 eV, thus allowing to infer a decrease in the energy barrier associated to the injection of electrons from VK monomer towards the TiO2-O surface. The charge density differences in all systems also revealed that the donor:acceptor model is plausible and concurs with previous results, suggesting promising features for potential application as a donor:acceptor systems in fullerene-free solar cells.

The properties of mid-band-gap electronic states are central to the potential application of self-assembled, hybrid organic–inorganic perovskite-like quantum wells in optoelectronic technologies. We investigate broadband light emission from mid-band-gap states in fast-forming hybrid organic lead iodide quantum wells at room temperature. By comparing temperature- and intensity-dependent photoluminescence (PL) spectra emitted from butyl ammonium spaced inorganic layers, we propose that structural defects in a metastable material phase trap excitons and cause broadband light emission spanning wavelengths between 600 and 1000 nm. We use temperature-dependent terahertz time-domain spectroscopy to correlate changes in the subgap PL emission with changes in the chemical bonding of the inorganic octahedral layer. Our results provide new fundamental physical insights into the array of mechanisms capable of inducing broadband light emission from low-dimensional perovskite-like materials central to their application in future optoelectronic technologies and novel spectroscopic tools to characterize these states.

Rosemary oils from cultivars of Rosmarinus officinalis L. growing in different areas of Uruguay and South of Brazil (Rio Grande do Sul State) were analyzed by GC and GC/MS. The oils from Uruguay were found to be rich in αpinene (37.8–46.2%) and 1,8-cineole (13.4–13.8%). The oil from R. officinalis which was cultivated in Brazil contained αpinene (32.2%) and 1,8-cineole (14.7%), while the oil from wild plants found in Brazil contained αpinene (12.4%), myrcene (22.7%) and 1,8-cineole (15.3%).

Three novel bimetallic hybrid crystals featuring rubidium–alkaline-earth metal pairs and trifluoroacetato ligands were synthesized, and their utility as self-fluorinating single-source precursors to the corresponding mixed-metal fluorides was demonstrated. Rb2Mg2(tfa)6(tfaH)2·3H2O, RbCa(tfa)3, and RbSr2(tfa)5 (tfa = CF3COO–; tfaH = CF3COOH) were synthesized in both single-crystal and polycrystalline forms via solvent evaporation. Their crystal structures were solved using single-crystal X-ray diffraction (XRD), and chemical purity was confirmed using thermal analysis (TGA/DTA). Metal–oxygen–metal connectivity in Rb2Mg2(tfa)6(tfaH)2·3H2O was restricted to four-metal building blocks. In contrast, RbCa(tfa)3 and RbSr2(tfa)5 were found to be extended inorganic hybrids (Cheetham et al. Chem. Commun. 2006, 0, 4780−4795) exhibiting infinite metal connectivity in three and two dimensions, respectively. Systematic analysis of the coordination modes of the trifluoroacetato ligand revealed its ability to bridge alkali and alkaline-earth metals. Rietveld analysis of powder X-ray diffraction data (PXRD) showed that thermal decomposition of Rb2Mg2(tfa)6(tfaH)2·3H2O and RbCa(tfa)3 under inert atmosphere yielded crystalline RbMgF3 and RbCaF3, respectively. This solid-state transformation occurred without the need for an external fluorinating agent because the trifluoromethyl group acted as a built-in fluorine source. Solid-state and solution thermolysis of Rb2Mg2(tfa)6(tfaH)2·3H2O provided access to the hexagonal and cubic polymorphs of the fluoroperovskite RbMgF3, respectively. Findings reported in this article highlight that bimetallic trifluoroacetates offer unique features from the standpoint of both crystal lattice topology and reactivity.

<p>In this study, optimization, extension, and validation of a streamlined, qualitative and quantitative multiclass, multiresidue method was conducted to monitor >100 veterinary drug residues in meat using ultrahigh-performance liquid chromatography–tandem mass spectrometry (UHPLC–MS/MS). Optimization centered on extensive ruggedness evaluation of the method. Various clean-up sorbents were tested and the amount of co-extractives were weighed, matrix effects were measured using post-column infusion of representative analytes, the effect of extract dilution before injecting was studied, and analyte recoveries and reproducibilities were determined. In order to extend our previous method, more drug analytes were added that possessed a wider range of chemical properties, and a re-appraisal of different types of C18 in dispersive solid-phase extraction clean-up and mobile phases in UHPLC–MS/MS was done. Ultimately, end-capped C18 and post-column infusion of ammonium formate as an ionization enhancer for the late-eluting anthelmintics were found to give improved qualitative results for greater analytical scope. A multi-day, multi-analyst validation demonstrated that the final method is suitable for screening of 113 analytes, identifying 98 and quantifying (recoveries between 70–120% and RSD < 25%) 87 out of the 127 tested drugs at or below US regulatory tolerance levels in bovine muscle</p>

The best demonstration of the ruggedness of a method is monitoring its performance on an ongoing basis as part of the analytical quality control applied in the laboratory. However, an initial demonstration of the ruggedness is often performed as one aspect of the method validation, to give confidence that the method should perform well under normal variations in conditions in routine application. This initial ruggedness testing is typically performed using either multiple replicate analyses or application of design of experiments (DoEs) which minimizes the number of analyses, time and effort required to detect influences on the measurement results. Two DoEs were applied for ruggedness testing for a modified QuEChERS multiresidue method for the detection of pesticide residues in potato by GC–MS/MS. Seven experimental factors were studied using an eight-run Plackett–Burman design replicated three times and an augmented definitive screening design with 34 runs. The relative effectiveness of the two approaches is discussed, in terms of their statistical significance, their cost-effectiveness and the richness of information they provide on the effects of the parameters investigated and the actual robustness of the method being tested.

Russian sturgeon (Acipenser gueldenstaedtii) has been successfully farmed in Uruguay for the past ten years. However, during the Uruguayan summer fish endure high water temperatures and increased bacterial infections that threaten aquaculture. Our understanding of sturgeon's immune system and its interplay with environmental factors like temperature is almost unknown. This study analysed the way in which seasonal variations affect enzymatic blood components of Russian sturgeon's innate defences, including the serum alternative complement pathway (ACP), ceruloplasmin (Cp) and lysozyme activities. Results showed that summertime conditions in the farm altered these defences in different ways, inducing a significant decrease in ACP and Cp, and an increase in lysozyme. In addition, serum levels of total protein and cortisol decreased in summer, suggesting a chronic stress response was induced in parallel. Subsequently, we analysed whether the increase in water river temperature during summer could account for the observed results. To that end, we acclimated juvenile sturgeons to mild (18 °C) or warm (24 °C) temperatures for 37 days. Like in summer, sturgeons exposed to 24 °C showed lower levels of serum ACP, Cp and total proteins, together with a progressive decrease in body weight and increased fish mortality. Administration of an immunostimulant containing Se and Zn slightly reverted the temperature-induced effects on sturgeon's defences. Altogether, our study provides novel data on various physiological parameters of the Russian sturgeon and highlights the impact warm temperature has on stress and innate immunity in this chondrostean fish.
