A summary of the resource.
[2 × 2] grids of general formula [Ln4L4Cl4(DMF)4].2H2O (Ln = Sm (1), Eu (2), Tb (3); H2L = (E)-N’-((E)- (hydroxyimino)butan-2ylidene)salicylolhidrazide; DMF = dimethylformamide) were successfully constructed using a lanthanide ion as corners and L2-, a Schiff base, as linkers. The single crystal X-ray diffraction study reveals that compounds 1 and 3 are neutral tetranuclear complexes. They feature a [2 × 2] grid distorted skeleton. Coordination sphere of Ln ions are completed by one chloro ligand and a solvent molecule. The grids were obtained after a careful study of the chemical speciation of these systems performed by potentiometric titrations (25.0 °C in 40:60 DMF:water mixture, I = 0.50 mol L-1 NMe4Cl).

A summary of the resource.
Heteropolynuclear metal organic frameworks (MOFs) of general formula [Ln2Ni3(oda)6(H2O)6]·xH2O (oda = oxydiacetate, Ln = +3 rare earth ion) were synthesized and characterized. Most of them bear a honeycomb structure with 1D channels containing 11 to 14 crystallization water molecules per formula unit. The size of the channels and the number of crystallization water molecules they contain decrease as the Ln ionic radii gets smaller along the series from La to Tm. Besides, the water content changes reversibly with temperature and relative humidity conditions. Accordingly, the as-synthesized [Tm2Ni3(oda)6(H2O)6]·11H2O picks-up water when exposed to 100% relative humidity atmosphere to become [Tm2Ni3(oda)6(H2O)6]·14H2O. Computational calculations show the formation of a hydrogen bond network in which layers of water molecules are arranged and associated primarily to the polar groups of the framework walls, anticipating good proton conduction along the [001] channels. Furthermore, higher confinement of water molecules in the smaller channels could favor the water molecules mobility. Indeed, the protonic conductivity of [Tm2Ni3(oda)6(H2O)6]·14H2O is 2.43 × 10−3 S·cm−1 at 70 °C, similar to standard conductive MOFbased materials reported. A vehicular carrier mechanism by means of H3O+ species diffusing along the 1D channels of the porous structure seems to be operative, with a high activation energy value of 1.46 eV.

The cupric reducing antioxidant capacity (CUPRAC) assay is an established procedure used to measure antioxidant content. Recently, the CUPRAC assay was adapted for use in post column derivatisation coupled with high performance liquid chromatography (HPLC-PCD). In prior works reported in the literature, CUPRAC HPLC-PCD assays used large post column mixing coils leading to a deterioration in the observed separation efficiency. In this work we used the CUPRAC PCD assay with reaction flow (RF) chromatography. No reaction loop was required, thus preserving the efficiency of the separation. We subsequently compared the performance of the CUPRAC assay to that of an antioxidant assay that we have previously converted to RF chromatography, namely the ferric reducing antioxidant potential (FRAP) assay, both in RF mode. The relative response factor with respect to trolox was measured for both RF assays via the separation of a standard mixture of 16 compounds. Sample analysis of coffee and green tea via both RF assays was used to compare their ability to screen for bioactive candidates in complex natural products. In general, the CUPRAC assay is recommended as the RF PCD methodology of choice as it provided a higher level of performance compared to the FRAP assay, demonstrating less baseline noise interference, greater sensitivity, wider linear dynamic range and better assay precision.

Abstract
A summary of the resource.
The aim of the present research was to explore consumers' conceptualization of feeling good in relation to food and beverages from a cross-cultural perspective. Participants from 14 countries across 5 continents and covering 10 languages (N = 8325) responded to an online survey including word association and free listing tasks related to feeling good in the context of food and beverages. Results were analyzed using inductive coding: a list of main codes was generated in English for each of the tasks, after which native speakers for each language coded the responses. Codes were grouped into categories reflecting common themes from which eight dimensions were identified. Results showed that in the context of foods and beverages, feeling good was mainly associated with specific foods and sensory and hedonic properties. Across the 14 countries, ‘Sweet and fat food’, ‘Fruit and vegetables’, and ‘Protein food’ were the three food categories most associated with feeling good. Emotional aspects of food consumption (‘Taste good’ and emotions) were also relevant. Health and nutrition-related aspects were more relevant for consumers when they were asked to think about how foods and beverages would make them feel good in the future. In other words, food-related feeling good seems to be mainly driven by sensory pleasure at present, but it is also related to nutrition and health in the future. Differences in the strength of the associations between feeling good and the identified categories were found between countries, in line with the existence of cultural differences in food habits, as well as in the importance people attach to the characteristics of foods and beverages. Results from the present work provide insights on the impact of eating and drinking on feeling good in terms of emotional, physical and social aspects, and increase knowledge about the way food and drink can contribute to general well-being.

We have used Salmonella as a delivery system for eukaryotic expression plasmids encoding cytokines, and assessed its capacity to modulate immune responses in different experimental models. Plasmids encoding mouse IL-4 and IL-18 under cytomegalovirus promoter were constructed and transformed into live attenuated Salmonella enterica serovar Typhi strain CVD 908-htrA, and Salmonella enterica serovar Typhimurium strain SL3261. We have shown that systemic as well as mucosal immunization with such constructs can influence the antibody and cytokine responses to the Salmonella carrier and to co-administered bystander antigens, as well as the specific immune response elicited during a parasitic infection. Further, we have shown that oral cytokine-therapy using Salmonella as gene vector induce antitumoral effect as demonstrated by extended survival time in melanoma-bearing mice. This approach may be particularly suited for the development of new immunotherapies with applications in parasitic infections and cancer, were alterations of the host’s immune responses are usually found, and therapy-induced modulation of the immune response is likely to be required

Triamcinolone acetonide (TA) is a powerful anti-inflammatory drug used in the treatment of inflammatory ocular disorders; however, its poor aqueous solubility and ocular anatomical barriers hinder optimal treatment. The aim of this work was to obtain triamcinolone acetonide nanocrystals (TA-NC) to improve ocular corticosteroid therapy. Self-dispersible TA-NC were prepared by the bead milling technique followed by spray-drying, exhaustively characterized and then evaluated in vivo in an ocular model of endotoxin-induced uveitis (EIU). Self-dispersible TA-NC presented an average particle size of 257 ± 30 nm, a narrow size distribution and a zeta potential of −25 ± 3 mV, which remained unchanged for 120 days under storage conditions at 25 ◦C. In addition, SEM studies of the TA-NC showed uniform and spherical morphology, and FTIR and XRDP analyses indicated no apparent chemical and crystallinity changes. The subconjunctival administration of TA-NC in albino male white rabbits showed no clinical signs of ocular damage. In vivo studies proved that treatment with self-dispersible TA-NC alleviated the inflammatory response in the anterior chamber and iris in EUI rabbit eyes. Dispersible TA-NC are a promising approach to obtaining a novel nanometric TA formulation for ocular disorders.

Time orientation can influence health-related behaviors, including food consumption. The aim of the present work was to study the influence of time orientation on food choice, using cookie labels as case study. A choice-conjoint task was designed using labels differing in type of cookie (chocolate chips vs. granola), front-of-pack nutrition information (nutritional warnings vs. Facts Up Front system) and nutritional claim (no claim vs. “0% cholesterol. 0% trans fat”). An online study was conducted, in which 155 participants evaluated 8 pairs of cookie labels and selected the one they would buy if they were in the supermarket. Then, they were asked to complete a consideration of future consequences scale (CFC) adapted to eating habits, as well as a questionnaire about socio-demographic characteristics. Time orientation influenced participants' choices of cookies labels; particularly the importance attached to type of cookie. Participants with greater consideration of future consequences preferred the granola cookies, associated with health, while those who prioritized immediate consequences preferred chocolate chip cookies. In addition, nutritional warnings discouraged choice regardless of participants' time orientation. Results from the present work provide additional evidence of the influence of time preferences on food choices and suggest that strategies to stimulate and generate a more future-oriented perspective on eating habits could contribute to more healthful food choices.

The variable VHH domains of camelid single chain antibodies have been useful in numerous biotechnology applications due to their simplicity, biophysical properties, and abilities to bind to their cognate antigens with high affinities and specificity. Their interactions with proteins have been well‐studied, but considerably less work has been done to characterize their ability to bind haptens. A high‐resolution structural study of three nanobodies (T4, T9, and T10) which have been shown to bind triclocarban (TCC, 3‐(4‐chlorophenyl)‐1‐(3,4‐dichlorophenyl)urea) with near‐nanomolar affinity shows that binding occurs in a tunnel largely formed by CDR1 rather than a surface or lateral binding mode seen in other nanobody‐hapten interactions. Additional significant interactions are formed with a non‐hypervariable loop, sometimes dubbed “CDR4”. A comparison of apo and holo forms of T9 and T10 shows that the binding site undergoes little conformational change upon binding of TCC. Structures of three nanobody‐TCC complexes demonstrated there was not a standard binding mode. T4 and T9 have a high degree of sequence identity and bind the hapten in a nearly identical manner, while the more divergent T10 binds TCC in a slightly displaced orientation with the urea moiety rotated approximately 180° along the long axis of the molecule. In addition to methotrexate, this is the second report of haptens binding in a tunnel formed by CDR1, suggesting that compounds with similar hydrophobicity and shape could be recognized by nanobodies in analogous fashion. Structure‐guided mutations failed to improve binding affinity for T4 and T9 underscoring the high degree of natural optimization.

<p>The dehydration of several sugars, including pentoses, hexoses, di, tri, and polysaccharides, in ionic liquids with acidic ion-exchange resins as heterogeneous catalysts was investigated. Good 2‐furaldehydes recovery yields, reaching 92% in some cases, were achieved when Dowex® 50W ion‐exchange resins and 1‐n‐butyl‐3‐ methylimidazoliumchloride ([C4mim]Cl)were used. Our results show that this aproach could represent a promising route towards the cost-efficient production of 2-furaldehydes from carbohydrate-based feedstocks</p>

A summary of the resource.
We detail a strategy for the preparation of bicyclo[4.1.0]heptanes, or norcaranes, based on Diels–Alder reactions and photochemical transformations. As the latter step of the process proceeds with unique regio and stereochemistries, the regio and stereochemical outcome of the overall sequence is dictated by the structures of the diene and dienophile and the conditions used in the initial cycloaddition reaction. This allows for the preparation of norcaranes with different substitution patterns and defined relative stereochemistries, and could be exploited in the design of complex structures bearing this bicyclic moiety.

Abstract
A summary of the resource.
Parasitic helminths use two benzoquinones as electron carriers in the electron transport chain. In normoxia, they use ubiquinone (UQ), but in anaerobic conditions inside the host, they require rhodoquinone (RQ) and greatly increase RQ levels. We previously showed the switch from UQ to RQ synthesis is driven by a change of substrates by the polyprenyltransferase COQ-2 (Del Borrello et al., 2019; Roberts Buceta et al., 2019); however, the mechanism of substrate selection is not known. Here, we show helminths synthesize two coq-2 splice forms, coq-2a and coq-2e, and the coq-2e-specific exon is only found in species that synthesize RQ. We show that in Caenorhabditis elegans COQ-2e is required for efficient RQ synthesis and survival in cyanide. Importantly, parasites switch from COQ-2a to COQ-2e as they transit into anaerobic environments. We conclude helminths switch from UQ to RQ synthesis principally via changes in the alternative splicing of coq-2.

A summary of the resource.
The 5¢-GCGTGGGCGT-3¢ (T4) oligomer found in the zinc ®nger 268±DNA complex was mutated into the sequence 5¢-GCGGGGGCGT-3¢ (G4). A 3D model was constructed from the T4 sequence using an X-ray structure as a template. Molecular dynamics simulations were used to test the thermal stability of the model. A 500-ps trajectory was obtained for the fully charged complex in water using GROMOS87. The complex and the G4 sequence are found to have dynamically stationary behavior. Comparisons made with a previous T4 sequence molecular dynamics simu- lation show both systems have similar thermal stability. The structure of DNA appears to be maintained by its global interactions with the protein although the mutated site does not contribute with its full potential for binding. The protein structure shows some small dierences compared to the T4 simulation. The simulation provided evidence for the role of a chloride ion interacting with the protein and helping in the recognition process.

A three-dimensional structure is engineered for the Trypanosoma congolense trypanothione reductase (TpR) using the sequence homology with glutathione reductase (GR) and lipoamide dehydrogenase, molecular graphics, energy optimization and molecular dynamics techniques. The model was extended to include the complex with the coenzyme nicotinamide adenine dinucleotide phosphate (NADP). The TpR-NADP structure is compared with X-ray data from the glutathione reductase complex with the reduced NADP (NADPH). A model of TpR-NADP including the trypanothione substrate is presented, and an electron-transfer mechanism is proposed.

In the search for new therapeutic tools against tuberculosis two novel iron complexes, [Fe(L–H)3], with 3- aminoquinoxaline-2-carbonitrile N1,N4-dioxide derivatives (L) as ligands, were synthesized, characterized by a combination of techniques, and in vitro evaluated. Results were compared with those previously reported for two analogous iron complexes of other ligands of the same family of quinoxaline derivatives. In addition, the complexes were studied by cyclic voltammetry and EPR spectroscopy. Cyclic voltammograms of the iron compounds showed several cathodic processes which were attributed to the reduction of the metal center (Fe(III)/Fe(II)) and the coordinated ligand. EPR signals were characteristic of magnetically isolated high-spin Fe(III) in a rhombic environment and arise from transitions between mS=±1/2 (geff~9) or mS=±3/2 (geff~4.3) states. Mössbauer experiments showed hyperfine parameters that are typical of high-spin Fe(III) ions in a not too distorted environment. The novel complexes showed in vitro growth inhibitory activity on Mycobacterium tuberculosis H37Rv (ATCC 27294), together with very low unspecific cytotoxicity on eukaryotic cells (cultured murine cell line J774). Both complexes showed higher inhibitory effects on M. tuberculosis than the “second-line” therapeutic drugs.

A bacterial strain (Chol-1ST) that is able to oxidize cholesterol to CO2 and reduce nitrate to dinitrogen was enriched and isolated from an upflow sludge bed (USB) anoxic reactor that treats sanitary landfill leachate from the city of Montevideo, Uruguay. Cells of strain Chol-1ST were Gram-negative, rod-shaped to slightly curved, measured 0?5–0?661?0-1?3 mm and were motile by a single polar flagellum. Strain Chol-1ST grew optimally at 30–32 °C and pH 7?0, with a doubling time of 44–46 h when cholesterol was used as the sole carbon and energy source. The metabolism of strain Chol-1ST was strictly respiratory, with oxygen or nitrate as the terminal electron acceptor. The presence of ubiquinone Q-8 as the sole respiratory lipoquinone indicated that strain Chol-1ST belonged to the b-subclass of the Proteobacteria. Phosphatidylethanolamine was the predominant polar lipid and the G+C content of the DNA was 65?3 mol%. The fatty acid profile of strain Chol-1ST, cultivated under denitrifying conditions by using a defined mineral medium supplemented with cholesterol, was characterized by the following major components: summed feature 4 (C16 : 1v7c and/or iso C15 : 0 2-OH), C16 : 0, C18 : 1v7c and hydroxy acid C10 : 0 3-OH. Minor components included C10 : 0, C11 : 0, C12 : 0, C14 : 0, C15 : 0, C19 : 0, C19 : 0 10-methyl and hydroxylated acids C8 : 0 3-OH and C16 : 0 3-OH. Analysis of the 16S rDNA sequence showed that strain Chol-1ST represents a separate lineage within the Thauera, Azoarcus, Zoogloea and Rhodocyclus assemblage of the b-Proteobacteria. Strain Chol-1ST had highest sequence similarity (96?5%) with strain 72Chol, a denitrifying b-Proteobacterium.On the basis of polyphasic evidence, strain Chol-1ST (=DSM 13999T=ATCC BAA-354T) is proposed as the type strain of Sterolibacterium denitrificans gen. nov., sp. nov

<p>Thione S-methylide, parent species of the thiocarbonyl ylide family, is a 1,3-dipolar species on the [C2SH4] potential energy surface, not so much studied as its isomers, thiirane, vinyl thiol, and thioacetaldehyde. The conrotatory ring-closure reaction toward thiirane was studied in the 90s, but no complete analysis of the potential energy surface is available. In this paper, we report a computational study of the reaction scheme linking all species. We employed several computational methods (density functional theory, CCSD(T) composite schemes, and CASSCF/CASPT2 multireference procedures) to find the best description of thione S-methylide, its isomers, and transition states. The barrier from thiirane to thione Smethylide amounts to 52.2 kcal mol−1 (against 17.6 kcal mol−1 for the direct one), explaining why thiocarbonyl ylides cannot be prepared from thiiranes. Conversion of thiirane to vinyl thiol implies a large barrier, supporting why the reaction has been observed only at high temperatures.<br />Fragmentations of thiirane to S(3P) or S(1D) and ethylene as well as decomposition to hydrogen sulfide plus acetylene were also explored. Triplet and singlet open-shell species were identified as intermediates in the fragmentations, with energies lower than the transition state between thiirane and vinyl thiol, explaining the preference of the latter at low temperatures.</p>

PURPOSE: To study the rhenium-188 labeling of polyamidoamine (PAMAM) generation 4 (G4) dendrimer and its evaluation on biodistribution and chromosomal aberrations in melanoma cells induced by ionizing radiation as potential treatment agent. MATERIALS AND METHODS: Dendrimers were first conjugated with Suc-HYNIC (succinimidyl 6-hydrazinopyridine-3-carboxylic acid hydrochloride). Dendrimer-HYNIC was then incubated with 188ReO4-. Biodistribution was performed administrating 188Re-dendrimer to normal (NM) or melanoma-bearing mice (MBM). Chromosome aberration test was conducted in order to measure treatment capacity of 188Re-dendrimer in melanoma cells. RESULTS: Radiolabeling yield of dendrimer was approx. 70%. Biodistribution studies in NM showed blood clearance with hepatic and renal depuration. MBM showed a similar pattern of biodistribution with tumor uptake of 6% of injected dose. Aberrant metaphases quantified in control cells were 7%, increasing to 29.5% in cells treated with 15μCi (0.555 MBq) of 188Re-dendrimer for 24 h. CONCLUSIONS: 188Re-dendrimer can produce double-stranded breaks in DNA induced by ionizing radiation in melanoma cells in vitro.

Abstract
A summary of the resource.
<p>La importancia de la presencia de gérmenes (bacterias, hongos y levaduras) en los alimentos, es cada día más notoria. Dadas las condiciones climáticas de nuestro país, la ingestión de ensaladas crudas es abundante durante el verano, por lo cual se eligieron lechugas, berros y escarolas como sustrato en la búsqueda de hongos y levaduras. Se trabajó con los productos lavados y sin lavar, habiéndose encontrado que la carga microbiana incial es considerablemente alta disminuyendo totalmente durante el lavado. Los materiales fueron tratados según técnicas standard. Todas las muestras consideradas dieron resultados positivos tanto para hongos filagéneros Trichoderma, Aspergillus, Penicillum, Alternaria, Geotrichum, Rohdotorula, Torulopsis. Si bien todos estos géneros están dentro de la flora normal del suelo nuestro interés se centró en la identificación de especies posibles productoras de microtoxinas.</p>
<p>No debe descartarse el riesgo potencial que puede significar una carga microbiana elevada sobre todo en personas cuyas defensas estén disminuidas.</p>
