El melanoma cutáneo es un tipo de cáncer de piel muy agresivo, altamente metastásico, con elevada mortalidad. La sobrevida se encuentra relacionada a un diagnóstico temprano. El diagnóstico temprano y preciso del melanoma es determinante en la selección del tratamiento óptimo y en la sobrevida del paciente. En este trabajo se evaluó, como agentes de diagnóstico específicos para melanoma, el desempeño in vitro e in vivo de cuatro nuevos péptidos análogos de la hormona alfa melanocito estimulante (α-MSH), ciclados mediante dos estrategias diferentes (enlace lactama y átomo metálico), marcados con 99mTc. Para ello, sobre modificaciones de los péptidos análogos de α-MSH, preparados principalmente a través de síntesis en fase sólida, se estudiaron nuevas estrategias de marcación con 99mTc, utilizando tres ligandos bifuncionales diferentes: uno derivado del sistema coordinante hidracino y otros dos derivados de un sistema tetramino. Los cuatro radiopéptidos se evaluaron fisicoquímica y biológicamente mostrando estabilidad y un buen perfil para la de detección in vivo de células que sobreexpresan el receptor de melanocortina MC1R. Los estudios in vivo de biodistribución y adquisición de imágenes centellográficas y micro-SPECT-CT en ratones portadores de melanoma mostraron concordancia con los perfiles fisicoquímicos y el comportamiento in vitro. Tres de los radiopéptidos presentan una buena captación tumoral, buena eliminación renal y metabolismo hepatobiliar. En comparación con otros radiopéptidos descritos en la bibliografía los desarrollados en este trabajo de Tesis Doctoral comparten características fundamentales como para ser potencialmente usados como agentes de diagnóstico para melanoma. Background: Lactam cyclized alpha-melanocyte stimulating hormone (α-MSH) analogues exhibit high stability and affinity for the MC1-R receptors over expressed in melanoma cells. Recently, we reported a novel 99mTc-HYNIC-cycMSH4-13 analogue with the HYNIC chelator directly attached to the lactam cyclized ring. Objective: In this study we proposed the introduction of a 6-aminohexanoic acid (Ahx) linker between the HYNIC chelator and lactam cyclized peptide cycMSH4-13 to reduce steric hindrance and improve the melanoma targeting and imaging proprieties of the radiolabeled peptide. Method: HYNIC-Ahx-cycMSH4-13 peptide was synthesized on an automated peptide synthesizer and displayed an IC50 of 0.3 nM using B16/F1 cells. The 99mTc/tricine radiolabeled peptide was examined for radiochemical purity, stability and cell binding. In vivo, biodistribution and planar gamma imaging studies were performed in B16/F1 melanoma tumor bearing C57BK mice. Results: 99mTc-HYNIC-Ahx-cycMSH4-13 was obtained with a radiochemical purity >95%, was stable up to 24 h at room temperature and exhibited high binding and rapid internalization in B16/F1 cells. In vivo biodistribution studies showed a tumor uptake of 4.92 ± 0.92 % ID/g and 2.78 ± 1.48 % ID/g at 2 h and 4 h post injection, respectively. Whole-body clearance was rapid through urinary excretion. The melanoma tumors were clearly visualized by planar gamma imaging. Conclusion: 99mTc-HYNIC-Ahx-cycMSH4-13 was shown radiochemically stability and exhibited rapid and selective uptake in melanoma cells and tumors. As a fundamental part of their chemical education, first-year undergraduate students are substantially involved in laboratory activities. Despite the specific teaching staff choices on the main laboratory aims, students normally receive a vast amount of information during these activities. Apart from understanding theoretical content, fundamental skills such as manipulation, data collection and interpretation should be developed. In this context, learners could feel overwhelmed since they can only process a few pieces of information at a time. Indeed, our experience at the Universidad de la República (Uruguayan public university) shows that many first-year students are in fact not able to cope with all the information they receive during laboratory activities. As a result, many of them only follow the experimental protocol automatically, without gaining significant knowledge or developing the necessary skills. In this work, we assessed the use of new online interactive pre-laboratory activities implemented for 252 first-year university students enrolled in a 12-module General Chemistry laboratory course. The student choice of interactive versus more traditional material was evaluated together with observed preferences regarding the different interactive tools offered. Besides, an online pre-laboratory discussion forum was also implemented and assessed. Both the interactive material and the discussion forum were chosen freely by the majority of students (61% and 79%, respectively). Interestingly, the choice was to some extent modulated by student previous performance. Interactive pre-laboratory material was more frequently chosen by low previous performance students, whereas pre-laboratory forum was preferentially used by high previous performance students. Finally, the influence of these new materials on student laboratory performance was statistically analyzed. Other personal and academic variables were also taken into account. Interactive material access was positively correlated with the final laboratory marks for medium previous performance learners. On the other hand, for lower previous performance students, the academic discussion between teachers and partners promoted by the online forum was positively correlated with their academic performance. Preliminary studies in mice bearing induced breast tumors rendered promising uptake values. Assessment of the presence of estrogen receptors in breast cancer is crucial for treatment planning. With the objective to develop a potential agent for estrogen receptors imaging, we present the development and characterization of a 99mTc‐tricarbonyl–labelled estradiol derivative. Using ethinylestradiol as starting material, an estradiol derivative bearing a 1,4‐disubstituted 1,2,3‐triazole–containing tridentate ligand system was synthesized by “Click Chemistry” and fully characterized. Labelling with high yield and radiochemical purity was achieved through the formation of a 99mTc‐tricarbonyl complex. The radiolabelled compound was stable, exhibited moderate binding to plasma protein (approximately 33%) and lipophilicity in the adequate range (logP 1.3 ± 0.1 at pH 7.4). Studies in MCF7 showed promising uptake values (approximately 2%). However, more than 50% of the activity is quickly released from the cell. Biodistribution experiments in normal rats confirmed the expected “in vivo” stability of the radiotracer but showed very high gastrointestinal and liver activity, which is inconvenient for in vivo applications. Taking into consideration the well‐documented influence of the chelating system in the physicochemical and biological behaviour of technetium‐labelled small biomolecules, research will be continued using the same pharmacophore but different complexation modalities of technetium. This research focuses on the identification and quantification of lithium-ion battery degradation indicators at low temperatures. We studied the cycling aging of 18,650 commercial NMC lithium-ion batteries at 10 ◦C. For this purpose, we carried out life cycle tests and performed Galvanostatic Intermittent Titration Technique (GITT) tests in the voltage range for the charge and discharge processes for different States of Health (SoH). The diffusion time constant and the ohmic overpotential were determined from GITT for different SoH. As the batteries degrade both parameters increase. A degradation mechanism associated with faradaic effects (increase in ohmic drop overpotential) and a thermodynamic effect associated with changes in the active material (diffusional time constant) is observed. Furthermore, we performed Electrochemical Voltage Spectroscopy studies through Incremental Capacity (IC) curves. IC curves peaks and valleys are associated with battery phase transformations due to aging phenomena. Each peak has a unique peak height, area, and position associated with a degradation mode. This research focuses on the IC curves derived from the discharge capacities at 3A and OCP (Open Circuit Potential) to study the thermodynamic and faradaic effects separately. The peak at 3.6 V and 3.4 V for the 3A and OCP studies, respectively, is the main feature for the detection of the degradation mechanism. The valley in the OCP curves intensifies with the decrease in SoH and is shifted to lower potentials, an effect that is not observed in the 3A curves. So, we could associate this with thermodynamic effects, Loss of Lithium Inventory (LLI) and Loss of Active Material (LAM) of both the positive electrode and the negative electrode. El hidrógeno es un vector energético limpio y desde el punto de vista energético, el hidrógeno es, de todos los combustibles, el que tiene la máxima relación energía/peso. Una manera eficiente de almacenarlo que cumple con muchos de estos requisitos es hacerlo en fase sólida en aleaciones formadoras de hidruros. Estas aleaciones tienen además la ventaja de poder almacenar hidrógeno tanto por la vía gaseosa como electroquímica. La vía electroquímica brinda la posibilidad de utilizar estos sistemas como electrodos de baterías alcalinas, substituyendo así a las baterías de níquel-cadmio. Molybdenum substitution by chromium in ZrCr1-xNiMox alloys produces the segregation of ZrxNiy phases, ZrCr2 and ZrMo2 Laves phases as it was stated in Part I of this paper. In order to study the contribution of these phases in ZrCr1-xNiMox alloys performance, secondary Zr9Ni11 and Zr7Ni10, together with ZrCr2 and ZrMo2 Laves phases were synthesized and their crystalline structures determined with XRD patterns. Zr7Ni10 exhibits an orthorhombic structure, whereas Zr9Ni11 depicts a tetragonal structure. ZrCr2 presents a hexagonal structure (C14), while ZrMo2 depicts a cubic structure (C15). ZrCr2 shows zero discharge capacities, while ZrMo2 exhibits a first capacity value of 245 mAh g−1. Zr9Ni11 and Zr7Ni10 phases exhibit maximum discharge capacities of 119 and 157 mAh g−1, respectively. Theoretical results show that the hydride stability for ZrCr2 is larger than that for ZrMo2. However, the great hydrogen storage capacity yields a slower desorption kinetics since it requires a considerable de-hydrogenation energy. The opposite effect is observed for the proposed hexagonal structure ZrMo2. The 100% charge of the alloys shows different results for the hydrogen evolution reaction. Zr7Ni10 phase depicts a better performance since the required potentials are lower than −1.05 V. There is a change in the rate determining step (rds) from a chemical recombination of surface hydrogen (Tafel pathway) in Zr7Ni10 to a fast first electron transfer (Volmer step) for Zr9Ni11. Thus, the rds is strongly affected by the type of secondary phase dominant in each sample, being Zr7Ni10 the most favorable. Hydrogen evolution reaction analysis showed that electrodes with 13% of molybdenum described the largest current intensities. The reaction rds was related to the type of secondary phase dominant in each sample, whereas self-discharge behavior was associated to the presence of molybdenum phases. Thereby, the alloy with 13% weight in molybdenum was the one which improves electrocatalytic performance for hydrogen storage purposes. Epilepsy, which is one of the most common neurological disorders, involves the occurrence of spontaneous and recurrent seizures that alter the performance of the brain and affect several sensory and behavioral functions. Oxidative damage has been associated with post-seizure neuronal injury, thereby increasing an individual’s susceptibility to the occurrence of neurodegenerative disorders. The present study investigated the possible anticonvulsive and neuroprotective effects of organic and conventional yerba mate (Ilex paraguariensis), a plant rich in polyphenols, on pentylenetetrazol (PTZ)-induced seizures in Wistar rats. The behavioral and polyphenolic profiles of the yerba mate samples were also evaluated. Infusions of yerba mate (50 mg/kg) or distilled water were given to rats for fifteen days by oral gavage. On the 15th day the animals were subjected to open field test, and exploratory behavior was assessed. Subsequently, 60 mg/kg PTZ (i.p.) was administered, and animals were observed for the appearance of convulsions for 30 min. Latency for the first seizure, tonic–clonic and generalized seizures time, frequency of seizures and mortality induced by PTZ were recorded. The animals were then sacrificed, and the cerebellum, cerebral cortex and hippocampus were quickly removed and frozen to study the neuroprotective effects of yerba mate. The oxidative damage in lipids and proteins, nitric oxide levels, the activities of the antioxidant enzymes superoxide dismutase (Sod) and catalase (Cat) and non-enzymatic cellular defense (sulfhydryl protein) were quantified in all the tissues. The results showed that organic and conventional yerba mate infusions were able to reduce the frequency of seizures when compared to the PTZ group. Besides, organic yerba mate infusion decreases the tonic–clonic seizures time in relation to the PTZ group. It was also shown that organic and conventional yerba mate infusions reduced the oxidative damage in lipids and proteins and nitric oxide levels and prevented the decrease in Sod and Cat activities and sulfhydryl protein content when compared to the PTZ group in all the CNS tissues assayed. Organic and conventional yerba mate commercial samples did not change the behavior (locomotion, exploration or anxiety) of the treated animals. In both organic and conventional infusions, the presence of the polyphenols rutin, chlorogenic acid and their acyl derivatives were detected, which could be associated with the biological effects observed. These data indicate that yerba mate may provide new perspectives for the development of therapeutic approaches with natural compounds in the pharmaceutical area, both to reduce the convulsions’ frequency and to minimize the neuronal damage associated with recurrent seizures. In the present work the feasibility of Tannat grape skin (TGS) as a functional ingredient in the formulation of two snacks (yogurt and biscuits) was studied. The research provided novel information on the effects of the food matrix and digestion process, under simulated human oral gastrointestinal conditions, in the bioaccessibility of TGS bioactive compounds composing of the snacks with health promoting properties (antioxidant, anti-inflammatory, and antidiabetic). TGS polyphenolic profile was analyzed by ultra-high performance liquid chromatography tandem mass spectrometry (UHPLC-MS/MS) finding mainly flavonoids, phenolic acids, and anthocyanins, which may exert antioxidant, anti-inflammatory, and carbohydrase inhibition capacities. TGS digest showed antioxidant and antidiabetic potential compared to the undigested sample (p < 0.05). Yogurt and biscuits with TGS were developed with the nutrition claims “no-added sugars” and “source of fiber” and were digested in vitro to evaluate the bioaccessibility of compounds with health promoting properties after food processing and digestion. After in vitro simulation of digestion, bioactive properties were enhanced for control and TGS snacks which may be attributed to the formation/release of compounds with health-promoting properties. Biscuits showed significant increase in ABTS antioxidant capacity and yogurt showed increased �-glucosidase inhibition capacity by the addition of TGS (p < 0.05). Polyphenols from TGS and bioactive peptides from snacks which may be released during digestion might be responsible for the observed bioactivities. Consumer’s acceptance of TGS yogurt and biscuits showed scores of 6.3 and 5.1 (scale 1–9), respectively, showing TGS yogurt had higher overall acceptance. Sensory profile assessed by check-all-that-apply + just-about-right (CATA+JAR) showed most of the attributes were evaluated as “just about right”, supporting good food quality. The developed yogurt presented adequate shelf-life parameters for 28 days. TGS yogurt with higher acceptability showed reduced ROS formation (p < 0.05) induced by tert-butyl hydroperoxide (1 mM) in CCD-18Co colon cells and RAW264.7 macrophages when pre-treated with concentrations 500–1000 and 100–500 �g/mL of the digests, respectively. Moreover, TGS yogurt digest pre-treatment reduced nitric oxide (NO) production (p < 0.05) in lipopolysaccharide (LPS)-induced RAW264.7 macrophages, showing anti-inflammatory potential. Bioactive peptides generated during lactic fermentation and digestion process may be contributors to intracellular effects. Inhibition of both effects was observed in the presence of distamycin suggesting the involvement of the DNA minor groove in the interaction with the nitrofurylsemicarbazone ruthenium complexes. In addition cleavage inhibition by dimethyl sulfoxide suggests an oxidative mechanism of action. The rate coefficients were determined using the relative method in a 480 L multipass glass reactor coupled to an FT-IR detection system. The rate coefficients found for oxolan 3-one and 2MTHF-one with • OH radicals (k1 and k2) and with • Cl atoms (k3 and k4) at 298 K and atmospheric pressure (in cm3 molecule− 1 s − 1 ) were: k1 = (1.86 ± 0.29) × 10− 11, k2 = (2.64 ± 0.47) × 10− 11, k3= (1.15 ± 0.28) × 10 − 10, and k4 = (1.33 ± 0.32) × 10− 10, respectively. Reactivity trends were developed by comparison with other similar structures and Fukui indices employed to determine the reactivity of different sites on the ring. The singularity of the reaction with • OH was assessed by computational studies which showed the formation of several stable hydrogen bonded complexes, explaining the difference with the reaction with the • Cl atom. SAR estimations of the rate coefficients were calculated and compared to the experimental values. We compared resistance to decay, mold fungi, termites and insect larvae of particleboards incorporated with the raw boron minerals ulexite and colemanite to that of particleboards impregnated with the commercial boron preservative zinc borate, or boric acid plus a borax mixture. We also quantified water absorption, thickness swell and boron release of particleboards. Ulexite had the best decay resistance, and colemanite had the best termite resistance. However, ulexite and colemanite were not as effective as zinc borate or the boric acid/borax mixture in preventing mold growth. In general, the boric acid/borax mixture combination was more effective against Anobium larvae than the other treatments. Less boron was released by specimens containing zinc borate and colemanite than by those containing ulexite or the boric acid/borax mixture. In general, water absorption and thickness swell were similar among the different treatments, but both were slightly higher in the ulexite-incorporated specimens. Further mechanical tests will be needed to evaluate the particleboard properties and thereby the compatibility of these boron minerals with various manufacturing processes.
