An efficient and facile general method for the synthesis of conduritol C analogs, taking advantage of an enantioselective biocatalysis process of monosubstituted benzenes, is described. The absolute stereochemical patterns of the target molecules (−)-conduritol C, (−)-bromo-conduritol C, and (−)-methyl-conduritol C were achieved by means of chemoenzymatic methods. The stereochemistry present at the homochiral cyclohexadiene-cis-1,2-diols derived from the arene biotransformation and the enantioselective ring opening of a non-isolated vinylepoxide derivative permitted the absolute configuration of the carbon bearing the hydroxyl groups at the target molecules to be established. All three conduritols and two intermediates were crystallized, and their structures were confirmed by X-ray diffraction. The three conduritols and intermediates were isostructural. The versatility of our methodology is noteworthy to expand the preparation of conduritol C analogs starting from toluene dioxygenase (TDO) monosubstituted arene substrates. The Scientific Advisory Board (SAB) of the Organisation for the Prohibition of Chemical Weapons (OPCW) has provided advice in relation to the Chemical Weapons Convention on assistance and protection. We present the SAB’s response to a request from the OPCW Director-General in 2014 for information on the best practices for preventing and treating the health effects from acute, prolonged, and repeated organophosphorus nerve agent (NA) exposure. The report summarises pre- and post-exposure treatments, and developments in decontaminants and adsorbing materials, that at the time of the advice, were available for NAs. The updated information provided could assist medics and emergency responders unfamiliar with treatment and decontamination options related to exposure to NAs. Trypanosoma cruzi, the causative agent of Chagas’ disease, affects tens of millions of South Americans. One of the most studied T. cruzi-biomolecules as a target for drug development is cruzipain, an essential cysteine proteinase of this parasite. Some of our recent studies identified amidine containing benzofuroxans as hit compounds for cruzipain inhibition with trypanosomicidal activities. Experimental and theoretical studies inspired us to modify these compounds by maintaining the amidine motif and using benzofuroxan and benzimidazole 1,3-dioxide systems as core scaffolds in order to obtain better cruzipain inhibitors. The new amidines had excellent trypanosomicidal activity, with good selectivity indexes, but without improved cruzipain-inhibitory activities compared with the parent compounds. The interaction of amidines with cruzipain has been investigated through a combined NMR -T1-differences, DOSY, and STD- and molecular docking approaches. Despite the low cruzipain-inhibition ability, our data suggest that these designed compounds have relevant structural features, i.e. aromatic groups and protonated moieties with stabilizing complex ability using stacking and electrostatic interactions, respectively, that bind reversibly to cruzipain. Whey proteins such as α-lactalbumin can be biologically active when the bioactive peptides encrypted in the amino acid sequence of the native protein are released by enzymatic hydrolysis. Peptides contained in the gastrointestinal digest of α-lactalbumin hydrolysate were separated by preparative RP-HPLC (55 fractions), and three peptides were identified by LC-MS/MS analysis from selected fractions: IWCKDDQNPH (MW: 1254.54 Da) f(59-68), KFLDDDLTDDIM (MW: 1439.64 Da) f(79-90), DKFLDDDLTDDIM (MW: 1554.67 Da) f(78-90). Among the chemically synthesized peptides, IWCKDDQNPH showed the highest antioxidant capacity determined byORAC,ABTS, andHORAC assays (IC50 0.015± 0.002, 0.45 ± 0.02, and 1.30 ± 0.05 mg/ml, respectively) and this activitymay be related to the amino acid sequence. Molybdenum is an essential trace element of biological and environmental relevance. Monitoring its content in water sources is very important from the standpoint of health and environment. Given the very low concentrations of Mo in water, it is necessary to use highly sensitive techniques such as ICP-MS or elaborate methods including preconcentration prior to detection. In this work, a new method for the determination of molybdenum in groundwater was developed, incorporating a preconcentration step based on dispersive liquid–liquid microextraction (DLLME) prior to detection by electrothermal atomic absorption spectrometry (ETAAS). After the injection of a ternary mixture formed by an aqueous solution of the complexing agent (potassium ethyl xanthate) and extraction and dispersion solvents (carbon tetrachloride, acetonitrile), the formation of the Mo–xanthate complex occurs followed by its extraction in fine droplets of carbon tetrachloride. After separating the phases the molybdenum content is measured by ETAAS. Several experimental parameters were optimised and the method was validated, presenting a detection limit (3s/b) of 0.017 μg L−1, a quantification limit (10s/b) of 0.062 μg L−1, and an enrichment factor of 165 for a sample volume of 6.0 mL. The calibration curve was linear in the range of 0.06–1.1 μg L−1, the precision (repeatability, sr (%), n = 10) was 2.98% at 0.1 μg L−1 and the recovery of spiked samples was 98.5–103%. This method was successfully applied to the determination of molybdenum in groundwater and the results were found to be equivalent to those obtained by ICP-MS. La concentración y las formas químicas de los elementos en el ambiente están en continuo cambio y las aguas naturales son el principal agente de movilización y transporte. Los cambios generados por las actividades humanas son más bruscos que los provocados por los ciclos naturales y en el caso de los elementos traza, su baja abundancia los hace especialmente sensibles a estas variaciones. En las aguas naturales del Uruguay en el correr de los años se ha dado un aumento de la concentración de elementos traza debido a factores antropogénicos. Para realizar un análisis completo del comportamiento de los elementos, a los efectos de su gestión, debe sumarse al monitoreo de las concentraciones totales, el conocimiento de las especies químicas predominantes del o los elementos de interés en las condiciones fisicoquímicas de la matriz en estudio. Por ello, no alcanza con conocer los valores de las concentraciones totales de estos elementos en cada muestra, sino también qué especies forman y con qué elementos o compuestos interaccionan para determinar su movilidad, en la medida que ambos factores son decisivos para determinar su biodisponibilidad y para detectar en forma temprana situaciones de desbalance, y aplicar técnicas de gestión apropiadas. Entre los elementos traza se seleccionó al Mo como elemento de estudio para esta tesis. Es de relevancia biológica y ambiental y su movilidad en la corteza terrestre depende en gran parte del ciclo hidrológico. Forma parte de diversos procesos bioquímicos, pero también es potencialmente tóxico. En las aguas superficiales y subterráneas poco profundas forma predominantemente aniones que podrían interaccionar con los cationes más abundantes. Estas reacciones de Mo han sido poco estudiadas en las condiciones de las aguas naturales. Este elemento se encuentra en niveles muy bajos de concentración, por lo que se requiere para su determinación analítica de una técnica muy sensible como ICP-MS. Al no estar fácilmente disponible en nuestro medio, el presente trabajo representó un desafío analítico importante que involucró el desarrollo de una metodología con preconcentración para la determinación de la concentración de Mo en muestras de aguas naturales por espectrometría de absorción atómica electrotérmica (ETAAS). Luego de evaluar las diferentes técnicas de preconcentración se optó por la microextracción dispersiva líquido líquido (DLLME). Se desarrolló y validó una metodología de determinación de Mo en agua con preconcentración por DLLME utilizando etilxantato (EX) como ligando para Mo, tetracloruro de carbono como solvente de extracción, acetonitrilo como solvente dispersivo y detección mediante ETAAS. Esta metodología demostró ser muy sensible, rápida, sencilla y amigable con el ambiente al usar volúmenes muy reducidos de solventes. Fue aplicada con éxito en el análisis de diferentes muestras de aguas naturales de diferentes regiones del país, permitiendo Resumen 20 realizar un relevamiento del contenido de este elemento así como también establecer las regiones en las que es posible encontrarlo. Para realizar el análisis de especiación del elemento de interés en las condiciones de las aguas naturales se realizaron las medidas de todas las constantes de equilibrio que determinan la formación de las especies en solución: equilibrios de protonación de los aniones, equilibrios de asociación con cationes y equilibrios de hidrólisis de los cationes si corresponde. Las medidas de las constantes fueron realizadas por medio de titulaciones potenciométricas en condiciones experimentales simuladas de las aguas naturales (20 °C y baja fuerza iónica). Asimismo se realizó el estudio de la variación de las constantes de protonación con la concentración de fuerza iónica según la teoría de interacción específica (SIT), mostrando que existe dependencia de los valores de las constantes de equilibrio con la concentración de la fuerza iónica del medio. Fueron identificados los sistemas y las condiciones en las que se detecta la formación de especies poco solubles (Cu y Cd principalmente) y se realizaron estudios de solubilidad de los molibdatos. Se sintetizaron y caracterizaron dichos sólidos, se determinaron los valores de las constantes de producto de solubilidad (Ks) y se demostró la validez del modelo químico establecido para los sistemas Mo(VI)-Cd y Mo(VI)-Cu que permite predecir la formación de fases sólidas. También se realizó un estudio comparativo de la química en solución acuosa de Mo con otros elementos formadores de aniones como W, As, Se y S. Los resultados de las muestras analizadas por la metodología desarrollada y los datos termodinámicos obtenidos fueron utilizados para evaluar la distribución de especies de Mo en los diferentes sistemas acuáticos de nuestro país. Se realizaron medidas de dureza de todas las muestras con el objetivo de caracterizarlas desde el punto de vista de la fuerza iónica y para evaluar como influye este parámetro en la especiación de Mo. A modo de reflexión final se desea destacar la importancia y el valor del trabajo interdisciplinario, que permitió integrar conocimientos de química analítica y de química en solución acuosa, para realizar el relevamiento del contenido de Mo en aguas de nuestro país así como también un estudio completo de la química de este elemento en las condiciones de las aguas naturales. An alkaline sample treatment using tetramethylammonium hydroxide was optimized and validated for the determination of Cu, Mo, and Zn in beef samples from Uruguay, aiming to contribute to the food quality evaluation of this product. The optimization of the developed method was performed by means ofmultivariate experiments. Copper and Zn were determined by flame atomic absorption spectrometry and Mo was determined by electrothermal atomic absorption spectrometry. The proposed method consisted of adding 5.0 mL of ultrapure water to 0.25 g of the sample, followed by 1.5 mL of tetramethylammonium hydroxide 25% w w−1 and 60 min at 85 °C in a water bath. After centrifuging, the obtained solution can be used for the analytical determinations of the three elements. Certified reference materials were analyzed and a comparison with microwave-assisted acid digestion was performed to guarantee reliability. At the 95% significance level, the obtained results were statistically equivalent to the certified values and to those obtained by total acid digestion for all the studied elements. Analytical precision expressed as relative standard deviation was less than 8% in all cases. Copper, Mo, and Zn were determined in Uruguayan beef samples using the proposed alkaline method. Moringa stenopetala is a tropical tree from the Moringaceae family, native to north-east Africa, that has widespread to many countries. It is highly valued for the associated nutritional and medicinal properties. The nutritional value of South American Moringa stenopetala was assessed in this work, giving some deeper insight into the expected biological accessibility of the main nutrients present in raw leaves and prepared infusions. Metal ions were determined by FAAS; L-ascorbic acid, leucine, and tryptophan by LC-DAD; rutin, isoquercitrin and neochlorogenic acid by LC-ESI-MS/MS. Metal ions content in analyzed specimens was in the same order of previously reported data, except for lower values for iron, probably associated to the different soil composition. Tested amino acids were also found in the previously reported range, while higher contents of L-ascorbic acid, rutin and neochlorogenic acid were determined. Interestingly, transfer factors to infusions resulted especially low for magnesium, calcium, iron, and L-ascorbic acid, indicating that the way of consumption exerts a strong influence on the nutritional value of the vegetal material. Chemical speciation modeling experiments to predict bioaccessibility showed that sodium, potassium, magnesium, and calcium ions to be predominantly free. On the other hand, the micronutrients are predicted to be partly associated to polyphenolic compounds. Finally, the antioxidant activity was studied both in the vegetal material and the prepared infusions by the ORAC assay, both showing relevant antioxidant activity probably associated to the high rutin content. This biomolecule, together with other polyphenolic compounds present, are expected to partially retain metal ions in solution, also contributing to the antioxidant beneficial properties of Moringa stenopetala. Theoretical ab initio calculations are done on different mechanisms for the conversion of vinyl alcohol to acetaldehyde, both in gas phase and in solution. Several basis sets are used in order to assess the accuracy of the results in gas phase and a continuum model of the solvent is employed to mimic reactions in water solution. The results indicate a catalytic action of water in hydrated clusters in gas phase, whereas in solution, and within the error limits of our calculations, both neutral water-chain and ionic mechanisms appear to be equally probable. Finally, the action of acids or bases is tested through the analysis of the reaction of vinyl alcohol with H3O+ and HO−. The results of the calculations are shown to be in qualitative agreement with the experimental facts when 6-31++G basis set is used but not when either STO-3G or 4-31G basis sets are employed. Chagas disease, caused by the protozoan parasite Trypanosoma cruzi, is a major health problem worldwide. In this work, we report the development of palladium and platinum metal complexes with 5-nitrofuryl-containing thiosemicarbazones (L) as bioactive ligands against T. cruzi and PTA (1,3,5-triaza-7-phosphaadamantane) as co-ligand. Eight new complexes of the formula [MCl(L)(PTA)] with M = Pd or Pt were synthesized and fully characterized. Most complexes showed similar activities against T. cruzi to those of the corresponding free thiosemicarbazone ligands. No significant differences between palladium and platinum complexes were observed. Metal compounds with the phenylthiosemicarbazone derivative were the most active ones (IC50 = 9.84 ± 0.32 and 4.94 ± 0.24 μM for Pd2+ and Pt2+, respectively). The prepared complexes were not toxic on mammalian cells, showing selective indexes of more than 10–20. The ability of the complexes to be reduced in the parasite, which leads to toxic free radical species, was confirmed by the detection of OH· and nitroanion free radical species by ESR spectroscopy experiments. Gel electrophoresis and fluorescence experiments were consistent with an intercalating-like mode of DNA interaction for the complexes, but DNA interaction does not seem to be the main mechanism of anti T. cruzi action for these compounds. The results obtained show that complexation of the bioactive ligands with the selected metals is a valid strategy to obtain improved metal-based antiparasitic compounds.
