Polychlorinated dibenzo-p-dioxins (PCDD) and dibenzofurans (PCDF) are persistent, toxic, and bioaccumulate in the environment. Due to their high analytical costs, these compounds are hardly regulated and mostly not monitored in the Third World. To overcome this, bioassays have been proposed as low-cost alternative methods. Two of the most established bioanalytical tools, the dioxin antibody-based enzyme-linked immunosorbent assay ELISA and the micro-EROD bioassay are evaluated and compared to high resolution gas chromatography and high resolution mass spectrometry (HRGC/HRMS) analytical methodology. The methods were tested using thirteen soils and sediment samples selected from diverse sites in Montevideo, Uruguay. The WHO₂₀₀₅ total toxic equivalent (WHO₂₀₀₅-TEQ) of soils ranged from 2.4 to 2212 (ng WHO₂₀₀₅-TEQ/kg dry sample) and from 0.14 to 9.4 (ng WHO₂₀₀₅-TEQ/kg dry sample) in sediments. This study shows significant contamination related to dioxin-like compounds, particularly in sites where uncontrolled burnings were carried out. ELISA and micro-EROD bioassay correlated well with HRGC/HRMS, R Spearman 0.773 and 0.913, respectively and were highly correlated to each other, R Spearman 0.879. Preliminary threshold values of bioassay toxic equivalents of 330 (ng/kg dry sample) for the micro-EROD bioassay and 220 (ng/kg dry sample) for ELISA are proposed.

A summary of the resource.
We evaluated the anti-insectan activity of extracts from different vegetative parts of ten plant species native to Uruguay. The selected plants belong to five families: Bignoniaceae: Clytostoma callistegioides, Dolichandra cynanchoides, Macfadyena unguis-cati; Sapindaceae: Dodonaea viscosa, Allophylus edulis, Serjania meridionalis; Lamiaceae: Salvia procurrens, Salvia guaranitica; Solanaceae: Lycium cestroides; and Phytolaccaceae: Phytolacca dioica. The extracts were evaluated in independent bioassays against four insect pests and one beneficial insect. Aphid settling inhibition was evaluated with a grass specialist, Rhopalosiphum padi, and a feeding generalist, Myzus persicae (both Hemiptera: Aphididae). Antifeedant activity was tested with adults of the specialist Epilachna paenulata (Coleoptera: Coccinellidae) and larvae of the generalist Spodoptera littoralis (Lepidoptera: Noctuidae). Finally, contact toxicity was assessed with honey bees, Apis mellifera (Hymenoptera: Apidae). Strong settling inhibition (SI) activity (expressed as %SI, where 100% means complete inhibition by the extract) was found only for the twig extracts of A. edulis (Sapindaceae) against M. persicae (%SI = 77±4). Antifeedant activity (expressed as % of feeding reduction (FR), where 100% means no consumption on extract-treated diet) against E. paenulata was significant for the leaf extracts of L. cestroides (Solanaceae) (%FR = 100±0) as well as of all Bignoniaceae and Sapindaceae species. No extracts were active against S. littoralis larvae, and most of them were innocuous to honey bees, with the exception of L. cestroides and S. meridionalis leaf extracts

Some results obtained during our research work in the search of anti-snake compounds from plant origin, allow us to propose sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) as a valuable method for a fast and reliable screening in order to evaluate plant extracts activity on snake proteins from Bothrops diporus (yarará chica). Such approach will allow to process a larger number of plant extracts and to select the active ones. Venoms used in this study came from B. diporus which was previously vacuum dried. Extracts (aqueous, alcoholic and hexanic) were from native plants: Aristolochia elegans, Aristolochia gibertii, Asclepia curassavica, Cissampelos pareira, Dorstenia brasiliensis, Eclipta prostrata, Iresine diffusa, Mikania micrantha, M. periplocifolia, M. coridifolia, Nectandra angustifolia, N. megapotamica, Sapium haematospermum and Trixis divaricata. The results obtained by SDS-PAGE were compared with those obtained from in vitro assays (coagulation and hemolysis inhibition). The correlation between results obtained from electroforetic and in vitro assays allowed to suggest SDS-PAGE as a suitable technique to assist in preliminary plant screenings for anti-snake activity by snake venom protein interaction with plant compounds.

An increasing share of fine wines is marketed through retail stores. In this context, packaging plays a major role in attracting consumers’ attention and influencing their purchase decisions. The aim of the present work was to identify a specific bottle for Tannat red wine, the typical Vitis vinifera grape of Uruguay, by studying the effect of bottle shape and colour on consumers’ sensory expectations and willingness to purchase. A consumer study was carried out with 64 fine wine consumers, who were first introduced a list of bottle colours and shapes and were asked to answer a check-all-that-apply question. Then, participants were asked to complete a conjoint analysis task about Tannat wine bottles. Results from the check-all-that-apply question showed that consumers associated the shape and colour of bottles with specific types and styles of wine. Conjoint analysis suggested that Bordeaux dark green or dark gray bottles might be the most appropriate for Tannat wine. Interestingly, results shown here may indicate the existence of risks if deep innovation is made in bottle shape designs, since consumers clearly preferred Bordeaux bottles for red wines.

In the search for gallium bioactive compounds five Ga(III) complexes, [GaIII(L–H)2](NO3), with tridentate salicylaldehyde semicarbazone derivatives as ligands (L) have been synthesized and characterized in the solid state and in solution by different techniques. The crystal structure of [GaIII(L4–H)2](NO3)�2H2O, where L4 is 3-ethoxysalicylaldehyde semicarbazone, was solved by X-ray diffraction methods. The gallium( III) ion is in a distorted octahedral environment, coordinated to two nearly planar and mutually perpendicular 3-ethoxysalicylaldehyde semicarbazonato anions acting as tridentate ligands through their phenol and carbonyl oxygen atoms and their azomethine nitrogen atom. Their biological potential has been explored by evaluating their activity on Mycobacterium tuberculosis, causative agent of tuberculosis, and their cytotoxicity on tumor cell lines. Three different human tumor cell lines were selected that show different degrees of resistance to metallodrugs: ovarian A2780 (low resistance), breast MCF7 (medium resistance) and prostate PC3 (high resistance) cells. Although the complexes have not shown activity on M. tuberculosis, complexation with gallium has led to the enhancement of the cytotoxic potencies of the organic compounds. Those complexes that contain a bromide substituent at the phenolate ring have shown the highest cytotoxicities. In particular, [GaIII(L2–H)2](NO3), where L2 is 5-bromosalicylaldehyde semicarbazone,�has shown a remarkable cytotoxicity on A2780 tumor cell line with an IC50 value of the same order than cisplatin (IC50 Ga–L2 = 2.4 ± 0.3 lM; IC50 cisplatin = 2.0 ± 0.1 lM, 72 h incubation at 37 �C). Interestingly, this complex has also shown moderate cytotoxicity against MCF7 and PC3 cells (IC50 MCF7 = 30 ± 6; IC50 PC3 = 18 ± 3 lM). Therefore, this gallium compound could be considered a promising wide spectrum potential anti-tumor agent.

Searching for new promising metal-based drugs for the treatment of parasitic diseases against Trypanosoma cruzi, three related oxidovanadium(IV) complexes, [VIVO(SO4)(H2O)2(NN)], with the phenanthroline derivatives (NN) 1,2,5thiadiazolo[3,4-f]1,10phenanthroline (tdzp), 1,10-phenanthroline-5, 6-dione (phendione), and 5,6-epoxy-5,6-dihydro-1,10-phenanthroline (epoxyphen) are synthesized, characterized, and evaluated in vitro as anti-T. cruzi agents. The compounds are characterized in the solid state and in solution by elemental analysis, electrospray ionization mass spectrometry (ESI-MS), conductimetric measurements, and infrared (FTIR), UV/Vis, and electronic paramagnetic resonance (EPR) spectroscopy. EPR spectroscopy suggests that the ligands act as bidentate, binding through both nitrogen donor atoms in an axial-equatorial mode. DFT calculations corroborate the structural assignments. The stability of the complexes in solution is evaluated by EPR and 51V- NMR spectroscopy and all complexes show reasonable stability. The anti-T. cruzi activity of the complexes was tested by measuring the growth inhibitory effect on the epimastigote life cycle form of the parasite (Dm28c strain). All complexes show IC50 values in the micromolar range against T. cruzi and display activities of the same order of that of Nifurtimox, but lower than that of the previously reported analogue [VIVO(SO4)(H2O)2(dppz)] (dppz = dipyrido[3, 2-a:2′,3′-c]phenazine). Furthermore, DNA was evaluated as a potential target by using atomic force microscopy (AFM), showing that the complexes display ability to interact with this biomolecule.

Rice is an important source of methane (CH4) and other crops may be sources of nitrous oxide (N2O), both of which are powerful greenhouse gases. In Uruguay, irrigated rice rotates with perennial pastures and allows high productivity and low environmental impact. A long-term experiment with contrasting rice rotation intensification alternatives, including rice–soybean and continuous rice, was recently carried out in an Argialboll located in a temperate region of South America. To know if rotation systems influence soil microbial activity involved in CH4 and N2O emissions, the abundance and potential rate for gas production or consumption of microbial populations were measured during the rice crop season. CH4 was only emitted when rice was flooded and N2O emission was not detected. All rotational soils showed the highest rate for methanogenesis at tillering (30 days after rice emergence), while for methanotrophy, the maximum rate was reached at flowering. The abundance of related genes also followed a seasonal pattern with highest densities of mcrA genes being observed at rice flowering whereas pmoA genes were more abundant in dry soils after rice harvest, regardless of the rotation system. Differences were foundmainly at tillering when soils with two consecutive summers under rice showed higher amounts ofmcrA and pmoA gene copies. The potential denitrification rate was highest at the tillering stage, but the abundance of nirK and nirS genes was highest in winter. Regarding ammoniumoxidation, bacterial amoA abundance was higher in winter while the archaeal amoA gene was similar throughout the year. A strong influence of the rice growth stage was registered for most of the parameters measured in rice paddy soils in this no-till rice intensification experiment. However, differences among rotations begin to be observed mainly at tillering when the abundance of populations of the methane and nitrous oxide cycles seemed to respond to the rice intensification.

The lipid content, lipid class composition, and fatty acid composition of hake (Merluccius hubbsi) liver oil was studied for a whole year. The lipid content of the liver ranged from 41 to 60%. Triacylglycerols were the main lipid class present in the oil, with a mean value of 80%. Docosahexaenoic acid was the main polyunsaturated fatty acid during the year and was twice that of the eicosapentaenoic acid content. The composition of hake liver oil makes it useful as a raw material for encapsulated pharmaceutical products.

Essential oils from the leaves of Thymus vulgaris L. collected from Caxias do Sul (Rio Grande do Sul State, Brazil) were isolated by hydrodistillation with an average oil yield of 0.25%. Twenty-two components were identified by GC and GC/MS analysis. Thymol was found to be the most abundant constituent (31.5–52.4%), followed by p-cymene (17.1–34.4%). Thyme possessed a higher oil yield and the oil was richer in oxygenated compounds when harvested in the spring.

Exploring the influence of different substitution patterns of 2H-benzimidazole 1,3-dioxide derivatives (BzNO) we prepared fifteen new derivatives. Initially the BzNO were tested against Trypanosoma cruzi Tulahuen 2 strain epimastigote form rendering very potent anti-T. cruzi agents. Moreover, the BzNO were able to inhibit the growth of virulent and resistant to Benznidazole strains (CL Brener clone, Colombiana, and Y strains) and to Leishmania braziliensis. Interestingly, BzNO exhibited very high selectivity index and particularly the spiro-BzNO 13 provokes an important diminution of amastigotes in Vero cells. Besides, it was found a diminution of acetate and glycine as excreted metabolites but without increase of parasite glucose uptake indicating that the glycosome is probably not involucrate in the 2H-benzimidazole 1,3- dioxides mechanism of action

In vitro growth inhibitory activity of 21 new 5-ethenylbenzofuroxan derivatives against the protozoan parasite Trypanosoma cruzi, the causative agent of American trypanosomiasis, was studied. The designed compounds possess the previously described exigencies for optimal anti-parasite activity, the 5-ethenylbenzofuroxanyl moiety with different substituents. The synthetic key for preparing the derivatives was the Wittig procedure, that when 5-formylbenzofuroxan was used as the electrophile the corresponding deoxygenated products were marginally generated. Four of the new derivatives displayed remarkable in vitro activities against the epimastigote form of three strains of T. cruzi, Tulahuen 2, CL Brener, and Y. While the three deoxygenated analogues biologically assayed resulted inactives. Unspecific cytotoxicity was evaluated using human macrophages and active derivatives were not toxic at a concentration at least 13 times that of its IC50 against T. cruzi (CL Brener strain). From the preliminary structure–activity relationship studies lipophilicity and electronic requirements were found relevant to anti-T. cruzi activity. Active compounds are more lipophilic than inactive ones and it was also identified that an optimum value of R Swain-Lupton’s descriptor is required for optimal activity

All products were tested for their in vitro NO-releasing capacities, vasodilating properties and mammal cytotoxic activities. The lipophilic–hydrophilic balance of all products was also evaluated. A new hybrid furoxan, phenol derivative 17, possesses adequate profile of the studied properties.

A summary of the resource.
Internal dosimetry intercomparisons are essential for the verification of applied models and the consistency of results'. To that aim, the First Regional Intercomparison was organised in 2005, and that results led to the Second Regional Intercomparison Exercise in 2013, which was organised in the frame of the RLA 9/066 and coordinated by Autoridad Regulatoria Nuclear of Argentina. Four simulated cases covering intakes of (131)I, (137)Cs and Tritium were proposed. Ninteen centres from thirteen different countries participated in this exercise. This paper analyses the participants' results in this second exercise in order to test their skills and acquired knowledge, particularly in the application of the IDEAS Guidelines. It is important to highlight the increased number of countries that participated in this exercise compared with the first one and, furthermore, the improvement in the overall performance. The impact of the International Atomic Energy Agency (IAEA) Projects since 2003 has led to a significant enhancement of internal dosimetry capabilities that strengthen the radiation protection of workers.

Entomopathogenic fungi such as Beauveria bassiana are extensively used for the control of insect pests worldwide. They infect mostly by adhesion to the insect surface and penetration through the cuticle. However, some insects, such as the red flour beetle Tribolium castaneum (Herbst), have evolved resistance by embedding their cuticle with antifungal compounds. Thus, they avoid fungal germination on the cuticle, which result in low susceptibility to entomopathogenic fungi. In adult T. castaneum, these antifungals are the well-known defensive compounds methyl-1,4- and ethyl-1,4- benzoquinone. In this study, we added B. bassiana conidia on the diet of adult beetles to study the effect of the entomopathogen on the secretion and detection of the beetle volatile blend containing both benzoquinones. The compounds were analyzed by solid phase microextraction coupled to gas chromatography–flame ionization detection, and were detected by electroantennography. In addition, we measured the expression level of four genes encoding for two odorant-binding proteins (OBP), one chemosensory protein (CSP), and one odorant receptor (OR) in both healthy and fungustreated insects. Significant alterations in the secretion of both benzoquinones, as well as in the perception of methyl-1,4-benzoquinone, were found in fungus-treated insects. TcOBP7D, TcOBP0A and TcCSP3A genes were down-regulated in insects fed conidia for 12 and 48 h, and the latter gene was up-regulated in 72 h samples. TcOR1 expression was not altered at the feeding times studied. We conclude that fungus-treated insects alter both secretion and perception of benzoquinones, but additional functional and genetic studies are needed to fully understand the effects of fungal infection on the insect chemical ecology.

The profiles of tocopherols and tocotrienols (vitamin E) in the seed oils recovered from apple (Malus domestica Borkh.), canary melon (Cucumis melo L.), gooseberry (Ribes uva-crispa L.), grape (Vitis vinifera L.), Japanese quince (Chaenomeles japonica), pomegranate (Punica granatum L.), red currant (Ribes rubrum L.), sea buckthorn (Hippophae rhamnoides L.), and watermelon (Citrullus lanatus) by-products of the fruit industry were studied. The highest concentration of tocochromanols was found in pomegranate seed oil (398.73 mg/100 g) and the lowest in gooseberry seed oil (69.13 mg/100 g). The γ and α homologues were the predominant forms of tocopherol and tocotrienol in all fruit seed oils studied. The pomegranate, canary melon, watermelon, gooseberry and red currants seed oils contained mainly γ-tocopherol (382.69, 63.08, 111.01, 60.35, and 156.39 mg/100 g, respectively). The profiles of tocopherols and tocotrienols (vitamin E) in the seed oils recovered from apple (Malus domestica Borkh.), canary melon (Cucumis melo L.), gooseberry (Ribes uva-crispa L.), grape (Vitis vinifera L.), Japanese quince (Chaenomeles japonica), pomegranate (Punica granatum L.), red currant (Ribes rubrum L.), sea buckthorn (Hippophae rhamnoides L.), and watermelon (Citrullus lanatus) by-products of the fruit industry were studied. The highest concentration of tocochromanols was found in pomegranate seed oil (398.73 mg/100 g) and the lowest in gooseberry seed oil (69.13 mg/100 g). The γ and α homologues were the predominant forms of tocopherol and tocotrienol in all fruit seed oils studied. The pomegranate, canary melon, watermelon, gooseberry and red currants seed oils contained mainly γ-tocopherol (382.69, 63.08, 111.01, 60.35, and 156.39 mg/100 g, respectively). In the apple, sea buckthorn and Japanese quince seed oils, α-tocopherol was the predominant tocochromanol (58.77, 121.79, and 198.94 mg/100 g, respectively).
