New thiazolyl derivatives (BT and TT) and their copper (II) complexes [Cu2Cl2(BT)2] (Cu-BT) and [Cu4ClO2(TT)2]PF6⋅3.5H2O (Cu-TT) were synthesized and characterized by elemental analysis, 1H NMR and 13C NMR, HRMS, X-ray diffraction, IR and UV–Vis spectroscopies. The crystal structure of Cu-BT shows the formation of a dinuclear complex where each copper(II) center is bonded to two thiazol N atoms, from different BT ligands, one deprotonated amide N atom, an O atom from the ester terminal groups and a chlorine atom. The structure found for Cu-TT is a positively charged tetranuclear moiety containing two deprotonated TT ligands, a chlorine anion, two hydroxide anions acting as bridges between the copper centers and a water molecule. The cytotoxic activity of both copper complexes was evaluated on metastatic breast cancer cell lines, characterized for its rapidly dividing behavior. Both, Cu-BT and Cu-TT, show higher cytotoxic activity against these tumor cells than free BT and TT and also than cisplatin. In addition, we found that both complexes interact with DNA. Consistently, they also show cytotoxicity against a rapidly dividing non-tumor cell line, although with higher IC50, being such interaction and selectivity an indicator of the possible coexistence of more than one mechanism of action.

Objective: The aim of this study was to develop an emulgel for the treatment of rosacea, applying quality by design (QbD). Methods: An emulgel designed to release the active pharmaceutical ingredients (APIs), metronidazole and niacinamide, via an emollient formulation that favors residence time and attenuates facial redness would be an excellent vehicle to develop to treat rosacea. It was decided to design first a vehicle presenting the attributes established in the quality target product profile, and then, after selecting the best formulation, to load the APIs in it to optimize the final emulgel. A design of experiments was introduced to study the effect of formulation variables on quality attributes (adhesion, phase separation by mechanical stress and viscosity) of the emulgels. Response surface methodology and desirability functions were applied for data analysis. After optimization, the final emulgel was further characterized by assay and in vitro release of APIs, attenuation of facial redness, and compared to commercially available metronidazole products regarding API release. Results: The final emulgel gradually released both APIs, reaching approximately 88% within the first 4 h, and their profiles were well described by the Higuchi model. Only a light attenuation effect to conceal facial redness was achieved. Conclusions: A metronidazole and niacinamide emulgel, also providing cosmetic assistance, was developed using QbD. The emulgel releases metronidazole faster than the creams, but more gradually than the commercially available gel, providing a realistic time frame of drug delivery in accordance with the expected time of residence of the adhesive emulgel over the affected facial area.

Los estudios de exploración y prospección realizados en Uruguay hasta el momento no han podido probar la existencia de yacimientos de combustibles fósiles tradicionales (petróleo, carbón mineral y gas natural) económicamente viables para su explotación. En el último siglo, Uruguay ha tenido que importar petróleo y gas natural para satisfacer gran parte de su demanda energética. Esto ha tenido un alto costo económico determinando una fuerte dependencia del exterior, pero también en los últimos tiempos ha sido impulsado el desarrollo y el uso de fuentes renovables de energía así como la exploración y estudio de aplicaciones de combustibles fósiles no convencionales tales como las Lutitas Pirobituminosas. Se han reportado numerosos estudios de co-combustión de Lutitas Pirobituminosas con otros combustibles como los lodos de tratamiento de efluentes y lignina, demostrando que esta tecnología puede ser una alternativa viable para la producción de energía. Sin embargo, existen pocos trabajos de investigación relacionados con la combustión de Lutitas Pirobituminosas con biomasa. Dada la abundancia de residuos de biomasa generados por la actividad agroforestal en Uruguay, este trabajo de tesis propone realizar un estudio riguroso de los fundamentos de la co-combustión de las Lutitas Pirobituminosas uruguayas con residuos de biomasa (Cáscara de Arroz, Eucalyptus grandis y Orujo de Uva), teniendo en cuenta parámetros ambientales. Se emplearon técnicas para caracterizar las Lutitas Pirobituminosas uruguayas, los residuos de biomasa y las mezclas preparadas a partir de estos, desde el punto de vista estructural, morfológico y fisicoquímico. Además, se evaluó la cinética y parámetros térmicos de diferentes mezclas de los combustibles. Durante el desarrollo de la tesis de posgrado se realizó una pasantía en la Universidad de Queensland con el fin de realizar otros análisis que permitieron obtener información importante de los combustibles y de la cinética de combustión mediante el empleo de análisis termogravimétrico acoplado a FTIR, Transient plane Source y Calorimetría de Cono. Los resultados indicaron que la relación entre el calor efectivo de combustión y el poder calorífico superior y las emisiones gaseosas de contaminantes ix atmosféricos de las Lutitas Pirobituminosas se ven mejoradas con la adicción de biomasa para la granulometría empleada para la realización de los análisis, tanto desde el punto de vista cinético como térmico. Se ha llegado a establecer un rango de composición de mezcla a ser utilizado principalmente con residuos de Eucaliptus grandis, la cual debe ser menor a 50 % de Lutitas Pirobitumino

Abstract
A summary of the resource.
Leaves extracts and essential oil of Nectandra angustifolia were explored for the first time for neutralization of Bothrops neuwiedi diporus snake venom. The ethanol extract was the most active and inhibited both venom activities (hemolytic and coagulant), while the oil was only active on the coagulant activity. These observations confirmed that certain medicinal plants from Corrientes and Chaco Provinces possess significant snake venom neutralizing capacity and need further examination for their active constituents. Analysis by GC and GC-MS of the essential oil and the enantiomeric excess found for -pinene, -pinene and limonene allowed a better characterization of this species.

<p>Medicinal plants encompass a rich source of active compounds that can neutralize snake venoms or toxins. Nectandra angustifolia (Schrad.) Nees & Mart. (Lauraceae), commonly known as “laurel amarillo”, “aju’yhû”, “laurel-hu”, “laurel<br />del río”, is used in traditional medicine as a digestive, for the treatment of rheumatism, arthritis, pain, and as antivenom. Snakebite envenomation was included in category A of neglected tropical diseases (NTD), in 2017, as they affect millions of people in the world (Chippaux, J Venom Anim Toxins 23:38–39: 2017). In Argentina, the snake genus Bothrops is responsible for the greatest number of accidents. Presently, the application of serum antivenom is the only specific treatment. This treatment does not protect over local effects, which are the main cause of morbidity. In the search for effective antivenoms, extracts of different polarity obtained from Nectandra angustifolia were evaluated both, in vitro and in vivo, against the venom of B. diporus "yarará chica". The effect of seasonal variability in this activity was also studied, to optimize both, the harvest period and the most appropriate type of extract. By successive bio-guided fractionations of the ethanol extract of N. angustifolia a fraction enriched in flavonoids responsible for the alexiteric activity of the species has been isolated. The phytochemical study of the volatile fraction of N. angustifolia and N. megapotamica allowed for the phytochemical characterization of farther interesting compounds.</p>

Antisnake activity of extracts and essential oil of Nectandra megapotamica was tested in vitro against the Bothrops diporus venom (yarará chica). Inhibitory activity of the hemolytic action on the aqueous extract and essential oil; inhibition of the procoagulant action on hexanic extract and condensed water from steam distillation; and inhibition of the proteolytic activity on alcoholic extract and condensed water was found. In all cases, the main antisnake venom activity was found on plant material collected in the autumn. The chemical composition of the N. megapotamica essential oil was characterized during different vegetative states finding a clear predominance of mono- (21.0–31.7) and sesquiterpene (58.5–68.9) hydrocarbons. The differences found with previous results published for this species, growing in other geographic places, opens the option for the existence of chemical types.

El samario-I53 Ipresenta optimas propiedades fisicas para su aplicacion como agente de radioterapia en cancer oseo secundario. Se trata de un emisor B de mediana energia y con un corto periodo de semidesintegracion. Emite ademas rayos y de energia adecuada como para generar imagenes centellogrMicas, 10 que permite el monitoreo continuo de la dosis recibida por el paciente. El desarrollo de nuevos farmacos, cada vez mas especificos, constituye un enorme desafio. El objetivo que se persigue es lograr agentes de radioterapia biolocalizados, capaces de entregar elevadas dosis de radiacion en las zonas de lesion, minimizando la depresion inducida por la radiacion en la medula osea. La captacion de arninoacidos y peptidos sencillos por parte de celulas anormales se encuentra favorecida, debido al metabolismo alterado de las celulas tumorales. Como consecuencia, el disefio de nuevos complejos de samario con aminoacidos y peptidos sencillos constituye una estrategia posible hacia la generacion de farmacos mas especificos para radioterapia osea. Por otra parte, el transporte de muchos metales in vivo, asi como la captacion de los rnismos por parte de receptores de membrana esta mediado por estas biomoleculas pequefias, 10 que podria tambien contribuir a la bioespecificidad buscada. Quimicamente, los arninoacidos presentan al menos un grupo carboxilato como po sible sitio de coordinacion al samario. Con el proposito de disefiar complejos mas estables, que sean capaces de alcanzar los sitios de accion sin sufrir descomposicion, surge la posibilidad de emplear ligandos que posean variado nfunero de grupos carboxilato. Estos ligandos son capaces de formar complejos quelato que estabi1izan al metal en solucion y disminuyen la probabilidad de degradacion in vivo. Finalmente, debido at alto indice de coordinacion del samaria (generalmente de ocho 0 nueve) resulta una hipotesis razonable el poder obtener complejos de ligandos mixtos de ese metal que contengan arninoacidos en su esfera de coordinacion. En estos complejos, un ligando policarboxilico estabiliza al atomo central. Los sitios de coordinacion atm disponibles (ocupados por moleculas de agua) pueden ser sustituidos por uno 0 mas aminoacidos. La coordinacion con estas biomoleculas podria aportar la especificidad deseada. Sobre la base de 10 antedicho, se estudia en este trabajo la formacion de complejos de samario con aminoacidos, peptidos sencillos y ligandos po licarboxHicos. Se explora tambien la formacion de complejos de ligandos mixtos de samario con ligandos policarboxilicos como ligandos primarios y arninoacidos como ligandos secundarios. Todos estos estudios se realizan en solucion acuosa y en condiciones sirnilares alas fisiologicas. Se deterrnina para cada sistema que especies complejas se forman, cuales predominan en condiciones fisiologicas y se evalua su estabilidad a partir de las constantes de formacion, mediante estudios potenciometricos. Adicionalmente, se Resumen sintetizan, aislan y caracterizan algunos de los complejos estudiados en los distintos sistemas propuestos, de manera de amp liar el conocimiento de la estructura tanto en soluci6n como en estado s6lido. Se logra asi disponer de un conjunto de nuevos complejos de samario con ligandos de interes biol6gico, muchos de los cuales resultan estables en condiciones fisiol6gicas. Este estudio sistematico permite ademas realizar un amilisis comparativo de los complejos obtenidos, expandiendo el conocimiento basico en esta area. l-Arabinose isomerase is an intracellular enzyme that can convert l-arabinose to l-ribulose and d-galactose to d-tagatose, a promising but rare nutraceutical. Most of l-arabinose isomerases purified up to date employed the combination between DNA recombinant technology and affinity chromatography based on poly-histidine tail recognition, but few of the enzymes were obtained and purified in a non-recombinant way. For these reasons, a specific affinity bioadsorbent containing l-arabitol as ligand, a competitive inhibitor of the enzyme, was designed and synthesized for achieving pure preparations of the enzyme l-arabinose isomerase from wild-type Enterococcus faecium DBFIQ E36 strain, isolated from raw cow milk. The two-step purification procedure consisted in fractionation by ammonium sulphate precipitation followed by affinity chromatography with obtained bioadsorbent, allowing the purification, to electrophoretical homogeneity, of target enzyme. Characterization studies were performed with purified l-arabinose isomerase in order to increase knowledge of their physicochemical properties. In this sense, enzyme exhibited an optimum temperature of 50 °C and optimum pH of 7.0, maintaining good stability in the ranges 20–45 °C and pH 6.5–8. Ki were calculated, employing d-galactose as substrate, for l-arabitol and l-ribitol, achieving values of 7.9 mM and 183 mM, respectively. Km and Vmax values obtained were 35 mM and 81 U mg−1 at 50 °C, respectively. Mass spectrometry assay revealed a 48 kDa monomer whereas gel permeation chromatography achieved a 187 kDa molecular weight for native enzyme. Finally, 2D-electrophoresis and isoelectrofocusing analysis revealed an isoelectric point value of 3.80. Results have unveiled both an acidic nature and promising properties for l-arabinose isomerase isolated from E. faecium DBFIQ E36.

The use of ketohexose isomerases is a powerful tool in lactose whey processing, but these enzymes can be very sensitive and expensive. Development of immobilized/stabilized biocatalysts could be a further option to improve the process. In this work, β-galactosidase from Bacillus circulans, l-arabinose (d-galactose) isomerase from Enterococcus faecium, and d-xylose (d-glucose) isomerase from Streptomyces rubiginosus were immobilized individually onto Eupergit C and Eupergit C 250 L. Immobilized activity yields were over 90% in all cases. With the purpose of increasing thermostability of derivatives, two post-immobilization treatments were performed: alkaline incubation to favor the formation of additional covalent linkages, and blocking of excess oxirane groups by reacting with glycine. The greatest thermostability was achieved when alkaline incubation was carried out for 24 h, producing l-arabinose isomerase-Eupergit C derivatives with a half-life of 379 h and d-xylose isomerase-Eupergit C derivatives with a half-life of 554 h at 50 °C. Preliminary assays using immobilized and stabilized biocatalysts sequentially to biotransform lactose at pH 7.0 and 50 °C demonstrated improved performances as compared with soluble enzymes. Further improvements in ketohexose productivities were achieved when the three single-immobilizates were incubated simultaneously with lactose in a mono-reactor system.

β-Galactosidase from Bacillus circulans, l-arabinose (d-galactose) isomerase from Enterococcus faecium and d-xylose (d-glucose) isomerase from Streptomyces rubiginosus, immobilized individually and co-immobilized onto Eupergit C and Eupergit C 250 L, were used batchwise in mono- and multi-bioreactor systems for lactolysis/isomerization processes in Mozzarella cheese whey. The degrees of lactolysis achieved with the multienzymatic systems were significantly higher than those obtained by using mono-enzymatic processes with immobilized β-galactosidase. This effect was observed when at least one of the isomerases was present in the reactor, suggesting a reduction in product inhibition of lactolysis as a result of decrease in d-galactose and/or d-glucose levels during reaction. Temperature was the main factor that affected d-tagatose/d-galactose and d-fructose/d-glucose ratios. Degree of lactolysis achieved at 50 °C was between 88% and 100%. Mono-reactor processes with co-immobilized stabilized tri-enzymatic systems exhibited the best performances regarding conversion degrees (producing complete lactolysis, 45% d-tagatose and 39% d-fructose against 93% lactolysis, 40% d-tagatose and 29% d-fructose for mono-reactor processes with individually immobilized enzymes). Stabilized co-immobilized enzymes were able to improve further to 53% the yield in d-fructose when demineralized Mozzarella cheese whey was employed as substrate. The productivities were moderate, ranging 9.5–10 g/Lh in terms of d-tagatose. Stabilized derivatives could be reused for at least 12 cycles in batch bio-reactors.

A summary of the resource.
<p>Ibogaine is a psychedelic alkaloid that has attracted large<br />scientific interest because of its antiaddictive properties in observational studies in humans as well as in animal models. Its subjective effect has been described as intense, vivid dream-like experiences occurring while awake; hence, ibogaine is often referred to as an oneirogenic psychedelic. While this unique dream-like profile has been hypothesized to aid the antiaddictive effects, the electrophysiological signatures of this<br />psychedelic state remain unknown. We previously showed in rats that ibogaine promotes a waking state with abnormal motor behavior along with a decrease in NREM and REM sleep. Here, we performed an indepth analysis of the intracranial electroencephalogram during “ibogaine wakefulness”. We found that ibogaine induces gamma oscillations that, despite having larger power than control levels, are less coherent and less complex. Further analysis revealed that this profile of gamma activity compares to that of natural REM sleep. Thus, our results provide novel biological evidence for the association between the psychedelic state and REM sleep, contributing to the understanding of the brain mechanisms associated with the oneirogenic psychedelic effect of ibogaine.</p>

<p>Tissue engineering (TE) and regenerative medicine offer strategies to improve damaged tissues by using scaffolds and cells. The use of collagen-based biomaterials in the field of TE has been intensively growing over the past decades. Mesenchymal stromal cells (MSCs) and dental pulp stem cells (DPSCs) are promising cell candidates for development of clinical composites. In this study, we proposed the development<br />of a bovine collagen type I: chondroitin-6-sulphate (CG) scaffold, obtained from Uruguayan raw material (certified as free bovine spongiform encephalopathy), with CG crosslinking enhancement using different gamma radiation doses. Structural, biomechanical and chemical characteristics of the<br />scaffolds were assessed by Scanning Electron Microscopy,<br />axial tensile tests, FT-IR and Raman Spectroscopy,<br />respectively. Once we selected the most appropriate scaffold for future use as a TE product, we studied the behavior of MSCs and DPSCs cultured on the scaffold by cytotoxicity, proliferation and differentiation assays. Among the diverse porous scaffolds<br />obtained, the one with the most adequate properties was the one exposed to 15 kGy of gamma radiation.</p>

A versatile palladium-catalyzed tandem synthetic sequence to afford E-stilbenes libraries has been developed. Excellent regio- and stereocontrol have been achieved by means of the sequence of Hiyama and Heck cross-couplings. Undesirable homocoupling byproducts were avoided employing immobilized substrates.

The aim of the present work was to study the hydrogel-forming polysaccharides carrageenan, guar and xanthan gum as potential absorbents in moisture activated dry granulation (MADG). Preferences of formulators regarding absorbents tend to incline towards microcrystalline cellulose and colloidal silicon dioxide. Besides, only few works exploring modified drug release achieved by MADG are reported in literature. A factorial design of experiments was introduced to study the effect of formulation variables on granulates and their tablets. Contact angle and drop penetration time provided useful information for guidance on excipient selection. Metronidazole was used as a highly soluble model drug to verify the release performance achieved with the polysaccharides. All of the granulates yielded tablets with appropriate physical properties, and significantly different disintegration behaviors and drug release profiles. Absorbent, binder and their mutual interaction had significant effects on formulation performance. The type of absorbent affected tablet disintegration time and drug release. The presence of copovidone, alone or in combination with povidone, proved to be a good choice for granulating a high load of not directly compressible materials by MADG. Carrageenan, guar and xanthan gum should be regarded as promising moisture absorbents for either immediate or extended release tablet formulations produced by MADG, an energy and time efficient granulation process. A broader range of absorbents could provide more versatility in MADG formulation and this advantageous process become more eligible for producing modified release products.

Compounds that cause powerful sensory irritation to humans were reviewed by the Scientific Advisory Board (SAB) of the Organisation for the Prohibition of Chemical Weapons (OPCW) in response to requests in 2014 and 2017 by the OPCW Director-General to advise which riot control agents (RCAs) might be subject to declaration under the Chemical Weapons Convention (the “Convention”). The chemical and toxicological properties of 60 chemicals identified from a survey by the OPCW of RCAs that had been researched or were available for purchase, and additional chemicals recognised by the SAB as having potential RCA applications, were considered. Only 17 of the 60 chemicals met the definition of a RCA under the Convention. These findings were provided to the States Parties of the Convention to inform the implementation of obligations pertaining to RCAs under this international chemical disarmament and non-proliferation treaty.

A summary of the resource.
The corrosion behavior of Zr-2.5%Nb pressure tube material and Zr-1%Nb aged at 560 °C for 4 months has been studied in 0.1 M lithium hydroxide solution at 343 °C. The results were compared with those obtained in steam at 400 °C. In the latter, aged alloys showed better behavior than the non-aged materials, whereas the ageing did neither delay the transition nor reduce the weight gains in 0.1 M lithium hydroxide. Second phase particles of Zr-Nb-Fe and Zr-Fe present in aged Zr-1Nb (α+β) and aged Zr–1Nb α’ seem to increase hydrogen uptake during post-transition in 0.1 M lithium hydroxide. Regarding the hydrogen absorption in the post-transition period, Zr-1Nb alloys show better performance than Zircaloy-4.
