The postharvest disease popularly known as gray mold is considered one of the most limiting factors strawberry fruit pro duction. The most efective way to control this disease is still the use of chemical fungicides. However, other alternative sources of control are being explored. Among these, psychrophilic yeasts adapted to extreme conditions, such as those found in the Antarctic region, may have great potential for use as biocontrol agents. Thus, the present study aimed to select psychrotolerant yeasts obtained from Antarctic region and to evaluate their potential for biocontrol under gray mold, caused by Botrytis cinerea in strawberries stored at low temperature. For this, 20 potential antagonist yeasts were evaluated in vitro (thermotolerance and enzymatic) assays. Debaryomyces hansenii, Rhodotorula mucilaginosa and Dioszegia hungarica were selected for growing in strawberry juice. However, only D. hansenii was selected for in vivo studies and showed a reduction in the incidence of gray mold by 82% for the tests performed on injury and 86% for the tests on non-injured fruits treated by immersion bath. Perceptions of essences for potential use in the development of a line of cosmetic emulsions containing olive oil were studied. Six cream samples prepared with six essences selected in a preliminary study were evaluated for overall liking and intention to purchase by a 63-women sample. A check-all-that-apply (CATA) question consisting of 32 terms was used to gather information about consumer perceptions of fragrance, affective associations, effects on the skin, price, target market, zones of application, and occasions of use. Hierarchical cluster analysis led to the identification of two consumer clusters with different frequency of use of face creams. The two clusters assigned different overall liking scores to the samples and used the CATA terms differently to describe them. A fragrance with jasmine as its principal note was selected for further development of cosmetic creams, as it was awarded the highest overall liking scores by respondents of the two clusters, and was significantly associated with cosmetic features including nourishing, moisturizing, softening, with a delicious and mild smell, and with a natural image, as well as being considered suitable for face and body creams. The use of CATA questions enabled the rapid identification of attributes associated by respondents with a cosmetic cream's fragrance, in addition to contributing relevant information for the definition of marketing and communication strategies. BACKGROUND: Increasing demand for sweet potato in regions with temperate climates has triggered interest in the development of new cultivars. Breeding of this crop should consider sensory characteristics in order to meet consumers’ expectations. This requires the application of simple and cost-effective methodologies that allow quality evaluation from a sensory perspective. RESULTS:With the objective of identifying the key sensory characteristics of different sweet potato genotypes, two commercial cultivars and seven clones were evaluated during three consecutive years using projectivemapping by an untrained consumer panel. This methodology allowed the discrimination of the genotypes, identifying similarities and differences among groups based on sensory terms selected by the assessors. Genotypes were differentiated in terms of texture and flavor characteristics (firmness, moisture, smoothness, creaminess, flavor intensity, sweetness and bitterness). Materials for future crossings were identified. CONCLUSIONS: The evaluation of the sensory characteristics of sweet potato clones and cultivars using projectivemapping is a quick, cost-effectiveandreliable tool for theselection ofnewadvancedsweet potato clones with superior sensory characteristics compared to the reference cultivars INIA Arapey and Cuarí. Aminophenylboronate-substituted agarose in 20 mM N-2-hydroxyethylpiperazine-N′-2-ethanesulphonic acid, pH 8.5, selectively adsorbs immunoglobulins and complement factors C3 and C4 from human serum. The selectivity of binding is strongly influenced by the presence of magnesium chloride in the sample buffer. Adsorbed immunoglobulins are quantitatively eluted by sorbitol, but only partially by ethylene glycol or methylcellosolve. Aniline-agarose of a similar degree of substitution shows only weak adsorption of serum proteins under similar experimental conditions, thus indicating the important contribution of the boronate moiety to this interaction. Immunoglobulin adsorption seems not to be due to the cis-diol complexation used extensively for the chromatographic determination of non-enzymatically glucosylated proteins. Hydrophobic and π-π interactions with the aromatic structure of the ligand seem also to contribute to protein binding. H-beta zeolite has been tested for the hydration of dihydromyrcene (DHM) to produce dihydromyrcenol (DHM-OH). By using a non-protic co-solvent like acetone, in order to work under a single phase system, reasonable conversion with excellent selectivity to DHM-OH is obtained, although some double bond isomerization of the terpene is also taking place. The reaction is not controlled by intrapore diffusion, and samples with different crystal sizes gave similar activities and selectivities. However, defect free beta samples with high hydrophobic character are preferred for the reaction due to their improved adsorption properties for non-polar molecules. Working with those materials more than 50% yield of DHM-OH was obtained with a selectivity close to 100%. A new approach for the control and interruption of enzymatic reactions via selective enzyme immobilization has been developed. The technique was exemplified by the use of three model enzymes with the corresponding macromolecular substrates: alpha-amylase/starch, trypsin/ insoluble collagen, and alkaline phosphatase/plasmid DNA. Prior to incubation with its substrate, each enzyme was provided with de novo thiol-groups by a two-step reaction involving N-succinimidyl-3-(2-pyridyldithio) propionate (SPDP) and DTT. The chemical modification was achieved such that at least 80% of the native enzyme activity was preserved in all cases. In order to interrupt rapidly the reactions in which the enzymes were used, the modified enzyme was immobilized by reaction via its thiol groups on a thiolsulfinate-agarose derivative. The gel-bound enzyme could then be easily removed from unreacted substrate and product by filtration or centrifugation. Comparative studies showed that the immobilized enzymes had much lower activities in the reactions studied than the corresponding soluble ones. The potential for enzyme reuse was also demonstrated with the alpha-amylase derivatives, which were quantitatively released and eluted in fully active form from the agarose. We have shown that it is possible to achieve practically complete enzyme immobilization in short times and thus to control the progress of the reactions. The solubility of different ethyl esters derivatized from hake liver oil in supercritical carbon dioxide was studied. A selectivity factor was used to determine optimal conditions to fractionate the ethyl ester mixture. A strong influence of solvent pressure and temperature was observed within8.63–18.04 MPa and 40–70 �C. The lowest total solubility of the ethyl ester mixture was obtained when using supercritical carbon dioxide at the lowest density (the lowest pressure and the highest temperatures value tested). Selenium is an essential trace element. Selenium speciation in natural environmental conditions has been poorly studied, mostly because its relevance has been only recently acknowledged. In this work, a complete thermodynamic study on selenium(IV) and selenium(VI) was carried out. It includes the protonation equilibria of selenium anions in aqueous solution at 20 °C. The interaction of selenium anions with some metal ions that are normally encountered in environmental scenarios was also studied. The strategy involves the potentiometric study of all the equilibria using different ionic strength values of a non interacting electrolyte. Distribution species diagrams allow the prediction of the predominant species of selenium both under reducing or oxidizing conditions and in the presence of these metal ions. The dependence of the thermodynamic magnitudes on the ionic strength value is also evaluated. The speciation of Se(IV) or Se(VI), 10−7 M, in the presence of mean concentrations of Ca2+ (16.5 mM) and Mg2+ (6.4mM) shows the predominance of HSeO3 −, [Ca(HSeO3)]+ and [Mg(HSeO3)]+, or of SeO4 2−, respectively. Results show that, depending on media pH and pE, interaction with metal ions can be either negligible or very important. A gold amalgamator, used conventionally for Hg vapor enrichment, is reported to preconcentrate Se after hydride generation. Optimum trapping and volatilization temperatures were 450 and 970 °C, respectively. Preconcentration processes are controlled by temperature only while carrier gas composition remains unchanged (75 ml min-1 H2). This is a significant advantage compared to the approach reported previously based on a gold wire, when Se preconcentration was controlled by both surface temperature and carrier gas composition. Moreover, the gold amalgamator offers higher capacity and better resistance towards interferences, at least by an order of magnitude. This approach to Se preconcentration is very well compatible with atomic absorption spectrometry using a modular trap-and-atomizer device reported recently. A limit of detection for 300 s sample introduction time reached 13 pg ml-1 Se (247 pg Se absolute). The method developed was validated by analysis of water certified reference materials. Mechanism of selenium hydride trapping was investigated employing 75Se radioactive indicator. Potential of coupling the gold amalgamator trap to an inductively coupled plasma mass spectrometer was also investigated. However, severe transport losses of volatilized Se species occur between the heated amalgamator and the torch due to analyte condensation on colder surfaces preventing this set-up from routine use. Selenoprotein T (SELENOT) is an endoplasmatic reticulum (ER)-associated redoxin that contains the amino acid selenocysteine (Sec, U) within a CXXU motif within a thioredoxin-like fold. Its precise function in multicellular organisms is not completely understood although it has been shown in mammals to be involved in Ca2+ homeostasis, antioxidant and neuroendocrine functions. Here, we use the model organism C. elegans to address SELENOT function in a whole organism throughout its life cycle. C. elegans possess two genes encoding SELENOT protein orthologues (SELT-1.1 and SELT-1.2), which lack Sec and contain the CXXC redox motif instead. Our results show that a Sec→Cys replacement and a gene duplication were two major evolutionary events that occurred in the nematode lineage. We find that worm SELT-1.1 localizes to the ER and is expressed in different cell types, including the nervous system. In contrast, SELT-1.2 exclusively localizes in the cytoplasm of the AWB neurons. We find that selt-1.1 and selt-1.2 single mutants as well as the double mutant are viable, but the selt-1.1 mutant is compromised under rotenone-induced oxidative stress. We demonstrate that selt-1.1, but not selt-1.2, is required for avoidance to the bacterial pathogens Serratia marcescens and Pseudomonas aeruginosa. Aversion to the noxious signal 2-nonanone is also significantly impaired in selt-1.1, but not in selt-1.2 mutant animals. Our results suggest that selt-1.1 would be a redox transducer required for nociception and optimal organismal fitness. Trypanosomatids previously examined harbor a full set of genes encodingthe machinery needed for selenocysteine biosynthesis and incorporation into three selenoproteins: SelK,SelT and, the parasite-specific, Seltryp. We investigated the selenoproteome of kinetoplastid speciesin recently sequenced genomes and assessed the in vivo relevance of selenoproteins for African try-panosomes. Database mining revealed that SelK, SelT and Seltryp genes are present in most kinetoplastids,including the free-living species Bodo saltans, and Seltryp was lost in the subgenus Viannia from theNew World Leishmania. Homology and sinteny with bacterial sulfur dioxygenases and sulfur transferasessuggest a putative role for Seltryp in sulfur metabolism. A Trypanosoma brucei selenocysteine synthase(SepSecS) null-mutant, in which selenoprotein synthesis is abolished, displayed similar sensitivity tooxidative stress induced by a short-term exposure to high concentrations of methylglyoxal or H2O2tothat of the parental wild-type cell line. Importantly, the infectivity of the SepSecS knockout cell line wasnot impaired when tested in a mouse infection model and compensatory effects via up-regulation ofproteins involved in thiol-redox metabolism were not observed. Collectively, our data show that seleno-proteins are not required for survival of African trypanosomes in a mammalian host and exclude a rolefor selenoproteins in parasite antioxidant defense and/or virulence. The cysteine protease cruzipain is an essential T. cruzi enzyme and one of the few validated drug targets for Chagas disease. Thiosemicarbazones have been described as cruzipain inhibitors. While searching for new antichagasic drugs, we synthesized a series of selenosemicarbazone analogs and demonstrated that the isosteric replacement of the sulfur atom with selenium resulted in an enhancement of the cysteine protease inhibitory effect. Three selenosemicarbazones were characterized enzymatically and proved to be reversible, slow-binding inhibitors for the Z-Phe-Arg- AMC substrate. Their KI values were in the low nM range (3.7 to 29.7 nM), suggesting a strong interaction with the enzyme, approaching a tight binding definition. All selenosemicarbazones tested showed better activities against epimastigotes than Benznidazole, the currently used drug, with IC50 values ranging from 1.2 to 5.9 mM (Bnz IC50 ¼ 12.5 mM). Three of these compounds showed a better Selectivity Index (SI) than Benznidazole. These compounds also displayed activity against the infective intracellular amastigote form at low micromolar range. Overall, our results support the role of these novel organoselenium compounds as promising lead candidates for further drug development studies. The most abundant member of this family in nature is InsP6 (phytate, L12− in its fully deprotonated form). Phytate interacts strongly with inorganic and organic cations, and this interaction is essential for determining the possible functions of this biomolecule. Herein, the chemical, thermodynamic, and structural characterization of phytate–MnII species is presented in a study aimed at understanding how the interaction of the two components modulates their biological roles and their bioavailability. Polynuclear complexes Mn5(H2L)⋅16 H2O (1) and (H2terpy)2[Mn(H6L)(terpy)(H2O)]⋅17 H2O (terpy=terpyridine) (2) were prepared and characterized by different techniques. The isolation of 1 and the determination of its solubility, together with potentiometric titrations of the MnII–phytate system, allow the full description of this binary system. In this competitive world, a package has only a few seconds to make an impact and catch the consumer’s eye. In that time, a successful label should communicate its message and convince the shopper. Considering the importance of transmitting an adequate, and in some occasions, the “perfect” message, it is crucial to understand how consumers perceive and interpret the labels froma semiotic perspective, which has not been considered in depth. In this context, the objective of the present study was to gather information on how consumers of different age groups read the profiles of symbolic labels and what they construe. A word association task was performed with 101 participants (50 aged below 35 years old, 51 aged 60 and over). Participantswere asked towrite down all that came to their minds when they sawfive simple images of yogurt labels, which were constructed by combining a variety of signs. All the elicited terms were grouped into 54 categories and then into nine general dimensions. Though some main messages were conveyed successfully, relevant differences were found in perceptions and interpretations of the two age cohorts. Youngsters made more references to the specific attributes of the expected product, whereas the elders assignedmore global and hedonic meanings.
