Here, we report the preparation of PEDOT:PSS-MoS2 nanocomposite thin films and their electrical resistance response under humidity conditions revealing their promising properties as humidity sensor materials. One of the most interesting features of our samples is the transition from positive to a negative electrical resistance response to humidity conditions with increasing MoS2 additions. Our confocal Raman imaging studies revealed that the presence of MoS2 yields a local charge rearrangement in the thiophenyl rings of PEDOT:PSS, in relation to the enhancement of the electrical resistance negative response as observed by impedance spectroscopy analysis. The enhancement on the negative response with increasing MoS2 additions could be explained through the increment of hole carriers in MoS2 nanosheets under humidity conditions, thus leading to an enhancement in the electrical transport along the PEDOT:PSS chains.

A summary of the resource.
H2/CO/CO2 gaseous mixtures obtained by biomass gasification (bio-syngas) could be used to obtain hydrocarbons by the Fischer-Tropsch synthesis (FTS). In this work, trimetallic Fe–La–Zr mixed oxides prepared by coprecipitation with oxalic acid were studied in the synthesis of hydrocarbons from bio-syngas at 300 °C and 10 bar. Catalysts obtained by calcination at 700 °C are activated by the bio-syngas and the one that contains 30 wt% of Fe attains steady CO conversion levels of 75% and a total hydrocarbon yield of 34%–36%. A liquid product containing hydrocarbons in the range C5–C13 is formed together with C1–C4 products. Potassium promotion (6 wt%) allows to increase CO conversion (∼89%) and hydrocarbons yield (∼44%). Catalysts obtained by calcination at 900 °C display higher activation times due to their crystalline structure, although they attain final activity levels close to the amorphous ones obtained by calcination at 700 °C.

The use of Cannabis for medical purposes is rapidly expanding and is usually employed as a self-medication for the treatment of insomnia disorder. However, the effect on sleep seems to depend on multiple factors such as composition of the Cannabis, dosage and route of administration. Vaporization is the recommended route for the administration of Cannabis for medical purposes; however, there is no published research about the effects of vaporized Cannabis on sleep, neither in laboratory animals, nor in humans. Because previous reports suggested that low doses of THC have sedating effects, the aim of the present study was to characterize in rats, the acute effects on sleep induced by the administration of low doses of THC by means of vaporization of a specific type of Cannabis (THC 11.5% and negligible amounts of other cannabinoids). For this purpose, polysomnographic recordings in chronically prepared rats were performed during 6 h in the light and dark phases. Animals were treated with 0 (control), 40, 80 and 200 mg of Cannabis immediately before the beginning of recordings; the THC plasma concentrations with these doses were low (up to 6.7 ng/mL with 200 mg). A quantitative EEG analyses by means of the spectral power and coherence estimations was also performed for the highest Cannabis dose. Compared to control, 200 mg of Cannabis increased NREM sleep time during the light phase, but only during the first hour of recording. Interestingly, no changes on sleep were observed during the dark (active) phase or with lower doses of Cannabis. Cannabis 200 mg also produced EEG power reductions in different cortices, mainly for high frequency bands during W and REM sleep, but only during the light phase. On the contrary, a reduction in the sleep spindles intra-hemispheric coherence was observed during NREM sleep, but only during the dark phase. In conclusion, administration of low doses of THC by vaporization of a specific type of Cannabis produced a small increment of NREM sleep, but only during the light (resting) phase. This was accompanied by subtle modifications of high frequency bands power (during the light phase) and spindle coherence (during the dark phase), which are associated with cognitive processing. Our results reassure the importance of exploring the sleep-promoting properties of Cannabis.

Minthostachys verticillata essential oil (EO) is a natural product that reports immunomodulatory effects on human T cells as well as anti-inflammatory activity. Bovine mastitis is a worldwide disease, mainly caused by bacteria, affecting milk quality and yield, leading to high economic losses. Environmental pathogens, as Enterococcus faecium, are implicated in the disease. Antibiotic therapy is adequate, although it can leave residues in milk, causing problems in human health. The search of immunomodulatory substances for bovine mastitis treatment is a promising alternative strategy. The aim of this study was to characterize the effect of M. verticillata EO on macrophage phagocytosis and evaluate its immunomodulatory and protective effects in mice challenged with E. faecium. The results showed that EO activated macrophage phagocytosis mechanisms inducing reactive oxygen species production. Moreover, EO modulated the innate immune response in mammary glands of female Balb/c mice challenged with E. faecium decreasing the infiltration of polymorphonuclear neutrophils and IL-1β and TNF-α mRNA expression. In addition, EO increased the expression of IL-10 in the last hours of infection. Treatment with EO did not increase the number of activated CD4+ or CD8+ T cells or the production of specific antibodies. These results suggest that EO play an important role in helping to resolve the infection in the first hours without activating adaptive immunity. In addition, a marked decrease of the bacterial count in the glands of mice treated with EO was observed. A natural product such as M. verticillata EO could have a potential use to control bovine mastitis.

Abstract
A summary of the resource.
Three years pesticide residues monitoring data in pollen, wax, bees and honey from sentinel apiaries in different agroecosystems in Uruguay were used for meta analysis. Support vector machines (SVM) algorithm was used previously to classify environments and is the basis of the present work. An useful indicator was developed to classify the contamination status of both beehives and associated environment. The index is a quotient between hive simple demographic parameters and the sum of toxic units for each pesticide found in it. The Agroecology Environment Quality quotient (AEQ) can be scaled in three different levels. When it is below 1, the hive will hardly develop in such environment. AEQ values between 1 and 2 indicate that the hive could have some restrictions to develop in such environment due to pesticide contamination but probably it will be productive. In the cases where the index is above 2, the hive will develop well in the environment where it is located. AEQ permits simultaneously to obtain different information. These are: the surveillance of good agricultural practices accomplishment in agroecosystems, the selection of the best environment to perform apicultural activities as well as to monitor the productivity and development of the beehives.

Suitable conditions of temperature and humidity are required to maintain wheat grains quality, but during processing and storage, the grains can be exposed to adverse environmental conditions and presence of infectious fungi. Fusarium graminearum, the main causal agent of Fusarium head blight on wheat, affects crop yields and grain quality by alteration of their biochemical components and mycotoxin contamination, which reduces the possibilities of wheat end use and compromises food safety. Lipid degradation by hydrolytic, oxidative and microbial deterioration is the predominant cause of the loss of sensory acceptability, nutritional value and baking quality. The aim of this research was to determine the influence of adverse environmental conditions –as the increasing moisture – on lipid patterns of whole wheat flours contaminated with F. graminearum in relation to the infection degree. In vitro cultures of F. graminearum were carried out on wheat grains under different degrees of relative humidity (11, 50, 75 and 100%) throughout 45 days of incubation at 28 °C. The fungal biomass measured by q-PCR increased proportionally with the humidity. A decrease in the signals of saturated (palmitic and estearic) and unsaturated (oleic, linoleic and linolenic) fatty acids, analyzed as fatty acid methyl esters (FAMEs) by GC–MS, was observed in relation with the humidity and infection degree. The degradation rate of the lipids was high during the first 15 days of incubation, reaching the fatty acids content, values around 20–40% of those found in the control. From that moment on, the rate of degradation was slower or even null. It was observed that in all treatments, the linolenic acid reached the highest degradation ratio in comparison with the other fatty acids, which may be caused by the action of lipoxygenases. The lipase activity and the content of deoxynivalenol were also determinate on the flours. The lipase activity increased until day 25 of incubation reaching twice the initial value. The deoxynivalenol content also increased along incubation while fatty acids decreased. Our results demonstrated that the magnitude in the signal of fatty acids in whole wheat flours varied in relation to the degree of humidity and fungal infection of the grains from which they were obtained. Otherwise, lipids and their oxidation products are related with the pathogenesis and production of mycotoxins. These observations highlight the importance of an adequate manipulation of wheat grains on the processing chain to prevent quality changes and mycotoxins contamination.

Two straightforward methods for glyphosate and AMPA analysis in honey through ion chromatography coupled to Q-Orbitrap accurate high-resolution mass spectrometry as well as their application to real samples are presented. The sample preparation step is accomplished through dilution with methanol–water or water. No pH adjustment is necessary. Recoveries were in the range 80 to 110% with RSDs < 20%. The LOQ of glyphosate was 0.005 mg kg−1, far below the allowed EU-MRL (0.05 mg kg−1) in honey, showing a linear range of 0.005–0.5 mg kg−1 and medium matrix effect. LOQ of AMPA was 0.02 mg kg−1. Glyphosate quantitation was performed with internal or external calibration yielding identical results, thus broadening the applicability of the method as isotopically labelled standards are no longer mandatory. The method was applied to 32 honey samples from different origins. Glyphosate was found in 81% of the samples and 41% were above the EU-MRL showing its applicability in routine work. AMPA was not detected in any of the samples.

We determined the antiproliferative and nitric oxide (NO)-releasing activity of furoxans and tocopherol analogs–furoxan hybrids by tandem Griess/resazurin/sulforhodamin B assays in HeLa, 253J, T24, and HepG2 cancer cells. In addition, to investigate the NO implications in the inhibition of cell growth, cells were pretreated with the NO scavenger hemoglobin and the genotoxic damage was determined. The compounds 1 and 3 emerged as good anticancer agents for bladder cancer treatment. The NO-releasing activity of these compounds appears to be necessary to obtain the antiproliferative effect. Although compound 1 exerted a DNA damage mechanism of action, compound 3 seemed to act in a different way. The low toxicity levels against normal cell line HaCaT point them out as a very promising scaffold for the further design of new anticancer agents.

Here, we report the preparation of carbon-coated cobalt-doped olivine nanoparticles (LiFePO4/C) by the citrate gel precursor combustion method. The major contribution of this work is to reveal insights and side effects of cobalt doping, not only in the structure, microstructure, and electrochemical performance of olivine nanoparticles, but also in the consequences on the carbon coating characteristics. Our structural and microstructural studies by means of several instrumentation techniques revealed that the presence of low amounts of cobalt during the synthesis leads to the apparent formation of more porous channels in the carbon coating. In consequence, the electrochemical characterization showed an enhancement in the capacity and diffusion constants for cobalt-doped LiFePO4/C samples in comparison with cobalt-undoped samples.

We report the effect of lithium titanate nanotubes (LiTNT) as active fillers for lithium-ion solid polymer electrolytes for lithium batteries. The interaction of LiTNT with polyacrylonitrile host material and enhancement on lithium perchlorate ionic-pair dissociation was clearly evidenced by our vibrational spectroscopy and confocal Raman microscopy studies. In strong correlation with the structural characterization, the active role of LiTNT fillers was evidenced by means of impedance spectroscopy analysis revealing the presence of two contributions to the ionic transport, one due to the bulk and the other possibly mediated by the LiTNT surface in the nanocomposites. A four-order-magnitude enhancement on the lithium-ion bulk conductivity was observed for 15% LiTNT loadings with respect to unloaded samples showing an increment up to ~ 7.1 × 10−4 S.cm−1. In addition, we also evidence that the LiTNT surface-mediated contribution to the lithium-ion transport yielded conductivities in the same order of magnitude.

Here, we report a synergistic in situ synthesis of lithium-ion poly (ethylene citrate) with embedded titania nanoparticles in which the esterification process itself triggered the hydrolysis of the titanium alkoxide precursor to form a promising fluorine-free lithium-ion solid polymer electrolyte. Structural characterization by means of X-ray diffraction, small angle X-ray scattering and also confocal Raman spectroscopy revealed the formation of the polyester with embedded titania nanoparticles in agreement with our Raman spectra simulation by means of DFT methodologies. Our theoretical modeling of the polyester also envisaged the interaction of lithium-ion via a tridentate coordination with oxygen atoms of carboxylate (–COO-) and ester (–COO–) groups. There is a slight increase in the degree of polymerization and thermal stability for our lithium-ion polyester with increasing titania nanoparticles concentration. In consequence, a two-order magnitude increase in the ionic conductivity was observed, yielding 1.74× 10−4 S cm−1 for a 20% weight fraction of titania nanoparticles. Thus, involving a novel approach based in the synergistic in situ preparation of lithium-ion poly (ethylene citrate) with titania nanoparticles, this procedure contributes to solve the many safety and environmental issues of the constitutive materials of lithium-ion solid polymer electrolytes.

Here we present a new analytical method where immunoconcentration of the analyte is coupled to quantitative matrix-assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF MS) analysis allowing in minutes the identification and highly sensitive quantitation of microcystins (MCs) as model targets. The key element is a site-specific in vivo biotinylated nanobody of broad cross-reactivity with microcystins. The single biotin moiety at the C-terminus and the small size of the nanobody (15 kDa) enable its oriented and tightly packed immobilization on magnetic beads, providing a highly efficient capture of the toxin. The binding capacity of the bioadsorbent is partially loaded with an easily synthesized internal standard for MS quantitation. After capture, the beads are directly dispensed on the MALDI-TOF MS target enabling the identification and sensitive quantitation of the microcystin (MC) congeners. Since salts and contaminants are removed during the concentration step, no cleanup or other sample treatments are needed. The method was validated with a large number of water and serum samples with excellent precision and recovery at quantitation limits of 0.025 μg/L of MC.

La integración enzimática en materiales híbridos permite capitalizar las ventajas sinérgicas de los diferentes materiales estructurales para mejorar las propiedades del biocatalizador. En esta tesis, hemos estudiado la síntesis de nanopartículas nanohíbridas [silica biomimética + nanopartículas magnéticas (MNPs)] como un vehículo de peroxidasa de rábano picante (HRP) para su uso en la terapia dirigida de profármacos de enzimas (DEPT). DEPT usa enzimas exógenas introducidas artificialmente en el cuerpo con el objetivo de convertir profármacos en su forma activa en el sitio deseado del organismo y, por lo tanto, tiene una ventaja sobre las terapias estándar no específicas por su selectividad potencial. El uso de enzimas enzapsuladas o unidas a nanopartículas (NPs) ha ganado interés en este tipo de terapia ya que permite mejorar la estabilidad y reducir la inmunogenicidad de la enzima terapéutica, a la vez que mejorar la bioacumulación y retención de la enzima en el tejido tumoral. Esto es debido al efecto de retención y permeabilidad aumentada (EPR) por la presencia de una abundante vascularización inmadura en el tumor que hace que de forma pasiva se acumulen mayor cantidad de NPs que en tejidos sanos. Por tanto, las enzimas terapéuticas transportada por NPs tienen una mayor facilidad en acumularse en tejidos tumorales que la misma enzima administrada en forma libre. Sin embargo, las NPs no pueden evitar la distribución no selectiva en el cuerpo, sobre todo en órganos que para filtrar la sangre también usan el efecto EPR (hígado, bazo)., Por lo tanto, el uso exitoso de modalidades de DEPT que utilicen nanomateriales como vehículos de la enzima terapéutica depende de la implementación de estrategias que puedan proporcionar un control espaciotemporal sobre su actividad enzimática. Para esto, en esta Tesis se ha optimizado la co-encapsulación de MNP junto con la enzima terapéutica HRP en nanopartículas de sílica biomimética. Se eligió HRP debido a su capacidad para convertir el ácido indol-3-acético (hormona vegetal) en radicales peroxilados con actividad citotóxica en varias líneas celulares tumorales. Sin embargo, la HRP tiene una temperatura óptima de 45ºC y a la temperatura corporal su actividad biocatalítica no es máxima. Debido a que las MNPs pueden absorber energía en presencia de un campo magnético alterno externo (AMF) y liberarla al medio ambiente en forma de calor, nuestra hipótesis de trabajo se centra en lograr utilizar este calor para 3 alcanzar la temperatura óptima de la HRP de forma localizada (en una escala nanométrica), y así incrementar su actividad enzimática. Tradicionalmente, el desencadenar el aumento macroscópico de la temperatura de tejidos tumorales ha sido la estrategia más difundida para utilizar el gradiente muy alto de temperatura que se genera por calentamiento magnético en pocos nanómetros desde la superficie de la MNPs. Esto ha permitido el desarrollo de una nueva terapia para el tratamiento del cáncer, conocida como hipertermia magnética (MHT), ya que las células tumorales tienen una mayor sensibilidad al calor que las células normales. En esta Tesis sin embargo proponemos aprovechar este gradiente de temperatura localizado alrededor de las MNPs no para desencadenar que se eleve de forma macroscópica la temperatura de un tejido tumoral sino para modular la aactividad de enzimas co-entrapadas con las MNPs en nanohíbridos de sílica biomimética. El uso de silica biomimética en el nanohíbrido se planteó bajo la premisa de utilizar un medio para estabilizar la enzima, que permita su probable inmunoaislación y, como se demostró más tarde, alcanzar a su vez un posible efecto de aislamiento térmico que disminuya la rápida disipación de calor que ocurre a pocos nm de la superficie de las MNPs y así sea posible modular la actividad de enzimas que no se encuentren unidas a la superficie de las MNPs. Es así que efectivamente la estrategia de co-encapsulación facilitó un aumento selectivo en la actividad biocatalítica de HRP al alcanzar localmente su temperatura óptima. Para llegar a esta conclusión, hemos optimizado la síntesis de los nanohíbridos variando diferentes parámetros tales como: i) tamaño del catalizador aminado requerido para la formación de NP de sílica (polietilenimina MW ~ 1300, 2000, 25000, 60000 Da), ii) metodología de inmovilización de la HRP (adsorción / unión covalente en MNP antes de la encapsulación, co-encapsulación con MNP), iii) relación HRP/ MNP. La inmovilización de HRP y los rendimientos de actividad después del atrapamiento son similares con o sin MNP (~ 60 ± 4% y 78 ± 1, respectivamente). Sin embargo, la estabilidad térmica de la enzima (50ºC) aumentó 532 (Factor de estabilización) cuando la HRP se co-encapsula con MNP. El uso de PEI de diferentes tamaños no afectó los rendimientos de la inmovilización de HRP ni el tamaño (diámetro hidrodinámico) de los híbridos. Sin embargo, las MNP parecen actuar como plantillas para la deposición de sílice ya que su co-encapsulación con HRP reduce el tamaño radio de los biohíbridos (de ~ 800-1000 a 500-600 nm). Todos los nanohíbridos obtenidos pudieron oxidar IAA, pero no se observó citotoxicidad en 4 ausencia del profármaco usando diferentes líneas celulares. También hemos estudiado el efecto del tamaño del catalizador aminado requerido para la formación del nanobiocatalizador (polietilenimina MW ~ 1300, 2000, 25000, 60000 Da), en su eficiencia de calentamiento magnético macroscópico (SAR, Specific Absorption Rate o tasa de absorción específica) y nanoscópico (activación enzimática). Los nanohíbridos (500-600 nm) mostraron activación enzimática (250-300%) sin un aumento significativo de la temperatura macroscópica y conservaron su actividad completa después de la aplicación de un ciclo de AMF (829 kHz, 25.2 mT) durante 20 min. Sin embargo, para que los nHs desarrollados puedan tener potencial para ser utilizados en DEPT es clave que presenten buena estabilidad coloidal en medios biológicos complejos. Para ello fue necesario bloquear grupos amino primarios presentes en la superficie de los mismos con glucosa. Este monósocaráido no solo permitió mejorar la estabilidad coloidal de los nHs en medio de cultivo completo, pero también sirve de biomoléculta para vectorización activa de células tumorales. Los nanohíbridos funcionalizados con glucosa lograron diferencias significativas en la reducción de la viabilidad celular de una línea celular de cáncer de colon humano (HCT-116) después de la exposición a un ciclo de AMF (80% versus 30% de reducción en controles sin aplicación de AMF). Estos resultados muestran por primera vez que es posible controlar de forma remota la actividad de HRP co-atrapada con MNP mediante calentamiento magnético para una potencial aplicación en terapia antitumoral DEPT.

La papa (Solanum tuberosum L.) es uno de los cultivos más importantes afectado por Ralstonia solanacearum, bacteria fitopatógena causante de la marchitez bacteriana. Una de las estrategias más efectivas para el control de esta enfermedad es el uso de cultivares resistentes. La estrategia adoptada por el programa nacional de mejoramiento genético de papa desarrollado en INIA, implica la introgresión de genes de resistencia a partir de la especie nativa silvestre Solanum commersonii, ampliamente distribuida y adaptada a nuestras condiciones ambientales. El objetivo general de esta tesis fue generar conocimiento sobre el patosistema R. solanacearum - S. commersonii para contribuir al desarrollo de cultivares con resistencia a esta bacteria fitopatógena y al control integrado de la marchitez bacteriana. Se evaluó la resistencia en germoplasma derivado del programa de mejoramiento y se seleccionaron genotipos con resistencia parcial altamente tolerantes que fueron utilizados como progenitores en los sucesivos cruzamientos. Además, se seleccionaron genotipos con diferentes niveles de resistencia para continuar con los estudios de las interacciones planta-patógeno. Para realizar el seguimiento de la colonización de la bacteria in planta, se construyeron y validaron cepas reporteras de R. solanacearum con actividad fluorescente (proteína verde fluorescente, GFP) y luminiscente (operón luxCDABE, LUX). Los resultados obtenidos sugieren que la resistencia a la marchitez bacteriana en papa está relacionada con la capacidad de la planta de restringir la colonización y multiplicación del patógeno, particularmente limitando la diseminación a lo largo del tallo. A su vez, se observaron varias respuestas de defensa inducidas luego de la infección con la bacteria en los genotipos con resistencia parcial como deposición de lignina, producción de especies reactivas del oxígeno, tílides, un aumento del intercambio de electrolitos a través de la membrana y cambios a nivel de la expresión de genes de defensa. También se analizó la composición mineral de la savia xilemática, hoja, tallo y raíz de los genotipos seleccionados encontrando mayores concentraciones de Ca a nivel de tallo y savia xilemática en el genotipo parcialmente resistente. Este mineral tiene un efecto inhibidor del crecimiento de R. solanacearum y sobre características asociadas a la virulencia como la formación de biofilm, adhesión y la movilidad tipo twitching. El agregado exógeno de sales de Ca retrasó y disminuyó la severidad de síntomas de marchitamiento en plantas susceptibles y resistentes inoculadas con R. solanacearum. Este efecto también se observó cuando se incubó a la bacteria en cámaras de microfluido, logrando optimizar por primera vez este modelo para el estudio de este patógeno bajo condiciones de flujo.

En el presente trabajo de tesis se implementó una prueba de concepto: el diseño y construcción de un analizador autónomo de campo para la determinación de fósforo y nitrógeno en aguas superficiales. El analizador construido permite la determinación de fosfato, nitrito y nitrato, está basado en un sistema de flujo pulsado con detección fotométrica y cuenta con comunicación de los resultados mediante mensaje de texto al teléfono móvil del usuario. Para cumplir los objetivos planteados, se profundizaron los conocimientos respecto a los perjuicios causados por las altas cargas de nutrientes en agua, su origen y las formas de determinación de los mismos; así como también lo que son los valores actuales de los niveles de fósforo y nitrógeno en nuestros principales cuerpos de agua. Se estudiaron las diferentes formas de detección del fósforo y del nitrógeno para su determinación analítica, particularmente en lo que refiere a su implementación en sistemas de análisis en flujo. Se seleccionaron los métodos químicos basados en la detección fotométrica por su sencillez de escalado y bajo costo de implementación en el desarrollo de sistemas analíticos portables y autónomos. Para llevar a cabo la construcción del analizador fue aplicar conocimientos de electrónica y programación lo cual brinda al presente trabajo un alto componente interdisciplinario.

Galectins are a family of carbohydrate-recognizing proteins that by interacting with specific glycoepitopes can mediate important biological processes, including immune cell homeostasis and activation of tolerogenic circuits. Among the different members of this family, Galectin 1 and 3 have shown pro-tumorigenic effects, being overexpressed in numerous neoplasic diseases, proving to be relevant in tumor immune escape, tumor progression and resistance to drug-induced apoptosis. Thus, generation of specific glycosides that could inhibit their pro-tumorigenic ability by blocking their carbohydrate recognition domain is one of the current major challenges in the field. Considering that galectin-ligand binding strength is closely related to the ligand structure, analysis of this relationship provides valuable information for rational design of high-affinity ligands that could work as effective galectin inhibitors. Taking profit of the ability of glycosidases to catalyze transglycosylation reactions we achieved the enzymatic synthesis of β-d-Galp-(1 → 6)-β-d-Galp-(1 → 4)-d-Glcp (2), a mixture of β-d-Galp-(1 → 6)-β-d-Glcp-(1 → 4)-d-Glcp (5) and β-d-Galp-(1 → 3)-β-d-Glcp-(1 → 4)-d-Glcp (6), and finally benzyl β-d-galactopyranoside (9), with reaction yields between 16 and 27%. All the galactosides were purified, and characterized using 1H and 13C nuclear magnetic resonance spectroscopy. Docking results performed between the synthesized compounds and human Galectin 1 (hGal-1) and human Galectin 3 (hGal-3) showed that the replacement of a glucose moiety linked to the terminal galactose with a galactose moiety, decreases the affinity for these galectins. Moreover, regarding the interglycosidic bond the most favorable β-Gal linkage seems to be β(1 → 4) followed by β(1 → 3) and β(1 → 6) for hGal-1, and β(1 → 4) followed by β(1 → 6) and β(1 → 3) for hGal-3. These results were in accordance with the IC50 values obtained with in vitro solid phase inhibition assays. Therefore, docking results obtained in this work proved to be a very good approximation for predicting binding affinity of novel galactosides.

Two flexible ligands derived from iminodiacetic acid, N-methlyiminodiacetic acid (H2mida) and N-(2-hydroxyethyl)iminodiacetic acid (H2hida) have been used as ligands towards lanthanide ions. Potentiometric studies (25.0 °C, 0.50 mol L−1 Me4NCl) revealed the predominance of stable species at acidic pH values. With this information, the synthesis of the coordination compounds was afforded in aqueous solution and the polymeric solids [Sm(Hmida)2(H2O)2]ClO4·1/6NaClO4·9H2O and [Ln(hida)(H2O)2](F3CSO3)·H2O (Ln = Ho, Yb) were prepared and characterized. The samarium compound consists of a cationic 2D polymeric structure in which the ligand is protonated in the N atom, preventing coordination to the central atom. The structure presents channels along one crystallographic direction. Ho and Yb coordination compounds are isostructural. A 2D polymeric cationic network is also observed. In this case, the additional hydroxyethyl group and the unprotonated N atom participate in the coordination.

Phytate (L12−) is a relevant natural product. It interacts strongly with biologically relevant cations, due to the high negative charge exhibited in a wide pH range. The synthesis and crystal structures of the mixed‐ligand Cu(II) polynuclear complexes K(H2tptz)0.5[Cu(H8L)(tptz)] ⋅ 3.6H2O (1), K(H2O)3{[Cu(H2O)(bpca)]3(H8L)} ⋅ 1.75H2O (2), and K1.5(H2O)2[Cu(bpca)](H9.5L) ⋅ 8H2O (3) (tptz=2,4,6‐tri(pyridin‐2‐yl)‐1,3,5‐triazine; Hbpca=bis(2‐pyridylcarbonyl) amine) are reported herein. They were obtained by the use of an aromatic rigid amine, which satisfies some of the metal coordination sites and promotes the hierarchical assembly of 2D polymeric structures. Speciation of phytate−Cu(II)−Hbpca system and determination of complex stability constants were performed by means of potentiometric titrations, in 0.15 M NMe4Cl at 37.0 °C, showing that, even in solution, this system is able to produce highly aggregated complexes, such as [Cu3(bpca)3(H7L)]2−. Furthermore, the Cu(II)‐mediated tptz hydrolysis was studied by UV‐vis spectroscopy at room temperature in 0.15 M NMe4Cl. Based on the equilibrium results and with the aid of molecular modelling tools, a plausible self‐assembly process for 2 and 3 could be proposed.

A summary of the resource.
The marine bacterium Stenotrophomonas rhizophila was assessed in vitro and in vivo as biocontrol agent against anthracnose disease of mango fruit caused by Colletotrichum gloeosporioides. The results showed that in vitro inhibition of the colony diameter and spore germination of the phytopathogen was due to the production of VOCs, competition for nutrients, and lytic enzymes. When a concentration of 1 × 108 cells ml−1 of the antagonist bacterium was applied to the fruit, disease incidence was reduced by 95%, and the lesion diameter of anthracnose decreased by 85%, which offered greater protection than the synthetic fungicide. This is the first report of antagonistic mechanisms of the marine bacterium S. rhizophila against anthracnose disease in mango, which in this study was found to be more effective than the synthetic fungicide.

The inhibitory effects of Cannabis flower extracts, obtained by supercritical carbon dioxide (scCO2) with and without modifier, on various human tumour cells and non-tumour cells were evaluated. Different techniques were used to optimize the polarity interaction between solute and solvent before extraction. An increase in the cannabinoid content of interest (CBD/THC) was evaluated by decarboxylation of the flowers at 110 and 140 °C. Extractions with pure scCO2 were conducted in a single step at temperatures of 50 and 70 °C and pressures of 22 and 40 MPa, varying the time of prior decarboxylation by 0.5 or 2.0 h. Sequential extractions were conducted in a first step at 35 °C and 10 MPa and then in a second step at 70 °C and 40 MPa, with and without modifier. Contents of the cannabinoids were evaluated using HPLC. Antitumor activity of the extracts was evaluated using the MTT assay. The highest yields and highest cannabinoid contents in the extracts were obtained with high solvent density values. Extracts with high concentrations of neutral cannabinoids showed high antitumor activity for cervical cancer cell lines.

Para extender y fortalecer la Mejora Continua en la “Unidad de Educación Permanente” (UNEP) de la Facultad de Química (FQ) de la Universidad de la República de Uruguay (UdelaR) se emplearon herramientas clásicas de ”Gestión de Calidad” que han demostrado su eficacia en distintas ramas de las actividades Empresarial y Educativa. La experiencia permitió identificar “oportunidades de mejora” e implementar las acciones de seguimiento correspondientes, La experiencia realizada es considerada una muy buena práctica de trabajo en consideración a los resultados obtenidos. Además, su ejecución implicó un uso mínimo de recursos disponibles, se aplicó fácilmente la metodología de trabajo, se logró una comprensión rápida de los participantes y gran empatía por parte de los actores involucrados. En resumen, se consiguió un excelente cumplimiento de los objetivos propios establecidos por el Consejo de Facultad de Química (CFQ) para la función de “Educación Permanente” (EP) de la FQ. La divulgación de esta experiencia puede servir como ejemplo para su aplicación en múltiples áreas de la actividad docente y no docente de la UdelaR.

Nematode parasites have a profound impact on humankind, infecting nearly one-quarter of the world’s population, as well as livestock. There is a pressing need for discovering nematicides due to the spread of resistance to currently used drugs. The free-living nematode Caenorhabditis elegans is a formidable experimentally tractable model organism that offers key advantages in accelerating nematicide discovery. We report the screening of drug-like libraries using an overnight high-throughput C. elegans assay, based on an automated infrared motility reader. As a proof of concept, we screened the “Pathogen Box” library, and identical results to a previous screen using Haemonchus contortus were obtained. We then screened an in-house library containing a diversity of compound families. Most active compounds had a conjugation of an unsaturation with an electronegative atom (N, O, or S) and an aromatic ring. Importantly, we identified symmetric arylidene ketones and aryl hydrazine derivatives as novel nematicides. Furthermore, one of these compounds, (1E,2E)-1,2-bis(thiophen-3-ylmethylene)hydrazine, was active as a nematicide at 25 µm, but innocuous to the vertebrate model zebrafish at 50 µm. Our results identified novel nematicidal scaffolds and illustrate the value of C. elegans in accelerating nematicide discovery using a nonlabor-intensive automated assay that provides a simple overnight readout.

Searching for a more effective chemotherapy for the treatment of Human African trypanosomiasis, the disease caused by the parasite Trypanosoma brucei, and cancer, in the current work five new [PtII(L)(dppf)](PF6) compounds, with HL = 8-hydroxyquinoline derivatives and dppf = 1,1′-bis(diphenylphosphino)ferrocene, were synthesized and fully characterized. Crystal structures of three compounds were solved by XRD. The compounds displayed fairly good activity against bloodstream T. brucei, with IC50 values in the submicromolar range (IC50: 0.14–0.93 μM), and good selectivity towards the pathogen (SI: 11 - 48) with respect to mammalian macrophages (cell line J774). Coordination to the {Pt-dppf} moiety led, in most cases, to an enhancement of the activity in respect to the bioactive ligands (11 to 41 fold). Cytotoxicity was assessed against wildtype (A2780) and cisplatin-resistance (A2780cisR) ovarian cancer cell lines. Four [PtII(L)(dppf)](PF6) compounds were more active (IC50: 1.2–4.4 μM) than cisplatin (IC50: 26.0 μM) on A2780 cells and showed far superior activity than the reference drug against A2780cisR cells. Platinum levels in A2780 cells showed poor correlation between cellular uptake and the cytotoxic activity. All the complexes interacted with DNA and the most active ones induced reactive oxygen species (ROS) formation which suggested that the mechanism of action for these complexes may be mediated by oxidative stress and interaction with DNA that could act as a potential molecular target for this type of complexes. Some complexes of this series could be considered new hits for the development of prospective agents against trypanosomatid parasites and ovarian cancer.

Las plantas medicinales y aromáticas (PAMs) han sido empleadas por las sociedades humanas desde sus inicios, y son el punto de partida para la investigación en productos naturales. En ésta tesis se trabajó sobre la fitoquímica del género Baccharis L. (Asteraceae) como modelo de PAMs, colectando in natura 19 especies nativas de Uruguay, del sur de Brasil y del nordeste de Argentina: B. articulata, B. crispa, B. cultrata, B. dentata, B. dracunculifolia, B. genistifolia, B. gibertii, B. gnaphalioides, B. linearifolia, B. microdonta, B. milleflora, B. ochracea, B. palustris, B. phyteumoides, B. punctulata, B. spicata, B. tridentata, B. trimera y B. uncinella. Para todas ellas se describió su composición química volátil (volatiloma) y se identificaron los principales metabolitos producidos por cromatografía gaseosa convencional y acoplada a espectrometría de masa (GC-FID y GC-MS). Para el estudio, se tomaron en cuenta diferentes variables que inciden directamente sobre la composición como: metodología de extracción (B. uncinella), origen geográfico de las poblaciones vegetales (B. trimera), estacionalidad (B. dracunculifolia y B. microdonta) y género de las plantas (masculino/femenino; B. articulata, B. crispa, B. dracunculifolia, B. linearifolia, B. spicata y B. tridentata). En particular, sobre éste último aspecto se trabajó en caracterizar el perfil de emisión volátil de ambos sexos empleando para ello cromatografía gaseosa/olfatometría (GC-O) y cromatografía gaseosa enantioselectiva (eGC). Para la especie B. trimera (―carqueja‖) se realizó un detallado estudio de la composición del aceite esencial, fraccionando el mismo por cromatografía en columna (CC) y estudiando la composición de cada una de las fracciones por GC-MS. Adicionalmente, se realizó un estudio de las características morfoanatómicas e histoquímicas de una población de referencia de B. trimera, lo que permitió su caracterización biotípica. Desde el aceite esencial de ésta especie fue aislado por CC su compuesto mayoritario acetato de carquejilo, y mediante reacciones químicas específicas fueron obtenidos sus derivados semi-sintéticos: carquejol, carquejifenol, carquejona, iso-carquejona y óxido de carquejol (los últimos tres no reportados anteriormente en la bibliografía). En todos los casos los productos fueron caracterizados por medio de técnicas espectroscópicas de resonancia magnética nuclear (RMN), infrarrojo (IR), ultravioleta (UV), dicroísmo circular (DC), Raman y espectrometría de masa (MS). También fue realizado un estudio de química computacional para predecir las propiedades moleculares del acetato de carquejilo, carquejol y carquejifenol, empleando la teoría del funcional de densidad (DFT). Fitoquímica de Baccharis spp. (Asteraceae): Metabolitos Secundarios, Semi-Síntesis y Bioactividad Manuel Minteguiaga, 2019 15 Para la especie B. palustris fue determinado por diferentes técnicas espectroscópicas la presencia de poliacetilenos en su volatiloma. Consecuentemente, fueron identificados los siguientes compuestos 1-nonen-3,5-diino, 1,7(Z)-nonadien-3,5-diino, 1,7(E)-nonadien-3,5-diino y 3,5-nonadiino; así como los (E)/(Z) ésteres de lachnophyllum. Para los tres primeros compuestos es su primer reporte en la literatura como productos naturales. Finalmente, se estudió la bioactividad de extractos y compuestos semi-sintéticos mediante técnicas in-vitro: antiradicalaria (metodología del DPPH) y neutralizante del veneno de serpientes del género Bothrops sp. (―yaras‖) (actividad alexitérica). Los extractos de B. dracunculifolia, B. punctulata y B. trimera demostraron la mejor actividad de captura del DPPH, en tanto la carquejona y el (Z)-éster de lachnophyllum exhibieron los mejores resultados de neutralización de los venenos. Los resultados de ésta tesis y sus correspondientes publicaciones demuestran la importancia del enfoque desarrollado en la búsqueda de metabolitos bioactivos provenientes de PAMs.
