A method was developed to separate Toxoplasma gondii cysts from mouse brain tissue and to liberate intact bradyzoites from cysts. Brains were blended in a Waring blender with 20% dextran solution and the homogenate was centrifuged at 4,000 g for 10 min. Cysts present in the sediment were digested by adding an equal amount of a solution containing 1 g NaCl, 1.4 ml HCl, and 1 mg pepsin (1:60,000 assay activity) per 100 ml water, and incubated at 37 C for 1 min. This method is harmless for bradyzoites, as tested by bioassays in mice. It compares favorably with published methods for separating cysts that require 3 centrifugations to achieve similar results. Controlled syntheses give unique block oligomers with alternating flexible ethylene glycol and rigid perylenetetracarboxylic diimide (PDI) units. The number of rigid units vary from n=1 to 10. PDI units were stitched together by using efficient phosphoramidite chemistry. The resulting oligomers undergo folding in most solvents, including chloroform. In their ground state, these folded oligomers were characterized by using Fourier transform ion cyclotron resonance mass spectrometry (FTICR-MS), NMR spectroscopy, and electronic absorption spectroscopy. FTICR-MS revealed the exact masses of these sequence-controlled oligomers, which confirmed the chemical composition and validated the synthetic strategy. The NMR neighboring ring-current effect (NRE) indicates the formation of cofacial p stacks; the stacked aromatic rings have nearly coaxial alignment akin to a nanosoleniod. Nanosolenoidal shielding in p stacks causes all aromatic protons to shift upfield, whereas NOE in a cyclic hetero-chromophoric dimer supports a rotated, cofacial p-stacking orientation separated by about 3.5 �. Electron–phonon coupling is much stronger than excitonic coupling in these self-folded PDI oligomers; thus, Franck–Condon factors dictate the observed spectral features in visible spectra. The absorbance spectrum exhibits weak hypochromism due to p stacking with increasing stacking units n. Finally, ab initio calculations support the experimental observations, indicating 3.5 � cofacial spacing in which one molecule is rotated 308 from the eclipsed orientation and higher oligomers can adopt, without a compensating energy penalty, either the right/left-handed helices or the 1,3-eclipsed structures. The Piperaceae family is represented by herbaceous plants, shrubs, and, more rarely, trees. Among the aromatic varieties of this genus, Piper hispidinervum C. DC. stands out. In this work, volatile and non‐volatile extracts from P. hispidinervum were obtained through the application of sequential extraction methods. The results of steam distillation experiments indicated that optimal conditions were achieved when the extractions were conducted at 101 kPa and an initial bed porosity of 0.91. Chemical analysis showed that safrole is the major component present in the essential oil. NMR analysis revealed the presence of 6‐methoxy eugenol for P. hispidinervum extracts obtained by supercritical fluid extraction using water as co‐solvent at 20 MPa and 313.15 K. The dot-ELISA reaction was used to study the dynamics of IgG titers in cattle naturally infected with Fasciola hepatica after anthelmintic treatment and during reinfection. Excretion/secretion products (ES) of the parasite were used as antigens for the dot-ELISA. IgG antibodies were no longer detectable by dot-ELISA, 4–6 months after nine animals received the first of three weekly doses of triclabendazole (15 mg kg−1) and were then maintained on a pasture free of F. hepatica metacercariae. Six fluke-free cattle began shedding F. hepatica eggs 3–6 months after grazing a pasture contaminated with metacercariae of the parasite. A detectable increase in dot-ELISA IgG antibody levels was observed 2–4 weeks after natural reinfection by grazing a similar pasture contaminated with F. hepatica metacercariae. The usefulness of the dot-ELISA system to diagnose chronic infection by serology is complicated by previous treatment against the parasite. The immune system of sturgeons, one of the most ancient and economically valuable fish worldwide, is poorly understood. The lack of molecular tools and data about infection biomarkers hinders the possibility to monitor sturgeon health during farming and detect infection outbreaks. To tackle this issue, we mined publicly available transcriptomic datasets and identified putative positive acute-phase proteins (APPs) of Russian sturgeons that could be induced by a bacterial infection and monitored using non-invasive methods. Teleost literature compelled us to focus on five promising candidates: hepcidin, a warm acclimation associated hemopexin, intelectin, serum amyloid A protein (SAA) and serotransferrin. Among them, SAA was the most upregulated protein at the mRNA level in the liver of sturgeons challenged with heat-inactivated or live Aeromonas hydrophila. To assess whether this upregulation yielded increasing SAA levels in circulation, we developed an in-house ELISA to quantify SAA levels in sturgeon serum. Circulating SAA rose upon bacterial challenge and positively correlated with hepatic saa expression. This is the first time serum SAA has been quantified in an Actinopterygii fish. Objectives: To confirm the influence of systemic inflammatory response syndrome (SIRS) on selenium (Se) levels and prospectively evaluate the relationship between serum Se concentration [Se], glutathione peroxidase activity [GPx-3] and injury severity in patients at the time of intensive care unit (ICU) admission. Design: Prospective, observational study. Setting: Multidisciplinary University Hospital ICU. Patients and participants: A total of 36 ICU patients and 23 healthy volunteer subjects (HVS). Measurements and results: Healthy volunteer subjects were designated as controls (Group 1). ICU patients were divided into three groups: without SIRS (Group 2); with SIRS (Group 3); with SIRS and multiple organ dysfunction syndrome (MODS) (Group 4). The latter groups had APACHE II scores [15. [GPx-3] and [Se] were determined by standard methods within the first 48 h of admission to ICU. Kruskal– Wallis and Mann–Whitney U test were used for analysis of non-parametric continuous variables. The predictive value of [Se] and [GPx-3] for SIRS was calculated using a receiver operating characteristics (ROC) analysis. In SIRS and MODS patients [GPx-3] and [Se] decreased significantly (P = 0.0001 and P = 0.002, respectively). After ICU admission [GPx-3] and [Se] had a predictive value for SIRS ([GPx-3] sensitivity: 90%, specificity: 86.2% (cut-off value: 0.5 U/mL); [Se]: sensitivity 90%, specificity 72.4% (cut-off value: 60 lg/L). [Se] had predictive value for ICU mortality (P = 0.034). Conclusions: Systemic inflammatory response syndrome and MODS were associated with early decreases in [Se] and [GPx-3]. Low [Se] and [GPx-3] after ICU admission had a predictive value for SIRS, which may aid future selection of patients who could benefit from Se supplementation. Food reformulation is widely recognised as a cost-effective policy to reduce sodium intake and improve public health outcomes. The objectives of the study were to: i) assess the sodium content of food products available in the Uruguayan market, ii) evaluate progress in achieving regional sodium reduction benchmarks; and iii) compare maximum targets and gradual percentage reductions considering potential effectiveness and sensory perception thresholds. Information about sodium content was extracted from the food labels of 4,719 products available at nine supermarkets. The median sodium content of the products was 389.3 mg/100g. When comparing median sodium content to the targets set by the Pan American Health Organisation for 2021, 32 of the 62 subcategories exceeded the established benchmarks, while 30 subcategories fell below them. Adopting regional targets would necessitate sodium reductions exceeding the 15% difference threshold for salty taste for a large proportion of products, potentially leading to noticeable changes in taste. At the aggregate level, percentage reductions were significantly more effective in reducing sodium content compared to maximum target strategies based on the 50th or 70th percentile. Results from the present work underscore the need to adapt regional targets for the development of a sodium reduction program in Uruguay. A delayed-release gastroresistant formulation of ketoprofen was administered under fasting and fed conditions. Cmax, AUC, Cmax/AUC, and kel obtained after food coadministration did not differ from those calculated under fasting administration. Ninety-five percent confidence intervals for fed/fasting geometric mean ratio of Cmax/AUC and AUC were 0.80–1.14 and 0.80–1.23, respectively. A significant difference (P < .01) was found between lag-time medians (T0), with a longer T0 after food intake (5.5 vs 2.5 hours). Also, a significant difference between the medians of Tmax was found (P < .01), being 7.0 hours after food coadministration and 4.0 hours under fasting administration, but this difference disappeared once T0 was subtracted from Tmax. Cmax/AUC, which is related to drug absorption rate, showed significant differences between sexes. Men showed higher (P =.006) Cmax/AUC means (0.468 ± 0.094 vs 0.361 ± 0.087 h−1. Tmax was also significantly different (P < .05), being 4.0 (3.0–5.0) hours for men and 8.0 (5.0–10.0) hours for women. In conclusion, men showed a faster intestinal absorption rate with earlier time-to-peak plasma concentration of ketoprofen. Food coadministration extended the gastric residence time of formulation but exerted no effect on its intestinal absorption pattern. Fourteen healthy subjects (seven women and seven men) were enrolled, food intake was standardized, blood samples were withdrawn up to 48 h post dosing and VPA plasma concentrations were analyzed by HPLC–UV method. Secondary peaks of VPA were observed in almost all subjects. A population pharmacokinetic analysis with sex, weight, age and contraceptive therapy as possible covariates was performed. Women without contraceptive therapy presented a longer lag time, a lower VPA hepatic output and a higher reabsorbed fraction than men. Weight affected both volume of the central compartment and the absorption rate constant. Women under contraceptive therapy showed similar disposition parameters to men. Reabsorbed fraction of bioavailable dose was 46.2 % in women and 21.8 % in men, revealing important sex differences in drug hepatobiliar output. These results showed that VPA displays sex-related pharmacokinetic differences due to different metabolic and transporter-mediated disposition. In vitro determination of the capsules opening time was performed in simulated gastric fluid without enzyme with USP Apparatus 2. The extent of absorption was similar between both products for all subjects or each sex-group. The absorption rate was similar for both products when considering all subjects, whereas a significant difference in the TMAX between the two products was observed for the male subjects only, which relates to its slower capsule opening time observed in vitro (12.4 versus 6.0 min). No differences were observed in women that could relate to their slower gastric emptying. Differences in drug exposure were observed between smokers and non-smokers. Sex- and smoke-related differences in the gastrointestinal transit should be considered when the on-set time would be determinant for the treatment success of a drug. On this work results from an average bioequivalence study between tablets containing 600 mg of the antiretroviral efavirenz (EFV), carried out with 14 healthy subjects (8 female and 6 men) in a randomized 2-period, 2-treatment crossover design, are analyzed from a sex-based approach. Sequences were balanced within each sex group. Considering all subjects, no differences were observed on EFV absorbed amount, as shown by the estimated 90CI of the AUC96 Test/Reference bioequivalence ratio (T/R): 0.950–1.05. However, results were not conclusive due to the 90CI for CMAX T/R was 0.743–1.07. Over this parameter, a significant sex-by-formulation interaction was detected: 90CI CMAX T/R was 0.838–1.36 in women and 0.540–0.920 in men; with a 52% relative difference between point estimates. Formulation differences were therefore evidenced only by male subjects. In vitro dissolution and disintegration tests for both products were carried out in two aqueous media: A) SLS 0.25% and B) HCl/KCl pH 1.2. T/R results for dissolution efficiency and tablet disintegration times of formulations in both A and B media were highly correlated with CMAX T/R bioequivalence results observed in women and men respectively, showing that a dissimilar gastrointestinal environment between sexes affected EFV oral absorption. This work shows how sex-by-formulation interaction can affect bioequivalence conclusions. Sex effect on product discrimination should be specially disclosed in bioequivalence studies, mainly for drugs aimed to be given to both sexes. Background: The goal of this study was to develop an in vivo-in vitro (IVIV) correlation, both in men and women, which allows constructing a model to predict bioequivalence assessments for drugs with narrow absorption windows. Besides, pharmacokinetic and pharmacodynamic equivalences were also investigated. Furosemide was chosen as a prototype. Methods: Twelve healthy Caucasian volunteers (8 women and 4 men) participated in a relative bioavailability study. Two oral formulations [Lasix® (Reference, R) and Furosemide EFA® (Test, T)] were administered under fasting conditions. Urinary excretion of unchanged drug (PK), and of chloride, sodium and potassium (PD) wasmonitored throughout time. PK and PD parameters were calculated from each respective excretion rate versus time curve. In vitro dissolution testing of both formulations was carried out using the USP apparatus 2 and 4 with fixed and variable dissolution media. Results: T and R could be considered bioequivalent since the 90% confidence intervals for the T/R ratio of geometric means for the area under the urinary drug excretion rate versus time curve and for the maximum excretion rate were within the 0.80-1.25 bioequivalence interval. However, T had faster initial absorption and higher levels in women, while R displayed such characteristics in men. Closer IVIV correlations in women were obtained when apparatus 4 with variable biorelevant dissolution media were used [going from fasting state simulated gastric fluid to fasting state simulated intestinal fluid]. Since R had faster disintegration time than T, a shorter stay of R under gastric conditions was required in order to obtain a good IVIV correlation in men. Saluretic effect displayed a typical clockwise hysteresis loop for the PKPD correlation assessed through chloride-versus-furosemide urinary excretion rates. Even though a higher amount of furosemide was excreted with the urine in men, differences in the excretion of electrolytes between sexes were almost negligible. Conclusions: Sex-differences in the gastrointestinal transit of formulations, under fasting conditions, determined the extent and the rate of furosemide absorption. The prolongation of the absorption process by mean of slowing the gastric emptying would make the formulation more effective. The USP-4 apparatus with variable dissolution media was able to discriminate the formulations even between sexes, becoming a promissory in vitro dissolution testing to predict bioequivalence. The arctiid moth Utetheisa ornatrix is protected against predation by pyrrolizidine alkaloids (PA) that it sequesters as a larva from its food plant. Earlier work had shown that males transmit PA to the female with the sperm package and that the female bestows part of this gift on the eggs, protecting these against predation as a result. We now show that the female herself derives protection from the gift. Females deficient in PA are vulnerable to predation from spiders (Lycosa ceratiola and Nephila clavipes). If mated with a PA-laden male, the females become unacceptable as prey. The effect takes hold promptly and endures; females are unacceptable to spiders virtually from the moment they uncouple from the male and remain unacceptable as they age. Chemical data showed that the female allocates the received PA quickly to all body parts. We predict that other instances will be found of female insects being rendered invulnerable by receipt of sexually transmitted chemicals. The precursor, Sgc8-c-NOTA, the radio probe Sgc8-c-NOTA-67Ga, and its nonradioactive complex, Sgc8-c-NOTA-69/71Ga, were purified by reverse-phase high-performance liquid chromatography and characterized by electrospray ionization mass spectrometry. The binding ability of Sgc8-c-NOTA-67Ga was studied in vitro against purified PTK7 receptor. In addition, the binding was also evidenced against the hemato-oncological A20 cell line, derived from B lymphocytes, and the corresponding A20-green fluorescent protein (GFP)-transfected cells. The proof of concept was performed on A20-GFP tumor-bearing mice, in which the biodistribution of the radiolabeled probe was evaluated through imaging, using X-ray, fluorescence, and γ modalities. The specific uptake of the probe was confirmed by blocking with the Sgc8-c aptamer in an in vivo competition assay. Results: The biodistribution results showed considerable uptake in tumor since 2 h, with highest at 48 h postinjection. However, the blood and muscle ID/g (injected dose per gram of tissue) activities were decreasing with time and tumor/no-target ratios increasing to 20 at 24 h postinjection. These results are consistent with the in vivo images. Conclusions: This study supports the utility of Sgc8-c-NOTA radiolabeled as a theranostic agent. In this study three different approaches—acceptability limit, failure cut-off point methodology, and survival analysis—to estimate the sensory shelf life of brown pan bread elaborated with different enzymes using consumer data were compared. The study was carried out in Spain and Uruguay independently, where four batches of bread were produced, with the same base formulation, one without enzyme, the others with the addition of maltogenic amylase, xylanase, and a mixture of both. The results showed that for Uruguayan consumers the mixture of enzymes provided better results in extending shelf life than the addition of amylase alone. For the Spanish consumers only the addition of amylase provided the desired results, and for consumers of both countries the use of xylanase did not extend the shelf life of bread. Among the methodologies used for the estimation, survival analysis provided the most adequate predictions considering consumer rejection of the product. Hedonic scales do not always reflect consumer behaviour when deciding whether to accept or reject a certain product for its consumption.
