A summary of the resource.
A bioequivalence study of carbamazepine in two conventional tablets commercially available in Uruguay was carried out in eight healthy volunteers using a crossover design. Saliva samples were collected at appropriate times |b and the samples were analyzed by high performance liquid chromatography (HPLC). The statistical evaluations of the pharmacokinetics parameters showed no significant differences |b either for the extent of absorption (AUC 0-48) or for the rate of absorption (as indicated by Cmax and Tmax) between both formulations. Significant differences were found between salivary concentrations 20 min.after the ingestion between both formulations. This was in agreement with the in vitro dissolution profiles.The mean elimination half-life for carbamazepine was found to be 28 hours+-11. Both formulations were bioequivalent according to Westlake's confidence interval of 20. It may be concluded from these results that saliva is useful for bioequivalence studies of carbamazepine using an appropriate sampling schedule.

A summary of the resource.
Inter- and intraindividual variability in the relationship between dose and clinical – or pharmacodynamic – response of a drug can be analysed in two steps: firstly, by considering the plasma pharmacokinetic response to a given dose and, secondly, by the connection between both pharmacokinetic and pharmacodynamic responses. As the cardiovascular system is the means of transport of endogenous and exogenous substances, blood flow fraction destined to each organ determines the relative mass of solute in plasma, which is constantly in contact with the tissue. Hence, not only the rate but also the extent of drug transfer would be increased when tissues are irrigated by a higher fraction of cardiac output. Aging and circadian rhythms present similar cardiac output distribution patterns when moving from young to aged adult and from nocturnal to diurnal hours. These two changes lead to an increased blood flow delivery to the extra-splanchnic-renal region in the elderly and in the morning, but with a decreased cardiac output in aged individuals and an increased one during the day. This scenario allows us to forecast substance concentrations outside the blood vessels, which are responsible for the extent of drug elimination and the intensity

Los mayores obstáculos para la toma de decisiones en bioequivalencia promedio están referidos al tamaño de la variabilidad residual. Se ha visto que en ella incide mucho las características de los individuos reclutados para el ensayo. El presente trabajo propone una metodología que estudia la varianza residual según el género de los voluntarios y construye intervalos de confianza para el cociente de medias Test / Referencia (T/R), de los parámetros de biodisponibilidad, tanto en hombres como en mujeres. Asimismo se analizan las varianzas totales producidas por los medicamentos T y R, a los efectos de complementar la toma de decisiones. Las anteriores pautas fueron aplicadas a 3 estudios de bioequivalencia llevados a cabo con fármacos sustratos de la isoenzima CYP3A4 (itraconazol, alprazolam, claritromicina), algunos de los cuales son también sustratos del transportador de eflujo P-glicoproteína. Las mujeres presentaron menor exposición a los fármacos sustratos de CYP3A4 que los hombres. Los medicamentos T y R, conteniendo tales fármacos, mostraron biodisponibilidades relativas dependientes del sexo de los voluntarios, principalmente a causa de probables diferencias de disolución, originadas por la diferente fisiología del tracto digestivo que presentan hombres y mujeres. Como conclusión de este trabajo se entiende que la toma de decisiones en bioequivalencia debería tener en cuenta los resultados obtenidos en cada género, para apoyar tanto que T y R sean bioequivalentes como que sean bioinequivalentes

Valproic acid, a branched short-chain fatty acid, has numerous action mechanisms which turn it into a broad spectrum anticonvulsant drug and make its use possible in some other pathologies such as bipolar disorder. It is extensively metabolized in liver, representing β-oxidation in the mitochondria one of its main metabolic route (40%). Carnitine is responsible for its entry into the mitochondria as any other fatty acid. Long-term high-dose VPA therapy or acute VPA overdose induces carnitine depletion, resulting in high levels of ammonia in blood. As a high correlation between salivary valproic acid levels and plasma ultrafiltrate levels was found in humans, saliva becomes a promising monitoring fluid in order to study valproic acid pharmacokinetics and its toxic effect. Extended-release (twice daily) formulations of valproic acid or carnitine supplementation are the proposed two therapeutic strategies in order to reverse hyperammonemia.

A summary of the resource.
Nevirapine (NVP) extensive data obtained after oral single dose administration of 200 mg in a crossover study involving 16 healthy subjects was used to develop a descriptive population pharmacokinetic model including drug recirculation, since secondary peaks were observed in plasma concentration-time profiles for all subjects. Through implementation of model event time feature in NONMEM 7.3.0, a simple mechanistic model physiologically consistent with the process of drug cycling was able to describe multiple peaks phenomena and quantify its pharmacokinetic parameters, achieving a better performance than its analogue conventional one. Absorption process, between subject and-between occasion variability of pharmacokinetic parameters was also assessed. Estimated mean fraction of NVP bioavailable dose undergoing recirculation process was 51.6 %, a magnitude which could hardly be explained by enterohepatic cycling. In this work we propose an alternative disposition process to explain NVP drug recirculation: gastric secretion with posterior intestinal reabsorption. Due to physicochemical and pharmacokinetic properties of the drug, this neglected phenomenon is more likely to explain the results obtained, and might be present in disposition of several basic drugs.

Se propone un modelo farmacocinético compartimental (SIMULFIS) en donde se consideran los flujos sanguíneos del organismo. Aplicado a la Digoxina, se estudiaron las consecuencias farinacocinéticas y los niveles plasmáticos y tisulares de la droga, al alterar el gasto cardíaco y los flujos sanguíneos en el cuerpo humano. Se analiza en base al modelo, la interacción Digoxina-Amiodarona. Los resultados de las simulaciones demuestran un aumento de la captación tisular y disminución de la concentración plasmática de Digoxina durante el ejercicio físico. El modelo asume un único mecanismo en la interacción Digoxina-Amiodarona, que explica el aumento de la biodisponibilidad oral, la disminución de la eliminación renal y hepática, y el inalterado volumen de distribución de la Digoxina.

Saliva and plasma profiles of Carbamazepine and Phenytoin were studied in healthy volunteers |b after the administration of a controlled-release tablet (Tegretol 400 mg) |b three conventional-release tablets (Comitoina Simple 100 mg) and a W/O emulsion of Phenytoin. Samples were analysed by High Performance Liquid Chromatography on reversed phase column C18 |b 5 micra |b 10 cm X 4 mm i.d.,mobil pha<se water/methanol/acetonitrile (50:45:5) |b flow rate 1 ml/min |b detection at 22onm. Salivary and plasma levels showed a lack of correlation |b evidenced through the variable saliva/plasma (S/P) ratios. The in vivo release process is well recorded by the S/P ratios |b which are higher during absorption periods. The technological development of pharmaceuticals destined for extavascular route |b can be evaluated in vivo by these S/P ratios. Constant S/P values related with the plasma unbound fraction of drug state absence of absorption process |b and allow the estimation of pharmacokinetic elimination parameters from time-concentration data.

SUMMARY<br />According to the literature, cyclosporine in healthy volunteers yielded significant lower clear-<br />ances than in patients with renal failure, uremic, or with kidney transplant. This could be due to a de-<br />creased renal blood flow these patients have and the negligible renal metabolism cyclosporine exhibits.<br />However, in the case of tacrolimus, literature shows almost the same clearance in healthy volunteers and patients with renal impairment. This fact might be related with its dual ability of being eliminated,<br />splanchnic and renal metabolism, and the relevant impact of CYP3A5 polimorfism. Eighty eight kidney<br />transplant recipients under tacrolimus treatment were enrolled. Whole blood tacrolimus levels were determined and evolution of apparent clearance with serum creatinine was assessed. In some patients an increase in serum creatinine produced an increase in tacrolimus clearance, whereas others displayed an inverse effect with increasing serum creatinine. This fact gives new evidence about the impact renal metabolism would have on tacrolimus pharmacokinetics.

A summary of the resource.
Objetivo: realizar una actividad de difusión a la población sobre el uso racional de medicamentos, mediante la inserción de los estudiantes en el equipo de salud y analizar los datos farmacoterapéuticos obtenidos de esta actividad educativa. Métodos: en la vía pública se instaló una carpa de Atención Farmacéutica durante<br />tres días donde participaron químicos farmacéuticos y médicos, quienes impartieron<br />charlas sobre diferentes temas de salud preponderantes en la población uruguaya.<br />Asimismo, los alumnos de Atención Farmacéutica, como parte de su formación práctica, participaron de esta actividad mediante la realización de entrevistas a los transeúntes, llenado de fichas de perfiles farmacoterapéuticos y de consumo de<br />plantas medicinales, elaboradas para este fin, y la entrega de folletos informativos.<br />A partir de las fichas farmacoterapéuticas se desarrolló un trabajo de investigación.<br />Resultados: los alumnos del curso participaron de forma activa en el llenado de las fichas farmacoterapéuticas y mantuvieron una comunicación fluida con los asistentes a la carpa y con los profesionales de la salud. Se completaron 117 fichas farmacoterapéuticas (90 mujeres y 27 hombres). El 60 % de los entrevistados<br />consumía plantas medicinales. Sesenta personas recibían cuatro o más especialidades farmacéuticas. Las drogas antihipertensivas resultaron las más utilizadas. Veintitrés personas presentaban hipotiroidismo y dos personas de este<br />grupo recibían litio para trastorno bipolar. Entre el grupo de mujeres: 18 tomaban<br />ansiolíticos, 12 antidepresivos, 7 hipnóticos y 2 antisicóticos. Diecinueve personas<br />manifestaron tener colesterol alto y 14 recibían medicación. Catorce presentaban<br />artrosis y 10 estaban en tratamiento con analgésicos. Nueve personas presentaban

In this report, we prepare LLTO ceramics by the sol-gel method in acetic medium. Raman spectroscopy showed the formation of lanthanum and titanium acetates precursors, which after calcination, lead to formation of the LLTO nanoparticles. Raman spectra were scanned directly over the LLTO pellets and the disappearance of impurities was observed during the microstructure evolution with increasing sintering temperature. X-ray diffraction characterization, including full pattern profile fitting refinements, showed no drastic changes in the unit cell parameters of the LLTO perovskite, but a large increase in the crystallite size domain was observed with increasing sintering temperature. Additionally, an interesting structural phase transition for the Li0.30La0.57TiO3 perovskite structure was observed, from tetragonal P4/mmm to distorted-cubic Pm-3m spacegroup, for the highest sintering temperature (Ts=1300 °C). Experimental and theoretical simulations of Raman spectroscopy confirmed the formation of a distorted-cubic phase and confocal Raman spectroscopy showed the presence of traces of impurities at the grain boundary region. In spite of the low total lithium conductivity observed, the electrochemical impedance spectroscopy analysis showed a remarkable increase in the lithium bulk conductivity for Ts=1300 °C. This fact could be attributed to the structural phase transition from tetragonal to the cubic crystal system.

Lead exposure remains an important public health problem. Contaminated foods may act as a source of lead exposure, while certain nutrients may reduce lead absorption. We examined the cross-sectional associations of dietary patterns and the intake of several nutrients and foods with blood (Pb-B) and urinary (Pb-U) lead concentrations in children (5–8 y) from Montevideo, Uruguay. From two 24-hour recalls completed by caregivers, we derived the mean daily intake of select nutrients and food groups (dairy, milk, fruit, root vegetables, foods rich in heme and non-heme iron), as well as “nutrient dense” and “processed” food patterns. Pb-B (n = 315) was measured using atomic absorption spectrometry; Pb-U (n = 321) using ICP-MS. Pb-U was adjusted for specific gravity and log-transformed to approximate a normal distribution. Iron deficiency (ID) and dietary variables were tested as predictors of Pb-B and log-Pb-U in covariate-adjusted regressions. Median [5%, 95%] Pb-B and Pb-U were 3.8 [0.8–7.8] μg/dL and 1.9 [0.6–5.1] μg/L, respectively; ~ 25% of Pb-B above current U.S. CDC reference concentration of 5 μg/dL. ID was associated with 0.75 μg/dL higher Pb-B, compared to non-ID (p < 0.05). Consumption of root vegetables was not associated with Pb-B or log-Pb-U. Higher scores on the nutrient-dense pattern were related with higher Pb-Bs, possibly due to consumption of green leafy vegetables. Dietary intake of iron or iron-rich foods was not associated with biomarkers of lead. Conversely, children consuming more calcium, dairy, milk and yogurt had lower Pb-B and log-Pb-U. Our findings appear consistent with existing recommendations on including calcium-rich, but not iron- or vitamin-C-rich foods in the diets of lead-exposed children, especially where the consumption of these foods is low.

1,4-Thiazepines derivatives are pharmacologically important heterocycles with different applications in medicinal chemistry. In the present work, we describe the preparation of new bicyclic thiazolidinyl-1,4-thiazepines 3 by reaction between azadithiane compounds and Michael acceptors. The reaction scope was explored and the yields were optimized. The activity of the new compounds was evaluated against Nippostrongylus brasiliensis and Caenorhabditis elegans as anthelmintic models and Trypanosoma brucei brucei. The most active compound was 3l, showing an EC50 = 2.8 ± 0.7 μM against T. b. brucei and a selectivity index >71.

Abstract
A summary of the resource.
The Cu(I)-catalyzed Huisgen [3+2] cycloaddition is the basis of click chemistry to synthesize triazole derivatives by coupling azides with ethynyl blocks. In the development of new compounds inspired by bioactive natural products, the synthesis of new oxazole building blocks containing azide moiety and coupling them with aromatic alkynes via triazole linker is described. These oxazole building blocks are synthesized using amino acids as chiral and inexpensive starting materials. Using this approach, 16 new triazole-oxazole hybrids were synthesized.

Abstract
A summary of the resource.
The induction periods for the peroxidation of various fish oils at 55–90°C were studied by the Rancimat test. The natural logarithms of the induction periods varied linearly with respect to temperature, with a mean coefficient of −7.5×10−2°C−1, which was significantly different from that reported for vegetable oils. The activation energy for the formation of volatile acids had a mean value of 38.9 kJ/mol and was independent of the fish oil source. Peroxide formation under Rancimat test conditions followed first-order kinetics. The same kinetics were followed under Schaal Oven test conditions (forced-air oven, 60°C). On the basis of the results obtained, the Rancimat test appears to be useful in determining the relative stabilities of fish oils without the change in peroxide decomposition kinetics that may occur at elevated temperatures.

Abstract

Hanseniaspora species can be isolated from grapes and grape musts, but after the initiation of spontaneous fermentation, they are displaced by Saccharomyces cerevisiae. Hanseniaspora vineae is particularly valuable since this species improves the flavour of wines and has an increased capacity to ferment relative to other apiculate yeasts. Genomic, transcriptomic, and metabolomic studies in H. vineae have enhanced our understanding of its potential utility within the wine industry. Here, we compared gene sequences of 12 glycolytic and fermentation pathway enzymes from five sequenced Hanseniaspora species and S. cerevisiae with the corresponding enzymes encoded within the two sequenced H. vineae genomes. Increased levels of protein similarity were observed for enzymes of H. vineae and S. cerevisiae, relative to the remaining Hanseniaspora species. Key di�erences between H. vineae and H. uvarum pyruvate kinase enzymes might explain observed di�erences in fermentative capacity. Further, the presence of eight putative alcohol dehydrogenases, invertase activity, and sulfite tolerance are distinctive characteristics of H. vineae, compared to other Hanseniaspora species. The definition of two clear technological groups within the Hanseniaspora genus is discussed within the slow and fast evolution concept framework previously discovered in these apiculate yeasts.

La formación de peroxinitrito en la mitocondria está favorecida por el suministro constante de radical superóxido a partir de la cadena de transporte electrónico mitocondrial y la difusión de óxido nítrico que alcanza a este organelo. La mayor parte de este trabajo fue realizado en un modelo de partículas submitocondriales (PSM), en presencia de succinato y un dador de óxido nítrico, el NOC-­‐7, de forma tal de estudiar la formación y cuantificar al peroxinitrito formado por una multiplicidad de métodos incluyendo fluorescencia, quimioluminiscencia y análisis inmunoquímicos. Mientras que todas las sondas que fueron evaluadas revelaron niveles casi estequiométricos de peroxinitrito respecto a sus radicales precursores, el ácido cumarin borónico (un compuesto que reacciona directamente con peroxinitrito) fue el que mostró un perfil de oxidación más consistente en las PSM formando radical superóxido, en función del flujo de óxido nítrico. Un resultado interesante surgió de los estudios inmunoquímicos que verificaron la generación de radicales proteicos en PSM inducida por peroxinitrito. En otro tipo de enfoque, utilizamos un análogo hidrofóbico de tirosina (N-­‐t-­‐BOC L-­‐tirosina tert-­‐ butil éster, BTBE) pre-­‐incorporado en mitocondrias y PSM, el cual se nitra y oxida por peroxinitrito. En las PSM conteniendo BTBE y formando radical superóxido, la exposición a flujos de óxido nítrico, causó la oxidación y posterior dimerización de la sonda en conexión con procesos de lipoperoxidación. Las PSM suplementadas con sustratos respiratorios redujeron a las Mn(III)porfirinas (MnP) a Mn(II)P, a niveles de oxígeno fisiológicamente relevantes (3-­‐30 μM); a continuación, las Mn(II)P fueron capaces de reducir al peroxinitrito a nitrito y proteger las PSM de la inhibición del consumo de oxígeno por complejo I, de la formación de radicales proteicos y de la nitración de proteínas de membrana. La pre-­‐ incubación de MnP en células endoteliales en cultivo protegió de la nitración de tirosinas proteicas mediada por peroxinitrito a partir de exposición a SIN-­‐1. A través de miscroscopía confocal Raman Resonante detectamos directamente a las MnP dentro de las células, tanto en forma reducida como oxidada; el predominio observado de la forma reducida es totalmente consistente con su rol citoprotector en procesos nitroxidativos. Globalmente, los datos apoyan la formación de peroxinitrito en mitocondrias sometidas a flujos óxido nítrico y demuestran que las MnP pueden participar de un ciclo catalítico redox a nivel mitocondrial para neutralizar el daño oxidativo dependiente de peroxinitrito.

Abstract
A summary of the resource.
An extract possessing proteolytic activity was obtained by grinding the ripe fruit Solanum granuloso-leprosum, a tree of the family Solnaceae. The crude extract had maximum activity at pH between 5.2 and 7.6, and temperatures between 50°C and 55°C. It was stable at pH 7.6 and freezing temperature. The proteolytic activity of this plant product was activated and stabilised by reducing agents. Purification was carried out using simple and economical procedures, including ethanolic precipitation and gel-filtration chromatography. The main proteolytically active fraction obtained was inhibited by iodoacetamide and E-64, indicating its cysteinic nature. This fraction had a pI range of 7.5–8.3. The enzyme preparation was unstable at temperature above 4°C, but was stabilised by adsorption onto alumina. Its hydrolytic action was studied on casein whey and haemoglobin substrates, which represent ecologically contaminating fractions of industrial waste.
