Amidasic activity was studied with pyroglutamyl-phenyl-leucyl-paranitroaniline (PFLNA) substrate having higher AntA catalytic efficiency of (kcat/Km=470s-1M-1) relative to stem bromelain (kcat/Km=305s-1M-1). Esterase activity using p-nitrophenyl esters of N-α-CBZ-l-Lysine (z-L-LysONp) showed a 10-fold higher catalytic efficiency for AntA (kcat/Km=6376s-1M-1) relative to stem bromelain (kcat/Km=688s-1M-1). Incubation with 8M Urea did not affect AntA activity and remained unchanged for 18h, with 6M GndHCl resulted in a 41% decrease in activity after 30min incubation, maintained this activity 18h. Crude extracts were partially purified by organic solvents fractionation: best results (96% of proteins, 91% of total caseinolytic activity) were obtained by adding four volumes of cold acetone to the crude extract. This preparation (redissolved acetone precipitate, RAP) showed maximum activity (>80%) at pH 5–9, and exhibited high thermal stability (>90% of residual activity after heating for 60 min at 60 ◦C). The enzyme was completely inhibited by E-64 trans-epoxysuccinyl-leucyl-amido(4-guanidino)-butane and iodoacetic acid and activated by the addition of cysteine or �-mercaptoethanol; these results strongly suggest that the isolated protease should be included within the cysteine group, as all the other studied proteases belonging to the family Bromeliaceae. IEF-zymogram of RAP showed five bands (pI 7.3 to <9.3), most of them proteolytically actives, but only three of which (pI 7.6, 8.2 and 8.8) proved to be important. Ralstonia solanacearum is the causative agent of bacterial wilt of potato. Ralstonia solanacearum strain UY031 belongs to the American phylotype IIB, sequevar 1, also classified as race 3 biovar 2. Here we report the completely sequenced genome of this strain, the first complete genome for phylotype IIB, sequevar 1, and the fourth for the R. solanacearum species complex. In addition to standard genome annotation, we have carried out a curated annotation of type III effector genes, an important pathogenicity-related class of genes for this organism. We identified 60 effector genes, and observed that this effector repertoire is distinct when compared to those from other phylotype IIB strains. Eleven of the effectors appear to be nonfunctional due to disruptive mutations. We also report a methylome analysis of this genome, the first for a R. solanacearum strain. A rapid and sensitive liquid chromatography–tandem mass spectrometry method, in electrospray ionization positive mode, has been developed for the determination of 160 selected multi-class pesticides over a 33-min run time. Extracts were obtained using the acetonitrile-based QuEChERS (quick, easy, cheap, effective, rugged and safe) sample preparation technique. The validation study was carried out on tomato, pear and orange matrices following DG SANCO/2007/3131 of the European Quality Control Guidelines. These matrices represent high water, high sugar and high acidic content commodities, respectively. Matrix influence on recoveries and its effects on ionization were evaluated for the three matrices. Ten out of the 160 pesticides showed very low intensity, linearity and/or sensitivity problems. Linearity was studied in the 5–500 μg kg−1 concentration range. Soft (50%) matrix effects were obtained for 69%, 20%, and 11% of the studied compounds, respectively. Recoveries were investigated at the 10 and 100 μg kg−1 levels, and depending on the commodity, 97%, 98% and 97% of the compounds in tomato, pear and orange, respectively, were in the 70–120% range. More than 90% of the investigated compounds had less or equal to a 5 μg kg−1 limit of detection in the studied matrices. The relative standard deviations obtained exceeded 20% in only very few cases. The overall standard deviation obtained in the survey study (0.1551) was used for the method's uncertainty estimation. The expanded uncertainty resulted as being 0.3002 (coverage factor K = 2, confidence level 95%). The method was applied on 59 real samples from 14 different kinds of fruits and vegetables. Thirty-three compounds were detected in 50 positive samples. Baccharis dracunculifolia, “Vassoura”, is a shrub with spontaneous occurrence in Brazil, Uruguay, Argentina, Paraguay, and Bolivia. The Vassoura oil, valuable in the perfumery industry for its exotic aroma, presents variations in composition depending on the geographical origin and extraction process. The commercial importance is related to the oxygenated fraction content, specifically to (E)-nerolidol and spathulenol. In this work we study the dependence of the Vassoura oil composition, obtained by supercritical CO2 (SCCO2) extraction, with the following parameters: pressure, temperature, SCCO2 flow, equilibrium, and extraction times. The flexibility of the extraction process using SCCO2 allowed one to study the Vassoura oil composition as a function of the extraction parameters. Whilst several are understood, how they act co-operatively in pathogenic strains is less clear. In some isolates, resistance profiles cannot always be explained by identifying the common resistance-determining pathways, suggesting that other mechanisms may be important. Pathogenic P. aeruginosa isolates from four countries were characterised by PCR. Quantitative expression analysis was also assessed for the activity of several pathways that influence antibiotic resistance, and culture experiments were conducted to test how random transposition of the insertion sequence IS26 during growth may influence resistance to some antibiotics. In most strains, antibiotic resistance was being driven by changes in multiple pathways and by the presence or absence of genes acquired by lateral gene transfer. Multiple mechanisms of resistance were prevalent in strains from all of the countries examined, although regional differences in the type of interacting mechanisms were apparent. Changes in chromosomal pathways included overexpression of AmpC and two efflux pumps. Also, gain or loss of IS26 at some chromosomal locations, most notably oprD, could influence resistance to carbapenems. IS26-related resistance was found in strains from Argentina and geographically linked Uruguay, but not in strains from either Colombia or Australia. Pseudomonas aeruginosa pathogenic strains are evolving to become multidrug-resistant in more complex ways. Global warming is causing serious problems, especially, in warm regions, where musts with excess sugars and high pH produce wines with decreased freshness and unstable evolution. This study aimed to determine biocompatibility between yeast species, the capacity for microbiological acidification, and the aromatic profile produced in ternary fermentations in which Lachancea thermotolerans has been co-inoculated with Hanseniaspora vineae, Torulaspora delbrueckii, or Metschnikowia pulcherrima, and the fermentation process is subsequently completed with sequential inoculation of Saccharomyces cerevisiae. For this purpose, different cell culture media and instruments were used such as infrared spectroscopy, enzymatic autoanalyzer, chromatograph coupled with a flame ionization detector, spectrophotometric analysis, among others. The behavior of these yeasts was evaluated alone and in co-inoculation, always finishing the fermentation with sequential inoculation of S. cerevisiae, at a stable temperature of 16°C and with a low level of sulfites (25 mg/L) in white must. Significant results were obtained in terms of biocompatibility using population counts (CFU/ml) in differential plating media that permitted monitoring. Quantification of the five species was studied. Concerning acidification by L. thermotolerans in co-inoculations, we showed some metabolic interactions, such as the inhibition of acidification when H. vineae/L. thermotolerans were used, generating just over 0.13 g/L of lactic acid and, conversely, a synergistic effect when M. pulcherrima/L. thermotolerans were used, achieving 3.2 g/L of lactic acid and a reduction in pH of up to 0.33. A diminution in alcohol content higher than 0.6% v/v was observed in co-inoculation with the L. thermotolerans/M. pulcherrima yeasts, with total sugar consumption and very slow completion of fermentation in the inoculations with H. vineae and T. delbrueckii. The aromatic composition of the wines obtained was analyzed and a sensory evaluation conducted, and it was found that both L. thermotolerans and co-inoculations retained more aromatic esters over time and had a lower evolution toward the yellow tones typical of oxidation and that the best sensory evaluation was that of the Lt + Mp co-inoculation. Lachancea thermotolerans and co-inoculations produced wines with low levels of volatile acidity (<0.4 g/L). This work shows that good consortia strategies with binary and ternary fermentations of yeast strains can be a powerful bio-tool for producing more complex wines. Ochenta y dos plantas silvestres se recolectaron de diversos orígenes geográficos y en diferentes épocas del año que se han caracterizado botánicamente y sus extractos etanólicos se evaluaron frente a la forma epimastigote de Trypanosoma cruzi. Las plantas pertenecen a las siguientes especies: Hydrocotyle bonariensis, Baccharis trimera, Baccharis articulata, Baccharis usterii, Taraxacum officinale, Plantago major y Achyrocline satureioides. Se encontró que las muestras de Hydrocotyle bonariensis, Baccharis trimera, Baccharis articulata y Baccharis usterii presentan significativa actividad anti-proliferativa. También se evaluó la actividad citotóxica en células mamíferas de los extractos con relevante actividad antiparasitaria para la determinación de la selectividad de los mismos. Se observó que las tres especies de Baccharis eran las que presentaban mejores selectividades. Los extractos que resultaron más activos y selectivos se evaluaron frente a amastigotes, la forma intracelular del parásito, encontrándose una muy buena correlación de actividades. Se observó que la actividad anti-Trypanosoma cruzi varía entre muestras de distinto tipo de suelo, por lo que se buscó determinar en cuales condiciones estas especies presentan mayor actividad biológica. También se encontró que en las diferentes estaciones climáticas de nuestra región las plantas modifican su metabolismo secundario produciendo diferentes compuestos químicos en cantidades también variables. El análisis de los cambios en el perfil metabólico de las plantas se realiza mediante la aplicación de la técnica de metabolómica por Resonancia Magnética Nuclear. Esta técnica permitió identificar los principios activos anti-Trypanosoma cruzi sin previo aislamiento de los mismos. A su vez fue posible identificar las condiciones óptimas para la máxima expresión de los principios activos mediante la aplicación de métodos quimiométricos con la información obtenida en metabolómica por RMN. Utilizando los datos obtenidos mediante metabolómica y realizando el fraccionamiento bioguiado se elucidó la estructura del principio activo de Baccharis articulata, 7-hidroxi-ent-clerodan-3-en-l6-lacton-18-al. También se realizó la estandarización del cultivo de Baccharis spp. mediante clonación por esqueje, donde se observó que la técnica empleada es eficaz para la multiplicación y estandarización de cultivos de Baccharis trimera y Baccharis usterii. The flavor of eight samples of commercial strawberry yogurt was studied by Free-Choice Profile analysis (FCP). Generalized Procrustes Analysis (GPA) applied to FCP allowed differentiation between samples and highlighted flavor attributes responsible for the observed differences. The relation between sensory and physicochemical datasets was studied by means of GPA. Those samples with higher carbohydrate content were perceived as sweeter, having stronger strawberry flavor, and with more dairy and yogurt flavors. In this study three different approaches—acceptability limit, failure cut-off point methodology, and survival analysis—to estimate the sensory shelf life of brown pan bread elaborated with different enzymes using consumer data were compared. The study was carried out in Spain and Uruguay independently, where four batches of bread were produced, with the same base formulation, one without enzyme, the others with the addition of maltogenic amylase, xylanase, and a mixture of both. The results showed that for Uruguayan consumers the mixture of enzymes provided better results in extending shelf life than the addition of amylase alone. For the Spanish consumers only the addition of amylase provided the desired results, and for consumers of both countries the use of xylanase did not extend the shelf life of bread. Among the methodologies used for the estimation, survival analysis provided the most adequate predictions considering consumer rejection of the product. Hedonic scales do not always reflect consumer behaviour when deciding whether to accept or reject a certain product for its consumption. As a case study, fifty consumers performed a projective mapping task with ten breakfast cereal packages while wearing a mobile eye-tracker. The combination of mobile eye-tracking and projective mapping enabled a more comprehensive analysis of the importance of package attributes for consumer perception. Eye tracking allowed the identification of the most relevant package features for perceived similarity and differences among products and spotted attributes that were attended to but were not relevant, as well as package features that were relevant for categorization but were not largely attended to. Results suggest that studying attentional capture could contribute to better understanding attribute importance for consumer perception. Irrespectively of the saliency, most consumers looked at the same key information, mainly located on the front-of-pack. Few consumers read the nutritional label and ingredient list (a much lower proportion than in previous static eye tracker studies). Results suggested that mobile eye-tracking has a great potential for assessing consumers' evaluation of packages in ecological settings. However, several disadvantages and limitations of the technique should be taken into account. The aim of the present work was to study the influence of brand on consumers’ expectations and liking of orange-flavoured powdered drinks. Ten samples of orange-flavoured powdered drinks were evaluated by 108 consumers who scored their expected liking (looking at the pack only) and the overall liking under blind and informed-testing conditions (tasting the products). Consumers were also asked to answer a check-all-that-apply question with eight statements related to some products’ characteristics and usage occasions. Results showed that brand and package information had a large impact on consumers’ liking scores only for well-recognized brands.
