Entomopathogenic fungi such as Beauveria bassiana are extensively used for the control of insect pests worldwide. They infect mostly by adhesion to the insect surface and penetration through the cuticle. However, some insects, such as the red flour beetle Tribolium castaneum (Herbst), have evolved resistance by embedding their cuticle with antifungal compounds. Thus, they avoid fungal germination on the cuticle, which result in low susceptibility to entomopathogenic fungi. In adult T. castaneum, these antifungals are the well-known defensive compounds methyl-1,4- and ethyl-1,4- benzoquinone. In this study, we added B. bassiana conidia on the diet of adult beetles to study the effect of the entomopathogen on the secretion and detection of the beetle volatile blend containing both benzoquinones. The compounds were analyzed by solid phase microextraction coupled to gas chromatography–flame ionization detection, and were detected by electroantennography. In addition, we measured the expression level of four genes encoding for two odorant-binding proteins (OBP), one chemosensory protein (CSP), and one odorant receptor (OR) in both healthy and fungustreated insects. Significant alterations in the secretion of both benzoquinones, as well as in the perception of methyl-1,4-benzoquinone, were found in fungus-treated insects. TcOBP7D, TcOBP0A and TcCSP3A genes were down-regulated in insects fed conidia for 12 and 48 h, and the latter gene was up-regulated in 72 h samples. TcOR1 expression was not altered at the feeding times studied. We conclude that fungus-treated insects alter both secretion and perception of benzoquinones, but additional functional and genetic studies are needed to fully understand the effects of fungal infection on the insect chemical ecology. Although several discrimination methods have been reported with children, the A-not-A test has not been studied yet. The aims of this work were to: (i) evaluate the feasibility of using the A-Not-A test with school aged children, and (ii) compare how the framing of the question (overall differences vs. differences in liking) may influence the results. A total of 126 children were involved in the study. They participated in three sessions, each composed of a familiarization task with a visual stimuli and sample tasting with one of three dairy products (vanilla milk desserts, chocolate-flavored milk, and vanillaflavored yogurt). Half of the children evaluated the samples in terms of overall differences and the other half in terms of differences in liking. Results from the familiarization step showed that children correctly identified the visual reference in the A-not-A test regardless of how the question was framed, suggesting that they were able to understand the methodology. In the case of tasting samples, children were significantly more likely to correctly identify the reference in two of the three studied dairy products, when the question was based on liking, as compared to the framing based on overall differences. Rice is an important source of methane (CH4) and other crops may be sources of nitrous oxide (N2O), both of which are powerful greenhouse gases. In Uruguay, irrigated rice rotates with perennial pastures and allows high productivity and low environmental impact. A long-term experiment with contrasting rice rotation intensification alternatives, including rice–soybean and continuous rice, was recently carried out in an Argialboll located in a temperate region of South America. To know if rotation systems influence soil microbial activity involved in CH4 and N2O emissions, the abundance and potential rate for gas production or consumption of microbial populations were measured during the rice crop season. CH4 was only emitted when rice was flooded and N2O emission was not detected. All rotational soils showed the highest rate for methanogenesis at tillering (30 days after rice emergence), while for methanotrophy, the maximum rate was reached at flowering. The abundance of related genes also followed a seasonal pattern with highest densities of mcrA genes being observed at rice flowering whereas pmoA genes were more abundant in dry soils after rice harvest, regardless of the rotation system. Differences were foundmainly at tillering when soils with two consecutive summers under rice showed higher amounts ofmcrA and pmoA gene copies. The potential denitrification rate was highest at the tillering stage, but the abundance of nirK and nirS genes was highest in winter. Regarding ammoniumoxidation, bacterial amoA abundance was higher in winter while the archaeal amoA gene was similar throughout the year. A strong influence of the rice growth stage was registered for most of the parameters measured in rice paddy soils in this no-till rice intensification experiment. However, differences among rotations begin to be observed mainly at tillering when the abundance of populations of the methane and nitrous oxide cycles seemed to respond to the rice intensification. Rice paddy soils are an important biogenic source of methane, a relevant greenhouse gas. Rice rotation with upland crops and pastures affects hydrolytic and fermentative bacteria and methanogenic archaea by sequential exposition to oxic and anoxic conditions. There are several ways to reduce methane emissions from rice paddy soils. Here we examine the effect of intensification in rice rotations with upland crops upon the microbial community structure and methanogenesis when dicarboxylic acids exudated by rice root were amended in microcosms assays. Soils under rotations rice-pasture, rice-soybean and continuous rice, with the last two more intensive systems recently installed in the field experiment, were sampled. Amplicon sequencing of 16S rRNA gene analysis showed that the rotation system was a main driver of the microbial community structure, explaining 33%, 38% and 55% of the differences in the structure of Bacteria, Archaea and methanogens, respectively. The density of mcrA gene copies was significantly higher in continuous rice soil than in other soils, and the relative abundance of methanogenic archaea was 33% for rice monoculture, whereas represented 15% and 14% for the rice-pasture and rice-soybean soils, respectively. The incubation of soils with tartrate and succinate confirmed functional differences among soils since rice monoculture showed similar or higher methane production and significantly less acetate and propionate accumulation than other soils. The T-RFLP analysis of mcrA gene in soils and incubated soils revealed that Rice Cluster I increased after incubation in all rotational soils, whereas Methanosarcinaceae and Methanobacteriaceae remained relevant after incubation. Furthermore, a more stable community of methanogenic archaea was established in soil under continuous rice, since T-RF profiles of soils and incubated soils with dicarboxylic acids grouped together with 62% similarity, while communities from other rotational soils experimented a great shift after incubation with these substrates. Altogether, these results showed that the conversion of rice-pasture rotational fields to a more intensive rice crop production strongly impacts the methanogenic community structure and their potential for methane emission. In the present study, we explored the reactivity of 5- hydroxymethylfurfural (5-HMF), a renewable platform chemical, as the aldehyde component in the Groebke-Blackburn-Bienaymé (GBB) multicomponent reaction. Given the limited stability of 5-HMF under conventional conditions used for this reaction, acidic milieu in combination with high temperature or long reaction times, we explored factors which influence the course of the reaction (solvent, catalyst, time, and temperature) within the guidelines of previously reported green reaction conditions. Mild conditions and short reaction times were achieved to obtain good conversion and selectivity values. The reaction scope was exemplified by preparing a small compound library based on the optimized conditions varying the amidine and the isocyanide component. The hydroxymethylfuran moiety present in the GBB products, imidazo[1,2-a]heterocycles, provides useful functionalization for further transformations, which could expand the diversity of the chemical space. The application of circular economy concepts has led to progress in production processes that use this residue as starting material. Propylene glycol has been identified as a potential destiny of glycerol surplus; however, the possibility of increasing this valorization from the existence of a stereogenic center has been scarcely explored. Zingiber officinale Rosc., “ginger”, is widely cropped for its use as a spice and raw material for phytotherapeuticals. During cropping or post-harvest, different phytosanitary products are used, whose residues can be found in the final product, lowering the, quality and safety of the raw material. To detect those residues, a multi-contaminant methodology for GC–MS/MS analysis was developed and validated. For the selection of most suitable methodology, the performance of three methods in the determination of 39 analytes were compared: QuEChERS, QuEChERS AOAC 2007.01, and a modification of the AcOEt method, based on the inclusion of Al2(SO4)3 in the extraction step to form stable and ethyl acetate insoluble Aluminum- (gingerols and/or polyphenols) complexes, lowering the co-extractives load injected in the instrument. Compared to the Acetonitrile based methods, the AcOEt method showed the best performance as 95% of the targeted analytes yielded recoveries between 70 and 120 and 80% of them between 90 and 110%). The method was validated for 57 contaminant residues by GC–MS/MS. The figures of merit (media of recoveries 96% and RSD 16%, back calculated concentration < 20%, matrix effects > 100%, linearity 5–300 μkg-1) of the method accomplished the SANTE guidelines and the LOQs for all compounds (10-25μkg-1) were below the ginger MRLs defined by food and medicinal herbs regulations, to ensure its safe consumption. The linear range was up to 25 mg L-1P for phosphate, up to 1600 μg L-1N-NO2for nitrite, and up to 2500 μg L-1N-NO3for nitrite plus nitrate. The analyzer can perform analysis cycles every 6 h with an autonomy of one week. Although the genetic and the biochemical process occurring during alternate bearing has been studied extensively, there is a lack of information identifying the presence of metabolic indicators during “on” and “off” years. In citrus plants, leaves play a central role in the metabolic pathway triggering the flflowering induction process. To investigate the changes during this transition, a liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS) analysis of the leaf profifiles of 20 compounds (17 polyphenols, two limonoids, and one furanocoumarin), in bearing and non-bearing branches arising from four different mandarin genotypes, was performed. The same metabolites were found in all the genotypes at both stages: both limonoids and 11 polyphenols.</div> <div>Using these compounds, the chemotaxonomic differentiation between cultivars was assessed. A path to enhance the efficiency of photocatalytic devices is by doping the active semiconductor in order to maximize the light absorption. However, impurities could increase the recombination of photoinduced charge carriers, in order that a careful analysis on the effects of those impurities it is necessary. On this research, density functional theory calculations were applied to analyze the structure and electronic characteristics of B–doped and C+B–codoped TiO2(B) considering the lowest formation energy of substitutional impurities. Our calculations suggest that the swapping of a boron or carbon atom for an oxygen atom (B@O or C@O) is energetically more favorable than the replacement of titanium atoms. The obtained results seem to indicate that the boron substitution into O2C and O3C1 sites leads to its migration towards interstitial sites in both mono and codoping, thus causing a great geometrical distortion compared to pure TiO2(B). Furthermore, impurity states in the band gap were obtained after B@O doping and C+B–codoping, producing an improvement of the optical properties, so it can be assumed that this nanomaterial might act as a photoelectrode after an adequate design optimization that facilitates the charge carrier separation. The first stage of the drug discovery process involves the identification of small compounds with biological activity. Iboga alkaloids are monoterpene indole alkaloids (MIAs) containing a fused isoquinuclidine-tetrahydroazepine ring. Both the natural products and the iboga-inspired synthetic analogs have shown a wide variety of biological activities. Herein, we describe the chemoenzymatic preparation of a small library of novel N-indolylethyl-substituted isoquinuclidines as iboga-inspired compounds, using toluene as a starting material and an imine Diels–Alder reaction as the key step in the synthesis. The new iboga series was investigated for its potential to promote the release of glial cell line-derived neurotrophic factor (GDNF) by C6 glioma cells, and to inhibit the growth of infective trypanosomes. GDNF is a neurotrophic factor widely recognized by its crucial role in development, survival, maintenance, and protection of dopaminergic neuronal circuitries affected in several neurological and psychiatric pathologies. Four compounds of the series showed promising activity as GDNF releasers, and a leading structure (compound 11) was identified for further studies. The same four compounds impaired the growth of bloodstream Trypanosoma brucei brucei (EC50 1–8 uM) and two of them (compounds 6 and 14) showed a good selectivity index. Biodiesel generated by transesterification of triglycerides from renewable sources is a clean form of energy that is currently used in many countries in blends with petrodiesel. It is mainly produced from food-grade vegetable oils obtained from oleaginous crops. High prices of these oils have made the sustainability of biodiesel production questionable. In this work, we propose intracellular lipids production by a previously selected oleaginous yeast strain in a medium composed only by sugar cane vinasse and crude glycerol. Different culture strategies were studied. The highest biomass and lipid yields were obtained when the yeast R. graminis S1/2R was cultivated in batch without control of dissolved oxygen. The fatty acid methyl esters obtained under these conditions met the specification of international biodiesel standards. The present research introduces text highlighting as a method for measurement of explicit attitudes using a case study on vertical farming (VF) with 837 UK consumers. They participated in an online survey, where they read a text about VF and used highlighting functions to mark text as ‘like’ and ‘dislike.’ Consumers approached the task in a systematic and logical way and desirable aspects of VF were frequently highlighted as ‘like’, whereas undesirable aspects were more frequently highlighted as ‘dislike’. The text highlighting responses were summarised using word clouds, frequency tables and through sentiment scores to reveal an overall positive attitude to VF among participants. Sentiment scores enabled the identification of consumer segments with interpretable differences in their attitude towards VF. Two approaches to method validation – comparison with direct attitude questions and consumer profiling – further confirmed the potential of the text highlighting method. The sentiment of specific sentences in the text highlighting task matched results from self-reported attitudinal based on Likert scales. Consumer segments with different sentiment in the text highlighting task also differed in their food technology neophobia scores in the expected direction. Future research should investigate methodological aspects of text highlighting and explore its suitability to other applications. Complexes [Ag(HPCPH)(PPh3)2]OTf (1), [Ag(HPCPH)((P(p-tolyl)3)2]OTf (2) and [Ag(HPCPH)(P(p-anisyl)3)2]OTf (3) were characterized in solid state and in solution by elemental analysis and FTIR and NMR spectroscopies (OTf––triflate). Crystal structures of (1,2) were determined by XRD. The Ag atom is coordinated to azomethine and pyridine nitrogen atoms of HPCPH ligand and to the phosphorous atom of each aryl-phosphine co-ligand. Although HPCPH did not show activity, the Ag(I) compounds demonstrated activity against Mycobacterium tuberculosis (MTB), H37Rv strain, and multidrug resistant clinical isolates (MDR-TB). Globally, results showed that the compounds are not only effective against the sensitive strain, but are more potent against MDR-TB than antimycobacterial drugs used in therapy. The compounds showed low to moderate selectivity index values (SI) towards the bacteria, using MRC-5 cells (ATCC CCL-171) as mammalian cell model. Interaction with DNA was explored to get insight into the potential mechanism of action against the pathogen. No significant interaction was detected, allowing to discard this biomolecule as a potential molecular target. Compound 1 was identified as a hit compound (MIC90 2.23 μM; SI 4.4) to develop further chemical modifications in the search for new drugs. The essential oil composition of the aerial parts from Baccharis palustris Heering (Asteraceae), a highly endangered species, was analyzed by GC/qMS, GC/HRMS-TOF, FT-IR, UV–vis and NMR spectroscopy using 1D and 2D techniques. To our knowledge, this is the first report on the essential oil composition of B. palustris, as well as the first phytochemical study on this plant species. The volatile extracts were obtained from different phenologicals stages of plant, at flowering stage (FS) and vegetative stage (VS), where 51 components were identified accounting for 96.3 % and 99.0 % of the oil, respectively. The oil was rich in polyacetylene compounds (> 75 %), being the main components identified the new natural C9-polyacetylenes 1-nonene-3,5-diyne (1, here named as baccharisdiyne) (52.7–65.0 %), 1,7(Z)-nonadiene-3,5-diyne [2, 7(Z)-dehydro-baccharisdiyne] (14.4–17.8 %), and 1,7(E)-nonadiene-3,5-diyne [3, 7(E)-dehydro-baccharisdiyne] (1.5–2.4 %). In addition, the known polyacetylenic compounds (Z)-lachnophyllum acid methyl ester (4) (4.3–5.3 %) and (E)-lachnophyllum acid methyl ester (5) (0.2 %) were also identified. Moreover, GC/qMS analysis allowed the identification of other 46 components in the essential oil samples, mainly mono- and sesquiterpenoids. In B. palustris, C9-polyacetylenes probably derive biogenetically from a C10 precursor: baccharisdiyne (1) would be derived from either or both lachnophyllum methyl ester geometric isomers (4 or 5), by enzymatic hydrolysis followed by decarboxylation. Dehydro-baccharisdiynes (2 and 3) could be produced by a similar  pathway starting from the corresponding matricaria acid methyl ester isomers, one of which was tentatively identified at trace-level in B. palustris essential oil. A hydride generation nitrogen microwave induced plasma optical emission spectrometry (HG-MIP OES) system was developed for lead determination. Plumbane chemical generation was performed using 0.045 mol L�1 K3Fe(CN)6 as the additive in acidic medium and 2.0% w/v NaBH4 in 1% w/v NaOH as the reductant. The additivated sample and the reductant were pumped towards a cyclonic chamber where hydride generation and separation from the solution occurred. Operating conditions such as the additive and reductant concentration, sample, reductant and nitrogen flow-rate, the cyclonic chamber stabilization time and the viewing position in the torch were optimized. A meticulous study of the variation of the nitrogen plasma response as a function of the hydrochloric acid concentration was carried out. The analytical performance of the method was studied and its applicability evaluated in a Trace Element in Water Certified Reference Material and a Soil Reference Material. The method performance in terms of limits of detection and quantification (0.54 mg L�1 and 1.8 mg L�1 Pb, respectively) was found comparable to other atomic spectrometry techniques. This is to our knowledge the first research on the analytical performance of lead determination by PbH4 generation and determination by MIP OES. Biofuel production from biomass sources emerges as a promising alternative to solve the actual environmental crisis and lead to a transition to a carbon-free future. In this sense, hydrogen produced through steam reforming of a biomass-derived biofuel, such as bio-oil, appears as an interesting energetic vector. The aim of this work is the hydrogen production through steam reforming of small organic model compounds present in the aqueous fraction of bio-oil (ethanol, acetic acid, acetone) and, for a better approximation to a real system, emulate such aqueous fraction by a representative mixture of these. Ternary mixed oxides Ni-La-Me catalysts (Me = Ce, Ti, Zr) were prepared, characterized through X-ray diffraction, N2 adsorption–desorption isotherms, thermogravimetric analysis, cyclic voltammetry, and scanning electron microscopy, and tested in the steam reforming reactions. Results indicate that, due to the intrinsic H2O adsorption capacity of ZrO2 as well to a higher number of oxygen vacancies in the catalyst structure, Ni-La-Zr presents interesting properties for its potential applicability as industrial catalyst for hydrogen production through steam reforming. On the other hand, the strong metal-support interactions on Ni-La-Ti led to a lower catalytic activity caused by a lower segregation of Ni crystallites from the mixed oxide structure. There is great interest in the study of the optical properties of materials such as glasses, due to their possible technological applications and from a fundamental point of view. For this reason, an attempt has been made to advance the understanding of the existence of optical band gap in this type of material. In recent times, several ways of analyzing the dependence of the absorption coefficient with energy have been proposed. Using lead, strontium, and barium borate glasses as a model, this dependence has been analyzed. For this, we considered the glass structure and the corresponding crystal structure, and the value of the optical band gap energy has been discussed considering the theory of Mott and Davis and comparing with values of crystals and glasses of similar composition. Here, we used a correction method, that takes into account intrinsic defects, to obtain the values of optical band gap energies in the framework of a non-direct transition. We also include DFT simulations of the corresponding crystalline compounds and the band gap was evaluated. The absorption coefficient obtained from simulations shows the same trend that the experimental one. The aim of the present work was to explore the potential of Biterm Topic Modelling (BTM) for analysing responses to open-ended survey questions in the pursuit of consumer insights. The selection of BTM was based on its suitability to the analysis of short texts compared to other approaches such as Latent Dirichlet Allocation. The empirical context was vertical farming (VF), which participants read a short text about and then answered a question about interest in buying fruits and vegetables from VF. The data came from US consumers (n = 1803) who provided answers with a median length of 9 words. Results were promising about the potential of BTM to uncover latent topics from responses to open-ended questions. The models predicted coherent and consistent topics relating to VF, including buying VF produce or not and year-round fruit and vegetable supply. Two variants of the text about VF were used (n ~ 900 per variant), and BTM models trained on responses to the variant which emphasised the possibility of using genetic engineering in VF consistently predicted topics related to genetic engineering. Models trained on responses to the variant which emphasized urban production and reduction of carbon emissions linked to product transportation, consistently revealed topics associated to production, environment, and local farming. The major limitation was not taking account of sentiment in consumers’responses, an issue that BTM shares with other topic models. This is a significant hindrance in the efforts to rely on machine learning for consumer insights. Benzimidazole methylcarbamate anthelmintics, including fenbendazole (FBZ), have only limited water solubility and small differences in drug solubility may have a major influence on their absorption, pharmacokinetic behavior and anthelmintic efficacy. To improve FBZ water solubility and dissolution rate, novel self-dispersible nanocrystals (SDNCs) of FBZ were recently described. In this work, the pharmacokinetic behavior of the SDNCs of FBZ and Poloxamer 188 was compared against a physical mixture (PM) of its components. The experiment was conducted following a crossover design with two different experimental phases. In phase I, sheep were treated with the SDNC (n = 3) or the PM (n = 3) formulations by the intraruminal route at the same dose rate (5 mg/kg). The treatment groups were reversed after a 7-days washout period. A non-compartmental analysis of the concentration in plasma versus time results showed that the calculated Cmax and AUC0-T were significantly higher (p < 0.05) for FBZ and its metabolites after the SDNC treatment compared to the PM (for FBZ: Cmax 0.346 μg/mL and AUC0-T 10.1 μg.h/mL after the SDNC vs Cmax 0.157 μg/mL and AUC0-T 5.1 μg.h/mL after the PM treatment). Additionally, population pharmacokinetic parameters of FBZ were estimated for the first time in sheep. In conclusion, the formulation of FBZ as SDNCs is a promising approach to improve FBZ dissolution reaching a higher drug plasma exposure in ruminants.
