Projective techniques have been increasingly used in marketing and consumer research. Despite their potentialities, few studies have been published comparing results from different techniques in a specific food product. In this context, the aims of this study were to uncover consumer perception and motivations to buy a ready-to-eat-salad using three different projective techniques (word association, completion and Haire’s shopping list), and to compare the results obtained by the three methodologies. The three tasks were completed through online questionnaires by groups of respondents within a sample of 320 consumers. Results from the three methodologies applied in this study revealed that convenience was the main motivation for purchasing ready-to-eat-salads, as expected. However, even though the three methodologies were useful, each of them brought to light consumer perception to a different extent. Word association enabled the elicitation of the largest number of categories, but only positive characteristics of the product were salient. Thus, inferences made from this methodology would differ to a great extent to those made from completion tasks’ results, where many limitations of the product were also relevant. Meanwhile, results from Haire’s shopping list were not as rich as those from the other methodologies, suggesting that it would be more appropriate for products which have a greater impact on consumers’ values and beliefs. Consumers’ perceptions of conventional (regular plain yogurt, low-calorie and low-fat yogurt) and functional yogurts (enriched with fibre and antioxidants) were studied using word association and hard laddering. Both methodologies provided similar results, being consumers’ perceptions of these products mainly related to health, nutrition, sensory characteristics and pleasure. However, both methodologies provided different information. Word association provided information about attributes that were relevant for consumers’ acceptance of a product, common defects of products in the marketplace, motives behind consumers’ choice and attributes that might limit consumers’ interest in the product. On the other hand, hard laddering allowed the identification of more consequences and values related to product attributes, and the links between them, which could be useful for understanding consumers’ choices. Besides, word association was more sensitive than hard laddering in determining differences between groups of consumers with different attitudes towards health and nutrition. Results suggested that word association might be an interesting and useful qualitative technique for product development. As the markets for functional foods increase, studying and understanding consumers’ perception of functional foods is essential to develop products that have a good consumer acceptance. The objective of this work was to provide a first insight into Uruguayan consumers’ perception of functional foods. A survey with 200 consumers was performed. The participants were not familiar with the term “functional food,” as only 37% related the term with the real concept behind it. However, the participants showed a positive attitude toward the functional foods concept. Preferred areas of action of functional foods at a generic level were not reflected in the ratings of perceived healthiness and willingness to try yogurt with different enrichments, probably because they did not associate the components with their health effect. These results suggest that the use of health claims would be essential to ensure the success of functional foods. The aims of the present work were to study consumer attitudes toward shelf-life labeling and to evaluate if shelf-life information affects consumer acceptability, rejection to consume and rejection to repeat purchase. Six samples of dulce de leche with different plastic flavor intensities were presented to a panel of 50 consumers, who scored their overall acceptability and indicated their intention to consume and to repeat purchase of the samples. They also answered a multiple-choice questionnaire regarding their attitude toward shelf-life labeling. Knowing that the samples were near the end of their shelf life did not affect acceptability scores or consumer rejection to consume. Thus, consumer perception of the product was not affected by knowing that it was close to expiration. This is in agreement with the fact that most consumers (75%) first try an expired product before deciding what to do. <h4>PRACTICAL APPLICATIONS</h4> Ninety-five and seventy-nine percent of the consumers stated that they read shelf-life information at purchase and consumption stage, respectively, which suggests the importance of proper shelf-life labeling. Consumer rejection to repeat purchase was higher than rejection to consume. Thus, consumers seemed to consume a product although they might not actually like it or buy it again. Thus rejection to purchase or repeat purchase could be a more conservative criterion than rejection to consume in order to estimate sensory shelf lives or acceptance limits for sensory defects. Sensory characterization provides a complete description of the sensory characteristics of food products. Descriptive Analysis is the most common methodology for this purpose. However, due to the cost and time associated with its application, several alternative methods have been recently developed. These methods do not require training, can be performed by trained assessors or consumers, and have been reported to be a good option when quick information about the sensory characteristics of a set of products is needed. There are basically four types of methodologies: methodologies based on the evaluation of specifi c attributes, on global differences among products, on the comparison with references, and methodologies that provide a verbal description of the products. In the present chapter these novel methodologies for sensory characterization of food products are described. Advantages and disadvantages of each methodology are discussed and recommendations for their application are provided. The application of check-all-that-apply (CATA) questions for sensory product characterisation is gaining acceptance and popularity. This question format has been reported to be a quick and reliable means of gathering sensory profiles from consumers, concurrently with hedonic assessment. However, a limitation of CATA questions is that they do not encourage deep processing by respondents. Forced-choice questions, where respondents answer “yes” or “no” for each term, may encourage systematic processing and be useful when consumers undertake sensory profiling tasks. This research compared sensory profiles elicited by consumers using CATA questions or forced-choice Yes/No questions and contribute to ongoing investigations of CATA questions and related question formats with a view to developing guidelines for best practise. Across seven consumer studies with 600+ consumers and multiple product categories, consistent evidence was obtained that forced-choice Yes/No questions are associated with higher term citation frequencies. However, this did not consistently translate into greater product discrimination. Conclusions regarding similarities and differences amongst samples and the stability of sample and term configurations were generally independent of question format (i.e., whether the sensory data were elicited by CATA or forced-choice Yes/No questions). Overall, the comparison of CATA and forced-choice Yes/No questions for sensory characterisation suggested parity of the two question formats. This extended to consumers’ perceived difficulty and tediousness for completing the test. Regardless of question format, consumers, on average, perceived the tests as easy and not tedious. Holistic methodologies such as Projective Mapping and sorting have gained popularity for sensory characterization of products with both trained assessors and consumers. One of their main disadvantage is that all samples should be simultaneously evaluated in the same session. An alternative to overcome this limitation is to evaluate samples by comparing them with a fixed set of products, as proposed in Polarized Sensory Positioning (PSP). In the present work a combination of Projective Mapping and Polarized Sensory Positioning, called polarized Projective Mapping (PPM), is presented and compared with the two original ways of performing PSP (scale based and triadic-PSP), in terms of conclusions regarding differences between samples and difficulty for consumers. Nine orange-flavored powdered drink samples (including one blind repeated sample) were evaluated by three groups of 45 consumers using PSP, triadic-PSP and PPM using a between-subjects design. Although the three methodologies provided similar sensory spaces, some differences were identified in terms of discriminative ability, conclusions regarding similarities between samples and perceived difficulty. Polarized Projective Mapping seems to be an interesting approach that combines the advantages of Polarized Sensory Positioning and the holistic character of Projective Mapping, providing the possibility of comparing samples evaluated in different sessions. Proteolytic biocatalysts extracted from the ripe fruits of Solanum granuloso-leprosum, a native Uruguayan tree of the Solanaceae family, were stabilized by adsorption onto gamma-alumina supports. Conditions for preparing the biocatalyst–adsorbant system were evaluated for different crude extract (CE) protein concentrations and at different pHs and temperatures. All systems assayed reached equilibrium within 2–4 h. The best proteolytic activity (measured as direct enzymatic activity) was found to be at pH 6.0 and 5 ◦C. The biocatalyst–adsorbant system was more stable upon storage than the free enzyme in solution, which loses activity over time due to autolytic processes. Stability (resistance to desorption) was studied after ten cycles of incubation at different pHs and temperatures. There was a general tendency to retain higher activity (more than 60%) when the incubation conditions were the same as those used for preparing the biocatalyst–adsorbant system. The best conditions for using this system to treat industrial effluents were evaluated using milk whey from cheese-making and hemoglobin (the main constituent of wastewater from cattle slaughter-houses) as trial substrates. Background: Several oral ivermectin (IVM) formulations for use in sheep are available in the pharmaceutical veterinary market in different countries. All of them are indicated at the same dose rate to treat the gastrointestinal nematodes. However, there is a lack of information on the relative systemic exposure (plasma bioavailability) and clinical efficacy among oral formulations routinely used in sheep. The main goal of the work reported here was to perform a pharmaco-parasitological assessment of three different IVM oral formulations in lambs infected with multiple resistant gastrointestinal nematodes. The comparative drug systemic exposure (IVM plasma concentrations) and nematodicidal efficacies (clinical efficacy) in lambs were determined for a reference (RF) and two different test (T1, T2) IVM oral formulations. One hundred and fifty six (n= 156) healthy Corriedale lambs, naturally infected with multiple resistant gastrointestinal nematodes were allocated into four experimental groups (n=39). Animals in each group received treatment (200 μg/kg) with either the RF, one of the test IVM formulations or were kept as untreated control. Blood samples were collected over 15 days post-treatment (n=8). The IVM plasma concentrations were measured by high performance liquid chromatography with fluorescence detection. The faecal nematode egg count reduction test (FECRT) (n=39) and evaluation of the clinical efficacy were performed at day 14 post-treatment (n=6), where a predominance of IVM highly resistant nematodes was observed. Results and conclusions: Neither the overall kinetic behaviour nor the IVM systemic exposure differed among all the tested oral formulations. Equivalent efficacy results were obtained for the different preparations, with an evident therapeutic failure to control Haemonchus spp. and Teladorsagia circumcincta, which correlates with a high degree of nematode resistance to IVM. Background: Bioavailability (F) and clearance (CL) are two pharmacokinetic parameters difficult to differentiate from simple plasma measurement when a drug is administered orally. Venous (V) / artery (A) concentration ratio of a drug could be a reliable index of its CL if measurements of plasma concentration were performed during a period of time where the absorption process was not longer operative, then during a pure elimination phase. Objective: A novel subrogate using two protocolized saliva samples sequentially collected (first, S1, and second, S2) was designed in order to replace V and A free plasma drug concentrations, respectively. Two drugs, phenytoin (PHT) and carbamazepine (CBZ), which are well-known for their inducer properties and their dose-dependent clearance variations, were studied taking into account the sex of individuals. Setting and patients: A multicentre two-phase collaborative study was done. The first phase was performed with healthy volunteers in order to determine salivary pharmacokinetic parameters after single dose administration. Twelve volunteers (6 male and 6 female) received 400 mg of CBZ (2 tablets x 200 mg, immediate release product). Twenty four volunteers (10 male and 14 female) received 100 mg of PHT. The second phase was carried out with epileptic patients under CBZ (11 male 15 female) or PHT (11 male and 11 female) monotherapy, in order to study dose-related and sex-related pharmacokinetic differences. Main outcome measures: In the single dose trials, peaks (Tmax, Cmax) were computed directly from the data. Areas under concentration-time curves (AUC∞), AUC∞xW (area corrected by weight) and half-lives (t1/2) were calculated. In the case of CBZ, AUCCBZ-10,11-epoxide/AUCCBZ metabolic ratios were also calculated. After multiple dose administration, S1 and S2 trough morning drug concentrations were measured. Results: Cmax and AUC differed significantly between sexes for the two drugs after single dose administration. Nevertheless, the apparent clearance (CL/F) per unit of body weight did not differ (CBZ) or slightly differed (PHT) between sexes. Higher metabolic ratio for CBZ in women would lead to lower F and therefore lower CL in this gender. In the case of PHT, women would have either lower F or higher CL than men. After multiple dose administration, S1/S2 saliva drug concentration ratio correlated positively with S2 for CBZ, showing that CBZ clearance increases with daily dose. Gender differences were also observed for CBZ-10,11-epoxide concentration, being bioavailability the main parameter responsible for this difference. S1/S2 saliva PHT concentration ratio correlated negatively with S2, showing that PHT clearance diminishes with dose as it has been previously reported. Since a significant difference was found for S1/S2 ratio between male and females, CL is the pharmacokinetic parameter influenced by gender in PHT disposition. Conclusion: S1/S2 saliva drug concentration ratio was sensitive enough for detecting systemic clearance changes. Both CBZ and PHT would modify their bioavailability and clearance by inducing efflux transporter throughout chronic treatments, from the first dose to multiple dose administration. The infl uence of key parameters in the kinetics of the transglycosylation system catalyzed by Aspergillus oryzae β -galactosidase when applied to the synthesis of galactosyl-ethylene glycol and galactosyl-glycerol has been studied. It was found that both acceptors were inhibitors of the enzyme, causing a decrease of 9 and 68 fold in enzyme catalytic effi ciency in the presence of 4.0 M ethylene glycol and glycerol, respectively. The effect was characterized as “ substrate-like inhibition ” , suggesting the formation of a nonproductive enzyme-acceptor complex. Inhibition constants for both acceptors were determined as 618 mM for ethylene glycol and 244 mM for glycerol.
