A summary of the resource.
The interaction of arsenic(V) and arsenic(III) oxyanions with metal cations was investigated by potentiometry under temperature and ionic strength conditions approaching those prevailing in natural waters. The selection includes the major metal cations and some other ions of high environmental relevance. Ionic pairs [M(AsVO4)]−, [M(HAsVO4)] and [M(H2AsIIIO3)]+ formation is suggested for all +2 metal cations, based on the potentiometric results. These ion-pairs between arsenic anions and other metal cations are hardly ever mentioned or taken into account when arsenic speciation in natural waters is considered. These results provide the basis for studying arsenic speciation in natural aquatic systems, on which environmental fate, bioavailability and toxicity of the element depend. Some extrapolations to the conditions of the natural waters are presented as well as some insights into the adsorption process onto hydrous oxides.

<p>Photolysis of 1,2-dithiane (1) in acetonitrile with single walled carbon nanotubes (SWCNTs) was earlier reported to form thiolfunctionalized SWCNTs via the butane-1,4-dithiyl diradical (2). The present study shows that 2 instead undergoes a facile rearrangement to thiophane-2-thiol (6). This photoreaction is clean, rapid, and irreversible under 313 nm irradiation. The secondary photolysis of 6 with SWCNTs at a shorter wavelength (254 nm) leads to 2-thiophanyl radicals 8, which derivatize SWCNTs by covalent attachment. Pyrolysis of the resulting “sulfurized SWCNTs” affords a mixture of organosulfur compounds, including thiophene formed by dehydrogenation. An unknown additional mechanism causes high TGA weight loss and a large incorporation of sulfur. ■ INTRODUCTION Single-walled carbon nanotubes (SWCNTs) have been functionalized by a variety of methods,1−6 many of which involve free radicals.7−18 In 2006 and 2007, Nakamura and coworkers19,20 published an unusual functionalization approach based upon biradicals. Irradiation of cyclic disulfides, e.g., 1 generated dithiyl biradicals 2, which were claimed to attack SWCNTs and produce thiol-functionalized SWCNTs 4, as illustrated in Figure 1. The stoichiometry and thermodynamic stability of copper(II), vanadium(IV/V), iron(II)/(III) and gallium( III) complexes of salicylaldehyde semicarbazone (SSC, HL) and its 5-bromo derivative (Br-SSC, HL) have been determined by pH-potentiometry, UV–Vis spectrophotometry, EPR, 1H and 51V NMR spectroscopy in 30% (w/w) dimethyl sulfoxide/water solvent mixture. Proton dissociation processes and lipophilicity of the ligands were also studied in detail. Formation of mono-ligand complexes such as [ML], [MLH�1], [MLH�2] was found with copper(II), vanadium(IV/V), while bis-ligand species of iron(II)/(III) and gallium(III) such as [ML2], [ML2H�1] and [ML2H�2] were also detected, in which the ligands coordinate via monoanionic (O�,N1,O) or dianionic (O�,N1,O�) modes. The bromine substituent on the phenol ring has no significant impact on the stability and binding modes but provides a remarkably enhanced lipophilic character, which is advantageous for the bioactivity. The Ga(III)–salicylaldehyde semicarbazone species show unambiguously higher stability; whereas Cu(II) species have somewhat lower stability relative to the corresponding thiosemicarbazone analogues, however no decomposition of the Cu(II) complex was observed even at micromolar concentrations at physiological pH.

Phytate (L12−) is a relevant natural product. It interacts strongly with biologically relevant cations, due to the high negative charge exhibited in a wide pH range. The synthesis and crystal structures of the mixed‐ligand Cu(II) polynuclear complexes K(H2tptz)0.5[Cu(H8L)(tptz)] ⋅ 3.6H2O (1), K(H2O)3{[Cu(H2O)(bpca)]3(H8L)} ⋅ 1.75H2O (2), and K1.5(H2O)2[Cu(bpca)](H9.5L) ⋅ 8H2O (3) (tptz=2,4,6‐tri(pyridin‐2‐yl)‐1,3,5‐triazine; Hbpca=bis(2‐pyridylcarbonyl) amine) are reported herein. They were obtained by the use of an aromatic rigid amine, which satisfies some of the metal coordination sites and promotes the hierarchical assembly of 2D polymeric structures. Speciation of phytate−Cu(II)−Hbpca system and determination of complex stability constants were performed by means of potentiometric titrations, in 0.15 M NMe4Cl at 37.0 °C, showing that, even in solution, this system is able to produce highly aggregated complexes, such as [Cu3(bpca)3(H7L)]2−. Furthermore, the Cu(II)‐mediated tptz hydrolysis was studied by UV‐vis spectroscopy at room temperature in 0.15 M NMe4Cl. Based on the equilibrium results and with the aid of molecular modelling tools, a plausible self‐assembly process for 2 and 3 could be proposed.

Sn-based nanocomposites attract extensive attention as a new generation of photoelectric hybrid materials due to their high photoelectric conversion efficiency. In this work we synthesized SnS2 nanoparticles by a hydrothermal method employing citric acid (capping agent) and performed a capping agent exchange by pyridine to improve their solution processing and final properties. We fabricated layers with pyridine-capped nanoparticles and poly (3-hexylthiophene-2,5-diyl) (P3HT), employing the classical partial film sequence of a solar cell (ITO/PEDOT: PSS/P3HT:Nps). We found a “coupling" between the nanoparticles and the polymer and highly ordered structures within the layers. The addition of nanoparticles improved the UV–vis absorption of P3HT and allowed the exploitation of the photoluminescence emission to promote extra charge carrier generation. Moreover, we found that SnS2_py nanoparticles form a nanoscopic mixture with the P3HT. Our pyridine-capped SnS2 nanoparticles are good candidates for fabricating hybrid materials, employing P3HT as the organic compound for application in polymeric-inorganic solar cells.

A summary of the resource.
The tautomeric equilibrium of the title triazole compound was computationally analyzed at the B3LYP/6-311++G(d,p) and MP2/6-311++G(d,p) levels of theory. The solvent effect was considered for three solvents (chloroform, methanol, and water). Two distinct mechanisms were applied: a direct intramolecular transfer using the polarizable continuum model (PCM) and a solvent-assisted mechanism. The calculations indicated that the keto form is more stable in all cases. It was found that the barrier heights for the tautomerization reaction are very high, indicating a relatively disfavored process. Although the barrier heights for solvent-assisted reactions are significantly lower than those for the unassisted tautomerization reaction, implying the importance of the superior catalytic effect of the solvents, monosolvation was not found to be sufficient for the reaction to occur. Finally, the two intermolecular hydrogen-bonding interactions in the crystal structure were investigated in the gas phase; according to the calculated energies and structural parameters, the order of stability is N3–H3···O1 > N1–H1···O1.

A summary of the resource.
Light chain deposition disease (LCDD) and light chain amyloidosis (AL) are disorders of monoclonal immunoglobulin deposition in which normally soluble serum precursors form insoluble deposits in tissues. A common feature in both is the clonal proliferation of B-cells that produce pathogenic light chains. However, the deposits in LCDD differ from those in AL in that they are ultrastructurally granular rather than fibrillar and do not bind Congo red or colocalize with amyloid P component or apolipoprotein E. The reason(s) for their differences are unknown but are likely multifactorial and related to their protein conformation and their interaction with other molecules and tissue factors in the microenvironment. Knowledge of the primary structure of the light chains in LCDD is very limited. In the present study two new κ1 light chains from patients with LCDD are described and compared to seven other reported κ-LCDD proteins. The N-terminal amino acid sequences of light chain GLA extracted from the renal biopsy and light chain CHO from myocardial tissue were each identical to the respective light chains isolated from the urines and to the V-region amino acid sequences translated from the cloned cDNAs obtained from bone marrow cells. The germline V-region sequences, determined from the genomic DNA in both and in MCM, a previously reported κ1 LCDD light chain, were identical and related to the L12a germline gene. The expressed light chains in all three exhibit amino acid substitutions that arise from somatic mutation and result in increased hydrophobicity with the potential for protein destabilization and disordered conformation.

Abstract
A summary of the resource.
An overall 44% transplacental transmission rate was observed in 221 rats fed cysts of 12 Toxoplasma strains at 15 days of pregnancy, with a range of 0–90% transmission. Considerable variability in the transmission rate was seen among different groups of rats that received similar Toxoplasma inocula; this is attributed to genetically based susceptibility to Toxoplasma among individuals of the outbred Wistar strain of rats. Transplacental transmission was more frequent in Long Evans than in Wistar rats. Significant differences in the rate of transmission were not found between rats that were fed similar Toxoplasma inocula 6–8 days or 15 days after conception. The frequency of transmission was not affected by the strain or dose of Toxoplasma used.

This paper described that glyoxyl activated supports are only able to quantitatively immobilize proteins at alkaline pH values and using highly activated supports (otherwise, immobilization is negligible). Furthermore, the immobilization rate depends on the surface density of glyoxyl groups and not on the total concentration of groups in the suspension. Moreover, the temperature presented a dramatic effect on enzyme immobilization rate and different enzymes become immobilized at very different rates. Finally, it was not possible to detect immobilization of mono aminated compounds at neither alkaline nor neutral pH values. In fact, these supports may be used to purify enzymes due to these great differences in immobilization rate (e.g., glutamate racemase). These special features are not shared with other supports usually utilized for covalent immobilization of enzymes (e.g., glutaraldehyde, cyanogen bromide supports) that are able to immobilize proteins by just one very stable bond and suggesting an unique mechanism for glyoxyl agarose supports. In fact, all these results suggested that each individual amino-glyoxyl bond is very weak. Therefore, the first immobilization of proteins on supports activated with glyoxyl groups, in absence of reducing reagents, needs the simultaneous establishment of, at least, two attachments between the protein and the support. This mechanism promotes that proteins become immobilized by the area/s where the highest lysine residues density is located and explain the high enzyme stabilization usually achieved by using this immobilization technique.

Abstract
A summary of the resource.
The ternary alloys PbxCd1−xTe were prepared by the vertical Bridgman method and studied from a morphological viewpoint. The evolution of the different defects with the Pb concentration was described. For x = 0.20, strong segregation was found. A supercrystal structure, formed by alternated layers of CdTe and PbTe was encountered in the PbTe rich phase. We propose some hypothesis about the mechanism for their formation. An interpretation of the optical behavior in the 0.2–0.6 eV infrared region will be also presented.

<p>Phage-borne peptides and antibody fragments isolated from phage display libraries have proven to be versatile and<br />valuable reagents for immunoassay development. Due to the lack of convenient and mild-condition methods for the labeling of the phage particles, isolated peptide/protein affinity ligands are commonly removed from the viral particles and conjugated to protein tracers or nanoparticles for analytical use. This abolishes the advantage of isolating ready-to-use affinity binders and creates the risk of affecting the polypeptide activity. To circumvent this problem, we optimized the phage display system to produce phage particles that express the affinity binder on pIII and a polyglycine short peptide fused to pVIII that allows the covalent attachment of tracer molecules employing sortase A. Using a llama heavy chain only variable domain (VHH) against the herbicide 2,4-D on pIII as the<br />model, we showed that the phage can be extensively decorated with a rhodamine-LPETGG peptide conjugate or the protein<br />nanoluciferase (Nluc) equipped with a C-terminal LPETGG peptide. The maximum labeling amounts of rhodamine-LPETGG and Nluc-LPETGG were 1238 ± 63 and 102 ± 16 per phage, respectively. The Nluc-labeled dual display phage was employed to develop a phage bioluminescent immunoassay (P-BLEIA) for the detection of 2,4-D. The limit of detection and 50% inhibition<br />concentration of P-BLEIA were 0.491 and 2.15 ng mL−1, respectively, which represent 16-fold and 8-fold improvement compared to the phage enzyme-linked immunosorbent assay. In addition, the P-BLEIA showed good accuracy for the detection of 2,4-D in spiked samples.</p>

Artisanal and small-scale gold mining (ASGM) in the Amazon has degraded tropical forests and escalated mercury (Hg) pollution, affecting biodiversity, ecological processes and rural livelihoods. In the Peruvian Amazon, ASGM annually releases some 181 tons of Hg into the environment. Despite some recent advances in understanding the spatial distribution of Hg within gold mine spoils and the surrounding landscape, temporal dynamics in Hg movement are not well understood. We aimed to reveal spatio-temporal trends of soil Hg in areas degraded by ASGM.,. Fusarium Head Blight (FHB) is a major constraint to barley production that substantially reduces yield and grain quality. FHB is also a major food safety concern because FHB pathogens contaminate grain with trichothecenes and other mycotoxins. DNA sequence-based analyses and in-vitro toxin assessments were used to characterize the species and trichothecene chemotype composition of FHB pathogens on barley in Uruguay. F. graminearum was the dominant species (89.7%), and three other members of the F. graminearum species complex (FGSC) were identified as FHB pathogens of barley in Uruguay for the first time. Other minor contributors to FHB species diversity included F. poae, F. avenaceum, F. pseudograminearum and an unnamed species from the F. incarnatum-equiseti species complex (FIESC). Most isolates (89.7%) had the 15-acetyldeoxynivalenol (15-ADON) trichothecene type. However, the results expanded the known area of occurrence within Uruguay for the nivalenol (NIV) toxin type, which was observed among isolates from three species of the FGSC, F. pseudograminearum, and F. poae. Isolates with the 3-acetyldeoxynivalenol (3-ADON) or NX-2 toxin types were not observed, although a previously published multilocus genotyping assay was updated to identify NX-2 strains. Analyses of population structure and comparisons with FHB isolates from wheat in Uruguay indicated that F. graminearum constitutes a single genetic population with no evidence of population differentiation related to the sampled hosts. Inter and intraspecific differences were observed in aggressiveness toward four barley genotypes with different levels of resistance to FHB, and in general nivalenol producers were the least aggressive isolates. Sensitivity to metconazole was approximately 10 times higher than was detected for tebuconazole. This is the first report regarding tebuconazole and metconazole sensitivity for Fusarium species causing FHB in barley in Uruguay, and constitutes an important starting point for monitoring temporal or spatial changes in FGSC sensitivity, which is critical to define FHB management practice

Cruzipain, the major cysteine proteinase of Trypanosoma cruzi, might have other biological roles than its metabolic functions. In this report, we have explored the interaction of cruzipain with molecules of the immune system. The enzyme was used to digest all human IgG subclasses at different pH values and lengths of time. At pH 7.3, all subclasses were readily split at the hinge region. Immunoblot and amino acid sequence analysis showed fragments of IgG1 and IgG3 to be compatible with Fab and Fc, whereas IgG2 and IgG4 rendered Fab2 and Fc. In all cases the fragments produced might impair the binding capacities and the effector functions of specific IgG. At these cleavage sites cruzipain displays cathepsin L and/or cathepsin B activities and shows a clear preference for Pro at the P�2 position and polar residues at P1. Despite the activity of cruzipain within the hinge, the enzyme also cleaved all heavy chains between the CH2 and CH3 domains; producing Fc�-like-fragments of 14 kDa. These fragments are potential candidates to block or saturate Fc receptors on immunocompetent cells. At mild acidic pH cruzipain produced further degradation of the Fc of all subclasses, the Fd of IgG4 and partially the Fd of IgG1, with the consistent loss of any antibody activity. The L chains apparently were not affected. Thus, cruzipain should be able to modulate, depending on the subclass selected and the pH of the environment, the production and the length of different biologically active/inactive IgG fragments

Vitellogenin (VTG) and choriogenin (CHO) are valuable biomarkers of endocrine-disrupting compound (EDC) exposure in fish. Existing immunoassays are limited to a few species, which restricts their use for the analysis of local wildlife sentinels. Using C. facetum as a relevant South American model fish, this work presents a new strategy for the preparation of antibodies to VTG and CHO, with zero cross-reactivity with fish serum components. Recombinant fragments of Cichlasoma facetum VTG (280-mer) and CHO (223-mer) were prepared by degenerate primer RT-PCR and expression in E. coli. Polyclonal and monoclonal antibodies prepared with these antigens were used to develop rapid dotblot assays for VTG and CHO. Both the polyclonal and monoclonal antibodies prepared with the recombinant antigens reacted against the native proteins adsorbed on to nitrocellulose allowing the set up of sensitive dotblot assays. The VTG assay was further validated with spiked samples and purified native VTG. Exposure experiments with several estrogenic compounds revealed the potential of C. facetum as a sensitive biomonitor that produced measurable responses at concentrations of 100 ng L−1 of 17-beta-estradiol, 100 ng L−1 of ethynylestradiol, and 6.6 μg L−1 of nonylphenol. The approach described here may be applied to other native species to produce highly specific and sensitive rapid tests. It may be particularly advantageous for species that cannot be kept in captivity or when homogeneous purification of the immunizing proteins is particularly challenging. In conclusion, we present a novel approach to develop a strategy for the generation of immunoassay reagents for vitellogenin (VTG) and choriogenin (CHO), which will facilitate regional studies on the impact of endocrine-disrupting chemicals on local wildlife.

The dihydrated potassium salt of the complex anion [VO(O2)NTA]2− (NTA= nitrilotriacetate anion, [N(CH2-COO)3]3−) was thoroughly characterized by electronic and vibrational (infrared and Raman) spectroscopies. The bioactivity of the complex on the cell proliferation was tested on three cell lines in culture (UMR106 rat osteosarcoma-derived cells, Caco-2 derived from a human colon adenocarcinoma, and RAW 264.7, a macrophage murine cell line).
