By means of first principle calculations, we have performed a theoretical characterization of the stability and electronic properties of sheets and nanoribbons that were recently synthesized employing octafunctionalized biphenylenes as building blocks. We found that the biphenylene sheet has a strong metallic character that is difficult to inhibit employing low levels of functionalization. Hydrogenation at full coverage induces a metal to insulator transition, but the band gap opened is very large, i.e., 6.6 eV. When other functional groups such as fluorine or chlorine are attached, the band gap can be regulated. The most effective chemical modification, in terms of gap opening, is the combination of hydrogen/chlorine or fluorine/chlorine. For the latter functional groups, band gaps similar to those of rutile were calculated at the HSEH1PBE/6-31G* level of theory. The biphenylene sheet functionalized on one side with fluorine and with chlorine on the other presented a CC bond length equal to 1.76 Å, one of the longest reported up to date. In contrast with recent claims, we found that, for armchair biphenylene nanoribbons, the twist induced by the functionalization of the edges does not increase the band gaps of the nanoribbons. Moreover, in some cases the gaps were reduced as we observed when the edges where saturated with hydrogen atoms. Finally, the high reactivity of the sheet indicated that it is may have promising applications in catalysis. It is well known that during the supercritical alcoholysis of vegetable oils the main transesterification reaction occurs simultaneously to several decomposition phenomena, In order to separately study the effect of such phenomena, pure ethyl esters from soybean oil (SBOEE) were mixed with ethanol at a molar ratio 40:3 (ethanol:SBOEE) and exposed for different periods to supercritical conditions in a continuous system, at 20MPa and different temperatures from 250 to 375 ◦C. It was experimentally observed that the ester content of the processed samples were lower than that corresponding to the original SBOEE, indicating the occurrence of decomposition processes, which were more important as the temperature increased and the flow rate diminished. The content of polyunsaturated esters of the treated SBOEE was lower than that of the starting mixture, showing that the decomposition rate was highly dependent on the nature and instauration degree of the alkyl chain. Projective mapping for sensory characterisation with consumers has been used for a relatively short period of time, which suggests that the development of guidelines regarding best practices is strongly needed. The present work aims to provide an insight on the minimum number of consumers needed to reach stable sample configurations. Data sets from 21 different consumer studies, differing in product category, number of samples and degree of difference among them, were used to evaluate the influence of the number of consumers on the stability of sample configurations by means of a resampling approach. For each study, 1000 random subsets of different number of consumers were generated from the original data set. For each virtual panel, sample configurations were obtained using Multiple Factor Analysis. The agreement between them and the reference configurations (obtained with all the consumers) was evaluated through the RV coefficient, using the first two and the first four dimensions of the MFA. Results showed that the stability of sample configuration clearly depended on the degree of difference and type of differences among samples and the number of samples in the dataset. Across the 21 data sets analysed, results suggested that when working with widely different samples, 50 consumers seems as a safe recommendation of minimum consumer panel size to obtain reliable results with projective mapping. However, after any characterisation by projective mapping is completed, it is highly recommended to check, a posteriori, the reliability of the sample space configuration using a bootstrapping procedure. The controlled and partial modification of epoxy groups of Eupergit C and EP-Sepabeads with sodium sulfide has permitted the preparation of thiol-epoxy supports. Their use allowed not only the specific immobilization of enzymes through their thiol groups via thiol– disulfide interchange, but also enzyme stabilization via multipoint covalent attachment. Penicillin G acylase (PGA) from Escherichia coli and lipase from Rhizomucor miehei were used as model enzymes. Both enzymes lacked exposed cysteine residues, but were introduced via chemical modification under very mild conditions. In the first moments of the immobilization, a certain percentage of immobilized protein could be released from the support by incubation with DTT; this confirms that the first step was via a thiol–disulfide interchange. Moreover, the promotion of some further epoxy-enzyme bonds was confirmed because no enzyme release was detected after some immobilization time by incubation with DTT. In the case of the heterodimeric PGA, it was possible to demonstrate the formation of at least one epoxy bond per enzyme subunit by analyzing with SDS-PAGE the supernatants obtained after boiling the enzyme derivatives in the presence of mercaptoethanol and SDS. Thermal inactivation studies showed that these multipoint enzyme-support attachments promoted an increase in the stability of the immobilized enzymes. In both cases, the stabilization factor was around 12–15-fold comparing optimal derivatives with their just-thiol immobilized counterparts. Controlled and directed immobilization plus post-immobilization techniques are proposed to get full stabilization of the quaternary structure of most multimeric industrial enzymes. The sequential utilization of two stabilization approaches is proposed: (a) Multi-subunit immobilization: a very intense multi-subunit covalent immobilization has been achieved by performing very long immobilization processes between multimeric enzymes and porous supports composed by large internal surfaces and covered by a very dense layer of reactive groups secluded from the support surface through very short spacer arms. (b) Additional cross-linking with poly-functional macromolecules: additional chemical modification of multi-subunit immobilized derivatives with polyfunctional macromolecules promotes an additional cross-linking of all subunits of most of multimeric enzymes. A number of homo and hetero-dimeric enzymes has been stabilized by the simple application of multi-subunit immobilization but more complex multimeric enzymes (e.g., tetrameric ones) were only fully stabilized after the sequential application of both strategies. After such stabilization of the quaternary structure these three features were observed: no subunits were desorbed from derivatives after boiling them in SDS, thermal inactivation becomes independent from enzyme concentration and derivatives became much more stable than soluble enzymes as well as than non-stabilized derivatives. For example, thermal stability of d-amino acid oxidase from Rhodotorula gracilis was increased 7.000 fold after stabilization of its quaternary structure. In anoxic environments, degradation of organic matter (OM) results in strong fractionation of carbon isotopes, with formation of 13C-depleted CH4. Propionate and acetate are important products of OM fermentation. Propionate is further fermented to acetate. Acetate is a direct precursor of CH4, the remainder usually being produced from H2-mediated CO2 reduction. There is a paucity of data for the turnover of acetate and, even more so, propionate. We therefore analyzed the d13C values of organic carbon, propionate, acetate and the methyl (Me) group of acetate (acetate-Me) during the production of CH4 in anoxic incubations of various flooded and non-flooded soils and various lake sediments. Incubation in the presence of CH3F, which inhibits CH4 production from acetate, allowed exclusion of isotope effects during aceticlastic methanogenesis. Despite the variation inherent in the wide diversity of sample type and origin, the data collectively showed that the d13C value of acetate was only marginally different (�2 ± 5&permil;) from that of OM, while propionate was depleted in 13C relative to total acetate (�6 ± 5&permil;). Acetate-Me was generally depleted in 13C relative to total acetate (�8 ± 5&permil;). Thus, isotopic enrichment factors during the degradation of OM to total propionate and acetate were much smaller than those during hydrogenotrophic and aceticlastic methanogenesis or the intramolecular difference in d13C between the carboxyl (CO2H) and Me of acetate, so that the d13C value of OM may be used as a proxy when data for acetate are not available. Steam reforming of crude glycerol, a side product of biodiesel industry, could be used to obtain H2 and H2/ CO mixtures using Ni based catalysts. In this work, carbon supported nickel catalysts prepared by a simple impregnation technique were studied in the crude glycerol steam reforming at 650 �C to yield H2 and CO2 as the main products. Promotion with different metal oxides (MgO, La2O3, Y2O3) is effective to increase H2 yield which attain more than 80% of the maximum theoretical amount with MgO. Rh promotion is also detected at the initial stages of the process although it is counteracted by the deleterious effect of carbon deposits on the active metal. Carbon deposits and sintering of the active metal are the main sources of deactivation. Several strains of endophytic bacteria and yeast were isolated from Daucus carota, Erythrina crista-galli, Curcubita maxima and Fortunella margarita, and tested for the reduction of two model �-alkyl-�- ketoesters. The yeast strains resulted better biocatalysts for the reduction of the tested substrates with the exception of Enterobacter agglomerans C8 strain, isolated from Erythrina crista-galli, that provided the best results for the preparation of syn (2R,3S) �-alkyl �-hydroxyesters. Among the isolated yeast strains, the Pichia sp. strain isolated from D. carota, was the best biocatalyst for production of anti (2S,3S) �-alkyl �-hydroxyesters. Our results show that the microbial endophytic community provides an interesting niche on the search for novel biocatalysts. Stereoselective hydrogenation of methylcyclohex-2-ene-1,4-diols used as important intermediates for the preparation of ampelomins and deoxy-carbasugars was studied. These olefins were obtained in few steps from a chiral cis-diol resulting from microbial oxidation of toluene. Although the stereoselective hydrogenation of this type of substrates is difficult, high yields were obtained for heterogeneous hydrogenation using Adam’s catalyst, where steric hindrance controlled the stereochemical outcome of the process. Background and Objectives Ketoprofen, a potent nonsteroidal anti-inflammatory drug, is clinically administered as a racemic mixture. One of the possible metabolism routes of ketoprofen is the inversion of the R- to S-enantiomer in the gastrointestinal tract. Ketoprofen, as a weak acid drug, might undergo recirculation through pancreatic/intestinal juices. The aim of the work was to investigate if a plasma-gastrointestinal tract recirculation of ketoprofen could explain its R-to-S chiral inversion after the oral administration of two modified-release formulations: a gastro-resistant delayed-release tablet (Reference) and an extended-release-plus-immediate-release bilayer tablet (Test). Methods Sixteen healthy Caucasian volunteers (eight women and eight men) participated in a ketoprofen bioequivalence study. Both formulations were administered with and without food. In both cases, standard meals were given throughout the experiment. R- and S-enantiomers were measured separately using a validated HPLC–UV chiral method. Mean concentration–time profiles of ketoprofen enantiomers in plasma were obtained for men and women. Area under the plasma concentration–time curve, maximum ketoprofen plasma concentration, and time-to-peak were also computed for both isomers, both modes of administration, and both sexes. S/R concentration ratio was assessed as an indicator of enantiomer chiral inversion rate. Results Differences in the pharmacokinetics of S- and R-ketoprofen enantiomers were found after the Test administration. S-Ketoprofen presented a lower plasma exposure compared to R-enantiomer. However, the S/R concentration ratio increased 1 h (in men) and 2 h (in women) after meal intakes. This was related to pancreatic and/or intestinal and/or biliary secretions of the drug, followed by reabsorption and conversion of the R- to the S-isomer. The lower intestinal pH reported for men would lead to a higher oral bioavailability of the Test formulation and a higher reabsorption of both ketoprofen isomers in this sex. Hence, a higher rise of the S/R concentration ratio could be observed in men. No significant differences between isomers exposure were detected in both sexes after the Reference administration. Different lag times were observed after fed and fasting administration of this formulation; however, drug absorption coincided with food ingestion. Then, drug recirculation affected the S/R ratio from the beginning of drug exposure, minimizing the difference between isomers disposition. Conclusions R-to-S conversion rate could be mainly associated with several passages of the drug through the intestinal mucosa. The concentration–time profiles of ketoprofen in plasma after the administration of both formulations evidenced R-to-S conversion of recirculating drug following meal intakes. A bacterial strain (Chol-1ST) that is able to oxidize cholesterol to CO2 and reduce nitrate to dinitrogen was enriched and isolated from an upflow sludge bed (USB) anoxic reactor that treats sanitary landfill leachate from the city of Montevideo, Uruguay. The presence of ubiquinone Q-8 as the sole respiratory lipoquinone indicated that strain Chol-1ST belonged to the b-subclass of the Proteobacteria. Phosphatidylethanolamine was the predominant polar lipid and the G+C content of the DNA was 65?3 mol%. The fatty acid profile of strain Chol-1ST, cultivated under denitrifying conditions by using a defined mineral medium supplemented with cholesterol, was characterized by the following major components: summed feature 4 (C16 : 1v7c and/or iso C15 : 0 2-OH), C16 : 0, C18 : 1v7c and hydroxy acid C10 : 0 3-OH. Stille reaction was performed with several halo cis-diol derivatives by reaction with allyltributyltin in the presence of a palladium catalyst forming allyl cis-dihydrodiol derivatives. These couplings were conducted with conventional heating as well as with microwave irradiation. Allylbenzene cis-dihydrodiol was obtained with excellent yield using mild conventional heating. However, if the diol moiety is protected with the isopropylidene group, the expected product is obtained only under microwave irradiation. The unusual reactivity observed for the polyoxygenated derivatives suggests assistance of the free hydroxyls in the catalytic cycle. Wet-mixing processes can be tuned to reduce energy consumption, although impacts on quality and stability should be addressed. This work evaluated the effect of wet-mix processing (total solids, TS: 50 or 60%; heat treatment: 75 or 100 C 18 s) and storage conditions (open package: 25 C, 58% RH, 4 weeks; closed package: 25 C, multilayer bag, 12 weeks) on the physicochemical stability and digestibility of infant formulas. Processing significantly influenced the physicochemical characteristics of the powders, with TS exerting a stronger impact than heat treatment. Powders produced from 60% TS wet mixes presented the fastest water sorption, which accelerated lactose crystallisation and fat liberation. Formulas in closed packages were stable for 12 weeks. While lactose crystallised before week 2 in openpackage storage, inducing major physical changes, only limited effects were observed on digestibility. These findings provide valuable insights to ensure consistent quality during shelf-life of infant formula. Contamination of honey with toxic pyrrolizidine alkaloids (PAs) and their N-oxides (PANOs) was assessed by dilution with acidic methanol and analysis through liquid chromatography (LC) coupled with tandem mass spectrometry (MS/MS) in three different modes. The hybrid linear trap/triple quadrupole (LC-QTRAP) instrument was used in precursor ion scan (PIS), enhanced product ion scan (EPI), and multiple reaction monitoring (MRM) mode. The parent ions from ions at m/z 120 or 138 amus, characteristic of all the toxic PAs and PANOs in the sample were first scanned by PIS. Then, the presence of each contaminant at specific retention times through its MS2 spectrum was confirmed by EPI. Finally, they were quantified in the MRM mode. The method was validated: recoveries 86–111%, relative standard deviation (RSD) <20%, at 20 and 40 μg/kg, except retrorsine, which showed a RSD of 30% at 20 μg/kg. Honey samples were analyzed and five of them showed levels of 40 μg/kg for the sum (PAs + PANOs). This approach allows the simultaneous determination of PAs and PANOs in honey, even if their chemical standards are not available.
