This work investigated the effect of temperature on the reaction efficiency of soybean oil transesterification in supercritical ethanol in a continuous catalyst-free process under different water concentrations and at varying substrate flow rates. Experiments were performed in the temperature range from 250 to 325 &deg;C, at 20 MPa, with an oil to ethanol molar ratio of 1:40. Results showed that temperature and substrate flow rates strongly affected the reaction conversion to fatty acid ethyl esters, decomposition, and trans-isomerization of unsaturated fatty acids, mainly for C18:2 and C18:3. Reduction of disulfide bonds and introduction of &ldquo;de novo&rdquo; thiol groups in cyclodextrin glucantransferase from Thermoanaerobacter sp. were assessed in order to perform reversible covalent immobilization onto thiol-reactive supports (thiolsulfinate-agarose). Only the thiolation process dramatically improved the immobilization yield, from 0 % for the native and reduced enzyme, up to nearly 90 % for the thiolated enzyme. The mild conditions of the immobilization process (pH 6.8&ndash;7.0 and 22 &deg;C) allowed the achievement of 100 % coupling efficiencies when low loads were applied. Ionic strength was a critical parameter for the immobilization process; for high activity recoveries, 50 mM phosphate buffer supplemented with 0.15 M NaCl was required. The kinetic parameters, pH and thermal stabilities for the immobilized biocatalyst were similar to those for the native enzyme. For &beta;-cyclization activity, optimal pH range and temperature were 4.0&ndash;5.4 and 85 &deg;C. Carbapenemase production in Enterobacterales clinical isolates is a global threat. Multi-drug resistant Klebsiella pneumoniae harboring carbapenemases are a major concern among the hospital settings in Latin America. Aim: The aim of this study was to analyze the genetic relatedness between three isolates of K. pneumoniae recovered from one patient in the same bacteriological round on the same day, which exhibited different susceptibility profiles to carbapenems (CP) and to colistin (Col). Isolates&rsquo; profiles were as follows (susceptible- S/resistant-R): CPS/ColR, CPR/ColR, and CPR/ColS. Pulse-field gel electrophoresis, multilocus sequence typing, and whole genome sequencing were performed. Conjugation assays were carried out and PCR determination in transconjugants (Tcs) was made for: blaCTX-M-groups, blaNDM, blaKPC, blaTEM, qnr alleles, aac(6&cent;)Ib-cr, ermB, and plasmid incompatibility groups (Inc). Results: All isolates belonged to the same clone, to ST258 and harbored blaCTX-M-14, blaCTX-M-15, qnrA1, qnrB1, aac(6&cent;)Ib-cr, and wzi154 (capsule-locus KL107). One isolate had additional wzi gene, wzi109 (capsulelocus KL36). In CPR isolates, the pattern was explained for blaNDM-1 or blaNDM-1/blaKPC-2 presence, and in ColR for IS5-like element insertion in mgrB at different positions. Co-mobilization of blaNDM-1/qnrA1 was associated to a different plasmid Inc (A/C-FII) in both blaNDM-1 donors. Mobilization of blaCTX-M-14 was related to IncI1 in one donor. Conclusion: These findings highlight the potential plasticity of ST258 K. pneumoniae clone. En el suero de humanos y murinos sanos se detectan anticuerpos contra antigenos propios, llamados autoanticuerpos naturales (revisado por Avrameas, 1991). En particular, en humanos se distinguen las aglutininas frias que reconocen el antigeno eritrocitario I (Wiener y co!., 1956). Se ha propuesto que las aglutininas frias naturales podrian participar en la eliminaci6n de eritrocitos envejecidos (Gofii y co!., 1990). Esta idea se apoya en el hecho de que el tratamiento de eritrocitos in vitro con proteasas aumenta la reactividad de las aglutininas frias, por 10 cual pod ria tratarse de antigeno previamente inaccesible. Se sugiere que in vivo podria ocurrir algun mecanismo semejante que permitiera exponer mayor concentraci6n de antigeno I en eritrocitos envejecidos. <br /><br />En este trabajo se estudi6 si las aglutininas frias reaccionan en forma diferencial frente a los eritrocitos con distinto grado de envejecimiento. <br /><br />Para ello en primer lugar se separaron eritrocitos de diferente grado de envejecimiento, utilizando gradientes de densidad, ya que esta descrito que existe correlaci6n entre densidad y edad. El grado de envejecimiento de las distintas fracciones se evalu6 de acuerdo a algunos marcadores de edad anteriormente descritos (Branch y co!., 1983). Con un nivel de significaci6n del 5 % se encontraron diferencias significativas, tanto en el porcentaje de reticulocitos como en el porcentaje de hemoglobina glicosilada, entre las poblaciones de edades extremas y la media de los eritrocitos. <br /><br />En segundo lugar, se estudi6 la interacci6n entre las aglutininas frias y los eritrocitos de edades extremas. Para ello se optimiz6 un ensayo de alta sensibilidad que permiti6 determinar el numero de sitios de antigeno I por eritrocito. Esto requiri6 en primer lugar purificar la aglutinina fria y en segundo lugar marcarla con 1251. Los resultados sugieren que la incorporaci6n de iodo en residuos de tirosina, ligadas al sitio activo, afect6 su actividad bio16gica en tanto que en residuos de lisina permaneci6 inalterada. <br /><br />En terminos generales se encontr6 que el numero de sitios de antigeno I por eritrocito en adultos normales fue de (27.000 &plusmn; 9.000) en tanto que con eritrocitos tratados con papain a fue de (80.000 &plusmn; 20.000), siendo en cada individuo el numero de sitios de antigeno I en eritrocitos sin tratar entre un medio y un tercio de los expuestos luego del tratamiento. Esto permite realizar transformaciones espec&iacute;ficas sobre los sustratos y obtener productos que muchas veces no son accesibles por m&eacute;todos cl&aacute;sicos, y principalmente permite obtener compuestos enantiom&eacute;ricamente puros de gran inter&eacute;s a nivel industrial. Entre las principales transformaciones en qu&iacute;mica org&aacute;nica se encuentran las reacciones de oxidaci&oacute;n, lo cual ha impulsado el estudio de las oxidorreductasas como biocatalizadores de inter&eacute;s, y es dentro de este grupo que se encuentran las dioxigenasas tipo Rieske con las que se trabaj&oacute; en esta tesis. Estas enzimas catalizan el primer paso para la degradaci&oacute;n de compuestos arom&aacute;ticos en bacterias, cuando son utilizados como fuente de carbono y energ&iacute;a. En 1968 el grupo del Dr. Gibson describi&oacute; la actividad dioxigenasa en una cepa mutante de Pseudomonas putida y a partir de ese momento comenzaron los estudios de esta vasta familia de enzimas, a la cual se le reconoci&oacute; un gran potencial en qu&iacute;mica sint&eacute;tica siendo utilizadas hasta el d&iacute;a de hoy para la producci&oacute;n de materiales de partida quirales. De los diferentes miembros de dicha familia de enzimas, el sistema Tolueno Dioxigenasa (TDO) es uno de los m&aacute;s utilizados a escala preparativa, pero no el m&aacute;s estudiado en cuanto a estructura del sitio activo y posible alteraci&oacute;n de su regio- o estereoselectividad. En esta tesis se abord&oacute; el estudio de tales aspectos, seleccionando por alineamiento de secuencias aquellos residuos del sitio activo identificados como claves en otras enzimas tipo Rieske, como Naftaleno &ndash; y Bifenilo Dioxigenasas (NDO y BPDO). El desarrollo de cuatro variantes de TDO (Q215A, I324F, T365N y F366V) permiti&oacute; evaluar la influencia de estos residuos en las propiedades de la enzima, identificando algunos importantes para la quimio- y estereoselectividad de la reacci&oacute;n de dihidroxilaci&oacute;n. Their action is counteracted by di�erent resistance mechanisms developed by bacteria. Among them, the most common strategy is the expression of �-lactamases, enzymes that hydrolyze the amide bond present in all �-lactam compounds. There are several inhibitors against serine-�-lactamases (SBLs). Metallo-�-lactamases (MBLs) are Zn(II)-dependent enzymes able to hydrolyze most �-lactam antibiotics, and no clinically useful inhibitors against them have yet been approved. Despite their large structural diversity, MBLs have a common catalytic mechanism with similar reaction species. Here, we describe a number of MBL inhibitors that mimic di�erent species formed during the hydrolysis process: substrate, transition state, intermediate, or product. Recent advances in the development of boron-based and thiol-based inhibitors are discussed in the light of the mechanism of MBLs. Expanding the reaction scope of natural metalloenzymes can provide new opportunities for biocatalysis. Mononuclear non‐heme iron‐dependent enzymes represent a large class of biological catalysts involved in the biosynthesis of natural products and catabolism of xenobiotics, among other processes. Here, we report that several members of this enzyme family, including Rieske dioxygenases as well as &alpha;‐ketoglutarate‐dependent dioxygenases and halogenases, are able to catalyze the intramolecular C&minus;H amination of a sulfonyl azide substrate, thereby exhibiting a promiscuous nitrene transfer reactivity. One of these enzymes, naphthalene dioxygenase (NDO), was further engineered resulting in several active site variants that function as C&minus;H aminases. Furthermore, this enzyme could be applied to execute this non‐native transformation on a gram scale in a bioreactor, thus demonstrating its potential for synthetic applications. These studies highlight the functional versatility of non‐heme iron‐dependent enzymes and pave the way to their further investigation and development as promising biocatalysts for non‐native metal‐catalyzed transformations. Galactomannans from the endosperm of mature seeds of Prosopis affinis were isolated following different extraction procedures in yields of up to 64%. The polysaccharides were characterized by HPLC, GC-MS, and NMR spectroscopy, and their physicochemical properties were studied by SEC/MALLS-QUELS/RI and rheological methods. The monosaccharide composition of the extracted galactomannans showed only mannose and galactose in a &sim;1.5 ratio, similar to galactomannans found in guar gum and obtained from other Prosopis species. The structural characterization showed that these polysaccharides are composed by a &beta;-(1&rarr;4)-linked mannose backbone with galactopyranosyl residues attached through &alpha;-(1&rarr;6) linkages. The weight-average molecular weight (Mw) of the P. affinis galactomannans went from (1.6 &plusmn; 0.2) x 106 to (1.8 &plusmn; 0.1) x 106 g/mol, and the intrinsic viscosity, in the 9.9 to 10.7 dL/g range, was comparable to that reported for other extracts from the Prosopis genre. Although the molecular weight distribution and intrinsic viscosity were dependent on the extraction procedure, the mannose-to-galactose ratio was almost invariant. Conformation plots showed that the polymer extracted at higher temperatures is more cohesive, a characteristic that could be attributed to a greater number of polymer-polymer interactions. Our findings show that the P. affinis galactomannan has nearly monomodal molecular weight distribution, functional properties similar to those of guar gum, and can be isolated from an abundant and readily available non-traditional source. It therefore represents a promising new biomaterial for the food and pharmaceutical industries that can be produced from the regional native flora in a sustainable way. Conjugated linoleic acid (CLA) is a prime substrate for intra-gastric nitration giving rise to the formation of nitro-conjugated linoleic acid (NO2-CLA). Herein, NO2-CLA generation is demonstrated within the context of acute inflammatory responses both in vitro and in vivo. Macrophage activation resulted in dose- and time-dependent CLA nitration and also in the production of secondary electrophilic and non-electrophilic derivatives. Both exogenous NO2-CLA as well as that generated in situ, attenuated NF-&kappa;B-dependent gene expression, decreased pro-inflammatory cytokine production and up-regulated Nrf2-regulated proteins. Importantly, both CLA nitration and the corresponding downstream anti-inflammatory actions of NO2-CLA were recapitulated in a mouse peritonitis model where NO2-CLA administration decreased pro-inflammatory cytokines and inhibited leukocyte recruitment. Taken together, our results demonstrate that the formation of NO2-CLA has the potential to function as an adaptive response capable of not only modulating inflammation amplitude but also protecting neighboring tissues via the expression of Nrf2-dependent genes. Los inhibidores pept&iacute;dicos de la enzima convertidora de angiotensina-I (ECA-I) son muy interesantes por ser compuestos naturales con efecto antihipertensivo. Muchos p&eacute;ptidos bioactivos han sido obtenidos por hidr&oacute;lisis enzim&aacute;tica de prote&iacute;nas alimentarias y las prote&iacute;nas l&aacute;cteas han tenido un papel destacado. Sin embargo, las peptidasas empleadas han sido fundamentalmente enzimas comerciales de origen animal o microbiano. El uso de peptidasas de plantas, obtenidas de fuentes menos convencionales, podr&iacute;a liberar nuevos p&eacute;ptidos inhibidores de ECA-I o aquellos que se sabe forman parte de la secuencia de las prote&iacute;nas del LS pero que a&uacute;n no han sido liberados con las enzimas cl&aacute;sicas. Las peptidasas de una especie de Bromeli&aacute;cea aut&oacute;ctona, recientemente caracterizadas por nuestro grupo de investigaci&oacute;n, presentan caracter&iacute;sticas interesantes para la hidr&oacute;lisis del LS y a&uacute;n no han sido utilizadas para la preparaci&oacute;n in vitro de hidrolizados de prote&iacute;nas. En este sentido, se estudi&oacute; el potencial de las peptidasas de Bromelia antiacantha Bertol., para hidrolizar el LS y evaluar la actividad inhibidora de la ECA-I. Demostramos que las peptidasas de B. antiacantha fueron capaces de hidrolizar a las prote&iacute;nas del LS con un importante efecto del pH de reacci&oacute;n para modificar la susceptibilidad de hidr&oacute;lisis de &alpha;-lactalbumina (ALA) y &beta;-lactoglobulina (BLG), las prote&iacute;nas mayoritarias del LS. Se obtuvieron hidrolizados de LS, ALA y BLG con potente actividad inhibitoria de ECA-I. En todos los casos, la fracci&oacute;n pept&iacute;dica de tama&ntilde;o menor a 2 kDa present&oacute; una actividad inhibitoria de ECA-I potente y esta fue resistente a un proceso de digesti&oacute;n gastrointestinal simulado in vitro. A partir del hidrolizado de ALA se capturaron por cromatograf&iacute;a de afinidad y posteriormente se identificaron por espectrofotometr&iacute;a de masas dos nuevos p&eacute;ptidos derivados de ALA con actividad inhibitoria de ECA-I. A partir de estudios de cin&eacute;tica de inhibici&oacute;n de ECA-I fue posible establecer que el p&eacute;ptido Thr-Thr-Phe-His-Thr-Ser-Gly-Tyr (TTFHTSGY) se comport&oacute; como un inhibidor total, mientras que el p&eacute;ptido Gly-Tyr-Asp-Thr-Gln-Ala-Ile-Val-Gln (GYDTQAIVQ) fue capaz de inhibir parcialmente a ECA-I y a concentraciones elevadas de inhibidor por lo que no se continu&oacute; su caracterizaci&oacute;n. Sin embargo, hay dos secuencias con actividad inhibitoria de ECA-I reportada encriptadas en el p&eacute;ptido GYDTQAIVQ (GY e IVQ) que podr&iacute;an ser liberadas durante su digesti&oacute;n gastrointestinal, convirti&eacute;ndolo en productos potencialmente m&aacute;s activos in vivo. Angiotensin converting enzyme (ACE) plays a key role in regulating blood pressure and ACE inhibitors are first-line drugs for treating hypertension. Food protein-derived peptides with ACE inhibitory activity are natural and safe alternatives for both prevention and treatment of hypertension. The second most abundant whey protein, &alpha;-lactalbumin (ALA) was hydrolysed with proteases from Bromelia antiacantha Bertol., a native plant. Two new peptides with potent ACE-inhibitory activity: TTFHTSGY (IC50 = 142 &mu;M) and GYDTQAIVQ (IC50 = 1.0 mM) were purified by affinity chromatography and sequences were determined by mass spectrometry. In silico analysis indicated that neither of these peptides was toxic (ToxinPred). Inhibition kinetic analysis showed that TTFHTSGY was a fully functional ACE competitive inhibitor, whereas GYDTQAIVQ caused only partial ACE inhibition. Our results indicate that cysteine peptidases from B. antiacantha hydrolyse ALA to produce novel ACE inhibitory peptides that could be useful even as functional food ingredients.
