Abstract
A summary of the resource.
The interaction of lanthanide ions with biologically active ligands is of the utmost relevance to understand and develop new complexes based on these ions. In this paper the thermodynamic and structural properties of Ln(III) complexes with amino acids and peptides is critically reviewed. Mixed-ligand and heteropolynuclear complexes containing amino acids or peptides in the coordination sphere are also included. Equilibrium data in aqueous solution have been collected, showing the formation of species with low thermodynamic stability. The solid state structures are dominated by the formation of carboxylate bridges which connect lanthanide cations. Different nuclearities are found, from discrete dimers to infinite 2D structures.

Abstract
A summary of the resource.
An immunoadjuvant preparation (named Fraction B) was obtained from the aqueous extract of Quillaja brasiliensis leaves, and further fractionated by consecutive separations with silica flash MPLC and reverse phase HPLC. Two compounds were isolated, and their structures elucidated using a combination of NMR spectroscopy and mass spectrometry. One of these compounds is a previously undescribed triterpene saponin (Qb1), which is an isomer of QS-21, the unique adjuvant saponin employed in human vaccines. The other compound is a triterpene saponin previously isolated from Quillaja saponaria bark, known as S13. The structure of Qb1 consists of a quillaic acid residue substituted with a -D-Galp-(1!2)-[ -D-Xylp-(1!3)]- -D-GlcpA trisaccharide at C3, and a -D-Xylp-(1!4)- -L-Rhap-(1!2)-[ -L-Arap-(1!3)]- -D-Fucp moiety at C28. The oligosaccharide at C28 was further substituted at O4 of the fucosyl residue with an acyl group capped with a -D-Xylp residue.

There is also an intramolecular hydrogen bond O2--H1...O3.

First |b we describe the isolation of three new diterpenoids with a hydroazulene skeleton |b dictyotriol C(1) D(2) |b and E(3). as well as the previously known pachydictyol A(4) dictyol B(5) dictyol C(6) and dictyotadiol(7) from a Dictyota sp. The structures of the new diterpenoids 1-3 were proposed on the basis of their spectral data and confirmed by chemical transformation of dictyol B(5). This is followed by absolute stereochemistries of the new compounds as well as that of the known dictyol B(5) and dictyotadiol (7) were determined. These absolute configurations were opposite to that reported for pachydictyol A(4).

A series of indazole N1-oxide derivatives has been spectroscopically studied in solution using 1H, 13C, and 15N NMR based on pulsed field gradient selected PFG 1HAX (X=13C and 15N) gHMQC and gHMBC experiments. Some indazoles were prepared using a new methodology to compare its spectral and structural data with the indazole N1-oxide parent compounds. The 13C resonances of the indazole N1-oxide carbon 3 and 7a demonstrate the N-oxide push-electron capability. The 15N resonances of the indazole N-oxide, nitrogen 1, are near to 30 ppm more shielded than the corresponding values in the indazole heterocycle (deoxygenated form). Moreover, the structures of one indazole and one indazole N-oxide were unambiguously confirmed by X-ray crystallography. The solid state structures were contrasted with the theoretical ones obtained in vacuo at different calculus level. The aromaticity of the derivatives was studied analyzing the HAH coupling constants of indazole’s aromatic hydrogens and measuring CAC distances in the solid state. The fragmentation that takes place in EI/MS was gathered for all the indazole N-oxide derivatives and the general fragmentation pattern analyzed.

The B rings have a sofa conformation. The two fused rings show a cis form of coupling. Despite the disorder both furan rings are planar. The hydroxyl groups are involved in hydrogen bonds with the carbonyl O atoms.

<p>An experimental investigation of the structure and crystal-chemistry of the triple perovskite BaSr2MgTa2O9 is reported. Polycrystalline BaSr2MgTa2O9 was synthesized via solid-state reaction and its structure quantitatively probed at 10 and 300 K using neutron diffraction. Monoclinic (A2/m, a0 b– b–) and trigonal structural models (P3c1, a–a–a–) were fit to diffraction data. The former was found to correctly reproduce experimental intensities, while the latter failed to do so. This difference is attributed to the ability of the monoclinic model to accurately reflect octahedral tilting. BaSr2MgTa2O9 features corner-sharing MgO6 and TaO6 octahedra with barium and strontium cations occupying cubooctahedral holes. Barium and strontium are disordered over the A site, whereas B site cations magnesium and tantalum are ordered. Chemical substitution of strontium for barium in Ba3MgTa2O9 induces tilting of MgO6 and TaO6 octahedra in two directions and geometric distortions<br />of MgO6 octahedra.</p>

These com pounds are active against Caenorhabditis elegans and parasitic nematodes by selectively inhibiting nematode complex II, and they display low toxicity in mammalian cells and vertebrate organisms. Although a big number of benzamides were tested against C. elegans in high-throughput screens, bioisosteres of the amide moiety were not represented in the chemical space examined. We thus iden tified an opportunity for the design, synthesis and evaluation of novel compounds, using bioisosteric replacements of the amide group present in benzamides. The compound Wact-11 was used as the reference scaffold to prepare a set of bioisosteres to be evaluated against C. elegans. Eight types of amide replacement were selected, including ester, thioamide, selenoamide, sulfonamide, alkyl thio and oxo-amides, urea and triazole. The results allowed us to perform a structure–activity relationship, highlighting the relevance of the amide group for nematicide activity. Experimental evidence was complemented with in silico structural studies over a C. elegans complex II model as a molecular target of benzamides. Importantly, compound Wact-11 was active against the flatworm Echinococcus granulosus, suggesting a previously unreported pan-anthelmintic potential for benzamides.

The eukaryotic translation Elongation Factor 2 (eEF2) is an essential enzyme in protein synthesis. eEF2 contains a unique modification of a histidine (His699 in yeast; HIS) into diphthamide (DTA), obtained via 3-amino-3-carboxypropyl (ACP) and diphthine (DTI) intermediates in the biosynthetic pathway. This essential and unique modification is also vulnerable, in that it can be efficiently targeted by NAD+-dependent ADP-ribosylase toxins, such as diphtheria toxin (DT). However, none of the intermediates in the biosynthesis path is equally vulnerable against the toxins. This study aims to address the different susceptibility of DTA and its precursors against bacterial toxins. We have herein undertaken a detailed in silico study of the structural features and dynamic motion of different His699 intermediates along the diphthamide synthesis pathway (HIS, ACP, DTI, DTA). The study points out that DTA forms a strong hydrogen bond with an asparagine which might explain the ADP-ribosylation mechanism caused by the diphtheria toxin (DT). Finally, in silico mutagenesis studies were performed on the DTA modified protein, in order to hamper the formation of such a hydrogen bond. The results indicate that the mutant structure might in fact be less susceptible to attack by DT and thereby behave similarly to DTI in this respect.

Electronic, lipophilic and steric descriptors included in QSAR-2D and -3D are analyzed for a set of ortho- and para-naphthoquinones that have proved to be powerful oxidative agents with potent trypanocidal activities specially against Leptomonas seymouri and Trypanosoma cruzi. Electronic properties are calculated by means of semiempirical (PM3), ab initio (HF/3-21G) and density functional theory (B3LYP/6-31 + G∗) methodologies. Three different electronic states, neutral quinones, hydroquinones and semiquinones, are studied to investigate if any one of them are statistically related with the biological activities. The best correlations were obtained at the B3LYP level of theory because it includes electronic correlation.<br /><br />The QSAR-2D indicates that the best trypanocidal growth inhibitors are molecules in the semiquinone electronic state, with the following properties: (a) high negative value of EHOMO, (b) high negative charge in the oxygen atoms of the carbonyl groups, (c) high positive charge in the carbon atom of one of carbonyl moieties and (d) high electronegativity (χ). In a complementary way, the QSAR-3D indicates that the electrostatic field correlates with trypanocidal activity and the presence of bulk moieties would increase activity.<br /><br />The idea of comparing the three electronic states may prove to be of most importance in the general strategy to the design of new trypanocidal drugs. In fact, the experimental results showed that semiquinone is the one really statistically relevant indicating a clear connection between biochemical and theoretical aspects. Finally, we demonstrated that to be a good anti-trypanosomatid compound, the molecule must be a good electron acceptor to reach easily the essential semiquinone state. We expect that the present results motivate new experimental as well as theoretical investigations that confirm our findings.<br /><br />

Five C13-norisoprenoid glycosides, isolated from lulo (Solanum quitoense) leaves, were subjected to enzymatic hydrolysis with a commercial glucosidase (emulsin) and also with an enzymatic preparation having glycosidase activity, isolated from lulo fruit pulp. The volatile compounds generated after reaction were characterized by capillary GC and capillary GC–MS. Lulo fruit glycosidases were extracted by ammonium sulfate precipitation at pH 6.5 and subjected to fractionation by hydrophobic interaction chromatography (HIC) on phenyl Sepharose1 gel, and some of their properties were determined. As the result of enzymatic hydrolysis with exogenous enzymes (emulsin), glycosides isolated from lulo leaves produced only the aglycone (glycosidically bound volatile); in contrast, enzymatic hydrolysis of glycosides, using endogenous enzymes (lulo fruit glycosidases), generated additional volatile compounds, some recognisable as components of fruit flavour. The role of lulo leaf glycosides as flavour precursors was confirmed.

In this work, the scope and limitations of Hantzsch, modified Hantzsch and Kelly methodologies for the synthesis of amino acid derived thiazoles are presented. In addition, the syntheses of bisthiazoles as key fragments of natural products and analogs are described. The Kelly’s methodology followed by oxidation provides the desired N-Cbz protected thiazole after purification. According with our results the Fmoc or Boc proctecting groups are not compatible with the conditions used in this methodology. Modifications of the temperature and reagents used in the Hantzsch thiazole synthesis enabled the preparation of chiral thiazole building blocks without racemisation and in good yields.

The larvae of the cestodes belonging to the genus Echinococcus are outwardly protected by the laminated layer (LL), a crucial but poorly understood carbohydrate-rich acellular structure. Carbohydrate structural data strongly suggest that the main components of the LL are mucins. The most massive LL in the genus is featured by E. granulosus, agent of cystic hydatid disease. No appropriate methods existed to date for the solubilisation of the E. granulosus LL and the electrophoretic visualisation of its proposed structural mucins. We report that reduction of disulphides greatly aids LL disassembly, resulting in almost full solubilisation in combination with moderate sonication. The structural mucins can then be visualised by agarose electrophoresis and blotting with galactose-binding lectins, which also react strongly with the LL in tissue sections. A substantial portion of the material migrates as if positively charged; since the LL glycans are neutral, this may correspond to mucins with cationic peptide backbones.

The combined use of metallic nanoparticles with protective systems has made it possible to manufacture new materials with the desired properties and besides, are a class of nanostructured materials that have recently received attention due to their interesting properties and wide range of applications in catalysis, biology, materials chemistry, sensors, among others. Silver (Ag)-coated iron (Fe) nanoparticles as functional antimicrobial agents have become an important research topic. However, there are synthesis methods and important properties and characteristics that still need to be investigated. In this study, a simple synthesis route for Fe2O3-Ag nanoparticles is presented using the microemulsion reaction (MR) technique, since these systems can be applied as antimicrobial agents in biological processes and subsequently recovered through the use of a magnet. The results obtained to date demonstrate the ability to use the MR method to design and synthesize nanoparticles composed of metals that synergize their properties.

Abstract
A summary of the resource.
New 5-nitroindazole derivatives were developed and their antichagasic properties studied. Eight compounds (14–18, 20, 26 and 28) displayed remarkable in vitro activities against Trypanosoma cruzi (T. cruzi). Its unspecific cytotoxicity against macrophages was evaluated being not toxic at a concentration at least twice that of T. cruzi IC50, for some derivatives. The electrochemical studies, parasite respiration studies and ESR experiment showed that 5-nitroindazole derivatives not be able to yield a redox cycling with molecular oxygen such as occurs with nifurtimox (Nfx). The study on the mechanism of action proves to be related to the production of reduced species of the nitro moiety similar to that observed with benznidazole.
