Several recombinant clones expressing antigens from Echinococcus granulosus were isolated previously from a parasite cDNA library using cystic hydatid disease (CHD) patients’ sera or rabbit hyperimmune antiserum against a lipoproteic fraction from bovine cyst fluid. Six of these antigens were expressed in Escherichia coli and the purified recombinant proteins were tested in enzyme-linked immunosorbent assay (ELISA) for specific IgG with a panel of sera from patients with surgically confirmed ( n = 58) or immunologically diagnosed ( n = 71) CHD. Sera from clinically normal individuals ( n = 203) and sera from individuals with other helminthic infections ( n = 65) were assayed for the assessment of specificity. A cut-off value was determined by receiver-operating-characteristic plots for each antigen. A recombinant antigen B subunit (AgB8/2) presented the highest sensitivity (93·1%), considering the group of sera from patients with CHD surgically confirmed, and specificity (99·5%) and is proposed as the basis for an immunodiagnostic test. The other recombinant antigens tested presented sensitivities between 58·6% and 89·7%, and three of them were considered of complementary value. In subclass-specific ELISA, different IgG isotypes showed dominance in the response for each of the recombinant antigens. There was a clear predominance of IgG4 response for all antigens tested, indicating that this would be the subclass of choice to be assessed for these recombinant proteins. We describe the first outbreak of Klebsiella pneumoniae carbapenemase-producing K. pneumoniae (KPC-KP), the infection control measures adopted and the shift in resistance patterns of isolates during antibiotic treatment. The ST258 KPC-KP strain exhibited a multiresistant antibiotic phenotype including co-resistance to gentamycin, colistin and tigecycline intermediate susceptibility. A progressive increase in the aminoglycosides (acquiring amicacin resistance) and β-lactam MICs, and a decreased susceptibility to fosfomycin was observed throughout the administration of combined antimicrobial regimens including meropenem. A high meropenem resistance KPC-KP homogeneous population (MIC 256 Jg/mL), could arise from the meropenem heterogeneous low-level resistance KPC-KP population (MIC 8 Jg/mL), by the selective pressure of the prolonged meropenem therapy. The kpc gene was inserted in a Tn4401 isoform a, and no transconjugants were detected. The core measures adopted were successful to prevent evolution towards resistance dissemination. La hipoalfalipoproteinemia se define como un trastorno que afecta al metabolismo de las partículas de HDL, alterando su composición, concentración y el tamaño de las mismas. El término hipoalfalipoproteinemia primaria corresponde a concentraciones plasmáticas de HDL-Cpor debajo del percentil 10 con un componente genético. El bajo HDL es uno de los factores de riesgo aterogénico mejor conocidos y forma parte de las dislipemias más comunes generadoras de enfermedades cardiovasculares en la población occidental. En las últimas 6 décadas, estas enfermedades son la principal causa de muerte en nuestro país. Se sabe que existe una relación inversa entre los niveles plasmáticos de las lipoproteínas de alta densidad y el riesgo de cardiopatía isquémica. Se calcula que entre 40 y 60% de la variación en los niveles de las HDL está determinada genéticamente y existe evidencia de que son varios los genes que podrían participar en la regulación de este rasgo fenotípico. Se sabe cuáles son las causas genéticas de esta patología. La ApoAI es la apolipoproteina más importante del HDL constituyendo el 70 % de su masa proteica, siendo el gen apoAI uno de los candidatos más importantes para estudiar la variabilidad genética de los niveles de HDL. Una causa más rara de deficiencia de HDL se debe a mutaciones en el transportador de membrana ATP Binding Casette 1 (ABCA1). Esta proteína interactúa directamente con la ApoAI para promover el eflujo de colesterol celular. Otro gen candidato es el que codifica la lecitin colesterol acil transferasa (LCAT). Esta enzima es la encargada de esterificar el colesterol de las HDL para generar las partículas maduras. Estas tres moléculas, ApoAI, ABCA1 y LCAT, parecen tener un papel fundamental en el transporte reverso del colesterol desde las células periféricas hacia el hígado y otros tejidos. Nuestro objetivo ha sido elaborar una metodología que contribuya al conocimiento de los componentes genéticos de la hipoalfalipoproteinemia primaria. Esto representa un campo enorme de investigación de vital importancia para la prevención y el tratamiento de la aterosclerosis y la cardiopatía isquémica en nuestro país. Huntington's disease (HD) is a hereditary neurodegenerative disorder. The genetic cause is an expansion of CAG repeats located in the IT15 gene. Though the number of CAG repeats ((CAG)n) can largely explain the age at onset (AAO) of symptoms, a percentage of its variation could be attributed to modifier genes and to environmental factors. The study aimed to evaluate the influence of genetic modifiers of the AAO of HD including: (CAG)n and del2642 in the IT15 gene, ADORA2A rs5751876, HAP1 rs4523977, PGC1-α rs7665116 and UCH-L1 rs5030732. Eighteen patients with positive family history and HD-suggestive symptoms were recruited. The (CAG)n and gene polymorphisms were determined by different molecular biology techniques. We observed that the (CAG)n influenced in a 64.5% of the variability in the AAO. We also showed that the rs5751876 variant significantly affected this variability. However, the influence of UCH-L1, del2642, HAP1 and PGC1-α gene polymorphisms could not be replicated, perhaps due to small sample size. Genetic studies including the molecular determination of (CAG)n, in addition to other genetic modifiers involved in the variability of the AAO, were first performed in Uruguay. We could replicate in our cohort the anticipation effect on the AAO by the ADORA2A rs5751876. Our results confirm the usefulness of an expanded molecular characterization in HD patients. Justificativa e Objetivos: O estudo objetivou a avaliação da atividade antibacteriana de extratos brutos e frações de Ibicella lutea, na inibição do crescimento de Staphylococcus aureus, avaliando a combinação destes compostos e sua atividade citotóxica. Métodos: Para a atividade antibacteriana dos extratos foi utilizado o Teste de Microdiluição em Caldo, seguido do Teste Checkerboard. Os extratos que apresentaram atividade antibacteriana foram submetidos ao teste de citotoxicidade, com células macrofágicas e determinação do seu Índice de Seletividade (IS). Resultados: A fração de acetato de etila(AcOE) obteve o melhor potencial antibacteriano (6,25 µg/mL), entretanto nenhum dos compostos testados apresentaram atividade bactericida nas concentrações empregadas. Neste estudo pode-se observar uma ação aditiva entre as frações AcOE e metanólica (MeOH), sendo a interação entre os extratos brutos indiferente. De acordo com o teste de citotoxicidade, a fração AcOE apresentou uma maior índice de sobrevivência das células macrofágicas (IC50%=30,35 µg/mL). Entretanto, quando calculado o IS, não houve resultados satisfatórios (IS < 10) para os extratos analisados. Conclusões: Neste estudo observou-se o potencial antibacteriano das frações AcOE e MeOH, assim como uma ação aditiva na combinação das frações, dando suporte para o isolamento e caracterização de seus componentes ativos. Apesar dos extratos não apresentarem um IS satisfatório, novos estudos de toxicidade devem ser realizados para determinar com segurança o potencial de uso dos produtos provenientes de I. lutea, como é o caso de medicamentos para uso tópico ou biocidas. The aim of the present study was to present new evidence supporting the use of saliva as a biological fluid in relative bioavailability studies. Carbamazepine was chosen as a model drug because of its suitability for salivary therapeutic drug monitoring and its well-documented plasma bioavailability. A relative bioavailability study of four different immediate release carbamazepine products was performed. Stimulated saliva samples were collected by chewing on parafilm wax and by the spitting method. In vitro dissolution testing of formulations, using 900 ml of 1% sodium lauryl sulphate in water, was also carried out. The in vitro– in vivo correlations obtained in this salivary study were consistent with previous correlations assessed using plasma. The aim of the present study was to evaluate the suitability of saliva as a biological fluid in relative bioavailability (RBA) studies, with the focus on the statistical design and data variability. A randomized, openlabel, four periods and two sequences (4 9 2) crossover RBA study in saliva of two phenytoin (PHT) 100 mg immediate-release capsules was performed. PHT is a narrow therapeutic index drug that has been widely used for epilepsy treatment for many years. Published information regarding its bioavailability is available, but plasma assessed. This study was designed and performed using saliva as the biological fluid and the simplest conditions that produce coherent results with previously published plasma studies. Pharmacokinetic parameters (Cmax, Tmax, AUC0-t, AUC0-inf, Cmax/AUC0-t, Ke, and t1/2) for each volunteer at each period were calculated. Four different BE calculations were performed: individual bioequivalence, by the method of moments, and three average bioequivalence with data averaged over the two administrations and with data of periods 1–2 and 3–4. ANOVA calculation showed no significant subject-by-formulation interaction, period and sequence effects. The intra-subject variabilities were at least 20-fold lower than the inter-subject ones for Cmax, AUC0-t and AUC0-inf. Rice bran oil was subject to different oxidation period under controlled condition to mimic the composition of the oil used in frying at low, medium and high oxidation degrees. By the hydrotreatment of the produced samples in a batch reactor at 100 bar and 350 ◦C, using Pd/Al2O3 as catalyst, they were converted to hydrocarbons with an efficiency over 95.5 %, regardless their oxidation degree. The chain length distribution of the hydrocarbon indicated that oil conversion took place preferentially through hydrodecarboxylation or hydrodecarbonylation or both combined, instead hydrodeoxigenation. Although the increase in the oxidation degree of the starting oil did not show any significant effect on the hydrocarbon yield, an enhanced effect on cracking was verified, suggesting that oxidized fatty acid chains were more vulnerable to be fractionated. Although all the products showed distillation curves similar to gasoil and over 85.5 % distilled within the diesel range, they reached 95 % distillate above the recommended temperature of 360 ◦C. Thus, they must be previously fractionated to be used as diesel fuels. Results demonstrate the convenience of the performed process for efficiently converting used cooking oils of different qualities to second-generation biogasoil. A key metabolic adaptation of some species that face hypoxia as part of their life cycle involves an alternative electron transport chain in which rhodoquinone (RQ) is required for fumarate reduction and ATP production. RQ biosynthesis in bacteria and protists requires ubiquinone (Q) as a precursor. In contrast, Q is not a precursor for RQ biosynthesis in animals such as parasitic helminths, and most details of this pathway have remained elusive. Here, we used Caenorhabditis elegans as a model animal to elucidate key steps in RQ biosynthesis. Using RNAi and a series of C. elegans mutants, we found that arylamine metabolites from the kynurenine pathway are essential precursors for RQ biosynthesis de novo. Deletion of kynu-1, encoding a kynureninase that converts l-kynurenine (KYN) to anthranilic acid (AA) and 3-hydroxykynurenine (3HKYN) to 3-hydroxyanthranilic acid (3HAA), completely abolished RQ biosynthesis but did not affect Q levels. Deletion of kmo-1, which encodes a kynurenine 3-monooxygenase that converts KYN to 3HKYN, drastically reduced RQ but not Q levels. Knockdown of the Q biosynthetic genes coq-5 and coq-6 affected both Q and RQ levels, indicating that both biosynthetic pathways share common enzymes. Our study reveals that two pathways for RQ biosynthesis have independently evolved. Unlike in bacteria, where amination is the last step in RQ biosynthesis, in worms the pathway begins with the arylamine precursor AA or 3HAA. Because RQ is absent in mammalian hosts of helminths, inhibition of RQ biosynthesis may have potential utility for targeting parasitic infections that cause important neglected tropical diseases. Echinococcus multilocularis metacestodes are fluid-filled, vesicle-like organisms, which are characterized by continuous asexual proliferation via external budding of daughter vesicles, predominantly in the livers of infected individuals. Tumor-like growth eventually leads to the disease alveolar echinococcosis (AE). We employed the monoclonal antibody (MAb) E492/G1, previously shown to be directed against a carbohydrate-rich, immunomodulatory fraction of Echinococcus granulosus, to characterize potentially related components in E. multilocularis. Immunofluorescence studies demonstrated that MAb E492/G1-reactive epitopes were found predominantly on the laminated layer and in the periphery of developing brood capsules. The respective molecules were continuously released into the exterior medium and were also found in the parasite vesicle fluid. The MAb E492/G1-reactive fraction in E. multilocularis, named Em492 antigen, was isolated by immunoaffinity chromatography. Em492 antigen had a protein/carbohydrate ratio of 0.25, reacted with a series of lectins, and is related to the laminated layer-associated Em2(G11) antigen. The epitope recognized by MAb E492/G1 was sensitive to sodium periodate but was not affected by protease treatment. Anti-Em492 immunoglobulin G1 (IgG1) and IgG2 and, at lower levels, IgG3 were found in sera of mice suffering from experimentally induced secondary, but not primary, AE. However, with regard to cellular immunity, a suppressive effect on concanavalin A- or crude parasite extract-induced splenocyte proliferation in these mice was observed upon addition of Em492 antigen, but trypan blue exclusion tests and transmission electron microscopy failed to reveal any cytotoxic effect in Em492 antigen-treated spleen cells. This indicated that Em492 antigen could be modulating the periparasitic cellular environment during E. multilocularis infection through as yet unidentified mechanisms and could be one of the factors contributing to immunosuppressive events that occur at the host-parasite interface. Introduction Quillaja brasiliensis (St. A. ‐Hil. & Tul) Mart (Quillajaceae) is a species native to South America, which is rich in saponins. Saponins are used in different industries, so there is a constant demand for this type of compound. Based on the wide range of applications for the saponins found in this species, notably as immunoadjuvants, we conducted a comprehensive study of this tree and its saponins. Objective The purpose of this work is to complete the characterisation of the immunoadjuvant saponin fraction from Q. brasiliensis leaves and further study the saponin fraction obtained from Q. brasiliensis bark. Methodology Saponin fractions were studied using mass spectrometry in combination with classical methods of monosaccharide and methylation analysis. We performed direct infusion and liquid chromatography/electrospray ionisation ion trap multiple‐stage mass spectrometry (DI‐ESI‐IT‐MSn and LC‐ESI‐IT‐MS2). Results Seventy‐five saponins, 21 from leaves and 54 from bark, were tentatively identified according to their molecular mass, fragmentation pattern and chromatographic behaviour. This work represents the first investigation of saponins from the bark of Q. brasiliensis and some of them presented new structural motifs not previously reported in the genus Quillaja. Conclusion The efficiency and selectivity of the data dependent LC‐MS2 method allowed the rapid profiling of saponins from Q. brasiliensis. The results of the monosaccharide and methylation analysis performed in saponins from Q. brasiliensis fractions and Q. saponaria Molina (Quillajaceae) fraction gives further support to the structures proposed according to the mass spectral data, validating the strategy used in the present work. Peptidomimetic and anti-immunocomplex peptides, which can be readily isolated from a phage-display library, have shown great potential for small-molecule immunoassay development because they typically improve the sensitivity and avoid the use of chemical haptens as coatings or tracer antigens. However, phage-borne peptides are unconventional immunoassay reagents, which greatly limits their use in commercial applications, and require secondary reagents for detection. In order to overcome these limitations, we used C2-15, a peptidomimetic of imidaclothiz, as a model peptide fused to emerald-green fluorescent protein (EmGFP) at the N-terminus (C2-15-EmGFP) and C-terminus (EmGFP-C2-15) to generate novel fluorescent-peptide tracers. Both recombinant fluorophores reacted with similar affinity to the anti-imidaclothiz monoclonal antibody 1E7, but because of its higher expression, C2-15-EmGFP was chosen to develop a competitive magnetic-separation fluorescence immunoassay (MSFIA). After a competitive step with the analyte, the C2-15-EmGFP–antibody complex bound to the magnetic beads was separated with a magnet, and because of the fast dissociation of the peptide–antibody interaction, the fluorescence signal was detected following the spontaneous dissociation of the complex in fresh buffer. The concentration of imidaclothiz causing the 50% inhibitory concentration (IC50) was 11.00 ng mL–1, and the MSFIA performed with excellent recovery and had a good correlation with high-performance liquid chromatography in different matrices.
