Several new pyrazolo[1,5-a]pyrimidines were obtained from the reaction of 1H-5-amino-3-phenylpyrazole (1) with β-dimethylaminopropiophenones 2 in pyridine. The structure elucidation of 6,7-dihydropyrazolo[1,5-a]pyrimidines 3 is based on nmr measurements. These compounds showed moderate anthelmintic in vitro activity against the Nipposirongylus brasiliensis model.

A summary of the resource.
[ReO2(PPh3)2I] has been used as starting material to prepare [RevO2(tn)2]I (tn = trimethylenediamine) in high yield. The complex was characterized by elemental analysis and spectroscopic methods. The complex [ReO2(en)2]I (en = 1,2-diaminoethane) was prepared analogously for comparison. The crystal structure of [ReO2(tn)2]I·H2O was determined from X-ray data. It consists of a six-coordinate rhenium atom bonded to two trans-oxo groups. The amines lie in the equatorial plane, almost perpendicular to the ReO2 core. The average value for the ReO bond is 1.76 Å, similar to that in other Re(V) dioxo complexes. The successive protonations of the oxo groups were determined for both complexes.

In this work, we report the synthesis, characterization and potentiometric studies of new Re(V) complexes of general formula [ReO2(amine)2]+ (amine = 1,2-diaminopropane, 1,4-diaminobutane, N,N′-diethylethylenediamine, diethylenetriamine, triethylenetetramine and N-tris(2-aminoethyl)amine). Compounds were prepared starting from [ReO2I(PPh3)2] and characterized by several techniques. The presence of different substituent groups in the diamines (including alkyl and amine groups) allows the presence of pendant arms. When these groups are free amino groups, they can react with acids such as H+ and M2+ cations and studied by potentiometry. The formation of very stable binuclear species was demonstrated.

Two new rhenium(IV) compounds of formula (AsPh4)(2)[ReCl4(ox)] (1) and [ReCl4(mu-ox)Cu(bipy)(2)] (2) (AsPh4 = tetraphenylarsonium cation, ox = oxalate anion, and bipy = 2,2'-bipyridine) have been synthesized and their crystal structures determined by single-crystal X-ray diffraction. 1 and 2 crystallize in the monoclinic system, space groups P2(1)/c and P2(1)/n, respectively, with a = 22.250(5) Angstrom, b = 11.245(3) Angstrom, c = 19.089(4) Angstrom, beta = 96.59(2)degrees, and Z = 4 for 1 and a = 9.421(2) Angstrom, b= 16.909(4) Angstrom, c = 16.179(4) Angstrom, beta = 93.97(2)degrees, and Z = 4 for 2. The structure of 1 is made up of [ReCl4(ox)](2-) anions and AsPh4+ cations held united by electrostatic forces. Rhenium(IV) is hexacoordinate, with two oxygens of a chelating ox and four chlorine atoms building adistorted octahedron around the metal atom. There is no contact between the [ReCl4(ox)](2-) anions, the shortest Re ... Re and Cl ... Cl distances being 10.345 and 7.209 Angstrom, respectively. This anionic complex is coordinated to a [Cu(bipy)(2)](2+) cation in 2, through one oxalate-oxygen, giving a neutral heterometallic dinuclear unit. The Cu(II) ion shows a very distorted five-coordinated geometry, four bipy-nitrogens occuping the equatorial positions and the oxygen atom the apical one. The basal plane geometry isdistorted toward the tetrahedron, the dihedral angle between the mean planes of the two bipy ligands is 37.6(2)degrees. These [ReCl4(mu-ox)Cu(bipy)(2)] units are arranged in such a way that a chlorine atom of one of them points toward the copper atom of the neighboring one, forming helicoid chains. The intrachain Re Cu distances through chloro and oxalato bridges are 4.658 and 4.798 Angstrom, respectively. The magnetic behavior of 1 and 2 has been investigated over the temperature range 1.8-300 K. 1 is a magnetically diluted Re(IV) complex, the great value of zero-field splitting of the ground level (D = 60 cm(-1)) accounting for the variation of chi(M)T with T in the low-temperature range. 2 behaves as a ferrimagnetic chain, with weak antiferromagnetic interactions between Re(IV) and Cu(II) through oxalato and single chloro bridges.

Abstract
A summary of the resource.
The preparation of the sodium salt of dicyano-acetic acid methyl ester (NaCH3OC(O)C(CN)2) (NaL) is reported. The structure of this anion is related to the structure of the dicyanamide, whose chelating capability has been used to develop 2D networks. Two new complexes of formula [M(L)2(H2O)2] (M=Mn2+ (1) and Cu2+ (2)) have been synthesized and their crystal structures determined by single-crystal X-ray diffraction. The coordination geometry of Mn atom in 1 is approximately octahedral, and the Mn atoms are μ2-bridged by two pairs of L− resulting in an infinite one-dimensional chain. Two types of hydrogen bonds link the chains within a 3-dimensional infinite lattice. The coordination geometry of copper in 2 is tetragonal bipyramidal, showing considerable Jahn–Teller distortion. Each copper atom is connected by four ligands to four different neighboring copper atoms, resulting in sheets of an infinite two-dimensional network. The large separation between the paramagnetic centers in both compounds produces a weak antiferromagnetic interaction.

Abstract
A summary of the resource.
The preparation of trans-[Co(NH3)4(CH3NH2)Br]2+ and trans-[Co(NH3)4(CH3NH2)-(NO3)]2+ complexes is described. The UV-VIS spectra of the complexes indicate a decrease of the ligand field compared to the parent pentaammines. Infrared spectra match with the pattern of the corresponding pentaammines. The catalyzed (by Hg2+) aquation of the trans-bromomethylamine complex go under retention of the stereochemical configuration. The base hydrolysis (studied at 25°C) products show trans to cis rearrangement for both complexes. 1H NMR spectroscopy is used for identification of the stereochemical configuration of the compounds.

Some novel valerolactam derivatives of 5(6)-substituted-(1H-benzimidazol-2-yl-amine) were constructed based on the union of two structural domains with anthelmintic activity. The tested hybrid compounds 7-9 exhibited greater activity using the Nippostrongylus brasiliensis physiology-based in vitro bioassay in comparison to commercial anthelmintic benzimidazoles. Moreover, the improved physicochemical properties of hybrid compounds compared with valerolactam domain allowed the penetration of parasite barriers, concurrently with an increased intraparasitary bioavailability.

Abstract
A summary of the resource.
Several novel semicarbazone derivatives were prepared from 5-nitro-2-furaldehyde or 5-nitrothiophene-2-carboxaldehyde and semicarbazides bearing a spermidine-mimetic moiety. All derivatives presented the E-configuration, as determined by NMR-NOE experiments. These compounds were tested in vitro as potential antitrypanosomal agents, and some of them, together with the parent compounds, 5-nitro-2-furaldehyde and 5-nitrothiophene-2-carboxaldehyde semicarbazone derivatives, were also evaluated in vivo using infected mice. Structure–activity relationship studies were carried out using voltammetric response and lipophilic–hydrophilic balance as parameters. Two of the compounds (1 and 3) displayed the highest in vivo activity. A correlation was found between lipophilic–hydrophilic properties and trypanocidal activity, high RM values being associated with low in vivo effects

Abstract
A summary of the resource.
In a search for antichagasic drugs, 14 new 4-(nitroarylidene)-1,2,6-thiadiazin-3,5-dione 1,1-dioxide derivatives were synthesized and tested in vitro against the epimastigote form of Trypanosoma cruzi and some of them showed important antiprotozoan activity. Attempts to synthesize new 4-(nitroarylidene)-3,5-diamino-1,2,6-thiadiazine 1,1-dioxides were unsuccessful. The antichagasic properties of nitroarylidene-malononitrile and nitroarylidene-cyanoacetamide derivatives, thus obtained, were also tested. The cytotoxic properties against Vero cells of compounds which showed remarkable in vitro antichagasic activity were evaluated. All compounds tested exhibited high toxicity percentages at 100 micrograms/ml. <p>During the last years, the progression to control malaria disease seems to be slowed and WHO (World Health Organization) reported a modeling analysis with the prediction of the increase in malaria morbidity and mortality in sub-Saharan Africa during the COVID-19 pandemic. A rapid way to the discovery of new drugs could be carried out by performing<br />investigations to identify drugs based on repurposing of “old” drugs. The 5-nitrothiazole drug, Nitazoxanide was shown to<br />be active against intestinal protozoa, human helminths, anaerobic bacteria, viruses, etc. In this work, Nitazoxanide and<br />analogs were prepared using two methodologies and evaluated against P. falciparum 3D7. A bithiazole analog, showed<br />attractive inhibitory activity with an EC50 value of 5.9 μM, low propensity to show toxic effect against HepG2 cells at<br />25 μM, and no cross-resistance with standard antimalarials.</p>

A new indole-4,9-dione and their phenoxy derivatives were synthesized and evaluated in vitro against the epimastigote form of Trypanosoma cruzi, Y strain. All of these novel compounds were found to be extremely potent and selective that the standard drug nifurtimox. Interestingly, phenoxyindole-4,9-dione 9d displayed excellent nanomolar inhibitory activity, IC50 = 20 nM, and high selectivity index, SI = 625. In silico studies using MOE program were performed to generate a preliminary pharmacophore model.

A summary of the resource.
With the aim to develop new potential 99mTc-radiopharmaceuticals for imaging hypoxia based on the formation of Tc-nitrido complexes, two novel dithiocarbamate containing metronidazole derivatives (L1 and L2) have been prepared and characterised. The synthesis of L1 and L2 was achieved in excellent yield and high purity. Labelling with 99mTc was successfully performed using a low ligand concentration (approximately 2–3 mg) and the desired products were obtained with high radiochemical purity (>90%). Lipophilicity, plasma protein binding, and biodistribution in normal- and tumour-bearing-CD1 mice studies were performed to asses the potentiality for nuclear medicine oncology. According to the physicochemical and biological behaviour both in healthy animals and in animals bearing solid tumours complex dtcTc1 could be considered as a starting point for the development of new radiopharmaceuticals for imaging hypoxia.

A summary of the resource.
Several new 1,2,5-oxadiazole N-oxide derivatives were synthesized to be tested both as potential selective hypoxic cell cytotoxins and as DNA-binding agents. The compounds prepared included bis(1,2,5-oxadiazole N-oxide) derivatives and oxadiazole rings linked to naphthyl residues. The compounds were tested for their cytotoxicity in oxia and hypoxia and they proved to be non-selective and less active than the parent compounds 3-formyl-4-phenyl-1,2,5-oxadiazole N2-oxide (3) and 3-chloromethyl-4-phenyl-1,2,5-oxadiazole N2-oxide (4). The DNA-affinity assays showed that the compounds tested have poor affinity for this biomolecule.<br /><br />

A summary of the resource.
A series of new 3-alkoxy- or 3-hydroxy-1-[x-(dialkylamino)alkyl]-5-nitroindazoles have been synthesized and their trichomonacidal, antichagasic and antineoplastic properties studied. Five derivatives (5, 6, 8, 9 and 17) showed remarkable trichomonacidal activity against Trichomonas vaginalis at 10 lg/mL concentration. Three compounds (8, 10, 11) exhibited interesting antichagasic activity and these same compounds moderate antineoplastic activity against TK-10 and HT-29 cell lines. Unspecific cytotoxicity against macrophages has also been evaluated and only compounds 9, 10 and 11 resulted cytotoxic at the higher dose evaluated (100 lg/mL), loosing cytotoxicity at lower doses. QSAR studies have been carried out. X-ray crystallographic study of compound 8 has been performed.

<p>Tissue engineering (TE) and regenerative medicine offer strategies to improve damaged tissues by using scaffolds and cells. The use of collagen-based biomaterials in the field of TE has been intensively growing over the past decades. Mesenchymal stromal cells (MSCs) and dental pulp stem cells (DPSCs) are promising cell candidates for development of clinical composites. In this study, we proposed the development<br />of a bovine collagen type I: chondroitin-6-sulphate (CG) scaffold, obtained from Uruguayan raw material (certified as free bovine spongiform encephalopathy), with CG crosslinking enhancement using different gamma radiation doses. Structural, biomechanical and chemical characteristics of the<br />scaffolds were assessed by Scanning Electron Microscopy,<br />axial tensile tests, FT-IR and Raman Spectroscopy,<br />respectively. Once we selected the most appropriate scaffold for future use as a TE product, we studied the behavior of MSCs and DPSCs cultured on the scaffold by cytotoxicity, proliferation and differentiation assays. Among the diverse porous scaffolds<br />obtained, the one with the most adequate properties was the one exposed to 15 kGy of gamma radiation.</p>

In the search for new therapeutic tools against parasitic diseases caused by the Kinetoplastids Leishmania spp. and Trypanosoma cruzi, a novel gold(I) triphenylphosphine complex with the bioactive coligand pyridine- 2-thiol N-oxide (mpo) was synthesized and characterized by using analytical and conductometric measurements, electrospray ionization-mass spectrometry (ESI) and electronic, FTIR and 1H and 31P NMR spectroscopies. A dinuclear structure is suggested for the complex. At a 1 microM concentration the complex induced in vitro after 30 min a potent leishmanicidal effect (LD50) against promastigotes of Leishmania (L.) mexicana while on Leishmania (V.) braziliensis with the same concentration only a leishmanistatic effect (IC75) was observed 48 h after treatment. Similar differential susceptibilities were also found when testing the ligand mpo, but at a higher dose (5 lM). In addition, the compound showed growth inhibitory effect on Dm28cT. cruzi epimastigotes in culture (IC50 0.09 lM), being even more active than the anti-trypanosomal reference drug Nifurtimox (IC50 6 lM). DNA interaction studies showed that this biomolecule does not constitute a main target for the mpo complex currently tested. Instead, the significant potentiation of the antiproliferative effect against both Leishmania species and T. cruzi could be associated to the inhibition of NADH fumarate reductase, a kinetoplastid parasite-specific enzyme absent in the host. Furthermore, due to its low unspecific cytotoxicity on mammalian cells (J774 macrophages), the new gold complex showed a selective anti-parasite activity. It constitutes a promising new potent chemotherapeutic alternative to be evaluated in vivo in experimental models of leishmaniasis and Chagas´ disease.

A summary of the resource.
Oxovanadium(IV) and oxovanadium(V) complexes of tropolone (Htrp, 2-hydroxy-2,4,6- cycloheptatrien-1-one), of stoichiometry VO(trp)2 and VO(trp)2Cl, were characterized by electronic, infrared, and Raman spectroscopies. Their electrochemical behavior was investigated by cyclic voltamperommetry in dimethylsulfoxide solutions. The magnetic susceptibility of VO(trp)2 was investigated between 2 and 300 K. The vibrational spectra of the related Co(trp)2, Cu(trp)2, and Zn(trp)2 complexes were also analyzed for comparative purposes.

The reaction of Cu(CH3COO)2·H2O and Cu(ClO4)2·6H2O with bis(3,5-dimethylpyrazol-1-yl)methane ligand (L) yielded complexes [Cu(CH3COO)2(L)] (1) and [Cu(L)2(H2O)](ClO4)2 (2), which were isolated as air-stable solids. The complexes were characterized by electrical conductivity; thermogravimetric analysis; cyclic voltammetry; and Raman, infrared and ultraviolet/visible spectroscopy. The structure of 2 was determined by single-crystal X-ray diffraction analysis. Graphite electrodes modified with each of the complexes were used to detect dopamine using differential pulse voltammetry. In the presence of ascorbic acid and uric acid, the highest sensitivity (2.01 μA μM− 1 cm− 2) and the lowest detection limit (0.5 μM) were obtained with the electrode modified with 1. The linearity of the signal, the sensitivity and the detection limit were significantly improved when the solution contained polystyrenesulfonate (PSS−), demonstrating the synergistic effect between the complexes modifying the electrode surface and the PSS− in solution.

Abstract
A summary of the resource.
Five new copper and zinc heteroleptic complexes with saccharin and aminoacids with general stoichiometry Na2[M(sac)2(aa)2]⋅nH2O (M denotes Cu or Zn, sac the saccharinate ion, and aa the aminoacids) were synthesized and characterized by elemental and thermogravimetric analysis, conductimetric measurements and IR, Raman and UV–vis spectroscopies. In all the complexes, copper and zinc ions coordinated with the aminoacids through the terminal amine and carboxylate residues and with saccharin through the heterocyclic nitrogen atom. Besides, the superoxide dismutase-like activity of the heteroleptic copper complexes was evaluated and compared with the homoleptic copper amino acid complexes with the aim to observe the influence of the saccharin coordination.
