Immunosensors for small analytes have been a great addition to the analytical toolbox due to their high sensitivity and extended analytical range. In these systems the analyte is detected when it competes for binding to the detecting antibody with a tracer compound. In this work we introduce the use of phage particles bearing peptides that mimic the target analyte as surrogates for conventional tracers. As a proof of concept, we developed a magneto-electrochemical immunosensor (EI) for the herbicide molinate and compare its performance with conventional formats. Using the same anti-molinate antibody and phage particles bearing a molinate peptidomimetic, the EI performed with an IC50 of 0.15 ng mL−1 (linear range from 4.4 × 10−3 to 10 ng mL−1). Compared to the conventional ELISA, the EI was faster (minutes), performed with a much wider linear range, and the detection limit that was 2500-fold lower. The EI produced consistent measurements and could be successfully used to assay river water samples with excellent recoveries. By using the same EI with a conventional tracer, we found that an important contribution to the gain in sensitivity is due to the filamentous structure of the phage (9 × 1000 nm) which works as a multienzymatic tracer, amplifying the competitive reaction. Since phage-borne peptidomimetics can be selected from phage display libraries in a straightforward systematic manner and their production is simple and inexpensive, they can contribute to facilitate the development of ultrasensitive biosensors. The aim of the present work was to apply survival analysis to estimate equivalent sweet concentration of low-calorie sweeteners in orange juice. Five low-calorie sweeteners were considered: sucralose and four samples of stevia. For each sweetener, a series of six orange juice samples with different sweetener concentration were prepared. Groups of 34 consumers evaluated the series of six pairs of samples, each composed of one reference sample (sweetened with 10% sucrose) and one sample with low-calorie sweetener. Consumers were asked to complete a paired-comparison test and to indicate which of the samples was sweeter. The elicited data were analyzed using survival analysis. For each sweetener, the percentage of assessors perceiving the sample with the low-calorie sweetener as sweeter than the sample with sucrose as a function of sweetener concentration was determined. Equivalent sweet concentrations were determined for each sweetener and compared with estimations obtained using logistic regression and published data. We have investigated whether the chemical components of fetal fluids (FFs), which elicit repulsion in late gestating ewes, are also those responsible for the attractiveness of fetal fluids at parturition. An aqueous fraction of FFs (A1), obtained after extraction with hexane, was tested for repulsion in late-pregnant ewes and for attraction at parturition. We also investigated if the repulsive and attractive characteristics of this A1 fraction were maintained after an additional extraction with dichloromethane (DCM, CH2Cl2) that produced two more fractions (aqueous/high polarity: A2 and dichloromethane/medium polarity: DCM). Thus, late-pregnant ewes were tested for repulsion of aqueous extracts of FFs (A1, A2 and DCM fractions) in a two-choice test of food preference, whereas parturient ewes were tested for attraction toward these same fractions in a two-choice test of licking warm spongy cloths. The A1 fraction was repulsive to late-pregnant ewes and attractive to parturient females. In contrast, neither the A2 nor the DCM fractions were repulsive to late-pregnant ewes, whereas both fractions were attractive to parturient ones. The discordance between the repulsive and attractive properties of the A2 and DCM fractions suggests that the attractiveness of FFs for parturient ewes and its repulsiveness for females outside the peripartum period depend on mixtures of substances that are at least partially different. Background: The body mass index (BMI) ≥30 kg/m2 is the universally accepted cut-off point for defining obesity; however, its accuracy in classifying obesity in older adults is poorly understood. Objectives: To assess the performance of the BMI cut-off point ≥30 kg/m2 in classifying obesity in older adults, using the fat mass index (FMI) and fat mass percentage (FM%) as reference criteria; and to establish region- and sex-specific BMI-based cut-off points to classify obesity in older adults. Methods: The present study is a secondary analysis derived from a cross-sectional project that included a sample of 1463 older adults from ten Latin American and Caribbean countries. Volunteers underwent total body water measurements using the deuterium dilution technique to determine FMI and FM%. Accuracy of the BMI and derived cutoff points was assessed by the area under the receiver operating characteristic curve (AUC).Results: The BMI cut-off point ≥30 kg/m2 had low sensitivity for classifying obesity in these older adults compared to the FMI and FM%. The AUC values for the optimal BMI-derived cut-off points showed an acceptable-to-outstanding discriminatory capacity in diagnosing obesity defined by the FMI. There was also a better balance between sensitivity and specificity than with the values obtained by a BMI ≥30 kg/m2 in older subjects in both regions. Conclusion: The BMI cut-off point ≥30 kg/m2 had poor sensitivity for accurately diagnosing obesity in older adults from two regions. The region- and sex-specific BMI-derived cut-off points for defining obesity using the FMI are more accurate in classifying obesity in older men and women subjects from both regions. Biogas upgrading through dry reforming is a promising technology for converting biomass into high-value products such as synthesis gas. In this work, Ni-La-Ti catalysts were synthesized using different preparation methods (impregnation, coprecipitation and modified Pechini method) and evaluated in clean biogas dry reforming reaction at 800 ◦C using two different CH4/CO2 molar ratios (1.5 and 3) to study the effects of the different biogas qualities on catalytic activity and stability. Fresh and used catalysts were examined by several techniques including the BET method, XRD, ICP-AES, H2-TPR, CO2-TPD, EPR, Raman, TGA, SEM and TEM. The different catalytic activities obtained could be attributed to the different degree of Ni-La-Ti interaction. The catalyst prepared by impregnation shows the highest initial catalytic activity for the two CH4/CO2 ratios tested. However, it was observed that the formation of solid carbon took place to a greater extent on this catalyst. The catalyst prepared by the modified Pechini method showed good stability through a 100 h stability test using CH4/CO2 = 1.5 feed ratio. Characterization of the used catalyst confirmed the formation of mainly filamentous type carbon and high resistance to Ni sintering. The bronze bug, Thaumastocoris peregrinus Carpintero & Dellape (Heteroptera: Thaumastocoridae), is an exotic emerging pest in Eucalyptus commercial forests in South America, Africa and southern Europe. Information on the chemical communication system and reproductive ecology of this insect is scant, and it may be relevant for designing management strategies for eucalypt plantations. Adults and nymphs usually aggregate in the field, possibly by means of chemical signals. Males emit large amounts of 3-methyl-2-butenyl butyrate, which attracts conspecific adult males but not females. The ecological role of this putative male aggregation pheromone remains unknown. Here, we report olfactometer bioassays showing that late-instarmale nymphs are also attracted to synthetic 3-methyl-2-butenyl butyrate and to adult male volatile extracts, which contain this compound as the major component. As previously shown for adult females, nymphs that moulted into females were not attracted to either volatile stimulus. The intra-gender attraction of nymphs and adults may be related to the exploitation of food resources, or as a reproductive strategy for newly emerged males. Further studies on the reproductive behaviour and mating system of T. peregrinus will contribute to understanding the ecological significance of male-male, adultnymph attraction, as well as the practical applications that may result from these findings. This study reports the effect of different doses of nitrogen applied to soil and/or leaves of Syrah and Chardonnay grapevines in the Languedoc-Roussillon (France) over two years. In 2011, nitrogen treatment involved both foliar urea sprayings and soil application at two different levels, with two controls - irrigated without nitrogen and no irrigation nor nitrogen. In 2012, the same grapevines received either soil or foliar nitrogen using the same controls. Results showed that foliar application increased the amino acid content to a greater extent than soil application, but that a combination of both was the most effective. For the first time, significantly elevated proline levels in response to drought were demonstrated for the grapevine. Increased contents of aromatic compounds and glycosylated precursors closely mirrored the applied nitrogen dose. Wines produced from N-fertilized Syrah grapes in 2011 showed a statistically significant effect of irrigation and fertilization on positive sensorial perception. Por otro lado no es ajena la mencionada línea al Laboratorio de Inmunotecnología dirigido por el otro copatrocinante, MsC. Battistoni. Mucho se ha investigado con el objetivo de obtener una cura para el Mal de Chagas. Esta Tesis es un aporte más, centrándose en el estudio de la molécula de ciclosporina (CsA), y compuestos estructuralmente relacionados. Estos compuestos han mostrado actividad inhibitoria sobre Trypanosoma cruzi. Si bien la CsA es una droga con actividad inmunosupresora, algunos de los compuestos estructuralmente relacionados activos contra T. cruzi no presentan efectos inmunosupresores. Se ha propuesto a CsA como compuesto líder para el desarrollo de nuevas drogas útiles en la farmacoterapias de la enfermedad de Chagas. Se conocen con cierto grado de detalle las bases bioquímicas de las actividades inmunosupresoras e inhibitorias sobre protozoarios de CsA. En ambas están involucradas una familia de proteínas, las ciclofilinas. Se han publicado datos de las actividades inhibitorias de CsA y compuestos relacionados sobre ciclofilinas presentes en T. cruzi, en particular sobre la ciclofilina mas expresada en T. cruzi, la TcCyP19. Las bases moleculares de la diferencia de actividad inmunosupresora y de inhibición sobre T. cruzi de CsA y de una serie análogos no han sido dilucidadas. Esta tesis pretende, mediante el uso de diversas herramientas de modelado molecular, docking y de mecánica molecular, estudiar el comportamiento de los complejos binarios de TcCyP19 con una serie de compuestos análogos a CsA para obtener información a nivel molecular que aporte a la comprensión de las diferencias encontradas. En forma complementaria, se estudian complejos de CsA y dos de sus análogos con una ciclofilina humana, así como el comportamiento de una proteína mutante de la ciclofilina humana con baja afinidad por CsA. Una vez elaborados los modelos de los sistemas a estudiar se analizaron los posibles sitios de unión de estos compuestos con las ciclofilina en estudio, así como los modos de unión a los mismos. Utilizando simulaciones de dinámica molecular, se obtuvo un muestreo del espacio de fases de los sistemas en estudio que permitió analizar estructuralmente y energéticamente dichos sistemas. Varios términos energéticos se calcularon, que incluyeron energías de interacción ligando-proteína y cálculos de energías libres de asociación por medio del método LIE (Linear Interacction Energy). Se discuten algunos aspectos metodológicos, como la necesidad de hacer varias simulaciones de dinámica molecular para obtener un muestreo mas apropiado del comportamiento del sistema y el cumplimiento de las premisas para usar el método LIE. Los resultados obtenidos muestran que se trata de sistemas con comportamiento muy similar, para los cuales los métodos de cálculo usados no son lo suficientemente sensibles para diferenciar energías libres de unión. Se demostró la posible existencia de lugares de unión de Modelado y estudio de complejos de Ciclosporina A y compuestos relacionados ….. 5 los compuestos estudiados diferentes al sitio catalítico, cuya relevancia deberá ser estudiada in vitro. Todo lo aprendido con estos sistemas permitirá abordar con mejores perspectivas ulteriores etapas del estudio de esta familia de compuestos que involucran complejos ternarios y sus potenciales actividades inmunosupresoras en el huésped o inhibitorias del crecimiento de T. cruzi. The antiproliferative effects of four series of VIVO- and VVO-based compounds containing 8-hydroxyquinoline ligands on the bacterium Mycolicibacterium smegmatis (M. smeg) were investigated. The effects on M. smeg were compared to the antiproliferative effects on the protozoan parasite Trypanosoma cruzi (T. cruzi), the causative agent for Chagas disease. In this study, we investigate the speciation of these compounds under physiological conditions as well as the antiproliferative effects on the bacterium M. smeg. We find that the complexes are more stable the less H2O is present, and that the stability increases in lipid-like environments. Only one heteroleptic complex and two homoleptic complexes were found to show similar antiproliferative effects on M. smeg as reported for T. cruzi so the responses generally observed by M.smeg. is less than observed by the pathogen. In summary, we find that M. smeg is more sensitive to the detailed structure of the V-complex but overall these complexes are less effective against M. smeg compared to T. cruzi. Entomopathogenic fungi such as Beauveria bassiana are extensively used for the control of insect pests worldwide. They infect mostly by adhesion to the insect surface and penetration through the cuticle. However, some insects, such as the red flour beetle Tribolium castaneum (Herbst), have evolved resistance by embedding their cuticle with antifungal compounds. Thus, they avoid fungal germination on the cuticle, which result in low susceptibility to entomopathogenic fungi. In adult T. castaneum, these antifungals are the well-known defensive compounds methyl-1,4- and ethyl-1,4- benzoquinone. In this study, we added B. bassiana conidia on the diet of adult beetles to study the effect of the entomopathogen on the secretion and detection of the beetle volatile blend containing both benzoquinones. The compounds were analyzed by solid phase microextraction coupled to gas chromatography–flame ionization detection, and were detected by electroantennography. In addition, we measured the expression level of four genes encoding for two odorant-binding proteins (OBP), one chemosensory protein (CSP), and one odorant receptor (OR) in both healthy and fungustreated insects. Significant alterations in the secretion of both benzoquinones, as well as in the perception of methyl-1,4-benzoquinone, were found in fungus-treated insects. TcOBP7D, TcOBP0A and TcCSP3A genes were down-regulated in insects fed conidia for 12 and 48 h, and the latter gene was up-regulated in 72 h samples. TcOR1 expression was not altered at the feeding times studied. We conclude that fungus-treated insects alter both secretion and perception of benzoquinones, but additional functional and genetic studies are needed to fully understand the effects of fungal infection on the insect chemical ecology.
