Trans-[Co(NH3)4(NH2CH3)(CH3)2 SO]3+ and trans-[Co(NH3)4(NH2CH3)(CH3) NCHO]3+ ions are prepared by Ag+-induced solvolysis of trans-[Co(NH3)4(NH2 CH3)X]2+ (X = Cl or Br) complexes in dimethylsulphoxide or dimethylformamide. The complexes were characterized by electronic, IR and 1H magnetic resonance spectroscopy. Both, dimethylsulphoxide and dimethylformamide are bonded through the oxygen atom. The trans-complexes obtained are not contaminated with the cis-isomers.

A polymeric heterometallic compound [{Mn(H2O)6} {MnCe(oda)3}2]·xH2O containing manganese(II) and cerium (III) cations bridged by carboxylate groups of oxydiacetate (oda) is presented. It was prepared by direct reaction of oda in aqueous solution with stoichiometric amounts of Mn(II) and Ce (III). The structure of the complex exhibits a highly ordered 3D structure with [MnCe(oda)3]- as building blocks. The charge is balanced with [Mn(H2O)6]2+ ions which occupy the holes of the anionic framework. The complex is thermally stable up to 230° C.

We report the synthesis and evaluation as potential anticancer agents of a series of tetracyclic indenoquinolines. The compounds, which are obtained through the photoisomerization of Diels–Alder adducts formed between purpurogallin derivatives and nitrosobenzene, have in vitro antiproliferative activities in the lM to nM range against breast (MCF-7), lung epithelial (A-549), and cervical (HeLa) adenocarcinoma cells. The cytotoxicities of several of the novel tetracycles are comparable to or better than that of camptothecin. A strong correlation between the activity of the compounds and their aromaticity and planarity was observed, suggesting a mode of action similar to that of topoisomerase poisons

Vascular endothelial growth factor (VEGF) is one of the classic factors involved in tumor-induced angiognesis in several solid tumors. Bevacizumab, a monoclonal antibody against VEGF, can be used as an imaging tool in preclinical studies. The aim of this study was to radiolabel Bevacizumab with (99m)Tc and to evaluate in vivo its imaging properties in an adenocarcinoma animal model. For this purpose, Bevacizumab was derivatized with Suc-HYNIC as a bifunctional coupling agent. A mixture of Tricine/SnCl(2).2H(2)O was added to Bevacizumab-HYNIC and radiolabeled with (99m)TcO(4)(-). The radiochemical stability of the radiolabeled antibody was assessed. Biodistribution and scintigraphy imaging were performed in normal CD1 female mice and in spontaneous adenocarcinoma tumor bearing CD1 mice (n = 5). We demonstrated that 99mTc-HYNIC-Bevacizumab was stable. In vivo biodistribution studies revealed that tumor uptake of (99m)Tc-HYNIC-Bevacizumab was 1.37 ± 0.51% and 5.33 ± 2.13% at 4 and 24 h postinjection, respectively. Scintigraphy image studies showed tumor selective uptake of (99m)Tc-HYNIC-Bevacizumab in the tumor-bearing mice. We conclude that (99m)Tc-HYNIC-Bevacizumb has the potential to be used as a tracer for tumor imaging in preclinical studies.

The objective of this work was to develop a novel 99mTc complex bearing the 5-nitroimidazol-1-yl moiety with recognised selectivity towards hypoxic tissue, as a potential radiopharmaceutical for imaging tumour hypoxia. The new metronidazole derivative (2-amine-3-[2-(2-methyl-5-nitro-1H-imidazol-1-yl)ethylthio]propanoic acid) (L) containing adequate groups to coordinate technetium through the formation of a Tc(I)-tricarbonyl complex was synthesised with adequate yield (33%) and characterised by spectroscopy. Labelling was performed by substitution of three labile water molecules of the technetium tricarbonyl precursor, fac-[99mTc(CO)3(H2O)3]+ with the ligand. A radiochemical purity higher than 90% was achieved and remained unchanged for more than 4 h. The complex has a high stability in plasma, a moderate plasma protein binding and a moderate hydrophilicity. In vitro cell uptake studies showed a ratio between the activity taken up by cells in hypoxia/normoxia of 1.6 ± 0.4 (p < 0.5). Biodistribution in normal mice showed rapid depuration and low uptake in all organs and tissues except liver. Biodistribution in mice bearing induced tumours showed a low tumour uptake, but tumour/muscle ratio was favourable thanks to depuration. Comparison with biological results of other metronidazole derivatives clearly shows that modifications of the chelator are very important and contribute to improve the biological behaviour.

Using different heterocycle formation methodologies (Deoxo-Fluor, DAST, POCl3, TosMIC), [2,5’]bis-1,3- azoles were prepared as stable analogs of bengazoles, a family of potent anthelmintic marine natural products. The cytotoxic activity of these heterocycles and their precursors on HCT-15 cells and the effect on the L4 larvae of Nippostrongylus brasiliensis were evaluated.

Abstract

A new series of heteroaryl nitrones were synthesized and evaluated as free radical traps due to the results showed in our previous report. The physicochemical characterization of these new nitrones by electron spin resonance (ESR) demonstrated their high capability to trap and stabilize different atom centered free radicals generated by the Fenton reaction. Additionally, we intensely studied them in terms of their physicochemical properties. Kinetic studies, including the use of a method based on competition and the hydroxyl adduct decay, gave the corresponding rate constants and half-lives at the physiological pH of these newly synthesized nitrones. New nitrones derived from quinoxaline 1,4-dioxide heterocycles were more suitable than DMPO to trap hydroxyl free radicals with a half-life longer than two hours. We explain some of the results using computational chemistry through density functional theory (DFT).

A summary of the resource.
Crocidosema (= Epinotia) aporema (Walsingham) (Lepidoptera: Tortricidae) is a bud borer that feeds on soybean and forage legumes. Its economic importance is restricted to South America, where it can alternate throughout the year between forage and grain legumes. The sex pheromone of C. aporema females is composed of a 15:1 mixture of (7Z,9Z)-dodeca-7,9-dien-1-ol and (7Z,9Z)- dodeca-7,9-dienyl acetate. Aiming at the development of a monitoring tool, it was synthesized both components of the pheromone and evaluated male captures in pheromone traps baited with different blends of synthetic pheromone, in an experimental soybean field in Uruguay. The conjugated dienes were obtained from 2-pentyn-1-ol and 1,7-heptanediol, by oxidation of the former, Wittig coupling and Zn-catalyzed reduction of the triple bond. The 1:1 mixture was the most efficient in capturing males. The pheromone traps were attractive for up to 40 days, even with small septum loads (0.1 mg) and low population levels.

As part of an ongoing program on the chemistry and biological activity of N-oxide-containing molecules, a number of novel 1,2,5-oxadiazole N-oxide, benzo[1,2-c]1,2,5-oxadiazole N-oxide, and quinoxaline N,N¢-dioxide derivatives were synthesized and evaluated for their herbicidal activity. Many of these compounds exhibited moderate to good herbicidal pre-emergence activity against Triticum aestivum. Dose-response studies were done on the more representative compounds (12, 20, and 26). The most active compound, butylcarbamoylbenzo[1,2-c]1,2,5-oxadiazole N-oxide, 26, displayed herbicidal activity at concentrations as low as 24 g/ha.

The synthesis and in vitro activity of R(þ)-Limonene derivatives against Leishmania and Trypanosoma cruzi strains are reported. Seven compounds have shown better in vitro activity against Leishmania (V.)braziliensis than the standard drug pentamidine. Additionally, we have identified two promising new anti-T. cruzi limonene derivatives.

A small library of compounds was prepared by a combination of toluene dioxygenase (TDO)-catalyzed enzymatic dihydroxylation and copper(I)-catalyzed Hüisgen cycloaddition. Some compounds were obtained by coupling an alkyne and a conduritol derivative, while more complex structures were obtained by a double Hüisgen reaction of a dialkyne and two molecules of the cyclitol. The compounds were fully characterized and subjected to preliminary biological screening

La4BaCu5O13+δ (LBCu) and La6.4Sr1.6Cu8O20±δ (LSCu) oxygen deficient perovskites, synthesized by a Pechini-type gel combustion method, have been investigated as potential cathode materials for IT-SOFC applications. The thermal stability and chemical compatibility with SOFC/PCFC electrolyte materials are examined using X-ray diffraction technique. For both compounds, no structural change is detected in air until 900 °C. The evolution of cell parameters with temperature shows thermal expansion coefficients (TEC) of 17.3 × 10− 6 K− 1 and 12.1 × 10− 6 K− 1 for LBCu and LSCu, respectively. For LBCu, the formation of oxygen vacancies is evidenced between 700 and 900 °C in air. Chemical reactivity tests with several SOFC electrolyte materials reveal a high reactivity of both cuprates with YSZ, GDC and, to a lesser extent, with LSGM. On the other hand, and contrary to what occurs for LSCu, LBCu presents an excellent chemical compatibility with doped barium cerate and zirconate, opening further perspectives for this material as possible SOFC or PCFC cathode. LBCu material presents a quite high electrical conductivity with metallic-type behavior

A series of Tc(V) cationic complexes was synthesized by electrochemical reduction of TcO4. The electrolysis was performed in aqueous media containing amines as ligands: en |b dien |b trien and 1,2-dap. The combination of different techniques alloes to propose the general formula [TcO2/amine/2]+ for these compounds. Electrodeposition of TcO2 was a competitive reaction. The UV spectra were compared with those for other Tc(V) amine complexes. The presence of two peaks |b one in the range 303-312nm and the other in 230-240nm |b could be verified. The latter was markedly sensitive to pH changes. Preliminary studies showed that kinetic stability decreases in the sequence en |b 1,3-dap |b trien |b dien. The decomposition rate increased when the pH was lowered.

The synthesis of fused heterocycles such as thiazolidinyl-oxazolidine 3 is described starting from Tris 3 HCl. The mercaptomethyl bisoxazolidine 8 was found to convert to the corresponding thiazolidinyloxazolidine 3 and the spiroheterocycle 4 by a ring-chain-ring tautomerism, depending on the electronic nature of the ring substituents as well as the reaction conditions. This equilibration pathway is absent in the hydroxymethyl bisoxazolidines 2. Computational studies confirm that both kinetic and thermodynamic control features play a role in the product distribution.

A new imidazolium hydrated salt (DCF-IMI-H2O) of the nonsteroidal anti-inflammatory drug Diclofenac (DCF) was synthesized by solvent evaporation. This salt was fully characterized by X-ray diffraction (SCXRD, PXRD), infrared<br />spectroscopic (FT-IR) and thermal analysis (TGA, DSC, HSM) techniques. The salt formation is driven by deprotonation of the drug's carboxylic group, with proton transfer to the nitrogen atom of the imidazole ring. The water molecule plays an<br />important role in the 3D arrange observed in the crystal lattice, acting as a bridge between adjacent DCF and coformer molecules. The DCF-IMI-H2O exhibits an improved solubility profile when compared with the pure form of DCF, and similar<br />values when compared with sodium and potassium salts. This new salt proved stable under 75% relative humidity for one week and is stable after six-months storage at room conditions. The study of its thermal stability showed that a dehydration<br />process starts at around 45 ºC, leading to the probably formation of an unstable anhydrous form, which melting around 92°C. Spectroscopic studies showed that most of the salt is ionized when in amorphous state (liquid). These behaviors<br />features the new compound as an ionic liquid (IL), i.e., organic salt with melting point below 100 ºC. Pharmaceutical ILs have been increasingly exploited in the past few years aiming improved properties regarding drug solubility, formulation and delivery (topical, transdermal and oral).

<p>A series of mixed ligand oxovanadium(V) complexes of the type [VO(NH2O)2(aa)], containing the hydroxylamido ligand and simple amino acids, were prepared and characterized. Seven of these complexes, with aa = valine, leucine, isoleucine, phenylalanine, tryptophan, cysteine and methionine, were described for the first time. Their infrared and Raman spectra, together with those of the previously investigated similar species with aa = glycine, serine, threonine and alanine, and those of the related [VO(NH2O)2(imidazole)2]Cl complex, were recorded and assigned. The spectroscopic behavior of the new complexes is similar to that of the other five, whose structures has been determined by X-ray crystallography, suggesting an identical structure for the full series of complexes. The spectroscopic results also give a better characterization of the vibrational behavior of the interesting O–N–“side-on” bonded hydroxylamido ligand.</p>

Four new lead(II) thiosaccharinate complexes: [Pb(tsac)2H2O] (1) (tsac: thiosaccharinate anion), [Pb2(tsac)4(py)4] (2) (py: pyridine), [Pb(tsac)(o-phen)2](tsac)·CH3CN (3) (o-phen: 1,10-phenantroline), and [Pb(tsac)2(bipy)] (4) (bipy: 2,2�-bipyridine) were prepared. The infrared and electronic spectra as well as the thermal analysis of all the compounds were recorded and discussed. The thiosaccharinate anion acts in three different coordination forms, one of then reported for the first time. The crystal structures of complexes 2 and 3 have been determined by single crystal X-ray diffractometry. In complex 2, two monomeric moieties are joined together forming a symmetric bis-μ-sulphur bridged dimer by interaction of two lead(II) atoms through the exocyclic sulphur atoms of two thiosaccharinate ligands. The seven-fold coordination sphere of each lead atom is completed by two pyridine nitrogen atoms and by another sulfur and two nitrogen atoms of the thiosaccharinate anions. In complex 3, the lead(II) atom is coordinated by four nitrogen atoms of two 1,10-phenantroline molecules and by the sulfur and nitrogen atoms of one thiosaccharinate ion. The second anion has an electrostatic interaction with the nucleus.

This work presents the synthesis and characterization of eight copper complexes [Cu(L-dipeptide)(neo)]·nH2O (neo = neocuproine) and their cytotoxic activities against tumor cell lines. The crystalline structure of [Cu(gly-val)(neo)]·3H2O, [Cu(gly-leu)(neo)]·H2O, [Cu(ala-gly)(neo)]·4H2O, [Cu(val-phe)(neo)]·4.5H2O and [Cu(phe-phe)(neo)]·3H2O were determined by single crystal X-ray diffraction. In all of them, the Cu(II) is pentacoordinated, in a square pyramidal environment. The coordination observed in solid state was retained in the major species in aqueous solution, as suggested by Electronic Paramagnet Resonance and UV–vis spectroscopies. The complexes were shown to have affinity for isolated DNA, as determined by Circular Dichroism experiments. Furthermore, biological experiments showed that all the complexes present high cytotoxic activity against the cell lines: MDA-MB-231, MCF-7 (human metastatic breast adenocarcinomas, the first triple negative), MCF-10A (human normal breast cells), A549 (human lung epithelial carcinoma) and MRC-5 (human lung epithelial cells). Together, these results suggest that these compounds are promising steps towards new effective drugs to treat cancer.

Two new N′-heteroarylidene-1-carbohydrazide derivatives, namely; E-N′-[(pyridine-3-yl)methylidene]adamantane-1-carbohydrazide (1) and E-N′-[(5-nitrothiophen-2-yl)methylidene]adamantane-1-carbohydrazide (2), were produced via condensation of adamantane-1-carbohydrazide with the appropriate heterocyclic aldehyde. Both compounds were chemically and structurally characterized by 1H-NMR, 13C-NMR, infrared and UV-vis spectroscopies, and single crystal X-ray diffraction. The study was complemented with density functional theory calculations (DFT). The results show an asymmetrical charge distribution in both compounds, with the electron density accumulated around the nitrogen and oxygen atoms, leaving the positive charge surrounding the N-H and C-H bonds in the hydrazine group. Consequently, the molecules stack in an antiparallel fashion in the crystalline state, although the contribution of the polar contacts to the stability of the lattice is different for 1 (18%) and 2 (42%). This difference affects the density and symmetry of their crystal structures. Both molecules show intense UV-Vis light absorption in the range 200–350 nm (1) and 200–500 nm (2), brought about by π → π* electronic transitions. The electron density difference maps (EDDM) revealed that during light absorption, the electron density flows within the π-delocalized system, among the pyridyl/thiophene ring, the nitro group, and the N′-methyleneacetohydrazide moiety. Interestingly, compounds 1 and 2 constitute broad-spectrum antibacterial candidates, displaying potent antibacterial activity with minimal inhibitory concentration (MIC) values around 0.5–2.0 μg/mL. They also show weak or moderate antifungal activity against the yeast-like pathogenic fungus Candida albicans

Nitrated fatty acids (nitroalkenes) have been recently detected and quantified in cell membranes and human plasma. However, nitration of arachidonate (AA), that could redirect AA-dependent cell signaling pathways, has not been studied in detail. Herein, we synthesized and determined for the first time the isomer distribution of nitroarachidonate (AANO2) and demonstrate its ability to modulate inflammation. Synthesis of AANO2 was achieved by AA treatment with sodium nitrite in acidic conditions following HPLC separation. Mass spectrometry (MS) analysis showed the characteristic MS/MS transition of AANO2 (m/z 348/301). Moreover, the IR signal at 1378.3 cm-1 and NMR studies confirmed the presence of mononitrated nitroalkenes. Positional isomer distribution was determined by NMR and MS fragmentation with lithium; four major isomers (9-, 12-, 14-, and 15-AANO2) were identified, which exhibited key anti-inflammatory properties. These include their ability to release biologically relevant amounts of nitric oxide, induce cGMP-dependent vasorelaxation, and down-regulate inducible nitric oxide synthase (NOS2) expression during macrophage activation, providing unique structural evidence and novel regulatory signaling properties of AANO2.

A microwave-assisted and base-catalyzed domino manifold to construct 2,4-cyclohexadienone derivatives has been implemented. The domino manifold uses easily accessible tertiary propargyl vinyl ethers bearing a methine group at the homopropargylic position and imidazole as the catalyst to deliver 2,4-cyclohexadienones featuring a key formyl group and a quaternized carbon atom in good yields.

Abstract
A summary of the resource.
Selective acetylation of 1 at C-28 afforded the monoacetate 2 |b which was oxidized to the 3-oxo compound 8. H,D exchange at C-2 in 8 followed by reduction with LiAIH4 led to [2-2H]-betulin (5). Reduction of 8 with NaBD4 or LiAID4 yielded [3-2H]betulin (6) besides different amounts of the respective 3alfa-hydroxy compound 7. For the introduction ofa CD2 or 13CH2 unit at C-29 betulin diacetate 4 was ozonolyzed to give the known compound 10 |b which was treated either with CD2Br2,Zn,and TiCl4 in THF to yield the [29-2H2]compound 11 or with [13C]methyltriphenylphosphonium iodide and potassium tert-butoxide to afford the [29-13C]-labeled compund 11. Solvolysis of 11 and 13 with methanol and potassium carbonate afforded the desired compounds [29-2H2]betulin (12)and [29-13C] betulin (14).
