A summary of the resource.
A study of 99mTc-adenosine-5'-diphosphate (99mTc-ADP) as a radiopharmaceutical for tumour diagnosis is presented. Two different labelling methods, using SnCl2 in alkaline solution and Zn as reducing agents, were developed. Reduction with Sn(II) alkaline solution was the selected method because a lower concentration of ADP (0.5 mg/mL) could be used and a higher radiochemical yield was achieved. A labelled molecule with a radiochemical purity higher than 95%, in vitro stability of at least 6 hours and an over all negative charge was obtained Biodistribution studies carried out in normal mice and rats revealed rapid urinary excretion and no specific accumulation of activity in any other particular organ. This behaviour was similar to that reported for 99mTc-adenosine-5'-triphosphate (99mTc-ATP). Rapid blood clearance, that could be fitted to a bicompartimental model, was also verified. No evidence of in vivo instability was observed. Studies in mice and rats bearing spontaneous mammary adenocarcinomas were performed and the results were compared to those from the 99mTc-ATP studies. Although the tumour models used were not the same, the incorporation of both labelled compounds was very similar. Radioactivity uptake in the tumour and the tumour-to-blood ratio were not notably high. However, a significant increment was observed in the tumour-to-muscle ratio (1.0 +/- 0.2 at 30 minutes to 2.7 +/- 0.4 at 240 minutes). Whole-body autoradiography enabled tumour visualization. Further investigations, including scintigraphic imaging, must be carried to complete the clinical evaluation of 99mTc-ADP as a tumour seeking agent.

En la actualidad, el consumo de vegetales en diversas formas y con mínimo procesamiento, es una práctica cada vez más extendida por la población, pues son reconocidos como fuente de principios activos benéficos para la salud. En general, son cultivados empleando prácticas agrícolas diversas, convencionales, integradas u orgánicas, con un claro predominio de la forma de cultivo convencional, donde el uso de agroquímicos es común. Considerando además la capacidad de estos vegetales de absorber metales y otros elementos del suelo donde se cultivan, algunos de ellos esenciales para la vida y otros potencialmente tóxicos, es que el interés en conocer los niveles de estos elementos se vuelve cada vez mayor. El desafío de este trabajo de posgrado fue obtener una visión global de los aportes tanto benéficos como negativos para la salud que el consumo de estos vegetales podía tener, mediante un estudio de la composición química de los nutracéuticos, así como de los contaminantes orgánicos e inorgánicos presentes en una planta con reconocidas propiedades benéficas como lo es la alcachofa. El fruto de la misma es un alimento muy valorado debido a sus propiedades nutricionales. Su elevada humedad, bajo contenido calórico, la práctica ausencia de grasas y su naturaleza fibrosa la han convertido en uno de los alimentos dietéticos por excelencia. A esto se suma los efectos antioxidantes, coleréticos y adelgazantes reportados en la literatura. Se estudiaron las propiedades de los frutos y de las hojas de alcachofa, así como las propiedades de las correspondientes infusiones de las hojas, debido a su amplio consumo bajo forma de té y a su empleo en productos fitoterápicos. Para lograr esta evaluación integrada, se estudió el contenido de elementos con valor nutricional como ser algunos metales esenciales (Cu, Fe, Ni y Zn) y algunos metabolitos secundarios como los polifenoles de reconocida capacidad antioxidante. También se estudió el contenido de aquellos elementos que podían representar un riesgo para la salud, como ser As, Cd y Pb, y los residuos de algunos agroquímicos comúnmente utilizados para proteger los cultivos y asegurar rendimiento y calidad en la producción. A lo largo de este trabajo se desarrollaron metodologías analíticas simples y originales que permitieron la evaluación de la presencia de contaminantes a nivel de trazas, orgánicos e inorgánicos, así como la determinación de compuestos orgánicos

Several Baccharis species are popularly known in traditional medicine as “carquejas”, “vassouras”, “ervas-santas” and “miomios”, and are used as anti-inflammatories, digestives, and diuretics. This study aimed to investigate the chemical compositions and cytotoxic activities of essential oils (EOs) of six Baccharis species belonging to subgenus Coridifoliae, namely B. albilanosa, B. coridifolia, B. erigeroides, B. napaea, B. ochracea, and B. pluricapitulata. GC/MS analyses of the EOs showed that the oxygenated sesquiterpenes spathulenol (7.32–38.22%) and caryophyllene oxide (10.83–16.75%) were the major components for all the species. The EOs of almost all species were cytotoxic against cancer (BT-549, KB, SK-MEL and SK-OV-3) and normal kidney (VERO and LLC-PK1) cell lines, whereas B. erigeroides EO showed cytotoxicity only against LLC-PK1. This article augments the current knowledge about the chemicalbiological properties of Baccharis subgenus Coridifoliae and discusses the therapeutic potentials of these economically unexploited plants.

En 1768 se autorizó la planteación de la primer botoica que tuvo esta ciudad, establecida por don José Gabriel Piedracueva. Hasta entonces habían carecido sus moradores de una farmacia donde poder obtener medicamentos para sus dolencias, estando recucidos al uso de yerbas silvestres para sus remedios, a excepción del que podía costearlos de Buenos Aires. Bien que en aquel tiempo había "peste de salud" en la población, computada en unos 1.200 habitantes, a pesar del desaseo, del lodoy de las aguas estancadas en charcos y zanjones, y por consecuencia eran pocas enfermedades que se conocían y ninguna epidémica. La botica de Piedracueva, la primera que tuvieron a su servicio nuestros antepasados , precedió con mucha antelación al establecimiento de la llamada del Rey, que fuera la segunda,. There are three basic steps to activated carbon fibers (ACF) manufacturing, from PAN fiber: oxidation/stabilization, carbonization and activation. Carbon material, specially ACF is a very attractive material to be used as supercapacitor electrode. The literature describes carbon material surface chemistry importance for supercapacitors application, mainly nitrogen groups by N-doping. Oxidation/stabilization is an important non-explored factor that influence the surface chemical functionality. This work describes the influence of oxidation/stabilization process on ACF production, from textile PAN fiber, and the non-doping nitrogen surface chemistry characteristic caused by the oxidation process design. Its textural, structural and surface was evaluated for supercapacitor electrode. The results show that the oxidation degree can be used as a mechanism of textural and surface chemistry control. The surface chemistry is the key of this work, different oxidation conditions can produce nitrogen compounds that help to increase specific capacitance. The tests showed an increase in capacitance higher than 100% in comparison to the standard oxidation.

The aim of this study was to develop an immunological model of avian Aspergillosis by studying the humoral response of pigeons toAspergillus fumigatus antigens. Immunization was performed by administering weekly injections ofA. fumigatus extracts for 70 days (10 weeks). A new booster injection was given 270 days (9 months) following the last immunization. Results showed an earlyAspergillus-specific humoral immunoresponse which reached a maximum level at 42–63 days (6–9 weeks) post-immunization. Using the ELISA method, it could be observed thatA. fumigatus-specific IgG became elevated in the 2nd week and reached a maximum titre at 63rd day (9th week). In contrast,A. fumigatus-specific IgM levels appeared early showing maximum levels at the 2nd week, after which they declined despite the maintenance of antigenic stimulation. Termination of immunization resulted in the decrease of specific humoral immunoresponse with minimal levels of specific antibodies detectable 210 days (7 months) later. A booster injection given at 270 days (9 months) induced a very fastAspergillus-specific IgM and IgG immunoresponse, reaching levels of antibodies similar to those observed during the immunization period.

Phenolic Profiling and Antioxidant Capacity of Eugenia uniflora L. (Pitanga) Samples Collected in Different Uruguayan Locations
Abstract
A summary of the resource.
The use of nutrient-rich foods to enhance the wellness, health and lifestyle habits of consumers is globally encouraged. Native fruits are of great interest as they are grown and consumed locally and take part of the ethnobotanic knowledge of the population. Pitanga is an example of a native fruit from Uruguay, consumed as a jelly or an alcoholic beverage. Pitanga has a red-violet pigmentation, which is a common trait for foods that are a good source of antioxidants. Hence, fruits from different Uruguayan regions were analyzed via miniaturized sample preparation method, HPLC-DAD-ESI/MSn and RP-HPLC-DAD techniques to identify and quantify phenolic compounds, respectively. The antioxidant capacity was evaluated via DPPH and ORAC (Oxygen Radical Absorbance Capacity) assays. A multivariate linear regression was applied to correlate the observed antioxidant capacity with the phenolic content. Furthermore, Principal Components Analysis was performed to highlight characteristics between the various samples studied. The main results indicated differences between northern and southern Uruguayan samples. Delphinidin-3-hexoside was present in southern samples (mean of 293.16 µmol/100 g dry weight (DW)) and absent in the sample collected in the north (sample 3). All the samples contain high levels of cyanidin-3-hexoside, but a noticeable difference was found between the northern sample (150.45 µmol/100 g DW) and the southern sample (1121.98 µmol/100 g DW). The antioxidant capacity (mean ORAC of 56370 µmol Trolox®/100 g DW) were high in all the samples compared to the Food and Drug Administration (FDA) database of similar berry-fruits. The results of this study highlight the nutraceutical value of a native fruit that has not been exploited until now.

CRISPR-Cas systems in prokaryotic cells provide an adaptive immunity against invading nucleic acids. For example, phage infection leads to addition of new immunity (spacer acquisition) and DNA cleavage (interference) in the bacterial model species Streptococcus thermophilus, which primarily relies on Cas9-containing CRISPR-Cas systems. Phages can counteract this defense system through mutations in the targeted protospacers or by encoding anti-CRISPR proteins (ACRs) that block Cas9 interference activity. Here, we show that S. thermophilus can block ACR-containing phages when the CRISPR immunity specifically targets the acr gene. This in turn selects for phage mutants carrying a deletion within the acr gene. Remarkably, a truncated acrIIA allele, found in a wild-type virulent streptococcal phage, does not block the interference activity of Cas9 but still prevents the acquisition of new immunities, thereby providing an example of an ACR specifically inhibiting spacer acquisition.

A summary of the resource.
Herein, we report the preparation and characterization of a novel polymeric blend between linear polyethylene imine (PEI) and polyacrylonitrile (PAN), with the purpose of facilitating the dissociation of lithium perchlorate salt (LiClO4) and thus to enhance Li ion transport. It is a joint theoretical and experimental procedure for evaluating and thus demonstrating the lithium salt dissociation. The procedure implies the correlation between the theoretical pair distribution function (PDF) and conventional X-ray diffraction (XRD) by means of a molecular dynamics (MD) approach. Additionally, we correlated the experimental and theoretical Raman and infrared spectroscopy for vibrational characterization of the lithium salt after dissociation in the polymeric blend. We also performed confocal Raman microscopy analysis to evidence the homogeneity on the distribution of all components and the LiClO4 dissociation in the polymer blend. The electrochemical impedance analysis confirmed that the Li–PAN–PEI blend presents a slightly better lithium conductivity of ∼8 × 10–7 S cm–1. These results suggest that this polymer blend material is promising for the development of novel fluorine-free solid polymer lithium ion electrolytes, and the methodology is suitable for characterizing similar polymeric systems.

A summary of the resource.
The presence of carbohydrate-binding proteins, namely lectins, ßgalactosidases and amylases, was determined in aqueous extracts of plants collected in Uruguay. Twenty-six extracts were prepared from 15 Uruguayan plants belonging to 12 Phanerogam families. Among them, 18 extracts caused hemagglutination (HAG) that was inhibited by mono- and disaccharides in 13 cases, indicating the presence of lectins. The other 8 extracts did not cause any HAG with the four systems used to detect HAG activity (rabbit and mouse red cells, trypsin-treated rabbit and mouse red cells). For the extracts prepared from Solanum commersonii, HAG activity and HAG inhibition were similar for those prepared from tubers, leaves and fruits, with the chitocompounds being responsible for all the inhibitions. Purification of the S. commersonii tuber lectin was carried out by affinity chromatography on asialofetuin-Sepharose, and SDS-PAGE under reducing conditions gave a single band of Mr of approximately 80 kDa. The monomer N-acetylglucosamine did not inhibit HAG induced by the purified lectin, but chitobiose inhibited HAG at 24 mM and chitotriose inhibited it at 1 mM. ß-Galactosidase activity was detected in leaves and stems of Cayaponia martiana, and in seeds from Datura ferox. Only traces of amylase activity were detected in some of the extracts analyzed. The present screening increases knowledge about the occurrence of carbohydrate-binding proteins present in regional plants

Searching for a more effective chemotherapy for the treatment of Human African trypanosomiasis, the disease caused by the parasite Trypanosoma brucei, and cancer, in the current work five new [PtII(L)(dppf)](PF6) compounds, with HL = 8-hydroxyquinoline derivatives and dppf = 1,1′-bis(diphenylphosphino)ferrocene, were synthesized and fully characterized. Crystal structures of three compounds were solved by XRD. The compounds displayed fairly good activity against bloodstream T. brucei, with IC50 values in the submicromolar range (IC50: 0.14–0.93 μM), and good selectivity towards the pathogen (SI: 11 - 48) with respect to mammalian macrophages (cell line J774). Coordination to the {Pt-dppf} moiety led, in most cases, to an enhancement of the activity in respect to the bioactive ligands (11 to 41 fold). Cytotoxicity was assessed against wildtype (A2780) and cisplatin-resistance (A2780cisR) ovarian cancer cell lines. Four [PtII(L)(dppf)](PF6) compounds were more active (IC50: 1.2–4.4 μM) than cisplatin (IC50: 26.0 μM) on A2780 cells and showed far superior activity than the reference drug against A2780cisR cells. Platinum levels in A2780 cells showed poor correlation between cellular uptake and the cytotoxic activity. All the complexes interacted with DNA and the most active ones induced reactive oxygen species (ROS) formation which suggested that the mechanism of action for these complexes may be mediated by oxidative stress and interaction with DNA that could act as a potential molecular target for this type of complexes. Some complexes of this series could be considered new hits for the development of prospective agents against trypanosomatid parasites and ovarian cancer.

The oxidation of Met to methionine sulfoxide (MetSO) by oxidants such as hydrogen peroxide, hypochlorite, or peroxynitrite has profound effects on protein function. This modification can be reversed by methionine sulfoxide reductases (msr). In the context of pathogen infection, the reduction of oxidized proteins gains significance due to microbial oxidative damage generated by the immune system. For example, Mycobacterium tuberculosis (Mt) utilizes msrs (MtmsrA and MtmsrB) as part of the repair response to the host-induced oxidative stress. The absence of these enzymes makes Mycobacteria prone to increased susceptibility to cell death, pointing them out as potential therapeutic targets. This study provides a detailed characterization of the catalytic mechanism of MtmsrA using a comprehensive approach, including experimental techniques and theoretical methodologies. Confirming a ping-pong type enzymatic mechanism, we elucidate the catalytic parameters for sulfoxide and thioredoxin substrates (kcat/KM = 2656 ± 525 M−1 s−1 and 1.7 ± 0.8 × 106 M−1 s−1, respectively). Notably, the entropic nature of the activation process thermodynamics, representing ∼85% of the activation free energy at room temperature, is underscored. Furthermore, the current study questions the plausibility of a sulfurane intermediate, which may be a transition-state-like structure, suggesting the involvement of a conserved histidine residue as an acid−base catalyst in the MetSO reduction mechanism. This mechanistic insight not only advances our understanding of Mt antioxidant enzymes but also holds implications for future drug discovery and biotechnological applications.

The electrokinetic and thermostability properties of phosphoglucose isomerase (PGI) in three species of temperate fishes (Perciformes, Acanthopterygii)--Cynoscion striatus, Macrodon ancylodon and Micropogonias furnieri--have been analyzed in order to study the adaptative 2. Unlike most diploid fishes the PGI of these species seemed to be encoded by three PGI loci. 3. The subunits encoded by these loci occurred at different levels in the different tissues and organs analyzed. 4. Genetic variants at two loci (Pgi-A and Pgi-B2) were detected in Cynoscion striatus, and at one (Pgi-A) in Micropogonias furnieri. 5. The product of these loci could be separated in three PGI regions based on their electrophoretic distribution and thermostability properties. 6. The more anodal region (including isozymes and allozymes) was more thermolabile than the less anodal one, which is predominant in a single tissue skeletal muscle. 7. Relative activities of PGI isozymes were compared by Klebe's method to determine pattern of divergence of duplicated gene expression in the three species studied.

El fabricante garantiza Ia calidad, tanto de los adhesivos comercializados (viscosidades, resistencia a mohos, etc.) como de las pe11culas secas. Geneeralmente emplea instrumental muy caro. El usuario no puede inncrementur sus costos con los mismos ensayos. La albúmina sérica humana (HSA), es una proteína monomérica compuesta por tres dominios, conocidos como dominios I, II y III. Presenta en su estructura 17 puentes disulfuro y una única cisteína libre (Cys34) que se localiza en el dominio I (DomI). La HSA es la proteína mayoritaria del plasma y su tiol se encuentra principalmente reducido (HSA-SH), constituyendo el 80% de los tioles reducidos en el compartimiento intravascular. Una fracción menor se encuentra formando disulfuros con tioles de bajo peso molecular (HSA-SSR) u oxidado a ácidos sulfínico (HSA-SO2H) y sulfónico (HSA-SO3H). En esta tesis se profundizó en el análisis de las isoformas de la HSA en plasma mediante cromatoenfoque y se trabajó en la expresión, purificación y caracterización del dominio I (DomI) de la HSA producido en Pichia pastoris. En cuanto al cromatoenfoque, se realizó una puesta a punto del análisis de muestras de plasma de donantes sanos mediante controles de precipitación de HSA en el amortiguador de trabajo y estableciendo la cantidad de HSA a inyectar para obtener una buena resolución. El cromatoenfoque es reproducible y puede utilizarse tal como se puso a punto, para estudiar el aumento de las fracciones oxidadas de la HSA de personas con sepsis. Por otra parte, se expresó el DomI en P. pastoris a partir de dos construcciones. Una conteniendo dos mutaciones sinónimas en su secuencia (DomIMS) y otra generada a partir de la secuencia de referencia de la HSA (DomISR). Ambos DomI se expresaron y secretaron correctamente hacia el medio de cultivo, y mostraron la presencia de un pigmento marrón. Respecto al DomIMS, la eliminación de la mayor parte del pigmento se logró con una cromatografía de intercambio catiónico a pH 5.0 y elución con NaCl 1 M. La reducción selectiva de dicha cisteína se logró por incubación con 2-mercaptoetanol, como se vio por reacción con 5,5´-ditiobis(2-nitrobenzoato) (DTNB) y espectrometría de masa. Además se determinó la constante de reacción de la Cys34 de DomI con DTNB. El valor de 229 M-1 s-1 resultó 16 veces mayor que el de la HSA deslipidada, en iguales condiciones experimentales. En relación al DomISR, se transformaron células de la cepa GS115 His- Mut+, se seleccionaron primero transformantes, y luego transformantes multicopia. Se prepararon crioinóculos que expresaron el DomISR correctamente. En un ensayo preliminar se vio que la Cys34 del DomISR se reduce en iguales condiciones que la del DomIMS y reacciona con DTNB con una constante similar, sugiriendo que no existen diferencias estructurales entre ambos

Abstract
A summary of the resource.
We report a new method for efficient and rapid synthesis of HYNIC-hydrazone derivatives using microwaveassisted solid-phase. In comparison to the conventional heating procedure, microwave irradiation considerably accelerated the formation of HYNIC analogues. The synthesis time and efforts are significantly reduced in the present method, without side product formation. We proved that the HYNIC-hydrazone clustering can be directly labeled with technetium without exposing the free hydrazine group. Therefore, this is a useful tool to obtain HYNIC-bioconjugates for use in 99mTc radiochemistry

Abstract
A summary of the resource.
The influence of the cooking method on the elemental composition of popcorn, prepared according to the way it is consumed, was evaluated. Arsenic, calcium, cadmium, copper, iron, lead, magnesium, manganese, potassium, selenium, sodium and zinc levels were determined using atomic absorption spectrometry. Results showed that the losses after cooking varied considerably (2.9%–48.3%) depending on the element. A bioaccessibility study of cooked samples was also performed, using an in vitro approach. The most bioaccessible elements were sodium (77.4%–88.5%) and potassium (63.7%–81.8%), while considerably low values were observed for calcium (5.5%–13.7%) probably due to strong interactions with the matrix. The contribution to the Recommended Dietary Allowance was evaluated, finding an important coverage for selenium (36.7%– 50.0%). Positive Pearson correlation coefficients were found for the pairs Cu/Fe, Cu/Mn, Fe/Mn Cu/K, Fe/K, K/Mn which can be explained by natural physiological mechanisms of the plant. To the best of our knowledge, this is the first study of bioaccessibility of essential elements in Uruguayan popcorn
