A summary of the resource.
While the contribution of 1,8-cineole to the aroma of wine has been reported, it is a matter of controversy that the vineyards producing such wines are surrounded by Eucalyptus trees, which may contribute their essence to the grapes. However, experimental information presented in this paper suggests that 1,8-cineole can be produced by chemical transformation of limonene and R-terpineol, and this process may be responsible for the occurrence of Eucalyptus-like aroma in Tannat wines from vines not grown in the vicinity of Eucalyptus trees. A mechanism for the chemical transformation of these aroma compounds is proposed.

A summary of the resource.
A relevant subject in biochemistry is the study of diverse effects of ultraviolet (UV) radiation on organisms. While short exposure of culture cells to UV radiation may produce point mutations, deletions, or rearrangements, long exposure may lead to cellular death. In this article, we describe a laboratory project quantifying the effects of UV radiation on a suspension of living cells. This laboratory experiment was designed for undergraduate students in biochemistry. The experiment was carried out employing different fungi strains, changing the dosage of radiation, the distance to the UV source, and the exposure period to the UV radiation. The quantity of viable spores was calculated and the dose–response curves were drawn. Students were able to prove that 100% of mortality was achieved with the longest time of exposure. The percent of cell survival decreased when the distance to the UV source was shorter. After the experimental manipulation, groups of students shared results with one another, and a final report with comparison of the results was to be handed in by each one. Thus, this experiment engaged the students in a mini-research experience. The pedagogic aim of this laboratory design is to familiarize students with chemical UV radiation reactions responsible for cellular and molecular damage.

During the iodoetherification of chiral and conformationally restricted 3‐butenyl carbinols to give 2,5‐disubstituted THFs, the presence of an ester in the side chain exerts a profound effect on the chemo‐ and diastereoselectivity of the reaction. When submitted to standard iodoetherification conditions, ester‐containing substrates gave diastereomeric mixtures of THFs, together with variable amounts of iodohydrins. Conversely, when the ester group was reduced to an alcohol or ether, the corresponding trans‐THFs were obtained more quickly, with complete diastereoselectivity and without contamination by the iodohydrin. In this paper, the mechanistic details of the iodoetherification of protected 3‐butenyl carbinols are revealed. The results show that the carbonyl group decreases the nucleophilicity of the acetonide oxygen atoms, and stabilizes the reactants by a carbonyl–iodonium through‐space interaction. Additionally, it promotes the formation of a stable seven‐membered‐ring carboxonium intermediate, causing an increase in the activation barriers and a decrease in the reaction diastereoselectivity compared to the non‐ester analogues.

New compounds to fght cancer are needed due to cancer high incidence and lack of curative treatments for several classes of this disease. Metal-based coordination compounds ofer a variety of molecules that can turn into drugs. Among them, coordi nation copper complexes are emerging as an attractive class of compounds for cancer treatment. A series of [Cu(l-dipeptide) (tmp)] (tmp=3,4,7,8-tetramethyl-1,10-phenanthroline) complexes were synthesized and characterized in the solid state, including the determination of the crystalline structure of [Cu(Gly-Gly)(tmp)]·3.5 H2O and [Cu2Cl4(tmp)2]. The complexes were studied in solution, where the major species are also ternary ones. The lipophilicity of the complexes was determined and the binding to the DNA was evaluated, suggesting that it occurs in the DNA’s major groove. The cytotoxicity of the com plexes was evaluated on diferent cancer cell lines: human metastatic breast adenocarcinoma MDA-MB-231 (triple negative, ATCC: HTB-26), MCF-7 (ATCC: HTB-22), SK-BR-3 (ATCC: HTB-30), human lung epithelial carcinoma A549 (ATCC: CCL-185), cisplatin resistant-human ovarian carcinoma A2780cis (SIGMA) and nontumoral cell lines: MRC-5 (lung; ATCC: CCL-171) and MCF-10A (breast, ATCC: CRL-10317). [Cu(l-dipeptide)(tmp)] complexes are highly cytotoxic as compared to [Cu(l-dipeptide)(phenanthroline)] and cisplatin. Therefore, [Cu(l-dipeptide)(tmp)] complexes are promising candidates to have their in vivo activity further studied toward new treatments for triple negative breast cancer and other aggressive tumors for which there is no curative pharmacological treatment to the date.

<p>Attitude measurement occupies a central position in consumer research. Concerns over the validity and reliability of traditional measures have motivated the development of alternative approaches. The present research introduces text highlighting as a method for measurement of explicit attitudes using a case study on vertical farming (VF) with 837 UK consumers. They participated in an online survey, where they read a text about VF and used highlighting functions to mark text as ‘like’ and ‘dislike.’ Consumers approached the task in a systematic<br />and logical way and desirable aspects of VF were frequently highlighted as ‘like’, whereas undesirable aspects were more frequently highlighted as ‘dislike’. The text highlighting responses were summarised using word clouds, frequency tables and through sentiment scores to reveal an overall positive attitude to VF among participants.<br />Sentiment scores enabled the identification of consumer segments with interpretable differences in their attitude towards VF. Two approaches to method validation – comparison with direct attitude questions and consumer profiling – further confirmed the potential of the text highlighting method. The sentiment of specific sentences in the text highlighting task matched results from self-reported attitudinal based on Likert scales. Consumer segments with different sentiment in the text highlighting task also differed in their food technology neophobia scores in the expected direction. Future research should investigate methodological aspects of text highlighting and explore its suitability to other applications.</p>

Text highlighting was explored as a new method for direct attitude measurement in cross-cultural research. Participants in an online survey read a text about vertical farming. They used highlighter functions to mark parts of the text they “liked” and “disliked”. Four countries were included in the research—United Kingdom, United States, Singapore, and China (637–683 adults per country). The percentage of participants who highlighted words as “like” and “dislike” was calculated for each of the sentences and countries. Analysis of the highlighting responses did not point to response style or systematic differences in responding. This supported the use of text highlighting in cross-cultural research. Groups of consumers with different sentiment towards specific sentences in the text highlighting task (Positive, Neutral/Ambivalent, or Negative) showed predictable differences in their average scores for attitudinal statements.

<p>Text highlighting is a novel method for measuring consumer attitudes where participants read information about a focal topic and use highlighting functions to select aspects of the text that they like and dislike. The present research contributed methodological knowledge about text highlighting by investigating how responses are influenced by two aspects of the texts — length and degree of reading difficulty. A case study pertaining to biodynamic agriculture was used to assess the research questions and empirical data were collected from 3718 consumers across four countries (United Kingdom, Australia, Germany, and Singapore). Results showed that both text length and reading difficulty influenced responses — overall frequency of highlighting, frequency of ‘like’ highlighting, frequency of ‘dislike’ highlighting, and sentiment scores — leading to recommendations about future implementations of the methodology. Specifically, a single highlighting task on a longer text is less recommended than consecutive highlighting tasks on shorter texts. Implementation of the latter increases highlighting frequency and is expected to be associated with greater participant task engagement. Text length also influenced sentiment scores but did so in a manner that was topic and content specific. The recommendation is to use simple and familiar language that is suited to the groups of participants taking part in the study. Text highlighting was recently introduced as a method for exploring consumer attitudes and perceptions. It consists of asking participants to read a piece of text on a digital device while using highlighting functions to indicate those parts of the text that they ‘like’/feel positive about, as well as those parts they ‘dislike’/feel negative about. As with any new method, methodological research helps to develop recommendations for best practice in use. In this regard, the present research addressed three research questions (RQs) in three studies with participants from New Zealand, United Kingdom, Singapore and Malaysia (ntotal = 5381). In a qualitative approach, Study 1 (RQ1) validated the notion that the text highlighting task is not expected to induce bias. Study 2 addressed RQ2 and explored whether the process of completing the text highlighting task influence consumers’ topic- and product-related responses? There was minimal evidence of this, meaning that text highlighting can reasonably be used together with other types of consumer responses without much the risk of biasing the latter. RQ3 (Study 3) investigated whether the type of highlighting task performed influence text selection. To date the method has been implemented as a dual approach (i.e., selecting both ‘like’ and ‘dislike’ aspects of the text), however, this could tentatively bias either or both responses compared to a single response task where partic ipants select only ‘like’ highlights or only ‘dislike’ highlights. Study 3 found no effect for ‘like’ highlights when comparing dual- and single-response highlighting. However, for ‘dislike’ highlighting, significantly fewer words were selected in the dual-response approach (although the difference in frequency of selection was < 10%).The findings were established in case studies pertaining to kiwifruit and biodynamic agriculture but require confirmation in studies with different products, texts and participants.

Considering that the memory encoding of perceptual components of our experience involves systems that are associated with the perceptual mechanism, textural terms might be organized in consumers’ minds in relation to their perceptual experiences. The aim of this work was to better understand the semantic organization of texture characteristics for Spanish-speaking consumers, trying to obtain a meaningful word categorization, and to study differences between Spanish and Uruguayan consumers. Fifty-two Spanish consumers and 47 Uruguayan consumers were asked to sort 37 textural terms according to their own criteria and to write down a few words related to or describing each group. Spanish and Uruguayan consumers categorized the textural terms in the same way, suggesting that they might have developed the same memory structure. Nevertheless, differences were identified in terms of the number of groups used to classify the textural terms and the words used to describe them.

<p>Baccharis L. (Asteraceae) contains more than 440 species of<br />exclusively American distribution, from USA to Argentina,<br />concentrated mostly in the highland areas of southern Brazil and throughout the Andes. Besides its ethnopharmacological/ medicinal importance, Baccharis volatile fractions (VFs) are important sources of bioactive compounds for flavor and fragrance industry, mainly the species B. dracunculifolia DC. and in a lesser extention B. trimera (Less.), whose essential oils (vassoura and carqueja oil, respectively) are in commerce. However, research and valorization of other Baccharis species as aroma sources remains scarce to date. In this chapter we discuss the relevance of B. uncinella DC., the species more<br />closely related genetically to B. dracunculifolia. Focusing our study on the different techniques and protocols applied to obtain B. uncinella VFs, namely steam distillation (SD, at both, laboratory- and pilot-scales to obtain the “other vassoura essential oil”), simultaneous distillation- extraction (SDE), and supercritical fluid extraction (SFE). The chemical identification and quantitation of VFs was carried out by conventional gas chromatography (GC-FID) and gas chromatography-mass spectrometry (quadrupole analyzer) (GC-qMS). A close look at the data indicated that the chemical composition of B. uncinella VFs largely depended on the extraction technique, thus significantly affecting their aromatic potential. Only SDE allowed the recovery of lighter volatile compounds as C6 components (i.e., “green leaf volatiles”), pilot-scale SD concentrated more hydrocarbons and fewer oxygenated compounds compared to laboratory-scale SD, and SFE recovered several oxygenated monoterpenes and polyquinane sesquitepenes (tentative identification) not obtained by the other techniques. The main components found in the VFs were α-pinene, β-pinene, limonene, (E)-caryophyllene, germacrene D, bicyclogermacrene, spathulenol, and caryophyllene oxide; most of them olfactorily relevant compounds according to previous studies. Therefore, the importance of B. uncinella, its VFs and the “other vassoura oil” as potential sources of aromatic compounds in the flavor and fragrance industry is highlighted.</p>

Herein, we perform a comparative study on the addition of azomethine ylides to graphene, carbon nanotubes, C60, ethene, pyrene and a C48H18 hydrocarbon. The calculated binding energies and free energy corrections suggest that the addition of azomethine ylide to perfect graphene is not spontaneous (�G � 0). However, the presence of Stones–Wales defects significantly increases reactivity: the binding energy between SW-defective graphene and the azomethine ylide is 0.83 eV, close to that determined for a (5,5) SWCNT. The electronic properties of the sheet are not modified by the 1,3 cycloaddition. The binding energies determined for the addition of an azomethine ylide to a (5,5) SWCNT are significantly lower than previously reported.

The 14-3-3 protein, already described in the metacestode of Echinococcus multilocularis, has been characterized in the Echinococcus granulosus adult worm. Immunolocalization studies show the presence of the 14-3-3 protein in the periphery of testes and externally associated with the apical rostellum and adjacent worm tegument. The alcian blue staining in consecutive parasite sections gave similar reactivity patterns, suggesting that the 14-3-3 protein is produced and secreted by rostellar glands. Immunoblot analysis showed the presence of the 14-3-3 protein in somatic and excretory-secretory worm products with higher and smaller apparent molecular masses, respectively, than those detected in E. multilocularis or E. granulosus metacestode tissues. Conversely, the 14-3-3 protein was not detected in metacestode secretory products. Detection of anti-E. granulosus 14-3-3 reactivity in sera of experimentally infected dogs was achieved at early stages of infection. Specific antibody titres decreased during the course of infection. The possible origin and functions of the 14-3-3 protein produced by the adult worm are discussed.

Cystic echinococcosis is caused by the tissue-dwelling larva (hydatid) of Echinococcus granulosus sensu lato. A salient feature is that this larva is protected by the acellular laminated layer (LL). As the parasite grows, the LL sheds abundant particles that can accumulate in the parasite's vicinity. The potential of LL particles to induce inflammation in vivo has not been specifically analysed. It is not known how each of its two major components, namely highly glycosylated mucins and calcium inositol hexakisphosphate (InsP6) deposits, impacts inflammation induced by the LL as a whole. In this work, we show that LL particles injected intraperitoneally cause infiltration of eosinophils, neutrophils and monocytes/macrophages as well as the disappearance of resident (large peritoneal) macrophages. Strikingly, the absence of calcium InsP6 enhanced the recruitment of all the inflammatory cell types analysed. In contrast, oxidation of the mucin carbohydrates caused decreased recruitment of neutrophils. The carbohydrate-oxidised particles caused cell influx nonetheless, which may be explained by possible receptor-independent effects of LL particles on innate immune cells, as suggested by previous works from our group. In summary, LL particles can induce acute inflammatory cell recruitment partly dependent on its mucin glycans, and this recruitment is attenuated by the calcium InsP6 component.

A series of Concanavalin A-Sepharose gels containing between 5 and 480 mg lectin/g of dry gel was prepared by varying the amount of Concanavalin A used in the coupling reaction after activation with 1-cyano-4-(dimethyl-amino)-pyridinium tetrafluoroborate (CDAP) in a modified procedure. The influence of the ligand content on the adsorption / desorption of two different glycoproteins horseradish peroxidase (HRP Mw 40 KDa) and porcine thyroglogulin (Mw 670 KDa) was studied. The adsorption of HRP was found to be dependent on the ligand density and the largest amount adsorbed was obtained when the molar ratio between immobilized Con A and applied HRP was 9/1. The results indicate that highly effective lectin affinity adsorbents can be rationally designed based on the amounts of immobilized lectin

Ethnopharmacological relevance: Lauraceae family includes Nectandra angustifolia a species widely used in the folk medicine of South America against various maladies. It is commonly used to treat different types of processes like inflammation, pain, and snakebites. Snakes of the Bothrops genus are responsible for about 97% of the ophidic accidents in northeastern Argentina. Aim of the study: To evaluate the anti-snake activity of the phytochemicals present in N. angustifolia extracts, identify the compounds, and evaluate their inhibitory effect on phospholipase A2 (PLA2) with in vitro and in silico assays. Methods: Seasonal variations in the alexiteric potential of aqueous, ethanolic and hexanic extracts were evaluated by inhibition of coagulant, haemolytic, and cytotoxic effects of B. diporus venom. The chemical identity of an enriched fraction obtained by bio-guided fractioning was established by UPLC-MS/MS analysis. Molecular docking studies were carried out to investigate the binding mechanisms of the identified compounds to PLA2 enzyme from snake venom. Results: All the extracts inhibited venom coagulant activity. However, spring ethanolic extract achieved 100% inhibition of haemolytic activity. Bio-guide fractioning led to an enriched fraction (F4) with the highest haemolytic inhibition. Five flavonoids were identified in this fraction; molecular docking and Molecular Dynamics (MD) simulations indicated the binding mechanisms of the identified compounds. The carbohydrates present in some of the compounds had a critical effect on the interaction with PLA2. Conclusion: This study shows, for the first time, which compounds are responsible for the anti-snake activity in Nectandra angustifolia based on in vitro and in silico assays. The results obtained in this work support the traditional use of this species as anti-snake in folk medicine.
