Fungal lectins constitute excellent ligands for development of affinity adsorbents useful in affinity chromatography. In this work, a lectin was purified from Pycnoporus sanguineus (PSL) mycelium using 3 procedures: by affinity chromatography, using magnetic galactosyl-nanoparticles or galactose coupled to Sepharose, and by ionic exchange chromatography (IEC). The highest lectin yield was achieved by IEC (55%); SDS-PAGE of PSL showed 2 bands with molecular mass of 68.7 and 55.2 kDa and IEC displayed 2 bands at pi 5.5 and 5.2. The lectin agglutinates rat erythrocytes, exhibiting broad specificity toward several monosaccharides, including galactose. The agglutination was also inhibited by the glycoproteins fetal calf fetuin, bovine lactoferrin, bovine transferrin, and horseradish peroxidase. The lectin was then used to synthesize an affinity adsorbent (PSL-Sepharose) and the interaction with glycoproteins was evaluated by analyzing their chromatographic behaviors. The strongest interaction with the PSL-derivative was observed with transferrin, although lower interactions were also displayed toward fetuin and lactoferrin. These results indicate that the purified PSL constitutes an interesting ligand for the design of affinity adsorbents to be used (i.e., in glycoprotein purification). Forest plantations inUruguay have doubled in the past decade, with Eucalyptus spp. leading this growth. The bronze bug, Thaumastocoris peregrinus (Heteroptera: Thaumastocoridae), originally restricted to Australia, is an important emerging pest of Eucalyptus plantations in the Southern hemisphere. T. peregrinus feeds onmature Eucalyptus leaves, causing them to turn brown and often fall from the tree. Although population dynamics and behavioural patterns are not clearly understood, circumstantial observations suggest that males and nymphs aggregate. We used gas chromatography coupled to mass spectrometry to analyze volatile organic compounds emitted by virgin males and females, and characterized a male-specific compound, 3-methylbut-2-enyl butanoate, based on mass spectral data and chromatographic comparison with a synthetic standard. We also performed Y-olfactometer bioassays to test the attraction of virgin males and females toward live virgin males, male volatile extracts, and synthetic 3-methylbut- 2-enyl butanoate. Males were attracted toward conspecific males, while virgin females showed no preference, suggesting that male volatiles are not involved in sexual communication. Further olfactometer tests showed that males were attracted to male volatile extracts and to synthetic 3-methylbut-2-enyl butanoate. The ecological significance of this compound and its potential use for the management of T. peregrinus in Eucalyptus forests will be further investigated. An automatic programmable-flow system capitalizing upon mesofluidic Lab-On-Valve (LOV) sample processing coupled on-line to inductively coupled plasma atomic emission spectrometry (ICP-AES) is proposed in this work as a quality control tool for expedient assessment of potential contamination episodes of food safety elements (namely, Cd and Pb) in a variety of undigested ripened honeys, taken as a model of an edible matrix. On-chip micro-solid phase extraction (μSPE) is effected in a bead-injection disposable sorbent mode using a cationic exchange restricted access-like material (viz., Bond Elut Plexa PCX). We have proven that the lifetime of the miniaturized solid reactor on-chip (11 ± 0.6 mg) is limited to processing less than 6 mL of unfiltered sample suspension (5% (w/w) honey buffered at pH 4.5) after which the analytical performance is severely deteriorated because of strong adherence of the sample matrix to the bead surface. Several physicochemical parameters including the nature of the sorbent material and its ionic form were investigated in detail so as to maximize absolute recoveries and enrichment factors of Pb and Cd. Variables for the elution process were explored by means of a full factorial design. Using 4.0 mL of sample and 200 μL of 3.0 mol L−1 HNO3 as an eluent, enrichment factors of ca. 15 with extraction/elution efficiencies close to 80% and relative recoveries ranging from 90–111% in honey were obtained for both Pb and Cd. The limits of detection (LODs), based on the 3sintercept criterion, were 26 ng g−1 and 68 ng g−1 for Cd and Pb, respectively, which are far below those endorsed by current regulatory agencies (namely, 100 and 300–500 ng g−1 for Cd and Pb, respectively). Demonstrated by the analysis of a suite of off-the-shelf honey brands, the proposed LOV-μSPE platform hyphenated to ICP-AES is deemed suitable for the reliable quantitation of trace level concentrations of Pb and Cd in honey and the detection of heavy metal contamination episodes. A 200 ps molecular dynamics trajectory for the 8634 atoms corresponding to the enzyme glutathione reductase were calculated with the GROMOS program. This enzyme is formed by two identical subunits, with 478 residues and a flavinadenine dinucleotide (FAD) molecule each. Four regions are well distinguished in its crystallographic structure, the dimer interface, the FAD binding site, the active site and the crystal contact region. The analysis shows an equilibrated trajectory after 40 ps. The r.m.s. difference between the X-ray structure and the conformation along the simulation is 2.4 Å when considering the r.m.s. of the whole molecule. The structure is locally well conserved. Cystic echinococcosis is still a major concern in South America. While some regions show advances in the control of the disease, others have among the highest incidence in the world. To reverse this situation the Pan American Health Organization (PAHO) has launched a regional project on cystic echinococcosis control and surveillance. An early concern of the program was the lack of a standardized diagnostic tool to monitor infection in dogs, a key target of control programs. Under this premise, we have developed a new copro-ELISA test after extensive screening of a large panel of monoclonal antibodies (MAbs) and polyclonal sera, which performs with high standards of sensitivity (92.6%) and specificity (86.4%) as established by necropsy diagnosis of dogs. The key component of the test, MAbEg9 has a convenient IgG isotype and reacts with a periodate-resistant epitope found in high molecular weight components of the worm. Time-course analysis of experimentally infected dogs showed that even animals with a very low number of parasites could be detected as early as day 20 post infection. The test was formulated in a ready-to-use kit format with proven stability of each component for a minimum of 3 months at room temperature. This characteristic facilitates its standardized use and shipping to other laboratories, which was demonstrated by the identical results obtained by two different laboratories in Peru and our own laboratory when a large number of field samples were analyzed independently in a blind fashion. The search for protective antigens of intestinal parasites is conditioned by the methodology used to induce a relevant local immune response against them. The present work describes the use of immuno stimulating complexes (iscoms) from tegumental antigens from protoscoleces (PSC) of the cestode Echinococcus granulosus as immunogens in dogs by the intranasal route. It also describes the evaluation of the immune response evoked at the antibody level (systemically and at a distant mucosal location) as well as at the level of antibody secreting cells in peripheral blood. Iscoms from both E. granulosus tegumental antigens and hen ovalbumin (OVA), given at 50 μg doses by intranasal route, evoked significant secretory IgA antibody responses detected in saliva. Specific IgA secreting cells in peripheral blood also increased 10–20-fold, although transiently, after primary and secondary stimulation, whereas specific IgG secreting cells in peripheral blood were only detected in some individuals after the second antigenic exposure. Generation of immune responses at a related mucosal site provides evidence of localised immunity. No significant increase in systemic antibody titers of either IgM, IgG or IgA isotype was detected in plasma as a result of the immunisation. The flow manifold was designed to implement the multicommutation approach and it comprised three 3-way solenoid valves controlled by a personal computer. The flow system presented allowed to process the oil samples to determine wear metals without any prior preparation. Aiming to assess accuracy the results were compared with those obtained by manual procedure using flame AAS. A new method is presented for the automation of the determination of dextrose in parenteral and hemodialysis solutions. The method is based on multicommutation flow analysis (MCFA) and exploits enzymatic reactions providing a colored derivative that is detected spectrophotometrically (Trinder’s method). The reagent, comprising glucose oxidase, peroxidase, 4-hydroxybenzoate and a buffer was obtained from a commercial kit for the determination of glycemia. The flow system used three 3-way solenoid valves operating under computer control. The necessary software was developed for the purpose and compiled in QuickBASIC 4.0 The influence of some operating variables (segment size, number of segments and reactor length) was studied. Calibration curves in the range 0–1 g/L presented a slight curvature and were fitted with a second-degree polynomial (h = −0.0632C2 + 0.6039C + 0.166, r2 = 0.9973, h being the peak-height (absorbance) and C the concentration in g/L). The method was validated by analyzing artificial samples presenting accurately known concentrations of dextrose, and comparing the results with the known value and with value obtained by polarimetry. Recoveries were in the range 96.6–100.2%, and the difference with the polarimetric analysis was in the range 0.1–3.3%. Precision (R.S.D.,%) was better than 2.4%. Sampling frequency of the system was 90 samples/h, with a reagent consumption of 0.14mL per sample. A multiproxy analysis of diatoms, chrysophyte cysts, opal phytoliths, and palynomorphs was undertaken in Negra Lagoon, to decipher Holocene changes in paleobotanical proxies associated with sea-level changes and paleolimnological conditions. Before the Holocene transgression (7000 yr BP), a terrestrial system was inferred as no aquatic palynomorphs or biogenic silica remains were recorded. During the sea-level maximum (5200 yr BP), marine/ brackish conditions were established as indicated by diatoms, Chenopodiaceae, and a high content of cysts of Peridinioideae. The catchment consisted of grasslands and wetlands as indicated by the opal phytolith data. The Holocene transgression was followed by a sea-level decrease, which led to the onset of brackish/freshwater conditions as inferred from the co-dominance of freshwater and marine/ brackish diatoms. This is also supported not only by the concomitant increase in non-siliceous freshwater microalgae and emergent macrophytes, but also by the presence of Myriophyllum. As sea level continued to decrease during late Holocene, a freshwater system was observed because of the complete separation from the ocean. Chrysophyte cysts were consistently recorded and brackish diatoms exhibited reduced abundances. The observed limnological changes are consistent with the alternative states hypothesis of Scheffer’s model (1998). That is, the clear water phases were explained because of high macrophyte abundances, low phytoplankton frequencies, high cyst to diatom ratios, and increase in frequencies of benthic diatom species. Conversely, turbid phases were likely to occur when low cyst to diatom ratios together with increase in planktonic diatoms and decrease in macrophytes values were detected. This highlights the importance of this approach to detect long-term changes in shallow lake alternative states. Antibiotic resistance is a major threat to global public health. β-lactamases, which catalyze breakdown of β-lactam antibiotics, are a principal cause. Metallo β-lactamases (MBLs) represent a particular challenge because they hydrolyze almost all β-lactams and to date no MBL inhibitor has been approved for clinical use. Molecular simulations can aid drug discovery, for example, predicting inhibitor complexes, but empirical molecular mechanics (MM) methods often perform poorly for metalloproteins. Here we present a multiscale approach to model thiol inhibitor binding to IMP-1, a clinically important MBL containing two catalytic zinc ions, and predict the binding mode of a 2-mercaptomethyl thiazolidine (MMTZ) inhibitor. Inhibitors were first docked into the IMP-1 active site, testing different docking programs and scoring functions on multiple crystal structures. Complexes were then subjected to molecular dynamics (MD) simulations and subsequently refined through QM/MM optimization with a density functional theory (DFT) method, B3LYP/6-31G(d), increasing the accuracy of the method with successive steps. This workflow was tested on two IMP-1: MMTZ complexes, for which it reproduced crystallographically observed binding, and applied to predict the binding mode of a third MMTZ inhibitor for which a complex structure was crystallographically intractable. We also tested a 12-6-4 nonbonded interaction model in MD simulations and optimization with a SCC-DFTB QM/MM approach. The results show the limitations of empirical models for treating these systems and indicate the need for higher level calculations, for example, DFT/MM, for reliable structural predictions. This study demonstrates a reliable computational pipeline that can be applied to inhibitor design for MBLs and other zinc-metalloenzyme systems. Colon cancer has a high prevalence worldwide and is a serious public health problem. Early diagnosis greatly improves its prognosis and, among the existing methods, the detection of fecal occult blood is the only noninvasive test recommended for screening of the disease. To promote its massive application as a screening tool for asymptomatic populations in low-resource settings, the availability of a reliable and cost-effective method is imperative. Here, we describe the development and validation of a sensitive nanobody-based immunoassay for the detection of hemoglobin in human fecal samples. The nanobodies were selected from a library generated from a llama immunized with human hemoglobin, using a high-throughput platform that enabled the identification of the best nanobody pair. The assay allowed a sub-ng/mL limit of detection to be reached in phosphate-buffered saline, and was validated with stool samples, showing excellent reproducibility (CV% < 15 inter-day precision) and accuracy at 2 and 4 μg of hemoglobin per gram of feces, which are well below the recommended cutoff for this test (10–20 μg/g). Moreover, no cross-reactivity was observed with a panel of dietary non-human hemoglobins removing the need for pre-test dietary restrictions. Considering that the monodomain nature of nanobodies facilitates their straightforward and low-cost production by bacterial fermentation, with their provided sequences and using synthetic genes, the assay reported here could be replicated in any laboratory to perform thousands of tests for early detection of colorectal cancer at almost no cost. The horn fly, Haematobia irritans (L.), is a blood-sucking ectoparasite that is responsible for sizeable economic losses in livestock. The salivary gland products facilitate blood intake. Taking advantage of the identification of novel H. irritans salivary antigens (Hematobin, HTB and Irritans 5, IT5), we investigated the parasite loads, H. irritans blood intake, and antibody response of naturally infected bovines during the fly season. Fly loads and fly hemoglobin content fluctuated during the trial. Each time horn fly loads exceeded 200 flies per cattle, a reduction in horn fly blood intake was observed three weeks later. All of the cattle elicited an antibody response against HTB and IT5 that declined once the fly season was over. Cattle anti-IT5 titers were positively correlated with parasite loads and negatively correlated with fly blood intake. These results suggest that the natural changes in the H. irritans blood intake observed in this study were associated with a natural host response against horn fly salivary antigens. Plants can be a useful source of these lead compounds. Xanthium cavanillesii Schouw, Asteraceae, grows wild in Uruguay and its infusion is used in traditional medicine as a skin antiseptic. A comprehensive study of the different classes of cycloaddition reactions ([3+2], [2+2], and [2+1]) of SO2 to acetylene and ethylene has been performed using density functional theory (DFT) and composite wavefunction methods. The [3+2] cycloaddition reaction, that was previously explored in the context of the cycloaddition of thioformaldehyde S-methylide (TSM) to ethylene and acetylene, proceeds in a concerted way to the formation of stable heterocycles. In this paper, we extend our study to the [2+2] and [2+1] cycloadditions of SO2 to acetylene, which would produce 1,1-oxathiete-2-oxide and thiirene-1,1-dioxide, respectively. One of the main conclusions is that cyclic 1,1-oxathiete-2-oxide can open through a relatively easy breaking of the S O single bond and rearrange toward sulfinyl acetaldehyde (SA). The SA molecule can easily undergo several internal rearrangements, which eventually lead to sulfenic acid and sulfoxide derivatives of ethenone, 1,2,3-dioxathiole, and CO plus sulfinylmethane. The most probable path, however, produces 2-thioxoacetic acid, whose derivatives (or those of the corresponding acetate) are usually obtained by Willgerodt–Kindler-type sulfuration of acetates. This product can in turn decompose, leading to the final products CO2 and H2C S. Comparison of this decomposition path with that of 2-amino-2-thioxoacetic acid shows that the process occurs through different H-transfer processes. In this study, we addressed the biochem. characterization of a new class of Trx-related protein (IsTRP) and a classical monothiol Grx (EgGrx5) from the human pathogen Echinococcus granulosus. Results: The dimeric form of IsTRP coordinates Fe S in a glutathione-independent manner; instead, Fe/S binding relies on the CXXC motif conserved among Trxs. This novel binding mechanism allows holo-IsTRP to be highly resistant to oxidn. IsTRP lacks canonical reductase activities. Mitochondrially targeted IsTRP aids growth of a Grx5 null yeast strain. Similar complementation assays performed with EgGrx5 revealed functional conservation for class II Grxs, despite the presence of nonconserved structural elements. IsTRP is a cestode lineage-specific protein highly expressed in the gravid adult worm, which releases the infective stage crit. for dissemination. Innovation: IsTRP is the first member from the Trx family to be reported to bind Fe/S. We disclose a novel mechanism of Fe/S coordination within the Trx folding unit, which renders the cluster highly resistant to oxidn.-mediated disassembly.
