The catalytic performance of trimetallic oxides MeLaZr (Me: Ni, Co) was evaluated in the steam reforming of ethanol for hydrogen production. Rh doped and non doped catalysts were prepared by the pseudo sol–gel like method, characterized by X-ray diffraction (XRD), thermo-programmed reduction (TPR), micro-homogeneity analysis (TEM-EDXS), specific surface areas (BET). A phase-segregation process takes place during the calcination for both trimetallic systems leading to the formation of La2Zr2O7 pyrochloretype compound in coexistence with the Me-oxide. NiLaZr and CoLaZr catalysts show high activity and high selectivity for hydrogen production. Addition of Rh promotes reducibility at lower temperatures for both trimetallic catalysts and remarkably improves the activity and selectivity for H2. The H2 selectivity is lowered by CO2 injection because of the increase of the rate of the reverse WGSR and/or methanation reaction. The CH4 selectivity is lowered by the dry reforming reaction. The key-role played by oxycarbonate species formed during the reforming tests leads to a highly efficient gasification of carbon deposits by the Boudouard reaction. Nitro-fatty acids represent endogenously occurring products of oxidant-induced nitration reactions. We have previously synthesized amixture of four isomers of nitroarachidonic acid, a novel anti-inflammatory signaling mediator. In this study, we synthesized and chemically and biologically characterized for the first time an esterified nitroalkene derived from the nitration of methylarachidonate (AAMet): 6-methylnitroarachidonate (6-AAMetNO2). Synthesis was performed by reacting AAMet with sodium nitrite under acidic conditions. Analysis by mass spectrometry (positive-ion ESI-MS) showed an [M+H]+ ion of m/z 364, characteristic of AAMetNO2. Fragmentation of this ion yielded a daughter ion at m/z 317, corresponding to the neutral loss of the nitro group ([M+H−HNO2]+). Furthermore, IR signal at 1378 cm−1 and NMR data confirmed the structure of a 6-nitro-positional isomer. This novel esterified nitroalkene was capable of promoting vascular protective actions including: (a) the induction of vasorelaxation via endothelium-independent mechanisms, associated with an increase in smooth muscle cell cGMP levels, and (b) a potent dose-dependent inhibition of human platelet aggregation. We postulate that 6-AAMetNO2 could be a potential drug for the prevention of vascular and inflammatory diseases, and the presence of the methyl group may increase its pharmacological potential. Background and Aims: Changes in different polyphenol families during grape ripening and vinification were determined in Tannat grapes. This was done to establish a polyphenol profile for Tannat grapes during ripening and for young wines. Methods and Results: We demonstrated, by high-performance liquid chromatography with diode array detection and mass spectrometry (HPLC-DAD-MS) analysis, that the content of flavan-3-ols in Tannat seeds was higher than that reported for a large number of other grape varieties analysed. Forty per cent of the total flavan-3-ols in seeds were galloylated compounds, whereas the flavan-3-ol profile in skins was characterised by the absence of galloylated forms. Prodelphinidins in skins ranged between 30 and 35% with very low values for epigallocatechin. Epicatechin gallate was not detected in Tannat wine and galloylated forms represented a low percentage of total flavan-3-ols. Tannat grapes had very high concentrations of anthocyanins in skins with levels increasing during ripening. Eleven phenolic acids were identified in Tannat grape skins and wines, but only gallic and protocatechuic acids were found in the seeds. Conclusions: Tannat wines were shown to have one of the highest levels of phenolic compounds reported for vinifera grape varieties. The profiles for different polyphenol families present in Tannat grapes (skins and seeds) and wines were determined. In most organisms, thioredoxin (Trx) and/or glutathione (GSH) systems are essential for redox homeostasis and deoxyribonucleotide synthesis. Platyhelminth parasites have a unique and simplified thiol-based redox system, in which the selenoprotein thioredoxin-glutathione reductase (TGR), a fusion of a glutaredoxin (Grx) domain to canonical thioredoxin reductase domains, is the sole enzyme supplying electrons to oxidized glutathione (GSSG) and Trx. This enzyme has recently been validated as a key drug target for flatworm infections. In this study, we show that TGR possesses GSH-independent deglutathionylase activity on a glutathionylated peptide. Furthermore, we demonstrate that deglutathionylation and GSSG reduction are mediated by the Grx domain by a monothiolic mechanism and that the glutathionylated TGR intermediate is resolved by selenocysteine. Deglutathionylation and GSSG reduction via Grx domain, but not Trx reduction, are inhibited at high [GSSG]/[GSH] ratios. We found that Trxs (cytosolic and mitochondrial) provide alternative pathways for deglutathionylation and GSSG reduction. These pathways are operative at high [GSSG]/[GSH] and function in a complementary manner to the Grx domain-dependent one. Despite the existence of alternative pathways, the thioredoxin reductase domains of TGR are an obligate electron route for both the Grx domain- and the Trx-dependent pathways. Overall, our results provide an explanation for the unique array of thiol-dependent redox pathways present in parasitic platyhelminths. Finally, we found that TGR is inhibited by 1-hydroxy-2-oxo-3-(N-3-methyl-aminopropyl)-3-methyl-1-triazene (NOC-7), giving further evidence for NO donation as a mechanism of action for oxadiazole N-oxide TGR inhibitors. Thus, NO donors aimed at TGR could disrupt the entire redox homeostasis of parasitic flatworms. The strong correspondence between the spatial arrangement of archaeological sites and wetland environments is one of the main axes of the Merín Lagoon basin archaeology. Without implying a deterministic simplification from the epistemological point of view, the cultural and environmental history of the region hold evidence suggesting a more complex scenario, where cultural responses to climate change were not so mechanical or direct as previously argued. The relationship between paleoclimate and prehistoric mound builders development between 7.0 and 0.6 ka 14C BP was studied in SE Uruguay. Paleoclimatic data were inferred from the phytolith record of three lagoon sediment cores. Four paleoclimatic periods were identified according to temperature/humidity changes. The first period, dated at 7.0e4.5 ka 14C BP, was characterized by a warm/humid climate. Between 4.5 and 3.5 ka 14C BP, the onset of a cool/dry climate was inferred. A hiatus between 3.5 and 2.6 ka 14C BP was identified. In the third period, 2.5e1.2 ka 14C BP, similar climate conditions to those of the present were inferred. Finally, the period 1.2 and 0.6 ka 14C BP, was characterized by warmer and wetter conditions than those of the present. According to these data, it was determined that mounds were first observed during the transition from warm/humid to cool/dry conditions, but the complexity of the mounds increased after the full establishment of cool/dry conditions. With the inception of warm/humid conditions, an increased number of mounds was observed in the plains located close to the Merín Lagoon littoral. The mounds showed lower density and height than those recorded for the cool/dry period. The aim of this work was to evaluate the potential of lufenuron, a benzylphenylurea with ability to interfere with the formation of insect exoskeleton, as a therapeutic drug for larval echinococcosis (hydatid disease). For this purpose lufenuron, alone or in combination with albendazole, was administered to CD1 mice bearing Echinococcus granulosus hydatid cysts in the peritoneal cavity. Neither of the drugs alone was able to exert parasiticidal effects. However, in combination with albendazole, lufenuron reduced the growth of cysts (30–40% in cyst diameter respect to control, p < 0.05). This effect was associated with ultrastructural alterations of the hydatid cyst wall and a reduction of the content of myo-inositol-hexakisphosphate, the major component of the electron dense granules of the laminated layer. Overall, this work provides evidence that lufenuron could represent a useful compound for the use in chemotherapy against larval echinococcosis, by enhancing albendazole parasiticidal activity. Nitric oxide donor tocopherol analogs were found to be incorporated in low-density lipoprotein to release nitric oxide into the hydrophobic core of the lipoprotein, thus inhibiting lipid oxidation processes associated with atheroma plaque formation. Previously, we studied their cytotoxicity against human and murine macrophages as first selection for in vivo studies. Herein, we examined both the in vitro mutagenic and DNA-damage effects of selected compounds to further evaluate drug potential. While the compounds of interest were nongenotoxics in both experimental tests (Ames and alkaline comet), one of the potential blood metabolites exhibited genotoxicity (alkaline comet test), and the furazan derivative was mutagenic (Ames test). Two selected (nitrooxy and furoxan) compounds were studied in long- and short-term in vivo treatment, and in these conditions, animal toxicity was not evidenced, suggesting the possibility of these compounds as potential antiatherogenic drugs. Some results obtained during our research work in the search of anti-snake compounds from plant origin, allow us to propose sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) as a valuable method for a fast and reliable screening in order to evaluate plant extracts activity on snake proteins from Bothrops diporus (yarará chica). Such approach will allow to process a larger number of plant extracts and to select the active ones. Venoms used in this study came from B. diporus which was previously vacuum dried. Extracts (aqueous, alcoholic and hexanic) were from native plants: Aristolochia elegans, Aristolochia gibertii, Asclepia curassavica, Cissampelos pareira, Dorstenia brasiliensis, Eclipta prostrata, Iresine diffusa, Mikania micrantha, M. periplocifolia, M. coridifolia, Nectandra angustifolia, N. megapotamica, Sapium haematospermum and Trixis divaricata. The results obtained by SDS-PAGE were compared with those obtained from in vitro assays (coagulation and hemolysis inhibition). The correlation between results obtained from electroforetic and in vitro assays allowed to suggest SDS-PAGE as a suitable technique to assist in preliminary plant screenings for anti-snake activity by snake venom protein interaction with plant compounds. Cyanobacterial 16S ribosomal RNA gene diversity was examined in a benthic mat on Fildes Peninsula of King George Island (62809054.400S, 58857020.900W), maritime Antarctica. Environmental DNA was isolated from the mat, a clone library of PCR-amplified 16S rRNA gene fragments was prepared, and amplified ribosomal DNA restriction analysis (ARDRA) was done to assign clones to seven groups. Low cyanobacterial diversity in the mat was suggested in that 83% of the clones were represented by one ARDRA group. DNA sequences from this group had high similarity with 16S rRNA genes of Tychonema bourrellyi and T. bornetii isolates, whose geographic origins were southern Norway and Northern Ireland. Cyanobacterial morphotypes corresponding to Tychonema have not been reported in Antarctica, however, this morphotype was previously found at Ward Hunt Lake (838N), and in western Europe (528N). DNA sequences of three of the ARDRA groups had highest similarity with 16S rDNA sequences of the Tychonema group accounting for 9.4% of the clones. Sequences of the remaining three groups (7.6%) had highest similarity with 16S rRNA genes of uncultured cyanobacteria clones from benthic mats of Lake Fryxell and fresh meltwater on the McMurdo Ice Shelf. Las bacterias endófitas son bacterias que viven dentro de los tejidos vegetales sin causar daño al organismo hospedero. Se ha postuladoque estas bacterias pueden aumentar la productividad de los cultivos por distintos mecanismos relacionados al aporte de nutrientes, producción de hormonas, y protección contra stress biótico y abiótico. En esta tesis se llevó a cabo el estudio de las comunidades de bacterias endófitas presentes en los distintos tejidos vegetales de arroz utilizando principalmente herramientas moleculares independientes del cultivo de microorganismos en el laboratorio. Se seleccionó este cultivo porque es de gran importancia económica y social para nuestro país y porque la planta y el suelo experimentan un cambio drástico durante el ciclo, como es la inundación. Se estudiaron la comunidad total y dos grupos fisiológicos particulares con relevancia biotecnológica y ambiental (bacterias diazótrofas y metanótrofas) así como distintos factores que pueden afectarlas (variedad de arroz utilizada, inundación del cultivo, tipo de suelo y su historia agrícola). Los resultados obtenidos revelaron que en las hojas de las tres variedades de arroz más sembradas en Uruguay la estructura de la comunidad de bacterias endófitases muy similar. Mientras Brevundimonas fue el género dominante en el análisis molecular (clonado del gen 16S rRNA) Pantoea ananatis y Pseudomonas syringae fueron las especies más frecuentemente aisladas en las tres variedades de arroz y en las dos zafras analizadas Se observó que la inundación como práctica habitual de manejo del cultivo de arroz tiene efectos muy importantes sobre la abundancia, diversidad y composición de las comunidades de bacterias diazótrofas endófitas de raíz. La abundancia de genes nifH, codificante para la dinitrogenasa reductasa, y su diversidad aumentan sustancialmente después de la inundación. Mientras que antes de inundar prevaleció la clase Gamaproteobacteria (66% de genoteca del gen nifH, dominando Stenotrophomonas con el 49% de las secuencias de nifH recuperadas), luego de la inundación se observó una gran diversidad de secuencias de genes nifH de bacterias anaerobias estrictas y facultativas (Spirochaetes, Chlorobi, Deltaproteobacteria y diversos Firmicutes). Estos resultados sugieren que los diazótrofos anaerobios podrían tener un rol importante en la fijación de nitrógeno en este ecosistema una vez que el cultivo de arroz es inundado. Asimismo, los diazótrofos endófitos de hoja se encuentran en una proporción casi 100 veces menor que en raíz y suelo de acuerdo a la cuantificación de genes nifH realizada por q-PCR. Las raíces de plantas de arroz sembradas en suelos de alta fertilidad con historia previa de cultivo de arroz presentaron una abundancia significativamente mayor que las raíces de las plantas sembradas en otros suelos, incluso mayor que aquellas sembradas en suelos similares que no habían sido utilizados previamente para el cultivo de arroz. La abundancia de las poblaciones diazótrofas endófitas de hoja parece ser independiente de las propiedades de los suelos en los las plantas se cultivan. Del mismo modo, no se observaron diferencias significativas en el número de copias del gen nifH en los distintos suelos, a pesar de sus características fisicoquímicas e historia de uso del suelo diferentes. 2 Por otra parte, las bacterias metanótrofas, de gran importancia porque contribuyen a mitigar las emisiones de metano producidas en el ecosistema arrocero, fueron detectadas en bajo número tanto en hojas como en raíces de plantas de arroz crecidas en suelo. Para ambos tejidos se obtuvieron cultivos altamente enriquecidos en metanótrofas capaces de crecer en ausencia de nitrógeno agregado indicando que se trata de metanótrofas diazótrofas. Esta propiedad podría ser importante en la interacción de este grupo fisiológico con la planta. La caracterización molecular de estos enriquecimeintos confirmó que Methylosinus trichosporium, una cepa metanótrofa C9 (aislada previamente de rizósfera de arroz en Uruguay) y Methylocystis hirsuta, se encuentran presentes en los tejidos internos de las hojas de arroz. El estudio realizado permitió generar nuevos conocimientos sobre las comunidades de bacterias endófitas nativas de arroz y profundizar en los factores que las afectan aportando información relevantesobre la interacción bacteria- planta. Asimismo, los conocimientos generados podrían ser útiles en la búsqueda de nuevas cepas con potencial biotecnológico y en la racionalización del uso potencial de inoculantes en el cultivo de arroz. In this study, the microbial community characteristics in continuous lab-scale anaerobic reactors were correlated to reactor functionality using the microbial resource management (MRM) approach. Two molecular techniques, denaturing gradient gel electrophoresis (DGGE) and terminal-restriction fragment length polymorphism (TRFLP), were applied to analyze the bacterial and archaeal communities, and the results obtained have been compared. Clustering analyses showed a similar discrimination of samples with DGGE and T-RFLP data, with a clear separation between the meso- and thermophilic communities. Both techniques indicate that bacterial and mesophilic communities were richer and more even than archaeal and thermophilic communities, respectively. Remarkably, the community composition was highly dynamic for both Bacteria and Archaea, with a rate of change between 30% and 75% per 18 days, also in stable performing periods. A hypothesis to explain the latter in the context of the converging metabolism in anaerobic processes is proposed. Finally, a more even and diverse bacterial community was found to be statistically representative for a wellfunctioning reactor as evidenced by a low Ripley index and high biogas production. Calcination was considered for the first time as an alternative to recover inorganic bone component from spent glucoamylase-bone derivatives. The subsequent adsorption of glucoamylase (GA) onto calcined bone particles was assessed. Adsorption capacity of the calcined matrix was found to be from 1.1- to 1.4-fold superior to that of non-calcined supports, and it was dependent on the applied load. Moreover, the expressed activity of GA derivatives on calcined matrix was, at least, 2-fold higher than that of biocatalysts onto non-calcined support. The optimization of the loading allowed the preparation of derivatives with 139 GA units per gram of support, which preserve 52% of the immobilized activity. Additionally, calcination of spent GA biocatalysts on calcined bone particles was performed, and adsorption of glucoamylase onto the bone particles calcined a second time was also found to be efficient. In addition to the improved catalytic properties, the half-life at 55°C of the GA biocatalysts on calcined bone was increased 1.7- fold in comparison with that of soluble GA and GA adsorbed onto non-calcined bone particles. Furthermore, the same cassava starch conversion can be achieved batchwise in a stirred-tank reactor using less insoluble biocatalyst, 37% of the GA-bone derivative, which represents an important saving for industrial applications. This research assesses the bench-scale application of a non-conventional support, bone particles, for glucoamylase (GA) immobilization and its subsequent use in cassava starch hydrolysis. Upon determining the appropriate conditions to immobilize GA onto chicken bone particles, such as pH, ionic strength, particle size, and enzyme load, bench-scale immobilization of commercial GA without further purification was performed. Optimal temperature and thermal stability of immobilized GA were only slightly different from those of the free enzyme, while optimal pH became more acidic by about one unit. The feasibility of the use of this immobilized biocatalyst for high glucose syrup production from liquefied cassava starch, at bench scale in batch process using a stirred-tank reactor, was demonstrated. Repeated use of the GA-bone derivative showed that similar conversions to those achieved with soluble enzyme (dextrose equivalent=98) were reached until the third batch and over 90% until the 25th batch. Although the ancient Vitis vinifera Tannat variety is virtually unknown in the international wine market, the strategy to promote it as the Uruguayan emblematic grape variety is starting to become successful in the red wines market. This paper presents a summary of the latest research and developments performed in Uruguay with Tannat since the late 1970s, its origin and production statistics. As an expression of the state-of-the-art viticultural technology developed in Uruguay for this variety during the last ten years, the chemical characterization of the polyphenolic and aroma profiles, typical for this variety, is also presented.
