A mixed Mg2+/Al3+ oxide featuring an incipient spinel phase was employed in the catalytic decomposition of a range of phosphate triesters, including the neurotoxic biocide chlorpyrifos methyl O-analog, with estimated catalytic effects up to 104-fold. The target reaction, namely, the transesterification with 1-propanol, was conveniently chosen as a means to convert toxic organophosphorus substrates into trialkyl phosphates structurally related to a family of flame retardants. Catalytic efficiency depends upon the stereo-electronic properties of the substrate, with both leaving group ability and geometric factors displaying pivotal effects on the rate constants. To evaluate the adsorption of 1-propanol/propoxide and the phosphotriester methyl paraoxon over the MgAl2O4(100) surface, density functional theory calculations with periodic boundary conditions were performed, which showed that the most probable reactants’ conformation prior to the reaction is such that propoxide is bound to a Mg2+ center with a juxtapositioned methyl paraoxon molecule at a close, neighboring Al3+ site.

Two fatty acid binding proteins, MvFABPa and MvFABPb were identified in the parasite Mesocestoides vogae (Platyhelmithes, Cestoda). Fatty acid binding proteins are small intracellular proteins whose members exhibit great diversity. Proteins of this family have been identified in many organisms, of which Platyhelminthes are among the most primitive. These proteins have particular relevance in flatworms since de novo synthesis of fatty acids is absent. Fatty acids should be captured from the media needing an efficient transport system to uptake and distribute these molecules. While HLBPs could be involved in the shuttle of fatty acids to the surrounding host tissues and convey them into the parasite, FABPs could be responsible for the intracellular trafficking. In an effort to understand the role of MvFABPs in fatty acid transport of M. vogae larvae, we analysed the intracellular localization of both MvFABPs and the co-localization with in vivo uptake of fatty acid analogue BODIPY FL C16. Immunohistochemical studies on larvae sections using specific antibodies, showed a diffuse cytoplasmic distribution of each protein with some expression in nuclei and mitochondria. MvFABPs distribution was confirmed by mass spectrometry identification from 2D-electrophoresis of larvae subcellular fractions. This work is the first report showing intracellular distribution of MvFABPs as well as the co-localization of these proteins with the BODIPY FL C16 incorporated from the media. Our results suggest that fatty acid binding proteins could target fatty acids to cellular compartments including nuclei. In this sense, M. vogae FABPs could participate in several cellular processes fulfilling most of the functions attributed to vertebrate’s counterparts.

Initiation of celiac disease is triggered in the gastrointestinal tract by transglutaminase 2 (TG2) assisted deamidation of gluten peptides. Deamidation is a side-reaction to transamidation and occurs if primary amines are absent. In contrast to deamidation, transamidation does not trigger an immune response. The aim of the study was to identify a suitable food additive that interacts with TG2 binding motives in gluten-derived peptides to prevent deamidation/transamidation. Homology modelling of α2-gliadin and computational screening of compounds for their binding affinity to a common TG2 binding motive (P)QLP were done by using computational approaches followed by experimental testing of TG2 activity. A database containing 1174 potential food grade ligands was screened against the model of α2-gliadin (27 out of 33 aa). Out of the five best ligands, ascorbyl palmitate, was observed to decrease TG2 transamidation of gliadin by 82% ± 2%. To completely silence the transamidation, we added zinc chloride (ZnCl2), and thereby reached a 99% ± 1% inhibition of TG2 activity. In addition, we conducted a pilot experiment in which ascorbyl palmitate was observed to decrease TG2 deamidation of gliadin completely. We propose ascorbyl palmitate in combination with ZnCl2 with the future perspective to become an additive in celiac-safe foods.

Abstract
A summary of the resource.
We have used Salmonella as a delivery system for eukaryotic expression plasmids encoding cytokines, and assessed its capacity to modulate immune responses in different experimental models. Plasmids encoding mouse IL-4 and IL-18 under cytomegalovirus promoter were constructed and transformed into live attenuated Salmonella enterica serovar Typhi strain CVD 908-htrA, and Salmonella enterica serovar Typhimurium strain SL3261. We have shown that systemic as well as mucosal immunization with such constructs can influence the antibody and cytokine responses to the Salmonella carrier and to co-administered bystander antigens, as well as the specific immune response elicited during a parasitic infection. Further, we have shown that oral cytokine-therapy using Salmonella as gene vector induce antitumoral effect as demonstrated by extended survival time in melanoma-bearing mice. This approach may be particularly suited for the development of new immunotherapies with applications in parasitic infections and cancer, were alterations of the host’s immune responses are usually found, and therapy-induced modulation of the immune response is likely to be required

Abstract
A summary of the resource.
A flow injection photochemical vapor generation atomic absorption spectrometry (FI-PVG-AAS) system was developed for selenium determination. The variables of influence of the photochemical generation process were evaluated and optimized. Sample already reduced to the Se(IV) state and mixed with the photochemical reagent (1.5% w/w HCOOH) was irradiated for 3 s. Selenium derivatives were transferred to an externally heated atomization cell by an argon-hydrogen mixture and selenium determined at 196.0 nm. Nitrogen was used as the carrier of the flow injection system. Nitrate is one of the most relevant interferents for the photochemical generation process; it is a very frequent concomitant as nitric acid is added for sample digestion or preservation. Hence, its removal before the vapor generation stage was studied. It was found that up to 3.0% w/w HNO3, it can be eliminated in the reduction stage of Se(VI) to Se(IV) with hydrochloric acid. For higher concentrations (up to 21% w/w HNO3), prior to the reduction step, the addition of 40% w/w H2CO and evaporation almost to dryness was successful for the abatement of the interference. The performance of the method was studied and its applicability evaluated with a Trace Element in Water Certified Reference Material (24.3 μg L−1 – Se, 2.8% w/w HNO3) and a spiked rice-flour microwave-assisted digest (103.1 μg L−1 – Se, 18.5% w/w HNO3).

<div class="paragraph">Propolis and grape pomace have significant amounts of phenols which can take part in anti-inflammatory mechanisms. As the cyclooxygenases 1 and 2 (COX-1 and COX-2) are involved in said mechanisms, the possibility for a selective inhibition of COX-2 was analyzed <em>in vitro</em> and <em>in silico</em>.</div>
<div class="paragraph">Propolis and grape pomace from Uruguayan species were collected, extracted in hydroalcoholic mixture and analyzed. Based on phenols previously identified, and taking as reference the crystallographic structures of COX-1 and COX-2 in complex with the commercial drug Celecoxib, a molecular docking procedure was devised to adjust 123 phenolic molecular models at the enzyme binding sites.</div>
<div class="paragraph">The most important results of this work are that the extracts have an overall inhibition activity very similar in COX-1 and COX-2, i.e. they do not possess selective inhibition activity for COX-2. Nevertheless, 10 compounds of the phenolic database turned out to be more selective and 94 phenols resulted with similar selectivity than Celecoxib, an outcome that accounts for the overall experimental inhibition measures.</div>
<div class="paragraph">Binding site environment observations showed increased polarity in COX-2 as compared with COX-1, suggesting that polarity is the key for selectivity. Accordingly, the screening of molecular contacts pointed to the residues: Arg106, Gln178, Leu338, Ser339, Tyr341, Tyr371, Arg499, Ala502, Val509 and Ser516, which would explain, at the atomic level, the anti-inflammatory effect of the phenolic compounds. Among them, Gln178 and Arg499 appear to be essential for the selective inhibition of COX-2.</div>

The mortality of 14–21-day-old Rhipicephalus (Boophilus) microplus larvae, and the mortality and fertility of groups of engorged adult females exposed to different concentrations of hexane, ethyl acetate and ethanol extracts of spiked pepper (Piper aduncum) were evaluated, using a completely randomized design with five treatment groups, two control groups, and two replicates for the larvae and five replicates for the adult females. Similar methodology was used to investigate the toxicity of the essential oil hydro-distillate (94.84% dillapiole) obtained from the P. aduncum crude hexane extract. The LC50 of the hexane extract was 9.30 mg ml�1 for larvae and the reproduction reduction ranged from 12.48% to 54.22%, while 0.1 mg/ml�1 of the essential oil induced 100% mortality in larvae. Literature reports on natural products active against R. microplus were listed and compared with the results presented here. These results indicate that P. aduncum extracts, and particularly its essential oil, are potential alternative control agents for R. microplus.

A summary of the resource.
The objective of the research was to test the hamster for a model of transmission of congenital toxoplasmosis. A non-invasive method for the diagnosis of pregnancy in hamsters was designed, with a specificity and a sensitivity of 70.2 and 94.7%, respectively (n = 168). Of 32 females with a chronic toxoplasma infection, 3 transmitted Toxoplasma congenitally during their first pregnancy, but not during the subsequent pregnancy. Congenital transmission rates of infections initiated during pregnancy with 2 stages of 2 strains of Toxoplasma were in the range of 33 to 100% of the 76 females inoculated. Only 1 of 17 females transmitted the parasite exclusively via milk. It was concluded that the hamster is a promising species for a model of transmission of congenital toxoplasmosis.

Eight of nine cats inoculated with strain ME-49 and challenged with three diVerent strains of Toxoplasma were immune to oocyst shedding, as ascertained with bioassays of their feces. In a second experiment, only toxoplasma asexual stages were seen in H&E stained gut sections of cats treated with suppressive doses of sulfamerazine and pyrimethamine starting 2 days after oral inoculation with cysts of the strain ME-49 and killed 6 days later. In a third experiment, four cats were similarly inoculated and treated for 20 days. Six weeks later, the cats received an oral homologous challenge with cysts, and none shed toxoplasma oocysts. An acceptable level of cross-protection was achieved with strain ME-49, and therefore, it can be used as a candidate strain from which antigens could be tested for enteric protection.

Abstract
A summary of the resource.
Rats were immunized with cysts of two Toxoplasma strains or with RH strain tachyzoites prior to pregnancy. The litters of the 13 rats that received homologous challenges with cysts during pregnancy, were all protected, whereas of 173 rats that received heterologous challenges with cysts or oocysts, only 21 protected their litters. 38.3 and 17% of rats immunized with the RH and with complete strains respectively, and 57% of control rats challenged with cysts, transmitted the infection congenitally. The percentages when similar groups were challenged with oocysts, were 33.3, 48.2, and 56.2%, respectively. Immunization with cysts did not completely protect against challenge with oocysts, even if the same strain was used. The divergence of these results from the complete protection against congenital toxoplasmosis observed in immune women and ewes, might be due to the use of excessive challenge doses in the model.<br /><br />

Abstract
A summary of the resource.
The objective was to test immune protection against the formation of Toxoplasma gondii tissue cysts in rats. It has been previously shown that 50 T. gondii tissue cysts of strain Me49 are not pathogenic for CF-1 mice, whereas 1 T. gondii tissue cyst of strain M-7741, can be lethal for mice 11–13 days after subcutaneous or oral administration. In the present study, ten rats were fed T. gondii oocysts of strain Me49 and after a further 30 days they were each orally challenged with T. gondii oocysts of strain M-7741. Thirty days after this, they were euthanased and brain and muscle samples inoculated subcutaneously or orally dosed, respectively, to mice for bioassay. None of the mice died, whereas all the mice that were inoculated with brain homogenates or were fed muscle samples from four non-immunized rats that had been inoculated with T. gondii oocysts of strain M-7741, died. These results encourage further research towards achieving vaccinal protection against the formation of T. gondii tissue cysts in meat animals and people.

A summary of the resource.
<p>Nine lambs were immunized with 106 oocysts of the relatively avirulent ME-49 strain of Toxoplasma gondii, and were challenged 45 days later with 4 � 106 oocysts of the M3 strain of this parasite. Less than 5 tissue cysts of the M3 strain formed per 8 g of muscle – if any – judging from a bioassay in mice in which all mice survived. In contrast, mortality occurred in mice fed muscle specimens from 4 lambs that had not been immunized with ME-49 prior to challenge with the M3 strain. This finding suggests the possibility of creating a vaccine for lambs that could diminish or abolish lamb and mutton as sources of human infection with T. gondii, and which could also prevent toxoplasmosis abortion in sheep. �</p>

A summary of the resource.
A multiresidue method was developed for the quantification and confirmation of 70 pesticides in paddy field water. After its filtration, water was injected directly in a liquid chromatograph coupled to a hybrid triple quadrupole-linear ion trap-mass spectrometer (QqLIT). The list of target analytes included organophosphates, phenylureas, sulfonylureas, carbamates, conazoles, imidazolinones and others compounds widely used in different countries where rice is cropped. Detection and quantification limits achieved were in the range from 0.4 to 80 ng L−1 and from 2 to 150 ng L−1, respectively. Correlation coefficients for the calibration curves in the range 0.1–50 �g L−1 were higher than 0.99 except for diazinon (0.1–25 �g L−1). Only 9 pesticides presented more than 20% of signal suppression/enhancement, no matrix effect was observed in the studied conditions for the rest of the target pesticides. The method developed was used to investigate the occurrence of pesticides in 59 water samples collected in paddy fields located in Spain and Uruguay. The study shows the presence of bensulfuron methyl, tricyclazole, carbendazim, imidacloprid, tebuconazole and quinclorac in a concentration range from 0.08 to7.20 �g L−1.
