Lung cancer remains the leading cause of cancer mortality worldwide. Thus, the development of strategies against this type of cancer is of high value. Parasite infections can correlate with lower cancer incidence in humans and their use as vaccines has been recently explored in preclinical models. In this study, we investigated whether immunisations with a Trypanosoma cruzi lysate from epimastigotes protect from lung tumour growth in mice. We also explore the role of parasite glycans in the induction of the protective immune response. A pre-clinical murine cancer model using the lung tumour cell line LL/2 was used to evaluate the anti-tumour potential, both in preventive and therapeutic settings, of a T. cruzi epimastigote-derived protein lysate. Immunisation with the parasite lysate prevents tumour growth and induces both humoral and cellular anti-tumour immune responses to LL-2 cancer cells. The induced immunity and tumour protection were associated with the activation of natural killer (NK) cells, the production of interferon- (IFN- ) and tumour cell cytotoxicity. We also show that mannose residues in the T. cruzi lysate induce Toll-like receptor (TLR) signalling. The evaluated T. cruzi lysate possesses anti-tumour properties likely by activating innate and adaptive immunity in a process where carbohydrates seem to be essential.

Background & Objective: Chagas disease or American trypanosomiasis is a major parasitic disease in Latin America with restricted available treatment: nifurtimox and benznidazole. These two drugs are ineffective in the chronic phase of the disease; therefore, there is a need for the development of new, efficient and safe drugs for the treatment of this pathology. With this goal, one of the promising targets is trypanothione reductase (TR), a key enzyme in the metabolism of Trypanosoma cruzi. Conclusion: In this review, we analyse the importance of TR as a drug target, as well as the well-known and new inhibitors reported in the last decade as potential therapeutic agents for Chagas disease.

<p>In this work, we perform first-principles density functional theory calculations to study siligraphene (SiC3)/ hexagonal boron nitride (hBN) vdW-heterostructures. Siligraphene has comparable electronic properties to graphene, including the presence of a Dirac-like cone, thus we investigate its potential as a replacement for graphene. We find that, when internal strains induced by the high lattice mismatch between the SiC3 and hBN layers are reduced, a very small and non-tunable gap is induced in a hybrid bilayer heterostructure. However, the<br />picture becomes quite different when structures with higher internal strains are considered. In this scenario, the<br />compressive strain induces a buckling in the SiC3 sheet, resulting in larger band gaps. Additionally, in many configurations, the gaps are found to be quite tunable by the application of external strains or an electrical field. Such tunability is absent in graphene/hBN heterostructures, thus giving SiC3/hBN heterostructures a clear edge. The origin of this gap and its tunability are discussed in terms of the buckling in the SiC3 layer and the interlayer charge transfer. Finally, the optical properties of these heterostructures were also investigated. Strong absorption peaks in both visible and deep UV regions were found, which could be explored for applications in future optoelectronic devices.</p>

Abstract

Heteronuclear zinc(II):mixed-lanthanide(III) metal organic frameworks (MOFs) bearing the formula [LnAx LnBy Zn3(oda)6(H2O)6]·nH2O (Ln = lanthanide(III) ion, LnB is smaller than LnA, x+y=2, n= 13 or 14) were obtained by direct reaction of the corresponding metal ion salts and oxydiacetate ( OOC-CH2-O-CH2-COO ) in aqueous solution, followed by slow solvent diffusion. This strategy led to the isolation of thirteen pure crystalline samples of isostructural hexagonal compounds (space group P6/mcc). Characterization by single-crystal X-ray diffraction was complemented by the analytical determination of the final LnB=LnA molar ratio by microwave induced plasma atomic emission spectrometry. Results show that the experimentally determined LnB=LnA molar ratio is systematically higher than that set out in the synthesis, especially for combinations of Ln that have larger size differences. Dual emission luminescence is observed for the compounds containing a combination of the more efficient visible-emitting Ln (Sm, Eu, Tb, Dy). The selected aliphatic ligand does not promote antenna effect. Thus, tunable color luminescence can be achieved by selectively targeting each Ln preferential excitation wavelength. Dual emission is also observed for CeEu mixed-MOF since the simultaneous excitation of both Ln is possible. Interestingly, GdEu mixed-MOF also shows dual emission when only Gd is directly excited.

Herein, we perform DFT calculations to study the stability and the electronic and optical properties of pristine and hybrid bilayers of hexagonal boron phosphide (h-BP) and hexagonal boron arsenide (h-BAs). The electronic and optical properties of all bilayers can be modulated by the introduction of an external perpendicular electric field. Consequently, band gaps can be tuned from 0.8 eV down to zero as the field increases up to a critical value. Above this value, the gap reopens as the valence and conduction bands exhibit an anticrossing with a “Mexican-hat” shape, in a similar fashion to biased graphene bilayers. For the optical properties, we report an intense peak in the absorption spectra around 2.5–2.6 eV, with a slight blueshift as a function of electric field. Above the critical field, a new peak is observed in the infrared region which exhibits a strong field dependence. Additionally, this peak is related to the optical transitions around the “Mexican-hat” region of the band structure and the corresponding electron-hole pair shows a layer separation, which may lead to larger recombination times, a critical factor in photovoltaics. We believe that, with such tunable properties, these bilayers can find interesting applications in future devices in nanoelectronics and optoelectronics.

Disulfide-bond formation is a major post-translational modification and is essential for protein folding, stability, and function. This is especially true for secreted proteins, many of which possess great potential for biotechnological applications. Focusing on the use of Escherichia coli for the production of this class of proteins, we describe the mechanisms that maintain redox compartmentalization in the cell, with an emphasis on those that promote the formation and isomerization of disulfide bonds in the bacterial periplasm, while presenting parallel pathways in the eukaryotic endoplasmic reticulum. Based on these concepts, we review the use of E. coli as a cell factory for the production of heterologous disulfide-containing proteins using either peri- or cytoplasmic expression and, in particular, how these compartments can be tuned to improve the yield of correctly folded recombinant proteins. Finally, we describe a few examples of the production of small disulfide- rich proteins (protease inhibitors) to illustrate how soluble, active, and fully oxidized recombinants may be successfully obtained upon peri- or cytoplasmic expression in E. coli.

In this report, we study the tuning of the electrical transport dimensionality of polyaniline–graphene oxide quantum dots nanocomposites (PANI-GOQD) for electronic device applications. We focused this study on the microstructure and its correlations with electrical transport properties. X-ray diffraction and small-angle X-ray scattering analyses showed the effect that caused the addition of GOQD on the structural and microstructural properties of polyaniline. Confocal Raman spectroscopy revealed that the presence of GOQD leads to a notorious decrease of the polaron population of polyaniline. In relation to this experimental evidence, a significant increase in resistivity was observed for PANI-GOQD nanocomposites with respect to pure polyaniline. Electrical transport showed a typical Arrhenius behavior at relatively high temperatures and a broad transition with a logarithmic dependence of the activation energy with temperature for the intermediate temperature regime. Additionally, PANI-GOQD showed an increase in the hopping transport dimensionality even in the case of low amounts of GOQD. The tuning of this dimensionality in these nanocomposites could be important for the development of novel organic electronic materials.

Bilayer graphene simultaneously doped with two different second-row atoms (aluminum and phosphorus) was investigated employing first-principle density functional calculations (LDA, VDW-DF and M06-L). In agreement with the results obtained for bilayer graphene doped with one type of heteroatom, we found that at 1.56% atomic doping of Al and 1.56% atomic doping of P, the structure with sheets not linked is more stable than that with an Al–P bond. While all methods agreed in predicting the most stable structure, the relative energies of the structural isomers were different at the three levels of theory considered. The electronic properties of doped bilayer graphene showed an unusual variation upon small conformational changes. In effect, metallic, semiconducting and semimetallic properties were observed upon variation of the protruding orientation of the heteroatoms. The electronic properties of bilayer graphene seem to be determined by the layer whose heteroatom protrudes into the interlayer region. When Al protrudes in and P out metallic properties are observed. However, when P protrudes in and Al in a small band gap semiconductor is obtained. This work shows that the electronic properties of bilayer graphene can be more easily tuned than those of monolayer graphene because a small conformational change can induce a metallic to semiconductor transition or vice versa.

We wish to report the isolation and identification of two new diterpenoids from the brown alga Glossophora kuntii(C.Agardh)J. Agardh(Dictyotaceae). Along with dictyotriol [5] and dictyotriol A diacetate[6] |b our compounds are the only examples of trihydroxylated perhydroazulene derivatives.

The structures of two new sulfate complexes are reported, namely di-μ-sulfato-κ3O,O′:O′′-bis{aqua­[2,4,6-tris(2-pyridyl)-1,3,5-triazine-κ3N1,N2,N6]­cadmium(II)} tetra­hydrate, [Cd2(SO4)2(C16H12N6)2(H2O)2]·4H2O, and di-μ-sulfato-κ2O:O′-bis­[(2,2′:6′,2′′-ter­pyridine-κ3N1,N1′,N1′′)­zinc(II)] dihydrate, [Cd2(SO4)2(C15H11N3)2]·2H2O, the former being the first report of a Cd(tpt) complex [tpt is 2,4,6-tris(2-pyridyl)-1,3,5-triazine]. Both compounds crystallize in the space group Pinline image and form centrosymmetric dimeric structures. In the cadmium complex, the metal center is heptacoordinated in the form of a pentagonal bipyramid, while in the zinc complex, the metal ion is in a fivefold environment, the coordination geometry being intermediate between square pyramidal and trigonal bipyramidal. Packing of the dimers leads to the formation of planar structures strongly linked by hydrogen bonding.

Abstract
A summary of the resource.
The cyclization of Schiff bases derived from phenethylamines to 1-methyl-, 1-phenyl- and 1-benzyl-tetrahydroisoquinolines has been examined. It was confirmed that a free OH group para or ortho to the place of ring closure is of fundamental importance for the possibility of cyclization under neutral or weekly acid conditions. The cyclization reaction with Schiff bases of phenylacetaldehydes synthesized via Darzens reaction is very easy. The IR, UV and NMR spectra of the Schiff bases and tetrahydroisoquinolines have been compared and from this methods deduced for following up the ring closure reaction.<br /><br />

Nitrogen cycle has been poorly investigated in Antarctic ecosystems. In particular, how extreme conditions of low temperature, dryness, and high radiation select the microorganisms involved in the cycle is not yet understood. Denitrification is an important step in the nitrogen cycle in which nitrate is reduced stepwise to the gases NO, N2O, and N2. Denitrification is carried out by a wide group of microorganisms spread in the phylogenetic tree. The aim of this work was to isolate and characterize denitrifying bacteria present in different cold environments from Antarctica. Bacterial isolates were obtained from lake, meltwater, sea, glacier ice, ornithogenic soil, and penguin feces samples from King George Island, Fildes peninsula in the Antarctic. Samples were taken during the deicing season in five sampling campaigns. From all the samples we were able to isolate denitrifying strains. A total of 199 bacterial isolates with the capacity to grow in anaerobic mineral media reducing nitrate at 4°C were obtained. The characterization of the isolates by 16S rRNA gene sequence analysis showed a high predominance of the genus Pseudomonas, followed by Janthinobacterium, Flavobacterium, Psychrobacter, and Yersinia. Other minor genera detected were Cryobacterium, Iodobacter, Kaistella, and Carnobacterium. The capacity to denitrify was not previously described for most of the bacteria related to our isolates and in many of them denitrifying genes were not present suggesting the presence of new genes in this extreme environment. Our work demonstrates the ubiquity of denitrification in the Maritime Antarctica and gives important information linking denitrification at cold temperature with taxa in an unequivocal way.

Immunosensors for small analytes have been a great addition to the analytical toolbox due to their high sensitivity and extended analytical range. In these systems the analyte is detected when it competes for binding to the detecting antibody with a tracer compound. In this work we introduce the use of phage particles bearing peptides that mimic the target analyte as surrogates for conventional tracers. As a proof of concept, we developed a magneto-electrochemical immunosensor (EI) for the herbicide molinate and compare its performance with conventional formats. Using the same anti-molinate antibody and phage particles bearing a molinate peptidomimetic, the EI performed with an IC50 of 0.15 ng mL−1 (linear range from 4.4 × 10−3 to 10 ng mL−1). Compared to the conventional ELISA, the EI was faster (minutes), performed with a much wider linear range, and the detection limit that was 2500-fold lower. The EI produced consistent measurements and could be successfully used to assay river water samples with excellent recoveries. By using the same EI with a conventional tracer, we found that an important contribution to the gain in sensitivity is due to the filamentous structure of the phage (9 × 1000 nm) which works as a multienzymatic tracer, amplifying the competitive reaction. Since phage-borne peptidomimetics can be selected from phage display libraries in a straightforward systematic manner and their production is simple and inexpensive, they can contribute to facilitate the development of ultrasensitive biosensors.

Porphyrin and perylene diimide have great application potential in organic optoelectronic devices. However, large plane frames of porphyrin and PDI tend to aggregate, resulting in low power conversion efficiency. In this work, four side chain modified porphyrins (H2TCPP, ZnTCPP, H2TCPP(CH3)4, ZnTCPP(CH3)4), and one perylene diimide (PDI-C5) were synthesized. The blended films of the donor and acceptor were prepared by a simple and low-cost liquid deposition method. Femtosecond transient absorption spectroscopy and global analysis revealed that ZnTCPP(CH3)4:PDI-C5 showed a higher rate of charge transfer and radical ions formation compared to the other porphyrin donors, which means that the ZnTCPP(CH3)4 has good compatibility with PDIC5. In addition, we also characterized these blended films by ultraviolet–visible absorption spectroscopy, cyclic voltammetry (CV), and AFM techniques. The CV results show that the difference between the LUMO of each porphyrin donor and PDI-C5 acceptor is about 0.3 eV, which is sufficient for exciton dissociation.

This work studied ultrasound assisted extraction (UAE) as a green chemistry technique to improve aqueous extraction of bioactive compounds from marcela (Achyrocline satureioides Lam.), a native plant from South America with reported beneficial health effects. Dried intact marcela inflorescences were extracted using solely water as solvent in an ultrasonic bath, applying 100 and 150 W/L ultrasound power at 40 and 60 °C, respectively. Extraction was followed throughout 30 min by determining antioxidant capacity (AOC) and total polyphenol content (TPC) every 5 min. Extracts were analysed by LC-MS and LC-DAD to identify extracted bioactive compounds and analyse the impact of temperature and ultrasound application on compound profile. Solidliquid extraction at 40 and 60 °C and maceration at room temperature were studied as reference extraction methods. UAE extracts were higher in AOC by 140% and 25% at 40 and 60 °C respectively, compared to solid-liquid extraction. For TPC, UAE showed an increase of 110% and 20% at 40 and 60 °C, respectively. LC results indicated that ultrasound application allowed faster and enhanced hydroxycinnamic acids extraction, whereas an increase in temperature resulted in a more diverse compound profile. UAE could be used to extract caffeic acid when applied at 40 °C.

Tannat red wine residues present a unique polyphenolic composition. Revalorization ofthis residue depends on adequate extraction technologies to assure process sustainability.Influence of temperature (15−60◦C), ultrasound (US) power (0−100 W) and US applicationtime (5−50 min) on Ultrasound Assisted Extraction (UAE) of fresh and freeze-dried Tan-nat grape pomace was evaluated. UAE was performed in a US bath, following extractionkinetics throughout 50 min by assessing: total polyphenol content (TPC), total monomericanthocyanins (TMAC) and total antioxidant capacity (AOC). Additionally, HPLC analysiswas performed to determine extracted compounds’ profile. Results showed UAE enhancedextraction in both fresh and freeze-dried pomace. In fresh pomace, UAE yielded extracts50% richer in TPC and TMAC on average when compared to conventional extractions, whilein freeze-dried pomace 180% increase in extraction was achieved. In fresh and freeze-driedpomace, US application allowed extraction of different compounds with respect to con-ventional extraction. This work shows that UAE has potential for Tannat grape pomacevalorization, yet UAE conditions should be carefully selected to favour desired extract com-position.

This study aimed to evaluate the waste cooking oil (WCO) hydrolysis in ultrasonic system using lipase as catalyst. Lipase was produced by the fungus Aspergillus niger via solid state fermentation (SSF) using canola meal as substrate. Prior to the hydrolysis reaction, the lipase behavior when subjected to ultrasound was evaluated by varying the temperature of the ultrasonic bath, the exposure time and the equipment power. Having optimized the treatment on ultrasound, the WCO hydrolysis reaction was carried out by evaluating the oil:water ratio and the lipase concentration. For a greater homogenization of the reaction medium, a mechanical stirrer at 170 rpm was used. All steps were analyzed by experimental design technique. The lipase treatment in ultrasound generated an increase of about 320% in its hydrolytic activity using 50% of ultrasonic power for 25 min. at 45 °C. The results of the experimental design conducted for ultrasound-assisted hydrolysis showed that the best condition was using an oil:water ratio of 1:3 (v:v) and enzyme concentration of 15% (v/v), generating 62.67 μmol/mL of free fatty acids (FFA) in 12 h of reaction. Thus, the use of Aspergillus niger lipase as a catalyst for hydrolysis reaction of WCO can be considered as a possible pretreatment technique of the oil in order to accelerate its degradation.

<p>Cystic echinococcosis is a globally distributed zoonosis caused by cestodes of the Echinococcus granulosus sensu lato (s.l.) complex, with Echinococcus ortleppi mainly involved in cattle infection. Protoscoleces show high developmental plasticity, being able to differentiate into either adult worms or metacestodes within definitive or intermediate hosts, respectively. Their outermost cellular layer is called the tegument, which is important in determining the infection outcome through its immunomodulating activities. Herein, we<br />report an in-depth characterization of the tegument of E. ortleppi protoscoleces performed through a combination of scanning and transmission electron microscopy techniques. Using electron tomography, a three-dimensional reconstruction of the tegumental cellular territories was obtained, revealing a novel structure termed the ‘tegumental vesicular body’ (TVB). Vesicle-like structures, possibly involved in endocytic/exocytic routes, were found within the TVB as well as in the parasite glycocalyx, distal cytoplasm and close inner structures. Furthermore, parasite antigens (GST-1 and AgB) were unevenly localised within tegumental structures, with both being detected in vesicles found within the TBV. Finally, the presence of host (bovine) IgG was also assessed, suggesting a possible endocytic route in protoscoleces. Our data forms the basis for a better understanding of E. ortleppi and E. granulosus s.l. structural biology.</p>

B polymorph of titanium dioxide (TiO2(B)) appears as a metastable phase during thermal annealing of low content sodium layered titanate nanostructures obtained via the widely used hydrothermal synthesis. In addition, the computed formation energy for the TiO2(B) (0 0 1) slabs in the order of the one calculated for anatase (1 0 1) which represents one of the most stable TiO2 surfaces. This encourages the study of this polymorph which could gain prominence in TiO2 applications at the nanoscale. In a first instance ultrathin TiO2(B) sheets, parallel to crystallographic planes (0 0 1) and (1 0 0), are investigated and compared to other well know TiO2 polymorphs, such as rutile and anatase surfaces. Then the adsorption of formic acid on (0 0 1) and (1 0 0) – less stable – TiO2(B) dry ultrathin sheets was studied as the first step for further evaluation of TiO2(B) nanostructures for dye sensitized solar cells (DSSC) applications. The results show that the monodentate through the carbonyl group configuration is the most stable for (0 0 1) sheet while bidentate dissociated configuration is the most stable for (1 0 0) sheet being the computed adsorption energies 0.51 eV and 1.49 eV, respectively. The evaluated reactivity of TiO2(B) slabs is comparable with anatase and indicates that it could be a good adsorbent for common dyes used for dye sensitized solar cells purposes.

Esta publicación tiene por objeto divulgar un procedimiento de igualación de ecuaciones químicas que he venido desarrollando en mis clases como Profesor Agregado de Análisis Químico Cualitativo desde el año 1939. El método fue expuesto en una conferencia pronunciada en la Facultad de Química y Farmacia de Montevideo y presentado a las Jornadas Químicas Rioplatenses, celebradas en dicha ciudad en diciembre de 1940.
