La hipercolesterolemia familiar (HF) es una enfermedad hereditaria, relativamente frecuente y asociada al desarrollo de ateroesclerosis y enfermedad coronaria prematuras. En este trabajo, describimos el hallazgo de una nueva mutación (–47C>A) en la región promotora del gen del receptor de las lipoproteínas de baja densidad (rLDL), principal encargado de este fenotipo. Esta mutación se identificó en una familia uruguaya con un fenotipo típico de HF, en la cual aparecen individuos heterocigotos y homocigotos para la mutación debido a la existencia de consanguinidad. El cambio nucleotídico ocurre en un sitio conservado y funcionalmente relevante del promotor; región en la que anteriormente se han descrito diversas mutaciones que provocan descensos drásticos en la actividad transcripcional del gen. No se han detectado otras variantes en las regiones analizadas del gen. Los análisis genéticos del rLDL asociados a otros genes de susceptibilidad permiten establecer un perfil genómico de riesgo cardiovascular, de aplicación muy necesaria en el tratamiento clínico de familias con HF. En este trabajo se describe el uso de una reacción multicomponente, la reacción de Passerini, como un experimento de laboratorio en un curso de grado de Química Orgánica. Es un proceso rápido, eficiente y simple que permite obtener productos complejos a partir de moléculas sencillas y en un paso de reacción. Diferentes parámetros fueron analizados para determinar cuán amigable es esta reacción con el medio ambiente, en las condiciones ensayadas. El estudiante tendrá la posibilidad de discutir sobre química verde, desarrollo de moléculas bioactivas, síntesis orientada a la diversidad, reacciones multicomponente y química en disolución acuosa. Chlorpyrifos is one of the most employed insecticides worldwide. It is moderately persistent in the environment. It degrades to 3, 5, 6- trichloro-2-pyridinol (TCP), the metabolite of toxicological concern to both human health and the environment. In the field, pesticides point contaminations occur during the operation of the application machinery. Biobeds made up of a biomixture containing straw, peat, and soil have been successfully developed to minimise these incidents. The retained pesticides are degraded by the microorganisms in the biomixture. Although the dissipation of chlorpyrifos in different biobeds has been studied, the fate of TCP received scarce attention. Biodegradation in specific bioreactors can only be assessed if it is evaluated through proper analytical methods. In this work chlorpyrifos and TCP dissipation in biobeds was studied with a previously fit for purpose analytical method. The lyophilised biomixture was extracted with ethyl acetate in an aqueous solution of NaCl and sodium tetraborate. The extract was cleaned up with alumina. Chlorpyrifos was determined by gas chromatography with electron capture and mass spectrometer detectors and TCP by liquid chromatography coupled to tandem mass spectrometry, using the negative Multiple Ion Mode. Recoveries of the two analytes, up to 2 mg kg−1, were between 70% and 113% with RSDs below 16% and RSDwR below 19%. Further TCP confirmation was evaluated through Enhanced Product Ion mode for each ion. The dissipation of both analytes in laboratory biobeds inoculated with Abortiporus biennis using two different types of Uruguayan soils was assayed. Both bioreactors were able to dissipate the compounds after 60 days following different dissipation kinetics. The larval stage of the cestode parasite Echinococcus granulosus causes hydatid disease in humans and livestock. This infection is characterized by the growth in internal organ parenchymae of fluid-filled structures (hydatids) that elicit surprisingly little inflammation in spite of their massive size and persistence. Hydatids are protected by a millimeter-thick layer of mucin-based extracellular matrix, termed the laminated layer (LL), which is thought to be a major factor determining the host response to the infection. Host cells can interact both with the LL surface and with materials that are shed from it to allow parasite growth. In this work, we analyzed the response of dendritic cells (DCs) to microscopic pieces of the native mucin-based gel of the LL (pLL). In vitro, this material induced an unusual activation state characterized by upregulation of CD86 without concomitant upregulation of CD40 or secretion of cytokines (interleukin 12 [IL-12], IL-10, tumor necrosis factor alpha [TNF-�], and IL-6). When added to Toll-like receptor (TLR) agonists, pLL-potentiated CD86 upregulation and IL-10 secretion while inhibiting CD40 upregulation and IL-12 secretion. In vivo, pLL also caused upregulation of CD86 and inhibited CD40 upregulation in DCs. Contrary to expectations, oxidation of the mucin glycans in pLL with periodate did not abrogate the effects on cells. Reduction of disulfide bonds, which are known to be important for LL structure, strongly diminished the impact of pLL on DCs without altering the particulate nature of the material. In summary, DCs respond to the LL mucin meshwork with a “semimature” activation phenotype, both in vitro and in vivo. The goal of our study was to improve Tannat grape and wine composition by achieving vine balance in high-capacity conditions. The current Uruguayan grape-growing paradigm accepts unpredictable periods of water deficit or excess in non-irrigated vineyards, only applying herbicides to eliminate weed competition (H). We used an under-trellis cover crop (UTCC) to limit vine water availability and to reduce vine growth rate and final canopy size. However, to avoid excessive vine water stress due to UTCC competition, irrigation was used as needed during water deficit periods. The experiment was conducted over three consecutive growing seasons from 2011 to 2013, in two experimental vineyards located in southern Uruguay (34° S 56° W). Treatments were: UTCC (full cover of the vineyard soil with tall fescue) versus conventional alleyway tall fescue with a 1.0 m-wide weed-free strip under the trellis. Deficit drip irrigation was provided at mid-day stem water potential (SWP) thresholds of −0.9 MPa early in the season and −1.1 MPa later. Shoot growth rate, SWP, berry size and berry composition (Brix, organic acids, total anthocyanins) were monitored over the season as well as final yield, cluster and pruning weights and wine sensory attributes. UTCC regulated vine vegetative growth and final canopy size and reduced bunch rot incidence as well as increasing fruit Brix and anthocyanin concentration in grapes and wines. Wines from UTCC treatments had an increased fruity aroma and overall aroma intensity levels and had distinctive sensory characteristics that exceeded those of H wines during overall palatability tests (liking tests). A total of 1025 Brazilian consumers completed word association task with lamb meat. The elicited words were analyzed using inductive coding. Participants' associations with lamb meat were mainly related to sensory characteristics and hedonic attitudes and feelings, indicating that they might be the mainmotivations for consuming this product. Participants strongly associated lamb meat with special consumption occasions, which suggests that lack of perceived appropriateness for everyday consumption situations might be a barrier for increasing lamb meat consumption. Conceptualization of lambmeat was strongly affected by frequency of consumption of this product. Consumers’ perceptions of conventional (regular plain yogurt, low-calorie and low-fat yogurt) and functional yogurts (enriched with fibre and antioxidants) were studied using word association and hard laddering. Both methodologies provided similar results, being consumers’ perceptions of these products mainly related to health, nutrition, sensory characteristics and pleasure. However, both methodologies provided different information. Word association provided information about attributes that were relevant for consumers’ acceptance of a product, common defects of products in the marketplace, motives behind consumers’ choice and attributes that might limit consumers’ interest in the product. On the other hand, hard laddering allowed the identification of more consequences and values related to product attributes, and the links between them, which could be useful for understanding consumers’ choices. Besides, word association was more sensitive than hard laddering in determining differences between groups of consumers with different attitudes towards health and nutrition. Results suggested that word association might be an interesting and useful qualitative technique for product development. Although fruits and vegetables are an essential component of a healthy diet from sustainable food systems, their consumption remains below recommended levels worldwide. In this context, the aim of the present work was to explore factors influencing fruit and vegetable consumption among Uruguayan adults. A telephone survey with 505 adults, living in Montevideo and involved in food purchasing for the household, was conducted by a specialized company in May 2024. Participants were asked to indicate the number of days they consumed fruit in a typical week and the number of servings they ate in each of those days. They were also asked to indicate whether they regarded their fruit and vegetable consumption as sufficient or not. Those answering negatively were asked to indicate the reasons underlying their insufficient consumption. Participants also answered a series of socio-demographic questions. Results showed that only 12.4% of the participants consumed at least 5 servings of fruit and vegetables per day. Gender and socio-economic status were identified as the main socio-demographic predictors of fruit and vegetable consumption. Fruit and vegetable intake was frequently perceived as sufficient, even if it was below national recommendations. A wide range of factors emerged as motives underlying insufficient consumption of fruits and vegetables, including lack of habits, cost, dislike, and perceived difficulty for preparing or cooking vegetables. These results stress the need to implement multicomponent strategies to address deficiencies in fruit and vegetable consumption in the country. The laminated layer (LL) is the massive carbohydrate-rich structure that protects Echinococcus larvae, which cause cystic echinococcosis (hydatid disease) and alveolar echinococcosis. Increased understanding of the biochemistry of the LL is allowing a more informed analysis of its immunology. The LL not only protects the parasite against host attack but also shapes the overall immune response against it. Because of its dense glycosylation, it probably contains few T-cell epitopes, being important instead in T-cell independent antibody responses. Crucially, it is decoded in non-inflammatory fashion by innate immunity, surely contributing to the strong immune-regulation observed in Echinococcus infections. Defining the active LL molecular motifs and corresponding host innate receptors is a feasible and promising goal in the field of helminth-derived immune-regulatory molecules. Durante este proyecto de tesis se ha desarrollado un novedoso soporte bifuncional para la inmovilizaci6n de proteinas, que permite combinar las ventajas de la uni6n sitio especifica con las de la inmovilizaci6n covalente multipuntual. EI mismo contiene: i) una pequena proporci6n de gropos tiol-reactivos, y ii) un alto numero de gropos ep6xidos. La optimizaci6n de su sintesis se llev6 a cabo utilizando ditiotreitol 0 sulfuro s6dico como agentes tiolantes, y resinas acrilicas (Eupergit C, EP-Sepabeads) como matrices epoxiactivadas. La inmovilizaci6n en este tipo de soportes es un proceso en dos etapas. Primero la proteina es inmovilizada en forma reversible a traves de un intercambio tiol-disulfuro entre los gropos tiol de la enzima y los gropos tiol-reactivos del soporte bajo condiciones suaves de temperatura (24°C) Y pH (7.0). Luego, en una segunda etapa los gropos ep6xido remanentes del soporte son capaces de formar uniones' irreversibles intramoleculares con los gropos nucleofilicos superficiales de la enzima (principalmente residuos de lisina), debido a su proximidad. Este mecanismo en dos etapas fue verificado por medio del estudio del proceso de inmovilizaci6n de proteinas modelo: i) que presentan naturalmente gropos tiol superficiales (~-galactosidasa de Escherichia coli), y ii) cuyos gropos tiol necesitan ser generados por reducci6n de disulfuros nativos (~-galactosidasa de Kluyveromices lactis) 0 par tiolaci6n con SPDP (penicilina G acilasa de E. coli y lipasa de Rhizomucor miehei). A su vez, estudios de inactivaci6n termica demostraron que las uniones adicionales multipuntuales que se establecen durante la segunda etapa de la inmovilizaci6n, son las responsables de un incremento en la estabilidad de las enzimas inmovilizadas con respecto alas enzimas solubles 0 alas inmovilizadas iInicamente a traves de enlaces disulfuros. Dado que la presencia de gropos sulfhidrilo en la superficie de la estructura proteica es una condici6n esencial para el uso de este nuevo soporte mixto, en el desarrollo de esta tesis se estudiaaran en forma complementaria dos estrategias para la generaci6n de gropos tiol: i) por reducci6n de puentes disulfuros, y ii) por mutagenesis dirigida. Es asi que por un lado, se evalu6 el uso de tiopraapil-agarosa, para la obtenci6n de gropos sulfhidrilo por reducci6n de disulfuros proteicos, como una altemativa al uso de reductores solubles. A su vez, con el fin de poder controlar totalmente la orientaci6n proteica sobre los soportes desarrollados, se estudi6 la viabilidad de introducir residuos de cisteina en la PGA de E. coli por medio de mutagenesis dirigida. Si bien los resultados obtenidos son muy preliminares, 10s mismos alientan a la realizaci6n de investigaciones que permitan estudiar el efecto que podria tener en la estabilidad enzimMica, el inmovilizar la PGA por distintas zonas de su superficie en los soportes desarrollados. The ubiquitous intracellular molecule myo-inositol hexakisphosphate (IP6) is present extracellularly in the hydatid cyst wall (HCW) of the parasitic cestode Echinococcus granulosus. This study shows that extracellular IP6 is present as its solid calcium salt, in the form of deposits that are observed, at the ultrastructural level, as naturally electron dense granules some tens of nanometers in diameter. The presence of a calcium salt of IP6 in these structures was determined by two different electron microscopy techniques: (i) the analysis of the spatial distribution of phosphorus and calcium in the outer, acellular layer of the HCW (the laminated layer, LL) through electron energy loss spectroscopy, and (ii) the observation, by transmission electron microscopy, of HCW that were selectively depleted of IP6 by treatment with EGTA or phytase, an enzyme that catalyses the dephosphorylation of IP6. Las mandarinas presentan una gran diversidad en características sensoriales y de calidad de fruta, así como una cosecha extendida. El mejoramiento genético de cítricos se focaliza en la búsqueda de nichos de cosecha para la obtención de mayores precios, así como en aspectos nutricionales y sensoriales, entre otras caracterís-ticas. En este contexto, la caracterización sensorial de nuevos híbridos y la identificación de las características sensoriales claves que influyen en la aceptabilidad de los consumidores proveen de una valiosa información para el mejoramiento y las estrategias de marketing. Estudios previos mostraron que las características senso-riales, especialmente el sabor, son claves en la aceptación de las mandarinas por los consumidores. Se llevaron a cabo dos estudios durante la estación de cosecha de mandarinas (media y tardía) aplicando la metodología «marque todo lo que corresponda» (CATA) y con la participación de al menos 100 consumidores. La aceptabi-lidad de los cultivares que fueron más aceptados se situó entre 6,5 y 7,7 en una escala hedónica de 9 puntos, evidenciando una reacción hedónica positiva. Los nuevos híbridos presentaron puntajes de aceptabilidad simi-lares o superiores a los cultivares de referencia Tango, Gold Nugget, Murcott y Ortanique. A su vez, los resul-tados mostraron que el color naranja, la forma regular, la piel lisa, el dulzor, la jugosidad, el sabor intenso y el sabor típico a mandarina son características claves que determinan la aceptabilidad de las mandarinas, y que la apariencia externa no es un buen predictor del sabor.
