A summary of the resource.
Activated carbon monoliths and oxidized carbon monoliths are prepared from Eucalyptus grandis wood by pyrolysis and subsequent activation under CO2 (aCMs) and further functionalization using nitric acid (ox-aCMs). Their textural and chemical properties are compared and the electrochemical characteristics as supercapacitor electrodes are tested. The nitric acid treatment does not produce substantial changes in the textural properties of the monolith obtained. Elemental analysis and X-ray photoelectron spectroscopy (XPS) reveal that the oxidized monolith presents a high content of oxygenated functional groups that can participate in pseudocapacitive reactions. This determines that the oxidized monoliths have higher electrical capacitance (80 F cm−3) at low current densities and higher energy density (up to Wv = 2.7 W h L−1) than the non-oxidized ones. The supercapacitor cell containing the oxidized monoliths presents higher total resistance values and response time compared to the non-oxidized monolith. The non-oxidized monolith showed the highest power density (up to PV = 94.5 W L−1).

GLUT’s (facilitative glucose transporters) over-expression in tumor cells has allowed the detection of several cancer types, using a glucose analogue (18F-FDG) with PET images, worldwide. New glucose analogs radiolabeled with 99mTc could be a less-expensive and more accessible alternative for diagnosis using SPECT imaging. D-Glucose (99mTc-IDAG) and 2-D-deoxyglucose (99mTc-AADG) organometallic complexes were proposed and studied as potential 18F-FDG surrogates. The glucose complexes were prepared and evaluated as potential cancer imaging agents, in a melanoma tumor model. Iminodiacetic acid (IDA) and aminoacetate (AA) moieties were chosen as chelating system for radiolabeling with 99mTc. Tumor uptake of the formed complexes was evaluated in B16 murine cell line in vitro and in vivo in melanoma bearing C57BL/6 mice. In vitro and in vivo studies were conducted with 18F-FDG in order to compare the uptake of 99mTc-glucose complexes in the tumor model. IDAG and AADG compounds were synthesized and radiolabeled with 99mTcO4 � to obtain the 99mTc-IDAG and 99mTc-AADG complexes in high yield and stability. In vitro cell studies showed maximum uptake at 60 min for complexes, 99mTc-IDAG and 99mTc-AADG, with 6% and 2%, respectively. Biodistribution studies showed high tumor uptake one hour post-injection, reaching tumor-to-muscle ratios of 12.1 ± 3.73 and 2.88 ± 1.40 for 99mTc-IDAG and 99mTc-AADG, respectively. SPECT and micro-SPECT-CT images acquired after the injection of 99mTc-IDAG showed accumulation in tumor sites, suggesting that this glucose complex would be a promising candidate for cancer imaging

New benzofuroxans were developed and studied as antiproliferative Trypanosoma cruzi agents. Compounds displayed remarkable in Vitro activities against different strains, Tulahuen 2, CL Brener and Y. Its unspecific cytotoxicity was evaluated using human macrophages being not toxic at a concentration at least 8 times, and until 250 times, that of its T. cruzi IC50. Some biochemical pathways were studied, namely parasite respiration, cysteinyl active site enzymes and reaction with glutathione, as target for the mechanism of action. Not only T. cruzi respiration but also Cruzipain or trypanothione reductase were not affected, however the most active derivatives, the vinylsulfinyl- and vinylsulfonyl-containing benzofuroxans, react with glutathione in a redox pathway. Furthermore, the compounds showed good in ViVo activities when they were studied in an acute murine model of Chagas’ disease. The compounds were able to reduce the parasite loads of animals with fully established T. cruzi infections.

A summary of the resource.
<p>The adjuvant activity of Chenopodium quinoa (quinoa) saponins on the humoral and cellular immune responses of mice subcutaneously immunized with ovalbumin (OVA) was evaluated. Two quinoa saponin fractions were obtained, FQ70 and FQ90, and 10 saponins were determined by UPLC/Q-TOF-MS. Mice were immunized subcutaneously with OVA alone or adjuvanted with Quil A (adjuvant control), FQ70, or FQ90. FQ70 and FQ90 significantly enhanced the amount of anti-OVAspecific antibodies in serum (IgG, IgG1, and IgG2b) in immunized mice. The adjuvant effect of FQ70 was significantly greater than that of FQ90. However, delayed type hypersensitivity responses were higher in mice immunized with OVA adjuvanted with FQ90 than mice treated with FQ70. Concanavalin A (Con A)-, lipopolysaccharide-, and OVA-stimulated splenocyte proliferation were measured, and FQ90 significantly enhanced the Con A-induced splenocyte proliferation. The results suggested that the two quinoa saponin fractions enhanced significantly the production of humoral and cellular immune responses to OVA in mice. Abstract
A summary of the resource.
The structure of the extracellular polysaccharide from Azotobacter vinelandii strain 1484 has been investigated, specific degradations and n.m.r. spectroscopy being the main methods used. It is concluded that the polysaccharide is composed of tetrasaccharide repeating-units having the following structure, in which Sug is 3-deoxy-d-threo-hexulosonic acid. The polysaccharide also contains a non-stoichiometric amount of O-acetyl groups, distributed over at least two positions.

A rapid quantitative TLC method for the determination of glycoalkaloids in potato leaves and tubers is described. The method includes a microscale extraction, a simple clean-up step and quantification by TLC scanning. The method is valuable for the determination of glycoalkaloids when many samples have to be evaluated as in potato breeding programmes.

A summary of the resource.
The isolation of three new N-methylated piperidine-3-ol alkaloids from the bark of Prosopis affinis Spreng. (Ñandubay) is described. Together with MS and IR data, results from 1D and 2D NMR experiments allowed for the identification of N‑methyl-2-isocassine, N‑methyl-6-isocassine, and N‑methyl-6-isocarnavaline. Inspection of 1D-NOESY spectra and coupling constant data revealed that the heterocyclic moiety of the alkaloids is in fast conformational equilibrium in solution, and this behavior had to be taken into account in order to determine the relative configuration of the chiral centers of the piperidine rings.

Antifeedant and repellent effects of two different extracts from native Solanum species Solanum bistellatum and Solanum sisymbrifolium on Cryptotermes brevis were evaluated. The extracts obtained, particularly the dichloromethane extracts and the enriched fraction of sugar esters from S. sisymbrifolium and S. bistellaltum, showed antifeedant and repellent action against the termite. The antifeedant effect of dichloromethane extract from S. sisymbrifolium at the concentration of 25.00mg/mL reached 100%, while the repellent action of the dichloromethane extracts and that of the enriched fraction of sugar esters at the concentration of 1.00mg/mL was higher than 90%. In case of S. bistellatum, the antifeedant effect of the dichloromethane extract and the enriched fraction of sugar esters at the concentration of 25.00mg/mL was 43.29% and 57.46%, respectively. The repellent action of the dichloromethane extracts and of the enriched fraction of sugar esters at a concentration level of 2.50mg/mL was higher than 92%.

Consumer perception of food products is a very complex phenomenon that is influenced by a wide range of characteristics. In particular, consumer perception of the sensory characteristics of food products can be strongly affected by information and expectations. Thus, the comparison of product characterizations from blind and informed conditions can provide a valuable insight about the impact of information on consumer perception of food products. In this context, the aim of the present work was to present a case study in which the influence of information on results of product characterizations of milk desserts from two methodologies, check-all-that-apply (CATA) questions and sorting, was studied. Six commercial vanilla milk desserts were evaluated under blind and informed conditions by two groups of 50 consumers using CATA questions and sorting. Results showed that information had a small impact on overall liking scores and product characterizations, suggesting that consumers’ perception of the desserts was mainly determined by their sensory characteristics.

Consumers' increasing demand for fresh, safe and high quality products requires food companies to accurately estimate sensory shelf life and tolerance limits for sensory defects. Sensory shelf-life and acceptance limits for sensory defects have been estimated with consumers' rejection data using survival analysis without considering contextual variables, which could have a major influence on consumers' perception. The aim of the present study was to study how consumers' rejection is affected by different evoked contexts in a laboratory setting. Two studies were carried out to study the influence of evoked contexts on consumers' rejection. In the first study consumers' rejection for consumption at home and purchase at a supermarket was compared using orange juice samples with different storage times. In the second study, consumers' rejection for consumption at home and purchase at a supermarket for brands of different familiarities was compared using dulce de leche with different plastic flavour intensities as samples. Consumers' rejection data were analysed using survival analysis. The use of written scenarios to evoke different contexts affected consumers' rejection of products with different storage times and intensities of a sensory defect. Shelf lives that were estimated based on consumers' rejection to purchase were shorter than those estimated considering consumers' rejection to consume. In the second study, consumers' rejection under the consumption at home evoked context was similar to rejection to repeat purchase of a usual brand. However, a large difference existed between the evoked contexts that involved a usual and a new brand. Consumers were harsher when considering to repeat purchase of a new brand compared to their usual brand. These results suggest that the consideration of evoked contexts could contribute to increase the accuracy of sensory shelf life estimation and acceptance limits of sensory defects, leading to more informed business decisions.

Temporal Check-All-That-Apply (TCATA) has been recently introduced as a method for temporal sensory product characterization. This method requires assessors to select all the terms they consider applicable at each moment of the evaluation, and to de-select terms when they are no longer applicable. In the present work a variant of TCATA, TCATA Fading, is presented and compared to TCATA. In TCATA Fading selected terms gradually and automatically become unselected over a predefined period of time. Eight studies were conducted with different product categories in three different countries. In Studies 1–3, TCATA and TCATA Fading were compared with trained assessors using within-subject experimental designs on the same set of products. In Studies 4–6, TCATA and TCATA Fading were compared with consumers following between-subject experimental designs. Comparison was performed in terms of citation proportion of the terms, significant differences among samples detected, dynamic sample profiles, and task perceptions. Across the eight studies, results suggested that automatic de-selection of attributes in a TCATA task can improve discrimination and provide a more accurate description of the dynamics of sensory characteristics of products than asking consumers to de-select attributes when they are no longer applicable. The present research represents a first attempt at implementing and evaluating TCATA Fading, and suggests that it is a useful variant of the TCATA method. Many avenues for methodological refinement are identified.

Sensory and consumer scientists disagree on the practice of concurrently obtaining sensory information in hedonic tests. This is in part due to different mindsets about what consumers are able to do and evidence that such co-elicitation may bias hedonic scores. Check-all-that-apply (CATA) questions have been claimed to have a smaller effect on hedonic scores than other attribute such as just-about-right or intensity scales. In this research, nine studies using consumers as participants examined effects on hedonic product scores when sensory attribute information was co-elicited using CATA questions. The use of CATA concurrently with hedonic was benchmarked against concurrent attribute liking scores, attribute intensity scores and just-about-right scaling. Across a range of product categories (beer, fresh fruit, tea, flavoured water, crackers, savoury dips), only weak and transient evidence of bias of hedonic scores when concurrently using CATA questions was established. This effect was independent on whether samples, on average were moderately liked or moderately disliked, and replicated when samples were assessed partially by the sense of smell only or via full product assessment (appearance, aroma, flavour, taste, aftertaste, mouthfeel). The present research suggests that co-elicitation of hedonic scores and product attribute information using CATA questions may bias the hedonic scores, but not that it certainly will do so. This needs to be recognised, leading to more widespread acceptance that co-elicitation has merit. Investigators should decide on whether or not to co-elicit product attribute information using CATA questions on a case-by-case basis, acknowledging that bias may occur. Further research is needed to understand when/when not bias is likely to occur.<br /><br />

Temporal Check-All-That-Apply (TCATA) has been recently introduced as a method for temporal sensory product characterization. Building on the standard Check-All-That-Apply (CATA) question format, assessors select all the terms they consider applicable for describing the sensations they perceive, and they do so at each moment of the evaluation process. This research further investigates the TCATA method, through its application to products of varying complexity (yogurt, salami, cheese, orange juice, French bread, and marinated mussels) using consumers and trained panellists as assessors. More importantly, to deliver new methodological insights we compare TCATA to Temporal Dominance of Sensations (TDS). This comparison will aid researchers to select the temporal method best suited to their needs. Across three countries, six studies were conducted. Within-subjects experimental designs were used in Studies 1–3 and involved trained panellists using both TCATA and TDS on the same set of products. In Studies 4–6, between-subjects experimental designs were used, and the assessors, who were consumers, evaluated samples using either TCATA or TDS. The results confirmed that TCATA is suitable for measuring the temporal sensory characteristics of products. By enabling identification of several sensory characteristics that are concurrently perceived in products, the results from this research also suggest that TCATA may provide a more detailed description of the dynamics of the sensory characteristics of products. The TDS concept of dominance appears to decrease detailed description and discrimination of attributes that are simultaneously perceived, particularly when dealing with multiple sensory modalities. The practical implications of these differences are discussed

Beehive products could be powerful monitors of pesticide residues originating in agroecosystems during production cycles. Their ready availability provides enough samples to perform analytical determinations, but their chemical complexity makes residue analysis a real challenge. Taking advantage of the plasticity of QuEChERS coupled to LC-MS/MS, validated methodologies were developed for bees, honey, beeswax, and pollen and applied to real samples for the simultaneous determination of 19 of the most employed pesticides in intensive cropping fields. Beehives placed in Uruguayan agroecosystems accumulated the pesticides thiacloprid, imidacloprid, methomyl, carbaryl, hexythiazox, azoxystrobin, pyraclostrobin, tebuconazole, and haloxyfop-methyl at 0.0001–0.01 mg/kg levels. The oscillations on the amount and occurrence of residue findings for specific apiaries were correlated statistically with the sampling season and the agroecosystem where the beehives were located, showing the potential of bees and bee products to record relevant information to survey the chemicals applied in their surroundings.

Grapes are one of the most commonly consumed fruit, in both fresh and processed forms; however, a significant amount is disposed of in the environment. Searching for a use of this waste, the antigenotoxic, antimutagenic, and antioxidant activities of aqueous extracts from organic and conventional Vitis labrusca leaves were determined using V79 cells as model. The antigenotoxic activity was analyzed by the alkaline comet assay using endonuclease III and formamidopyrimidine DNA glycosylase enzymes. The antimutagenic property was assessed through the micronucleus (MN) formation, and antioxidant activities were assessed using 2',7'-dichlorodihydrofluorescin diacetate (DCFH-DA) assay and 2,2-diphenyl-1-picrylhydrazyl (DPPH(●)) radical scavenging, as well as with superoxide dismutase (SOD) and catalase (CAT) activity assays. In addition, phenolic content and ascorbic acid levels of both extracts were determined. Data showed that both organic and conventional grapevine leaves extracts possessed antigenotoxic and antimutagenic properties. The extract of organic leaves significantly reduced intracellular reactive oxygen species (ROS) levels in V79 cells, and displayed greater ability for DPPH(●) scavenging and higher SOD and CAT activities than extract from conventional leaves. Further, the extract from organic leaves contained higher phenolic and ascorbic acid concentrations. In summary, extracts from organic and conventional grape leaves induced important in vitro biological effects.
