A new method for the reversible immobilization of thiol bimolecules |b e.g.,thiolpeptides and thiolproteins |b to beaded agarose and other solid phases is reported. The method consists of an activation and a coupling step. The activation is based on oxidation of disulfides (or thiol groups via disulfides) present in a solid phase by hydrogen peroxide at moderately acidic pH. This oxidation leads to disulfide oxides (thiolsulfinate groups of which the majority are further oxidized to thiolsulfonate). The thiolsulfonate groups react easily with thiol compounds |b which become immobilized via disulfide bonds. The pH range for thiol coupling is wide (pH 5-8) |b but for most thiols the reaction seems to proceed faster at pH>7. The stability of the reactive group to hydrolysis |b especially at neutral and weakly acidic pH |b is very high. The activated gel |b therefore |b can be stored as a suspension at pH 5 for extended periods. The method has been used to reversibly immobilize glutathione |b Beta-galactosidase |b alcohol dehydrogenase |b urease |b and papain |b all with exposed thiol groups as well as thiolated bovine serum albumin and sweet-potato Beta-amylase. Depending on the thiol content of starting thiol-agarose |b thiol-sulfonate-agarose derivatives with different binding capacities can be obtained. Thus |b up to 5.0 mg (16 umol) glutathione and 15 mg thiol-protein/mL gel derivative have been immobilized. The gel bead can be regenerated and reused at least twice. Besides agarose |b cellulose |b crosslinked dextran |b and polyacrylamide were shown to be very suitable as supports for solid-phase thiolsulfonates.

<p>Classic acetyl thioester protection/deprotection methodologies are widely used in organic synthesis, but deprotection step usually requires harsh conditions not suitable for labile substrates. In this work, a new method for thioester deprotection using a thiotransesterification approach is described. Firstly, thioglycolic acid (TGA) was identified as a good deprotecting reagent in solution. In order to develop a thiol polymer-supported reagent, TGA was anchored to a PEG-based resin through an amide bond (TG-NCOSH).<br />Both homogeneous and heterogeneous approaches were conveniently carried out at room temperature, in aqueous buffer at pH 8. The mild conditions were suitable for alkyl and phenyl thioesters. Moreover labile thioesters containing thiazolidine and oxazolidine scaffolds, bearing amine, ester and<br />acetal functionalities were also deprotected. The polymer-supported TGA gave better deprotection yields compared to TGA in solution, yields ranging from 61 to 90%. The feasibility of the recovery and reuse of TG-NCO-SH reagent was explored, showing it can be reused at least five times without lossing the<br />activity.</p>

As part of our efforts to develop organometallic ruthenium compounds bearing activity on both trypanosomatid parasites and tumor cells, a new Ru(II)–cyclopentadienyl clotrimazole complex, [RuCp(PPh3)2(CTZ)](CF3SO3), where Cp = cyclopentadienyl, CTZ = clotrimazole, was synthesized and characterized. The compound was evaluated in vitro on T. cruzi (Y strain), the infective form of T. brucei brucei strain 427 (cell line 449), on three human tumor cell lines with different sensitivity to cisplatin (A2780, ovary; MCF7, breast; HeLa, cervix) and on J774 murine macrophages as mammalian cell model. The new compound is more cytotoxic on T. cruzi and on the tumor cell lines than the reference drugs (Nifurtimox and cisplatin, respectively). In addition, complexation of the bioactive CTZ to the {RuCp(PPh3)} moiety leads to significant increase of the antiparasitic and antitumoral activity. To get insight into the potential “dual” mechanism of antiparasitic action emerging from the presence of Ru(II) and CTZ in a single molecule, the inhibitory effect of this new complex on the biosynthesis of T. cruzi sterols of membrane and the interaction with DNA were studied. Although the tested complex does not affect DNA, it affects the T. cruzi biosynthetic pathway of conversion of squalene to squalene oxide. According to the results reported here, [RuCp(PPh3)2(CTZ)][CF3SO3] could be considered a prospective antiparasitic and/or antitumoral agent that deserves further evaluation

Searching for prospective metal-based drugs for the treatment of Chagas disease, a new series of ten mixed-ligand oxidovanadium(IV) complexes, [VIVO(L-2H)(NN)], where L is a tridentate salicylaldehyde semicarbazone derivative (L1–L5) and NN is either 5-amine-1,10-phenanthroline (aminophen) or 5,6-epoxy-5,6-dihydro-1,10-phenanthroline (epoxyphen), were synthesized. The compounds were characterized in the solid state and in solution. EPR spectroscopy suggests that the NN ligands act as bidentate through both nitrogen donor atoms in an axial–equatorial mode. The stability of the complexes in solution was investigated by EPR and 51V-nuclear magnetic resonance spectroscopies. The complexes were evaluated in vitro for their activities against Trypanosoma cruzi (T. cruzi), the parasite responsible for the disease, and their selectivity was analyzed using J-774 murine macrophages, as a mammalian model. All the complexes are more active than both the reference drug Nifurtimox and the previously reported [VIVO(L-2H)(NN)] complexes. In general they are more active than the corresponding free NN ligands. Complexation led to highly increased selectivities towards the parasite. In addition, the lipophilicity of the compounds was determined and correlated with the observed activity in order to perform a QSAR (quantitative structure–activity relationship) study. A clear quadratic correlation is found. This study also confirms the influence of the structure of the co-ligand on the anti-T. cruzi effect. To get insight into the mechanism of action of the compounds, the changes in biochemical pathways promoted by two of the most active and most selective complexes are studied by analyzing a few of the parasite excreted metabolites by 1H NMR spectroscopy. The combined information suggests that the mitochondrion could be a target for these complexes. Furthermore, DNA was preliminarily evaluated as a potential target by using atomic force microscopy (AFM), which showed that the complexes display an ability to interact with this biomolecule.

Motor neuron degeneration and neuroinflammation are the most striking pathological features of amyotrophic lateral sclerosis (ALS). ALS currently has no cure and approved drugs have only amodest clinically therapeutic effect in patients. Drugs targeting different deleterious inflammatory pathways in ALS appear as promising therapeutic alternatives. Here, we have assessed the potential therapeutic effect of an electrophilic nitroalkene benzoic acid derivative, (E)-4-(2-nitrovinyl) benzoic acid (BANA), to slow down paralysis progression when administered after overt disease onset in SOD1G93A rats. BANA exerted a significant inhibition of NF-κB activation in NF-κB reporter transgenicmice and microglial cell cultures. Systemic daily oral administration of BANA to SOD1G93A rats after paralysis onset significantly decreased microgliosis and astrocytosis, and significantly reduced the number of NF-κB-p65-positive microglial nuclei surrounding spinal motor neurons. Numerous microglia bearing nuclear NF-κB-p65 were observed in the surrounding of motor neurons in autopsy spinal cords from ALS patients but not in controls, suggesting ALS-associated microglia could be targeted by BANA. In addition, BANA-treated SOD1G93A rats after paralysis onset showed significantly ameliorated spinal motor neuron pathology as well as conserved neuromuscular junction innervation in the skeletal muscle, as compared to controls. Notably, BANA prolonged post-paralysis survival by ~30%, compared to vehicle-treated littermates. These data provide a rationale to therapeutically slow paralysis progression in ALS using small electrophilic compounds such as BANA, through a mechanism involving microglial NF-κB inhibition.

This paper describes a non-destructive “peak parking” protocol in order to assess the axial heterogeneity of an in situ modified monolithic column for high performance liquid chromatography; a “gradient stationary phase” was designed whereby the ligand density decreases along the length of the rod in the “forward flow” configuration. The results of multi-location peak parking demonstrated a consistent increase in peak variance from the 1 cm position of the column to the 9 cm location. This increase in band broadening supported the theory of a decreasing ligand density along the length of this gradient column. This is consistent with efficiency measurements performed in both the forward and reverse flow directions, with an improved efficiency (15% increase in N m−1) in the reverse direction. These results are consistent with theoretical investigations into stationary phase gradients.

It is urgent yet challenging to develop new environmentally friendly and cost-effective sources of energy. Molecular solar thermal (MOST) systems for energy capture and storage are a promising option. With this in mind, we have prepared a new water-soluble (pH > 6) norbor nadiene derivative (HNBD1) whose MOST properties are reported here. HNBD1 shows a better matching to the solar spectrum compared to unmodified norbornadiene, with an onset absorbance of λonset = 364 nm. The corresponding quadricyclane photoisomer (HQC1) is quantitatively generated through the light irradiation of HNBD1. In an alkaline aqueous solution, the MOST system consists of the NBD1−/QC1− pair of deprotonated species. QC1− is very stable toward thermal back-conversion to NBD1−; it is absolutely stable at 298 K for three months and shows a marked resistance to temper ature increase (half-life t1/2 = 587 h at 371 K). Yet, it rapidly (t1/2 = 11 min) releases the stored energy in the presence of the Co(II) porphyrin catalyst Co-TPPC (∆Hstorage = 65(2) kJ·mol−1 ). Under the explored conditions, Co-TPPC maintains its catalytic activity for at least 200 turnovers. These results are very promising for the creation of MOST systems that work in water, a very interesting solvent for environmental sustainability, and offer a strong incentive to continue research towards this goal.

Since most Saccharomyces strains show no �-glucosidase activity, the importance of non-Saccharomyces �-glucosidases in the development of wine aroma has been highlighted. However, only a few enzymes from yeasts isolated from grape-must and enological ecosystems are active in the stringent conditions of wine and the wine-making process (low pH, high concentrations of ethanol or glucose). A purified extract of extracellular �-glucosidase from Issatchenkia terricola, proved to be very active in the presence of glucose (100 g/L), ethanol (18%) and metabisulfite (60 mg/L). It is also active and relatively stable at acidic pH over 3.0. Immobilization onto Eupergit C greatly improved its stability, allowing the aromatization of white Muscat wine over a 16-day experiment. The released aroma compounds of control and treated wine were analyzed by GC–MS. The enzymatic treatment significantly increased the amount of monoterpenes and norisoprenoids, showing the potential of the immobilized enzyme for aroma development in wines.

The structure of the novel cadmium phenanthroline thio­sulfate poly­[[(1,10-phenanthroline-[kappa]2N,N')­cadmium(II)]-[mu]3-thio­sulfato-[kappa]3S:S:O], [Cd(S2O3)(C12H8N2)]n, with a pentacoordinated Cd centre, is reported. It forms linear chains built up around a 21 axis and is isostructural with the known bi­pyridine homologue. The structure is also compared with a previously reported closely related mono­aqua monohydrated phase, where the Cd2+ cation is hexacoordinated. The incidence of weak C-H...O interactions in the determination of its general packing properties is discussed.

A summary of the resource.
<p>Chronic metabolic diseases, like obesity, type II diabetes and atherosclerosis often involve a low-grade and sterile systemic inflammatory state, in which activation of the pro-inflammatory transcription factor NF-kB and the NLRP3 inflammasome play a major role. It is well established that genetic inhibition of the NLRP3 inflammasome ameliorates acute and chronic inflammation. Indeed, accumulating experimental evidences in murine models and also in humans suggest that inhibition of the NLRP3 inflammasome might be a suitable approach to tackle the deleterious effects of chronic metabolic diseases. In this work, we explored our previously synthesized nitroalkene-Trolox™ derivative named NATx0, as a non-conventional anti-inflammatory strategy to treat chronic inflammatory diseases, such as obesity-induced glucose intolerance. We found that NATx0 inhibited NF-kB nuclear translocation and pro-inflammatory gene expression in macrophages in vitro. In addition, treatment with NATx0 prevented NLRP3 inflammasome activation after LPS/ATP stimulation in macrophages in vitro. When tested acutely in vivo, NATx0 inhibited neutrophil recruitment in zebrafish larvae, and also diminished IL- 1β production after LPS challenge in mice. Finally, when NATx0 was administered chronically to diet-induced obese mice, it decreased muscle tissue inflammation and glucose intolerance, leading to improved glucose homeostasis. In conclusion, we propose that this novel nitroalkene-Trolox derivative is a suitable tool to tackle acute and chronic inflammation in vitro and in vivo mainly due to inhibition of NF-kB/NLRP3 activation.</p>

BACKGROUND AND PURPOSE: Atherosclerosis is characterized by chronic low-grade inflammation with concomitant lipid accumulation in the arterial wall. Anti-inflammatory and anti-atherogenic properties have been described for a novel class of endogenous nitroalkenes (nitrated-unsaturated fatty acids), formed during inflammation and digestion/absorption processes. The lipid-associated antioxidant α-tocopherol is transported systemically by LDL particles including to the atheroma lesions. To capitalize on the overlapping and complementary salutary properties of endogenous nitroalkenes and α-tocopherol, we designed and synthesized a novel nitroalkene-α-tocopherol analogue (NATOH) to address chronic inflammation and atherosclerosis, particularly at the lesion sites. EXPERIMENTAL APPROACH: We synthesized NATOH, determined its electrophilicity and antioxidant capacity and studied its effects over pro-inflammatory and cytoprotective pathways in macrophages in vitro. Moreover, we demonstrated its incorporation into lipoproteins and tissue both in vitro and in vivo, and determined its effect on atherosclerosis and inflammatory responses in vivo using the Apo E knockout mice model. KEY RESULTS: NATOH exhibited similar antioxidant capacity to α-tocopherol and, due to the presence of the nitroalkenyl group, like endogenous nitroalkenes, it exerted electrophilic reactivity. NATOH was incorporated in vivo into the VLDL/LDL lipoproteins particles to reach the atheroma lesions. Furthermore, oral administration of NATOH down-regulated NF-κB-dependent expression of pro-inflammatory markers (including IL-1β and adhesion molecules) and ameliorated atherosclerosis in Apo E knockout mice. CONCLUSIONS AND IMPLICATIONS: In toto, the data demonstrate a novel pharmacological strategy for the prevention of atherosclerosis based on a creative, natural and safe drug delivery system of a non-conventional anti-inflammatory compound (NATOH) with significant potential for clinical application.

Four novel rhenium complexes of formula [ReCl4(bpym)] (1), [ReBr4(bpym)] (2) PPh4[ReCl4(bpym)] (3) and NBu4[ReBr4(bpym)] (4) (bpym= 2,2�-bipyrimidine, PPh4 = tetraphenylphosphonium cation and NBu4 = tetrabutylammonium cation), have been synthesized and their crystal structures determined by single-crystal X-ray diffraction. The structures of 1 and 2 consist of [ReX4(bpym)] molecules held together by van der Waals forces. In both complexes the Re(IV) central atom is surrounded by four halide anions and two nitrogen atoms of a bpym bidentate ligand in a distorted octahedral environment. The structures of 3 and 4 consist of [ReX4(bpym)]− anions and PPh4 + (3) or NBu4 + (4) cations. The coordination sphere of the Re(III) metal ion is the same as in 1 and 2, respectively. However, whereas the Re–X bonds are longer the Re–N bonds are shorter than in 1 and 2. This fact reveals that the bpym ligand forms a stronger bond with Re(III) than with Re(IV) resulting in a stabilisation of the lower oxidation state. [ReX4(bpym)] complexes are easily reduced, chemically and electrochemically, to the corresponding [ReX4(bpym)]− anions. A voltammetric study shows that the electron transference is a reversible process characterized by formal redox potentials of +0.19 V (1) and +0.32 V (2) vs. NHE, in acetonitrile as solvent

Janibacter sp. strain R02 (BNM 560) was isolated in our laboratory from an Antarctic soil sample. A remarkable trait of the strain was its high lipolytic activity, detected in Rhodamine-olive oil supplemented plates. Supernatants of Janibacter sp. R02 displayed superb activity on transesterification of acyl glycerols, thus being a good candidate for lipase prospection. Considering the lack of information concerning lipases of the genus Janibacter, we focused on the identification, cloning, expression and characterization of the extracellular lipases of this strain. By means of sequence alignment and clustering of consensus nucleotide sequences, a DNA fragment of 1272 bp was amplified, cloned and expressed in E. coli. The resulting recombinant enzyme, named LipJ2, showed preference for short to medium chain-length substrates, and displayed maximum activity at 80 °C and pH 8–9, being strongly activated by a mixture of Na+ and K+. The enzyme presented an outstanding stability regarding both pH and temperature. Bioinformatics analysis of the amino acid sequence of LipJ2 revealed the presence of a consensus catalytic triad and a canonical pentapeptide. However, two additional rare motifs were found in LipJ2: an SXXL β-lactamase motif and two putative Y-type oxyanion holes (YAP). Although some of the previous features could allow assigning LipJ2 to the bacterial lipase families VIII or X, the phylogenetic analysis showed that LipJ2 clusters apart from other members of known lipase families, indicating that the newly isolated Janibacter esterase LipJ2 would be the first characterized member of a new family of bacterial lipases.

Several breeding programs are under way to introduce resistance to bacterial wilt caused by Ralstonia solanacearum in solanaceous crops. The lack of screening methods allowing easy measurement of pathogen colonization and the inability to detect latent (i.e. symptomless) infections are major limitations when evaluating resistance to this disease in plant germplasm. We describe a new method to study the interaction between R. solanacearum and potato germplasm that overcomes these restrictions. The R. solanacearum strain UY031 was genetically modified to constitutively generate light from a synthetic luxCDABE operon stably inserted in its chromosome. Colonization of this reporter strain on different potato accessions was followed using life imaging. Bacterial detection in planta by this non-disruptive system correlated with the development of wilting symptoms. In addition, we demonstrated that quantitative detection of the recombinant strain using a luminometer can identify latent infections on symptomless potato plants. We have developed a novel, unsophisticated and accurate method for high-throughput evaluation of pathogen colonization in plant populations. We applied this method to compare the behavior of potato accessions with contrasting resistance to R. solanacearum. This new system will be especially useful to detect latency in symptomless parental lines before their inclusion in long-term breeding programs for disease resistance.

The third is a complex isoflavan where the isoprenyl substituent and the aromatic A-ring yielded a p-quinol nucleus.

<p>Interfacing a microflow velocity < 100 μL/min analyses between the separation technique and MS detection for environmental organic contaminant analyses have shown potential to decrease the matrix effect. However performing such volumetric flow rates typically involves the upgrade of equipment. A plug and play study to demonstrate the potential of the Parallel Segmented Flow (PSF) column technology without any changes to the separation or detection programmed settings can facilitate microflow separations on a HPLC 400 bar hyphenated MS system. Here, we present the PSF column that interfaced 54 μL/min with electrospray tandem mass spectrometry (MS/MS) detection. Advantages of this alternative plug and play PSF microflow interfacing strategy include robust HPLC separation % RSD retention times ≤ 0.38% and mobile phase consumption comparable to UHPLC consumption rates of 9 mL per analysis. The calculated matrix effect for microflow PSF was representative of reduced ion suppression/enhancement (< ± 20%), with the exception of atrazine in river water which experienced medium suppression (< ± 30%). The %ME is not yet competitive with capillary scale columns. The instrument detection limits (IDLs) calculated takes into account the variability (%RSD) and amount reaching the detector. The PSF had lower IDLs (3.09–7.66 pg) compared to the conventional column (1.53–31.61 pg), with the exception of atrazine’s peak height calculation. This communication focused on the microflow PSF operation strictly on conventional HPLC tandem MS system, without hardware upgrades, and applicability for robust organic contaminant separations in environmental matrices (soil and river water).</p>

A summary of the resource.
Yeasts of the genus Hanseniaspora gained notoriety in the last years due to their contribution to wine quality, and their loss of several genes, mainly related to DNA repair and cell cycle processes. Based on genomic data from many members of this genus, they have been classified in two well defined clades: the “faster-evolving linage” (FEL) and the “slower-evolving lineage” (SEL). In this context, we had detected that H. vineae exhibited a rapid loss of cell viability in some conditions during the stationary phase compared to H. uvarum and S. cerevisiae. The present work aimed to evaluate the viability and cell cycle progression of representatives of Hanseniaspora species along their growth in an aerobic and discontinuous system. Cell growth, viability and DNA content were determined by turbidity, Trypan Blue staining, and flow cytometry, respectively. Results showed that H. uvarum and H. opuntiae (representing FEL group), and H. osmophila (SEL group) exhibited a typical G1/G0 (1C DNA) arrest during the stationary phase, as S. cerevisiae. Conversely, the three strains studied here of H. vineae (SEL group) arrested at G2/M stages of cell cycle (2C DNA), and lost viability rapidly when enter the stationary phase. These results showed that H. vineae have a unique cell cycle behavior that will contribute as a new eukaryotic model for future studies of genetic determinants of yeast cell cycle control and progression.

A simple sticking probability model is used for deducing a kernel capable to describe the kinetics of computer-simulated irreversible aggregation processes. Not only the diffusion- and reaction-limited aggregation regimes were fitted but also the whole transition region. The deduced kernel establishes λ = 0 for the entire range of sticking probabilities and helps to understand how irreversible cluster-cluster aggregation works.

A summary of the resource.
The increasing concern about accidents involving radioactive materials has led to the search of alternative methods to quickly identify and quantify radionuclides in workers and in the population. One of the options to face up an eventual demand for mass monitoring of internal contamination is the use of a nuclear medicine diagnostic equipment known as gamma camera, a device used to scan patients who have been administered specific amounts of radioactive materials for medical purposes. The components of a gamma camera are the collimator, detector crystal, photomultiplier tube array, position logic circuits, and the data analysis computer. A Thallium-activated Sodium Iodide [NaI (Tl)] detector crystal is generally used in Gamma cameras. This is due to this crystal's optimal detection efficiency for the gamma ray energies of radionuclide emission common to Nuclear Medicine. Although the gamma camera is used for image diagnosis it can be calibrated with anthropomorphic phantoms or point sources for the quantification of radionuclide activities in the human body. Therefore, a wide range of radioactive materials can be detected by means of this technique. This work presents a protocol for the calibration of gamma cameras for such application. In order to evaluate the suitability of this type of equipment, a gamma camera available in a public hospital located in Rio de Janeiro was calibrated for the in vivo measurement of 131I. The calibration includes the determination of detection efficiencies and minimum detectable activities for each radionuclide. The results show that the gamma camera presents enough sensitivity to detect activity levels corresponding to effective doses bellow 1 mSv. The protocol is the basis to establish a network of nuclear medicine centers, located in public hospitals in 8 countries of South America (Argentina, Brazil, Colombia, Cuba, Chile, Mexico, Peru, Uruguay) and in Spain that could be requested to collaborate in remediation actions in the event of an accident involving incorporation of radioactive materials. This protocol is one of the most significant outputs of the IAEA-ARCAL Project (RLA/9/049-LXXVIII) aimed to the Harmonization of Internal Dosimetry Procedures
