This study investigated the microbial community developed in a UASB reactor for hydrogen production and correlated it to reactor performance. The reactor was inoculated with kitchen waste compost and fed with raw cheese whey at two organic loading rates, 20 gCOD/Ld and 30 gCOD/Ld. Hydrogen production was very variable, using an OLR of 30 gCOD/Ld averaged 1.0 LH2/Ld with no methane produced under these conditions. The hydrogen yield was also very variable and far from the theoretical. This low yield could be explained by selection of a mixed fermentative population with presence of hydrogen producing organisms (Clostridium, Ruminococcus and Enterobacter) and other non-hydrogen producing fermenters (Lactobacillus, Dialister and Prevotella). The molecular analysis of the raw cheese whey used for feeding revealed the presence of three predominant organisms that are affiliated with the genera Buttiauxella (a low-yield hydrogen producer) and Streptococcus (a lactic acid-producing fermenter). Although these organisms did not persist in the reactor, the continuous addition of these fermenters could decrease the reactor’s hydrogen yield. Leaves of Palicourea marcgravii were extracted successively with hexane, ethyl acetate and ethanol in order to evaluate their acaricidal activity on larvae and adult stages of the cattle tick Rhipicephalus (Boophilus) microplus. The ethyl acetate extract showed the highest bioactivity of the tested extracts, which contained 0.12% monofluoroacetic acid. On engorged female, the ethyl acetate extract showed a lethal concentration 50% – LC50 = 30.08mgml−1, inhibitory concentration 50% – IC50 = 5.79mgml−1 and lethal time 50% – LT50 = 4.72 days; 100% reproduction was controlled at concentrations of 50mgml−1 and on larvae the ethyl acetate extract showed a LC50 = 2.46mgml−1. No alkaloids were detected in any of the extracts. This is the first report on the acaricidal activity of P. marcgravii extracts against R. microplus as well as the acaricidal properties of a plant species containing monofluoroacetic acid. The aetiological agent of cystic hydatid disease, the platyhelminth parasite Echinococcus granulosus, undergoes a series of metamorphic events during its complex life cycle. One of its developmental stages, the protoscolex, shows a remarkable degree of heterogeneous morphogenesis, being able to develop either into the vesicular or strobilar direction. Another level of complexity is added by the existence of genotypes or strains that differ in the range of intermediate hosts where they can develop and form fertile cysts. These features make E. granulosus an interesting model for developmental studies. Hence, we focused on the study of the regulation of gene expression by microRNAs (miRNAs), one of the key mechanisms that control development in metazoans and plants and which has not been analysed in E. granulosus yet. In this study, we cloned 38 distinct miRNAs, including four candidate new miRNAs that seem to be specific to Echinococcus spp. Thirty-four cloned sequences were orthologous to miRNAs already described in other organisms and were grouped in 16 metazoan miRNA families, some of them known for their role in the development of other organisms. The expression of some of the cloned miRNAs differs according to the parasite life cycle stage analysed, showing differential developmental expression. We did not detect differences in the expression of the analysed miRNAs between protoscoleces of two parasite genotypes. This work sets the scene for the study of gene regulation mediated by miRNAs in E. granulosus and provides a new approach to study the molecules involved in its developmental plasticity and intermediate host specificity. Understanding the developmental processes of E. granulosus may help to find new strategies for the control of cystic hydatid disease, caused by the metacestode stage of the parasite. Hypoxic regions of tumours are associated with increased resistance to radiation and chemotherapy. Nevertheless, hypoxia has been used as a tool for specific activation of some antitumour prodrugs, named bioreductive agents. Phenazine dioxides are an example of such bioreductive prodrugs. Our 2D-quantitative structure activity relationship studies established that phenazine dioxides electronic and lipophilic descriptors are related to survival fraction in oxia or in hypoxia. Additionally, statistically significant models, derived by partial least squares, were obtained between survival fraction in oxia and comparative molecular field analysis standard model (r2 = 0.755, q2 = 0.505 and F = 26.70) or comparative molecular similarity indices analysis-combined steric and electrostatic fields (r2 = 0.757, q2 = 0.527 and F = 14.93), and survival fraction in hypoxia and comparative molecular field analysis standard model (r2 = 0.736, q2 = 0.521 and F = 18.63) or comparative molecular similarity indices analysis-hydrogen bond acceptor field (r2 = 0.858, q2 = 0.737 and F = 27.19). Categorical classification was used for the biological parameter selective cytotoxicity emerging also good models, derived by soft independent modelling of class analogy, with both comparative molecular field analysis standard model (96% of overall classification accuracy) and comparative molecular similarity indices analysis-steric field (92% of overall classification accuracy). 2D- and 3D-quantitative structure-activity relationships models provided important insights into the chemical and structural basis involved in the molecular recognition process of these phenazines as bioreductive agents and should be useful for the design of new structurally related analogues with improved potency. Tradicionalmente, los extractos y plantas frescas de Mentha han sido usados para prolongar la vida útil de los alimentos y como componentes de productos farmacéuticos y cosméticos, por sus propiedades antimicrobianas y antioxidantes. Una alternativa viable para la comercialización de menta es su presentación como producto fresco, cortado, envasado y listo para usar, con un mayor valor agregado. El objetivo del presente trabajo fue evaluar las variaciones en el rendimiento y la composición de los aceites esenciales de Mentha×piperita y M. spicata frescas, cortadas, envasadas y almacenadas a 0°C. Para ello, los tallos herbáceos y hojas de ambas especies fueron cosechados, seleccionados, lavados con agua corriente y desinfectados por inmersión en agua clorada (200 ppm de cloro activo; pH 6-7; 1 min). Se secaron mediante corriente de aire forzado y se envasaron en bandejas cubiertas con película de cloruro de polivinilo, conteniendo 30-35g del producto así procesado. Se conservaron durante 21 días a 0ºC. Las experiencias de almacenamiento se realizaron por duplicado. Al inicio y al final de la conservación se extrajeron los aceites esenciales mediante una trampa graduada tipo Clevenger, apta para esencias de menor densidad que el agua. El rendimiento se expresó como mL de aceite esencial/100g de materia seca. Los componentes de los aceites esenciales se analizaron mediante HRGC-MS, utilizando una columna capilar de sílica fundida, con fase estacionaria ligada BP 20 de tipo polietilenglicol. Los resultados se sometieron a análisis de varianza y se compararon las medias mediante el test de la DMS (p<0,05). El rendimiento de aceite esencial estuvo comprendido entre 1,5 y 3,2 mL/100g (base seca), dependiendo de la especie de menta. En el caso de M. piperita fueron identificados 24 componentes al principio y 23 al final del almacenamiento, correspondiendo respectivamente al 89,7% y 82,4% del total de los componentes analizados. En M. spicata fueron identificados 11 y 16 constituyentes, representando el 60,8% (día 0) y 53,1% (día 21) del total de los componentes del aceite. Los principales compuestos responsables del flavor de M. piperita variaron apreciablemente durante el almacenamiento: la mentona disminuyó 29,4 % con respecto al porcentaje inicial, luego de 21 días de almacenamiento, al igual que el mentol que lo hizo en menor medida (7,2 % del porcentaje inicial, en el mismo período). Por otro lado, la piperitona mostró un leve incremento. De los componentes que dan lugar al flavor característico de M. spicata, los más afectados durante el almacenamiento fueron la carvona, que se redujo un 17% con respecto al porcentaje inicial después de 21 días a 0°C, y el limoneno que también sufrió una notoria disminución (22% con relación al inicio) en el mismo período. Estas modificaciones, vinculadas al metabolismo de los terpenos, serían relevantes para el diseño del producto. Myrcianthes cisplatensis (Cambess.) O. Berg (Myrtaceae) grows freely in Uruguay especially in the banks of rivers and streams. It is locally known as guayabo colorado and it's fruits are edible and used for the preparation of marmalades. In this work we present the results of the bioguided isolation and structural elucidation of the main active compound as well as it's antibacterial activity. Through repeated chromatography a pure compound could be obtained. The compound was studied by different spectroscopic techniques and could be unambiguously identified as α-methyl-1-(2′, 4′, 6,-trimethoxyphenyl)-1-propanone. When assaying for antistaphylococcal activity, it showed MICs of 62.5 μg/mL for the sensible strain (ATC 6538p) and 250 μg/mL for the multirresistant ones (ATCC 43300 and ATC 700699). Bilayer graphene simultaneously doped with two different second-row atoms (aluminum and phosphorus) was investigated employing first-principle density functional calculations (LDA, VDW-DF and M06-L). In agreement with the results obtained for bilayer graphene doped with one type of heteroatom, we found that at 1.56% atomic doping of Al and 1.56% atomic doping of P, the structure with sheets not linked is more stable than that with an Al–P bond. While all methods agreed in predicting the most stable structure, the relative energies of the structural isomers were different at the three levels of theory considered. The electronic properties of doped bilayer graphene showed an unusual variation upon small conformational changes. In effect, metallic, semiconducting and semimetallic properties were observed upon variation of the protruding orientation of the heteroatoms. The electronic properties of bilayer graphene seem to be determined by the layer whose heteroatom protrudes into the interlayer region. When Al protrudes in and P out metallic properties are observed. However, when P protrudes in and Al in a small band gap semiconductor is obtained. This work shows that the electronic properties of bilayer graphene can be more easily tuned than those of monolayer graphene because a small conformational change can induce a metallic to semiconductor transition or vice versa. Herein, we perform a M06-2X/6-311G investigation of the homo and mixed dimers formed between corannulene, sumanene, and pentaindenocorannulene; also, their complexes with C60 and C70 were studied. Despite the lower area of sumanene, the interaction of the sumanene-dimer is stronger that of the corannulene- dimer because of CH–p interactions. For mixed-dimers, when the concave and convex sides perfectly match the interactions are similar to those of the homodimers. However, when the concave area is larger than the convex one, CH–p interactions become important and perfect stacking is lost. The interaction between pentaindenocorannulene-C60 is twice that of corannulene-C60, and DG298 = �10.8 kcal/ mol. The addition of oxygen-centered radicals to fullerenes has been intensively studied due to their role in cell protection against against hydrogen peroxide induced oxidative damage. However, the analogous reaction of sulfur-centered radicals has been largely overlooked. Herein, we investigate the addition of S-centered radicals to C50, C60, C70, and C100 fullerenes by means of DFT calculations. The radicals assayed were: S, SH, SCH3, SCH2CH3, SC6H5, SCH2C6H5, and the open-disulfide SCH2CH2CH2CH2S. Sulfur, the most reactive species, prefers to be attached to a 66- bond of C60 with a binding energy (Ebind) of 2.4 eV. For the SR radicals the electronic binding energies to C60 are 0.77, 0.74, 0.58, 0.67, and 0.35 eV for SH, SCH3, SCH2CH3, SCH2C6H5, and SC6H5, respectively. The reactivity of C60 toward SR radicals can be increased by lithium doping. For Li@C60, the Ebind is increased by 0.65 eV with respect to C60, but only by 0.33 eV for the exohedral doping. Fullerenes act like free radical sponges. Indeed, the C60-SR Ebind can be duplicated if two radicals are added in ortho or para positions. The enhanced reactivity because of multiple additions is mostly a local effect, although the addition of one radical makes the whole cage more reactive. Therefore, as observed for hydroxylated fullerenes, they should protect cells from oxidative damage. However, the thiolated fullerenes have one advantage, they can be easily attached to gold nanoparticles. For the addition on pentagon junctions smaller fullerenes like C50 are more reactive than C60. Interestingly, C70 is as reactive as C60, even for the addition on the equatorial belt. For larger fullerenes like C100, reactivity decreases for the carbon atoms belonging to hexagon junctions.
