A summary of the resource.
Peroxisome proliferator-activated receptors (PPARs) are multi-domains proteins, belonging to the superfamily of nuclear receptors, which mainly act as ligand-activated transcription factors. A variety of lipophilic molecules, including long-chain polyunsaturated fatty acids and eicosanoids, are capable of binding to PPAR, although the nature of the physiological ligands is still under debate. PPARs regulate the expression of a set of genes involved in glucose and lipid metabolism as well as in the control of inflammatory responses. Herein we review the main molecular and cellular events associated with the activation of PPARs and their effects on metabolism.

A summary of the resource.
The laminated layer (LL) is the massive carbohydrate-rich structure that protects Echinococcus larvae, which cause cystic echinococcosis (hydatid disease) and alveolar echinococcosis. Increased understanding of the biochemistry of the LL is allowing a more informed analysis of its immunology. The LL not only protects the parasite against host attack but also shapes the overall immune response against it. Because of its dense glycosylation, it probably contains few T-cell epitopes, being important instead in T-cell independent antibody responses. Crucially, it is decoded in non-inflammatory fashion by innate immunity, surely contributing to the strong immune-regulation observed in Echinococcus infections. Defining the active LL molecular motifs and corresponding host innate receptors is a feasible and promising goal in the field of helminth-derived immune-regulatory molecules.

A novel oxalato-bridged rhenium(IV)-copper(II) compound, namely [ReIVBr4(l-ox)CuII(bpy)2] (1), has been obtained by reacting (PPh4)2[ReBr4(ox)] with Cu(CF3SO3)2 and 2,20-bpy in CH3CN, and its crystal structure determined by single-crystal X-ray diffraction. Intermolecular Br� � �Br interactions and nonbonding Cu� � �Br type contacts between the heterobimetallic dinuclear units lead to a two-dimensional supramolecular structure. Compound 1 behaves magnetically as a [ReIVCuII]2 tetranuclear species with weak antiferromagnetic interactions through the oxalato bridge and intermolecular Br� � �Br contacts

Durante este proyecto de tesis se ha desarrollado un novedoso soporte bifuncional para la inmovilizaci6n de proteinas, que permite combinar las ventajas de la uni6n sitio especifica con las de la inmovilizaci6n covalente multipuntual. EI mismo contiene: i) una pequena proporci6n de gropos tiol-reactivos, y ii) un alto numero de gropos ep6xidos. La optimizaci6n de su sintesis se llev6 a cabo utilizando ditiotreitol 0 sulfuro s6dico como agentes tiolantes, y resinas acrilicas (Eupergit C, EP-Sepabeads) como matrices epoxiactivadas. La inmovilizaci6n en este tipo de soportes es un proceso en dos etapas. Primero la proteina es inmovilizada en forma reversible a traves de un intercambio tiol-disulfuro entre los gropos tiol de la enzima y los gropos tiol-reactivos del soporte bajo condiciones suaves de temperatura (24°C) Y pH (7.0). Luego, en una segunda etapa los gropos ep6xido remanentes del soporte son capaces de formar uniones' irreversibles intramoleculares con los gropos nucleofilicos superficiales de la enzima (principalmente residuos de lisina), debido a su proximidad. Este mecanismo en dos etapas fue verificado por medio del estudio del proceso de inmovilizaci6n de proteinas modelo: i) que presentan naturalmente gropos tiol superficiales (~-galactosidasa de Escherichia coli), y ii) cuyos gropos tiol necesitan ser generados por reducci6n de disulfuros nativos (~-galactosidasa de Kluyveromices lactis) 0 par tiolaci6n con SPDP (penicilina G acilasa de E. coli y lipasa de Rhizomucor miehei). A su vez, estudios de inactivaci6n termica demostraron que las uniones adicionales multipuntuales que se establecen durante la segunda etapa de la inmovilizaci6n, son las responsables de un incremento en la estabilidad de las enzimas inmovilizadas con respecto alas enzimas solubles 0 alas inmovilizadas iInicamente a traves de enlaces disulfuros. Dado que la presencia de gropos sulfhidrilo en la superficie de la estructura proteica es una condici6n esencial para el uso de este nuevo soporte mixto, en el desarrollo de esta tesis se estudiaaran en forma complementaria dos estrategias para la generaci6n de gropos tiol: i) por reducci6n de puentes disulfuros, y ii) por mutagenesis dirigida. Es asi que por un lado, se evalu6 el uso de tiopraapil-agarosa, para la obtenci6n de gropos sulfhidrilo por reducci6n de disulfuros proteicos, como una altemativa al uso de reductores solubles. A su vez, con el fin de poder controlar totalmente la orientaci6n proteica sobre los soportes desarrollados, se estudi6 la viabilidad de introducir residuos de cisteina en la PGA de E. coli por medio de mutagenesis dirigida. Si bien los resultados obtenidos son muy preliminares, 10s mismos alientan a la realizaci6n de investigaciones que permitan estudiar el efecto que podria tener en la estabilidad enzimMica, el inmovilizar la PGA por distintas zonas de su superficie en los soportes desarrollados.

The ubiquitous intracellular molecule myo-inositol hexakisphosphate (IP6) is present extracellularly in the hydatid cyst wall (HCW) of the parasitic cestode Echinococcus granulosus. This study shows that extracellular IP6 is present as its solid calcium salt, in the form of deposits that are observed, at the ultrastructural level, as naturally electron dense granules some tens of nanometers in diameter. The presence of a calcium salt of IP6 in these structures was determined by two different electron microscopy techniques: (i) the analysis of the spatial distribution of phosphorus and calcium in the outer, acellular layer of the HCW (the laminated layer, LL) through electron energy loss spectroscopy, and (ii) the observation, by transmission electron microscopy, of HCW that were selectively depleted of IP6 by treatment with EGTA or phytase, an enzyme that catalyses the dephosphorylation of IP6. The deposits of the IP6-Ca(II) salt are also observed inside membrane vesicles in cells of the germinal layer (the inner, cellular layer of the HCW), indicating that IP6 precipitates with calcium within a cellular vesicular compartment and is then secreted to the LL. Thus, much as in plants (that produce vesicular IP6 deposits), the existence of transporters for IP6 or its precursors in internal membranes is needed to explain the compound's cellular localisation in E. granulosus.

NAMD is a parallel molecular dynamics software designed for high-performance simulations of large biomolecular systems. It scales from single computer up to hundreds of processors as high-end parallel platforms. Additionally, considering the evolution of Graphics Processing Units (GPUs) as a general purpose massively parallel co-processors, NAMD has included this kind of devices to leverage its computational power. In this work we analyze current NAMD GPU solution and develop an alternative based on Newton’s third law. The results shows a significant reduction of the execution time of GPU computations, of up to 20 % when compared with a highly tuned version of the original GPU-enabled NAMD.

Las mandarinas presentan una gran diversidad en características sensoriales y de calidad de fruta, así como una cosecha extendida. El mejoramiento genético de cítricos se focaliza en la búsqueda de nichos de cosecha para la obtención de mayores precios, así como en aspectos nutricionales y sensoriales, entre otras caracterís-ticas. En este contexto, la caracterización sensorial de nuevos híbridos y la identificación de las características sensoriales claves que influyen en la aceptabilidad de los consumidores proveen de una valiosa información para el mejoramiento y las estrategias de marketing. Estudios previos mostraron que las características senso-riales, especialmente el sabor, son claves en la aceptación de las mandarinas por los consumidores. Se llevaron a cabo dos estudios durante la estación de cosecha de mandarinas (media y tardía) aplicando la metodología «marque todo lo que corresponda» (CATA) y con la participación de al menos 100 consumidores. La aceptabi-lidad de los cultivares que fueron más aceptados se situó entre 6,5 y 7,7 en una escala hedónica de 9 puntos, evidenciando una reacción hedónica positiva. Los nuevos híbridos presentaron puntajes de aceptabilidad simi-lares o superiores a los cultivares de referencia Tango, Gold Nugget, Murcott y Ortanique. A su vez, los resul-tados mostraron que el color naranja, la forma regular, la piel lisa, el dulzor, la jugosidad, el sabor intenso y el sabor típico a mandarina son características claves que determinan la aceptabilidad de las mandarinas, y que la apariencia externa no es un buen predictor del sabor. Los híbridos F3P8, F5P8 y F2P3 fueron descritos por los consumidores con términos sensoriales positivos, recibiendo valores de aceptabilidad similares o mayores que los cultivares de referencia como tango, reafirmando la importancia de la incorporación de estudios senso-riales en el contexto del mejoramiento genético

<p>Echinococcus granulosus sensu lato (s.l.) is the helminth parasite responsible for cystic echinococcosis, a neglected<br />tropical disease currently affecting millions of people worldwide. Incomplete knowledge on the parasite biochemistry contributes, at least partially, to the limited development of useful biotechnological advances for the infection control. In this sense, little information is available regarding post-translational modifications (PTMs) occurring in E. granulosus s.l. proteins, which ultimately may affect the performance of biotechnological products to be developed. Therefore, we report here a proteomic analysis of the parasite PTMs identified through FindMod software applied to a set of tegumental proteins previously characterized by mass spectrometry<br />(MALDI-TOF/TOF) analysis of protein spots from a 2D electrophoresis gel. Manual searches for already annotated<br />proteins exhibiting such PTMs were also performed within proteome databases of E. granulosus s.l. and other platyhelminthes. In addition, key enzymes involved in PTMs modifications were searched for within E. granulosus s.l. proteome. Finally, the presence of selected PTMs was further confirmed by a high-resolution proteomic approach (nanoLC-MS/MS). A set of 22 different PTMs most likely to be present in the parasite was suggested, 9 of them with high confidence as they were identified in the same m/z fragment by both proteomic techniques (acetylation, deamidation, deamidation followed by methylation, mono- and dihydroxylation,<br />mono- and di-methylation, S-nitrosylation and phosphorylation). Interestingly, 5 PTMs were herein identified for the first time in E. granulosus s.l. proteins. Our results expand the scarcely studied topic of PTMs in platyhelminthes.</p>

We have employed density functional theory to study the structural, electronic and magnetic properties of the first all-carbon layer grown epitaxially on 6H-SiC(0 0 0 1). Using VDW-DF, M06-L, LSDA, LSDA+U, PBE and PBE-D2 methods we have performed a comparative study of the preferable magnetic configuration of the system. In this work, for the first time, we report a stable antiferromagnetic (AF) ordering in the buffer layer caused by the presence of silicon dangling bonds in the SiC top layer. This state is nearly degenerated with the ferromagnetic state with a magnetic moment equal to the number of silicon dangling bonds. A net magnetic moment of 0.55 µb per Si dangling bond was found for both states. However, only for the ferromagnetic state the carbon atoms of the buffer layer exhibited a magnetic moment. The magnetic configuration is much more stable than the non-polarized one and might explain SQUID results and spin transport experiments with epitaxial graphene. Furthermore, we found that, as previously observed experimentally, the buffer layer is a true semiconductor.

In the present report, we present a combined experimental and theoretical study on the lithium bis(trifluoromethanesulfonyl)imide (LiTFSI) doping mechanism of regioregular poly(3-hexyl thiophene) (P3HT). First, we focus on the effects of LiTFSI doping in both crystalline and amorphous structures of P3HT by performing a complete structural analysis supported by classical molecular dynamics (MD) calculations. Then, we study the effects of LiTFSI doping on electronic properties such as charge transfer and charge transport by performing Raman and impedance spectroscopy, in both cases supported by density of functionals theory (DFT) calculations using periodic boundary conditions. Our structural analysis suggests that the LiTFSI dopant is mainly located in the amorphous region and only a small fraction is located in the crystalline region. In addition, our DFT calculations also suggest that the LiTFSI dopant can effectively act as an electronic acceptor only when it is located in the vicinity of and is accessible to the thiophene rings of P3HT due to the formation of a π···Li chemical bond as an anchoring mechanism, permitting the electronic charge loss of thiophene rings through the sulfonyl groups. A thorough understanding of the LiTFSI doping mechanism of poly(alkyl thiophenes) (P3HT in this particular case) is crucial to elucidating not only the electronic but also the eventual mixed ionic–electronic transport mechanism and its promising properties, particularly as electrodes for lithium ion battery applications.

The trichalcogenides Sb2S3, Sb2Se3, Bi2S3, and Bi2Se3 share an orthorhombic crystal structure and have recently been pointed out as promising materials for application in solar energy harvesting, such as photovoltaic solar cells, because of their ultimate structural and electronic/optical properties. In this work, using a first-principles theoretical approach, we investigated the origin of the electrical conduction in bulk systems as well as the energy band alignment in different heterostructures composed of these compounds. In the first part, formation energy and thermodynamic transition energy of native point defects are evaluated. In the second part, surface properties such as free energy and electron affinity were obtained. In the third part, the energy alignments of some possible heterostructures were proposed. The excellent agreement between theoretical results and reported experimental values indicates that these trichalcogenides have their electrical properties ruled by native point defects, mainly antisites. The energy alignment between the trichalcogenides and usual photovoltaic substrates shows that these materials can be successfully applied to the construction of type-II staggered heterojunctions. A last analysis is done by considering only homo- and heterojunction of trichalcogenides, showing that these materials could lead to high-efficiency cells with broad spectral absorption and high conduction/valence band offsets.

Dimethyl sulfide (DMS) is the main organosulfur compound in the atmosphere. Oxidation by OH radicals to form the methyl thiomethyl species (MTMr) has been studied under normal atmospheric conditions and in reaction chambers at different O2 partial pressure, including complete absence of oxygen. Scarce attention has been devoted however to the possibility of further reaction of OH with MTMr. We present here a computational study using DFT, CCSD(T) and composite methods, on the properties of two stable intermediates never fully investigated before, methanesulfenyl methanol (MSMOH) and S-methyl-methanesulfenic acid (SMMSA), arising from addition of a second OH radical to MTMr.

Alzheimer’s disease (AD) is the most common form of dementia, affecting approximately 47 M people worldwide. Histological features and genetic risk factors, among other evidence, supported the amyloid hypothesis of the disease. This neuronocentric paradigm is currently undergoing a shift, considering evidence of the role of other cell types, such as microglia and astrocytes, in disease progression. Previously, we described a particular astrocyte subtype obtained from the 3xTg-AD murine model that displays neurotoxic properties in vitro. We continue here our exploratory analysis through the lens of metabolomics to identify potentially altered metabolites and biological pathways. Cell extracts from neurotoxic and control astrocytes were compared using high-resolution mass spectrometrybased metabolomics. Around 12 % of metabolic features demonstrated significant differences between neurotoxic and control astrocytes, including alterations in the key metabolite glutamate. Consistent with our previous transcriptomic study, the present results illustrate many homeostatic and regulatory functions of metabolites, suggesting that neurotoxic 3xTg-AD astrocytes exhibit alterations in the Krebs cycle as well as the prostaglandin pathway.This is the first metabolomic study performed in 3xTg-AD neurotoxic astrocytes. These results provide insight into metabolic alterations potentially associated with neurotoxicity and pathology progression in the 3xTg-AD mouse model and strengthen the therapeutic potential of astrocytes in AD. Biological significance: Our study is the first high-resolution metabolomic characterization of the novel neurotoxic 3xTg-AD astrocytes. We propose key metabolites and pathway alterations, as well as possible associations with gene expression alterations in the model. Our results are in line with recent hypotheses beyond the amyloid cascade, considering the involvement of several stress response cascades during the development of Alzheimer’s disease. This work could inspire other researchers to initiate similar studies in related models. Furthermore, this work illustrates a powerful workflow for metabolite annotation and selection that can be implemented in other studies.

Herein, we have employed first-principles calculations to investigate the interaction between XY dual-doped graphene (DDG) (X = AL, Si, P, S; Y = B, N, O) and sodium/potassium. The introduction of two dopants alters the adsorption energy (AE) of sodium and potassium with respect to perfect graphene by an average of 0.88 and 0.66 eV, respectively. The systems that display the strongest interactions with the two alkalies assayed are SN and SiB DDG. Although the adsorption energy of sodium on graphene is weaker in comparison to that of potassium, the introduction of these dopants significantly reduces this difference. In effect, in some cases, the AE-K and AE-NA differ by less than 0.05 eV. The protrusion of the 3p dopants out of the graphene plane creates a hole where sodium and potassium can easily be intercalated between two layers of dual-doped graphene. The interlayer distances are reduced by less than 0.4 Å after K intercalation, making the process very favorable. Finally, most importantly, this eminent rise in adsorption energies guarantees exceptional storage capacities at the cost of low doping concentration.

Four unusual terpenylated acylphloroglucinols were isolated from the diethyl ether extract of the scales and rhizomes of the fern <em>Dryopteris wallichiana</em> together with the known compounds albaspidins AA and AB, and filixic acids ABA and ABB. Structures of the isolated compounds were established by extensive spectroscopic analysis and their absolute configuration at C-14″ was determined by comparing their CD spectra with those simulated for the respective isomers. Pure acylphloroglucinols displayed moderate <em>in vitro</em> nematocidal activity against L4 stage larvae of <em>Nippostrongylus brasiliensis</em> (LD<sub>50</sub> = 22–121 μM).

The respiratory infection COVID-19 is caused by the severe acute respiratory syndrome coronavirus 2 (SARSCoV- 2), which continues to spread worldwide causing more than a million deaths yearly. Despite the rapid development of vaccines for prevention, a critical need remains for therapeutic agents to combat the established infection. With this goal in mind, we conducted a target- (SARS-CoV-2 chemotrypsin-like protease, MPro) and phenotypic-based screening of a subset of compounds from our in-house chemo-library. The selected compounds have been previously studied in different diseases’ models. About 45 % of the analyzed compounds displayed inhibitory activity against MPro in the low μM range. Additionally, 3 out of 26 tested compounds (11 %) against SARS-CoV-2 displayed true anti-viral activity and (SI > 2). One of them belongs to the chloroquine-heterocycle chemotype, which was previously reported to hold anti-SARS-CoV-2. The remaining two novel hits belong to the polyhedral‑boron system, metallacarborane and dicarba-closo-dodecaborane chemotypes, which have not been previously reported for this anti-viral activity. In addition to targeting MPro, the chloroquine- and the metallacarborane-derivative, but not the dicarba-closo-dodecaborane, also inhibited the papain-like protease of SARS-CoV-2.

A summary of the resource.
One hundred four bacterial strains mediating urinary tract infections in separate individuals from a Uruguayan community were isolated. Forty-six strains conferred a multidrug resistance phenotype. All 104 strains were examined for the presence of class 1, 2, and 3 integrons. Class 1 integrons were found in 21 isolates across four distinct bacterial genera. A large class 1 integron in a Klebsiella pneumoniae strain was fully sequenced and was 29,093 bp in length. This integron probably arose by homologous recombination since it was embedded in a hybrid Tn21-like transposon backbone which comprised a Tn5036-like tnp transposition module at the IRi integron end and a Tn21 mer module at the IRt integron end. The parent integron/transposon that contributed the Tn5036 module was not related to Tn1696 since the integron insertion points in the transposon backbones were 16 bases apart. Examination of the other 20 class 1 integron-containing strains revealed further evidence of genetic exchange. This included a strain that possessed a Tn5036 module at the IRt end but not at the IRi end and another that possessed a tnp module beyond IRi that was a hybrid of Tn21 and Tn5051 and that is presumed to have arisen by site-specific recombination. This study highlights the ability of different genetic elements to act cooperatively to spread and rearrange antibiotic resistance in a community.

A summary of the resource.
As the markets for functional foods increase, studying and understanding consumers’ perception of functional foods is essential to develop products that have a good consumer acceptance. The objective of this work was to provide a first insight into Uruguayan consumers’ perception of functional foods. A survey with 200 consumers was performed. The participants were not familiar with the term “functional food,” as only 37% related the term with the real concept behind it. However, the participants showed a positive attitude toward the functional foods concept. Preferred areas of action of functional foods at a generic level were not reflected in the ratings of perceived healthiness and willingness to try yogurt with different enrichments, probably because they did not associate the components with their health effect. These results suggest that the use of health claims would be essential to ensure the success of functional foods.

A summary of the resource.
Myrcianthes cisplatensis leaf oil, obtained by steam distillation, was analysed by GC– FID and GC– MS. Twenty-six components were identified in the oil (90% of the total composition); the enantiomeric dis- tribution of -pinene, -pinene, limonene, linalool, terpinen-4-ol and -terpineol was studied by multidimensional HRGC – HRGC. The major component was 1,8-cineole (54%). The enantiomeric purity for the . C / enantiomers for the monoterpenes studied was 96% for -pinene, 49% for -pinene, 100% for limonene, 94% for linalool, 50% for terpinen-4-ol and 64% for -terpineol.

Abstract
A summary of the resource.
The composition of 31 genuine Uruguayan lemon oils obtained by industrial processing (FMC on line) during the 1995 season is reported. The samples were representative of all production areas situated in the North and South of Uruguay. The volatile fraction was analysed by HRGC–FID and GC–MS; the enantiomeric distribution of β-pinene, sabinene, limonene, linalol, terpinen-4-ol and α-terpineol was studied by multidimensional HRGC–HRGC; coumarins and psoralens present in the non-volatile residue were analysed by normal phase HPLC. The results relative to the volatile fraction were compared with those obtained for Uruguayan oils produced in the 1992 and 1993 seasons and for Italian FMC oils. Coumarins and psoralens have been analysed for the first time in Uruguayan oils; their content is compared with that of Italian oils.

Abstract
A summary of the resource.
Hyptis floribunda leaf oil, obtained by hydrodistillation, was analyzed by GC and GC/MS. Forty-three components were identified in the oil. The main components were germacrene B (40.2%), β-caryophyllene (9.9%), germacrene D (9.8%) and α-copaene (8.0%).
