Levels of specific antibodies (Ab) and circulating antigens (CAg) were tested by ELISA in sera from 115 surgically confirmed hydatid patients, 41 individuals exhibiting other parasitic and unrelated diseases and 69 healthy subjects. Addition of CAg data to Ab detection in this sera collection increased sensitivity from 85% (only Ab) to 89% (Ab + CAg). Combination of ultrasonography with Ab and CAg serology for diagnosis of asymptomatic population in endemic areas was analyzed. One field survey (163 persons) involved both blood extraction and ultrasonography to all the population. Three people exhibited cyst images and all of them were Ab positive, while 6 Ab and 1 CAg positive individuals exhibited no cyst image. Another survey (1620 persons) involved a selection of 85 subjects for serology according to ultrasound data and record of family hydatid history. Twelve per cent exhibited no hydatid image being serologically positive and 14% were serologically negative but exhibited cyst image. Ultrasonography and serology (Ab and CAg) should be used in combination to maximize the diagnostic yield in asymptomatic population.

Debido a lo dificultosa de la determinación de compuestos polares (PC) por el método IUPAC 2.507 se comercializan dispositivos sencillos y de bajo costo como el Viscofrit, para determinaciones in situ en los locales elaboradores de alimentos fritos. Este dispositivo está calibrado para aceites y no para grasas, como la grasa vacuna, ya que el uso de estos materiales con alto punto de fusión es de uso poco frecuente a nivel mundial. Se observó una gran dispersión en las medidas realizadas con el Viscofrit a 48 °C sobre la grasa vacuna termoxidada. El máximo de tiempo de vaciado se alcanzó a unas 6 horas de termoxidación, tiempo al que la grasa aún no alcanza el nivel de PC máximo admitido. En este ensayo se observó una gran dispersión en los datos y un alto porcentaje de “valores falsos”, lo que pudo deberse a la presencia de sólidos grasos a la temperatura de medida que afectan el tiempo de vaciado. Con el fin de utilizar el Viscofrit para grasas, como la grasa vacuna, se sugiere realizar una calibración a una temperatura fuera del rango establecido por el fabricante que asegure la ausencia de sólidos grasos, en este caso 60 °C. En estas condiciones se obtuvo un tiempo máximo de vaciado para la grasa vacuna de 37 segundos.

Background: Bioavailability (F) and clearance (CL) are two pharmacokinetic parameters difficult to differentiate from simple plasma measurement when a drug is administered orally. Venous (V) / artery (A) concentration ratio of a drug could be a reliable index of its CL if measurements of plasma concentration were performed during a period of time where the absorption process was not longer operative, then during a pure elimination phase. Objective: A novel subrogate using two protocolized saliva samples sequentially collected (first, S1, and second, S2) was designed in order to replace V and A free plasma drug concentrations, respectively. Two drugs, phenytoin (PHT) and carbamazepine (CBZ), which are well-known for their inducer properties and their dose-dependent clearance variations, were studied taking into account the sex of individuals. Setting and patients: A multicentre two-phase collaborative study was done. The first phase was performed with healthy volunteers in order to determine salivary pharmacokinetic parameters after single dose administration. Twelve volunteers (6 male and 6 female) received 400 mg of CBZ (2 tablets x 200 mg, immediate release product). Twenty four volunteers (10 male and 14 female) received 100 mg of PHT. The second phase was carried out with epileptic patients under CBZ (11 male 15 female) or PHT (11 male and 11 female) monotherapy, in order to study dose-related and sex-related pharmacokinetic differences. Main outcome measures: In the single dose trials, peaks (Tmax, Cmax) were computed directly from the data. Areas under concentration-time curves (AUC∞), AUC∞xW (area corrected by weight) and half-lives (t1/2) were calculated. In the case of CBZ, AUCCBZ-10,11-epoxide/AUCCBZ metabolic ratios were also calculated. After multiple dose administration, S1 and S2 trough morning drug concentrations were measured. Results: Cmax and AUC differed significantly between sexes for the two drugs after single dose administration. Nevertheless, the apparent clearance (CL/F) per unit of body weight did not differ (CBZ) or slightly differed (PHT) between sexes. Higher metabolic ratio for CBZ in women would lead to lower F and therefore lower CL in this gender. In the case of PHT, women would have either lower F or higher CL than men. After multiple dose administration, S1/S2 saliva drug concentration ratio correlated positively with S2 for CBZ, showing that CBZ clearance increases with daily dose. Gender differences were also observed for CBZ-10,11-epoxide concentration, being bioavailability the main parameter responsible for this difference. S1/S2 saliva PHT concentration ratio correlated negatively with S2, showing that PHT clearance diminishes with dose as it has been previously reported. Since a significant difference was found for S1/S2 ratio between male and females, CL is the pharmacokinetic parameter influenced by gender in PHT disposition. Conclusion: S1/S2 saliva drug concentration ratio was sensitive enough for detecting systemic clearance changes. Both CBZ and PHT would modify their bioavailability and clearance by inducing efflux transporter throughout chronic treatments, from the first dose to multiple dose administration.

The Raigón aquifer is an important groundwater system in southern Uruguay. The increasing use of groundwater resources in the last decades has provoked changes in the concentration of many elements which are strongly related to anthropogenic pollution sources. Concentration levels are useful to detect changes in reservoir status but it is also necessary to analyze their chemical significance in order to make an accurate assessment of the sources of contamination and the causes of changes. In this work we use the available thermodynamic data to calculate chemical speciation on these groundwater samples. Trace elements present as anions, in particular Se and Mo, are especially focused to show the chemical modeling possibilities. Both elements form anionic species, predominantly MoO42- and SeO42-. Results show that these anions interact in solution and are greatly influenced by the concentration of the abundant calcium ion. Localized changes in pH can strongly affect the situation. The same is observed with the pE parameter, but only in the case of Se. Chemical speciation of trace elements is in general highly dependent on pH, pE and concentration of major elements. In consequence, for a fixed analytical total concentration, these parameters can markedly change the situation, affecting the mobility, the bioavailability and environmental fate of these elements. The strategy employed in this work can also be extended to the study of many other environmental water scenarios.

Reaction fl ow (RF) chromatography with fl uorescamine reagent and UV – vis detection was used for the analysis of amino acids. The performance advantage of RF chromatography was tested against the conventional post- column derivatisation (PCD) methods. The results of the study veri fi ed that greater sensitivity could be obtained using RF columns compared to conventional PCD methods. The reaction loops employed in the conventional PCD methods did not need to be used, which resulted in more ef fi cient separations using RF columns. The selective detection of amino acids in this RF study reduced limits of detection (LOD) and limits of quanti fi cation (LOQ) by about 50% compared to conventional methods of PCD. The calibration data of the RF-PCD technique also yielded R 2 ≥ 0.99 and %RSD in peak areas ranging from 3.0 – 8.5%. The RF method also enabled multiplexed detec- tion since both the derivatised and non-derivatised fl ow streams could be analysed simultaneously.

Text highlighting is a new method for online sensory-consumer research where participants read a text while making use of highlighting functions to indicate content that they, respectively, ‘like’ and ‘dislike.’ In a recent series of papers, consumers have been found to approach the text highlighting task in a logical and systematic manner, and the results have been shown to deliver attitudinal insights about the focal topic that fit expectations. Building on the early successes of text highlighting, it was an obvious step to try to extend it to applications in product research which occupies a central role in sensory-consumer research. A case study with kiwifruit was performed with consumers in Singapore and Malaysia (n = 1140). Participants read a text (288 words, 19 sentences) which primarily described sensory, nutrition and health characteristics of green and gold kiwifruit. Focusing on 43 specific fruit characteristics, sentiment (difference between the frequency of ‘like’ and ‘dislike’ highlights) was higher for nutrition and health characteristics than for those describing sensory characteristics. Only the Chinese origin of kiwifruit and the hairy skin of green kiwifruit were negatively perceived overall. A novel use of penalty/lift analysis to identify the specific fruit characteristics that significantly influenced ex pected liking of kiwifruit confirmed the results based on sentiment scores. Moreover, a difference between the drivers of liking for gold and green kiwifruit was identified; the use of ‘like’ highlighting for sensory properties positively contributed to expected liking for gold kiwifruit only. The case study also demonstrated the potential to meaningfully segment consumers based on their highlighting responses to product specific characteristics. The results are discussed in the context of future applications of text highlighting in product research.

While emotional associations to products are increasingly used alongside sensory product characterisation in consumer research, studies that link these product responses remain less common. Six studies (104–270 people per study) were conducted using diverse products (salted snacks, potato chips, yoghurt, cheese, snack bars and fruit; 5–10 samples per study). To indicate their emotional response during product tasting, consumers used the emotion circumplex which is a single-response questionnaire that spans the dimensions of valence (pleasure to displeasure) and arousal (activation to deactivation). Sensory characterisation was obtained using check-all-that-apply (CATA) questions. Through regression analysis, sensory drivers of emotional associations were established, with different linkages to valence and arousal being identified. Many study-specific linkages were found, but the main result pertained to generalised patterns in some linkages including positive associations between ‘sweet’ and “pleasure”, often combined with “pleasant activation”. For ‘sour’, positive linkages were established to “activation”, typically with, or without, “negative pleasure”. Low flavour intensity was positively linked to the emotion circumplex quadrant that connected “deactivation” to “displeasure.” In general, linkages had straightforward and meaningful interpretations, confirming suitability of the emotion circumplex for use in product-focused emotion research.

Twitter data is emerging as a source of insight regarding food-related consumer behaviour, and as such merit consideration regarding inclusion in the set of methodologies used by sensory and consumer researchers. Contributing to the evaluation of the pros/cons of Twitter as a research tool, we present a case study on the topic: “what people say when they tweet about different eating situations”. Rather than focusing on specific food/beverage products, we consider eating/drinking more widely, and hereby adopt a broader lens through which to evaluate this data gathering platform. Using the search words breakfast, lunch, dinner and snack, a total of 69,961 tweets were retrieved, of which 48,746 corresponded to original tweets and were subject to automated word analysis. However, manual content analysis of a subset of 16,000 tweets randomly selected provided more detailed insight. Mirroring results from past food choice research, information about what was consumed, when, where, with whom, and why was contained within the tweets, hereby confirming the potential of Twitter data as a source of information about food choice decisions. Despite access to free data, labour costs associated with data analysis were high and other limitations were also identified. While the content of tweets was largely free of written emotional expressions, emoticons were used with greater frequency. Some tweets also contained references to the (un)healthiness of foods/beverages being consumed and may be useful for exploring consumer perceptions of health/wellbeing, a topic which, like emotions, is currently attracting considerable attention. Overall, we find that Twitter data merit inclusion in the researcher’s toolbox, but that it is no panacea.

Abstract
A summary of the resource.
The wax hydrocarbon fractions of native Butia and Syagrus species collected from Palms in different regions of the of Rio Grande do Sul state (Brazil) and in Rocha (Uruguay) were analyzed to evaluate their potential as chemotaxonomic markers. The wax was extracted with chloroform and the resulting wax was fractionated by preparative TLC. The hydrocarbon fractions were analyzed by GC-MS. Statistical analyses were completed with the Statistica 5.0 program. The total crude wax yields averaged 0.31% w.w–1 dried leaves for Butia samples and 0.28% for Syagrus samples. The linear hydrocarbons represented on average 15% of the total waxes in the case of Butia samples and 13.7% in Syagrus samples. Hentriacontane and triacontane were the main components of all samples. The comparison of the means showed significant differences among Butia and Syagrus samples, and amongst Butia samples collected in different localities. In the case of the Syagrus collections no consistent groupings could be made. In the case of Butia samples the formation of three groupings could be observed, which were consistent with the species described for their geographical distribution. A summary of the resource.
The application of diagnostic evaluations in advanced courses of organic chemistry revealed a poor learning of some concepts presented in previous organic chemistry courses. Taking this into consideration and aiming at overcoming these difficulties, a new aula methodology was implanted in the first organic chemistry course. By using molecular modeling as a didactic tool our objective was to improve the student’s ability to visualize the tridimensional structure of molecules and correlate it with their reactivity. Also, the use of this interactive graphical methodology allows the students to solve problems (and not only simple exercises) and increases their capacity at interdisciplinary thinking. In addition, the students are exposed to new techniques used in current research in organic chemistry. This methodology was implanted in a so called ‘‘virtual laboratory’’. This experience was evaluated as very good. It is worth to mention the high receptivity and enhanced motivation found in the participants related to the didactic tool used, which offered a visual aid to facilitate the learning of different topics in the course.

Rice bran oil usually shows a deep green colour, whose bleaching is still a problem to be solved. In this research an Uruguayan montmorillonite, modified by different treatments, pillaring (P-120) and acid-activation (Act), is tested for bleaching rice bran oil and refined sunflower oil enriched with chlorophyll a. A commercial bleaching earth (Ts) was used as reference. For equal total amounts of the same clay, the results obtained depend upon the number of stages of the process of bleaching. High percentages of removal of chlorophyll a in rice bran oil were achieved when employing P-120 during the first stage and either Ts or Act in the second one.

Saliva and plasma profiles of Carbamazepine and Phenytoin were studied in healthy volunteers |b after the administration of a controlled-release tablet (Tegretol 400 mg) |b three conventional-release tablets (Comitoina Simple 100 mg) and a W/O emulsion of Phenytoin. Samples were analysed by High Performance Liquid Chromatography on reversed phase column C18 |b 5 micra |b 10 cm X 4 mm i.d.,mobil pha<se water/methanol/acetonitrile (50:45:5) |b flow rate 1 ml/min |b detection at 22onm. Salivary and plasma levels showed a lack of correlation |b evidenced through the variable saliva/plasma (S/P) ratios. The in vivo release process is well recorded by the S/P ratios |b which are higher during absorption periods. The technological development of pharmaceuticals destined for extavascular route |b can be evaluated in vivo by these S/P ratios. Constant S/P values related with the plasma unbound fraction of drug state absence of absorption process |b and allow the estimation of pharmacokinetic elimination parameters from time-concentration data.

Nuestro primer curso de Química orgánica, donde se presentan los principales grupos funcionales, sus propiedades fisicoquímicas y sus síntesis, poseía un contenido estrictamente teórico. Por este motivo, en muchas temáticas, especialmente propiedades físicas y propiedades ácido-base de los compuestos orgánicos y reacciones de reconocimientos de grupos funcionales los estudiantes no conseguían traspasar la barrera del aprendizaje significativo. Por eso, como forma de complementar y fortalecer la instancia de adquisición de conocimientos, decidimos implementar una serie de seminarios experimentales sencillos y atractivos. La asistencia a los mismos y la realización de pequeñas pruebas individuales y experimentales fueron incluidas como exigencia para aprobar el curso. Tanto los resultados de las pruebas experimentales como los de las pruebas teóricas indicaron una mejoría en el aprendizaje significativo de la temática respecto a años anteriores. Por otro lado, el grado de satisfacción y aceptación por parte de los estudiantes de este tipo de sistema fue muy alto.

La selenocisteína (Sec) es incorporada a la cadena polipeptídica mediante un mecanismo no canónico: una secuencia de incorporación de Sec (SECIS) presente en el ARNm de la selenoproteína reprograma al codón UGA para incorporar Sec. Si bien este mecanismo ha sido completamente elucidado en bacterias, en eucariotas aún restan aspectos por comprender. La proteína de unión al elemento SECIS (SBP2) media la incorporación de Sec, sin embargo los roles que cumplen sus dominios y el impacto de su deficiencia en la inserción de Sec no se conoce completamente. Por otra parte, existen genes que se han reportado como involucrados o presuntamente involucrados en la decodificación e incorporación de Sec cuyas funciones no han sido esclarecidas. Para estudiar algunos de estos aspectos, utilizamos Caenorhabditis elegans como modelo ya que se trata de un organismo eucariota simple que posee toda la maquinaria de incorporación de Sec a disposición de una única selenoproteína: la tiorredoxina reductasa citosólica (TRXR-1). Todas las SBP2 descritas hasta el inicio de esta tesis poseían un dominio de unión al ARN (RBD) y uno de incorporación de Sec (SID). Mediante análisis in silico identificamos la SBP2 de C. elegans y, sorprendentemente, encontramos que carece de SID y que consiste únicamente en un RBD. Generamos una estirpe mutante en sbp2 que resultó incapaz de incorporar Sec, demostrando así que este gen es esencial para este fin. El análisis de genomas de otros nematodos reveló que la ausencia de SID es característica del linaje y que el mantenimiento de la incorporación de Sec se encuentra ligado a la TRXR-1. Se deduce entonces que el mecanismo de incorporación de Sec en este filo difiere, al menos en parte, del ya reportado para otros eucariotas; SID podría ser completamente prescindible, u otra proteína, no identificada aún, podría suplir sus funciones. Encontramos también que los nematodos del clado Tylenchina han perdido la capacidad de incorporar Sec. Estos son los primeros animales no artrópodos que se reportan que perdieron esta característica; a nivel genético perdieron todos los genes involucrados en su decodificación, excepto SecP43, que se presume tendría un rol adicional a la metilación de una base del anticodón del ARNtSec y el transporte de complejos supramoleculares asociados a los ARNm de selenoproteínas. Mediante ensayos con una estirpe mutante en SecP43 encontramos que si bien no este gen no es esencial para la incorporación de Sec en condiciones óptimas de laboratorio, es necesario para la correcta expresión de TRXR-1 en condiciones desafiantes. 4 Análogamente, demostramos que R13A5.9 , un gen asociado a la incorporación de Sec, pero de función desconocida, es requerido para la incorporación de Sec en condiciones desafiantes. Así, es probable que R13A5.9, como SecP43, tenga funciones regulatorias en el metabolismo de selenio y/o la incorporación de Sec. Globalmente, nuestros resultados indican que C. elegans y el linaje nematodo proveen indicios claves para comprender el mecanismo de incorporación de Sec y la evolución de la utilización de Sec y de los genes asociados a su incorporación, así como de las selenoproteínas y los selenoproteomas. Finalmente nuestros estudios mostraron evidencia de inicio no canónico de la traducción en C. elegans , mecanismo que no había sido previamente descrito en este organismo modelo. Se identificaron dos genes (uno de ellos sbp2 ) cuya traducción no iniciaría en AUG sino en AUU. Se prevé que el número de transcriptos que tengan inicio no canónico de la traducción sea aún mayor.

Abstract

Las enzimas catalizan un importante número de reacciones, a menudo con regio o estero selectividad muy precisa, hacienda posible una variedad de procesos catalíticos que no serían posibles o son impracticables por síntesis convencional. El uso aplicado de enzimas en biocatálisis se afianza año a año con la aparición de nuevos y mejores biocatalizadores que facilitan la producción de moléculas de interés biotecnológico. Cada vez son más comunes los procesos en los que se combinan varias enzimas actuando secuencialmente. El uso de sistemas enzimáticos acoplados puede mejorar la cinética de las reacciones, disminuir la pérdida de intermediarios o evitar la inactivación o inhibición de una de las enzimas del sistema por eliminación rápida de algún intermediario inactivante. Sin embargo, la naturaleza soluble de las enzimas presenta restricciones específicas para su uso aplicado: pobre estabilidad, dificultad de separación, contaminación de productos y reuso limitado. La inmovilización de enzimas es usada comúnmente para mejorar estas limitaciones ya que muchas veces provee efectos estabilizantes, facilita su separación del medio de reacción y su reuso. En este trabajo de Maestría hemos abordado el estudio de sistemas enzimáticos que acoplados funcionan para producir moléculas de interés industrial. Nuestros estudios incluyeron la inmovilización de todas las enzimas de las vías sintéticas seleccionadas, caracterización y utilización in vitro. Muchos de los biocatalizadores utilizados debieron ser expresados de manera recombinante y purificados parcialmente previamente a su utilización. Las estrategias de inmovilización utilizadas incluyeron interacción covalente y atrapamiento físico. Además de utilizar agarosa como soporte de inmovilización se planteó el uso de un nuevo soporte para la unión de enzimas: las nanopartículas de sílica biomimética. Para mejorar las propiedades de este soporte para la unión de proteínas hemos propuesto una estrategia de síntesis novedosa que incluye moldes proteicos y genera nanopartículas más pequeñas y más homogéneas en tamaño. Para todas las enzimas con las que se trabajó se lograron preparar inmovilizados activos, en algunos casos muy estables, alcanzándose factores de estabilización sin precedentes en la literatura. Nuestros estudios permitieron el acoplamiento de 5 enzimas que en una secuencia no natural funcionaron en la producción enantioselectiva del L-ácido láctico a partir de mezclas racémicas de Alanina. Además, la co-inmovilización en un mismo soporte de dos enzimas que en la naturaleza funcionan secuencialmente en la vía de síntesis del antibiótico butirosina B, permitió la producción del antibiótico in vitro a partir de un Tesis de Maestría en Química Lic. Erienne Jackson sustrato alternativo que debimos sintetizar químicamente. Asimismo, hemos sentado las bases para la inmovilización de uno de los sistemas enzimáticos más complejos de la naturaleza, las poliquétido sintasas demostrando mediante el seguimiento de actividades parciales su estabilización frente al complejo no inmovilizado. En todos estos sistemas hemos demostrado que un diseño adecuado de los procesos de inmovilización facilita el acoplamiento de múltiples enzimas trabajando en tándem para la síntesis in vitro de moléculas de interés biotecnológico.
