Warning labels have been implemented in different countries around the globe, including Brazil, to encourage healthier food choices. The aims of the present study were to evaluate the relative effect of different warning labels, brands and nutrition marketing claims on the food choice after the implementation of the Brazilian frontof package nutrition labelling regulation. Packages of four categories were designed considering the following factors: brand (familiar vs. unfamiliar), nutritional warning (present or absent), textual nutrition marketing claims (present or absent) and visual nutritional marketing claims (present or absent). A total of 850 Brazilian adults were recruited by a marketing agency. Participants were randomly allocated to one of two experimental conditions: black magnifier (n = 403) and black octagon (n = 447). They completed a choice task involving two out of four product categories: chocolate flavored milk, cookies, granola, and iced tea. The packaging was designed considering four factors: The presence of warning labels encouraged product choice in three of the four categories. However, brand and textual nutrition marketing claims had a larger effect. These results suggest the need to introduce comprehensive packaging regulations to reduce the prevalence of nutrition marketing claims on unhealthy products, as well as communication campaigns to raise awareness of the benefits of taking into account warning labels when making food purchases.

Salto district, a key horticultural area in Uruguay, has crucial water sources such as the Guarani Aquifer System, the Salto-Arapey Aquifer System (SAAS), and the San Antonio Creek (SAC). Agricultural and livestock activities have increased in recent years, leading to demographic growth and housing expansion. Concerns about water quality arise due to limited studies on environmental exposure to harmful chemicals and microbiological factors from agriculture, inadequate wastewater treatment, and septic tank seepage in rural areas. The present study collected hydrochemical background and new sampling data for hydrogeochemical and water quality evaluation (28 parameters) from SAAS (73 wells) and SAC (6 sites). Results indicated groundwater from SAAS is mainly sodium bicarbonate type, RAS index shows low saline waters and microbiological contamination were detected in half of the sampled wells, As levels (4.0–49.3 μgL−1), total phosphorus (P) (50–178.5 mg/L), and nitrate (NO− 3 ) (7.0–168.5 mg/L). SAC hydrochemistry revealed high levels of P (77.5–163.5 μg/L), which could be one of the factors that promote the appearance of signs of eutrophication towards the mouth of the Uruguay River. Spatial distribution of total arsenic (As) in SAAS and water quality evaluation of SAC assess future human health risks. These results represent a challenge to foster Uruguayan water education and raise public awareness about the importance of using safe water for human consumption and irrigation.

Sturgeon aquaculture faces challenges due to global warming, as chronic heat stress (CHS) jeopardises fish health and production sustainability. To tackle this issue, we analysed serum innate components and the cellular composition of the peritoneum in Russian sturgeons (Acipenser gueldenstaedtii) subjected to CHS and challenged with Aeromonas hydrophila. Russian sturgeons exposed to CHS exhibited a reduction in the serum levels of total protein and alkaline phosphatase activity, trending to a higher decrease in sturgeons exposed to both bacterial challenge and CHS. This sturgeon group showed a worsened health status, reduced growth and a higher mortality rate. Under tolerable temperatures, the bacterial challenge increased serum ceruloplasmin, lysozyme and serum amyloid A levels, which tended to be lower under CHS. The naive peritoneal cellular composition of the Russian sturgeon revealed a predominance of myeloid cells, with lymphoid cells present in a smaller proportion. CHS altered this distribution, increasing lymphoid while reducing myeloid cell percentages and A. hydrophila challenge reverted these alterations. Myeloid cells exhibiting the highest phagocytic potential were reduced by CHS, potentially compromising peritoneal bacterial clearance. Principal component analysis revealed that sturgeons’ integral innate response varied according to temperature exposure, evidencing CHS’s impact on the antibacterial response. Overall, this study provides a first picture of the peritoneal cellular composition of a chondrostean fish. Furthermore, it demonstrates the disruption caused by CHS on the sturgeon’s early systemic and peritoneal responses mounted against A. hydrophila.

Background: Prenylated flavanones represent a structurally diverse class of natural compounds with significant biological potential. Among them, chromene flavanones (CFs) constitute a rare and specialized subgroup with promising therapeutic applications. These molecules have gained attention due to their potential to inhibit xanthine oxidase (XO), a key enzyme involved in oxidative stress-related disorders such as gout and hyperuricemia. Their distinctive structural features, combined with notable bioactivity, make them compelling candidates for further pharmacological exploration. Given their potential relevance, this study focuses on the in vitro and in silico evaluation of three CFs isolated from Dalea boliviana Britton [Fabacea], assessing their capacity to inhibit XO and elucidating key structure–activity relationships (SARs) that contribute to their biological effectiveness. Purpose: This study aims to investigate the in vitro and in silico interactions of the chromene flavanones, namely, (2S) 5,2′-dihydroxy-6″,6″-dimethylchromeno- (7,8:2″,3″)-flavanone (1), (2S) 5,2′-dihydroxy-6″,6″-dimethylchromeno-(7,8: 2″,3″)-3′-prenylflavanone (2), and obovatin (3), obtained from D. boliviana, with XO, in order to explore their potential as XO inhibitors and their potential therapeutic applications for hyperuricemic diseases. Material and Methods: XO inhibition by the three chromene flavanones was measured spectroscopically. The relationships between their structures and inhibitory activities were evaluated. Moreover, molecular docking studies were performed to propose the binding modes of the most active natural compounds. Results and discussion: Compounds 1 and 2 exhibited potent inhibition, with IC50 values in the nanomolar range (0.5 ± 0.01 nM and 1.7 ± 0.46 nM, respectively), demonstrating significantly higher activity than allopurinol (AL), the reference inhibitor (IC50 = 247 ± 4 nM). In contrast, compound 3 displayed only weak inhibition. SAR analysis revealed that the presence of a chromene moiety in the A-ring, combined with hydroxyl and prenyl groups in the B-ring, played a crucial role in enhancing inhibitory activity. Molecular docking studies confirmed the strong binding affinities of compounds 1 and 2 within the active site of XO (PDB ID: 3NVY), with binding energies of −6.1687 kcal/mol and −6.7820 kcal/mol, respectively. Key stabilizing interactions involved π–π interactions with Phe914 and hydrogen bonding with residues such as Leu873 and Leu1014. These findings highlight the structural features essential for potent XO inhibition and suggest that chromene flavanones represent a valuable scaffold for the development of novel inhibitors. Further molecular dynamics simulations could provide deeper insights into their stability and interaction dynamics, aiding in the rational design of more effective XO inhibitors. Conclusion: Our findings lead us to propose these chromene flavanones as lead compounds for the design and development of novel XO inhibitors for treating diseases in which exacerbated activity of this enzyme is involved.

Although fruits and vegetables are an essential component of a healthy diet from sustainable food systems, their consumption remains below recommended levels worldwide. In this context, the aim of the present work was to explore factors influencing fruit and vegetable consumption among Uruguayan adults. A telephone survey with 505 adults, living in Montevideo and involved in food purchasing for the household, was conducted by a specialized company in May 2024. Participants were asked to indicate the number of days they consumed fruit in a typical week and the number of servings they ate in each of those days. They were also asked to indicate whether they regarded their fruit and vegetable consumption as sufficient or not. Those answering negatively were asked to indicate the reasons underlying their insufficient consumption. Participants also answered a series of socio-demographic questions. Results showed that only 12.4% of the participants consumed at least 5 servings of fruit and vegetables per day. Gender and socio-economic status were identified as the main socio-demographic predictors of fruit and vegetable consumption. Fruit and vegetable intake was frequently perceived as sufficient, even if it was below national recommendations. A wide range of factors emerged as motives underlying insufficient consumption of fruits and vegetables, including lack of habits, cost, dislike, and perceived difficulty for preparing or cooking vegetables. These results stress the need to implement multicomponent strategies to address deficiencies in fruit and vegetable consumption in the country.

Monitoring economic access to food is fundamental to implement strategies to reduce food insecurity and enhance the affordability of healthy diets. The aims of the present study were to develop a healthy food basket (HFB) and analyze its cost from 2011 to 2024, in Uruguay, a highincome Latin American country. The HFB was conceptualized as a set of foods that allows the achievement of the nutritional needs of a person at the lowest possible cost, considering the dietary patterns of the local population. Its construction was based on national and international dietary intake recommendations, local food acquisition patterns, and food prices. The daily cost of the HFB in May 2024 was USD 7.5, considering purchasing power parity (PPP) conversion factors. The cost of the basket was higher than the Cost and Affordability of a Healthy Diet elaborated by FAO for the country and the region and higher than the cost of the official food baskets of the country. The cost of the HFB increased 202.3% from January 2011 to May 2024, while inflation increased by 178.4%. Fruits and vegetables, meat, dairy products, and processed cereals made the largest contribution to the cost of the HFB. Results highlight the need to reassess the country’s poverty estimation criteria and implement strategies to improve the affordability of healthy foods, such as adjusting cash transfer amounts and introducing targeted subsidies. The proposed methodological approach can contribute to the standardization of approaches to measure and monitor the cost of healthy diets in other countries.

A summary of the resource.
This research was motivated by the sustainable food transition and focused on plant-based (PB) alternatives to yoghurt, cheese, eggs and fish (salmon). An online study was conducted with 1667 consumers from the United Kingdom who were omnivores. Among participants, 24 % were restricted omnivores (or flexitarians) and made some efforts to reduce their animal food intake. Written names were used as product stimuli and varied according to protein type (PB or animal-based). Compared with animal foods, the findings revealed a strong negative perception of PB variants that spanned the four product categories and seven response variables – overall appeal, healthy, affordable, good for the environment, tasty, natural, and versatile. Further, it extended to behavioural intention, where willingness to eat the PB variants was much lower than for the animal-based counterparts. The effect of nutrition and/or ingredient information on product perceptions and behavioural intent was examined, but no major effect was found. When participants were segmented according to their stated willingness to transition to a vegetarian diet, those with a higher willingness (~1/3 of the total sample) perceived the PB variants more positively than those with lower transition willingness. This latter group (19 % of participants) also showed a significantly higher willingness to eat the PB food alternatives. Collectively, the findings contribute to the accumulating evidence that PB alternatives to animal foods may face many barriers to uptake among consumers who are currently not open to major dietary changes towards sustainability.

Nutrition plays a crucial role in maintaining health and quality of life in old age. Therefore, strategies aimed at improving diet quality are highly relevant. The objective of this work was to evaluate the relative effect of frontof- package warning labels on the food choices of consumers over 60 years of age living in Brazil. A total of 1125 participants over 60 years old were recruited using an Instagram advertisement. They were randomly allocated to four experimental groups, defined considering two 2-level variables: type of warning label (black magnifier vs. black octagon) and information about warning labels (with information vs. no information). Participants in each of the four groups completed a choice-conjoint task involving pairs of product categories (cream cheese, juice, toasts, and yogurt) differing in four variables: commercial “Brand” (familiar vs. unfamiliar), “Nutrition claims” (present vs. absent), “Sensory claims” (present vs. absent) and “Warning label” (present vs. absent). Data were analyzed using mixed logit models. Results revealed a significant effect of type of warning label in some categories, with the magnifier being more effective than the octagon in discouraging unhealthy food choices. However, “Brand” was the most relevant variable and, in some categories, “Nutrition claims” outweighed the effect of “Warning labels”. Exposure to information about the warnings prior to the choice-task reduced the importance of the “Brand” and tended to increase the effect of the “Warnings labels” and “Nutrition claims”. These results highlight the need to implement strategies to promote the use of warnings among older consumers.

Abstract

The transformation of the first-food systems and the widespread availability and marketing of breastmilk substitutes undermine efforts to promote adequate breastfeeding practices. The objective of the present research was to explore mothers’ perspectives on infant formula use in Montevideo, the capital of Uruguay. A generic qualitative design was used to explore mothers’ perspectives on the factors that act as enablers or barriers for infant formula use. Semi-structured interviews with thirty-four mothers of 0–23 months old children, aged between 25- and 40-years old, were conducted. The interview transcripts were analyzed using content analysis based on inductive-deductive coding considering the framework of the Capability, Opportunity, Motivation, and Behavior (COM-B) model. The discourse of the interviewees enabled the identification of a wide range of barriers and enablers for the use of infant formula, which were related to the three components of the model: capabilities, opportunity, and motivation. The health system emerged as the key determinant of infant formula use. Most of the participants who used infant formula referred to medical indication when explaining the reasons underlying their infant feeding decisions. The working status of the mother and emotional aspects of the child feeding experience were also identified as key enablers of infant formula use, whereas knowledge about health benefits and breastfeeding techniques, support from families and health-professionals, and motivation to breastfeed were key barriers. Taken together, these results suggest that strategies to reduce the use of infant formula in Uruguay should mainly focus on providing opportunities to breastfeed by improving the quality of the support and guidance provided to families in the health system and facilitating greater balance between breastfeeding and work.

Natural products play a crucial role in drug discovery, primarily due to their structural complexity. The prenylated flavanone glabranin ((S)-5,7-dihydroxy-8-(3-methylbut- 2-en-1-yl)-2-phenylchroman-4-one), isolated from the Dalea elegans plant species, has demonstrated neuroprotective effects, attributed to its inhibition of GSK-3β, as per our previous in silico studies. Given the enzyme’s diverse functions and its potential as a target for neurodegenerative diseases, our group synthesized and evaluated an 11C-labeled derivative of glabranin. We present its in vitro biological activity, including IC50, neuronal uptake in Alzheimer’s-affected brain regions, key physicochemical properties, and animal studies. This study confirms [11C]FLA’s interaction with GSK-3β in vitro, highlighting the potential of radiotracers in bioactive compound research
