Noribogaine (Noribo) is the primary metabolite from ibogaine, an atypical psychedelic. The main objective of this study was to evaluate the effect of Noribo on thalamic physiology of calcium channels voltage activation and gene expression and the potential involvement of the 5-HT2A receptor on those effects. Thus, we used male and female, 5-HT2AR knockout (KO) and wild type (WT) mice, injected with a single Noribo dose (10 or 40 mg/kg; i. p.) and measured thalamic mRNA expression of T-type and P/Q-type calcium channels, as well as HCN2. For CACNA1g (T- type) we found that Noribo 10 increased gene expression of this channel; however, this effect was observed only in WT males and KO females. This result suggests that the effect of Noribo 10 on gene expression is both receptor- and sex-dependent. For CACNA1a (P/Q type), we found that Noribo 10 increased the mRNA expression in both sexes. For HCN2, we found that Noribo 10 decreased the mRNA expression in WT and KO females but only in KO males. We also investigated calcium channel current density by patch-clamp recordings of thalamocortical ventrobasal neurons after bath application of Noribo (50 μM) in slices obtained from WT and KO 5-HT2AR mice, male and female. We found that bath-applied Noribo blocked T-type calcium current density only in KO females but not in WT. However, in males no blocking effect of Noribo was observed for both genotypes. Also, calcium current density values appeared to be drastically increased in ventrobasal neurons from female mice, another indication of sex-dependent differences. BACKGROUND: Piezodorus guildinii (Westwood, Hemiptera: Pentatomidae) causes economic losses in soybean crops by diminishing productivity, impacting grain size, oil content, and germination power. RNA interference (RNAi), a conserved gene regulation process initiated by double-stranded RNA (dsRNA), may be used as a safe technique in integrated pest management strategies. Conjugating dsRNA with cationic polymers, which protect dsRNA from degradation has shown to enhance this response. This study aims to assess the impact of dsRNA administration on the viability of P. guildinii and evaluate dsRNAchitosan nanocomplexes as a pest control strategy. RESULTS: Ten target genes were selected, dsRNAs were designed and synthesized. In injection assays, significant mortality exceeding 76% was observed, while mortality from ingestion also showed significant differences, reaching 49%. The dsRNA targeting Srp54k gene was selected and a chitosan-dsRNA nanocomplex was synthesized. These nanocomplexes demonstrated enhanced stability under RNase treatment in vitro, and, in in vivo assays, a cumulative corrected mortality rate of 66% was reported. Consistent with these findings, RT-qPCR analysis revealed that gene expression levels in treated insects were significantly reduced compared to control at 24, 48, and 72 h post treatment for targeted genes. CONCLUSION: In this work, we evaluated the effect of dsRNA administration by injection and ingestion on the viability of P. guildinii and determined its molecular silencing effects. Additionally, we synthesized and characterized chitosan-dsRNA nanoparticles, which demonstrated improved efficacy in terms of stability. These findings are promising for the development of new integrated management strategies for stink bugs control. Chagas disease and Leishmaniasis, caused by Trypanosoma cruzi and Leishmania spp., respectively, are highly prevalent neglected tropical diseases (NTDs) that pose significant global health challenges. In our pursuit of effective vanadium-based therapeutics against these diseases, we previously developed several series of oxidovanadium( V) complexes featuring bidentate bioactive ligands and Schiff base tridentate ligands. The current study extends our previous research by incorporating in the same molecule, a tridentate bromo-substituted isonicotinyl hydrazone Schiff base ligand, BrIS, and a 8-hydroxyquinoline derivative (L), leading to the synthesis and comprehensive characterization of five new complexes, [VVO(BrIS-2H)(L-H)]. Most of new complexes exhibited activity in the micromolar range against the infective trypomastigote form of T. cruzi (EC50, 24h: 0.73–7.95 μM) and against L. infantum promastigotes (IC50, 5 days: 1.14–1.16 μM) and some of them showed good selectivity indexes towards the parasites (SI up to 52). Notably, the vanadium uptake by the parasites was higher for the new [VVO(BrIS-2H)(L-H)] compounds compared to [VVO(IN-2H)(L-H)] analogues previously developed, where IN is the structurally related 2-hydroxy-1-naphtaldehyde isonicotinoylhydrazone ligand, with accumulation in the soluble cell fraction. High-dose incubations resulted in trypanocidal effects and suggested the generation of reactive oxygen species (ROS). Further analysis revealed that [VVO(BrIS-2H)(L-H)] complexes induced a higher percentage of apoptosis, whereas the [VVO(IN-2H)(L-H)] series was associated with autophagic cell death. These findings highlight the potential of the [VVO(BrIS-2H)(L-H)] series as promising anti-T. cruzi agents and underscore the need for further research to optimize their therapeutic efficacy and explore their mechanisms of action. Niobium-based oxides are being increasingly evaluated as materials for energy storage applications. Additionally, the use of these oxides as cathodes in aqueous electrolytes has shown promise. Based on this, the pseudocapacitive behavior of protonic niobate nanowires in an aqueous acidic electrolyte (1 M H2SO4) was evaluated for the first time. The material was obtained in two simple sequential steps. First, hydrothermal synthesis resulted in sodium niobate; second was ionic exchange (in two concentrations of 2 M and 0.1 M HNO3), where the protonic niobate was obtained. The resulting protonic niobate was characterized by FEG-SEM, the results demonstrated that the morphology of the oxide was concentration-dependent in the ionic exchange step, and EDS analysis was used to validate the procedure. Using DRX, Raman spectroscopy, and FTIR analysis, the transformation of sodium niobate to protonic niobate was evidenced. The electrochemical tests demonstrated that the protonic niobate presented pseudocapacitive behavior when employed as the cathode in 1 M H2SO4, and the ionic exchange in 2 M HNO3 promoted a better specific capacitance, reaching 119.8 mF·cm−2 at a 1 mA·cm−2 current density. Background: Nutritional warnings are one of several front-of-package labelling schemes implemented worldwide. Their adoption has often been complex and contested, particularly due to trade concerns and strong industry opposition. In this context, citizen support is considered critical. Understanding how people express views, motivations, and trust in institutions through everyday social talk can inform strategies to bridge opinion groups. Objective: To explore narratives about nutritional warnings on Twitter throughout the Uruguayan policy process. Methods: Tweets from Uruguay between May 2018 and April 2021 were retrieved using Synthesio®. Content analysis followed a deductive–inductive approach. First, references to the four elements of the Narrative Policy Framework (Setting, Characters, Plot, Moral) were identified. Then, inductive coding was used to generate categories within each element. Results: A total of 1013 tweets were retrieved, of which 752 related to the Uruguayan warning label policy. Most tweets were posted by regular citizens without explicit links to key stakeholders. Tweets commonly included narrative elements, particularly a character and a moral or policy solution. As the policy process advanced, narrative elements shifted: regulatory changes prompted higher engagement, stronger polarization, and increasingly divergent arguments. Conclusions: Findings highlight the value of examining public discourse on social media during the policy process. Understanding how nutritional warning labels are framed by citizens can help researchers and policymakers develop advocacy and communication strategies to shape opinion and support policy implementation in other countries. The classification of processed foods plays a pivotal role in shaping public health policies and research. While systems such as NOVA categorize foods based on processing level, they have been criticized for definitional ambiguity and for conflating technological, nutritional, and commercial dimensions. This study aimed to contribute to the ongoing debate by applying a data-driven, unsupervised clustering approach to a comprehensive database of 7284 packaged food and beverage products available in the Uruguayan market. Products were grouped based on nutritional composition and food additive disclosure using non-hierarchical clustering based on Gower distances. Nineteen distinct clusters were identified, each characterized by unique nutrient and additive profiles. Principal Component Analysis and heatmap visualizations revealed substantial heterogeneity in formulation both within and across traditional product categories and subcategories. Clusters differed markedly in their use of specific additives, such as acidity regulators, antioxidants, and humectants, demonstrating that additive combinations reflect distinct technological strategies. Notably, additive disclosure did not consistently align with nutrient profiles, reinforcing the multidimensional nature of food formulation. These results call for more granular classification approaches that move beyond prototypical products within categories and explicitly integrate both nutrient profiles and additive disclosure. Developing multidimensional indexes of food formulation could enhance risk assessment and clarify how specific formulation strategies relate to health outcomes, ultimately enabling more precise and effective public health nutrition policies. Different front-of-package labelling (FOPL) schemes have been developed worldwide to encourage informed and healthier food purchase decisions. However, few studies have formally compared different schemes, particularly in the Latin American context. This study aimed to assess the effects of four different FOPL schemes on the objective understanding of the nutritional content and intention to purchase products. This single-blinded multiarm randomised controlled trial was conducted using a face-to-face survey with adult shoppers, recruited at supermarkets in Costa Rica (n = 1350). Participants randomly assigned to intervention groups saw 12 mock-up products presented at random and balanced orders featuring one FOPL scheme or none: black octagonal warning labels (OWL), Nutri-Score (NUS), traffic-light labelling (TFL), guideline daily amounts (GDA), or no FOPL scheme (control group). Similar number of participants were analysed in each group: OWL (n = 264), NUS (n = 279), TFL (n = 263), GDA (n = 273), and control (n = 271). Compared to the control group, the odds for correctly identifying the least harmful option more often were three times higher in the OWL group (OR 3.08; 95% CI, 2.26–4.20), and 89%, 57% and 19% higher in the TFL (1.89; 95% CI, 1.40–2.56), the GDA (1.57; 95% CI, 1.16–2.21) and the NUS (1.19; 95% CI 0.89–1.60), respectively. OWL also was more efficacious in helping participants to correctly identify a product with excessive amounts of sugars, sodium, and/or saturated fats, as well as in encouraging the intention to purchase the least harmful or the intention to choose none of the options in the choice task. OWL performed best in helping shoppers to correctly identify when a product contained excessive amounts of nutrients to limit, to correctly identify the least harmful option, and to intend to purchase the least harmful option, more often. The proboscis extension reflex (PER) conditioning protocol is a widely used paradigm for studying learning and memory in honeybees. Accurate and consistent delivery of odor stimuli is essential for ensuring reproducible conditioning results, yet many existing systems require complex and expensive equipment. Here, we present a low-cost, fully documented Arduino-based system for automated volatile delivery during PER conditioning assays. The system integrates solenoid and servo valves, allowing controlled airflow between clean air and odorant channels, with stimulus timing managed by easily modifiable open-source code. Using this setup, we successfully conditioned Apis mellifera workers to associate 1-hexanol with a sucrose reward, achieving response rates comparable to those reported in previous studies. After three conditioning trials, 90% of honeybees exhibited conditioned responses, and 75% retained the association after five trials. This accessible and flexible system provides a reliable tool for researchers and educators aiming to implement classical conditioning protocols without requiring specialized technical expertise or infrastructure. Cannabis use during pregnancy has recently become an important concern due to its increasing trend and association with neurodevelopmental alterations of exposed children. Although inhalation is the primary route of cannabis consumption in humans, few studies replicated this route in preclinical models of prenatal exposure. This study aimed to analyze the effects of chronic prenatal exposure to vaporized cannabis (PEVC) on hippocampal neurodevelopment, functionality, and learning and memory processes. Using a commercially available high-potency cannabis strain (THC 14.7%), pregnant rats were exposed to cannabis vapor from gestational day 8 to 21. A combination of primary hippocampal cultures, electrophysiological studies in slices, and behavioral tests was employed to assess the impact of PEVC. We found that PEVC induced presynaptic remodeling of hippocampal neurons in offspring under both basal and activity-dependent conditions by increasing synaptic vGlut1 abundance and the total recycling pool of synaptic vesicles. Chronic PEVC also induced a downregulation of CB1R at glutamatergic and GABAergic synapses. Additionally, an increase in axonal recruitment and synaptic efficacy was observed at hippocampal CA1 synapses of juvenile offspring. Moreover, long-lasting cognitive impairments were identified during adolescence, including deficits in spatial memory in male and female offspring. In summary, these findings demonstrate that exposure to high-potency cannabis vapor during pregnancy leads to significant changes in synaptic remodeling and efficacy, axonal recruitment, and long-term cognitive consequences in offspring. Neurodegenerative diseases affect millions worldwide, posing significant challenges to public health. Oxidative stress and increased caspase-3 activity are key features in the development and progression of neurological disorders leading to neuronal damage and cell loss. As part of our ongoing efforts to identify new molecules with enhanced neuroprotective potential, we have synthesized six novel thiazole-nitrone hybrid compounds, designed as dual-action antiapoptotic and antioxidant agents. The nitrone moiety is known for its spin-trapping ability, which confers antioxidant and neuroprotective properties, while thiazole-containing compounds have been described as antioxidants and potent caspase-3 inhibitors. Our findings demonstrate that these hybrid compounds effectively reduce intracellular reactive oxygen species (ROS) levels and mitigate apoptosis rates in a hippocampal cell line by decreasing active caspase-3 activity, confirming their antioxidant and antiapoptotic properties. Notably, two of the six evaluated compounds exhibited the most promising neuroprotective profiles, including favorable blood-brain barrier permeability. Altogether, these results position the identified molecules as excellent candidates for further structure-based optimization, contributing to the development of more efficient therapeutic strategies against neurodegenerative disorders. Alzheimer’s disease (AD) is the most common form of dementia, affecting approximately 47 M people worldwide. Histological features and genetic risk factors, among other evidence, supported the amyloid hypothesis of the disease. This neuronocentric paradigm is currently undergoing a shift, considering evidence of the role of other cell types, such as microglia and astrocytes, in disease progression. Previously, we described a particular astrocyte subtype obtained from the 3xTg-AD murine model that displays neurotoxic properties in vitro. We continue here our exploratory analysis through the lens of metabolomics to identify potentially altered metabolites and biological pathways. Cell extracts from neurotoxic and control astrocytes were compared using high-resolution mass spectrometrybased metabolomics. Around 12 % of metabolic features demonstrated significant differences between neurotoxic and control astrocytes, including alterations in the key metabolite glutamate. Consistent with our previous transcriptomic study, the present results illustrate many homeostatic and regulatory functions of metabolites, suggesting that neurotoxic 3xTg-AD astrocytes exhibit alterations in the Krebs cycle as well as the prostaglandin pathway.This is the first metabolomic study performed in 3xTg-AD neurotoxic astrocytes. These results provide insight into metabolic alterations potentially associated with neurotoxicity and pathology progression in the 3xTg-AD mouse model and strengthen the therapeutic potential of astrocytes in AD. Biological significance: Our study is the first high-resolution metabolomic characterization of the novel neurotoxic 3xTg-AD astrocytes. We propose key metabolites and pathway alterations, as well as possible associations with gene expression alterations in the model. Our results are in line with recent hypotheses beyond the amyloid cascade, considering the involvement of several stress response cascades during the development of Alzheimer’s disease. This work could inspire other researchers to initiate similar studies in related models. Furthermore, this work illustrates a powerful workflow for metabolite annotation and selection that can be implemented in other studies. Heteronuclear zinc(II):mixed-lanthanide(III) metal organic frameworks (MOFs) bearing the formula [LnAx LnBy Zn3(oda)6(H2O)6]·nH2O (Ln = lanthanide(III) ion, LnB is smaller than LnA, x+y=2, n= 13 or 14) were obtained by direct reaction of the corresponding metal ion salts and oxydiacetate ( OOC-CH2-O-CH2-COO ) in aqueous solution, followed by slow solvent diffusion. This strategy led to the isolation of thirteen pure crystalline samples of isostructural hexagonal compounds (space group P6/mcc). Characterization by single-crystal X-ray diffraction was complemented by the analytical determination of the final LnB=LnA molar ratio by microwave induced plasma atomic emission spectrometry. Results show that the experimentally determined LnB=LnA molar ratio is systematically higher than that set out in the synthesis, especially for combinations of Ln that have larger size differences. Dual emission luminescence is observed for the compounds containing a combination of the more efficient visible-emitting Ln (Sm, Eu, Tb, Dy). The selected aliphatic ligand does not promote antenna effect. Thus, tunable color luminescence can be achieved by selectively targeting each Ln preferential excitation wavelength. Dual emission is also observed for CeEu mixed-MOF since the simultaneous excitation of both Ln is possible. Interestingly, GdEu mixed-MOF also shows dual emission when only Gd is directly excited. Title A name given to the resource <strong>Use of epilobium parviflorum in the treatment of benign prostatic hyperplasia in canines</strong> Abstract A summary of the resource. The objective of the study was to evaluate the effect of Epilobium parviflorum (EP) as a treatment for benign prostatic hyperplasia (BPH) in dogs based on two clinical case reports. Two dogs with BPH were treated for 2 months with EP extract. For both cases, follow-up was performed with B-mode ultrasound on Days 7, 15, 30 and 60 after starting treatment, and measurements of the length, width and height of the prostate were taken to calculate its volume (cm3). A complete blood count, liver and kidney biochemistry, urinalysis and serum testosterone concentration (ng/dL) were completed at the same time points. EP was well tolerated with no side effects observed during the 60-day oral administration period. In both cases, prostate volume decreased by 40% (Case 1: 81–50 cm3; Case 2: 79–44 cm3) by Day 60. Serum testosterone concentration decreased by 30% for Case 1 (749– 512 ng/dL) and 20% for Case 2 (220–188 ng/dL). This case report describes an alternative therapeutic method effective for treating canine BPH without apparent adverse reactions or complications. The respiratory infection COVID-19 is caused by the severe acute respiratory syndrome coronavirus 2 (SARSCoV- 2), which continues to spread worldwide causing more than a million deaths yearly. Despite the rapid development of vaccines for prevention, a critical need remains for therapeutic agents to combat the established infection. With this goal in mind, we conducted a target- (SARS-CoV-2 chemotrypsin-like protease, MPro) and phenotypic-based screening of a subset of compounds from our in-house chemo-library. The selected compounds have been previously studied in different diseases’ models. About 45 % of the analyzed compounds displayed inhibitory activity against MPro in the low μM range. Additionally, 3 out of 26 tested compounds (11 %) against SARS-CoV-2 displayed true anti-viral activity and (SI &gt; 2). One of them belongs to the chloroquine-heterocycle chemotype, which was previously reported to hold anti-SARS-CoV-2. The remaining two novel hits belong to the polyhedral‑boron system, metallacarborane and dicarba-closo-dodecaborane chemotypes, which have not been previously reported for this anti-viral activity. In addition to targeting MPro, the chloroquine- and the metallacarborane-derivative, but not the dicarba-closo-dodecaborane, also inhibited the papain-like protease of SARS-CoV-2. The objective of the study was to evaluate the effect of food education seminars on the introduction of bread with OPN flour added to public school meals in Brazil. A product-specific emoji list, facial hedonic scale, and word association (WA) were used. To evaluate the effects of the food education intervention, seminars were applied at only one of the schools. One school was selected as an intervention group (IG) (n = 398) and the other one as a control group (CG) (n = 441). The food education seminar revealed itself to be an essential strategy for willingness to try and acceptance of the bread. The product-specific emoji scale helped to understand children's emotions regarding the new food. The WA task brought a holistic insight into the evaluation of bread by children, confirming the hedonic data. The introduction of new food in school meals is positively influenced by food literacy, and all sensory evaluations corroborate this.
