A novel method was developed for the determination of zinc in wine by direct-injection electrothermal atomic absorption spectrometry (ETAAS). Unlike the OIV (Organisation Internationale de la Vigne et du Vin) reference method, which is based on flame atomic absorption spectrometry (FAAS), the proposed method does not require any sample preparation. Previous to the development of this method a reappraisal of the possible interferences found during the determination of zinc in wine by the reference OIV method was undertaken. Aiming at minimising sample preparation during FAAS method, alternatives such as the use of matched-matrix standards and the use of the Standard Additions Method (SAM) were studied, however different samples of Tannat wine produced in Uruguay exhibited widely different interference patterns, precluding the possible development of an alternate sample preparation-free FAAS method. Thus a new method based on direct-injection ETAAS was developed. The method is based on the atomic absorption of zinc at the alternate resonance wavelength of 307.6 nm. Samples are injected in the graphite furnace without preparation and the determination carried out by SAM. Quantification limit was 0.10 mg L− 1 and linearity was confirmed in the range 0.10–10 mg L− 1. Precision (repeatability, sr (%), n = 5) was calculated at three concentration levels and found to be better than 5%. When compared with the OIV reference method, results agreed within 10%. The method is fit for the purpose of routine analytical quality control, requiring no sample preparation and thus less labour and producing less chemical waste.

Mercury (Hg) is a global pollutant that is released into the environment from geological and anthropogenic sources and its compounds are highly mobile and toxic at trace level. A simple method based on cold vapour atomic absorption spectrometry (CV AAS) for the determination of total mercury in irrigation waters and extractable mercury in paddy soil was validated, using a low-cost sample introduction -1 device. The validated method presented a linear range from 0.2 to 2.0 µg L ; detection limits were 0.03 -1 -1 µg L and 0.012 mg kg (dry basis) for irrigation water and paddy soil respectively. Trueness was evaluated using spiked samples; the mean recovery was 101% (n=8) and 99.4% (n=6) for irrigation water and paddy soil respectively. Precision, expressed as relative standard deviation (RSD), was 9.6% (n=10) and 24% (n=8) for irrigation water and paddy soil respectively. These figures of merit were adequate for the determination of total Hg levels in irrigation waters and extractable Hg in paddy soils established by the regulations. To assess analytical performance several samples from the Eastern zone of Uruguay were analysed. Sample preparation consisted of an acid digestion using an economical pressurized reactor. Hg levels in waters and paddy soils were all below the maximum allowed and therefore were apt for agricultural use. These results are very important not only locally but also for the world since Uruguay is currently among the eight-largest paddy rice and milled rice exporter in the world. This method can be an alternative instead of the more expensive techniques mostly not available in developing countries and very easy to implement in environmental laboratories. Penicillin is one of the most widely used antibiotics to treat bacterial infections in clinical practice. The antibiotic undergoes degradation under physiological conditions to produce reactive compounds that in vivo bind self proteins. These conjugates might elicit an immune response and trigger allergic reactions challenging to di agnose due to the complex immunogenicity. Penicillin allergy delabeling initiatives are now part of antibiotic stewardship programs and include the use of invasive and risky in vivo tests. Instead, the in vitro quantification of specific IgE is highly useful to confirm immediate allergy to penicillins. However, discrepant results associated with the low sensitivity and accuracy of penicillin allergy in vitro tests have limited their routine diagnostic use for delabeling purposes. We aimed to develop a homologous chemiluminescence-based immunochemical method for the reliable determination of specific IgE to penicillin G, using unprecedented synthetic human-like standards. The synthetic standard targets the major antigenic determinant of penicillin G and the paratope of Omalizumab, acting as human-like specific IgE. It is a potent calibrator, highly stable, easy, and inexpensive to produce, overcoming the limitations of the pooled human serum preparations. The developed method achieved a good agreement and strong positive relationship, reaching a detection limit below 0.1 IU mL− 1 and excellent reproducibility (RSD <9%). The clinical sensitivity of the assay significantly increased (66%), doubling the accuracy of the reference method with an overall specificity of 100%. The new diagnostic strategy compares favorably with results obtained by the standard procedure, paving the way towards the standardization of penicillin allergy testing, and enhancing the detection sensitivity of specific IgE in serum to tackle reliably β-lactam allergy delabeling.

Background: The body mass index (BMI) ≥30 kg/m2 is the universally accepted cut-off point for defining obesity; however, its accuracy in classifying obesity in older adults is poorly understood. Objectives: To assess the performance of the BMI cut-off point ≥30 kg/m2 in classifying obesity in older adults, using the fat mass index (FMI) and fat mass percentage (FM%) as reference criteria; and to establish region- and sex-specific BMI-based cut-off points to classify obesity in older adults. Methods: The present study is a secondary analysis derived from a cross-sectional project that included a sample of 1463 older adults from ten Latin American and Caribbean countries. Volunteers underwent total body water measurements using the deuterium dilution technique to determine FMI and FM%. Accuracy of the BMI and derived cutoff points was assessed by the area under the receiver operating characteristic curve (AUC).Results: The BMI cut-off point ≥30 kg/m2 had low sensitivity for classifying obesity in these older adults compared to the FMI and FM%. The AUC values for the optimal BMI-derived cut-off points showed an acceptable-to-outstanding discriminatory capacity in diagnosing obesity defined by the FMI. There was also a better balance between sensitivity and specificity than with the values obtained by a BMI ≥30 kg/m2 in older subjects in both regions. Conclusion: The BMI cut-off point ≥30 kg/m2 had poor sensitivity for accurately diagnosing obesity in older adults from two regions. The region- and sex-specific BMI-derived cut-off points for defining obesity using the FMI are more accurate in classifying obesity in older men and women subjects from both regions.

First-principles density functional theory (DFT) calculations were performed to investigate the lithium (Li) adsorption upon beryllium (Be) doped graphene. Be acts as hole doping in graphene leaving the structure as electron deficient, offering a greater tendency for Li adsorption than in pristine and boron (B) doped graphene. The introduction of Be augments the adsorption energy of Li from −1.11 to −2.53 eV/Li. Furthermore, 12, and 16 Li ions can easily be captured by one Be center in the single and double vacancy case, respectively, with the adsorption energies of −1.33 eV/Li (for both the cases), showing that Be doped graphene is an excellent anode material for lithium ion batteries (LIBs). Consequently, the presence of structural defects, in particular, a divacancy is found to be more efficient in terms of Li storage capacity. A huge Li storage capacity (2303.295 mAh/g) is calculated for Li8BeC7 having reasonable adsorption energy (−1.47 eV/Li). Our calculated capacity is 6.19 times greater than that of the graphite.

Abstract
A summary of the resource.
Microorganisms were isolated from industrial wool scouring effluents and from the soil adjacent to the wastewater treatment lagoon, both sterols-rich environments, in order to search for novel biocatalysts able to transform cholesterol. The isolates were identified on the basis of morphological and biochemical characteristics and phylogenetic analysis. Furthermore, a rapid and accurate bacteria identification by matrix assisted laser desorption/ionization-time-of-flight mass spectrometry was carried out. Bacteria and fungi including representatives of the genera Fusarium, Talaromyces, Trichoderma, Mucor, Aspergillus, Citrobacter, Proteus, Klebsiella, Exiguobacterium, Acinetobacter, Tsukamurella, Bacillus, and Streptomyces were found and evaluated for their ability to biotransform cholesterol by whole-cell treatment system. The results show that a Trichoderma koningiopsis strain, as well as two strains of Mucor circinelloides were able to transform cholesterol into value-added products. The major products were characterized as 7β-hydroxycholesterol, 4-cholesten-3-one, 5α,6α-epoxycholestan-3β-ol and 5β,6β-epoxycholestan-3β-ol. To the best of our knowledge, the present study is the first report of cholesterol biotransformation by representatives of Trichoderma and Mucor genera

Abstract
A summary of the resource.
This paper deals with the role of dispersed lanthana as an active phase in several catalytic reactions: CO hydrogenation, CO oxidation, and oxidative dimerization of methane. Characterization of the prepared catalysts indicates that lanthana can be effectively dispersed on silica and on ceria. While in the case of silica-supported catalysts lanthana appears at the surface, leading to an almost full coverage for loadings higher than 40%, in the case of ceria-based systems, lanthana forms a solid solution with the support. In all the reactions studied, the presence of lanthana can be related to significant changes in the catalytic properties of the bare supports. Thus, the selectivity towards the total oxidation products observed on pure ceria is decreased, and the low activity shown by silica is enhanced. For the CO + H{sub 2} reaction, the addition of lanthana also generates upgraded products. (orig.).

A summary of the resource.
Flavonoids are an important group of recognized antioxidants ubiquitous in fruits, vegetables and herbs. There are epidemiological evidences for the stroke-protecting capacity of flavonoids and while the neuroprotective power of complex extracts rich in flavonoids like those of Ginkgo biloba, green tea or lyophilized red wine have been demonstrated in several studies, neuroprotection by individual flavonoids has been poorly studied in vivo. The neuroprotective capacity of individual flavonoids was studied in PC12 cells in culture and in a model of permanent focal ischemia (permanent Middle Cerebral Artery Occlusion - pMCAO). In the in vivo experiments, flavonoids were administered in lecithin preparations to facilitate the crossing of the blood brain barrier. The simultaneous incubation of PC12 cells with 200 μM hydrogen peroxide (H2O2) and different flavonoids for 30 min resulted in a conspicuous profile: quercetin, fisetin, luteolin and myricetin significantly increased cell survival while catechin, kaempherol and taxifolin did not. Quercetin was detected in brain tissue 30 min and 1 h after intraperitoneal administration. When one of the protective flavonoids (quercetin) and one of those that failed to increase PC12 cell survival (catechin) were assessed for their protective capacity in the pMCAO model, administered i.p. 30 min after vessel occlusion, quercetin significantly decreased the brain ischemic lesion while catechin did not. It is concluded that when administered in liposomal preparations, flavonoids structurally related to quercetin could become leads for the development of a new generation of molecules to be clinically effective in human brain ischemia

Interest in methodologies for sensory product characterization by consumers is increasing, and check-allthat- apply questions (CATA or checklists) have gained some popularity. This research studied bias pertaining to: (i) the order in which sensory attributes are placed within a CATA question, and (ii) the order of a sensory CATA question within an extended product assessment ballot (including product-elicited emotions, purchase intention and overall liking). In three studies including 335 consumers and using fresh fruit as samples, evidence of attribute order bias was established. In each study, two ballot versions were used in a between-subjects design. Primacy effects linked to attribute salience could explain some of the results. For example, differences in frequency of use of an attribute would be higher on the ballot version where it was placed nearer to the top of the list. However, this type of bias was not sufficient to explain all observed effects. It was found that a random ordering of sensory attributes in a CATA question reduced the total frequency of usage of terms compared with when attributes were grouped with similar terms (e.g. flavour/taste terms grouped together and texture terms grouped together). Some evidence was obtained to suggest that listing sensory attributes in the order that approximate the time when they would be perceived during the course of consuming the sample is necessary unless consumers are given explicit instructions to recall all sensory perceptions and evaluate attributes on the list from that point of reference. It was also found that conclusions regarding differences between samples depended on which ballot version was used. Few order effects were uncovered when the sensory CATA question was prior to or subsequent to other product evaluations (product-elicited emotions and purchase intention). There was no significant effect on hedonic scores of the tested products linked to the attribute order within the sensory CATA question. This research can help to inform best practices in the design of CATA questions for sensory product characterization

Objective The current study aimed to assess Uruguayan consumers’ accounts of their own need to change their dietary patterns, their intended changes and the barriers related to doing so, and to compare the intentions and barriers with the recommendations of the national dietary guidelines. Design An online survey with 2381 Uruguayan employed adults, aged between 18 and 65 years, 65 % females, was conducted. Participants had to answer two open-ended questions related to changes they could make in the foods they eat and/or the way in which they eat to improve the quality of their diet and the reasons why they had not implemented those changes yet. Content analysis using inductive coding by two researchers was used to analyse the responses. Results Consumers mainly intended to change consumption of types of foods, particularly eating more fruits, vegetables and legumes and consuming less flour, but also intended to alter their eating patterns. Lack of time and the fact that healthy foods are perceived as being more expensive than unhealthy foods were major barriers to behaviour change. Some of the recommendations of the dietary guidelines, particularly those related to enjoying cooking and meals and engaging in it as a social activity, were not represented in consumer accounts. Conclusions Accompanying policies to the dietary guidelines need to underline the importance of changes in dietary patterns, including greater enjoyment and sharing food preparation and meals in the company with others, address misconceptions about flour, and provide concrete, consumer-derived recommendations on how to enact the guidelines.

The extent of food waste raises concern about its effect on natural resource use, food security and future sustainability of the international food sector. It is expected that emerging countries with a growing middle class will exhibit a similar extent and pattern of consumer-related food waste, which is disturbing given that food waste coexists with food insecurity. However, the topic is yet under-researched, as are potential actions against food waste. Through an experimental survey, individual consumer characteristics explaining self-reported food waste at home and choice of price-reduced suboptimal food otherwise wasted in stores were explored among a sample of 1124 Uruguayan consumers. Results showed that food waste was higher among price and convenience-oriented, but lower for value conscious consumers. Choice of suboptimal food was more likely for price-oriented consumers and less likely for convenience-oriented ones. Gender and age explained wastage and choice of suboptimal food. Perceived norms did not play any role, but perceived quality of suboptimal food had a major effect. For packaged foods, value consciousness decreased suboptimal food choice likelihood, while convenience orientation decreased the latter for fresh foods. Consumer price orientation impacted food waste when related to eating food waste incidents more than to storage food waste incidents. The findings show that convenience orientation is a driver of food waste behaviours both when shopping and at home. However, findings imply that tackling food waste requires context-specific consumer-targeted strategies: food market stakeholders should appeal to value consciousness in the household, and perceived quality and price orientation in the store.

Metallo-β-lactamases (MBLs) hydrolyze almost all β-lactam antibiotics and are unaffected by clinically available β-lactamase inhibitors (βLIs). Active-site architecture divides MBLs into three classes (B1, B2, and B3), complicating development of βLIs effective against all enzymes. Bisthiazolidines (BTZs) are carboxylate-containing, bicyclic compounds, considered as penicillin analogs with an additional free thiol. Here, we show both l- and d-BTZ enantiomers are micromolar competitive βLIs of all MBL classes in vitro, with Kis of 6-15 µM or 36-84 µM for subclass B1 MBLs (IMP-1 and BcII, respectively), and 10-12 µM for the B3 enzyme L1. Against the B2 MBL Sfh-I, the l-BTZ enantiomers exhibit 100-fold lower Kis (0.26-0.36 µM) than d-BTZs (26-29 µM). Importantly, cell-based time-kill assays show BTZs restore β-lactam susceptibility of Escherichia coli-producing MBLs (IMP-1, Sfh-1, BcII, and GOB-18) and, significantly, an extensively drug-resistant Stenotrophomonas maltophilia clinical isolate expressing L1. BTZs therefore inhibit the full range of MBLs and potentiate β-lactam activity against producer pathogens. X-ray crystal structures reveal insights into diverse BTZ binding modes, varying with orientation of the carboxylate and thiol moieties. BTZs bind the di-zinc centers of B1 (IMP-1; BcII) and B3 (L1) MBLs via the free thiol, but orient differently depending upon stereochemistry. In contrast, the l-BTZ carboxylate dominates interactions with the monozinc B2 MBL Sfh-I, with the thiol uninvolved. d-BTZ complexes most closely resemble β-lactam binding to B1 MBLs, but feature an unprecedented disruption of the D120-zinc interaction. Cross-class MBL inhibition therefore arises from the unexpected versatility of BTZ binding.

A summary of the resource.
The enthalpy of formation of methanethial-S,S-dioxide (CH2SO2) has been determined by the use of isodesmic and nonisodesmic reactions, with individual enthalpies calculated by employing density functional methods and the CBS-QB3 model chemistry. After assessing the possible errors in the methodology, it was concluded that this value can be expressed as ¢fH°298(CH2SO2) ) -144.7 ( 8.4 kJ/mol.
